Traceability of primordial germ cells in three neotropical fish species aiming genetic conservation actions.

Primordial germ cells (PGCs) are embryonic pluripotent cells that can differentiate into spermatogonia and oogonia, and therefore, PGCs are a genetic source for germplasm conservation through cryobanking and the generation of germline chimeras. The knowledge of PGC migration routes is essential for transplantation studies. In this work, the mRNA synthesized from the ddx4 3'UTR sequence of Pseudopimelodus mangurus, in fusion with gfp or dsred, was microinjected into zygotes of three neotropical species (P. mangurus, Astyanax altiparanae, and Prochilodus lineatus) for PGC labeling. Visualization of labeled PGCs was achieved by fluorescence microscopy during embryonic development. In addition, ddx4 and dnd1 expressions were evaluated during embryonic development, larvae, and adult tissues of P. mangurus, to validate their use as a PGC marker. As a result, the effective identification of presumptive PGCs was obtained. DsRed-positive PGC of P. mangurus was observed in the hatching stage, GFP-positive PGC of A. altiparanae in the gastrula stage, and GFP-positive PGCs from P. lineatus were identified at the segmentation stage, with representative labeling percentages of 29% and 16% in A. altiparanae and P. lineatus, respectively. The expression of ddx4 and dnd1 of P. mangurus confirmed the specificity of these genes in germ cells. These results point to the functionality of the P. mangurus ddx4 3'UTR sequence as a PGC marker, demonstrating that PGC labeling was more efficient in A. altiparanae and P. lineatus. The procedures used to identify PGCs in P. mangurus consolidate the first step for generating germinal chimeras as a conservation action of P. mangurus.

Dnd is required for primordial germ cell specification in Oryzias celebensis

Dead end (dnd) is a germ plasm component that plays an essential role for primordial germ cell (PGC) development in vertebrates. Previously, we have found that dnd is the first fish PGC specifier in medaka. Here, we present an additional evidence that dnd is the determinant for PGC specification in Oryzias celebensis. In adult tissues, the O. celebensis dnd (Ocdnd) RNA shows germ cells specific expression in gonads. In the testis, Ocdnd RNA is strongly detected in spermatogonia and meiotic cells and gradually decreases during the spermatogenesis. In the ovary, Ocdnd RNA is present throughout oogenesis. In the embryos, Ocdnd RNA is maternally provided and asymmetrically localized to prominent particles of presumptive PGCs before gastrulation stage and restricted to PGCs subsequently. In addition, Ocdnd 3′ UTR can induce specific and stabilized GFP reporter expression in PGCs. Furthermore, knockdown of Ocdnd by morpholino (MO) injection abolishes the PGCs formation and this can be rescued by co-injection of medaka dnd (Oldnd) mRNA. More importantly, overexpression of Oldnd mRNA surprisingly boosts PGCs number. These results provide insights into function of dnd as a conserved specifier of PGCs in the genus Oryzias.

Presumptive primordial germ cells (pPGCs) were examined during migration from their deep endodermal position to the endodermal crest in Xenopus laevis, using light and electron microscopy with Epon sections, and several morphological characteristics of pPGCs, associated with their migration, were revealed. pPGCs displayed polymorphism, with smooth contours. The intercellular space around the PGCs was large and variable in width and cytoplasmic processes from pPGCs were occasionally observed in it. It was shown quantitatively that pPGCs at the migratory stage had a tendency to move with the leading end, towards which the nucleus was localized, dragging the germinal plasm behind. These polarized pPGCs were frequently associated with large intercellular spaces, both at their leading and trailing ends. Cytoplasmic processes of polarizing pPGCs found in the large intercellular space at the leading end were conspicuous. Ultrastructurally, the nuclei of pPGCs were euchromatic, and the nucleolus was prominent. The germinal plasm at the light microscope level corresponded to the cytoplasmic area near the nucleus where a large number of mitochondria with well-developed cristae and most of the other organelles were aggregated. Centrioles and centriole-associated microtubules observed in the aggregate were thought to be important structures responsible for the cell polarization mentioned above. It was demonstrated quantitatively that the size of mitochondria in pPGCs was larger on average than that of mitochondria in neighbouring somatic endodermal cells. Numerous irregularly shaped small yolk platelets characterized pPGCs. These ultrastructural features suggested that pPGCs were in an activated metabolic state. It was concluded that the migration of pPGCs was attributable to active movement with high cell metabolism, causing the formation of cell processes and intracellular polarization.

Dnd Is a Critical Specifier of Primordial Germ Cells in the Medaka Fish.

SummaryPrimordial germ cell (PGC) specification occurs early in development. PGC specifiers have been identified in Drosophila, mouse, and human but remained elusive in most animals. Here we identify the RNA-binding protein Dnd as a critical PGC specifier in the medaka fish (Oryzias latipes). Dnd depletion specifically abolished PGCs, and its overexpression boosted PGCs. We established a single-cell culture procedure enabling lineage tracing in vitro. We show that individual blastomeres from cleavage embryos at the 32- and 64-cell stages are capable of PGC production in culture. Importantly, Dnd overexpression increases PGCs via increasing PGC precursors. Strikingly, dnd RNA forms prominent particles that segregate asymmetrically. Dnd concentrates in germ plasm and stabilizes germ plasm RNA. Therefore, Dnd is a critical specifier of fish PGCs and utilizes particle partition as a previously unidentified mechanism for asymmetric segregation. These findings offer insights into PGC specification and manipulation in medaka as a lower vertebrate model.

Expression patterns of prdm1 during chicken embryonic and germline development

PRDM1 (PR domain containing 1) is a transcriptional repressor that has been identified in various species and is crucial for cell growth, differentiation and development. However, the expression pattern and role of PRDM1 in development has not been sufficiently established in birds. We therefore investigate the spatio-temporal expression of PRDM1 in various tissues, especially in the germline, during chicken development, providing the basis for functional study. Our results show that prdm1 mRNA was expressed in blastodermal cells (BCs) at stage X and in various tissues including the liver, skin, lung, kidney, eye, bursa of fabricius, spleen, proventriculus, gizzard, intestine, testis, ovary, tongue, feathers and thymus but was not or was only sparcely present in the heart, brain and skeletal muscle. The level of prdm1 mRNA was highest in the BCs among all tissues tested and significantly changed during development in many tissues, such as the blastoderm, bursa of fabricius, spleen, feathers and germline. Furthermore, the expression of the PRDM1 protein generally paralleled the mRNA results, except for in the gizzard. Immunohistochemistry also revealed that PRDM1 was localized in the smooth muscle. In addition, during germline development, PRDM1 was found to be continuously expressed in the presumptive primordial germ cells (PGCs) at stage X, the circulating PGCs in blood and the germ cells in the gonads from embryonic day 6 to adult in both males and females. Conserved Mechanisms for Germ Cell-Specific Localization of nanos3 Transcripts in Teleost Species with Aquaculture Significance

The importance of the aquaculture production is increasing with the declining global fish stocks, but early sexual maturation in several farmed species reduces muscle growth and quality, and escapees could have a negative impact on wild populations. A possible solution to these problems is the production of sterile fish by ablation of the embryonic primordial germ cells (PGCs), a technique developed in zebrafish. Cell-specific regulation of mRNA stability is crucial for proper specification of the germ cell lineage and commonly involves microRNA (miRNA)-mediated degradation of targeted mRNAs in somatic cells. This study reports on the functional roles of conserved motifs in the 3' untranslated region (UTR) of the miRNA target gene nanos3 identified in Atlantic cod, Atlantic salmon, and zebrafish. The 3'UTR of cod nanos3 was sufficient for targeting the expression of green fluorescent protein (GFP) to the presumptive PGCs in injected embryos of the three phylogenetically distant species. 3'UTR elements of importance for PGC-specific expression were further examined by fusing truncated 3'UTR variants of cod nanos3 to GFP followed by injections in zebrafish embryos. The expression patterns of the GFP constructs in PGCs and somatic cells suggested that the proximal U-rich region is responsible for the PGC-specific stabilization of the endogenous nanos3 mRNA. Morpholino-mediated downregulation of the RNA-binding protein Dead end (DnD), a PGC-specific inhibitor of miRNA action, abolished the fluorescence of the PGCs in cod and zebrafish embryos, suggesting a conserved DnD-dependent mechanism for germ cell survival and migration.

Germline-specific and sexually dimorphic expression of a dead end gene homologue in turbot (Scophthalmus maximus)

Germ cells are indispensable for gonadal development and fertility. However, the physiological mechanisms regulating germ cell development in marine fish are poorly understood due to a lack of germ cell markers. The dead end (dnd) gene is a vertebrate-specific component of germplasm crucial for primordial germ cells (PGCs) migration and development in teleosts. In this study, we identified a dnd homologue (Smdnd) in turbot (Scophthalmus maximus) and investigated its expression pattern during embryogenesis and gonadal development. The deduced amino acid sequence of Smdnd shared several conserved motifs of Dnd homologues as well as high identity to other Dnd proteins. Phylogenetic analysis revealed that the SmDnd was closely related to its teleost counterparts. Reverse transcription polymerase chain reaction (RT-PCR) and in situ hybridization revealed that Smdnd transcripts could be exclusively detected in germ cells, including presumptive PGC and adult male and female germ cells. In addition, an interesting sexually dimorphic expression of Smdnd during gonadal development was observed by real-time PCR. Female turbot showed greater (P < 0.05) Smdnd expression than male before sex maturation. This difference reduced gradually due to the upregulation of Smdnd in the male during the period corresponding to spermatogonia proliferation and meiosis. These results indicate that Smdnd can be used as a germ cell marker in turbot. In addition, the temporal and sex differences in Smdnd expression indicate that this gene may play different roles in gonadal development in both sexes.

Primordial germ cells in an oligochaete annelid are specified according to the birth rank order in the mesodermal teloblast lineage

The primordial germ cells (PGCs) in the oligochaete annelid Tubifex tubifex are descentants of the mesodermal (M) teloblast and are located in the two midbody segments X and XI in which they serve as germline precursors forming the testicular gonad and the ovarian gonad, respectively. During embryogenesis, vasa-expressing cells (termed presumptive PGCs or pre-PGCs) emerge in a variable set of midbody segments including the genital segments (X and XI); at the end of embryogenesis, pre-PGCs are confined to the genital segments, where they become PGCs in the juvenile. Here, using live imaging of pre-PGCs, we have demonstrated that during Tubifex embryogenesis, pre-PGCs (defined by Vasa expression) stay in segments where they have emerged, suggesting that it is unlikely that pre-PGCs move intersegmentally during embryogenesis. Thus, it is apparent that pre-PGCs derived from the 10th and 11th M teloblast-derived primary m blast cells (designated m10 and m11) that give rise, respectively, to segments X and XI are specified in situ as PGCs and that those born in other segments become undetectable at the end of embryogenesis. To address the mechanisms for this segment-specific development of PGCs, we have performed a set of cell-transplantation experiments as well as cell-ablation experiments. When m10 and m11 that are normally located in the mid region of the embryo were placed in positions near the anterior end of the host embryo, these cells formed two consecutive segments, which exhibited Vasa-positive PGC-like cells at early juvenile stage. This suggests that in terms of PGC generation, the fates of m10 and m11 remain unchanged even if they are placed in ectopic positions along the anteroposterior axis. Nor was the fate of m10 and m11 changed even if mesodermal blast cell chains preceding or succeeding m10 and m11 were absent. In a previous study, it was shown that PGC development in segments X and XI occurs normally in the absence of the overlying ectoderm. All this strongly suggests that irrespective of their surrounding cellular environments, m10 and m11 autonomously generate PGCs. We propose that m10 and m11 are exclusively specified as precursors of PGCs at the time of their birth from the M teloblast and that the M teloblast possesses a developmental program through which the sequence of mesodermal blast cell identities is determined.

The expression of a novel cxcr4 gene in Xenopus embryo.

The aim of the present work was to identify a homologue of zebrafish cxcr4b in Xenopus, which could be involved in primordial germ cell (PGC) guidance migration. Following a BLAST analysis, the clone gi 27519681, homologous to the zebrafish gene z-cxcr4b, was identified, inserted into pCMV-SPORT6 plasmid and cloned in Escherichia coli. Embryonic expression of x-cxcr4b was analyzed by RT-PCR. X-cxcr4b was weakly expressed maternally but sharply increased after the mid-blastula transition (MBT), declining significantly at stage 45 when PGCs migration is complete. In contrast, RT-PCR of isolated presumptive PGCs showed strong maternal expression at stage 8, which decreased by stage 10 post-MBT and was not detected at stage 14. Whole mount in situ hybridization of x-cxcr4b mRNA showed that this gene is expressed in neural and haematopoietic tissues, and should be linked to important processes during embryonic development of these organs. Although weak staining could be seen in some samples within the anterior endoderm, expression of x-cxcr4b was never coincident with that of Xpat mRNA, which labels PGCs restricted to the posterior endoderm. Therefore, maternal x-cxcr4b is specifically downregulated within PGCs at pre-migratory stages while it is expressed in other tissues.

Vasa and the germ line lineage in a colonial urochordate

Germ cell sequestering in Animalia is enlightened by either, launching true germ line along epigenetic or preformistic modes of development, or by somatic embryogenesis, where no true germ line is set aside. The research on germ line-somatic tissue segregation is of special relevancy to colonial organisms like botryllid ascidians that reconstruct, on a weekly basis, completely new sets of male and female gonads in newly formed somatic tissues. By sequencing and evaluating expression patterns of BS-Vasa, the Botryllus schlosseri orthologue of Vasa, in sexually mature and asexual colonies during blastogenesis, we have demonstrated that the BS-Vasa mRNA and protein are not expressed exclusively in germ cell lineages, but appeared in cells repeatedly emerging de novo in the colony, independently of its sexual state. In addition, we recorded an immediate Vasa response to cellular stress (UV irradiation) indicating additional functions to its germ line assignments. To confirm germ lineage exclusivity, we examined the expression of three more stem cell markers (BS-Pl10, Bl-piwi and Oct4). Vasa co-expression with Pl10 and Oct4 was detected in germ line derivatives and with Bl-piwi in somatic tissues. Presumptive primordial germ cells (PGC-like cells), that are Vasa+/Pl10+/Oct4+ and 6–12 μm in diameter, were first detected in wrapped-tail embryos, in oozooids, in sexual/asexual colonies, within a newly identified PGC niche termed as ‘budlet niche’, and in circulating blood borne cells, indicating epigenetic embryogenesis. Alternatively, BS-Vasa co-expression with piwi orthologue, an omnipresent bona fide stemness flag, in non germ line cell populations, may indicate germ cell neogenesis (somatic embryogenesis) in B. schlosseri. Both alternatives are not necessarily mutually exclusive.

Embryonic expression of p68, a DEAD-box RNA helicase, in the oligochaete annelid Tubifex tubifex

We have cloned and characterized the expression of a p68 homologue (designated Ttu-p68) from the oligochaete annelid Tubifex tubifex. Ttu-p68 mRNA is distributed broadly throughout the early stages. Ttu-p68 is expressed in all of the early blastomeres, in which Ttu-p68 RNA associates with pole plasms. Ttu-p68 transcripts are concentrated to 4d cell but not to 2d cell. During gastrulation, expression of Ttu-p68 is restricted to elongating germ bands (GBs) and an anteriormost crescent of micromere descendants on both sides of the embryo. During body elongation that follows gastrulation, expression of Ttu-p68 is further restricted to the stomodaeum (derived from the micromere crescent), ventral ganglia, lateral dots (corresponding to dorsal and ventral setal sacs), ventral large cells (that resemble presumptive primordial germ cells) in segments VIII–XII, and a bilateral pair of cell clusters at the caudal end. At the end of embryogenesis, Ttu-p68 expression persists exclusively in the tail and the lining epithelium of the pharynx.

Transient occurrence of vasa-expressing cells in nongenital segments during embryonic development in the oligochaete annelid Tubifex tubifex

The primordial germ cells (PGCs) in the oligochaete annelid Tubifex tubifex are mesodermal in origin and are located in the two midbody segments X and XI in which the testis and the ovary are formed, respectively. To identify a molecular marker for the Tubifex PGCs, we isolated the Tubifex homologue (Ttu-vas) of the Drosophila vasa gene. Using whole-mount in situ hybridization, we examined the spatial expression patterns of Ttu-vas from one-cell stage through juvenile stage. Ttu-vas messenger ribonucleic acid (RNA) is present as a maternal transcript distributed broadly throughout the early stages. Ttu-vas is expressed in all of the early cleavage blastomeres, in which Ttu-vas RNA associates with mitotic spindles and pole plasms. Expression of Ttu-vas gradually becomes restricted, first to teloblasts, then to their blast cell progeny comprising the germ bands (GBs), and finally to a set of large ventral cells (termed VE cells) in a variable set of midbody segments including the genital segments (X and XI). At the end of embryogenesis, VE cells are confined to genital segments where they are presumably germline precursors in the juvenile. Staining with a cross-reacting anti-Vasa antibody suggested that VE cells express Ttu-vas protein to the same extent irrespective of their positions along the anteroposterior axis. A set of cell ablation experiments suggested that VE cells are derived from the mesodermal teloblast lineage and that the emergence of VE cells takes place independently of the presence of the ectodermal GBs that normally overlay the mesoderm. These results suggest that T. tubifex generates supernumerary presumptive PGCs during embryogenesis whose number is variable among embryos.

Ectopic germline cells in embryos of Xenopus laevis

Whether all descendants of germline founder cells inheriting the germ plasm can migrate correctly to the genital ridges and differentiate into primordial germ cells (PGCs) at tadpole stage has not been elucidated in Xenopus. We investigated precisely the location of descendant cells, presumptive primordial germ cells (pPGCs) and PGCs, in embryos at stages 23-48 by whole-mount in situ hybridization with the antisense probe for Xpat RNA specific to pPGCs and whole-mount immunostaining with the 2L-13 antibody specific to Xenopus Vasa protein in PGCs. Small numbers of pPGCs and PGCs, which were positively stained with the probe and the antibody, respectively, were observed in ectopic locations in a significant number of embryos at those stages. A few of the ectopic PGCs in tadpoles at stages 44-47 were positive in TdT-mediated dUTP digoxigenin nick end labeling (TUNEL) staining. By contrast, pPGCs in the embryos until stage 40, irrespective of their location and PGCs in the genital ridges of the tadpoles at stages 43-48 were negative in TUNEL staining. Therefore, it is evident that a portion of the descendants of germline founder cells cannot migrate correctly to the genital ridges, and that a few ectopic PGCs are eliminated by apoptosis or necrosis at tadpole stages.

The mRNA coding for Xenopus glutamate receptor interacting protein 2 (XGRIP2) is maternally transcribed, transported through the late pathway and localized to the germ plasm

Using a large-scale in situ hybridization screening, we found that the mRNA coding for Xenopus glutamate receptor interacting protein 2 (XGRIP2) was localized to the germ plasm of Xenopus laevis. The mRNA is maternally transcribed in oocytes and, during maturation, transported to the vegetal germ plasm through the late pathway where VegT and Vg1 mRNAs are transported. In the 3′-untranslated region (UTR) of the mRNA, there are clusters of E2 and VM1 localization motifs that were reported to exist in the mRNAs classified as the late pathway group. With in situ hybridization to the sections of embryos, the signal could be detected in the cytoplasm of migrating presumptive primordial germ cells (pPGCs) until stage 35. At stage 40, when the cells cease to migrate and reach the dorsal mesentery, the signal disappeared. A possible role of XGRIP2 in pPGCs of Xenopus will be discussed.

The Xdsg protein in presumptive primordial germ cells (pPGCs) is essential to their differentiation into PGCs in Xenopus

In order to know the role of the Xdsg gene in presumptive PGCs (pPGCs) of Xenopus, we attempted to inhibit the translation of Xdsg mRNA in pPGCs by injecting antisense morpholino oligo (asMO), together with Fluorescein Dextran-Lysine (FDL), into single germ plasm-bearing cells of 32-cell embryos. Among three types of asMOs complementary to different parts of the 5′-untranslated region of Xdsg mRNA tested, only one asMO, designated as Xdsg-3, inhibited the translation of the mRNA in FDL-labeled pPGCs, resulting in the absence of labeled PGCs in experimental tadpoles. On the other hand, two other asMOs, Xdsg-1 and -2, did not inhibit the translation, so that a similar number of labeled PGCs found in FDL-injected but asMO-uninjected control tadpoles were observed in experimental tadpoles derived from asMO-injected embryos. Surprisingly, use of Xdsg-3 asMO resulted in the disappearance of the protein of Xenopus vasa homolog ( Xenopus vasa-like gene 1, XVLG1) from FDL-labeled pPGCs by inhibiting the translation of XVLG1 mRNA. However, the effect of Xdsg-3 asMO on the translation of Xdsg and XVLG1 mRNAs and PGC formation could be canceled by the coinjection with Xdsg mRNA. Consequently, the Xdsg protein in pPGCs may play an important role in the formation of PGCs by regulating the production of XVLG1 protein.

Isolation of germline cells from Drosophila embryos by flow cytometry

Primordial germ cells (PGC) are the earliest identifiable germ cells in the embryo. To understand the molecular basis of germline development, isolation of pure PGC is required. We report here the use of fluorescence-activated cell sorting (FACS) to isolate pure populations of Drosophila pole cells, which are the presumptive primordial germ cells in flies. In order to fluorescently mark pole cells, we used an EGFP-vasa transgenic line that expresses green fluorescent protein (GFP) specifically and continuously in the germ line throughout the life cycle. The purity of FACS-sorted pole cells from embryos was confirmed by microscopic inspection and quantitative polymerase chain reaction. Moreover, by optimizing the sample preparation and the sorting protocol, embryonic gonads could also be isolated. This technique opens the way for genome-wide transcriptome analysis of germline cells. In a pilot experiment, we generated a cDNA library from purified embryonic gonad and identified a novel germline-specific gene, RpL22-like.

140 LOCALIZATION OF PRIMORDIAL GERM CELLS IN DAY 21 BOVINE EMBRYOS

In most animal species, germ cell precursors, i.e., primordial germ cells (PGCs), arise from the epiblast and then migrate to the future gonadal ridge during development. At least in the mouse, PGCs may be cultured as embryonic germ cells that remain pluripotent. PGCs are the only cells in which OCT4 expression is maintained after gastrulation. The present study aimed at identifying the localization of PGCs in Day 21 in vivo-derived bovine embryos by immunohistochemical staining against OCT4. Six embryos were obtained after slaughter of superovulated heifers 21 days after insemination. The uterine tracts were flushed and embryos fixed, paraffin-embedded, and processed for immunohistochemistry. Embryos were sagitally sectioned, and selected serial sections were immunohistochemically stained for OCT4 to identify potential PGCs. Two embryos were at the neural groove stage. At this stage of development, the primitive gut had not yet been abstricted from the yolk sac and the allantois was not visible. A weak homogeneous OCT4 staining was localized to nuclei in a well-defined region of the epiblast, which was in the process of a gradual anterior to posterior differentiation into neural and surface ectoderm. Moreover, a strong OCT4 staining was localized to a few scattered cells found in the visceral mesoderm associated with the yolk sac in the region of the endoderm-hypoblast transition at some distance from the embryo proper. Four embryos were at the neural tube/somite stage. At this stage of development, the primitive gut had been defined and only the midgut was connected to the yolk sac. Furthermore, the allantois was visible as an anchor-shaped structure at the posterior end of the embryo. A strong OCT4 staining was found in nuclei of solitary cells in the endoderm and its associated visceral mesoderm of the ventral aspect of the mid and hindgut. The described OCT4 staining corresponds well with previous findings in the pig, in which presumptive PGCs are found in the endoderm epithelium during the neural groove stage. Later, during the early somite stages, they are localized in the endoderm and visceral mesoderm of the yolk sac and allantois, and in later somite stages, they are found in the developing genital ridge. This is, however, the first study to demonstrate the localization of these cells, at least by OCT4 staining, in bovine embryos at the neural groove and neural tube/somite stages.

A Simple Method for Estimation of the Effects of Environmental Chemicals on the Formation of Primordial Germ Cells (PGCs) Using Embryos and Larvae of Xenopus laevis

To estimate the developmental toxicity of environmental chemicals on the formation of primordial germ cells (PGCs), Xenopus laevis embryos were exposed to caffeine at 200 mg/l during the migratory stage of presumptive PGCs. The PGC-containing region of the larvae at stage 46, which corresponds to genital ridge, was illuminated by cold light using a halogen lamp and photographed using a digital camera under a dissecting microscope. The length along the cephalocaudal axis and area of the PGC-containing region were measured using image-measuring software. The length and area of the PGC-containing region as well as its relative length compared to the 100-μm body length of caffeine-exposed larvae were significantly shorter and smaller, respectively, than those of the control. Though further studies concerning the effect of caffeine on PGC formation are needed, these findings suggest that the Xenopus embryo and larva system examined in this study is useful as a simple, rapid and low-cost initial method for the estimation of the developmental toxicity of environmental chemicals on PGC formation.

Vasa and nanos expression patterns in a sea anemone and the evolution of bilaterian germ cell specification mechanisms

Most bilaterians specify primordial germ cells (PGCs) during early embryogenesis using either inherited cytoplasmic germ line determinants (preformation) or induction of germ cell fate through signaling pathways (epigenesis). However, data from nonbilaterian animals suggest that ancestral metazoans may have specified germ cells very differently from most extant bilaterians. Cnidarians and sponges have been reported to generate germ cells continuously throughout reproductive life, but previous studies on members of these basal phyla have not examined embryonic germ cell origin. To try to define the embryonic origin of PGCs in the sea anemone Nematostella vectensis, we examined the expression of members of the vasa and nanos gene families, which are critical genes in bilaterian germ cell specification and development. We found that vasa and nanos family genes are expressed not only in presumptive PGCs late in embryonic development, but also in multiple somatic cell types during early embryogenesis. These results suggest one way in which preformation in germ cell development might have evolved from the ancestral epigenetic mechanism that was probably used by a metazoan ancestor.
