Porous hydroxyapatite-titanium dioxide composites prepared by a sol-gel / supercritical CO2-drying combined process

In this work, porous nanostructured materials based on hydroxyapatite (HA) and titanium dioxide (TiO2) were prepared by integrating the sol-gel process and supercritical CO2 drying. These combined processes constitute a synergistic methodology that allows modulating the porosity generated in the gel and preserving it after drying. HA nanoparticles and a pore-generating agent (Polyvinylpyrrolidone) were included without significant modifications to gel consolidation times or integrity by using this methodology. The presence of HA nanoparticles within the TiO2 matrix helped preserving the anatase phase after the calcination stage. No reactivity between the present phases was detected, indicating their successful integration in the final composite material structure, which was one of the most challenging aspects of the research. This allowed HA-TiO2 composites to improve the adhesion of MG63 osteoblast-like cells and their proliferation, being the sample with high HA content (∼37 % wt/wt) the one that presented the best response. The present work puts forward a new way for the synthesis of scaffolds composed of HA and TiO2 with retention of the anatase phase for applications in bone tissue engineering.

Fabrication of osteogenesis induced WE43 Mg-Hydroxyapatite composites with low biodegradability and increased biocompatibility for orthopedic implant applications

Good biocompatibility, low biodegradation rate and excellent mechanical properties are essential for desirable performance of metallic biomaterials during osseointegration process. In the present study, bioabsorbable WE43 Mg alloy was used for fabrication of bio-composites with addition of nanosized hydroxyapatite (HA) particles by means of multi pass friction stir processing (FSP) for orthopedic implant applications. Microstructure, in vitro biodegradation and biocompatibility, wettability, and microhardness of resultant bio-composites were characterized. Results showed that applying severe plastic deformation through FSP caused significant grain refinement of Mg matrix and homogenous dispersion of fragmented secondary phase particles. Moreover, applying higher number of FSP passes increased uniformity of distribution of HA nanoparticles throughout Mg matrix. Compared to coarse grain samples, grain refined samples showed lower Mg2+ concentration and H2 evolution in simulated body fluid (SBF). Moreover, addition of HA into Mg matrix, reduced Mg2+ release and H2 evolution. The bio-composites illustrated uniformly and flatly corroded surface compared to samples without HA nanoparticles. During immersion of the bio-composites in SBF solution, a cauliflower structure of Ca–P compounds deposited on the surface of bio-composites which confirmed acceptable biomineralization. Contact angle measurement demonstrated that wettability of fabricated bio-composites increased by increasing number of FSP passes. Mouse osteoblast MC3T3 cell culture indicated that the fabricated bio-composites with uniform distribution of HA nanoparticles showed excellent in vitro biocompatibility, cell viability, and cell proliferation. Eventually, it was found that applying further FSP and presence of HA nanoparticles resulted in increased microhardness which is an indication of higher load-bearing capability of the fabricated bone implants.

Enhancing structural strength and improving cell survival through Polycaprolactone/(gelatin/hydroxyapatite) Core‐Shell nanofibers for tissue engineering

AbstractBone tissue engineering is a practical approach to repairing broken or damaged bones that combines scaffold, cells, and growth factors for treatment. In this study, polycaprolactone (PCL)/gelatin (Ge)/hydroxyapatite (HA) core‐shell nanocomposites have been produced by the coaxial electrospinning method. Coaxial electrospinning is an efficient method for scaffold preparation to provide an interconnected porous fibrous scaffold. The prepared nanocomposite simultaneously benefited from the good mechanical behavior of the core PCL polymer. The desired biological properties also originated from the outer layer of the Ge/HA nanocomposite. Nanofibrous scaffolds' properties were characterized using SEM, TEM, FTIR, TGA, DSC, tensile test, contact angle, and MTT assay. The morphology of the as‐electrospun nanofibers was investigated using SEM and TEM, which revealed a defect‐less fibrous morphology. TEM images showed the core‐shell structure of the prepared scaffold nanofibers. The contact angle test showed that the presence of HA nanoparticles has improved the wettability of fibrous composites. In addition, HA nanoparticles could effectively strengthen the polymer scaffolds. The highest UTS value of 4.1 MPa was obtained in the PCL/(Ge+10%HA) sample. The cytotoxicity results revealed that the prepared scaffolds were utterly biocompatible. Moreover, significant cell proliferation of osteosarcoma cells was observed at high HA contents. The interconnected pores allowed cells to migrate into the scaffolds and grow inside. Based on the obtained results, PCL/(Ge/ HA) core‐shell nanofibers could be a promising candidate for bone scaffolds.

Green synthesis and characterization of poly(glycerol‐azelaic acid) and its nanocomposites for applications in regenerative medicine

AbstractA series of novel bio‐polyester nanocomposites based on glycerin and azelaic acid as monomers incorporating hydroxyapatite (HA) nanoparticles were fabricated via in situ polymerization method. Chemical structure of the samples was investigated by 1H‐NMR, 13C‐NMR, and Fourier‐transform infrared spectroscopy (FTIR). Energy dispersive X‐ray‐mapping analysis illustrated that the nanoparticles were well dispersed in the poly (glycerol azelaic acid) (PGAZ) matrix. Viscoelastic properties of the samples under various frequencies were examined in which the PGAZ specimen containing 1.0 wt% of HA nanoparticles (PGAZH1.0) exhibited superlative properties. Furthermore, the alterations in the glass transition temperature of the samples were comprehensively discussed. Thermal gravimetric analysis displayed that nanocomposites generally have a difference in degradation patterns from that of the pristine sample. Dynamic contact angle demonstrated that the presence of HA nanoparticles imposed a significant influence on hydrophilicity. The hydrolytic degradation values at pH = 7 and pH = 11 were measured and determined that the degradation rate for the PGAZ sample containing 1.5 wt% HA (PGAZH1.5) was higher than those of the other samples. Moreover, in vitro studies elucidated that cell attachment on PGAZH1.0 and PAZH1.5 surfaces were acceptable.

Poly(ε-Caprolactone)/Poly(Glycerol Sebacate) Composite Nanofibers Incorporating Hydroxyapatite Nanoparticles and Simvastatin for Bone Tissue Regeneration and Drug Delivery Applications.

Herein, we report a drug eluting scaffold composed of a composite nanofibers of poly(ε-caprolactone) (PCL) and poly(glycerol sebacate) (PGS) loaded with Hydroxyapatite nanoparticles (HANPs) and simvastatin (SIM) mimicking the bone extracellular matrix (ECM) to improve bone cell proliferation and regeneration process. Indeed, the addition of PGS results in a slight increase in the average fiber diameter compared to PCL. However, the presence of HANPs in the composite nanofibers induced a greater fiber diameter distribution, without significantly changing the average fiber diameter. The in vitro drug release result revealed that the sustained release of SIM from the composite nanofiber obeying the Korsemeyer–Peppas and Kpocha models revealing a non-Fickian diffusion mechanism and the release mechanism follows diffusion rather than polymer erosion. Biomineralization assessment of the nanofibers was carried out in simulated body fluid (SBF). SEM and EDS analysis confirmed nucleation of the hydroxyapatite layer on the surface of the composite nanofibers mimicking the natural apatite layer. Moreover, in vitro studies revealed that the PCL-PGS-HA displayed better cell proliferation and adhesion compared to the control sample, hence improving the regeneration process. This suggests that the fabricated PCL-PGS-HA could be a promising future scaffold for control drug delivery and bone tissue regeneration application.

Physico-chemical and biological properties of electrospun poly-L-lactic acid/hydroxyapatite nano-composite scaffold for bone regeneration

The main goal of tissue engineering is to design scaffolds with the suitable microenvironment, prominent mechanical properties, adequate biocompatibility, and biodegradability. In the present study, Poly-L-lactic acid (PLLA)/10% hydroxyapatite (HA) nano-composite (PLLA/HA) scaffold and pure PLLA scaffold were fabricated via electrospinning technique. They were then assessed for their potential as substrates for the cell culture. The scaffolds were characterized by scanning electron microscope, thermogravimetric analysis, differential thermal analysis, and fourier transform infrared spectroscopy. The results showed the dispersion and integration of HA nanoparticles in the composite scaffold. Pore size distribution measurements indicated that the scaffold pore size in terms of Ferret diameter varies between 0.1 and 22.0 µm. In vitro degradation behavior of the scaffold in phosphate buffered saline (PBS) was obtained by measuring water uptake, weight loss, and pH change of the PBS during the degradation period. We observed that the degradation of the scaffold could be markedly accelerated by HA nanoparticle (10%). It was also shown that acidification of the PBS due to the PLLA degradation is suppressed by the HA nanoparticles. Finally, human umbilical-cord-derived mesenchymal stem cells (hUC-MSCs) were used to evaluate the cellular support of the composite scaffolds. The presence of HA nanoparticles in the PLLA fibrous scaffolds resulted in promoting the attachment and the proliferation of the hUC-MSCs. This study suggests that the PLLA/HA scaffold could be the material of choice for functional bone tissue engineering.

Fabrication and characterization of a starch-based nanocomposite scaffold with highly porous and gradient structure for bone tissue engineering

Starch based scaffolds are considered as promising biomaterials for bone tissue engineering. In this study, a highly porous starch/polyvinyl alcohol (PVA) based nanocomposite scaffold with a gradient pore structure was made by incorporating different bio-additives, including citric acid, cellulose nanofibers, and hydroxyapatite (HA) nanoparticles. The scaffold was prepared by employing unidirectional and cryogenic freeze-casting and subsequently freeze-drying methods. Fourier transform infrared (FTIR) spectroscopy confirmed the cross-linking of starch and PVA molecules through multiple esterification phenomenon in the presence of citric acid as a cross-linking agent. Field emission scanning electron microscopy (FE-SEM) observations showed formation of aligned lamellar pores with a gradient pore width in the range of 80 to 292 μm, which well meets the pore size requirement for bone regeneration, and also well dispersion of cellulose and HA nanofillers within the scaffold matrix. Based on the mechanical testing results, the cellulose-HA reinforced scaffold possesses sufficient compressive modulus and yield strength for non-load bearing applications in the dry state; and also it presents fast responsive shape recovery in wet state. According to in-vitro assessments, apatite phase mineralization was extensively induced in the presence of HA nanoparticles as heterogeneous nucleating sites. Also, it was revealed that cellulose and HA nanofillers decelerate and accelerate the scaffold biodegradation rate, respectively. MTT assay proved good cytocompatibility of the nanocomposite scaffold with osteoblast cells. Finally, it was shown that the introduced scaffold provides a suitable platform for the cells adhesion.

Synergistic toughening of nanocomposite hydrogel based on ultrasmall aluminum hydroxide nanoparticles and hydroxyapatite nanoparticles

The development of hydrogels combining superior mechanical property attracts researchers' attention in recent years. Here, a novel nanocomposite hydrogel is prepared by one‐step photo‐polymerization of acrylic acid (AA) and N,N‐dimethylacrylamide (DMAA) in the presence of hydroxyapatite nanoparticles (HAp NPs) and aluminum hydroxide nanoparticles (Al(OH)3 NPs). The resulting transparent hydrogel exhibits ultrahigh toughness of 28.3 MJ m−3, high strength of 2.8 MPa, large elongation of 1300%, excellent self‐recovery and good biocompatibility. The crosslinking mechanism is contributed to the ionic coordination interaction between cations absorbing on the surface of nanoparticles and carboxyl groups of the polymer chain, and the synergistic toughening effect between HAp NPs and Al(OH)3 NPs is responsible for improving the strength and toughness simultaneously. Furthermore, this synergistic toughening strategy may provide a universal method for designing and constructing high strength nanocomposite hydrogels in many applications, especially for artificial cartilages. Integration of Stem Cell to Chondrocyte-Derived Cartilage Matrix in Healthy and Osteoarthritic States in the Presence of Hydroxyapatite Nanoparticles.

We investigated the effectiveness of integrating tissue engineered cartilage derived from human bone marrow derived stem cells (HBMSCs) to healthy as well as osteoarthritic cartilage mimics using hydroxyapatite (HA) nanoparticles immersed within a hydrogel substrate. Healthy and diseased engineered cartilage from human chondrocytes (cultured in agar gels) were integrated with human bone marrow stem cell (HBMSC)-derived cartilaginous engineered matrix with and without HA, and evaluated after 28 days of growth. HBMSCs were seeded within photopolymerizable poly (ethylene glycol) diacrylate (PEGDA) hydrogels. In addition, we also conducted a preliminary in vivo evaluation of cartilage repair in rabbit knee chondral defects treated with subchondral bone microfracture and cell-free PEGDA with and without HA. Under in vitro conditions, the interfacial shear strength between tissue engineered cartilage derived from HBMSCs and osteoarthritic chondrocytes was significantly higher (p < 0.05) when HA nanoparticles were incorporated within the HBMSC culture system. Histological evidence confirmed a distinct spatial transition zone, rich in calcium phosphate deposits. Assessment of explanted rabbit knees by histology demonstrated that cellularity within the repair tissues that had filled the defects were of significantly higher number (p < 0.05) when HA was used. HA nanoparticles play an important role in treating chondral defects when osteoarthritis is a co-morbidity. We speculate that the calcified layer formation at the interface in the osteoarthritic environment in the presence of HA is likely to have attributed to higher interfacial strength found in vitro. From an in vivo standpoint, the presence of HA promoted cellularity in the tissues that subsequently filled the chondral defects. This higher presence of cells can be considered important in the context of accelerating long-term cartilage remodeling. We conclude that HA nanoparticles play an important role in engineered to native cartilage integration and cellular processes.

Parallel nano-assembling of a multifunctional GO/HapNP coating on ultrahigh-purity magnesium for biodegradable implants

This work reports the one-step fabrication of a novel coating on ultra high purity magnesium using a parallel nano assembling process. The multifunctional biodegradable surface was obtained by adding hydroxyapatite nanoparticles (HapNP) plus graphene oxide (GO). The coating was characterized by scanning electron microscope (SEM), transmission electron microscope (TEM), X-ray diffractometer (XRD), micro-Raman spectroscopy. The thin phosphate coating (thickness of 1μm) reveals a uniform coverage with cypress like structures. The incorporation of HapNP and GO promotes the hydrophilic behavior of the coating surface. The results revealed that the proposed coating can be used to tailor the surface properties such as wettability by adjusting the contents of HapNP and GO. The in vitro degradation rate of the coated magnesium suggests that the presence of HapNP and GO/HapNP in the phosphate coating decreased the current density compared to the single phosphate coating and uncoated magnesium. This study also reveals the HapNP/GO/phosphate coating induces apatite formation, showing suitable degradability that makes it a promising coating candidate for enhanced bone regeneration.

Effect of Hydroxyapatite Nanoparticles on Biotransport Phenomena in Freezing HeLa Cells

The effect of nanoparticles on subzero biotransport phenomena of living cells is very rare in the literature, although the information is of great importance for the application of nanotechnology in the field of cryobiology. In this study, subzero water transport phenomena in freezing HeLa cells in 1 × phosphate buffered saline (PBS) containing 0%, 0.05%, and 0.1% (w/w) hydroxyapatite (HA) nanoparticles with and without pre-incubation at 37 °C was quantitatively investigated. The results reveal that the presence of HA nanoparticles slightly facilitates the subzero water transport of HeLa cells.

Effects of nanoparticles on devitrification and recrystallization of aqueous glycerol and PEG-600 solutions

Nanoparticles in solution offer unique electrical, mechanical and thermal properties due to their physical presence and interaction with the state of dispersion. This work is aimed to study the effects of hydroxyapatite (HA) nanoparticles on the behavior of devitrification and recrystallization of glycerol (60% w/w) and PEG-600 (50% w/w) solutions during warming. HA nanoparticles of different sizes (20, 40, 60 nm) and concentrations (0.1%, 0.5%, w/w) were incorporated into solutions, and were studied by calorimetric analysis coupled with cryomicroscopy. The presence of HA nanoparticles has little effect on the devitrification end temperatures, but affects the devitrification onset temperatures of glycerol and PEG-600 solutions. The investigation with the cryomicroscope observed that the ice morphologies of glycerol and PEG-600 solutions are dendritic and spheric respectively. The ice fraction of glycerol solution containing 0.1% HA with the size of 60 nm decreased to 2/5 of that of the solution without nanoparticles at −45°C. The ice fractions of PEG-600 solutions increased significantly between −64°C and −54°C, and the ice fraction of PEG-600 solution without nanoparticles increased by 92% within the temperature range. The findings have significant implications for biomaterial cryopreservation, cryosurgery, and food manufacturing. The complexity of ice crystal growth kinetics in nanoparticle-containing solutions awaits further study.

Synthesis of Hydroxyapatite Nanoparticles in Presence of a Linear Polysaccharide

Hydroxyapatite nanoparticles compounds were synthesized. Natural hydroxyapatite and a linear polysaccharide (1–3 linked β-D galactopyranose and 1,4 linked 3,6 anhydro-α-L-galactopyranose) were used as a precursor in its formation. Our purpose was to produce nanoparticles in the presence of a linear polysaccharide with the use of a gelification method. The powder sample was evaluated by scanning tunneling microscope (STM), Brunauer-Emmett-Teller (BET) analysis, X-ray diffraction pattern (XRD), differential thermal analysis (DTA), infrared (IR) analysis, and thermal gravimetric analysis (TGA). According to the results, it was found that these nanoparticles can successfully be synthesized using a polysaccharide in a solution. On the other hand, the XRD peak intensity corresponds to hydroxyapatite structure in the range of temperature of 810°C. The influence of the polysaccharide on the evolution of the nanoparticles has been demonstrated. This observation opens up new routes for the fabrication of nanoparticles using polysaccharides network. The synthesized nanoparticles have diameters ranging from 10 nm to 11 nm approximately. The elaboration conditions such as pH and concentration were optimized in this solution.

Synthesis of hydroxyapatite nanoparticles trough polyelectrolyte-modified microemulsions

The paper is focused on the formation of hydroxyapatite nanoparticles (HAp) in polyelectrolyte-modified microemulsions, in a microemulsion template phase consisting of cyclohexane, water, cationic surfactant and cosurfactant, in the presence of Na-polyacrylate (PAA) as an anionic polyelectrolyte. It is shown that PAA, can be incorporated into the individual inverse microemulsion droplets. The microemulsion droplets and PAA-filled microemulsion droplets can be successfully used as a template phase for the nanoparticles formation. Prepared HAp in presence of polyelectrolyte has a different morphology from samples which are synthesized in absence of polyelectrolyte. PAA leads to formation of needle-like HAp (20-30nm in diameter and 100-200nm in length). Formation of HAp at room temperature was confirmed by X-ray diffraction (XRD) and Fourier transform infrared spectroscopy (FT-IR). Size and morphology of the HAp samples were characterized using transmission electron microscopy (TEM).

Improvement of thermostability and activity of pectate lyase in the presence of hydroxyapatite nanoparticles

The activity and half-life of pectate lyase (PL) from Bacillus megaterium were nine- and 60-fold, respectively, higher at 90°C in the presence of hydroxyapatite nanoparticles (NP-PLs) than in the presence of 1mM CaCl2. Thermodynamic analysis of the nanoparticle-induced stability revealed an enhanced entropy–enthalpy compensation by the NP-PLs since a reciprocal linearity of the enthalpy–entropy change to 90°C was observed. Without nanoparticles, the linearity range was 70°C. Such compensation reflected the maintenance of the native structure of proteins. The remarkable enhancement of activity and stability of the NP-PL system at high temperatures may be utilized commercially e.g. in the food industry or the processing of natural fibers that may require a thermotolerant enzyme.

Synthesis of hydroxyapatite nanoparticles by a novel ultrasonic assisted with mixed hollow sphere template method

Hydroxyapatite (HAP) is the main inorganic component of bone material and is widely used in various biomedical applications due to its excellent bioactivity and biocompatibility. In this paper, we have reported the synthesis of hydroxyapatite nanoparticles by a novel ultrasonic assisted mixed template directed method. In this method glycine–acrylic acid (GLY–AA) hollow spheres were used as an organic template which could be prepared by mixing of glycine with acrylic acid. The as-synthesized HAP nanoparticles were characterized by X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FT-IR), scanning electron microscope (SEM) and tunnelling electron microscope (TEM) to investigate the nature of bonding, crystallinity, size and shape. The thermal stability of as-synthesized nanoparticles was also investigated by the thermo gravimetric analysis (TGA). The effect of ultrasonic irradiation time on the crystallinity and size of the HAP nanoparticles in presence of glycine–acrylic acid hollow spheres template were investigated. From the inspection of the above results it is confirmed that the crystallinity and size of the HAP nanoparticles decrease with increasing ultrasonic irradiation time. Hence the proposed synthesis strategy provides a facile pathway to obtain nano sized HAP with high quality, suitable size and morphology.

Hydroxyapatite-armored poly(ε-caprolactone) microspheres and hydroxyapatite microcapsules fabricated via a Pickering emulsion route

Hydroxyapatite (HAp) nanoparticle-armored poly(ε-caprolactone) (PCL) microspheres were fabricated via a “Pickering-type” emulsion solvent evaporation method in the absence of any molecular surfactants. It was clarified that the interaction between carbonyl/carboxylic acid groups of PCL and the HAp nanoparticles at an oil–water interface played a crucial role in the preparation of the stable Pickering-type emulsions and the HAp nanoparticle-armored microspheres. The HAp nanoparticle-armored PCL microspheres were characterized in terms of size, size distribution, morphology, and chemical compositions using scanning electron microscopy, laser diffraction, energy dispersive X-ray microanalysis, and thermogravimetric analysis. The presence of HAp nanoparticles at the surface of the microspheres was confirmed by scanning electron microscopy and energy dispersive X-ray microanalysis. Pyrolysis of the PCL cores led to the formation of the corresponding HAp hollow microcapsules.

Mechanical properties of (poly(L‐lactide‐co‐glycolide))‐based fibers coated with hydroxyapatite layer

AbstractThis article presents the results of the experimental study on manufacturing and mechanical evaluation of poly(L‐lactide‐co‐glycolide) (PLGA)‐based fibers modified with ceramic nanoparticles. Study was conducted to establish the effect of biomimetic formation of apatite layers on polymeric fibers on their mechanical properties. The tensile tests were performed to determine the influence of polymer crystallinity and the presence of hydroxyapatite nanoparticles (nanoHAp) on mechanical properties of PLGA fibers coated with hydroxyapatite (HAp) layer. HAp deposits on the surfaces of the fibers precipitated from simulated body fluid (SBF). Three types of fibers coated with HAp layers were compared in mechanical tests. The results indicated that by using a biomimetic fiber coating method the mechanical properties of the fibers are affected by their crystallinity. The nanoHAp modified polymer fibers after incubation in SBF were found to have a continuous HAp layer. The layer affected the mechanical behavior (force–strain function) of the fibers from nonlinear to linear, typical of ceramic materials. The tensile modulus of the fibers with a continuous layer was found to increase with the apatite layer thickness, whereas the tensile strength decreases. © 2011 Wiley Periodicals, Inc. Fibrous Composites With Anisotropic Distribution of Mechanical Properties After Layer‐by‐Layer Deposition of Aligned Electrospun Fibers

AbstractNanofibers and composite nanofibers have been used as mechanical reinforcement in a bulk matrix, but there is little research done on bulk nanocomposites made out of nanofibers. In the current study, in situ grown composite fibers of poly(DL‐lactide) (PDLLA) and hydroxyapatite (HA) were obtained with uniaxial distribution, which were deposited by layer‐by‐layer with different interlayer angles, and hot‐pressed into fibrous composites with anisotropic distribution of mechanical properties. Composite fibrous mats with the HA loading of 25.2 ± 2.1% and orientation degree of 75.4 ± 3.6% were obtained. Orthogonal experiments design was employed to qualitatively analyze effects of the hot‐pressing parameters on the mechanical properties. The holding temperature, pressure, and duration were optimized as 60 °C, 2 MPa, and 10 min, respectively, to maintain the fibrous structure, strengthen the interfiber bondings, and eliminate the dimensional shrinkage of the hot‐pressed fibrous composites. The ultimate tensile strength along the axial direction was 36.2 ± 2.2 MPa, which was significantly higher than that along the transverse direction. Furthermore, the aligned composite fibers were deposited by layer‐by‐layer with different interlayer angles, and the quantitative equations of the ultimate tensile strength along the axial and transverse directions were drafted as a function of the interlayer deposition angles. The results of cell culture indicated that the presence of HA nanoparticles in the fibrous composites improved the hydrophilicity and provided favorable conditions for cell proliferation and osteogenic differentiation. It is suggested that hot‐pressed fibrous composites with anisotropic distribution of mechanical properties be potential as bone substitutes and repairing devices.
