Preparation of Chitosan Nanoparticles through a Readily Solvent-Exchange Process for Efficient and Enhanced Gene Delivery.

In view of the excellent prospects of gene therapy and the potential safety and immunogenicity issues challenged by viral vectors, it is of great significance to develop a nonviral vector with low toxicity and low cost. In this work, we report a chitosan nanoparticle (CSNP) to be used as a gene vector prepared through a facile solvent-exchange strategy. Chitosan is first dissolved in ionic liquid 1-ethyl-3-methylimidazolium acetate (EMIM Ac), and then, the solvent is exchanged with water/phosphate-buffered saline (PBS) to remove ionic liquid, forming a final CSNP dispersion after ultrasonication. The prepared CSNP shows a positive surface charge and can condense green fluorescent protein-encoding plasmid (pGFP) at weight ratios (CSNP/pGFP) of 5/1 or higher. Dynamic light scattering size and ζ-potential characterization and gel retardation results confirm the formation of CSNP/pGFP complexes. Compared with plain pGFP, efficient cellular internalization and significantly enhanced green fluorescent protein (GFP) expression are observed by using CSNP as a plasmid vector. Benefitting from the intrinsic biocompatibility, low cost, low immunogenicity, and abundant sources of chitosan, as well as the facile preparation and the efficient gene transfection capacity of CSNP, it is believed that this CSNP could be used as a nonviral gene vector with great clinical translational potentials.

Preparation of Chitosan Nanoparticle from Crab Shells and Evaluation of ‎Analgesic and Anti-inflammatory Activities of Chitosan-loaded ‎Haematostaphisbarteri Hook. f. Stembark Extract

Haematostaphis barteri Hook. f. is a tree belonging to the Family Anacardiaceae whose morphological parts have been used for decades for treating various diseases in Northern Nigeria. This study aimed to prepare chitosan nanoparticles from crab shells and evaluate the analgesic and anti-inflammatory activities of chitosan loaded with stembark extract from H. barteri. Phytochemical screening of extract, preparation of chitosan, and evaluation of analgesic and anti-inflammatory activities were carried out following standard procedures. The qualitative phytochemical analysis of the extract revealed the presence of alkaloids, saponins, tannins, flavonoids, and cardiac glycosides while steroids, triterpenes and anthracenes were absent. The LD50 of the crude methanol extract in Wistar albino rats was determined to be greater than 5000 mg/kg body weight and after 14 days of single dose oral administration. No signs of toxicity were recorded. Prepared chitosan nanoparticle loaded with extract has a yield of 26.54 %, particle size of 210 ± 1.01 nm, zeta potential of 25.4 ± 1.02 V, drug entrapment efficiency of 68.4 %, cumulative drug release of 88.24 % and swelling index of 58.14 %. The extract produced a dose-dependent analgesic and anti-inflammatory activity. The chitosan-loaded extract produced the highest inhibition of writhing and paw oedema diameter in the rats, which was significant (p < 0.05) when compared with the normal controls and standards. In conclusion, the study showed that chitosan-loaded stembark extract from H. barteri possessed more potential analgesic and anti-inflammatory activities than those delivered without chitosan nanoparticles. The study further affirms the potential usefulness use of the stembark extract for the treatment of pain and inflammation.

Preparation of chitosan nanoparticles loaded with Balanites aegyptiaca extract for treatment of streptozotocin-induced diabetes in rats

Diabetes mellitus is characterized by elevated blood sugar level due to a deficiency in insulin production and/or action. Balanites aegyptiaca (BA) has been employed as a hypoglycemic medication. Nanoparticles (NPs) have many advantages like minimized drug dose, sustainable drug release, maximized bioavailability and delivery of drugs. The study aimed to synthesize novel chitosan (CS) NPs loaded with BA extract (BA Ex). The prepared NPs were examined in treatment of streptozotocin-induced diabetes in rats. The anti-diabetic efficiency was evaluated through measuring of levels of blood glucose, insulin, lipid profile, oxidative stress markers, pro-inflammatory cytokines. GC-MS, HPLC and ICP techniques showed the presence of numerous bioactive components that have an anti-diabetic effectiveness. BA Ex-CS NPs succeeded in treatment of diabetes; where, it increased insulin secretion, lowered both FBG and FTA levels and helped in neogenesis of pancreatic islets beta cells. The regenerative activity of BA Ex-CS NPs is attributed to its high antioxidant and anti-inflammatory properties. This antioxidant activity scavenged the generated free radicles that resulted from STZ administration. CS NPs raised the plant extract efficacy, prevented its degradation, and regulated the release of its components. The delivery of BA Ex bioactive components has been revolutionized by CS NPs.

Preparation of Chitosan Nanoparticles Loaded with Vascular Endothelial Growth Factor and Its Application in the Treatment of Skin Injury

This research was aimed to explore the application of chitosan (CS) nanoparticles (NPs) loaded with vascular endothelial growth factor (VEGF) in rat skin injury. Method: the VEGF-loaded CS NPs (VEGF/CS-NPs) were prepared, and a skin injury model on the back skin of rats was established. The VEGF/CS-NPs were applied to the injury model to observe the skin changes in the wound area of each group, compare the healing rate, the number of micro-vessels, and VEGF expression in each group of rats, so as to explore the intervention effect of VEGF/CS-NPs in skin injury. Results: the prepared VEGF/CS-NPs were spherical or quasi-spherical in morphology, uniformly dispersed, with smooth surfaces and relatively uniform size. The average particle size (PS) of the blank CS-NPs was 267.5 nm± 25.3 nm, while that of the drug-loaded CS-NPs was 298.6 nm ±44.7 nm, and the PS distribution was also very concentrated. The genipin cross-linked NPs could effectively prevent the degradation of the spherical wall and achieve the slow release of VEGF. After VEGF was applied to rats in the form of NPs, its healing rate and micro-vessel count were markedly superior to those of VEGF injection alone and blank group (P < 0.05), and VEGF expression in skin injury tissues was also greatly increased (P < 0.05). Conclusion: VEGF/CS-NPs could improve the bioavailability of VEGF and promote the healing of rat skin injuries.

Electrosprayed Chitosan Nanoparticles for Drug Carriers in Cancer Treatment - A Mini Review

Nanoparticles have emerged as promising tools for cancer treatment due to their ability to selectively deliver drugs to the tumor site while avoiding significant systemic side effects. Chitosan nanoparticles, among various types of nanoparticles, have gained significant attention due to their biocompatibility, biodegradability, and local drug delivery capacity. The electrospray technique is an efficient method for preparing chitosan nanoparticles, offering reproducibility, scalability, and high drug encapsulation efficiency. This technique has gained popularity due to its ease of use and flexibility in meeting various demands of nanoparticle production. Recent studies have investigated the potential of chitosan nanoparticles prepared by electrospray technique to encapsulate a range of drugs. The method leverages active surface absorption, binding, or complexation with drugs. For example, chitosan-based nanoparticles loaded with DOX and QUE achieved high encapsulation efficiency of 83% and effectively inhibited the growth of HCT-116 cancer cells. Similarly, SNP-CH-DOX-CM nanoparticles showed significant anti-cancer activity against HepG2 tumors. However, it should be noted that the toxicity of nanoparticles is directly related to the concentration of the active substance. Therefore, careful optimization of drug dosing is necessary to minimize any potential toxicity.

THMs removal from aqueous solution using hydrochar enhanced by chitosan nanoparticles: preparation, characterization, kinetics, equilibrium studies

In this study, novel green and economical techniques were established to synthesize modified banana peel hydrochar (HBP) through hydrothermal carbonization. Chitosan nanoparticles, a green adsorbent with multiple features, were prepared, characterized, and used to enhance the performance of the hydrochar in THMs removals. The optimal method for hydrothermal carbonization was determined based on previously cited methods. The dried banana peels and HBP were characterized using Fourier transform infrared spectrometer (FTIR), pH, zero point of charge, and bulk density. The prepared chitosan nanoparticles were characterized using scanning electron microscope (SEM), transmission electron microscope (TEM), and zeta potential techniques. The average size of chitosan nanoparticles was 12.1 nm, and a 1:1 mixture of HBP and chitosan nanoparticles was used for the removal of THMs from an aqueous solution. The concentrations of THMs were measured using gas chromatography, and the effect of THMs’ initial concentrations, adsorbent doses, temperature, and pH value on the adsorption capacity was studied. The results showed that the 1:1 mixture of HBP and chitosan nanoparticles was an efficient adsorbent for removing THMs from aqueous solutions. This adsorbent was found to be effective in removing THMs from the aqueous solution, even at low temperature and pH values, with an estimated maximum sorption capacity (qm) of 588.23 g g−1, exceeding numerous other adsorbents previously studied, and the regression coefficient (R2 = 0.9959) for data fitting with the Freundlich isotherm. This research has the potential to offer a great avenue for future development and implementation in water treatment. It could lead to improved methods of treating water, which would be beneficial for both environmental protection and economic growth. The developed adsorbent had high adsorption affinity toward the THMs compounds, making the adsorption method efficient and viable technique for treating water containing such contaminants.

Sustainable functionalized chitosan based nano-composites for wound dressings applications: A review

Functionalized chitosan nanocomposites have been studied for wound dressing applications due to their excellent antibacterial and anti-fungal properties. Polysaccharides show excellent antibacterial and drug-release properties and can be utilized for wound healing. In this article, we comprise distinct approaches for chitosan functionalization, such as photosensitizers, dendrimers, graft copolymerization, quaternization, acylation, carboxyalkylation, phosphorylation, sulfation, and thiolation. The current review article has also discussed brief insights on chitosan nanoparticle processing for biomedical applications, including wound dressings. The chitosan nanoparticle preparation technologies have been discussed, focusing on wound dressings owing to their targeted and controlled drug release behavior. The future directions of chitosan research include; a) finding an effective solution for chronic wounds, which are unable to heal completely; b) providing effective wound healing solutions for diabetic wounds and venous leg ulcers; c) to better understanding the wound healing mechanism with such materials which can help provide the optimum solution for wound dressing; d) to provide an improved treatment option for wound healing.

Preparation, characterization and protective effect of chitosan - Tripolyphosphate encapsulated dihydromyricetin nanoparticles on acute kidney injury caused by cisplatin

Dihydromyricetin (DMY) is a natural dihydroflavonol compound known for its diverse pharmacological benefits. However, its limited stability and bioavailability posed significant challenges for further applications. To address these issues, in this study, an ion crosslinking method was utilized to prepare chitosan nanoparticles that were loaded with DMY. The synthesized chitosan nanoparticles (CS-DMY-NPs) were spherical in shape with particle size and ζ potential of 198.7 nm and 45.05 mV, respectively. Furthermore, in vitro release experiments demonstrated that CS-DMY-NPs had sustained release and protective effects in simulated gastric and intestinal fluids. CS-DMY-NPs exhibited better antioxidant activity by ABTS and DPPH radical scavenging activity than free DMY. In vivo study showed that CS-DMY-NPs alleviated cisplatin-induced kidney damage by inhibiting oxidative stress and proinflammatory cytokines, and had better activity compared to DMY (free). Immunofluorescence data showed that CS-DMY-NPs activated the Nrf2 signaling pathways in a dose-dependent manner to combat cisplatin-induced kidney damage. Our results demonstrate that CS-TPP has good compatibility with DMY, and CS-DMY-NPs exhibited better protective effects against cisplatin-induced acute kidney injury (AKI) than free DMY.

Chitosan Nanoparticles-Preparation, Characterization and Their Combination with Ginkgo biloba Extract in Preliminary In Vitro Studies.

Nanoparticles (NPs), due to their size, have a key position in nanotechnology as a spectrum of solutions in medicine. NPs improve the ability of active substances to penetrate various routes: transdermal, but also digestive (active endocytosis), respiratory and injection. Chitosan, an N-deacetylated derivative of chitin, is a natural biodegradable cationic polymer with antioxidant, anti-inflammatory and antimicrobial properties. Cross-linked chitosan is an excellent matrix for the production of nanoparticles containing active substances, e.g., the Ginkgo biloba extract (GBE). Chitosan nanoparticles with the Ginkgo biloba extract (GBE) were obtained by ion gelation using TPP as a cross-linking agent. The obtained product was characterized in terms of morphology and size based on SEM and Zeta Sizer analyses as well as an effective encapsulation of GBE in nanoparticles-FTIR-ATR and UV-Vis analyses. The kinetics of release of the active substance in water and physiological saline were checked. Biological studies were carried out on normal and cancer cell lines to check the cytotoxic effect of GBE, chitosan nanoparticles and a combination of the chitosan nanoparticles with GBE. The obtained nanoparticles contained and released GBE encapsulated in research media. Pure NPs, GBE and a combination of NPs and the extract showed cytotoxicity against tumor cells, with no cytotoxicity against the physiological cell line.

Preparation of chitosan nanoparticles for simultaneous drug delivery of dacarbazine and enoxaparin in melanoma

The aim of this study was to investigate the anti-melanoma and anti-angiogenic effects of enoxaparin surface-coated dacarbazine-loaded chitosan nanoparticles (Enox-Dac-Chi NPs). The prepared Enox-Dac-Chi NPs had a particle size of 367.95 ± 1.84 nm, zeta potential of −7.12 ± 0.25 mV, efficiency of drug loading (DL%) of 73.90 ± 3.84 %, and attached enoxaparin percentage of 98.53 ± 0.96 %. Both drugs had extended-release profiles and approximately 96 % of enoxaparin and 67 % dacarbazine were released within 8 h. The Enox-Dac-Chi NPs with IC50 of 59.60 ± 1.25 μg/ml were the most cytotoxic against melanoma cancer cells compared with chitosan nanoparticles containing only dacarbazine (Dac-Chi NPs) and free dacarbazine. There was no significant difference between the cellular uptake of Chi NPs and enoxaparin coated Chi NPs (Enox-Chi NPs) in B16F10 cells. Enox-Chi NPs with an average anti-angiogenic score of 1.75 ± 0.125 had more anti-angiogenic effect than enoxaparin. The results showed that simultaneous delivery of dacarbazine and enoxaparin by chitosan nanoparticles can enhance the anti-melanoma effect of dacarbazine. Additionally, enoxaparin can prevent the melanoma metastasis by its anti-angiogenic activity. Thus, the designed nanoparticles can be introduced as effective drug delivery vehicles for the treatment and prevention of metastatic melanoma.

Fabrication of Nanoformulation Containing Carvedilol and Silk Protein Sericin against Doxorubicin Induced Cardiac Damage in Rats.

Nanotechnology has emerged as an inspiring tool for the effective delivery of drugs to help treat Coronary heart disease (CHD) which represents the most prevalent reason for mortality and morbidity globally. The current study focuses on the assessment of the cardioprotective prospective ofanovel combination nanoformulation of sericin and carvedilol. Sericin is a silk protein obtained from Bombyx mori cocoon and carvedilol is a synthetic nonselective β-blocker. In this present study, preparation of chitosan nanoparticles was performed via ionic gelation method and were evaluated for cardioprotective activity in doxorubicin (Dox)-induced cardiotoxicity. Serum biochemical markers of myocardial damage play a substantial role in the analysis of cardiovascular ailments and their increased levels have been observed to be significantly decreased in treatment groups. Treatment groups showed a decline in the positivity frequency of the Troponin T test as well. The NTG (Nanoparticle Treated Group), CSG (Carvedilol Standard Group), and SSG (Sericin Standard Group) were revealed to have reduced lipid peroxide levels (Plasma and heart tissue) highly significantly at a level of p < 0.01 in comparison with the TCG (Toxic Control Group). Levels of antioxidants in the plasma and the cardiac tissue were also established to be within range of the treated groups in comparison to TCG. Mitochondrial enzymes in cardiac tissue were found to be elevated in treated groups. Lysosomal hydrolases accomplish a significant role in counteracting the inflammatory pathogenesis followed by disease infliction, as perceived in the TCG group. These enzyme levels in the cardiac tissue were significantly improved after treatment with the nanoformulation. Total collagen content in the cardiac tissue of the NTG, SSG, and CSG groups was established to be highly statistically significant at p < 0.001 as well as statistically significant at p < 0.01, respectively. Hence, the outcomes of this study suggest that the developed nanoparticle formulation is effective against doxorubicin-induced cardiotoxicity.

Chitosan nanoparticles: Current aspects

Nanoparticles are currently becoming one of the important parts for various segments. Much more process/application becomes effective or more economical and time-saving because of nanoparticles. Polymers with distinct characteristics are widely used but nature-origin polymer has important properties that make a polymeric material for nanoparticles. Many methods were successfully evaluated to prepare chitosan nanoparticles for loading various compounds including ionotropic gelation, micelles, and emulsification methods. In this article, we have reviewed the current aspects made-up of chitosan nanoparticles. Chitosan nanoparticles have an application to improve bioavailability, controlled release of the loaded drug also increases cellular uptake and targeting to cancer cells, stabilization of proteins, and enhanced the effectivity of anti-microbial agents. In agriculture, chitosan nanoparticles are used for herbicides, insecticides, and pesticide loading to improve the cultivation of crops and are used in food packaging. Chitosan nanoparticles also have wide applications as implant and therapeutic agent.

Curcumin-Loaded Chitosan Nanoparticle Preparation and Its Protective Effect on Celecoxib-induced Toxicity in Rat isolated Cardiomyocytes and Mitochondria.

Curcumin has a wide range of pharmacological activities, including antioxidant, anti-inflammatory and tissue protective. In here we hypothesized that curcumin-loaded chitosan-coated solid lipid nanoparticles (CuCsSLN) are able to increase its overall bioavailability and hence its antioxidant and mitochondria;/lysosomal protective properties of curcumin. CuCsSLN were prepared using solvent diffusion technique for formation of solid lipid nanoparticles (SLNs) and electrostatic coating of positive-charged chitosan to negative surface of SLNs. CuCsSLN showed the encapsulation efficiency of 91.4±2.7%, the mean particle size of 208±9 nm, the polydispersity index of 0.34±0.07, and the zeta potential of+53.5±3.7 mV. The scanning electron microscope (SEM) images of nanoparticles verified their nanometric size and also spherical shape. Curcumin was released from CuCsSLN in a sustain release pattern up to 24 hours. Then isolated cardiomyocytes and mitochondria were simultaneously treated with (1) control (0.05% ethanol), (2) celecoxib (20 µg/ml) treatment, (3) celecoxib (20 µg/ml)+++CuCsSLN (1 µg/ml) treatment, (4) CuCsSLN (1 µg/ml) treatment, (5) celecoxib (20 µg/ml)+++curcumin (10 µM) treatment and (6) curcumin (10 µM) treatment for 4 h at 37°C. The results showed that celecoxib (20 µg/ml) induced a significant increase in cytotoxicity, reactive oxygen species (ROS) formation, mitochondria membrane potential (ΔΨm) collapse, lipid peroxidation, oxidative stress and mitochondrial swelling while CuCsSLN and curcumin reverted the above toxic effect of celecoxib. Our data indicated that the effect of CuCsSLN in a number of experiments, is significantly better than that of curcumin which shows the role of chitosan nanoparticles in increasing effect of curcumin.

Transferrin-modified chitosan nanoparticles for targeted nose-to-brain delivery of proteins

Nose-to-brain delivery presents a promising alternative route compared to classical blood–brain barrier passage, especially for the delivery of high molecular weight drugs. In general, macromolecules are rapidly degraded in physiological environment. Therefore, nanoparticulate systems can be used to protect biomolecules from premature degradation. Furthermore, targeting ligands on the surface of nanoparticles are able to improve bioavailability by enhancing cellular uptake due to specific binding and longer residence time. In this work, transferrin-decorated chitosan nanoparticles are used to evaluate the passage of a model protein through the nasal epithelial barrier in vitro. It was demonstrated that strain-promoted azide–alkyne cycloaddition reaction can be utilized to attach a functional group to both transferrin and chitosan enabling a rapid covalent surface-conjugation under mild reaction conditions after chitosan nanoparticle preparation. The intactness of transferrin and its binding efficiency were confirmed via SDS-PAGE and SPR measurements. Resulting transferrin-decorated nanoparticles exhibited a size of about 110–150 nm with a positive surface potential. Nanoparticles with the highest amount of surface bound targeting ligand also displayed the highest cellular uptake into a human nasal epithelial cell line (RPMI 2650). In an air–liquid interface co-culture model with glioblastoma cells (U87), transferrin-decorated nanoparticles showed a faster passage through the epithelial cell layer as well as increased cellular uptake into glioblastoma cells. These findings demonstrate the beneficial characteristics of a specific targeting ligand. With this chemical and technological formulation concept, a variety of targeting ligands can be attached to the surface after nanoparticle formation while maintaining cargo integrity.Graphical abstract

Enhancement of Antimicrobial and Dyeing Properties of Cellulosic Fabrics via Chitosan Nanoparticles.

The primary goal of this study is to prepare chitosan nanoparticles (CSNPs) by the ionic gelation method via the treatment of chitosan (0.2 wt.%) with tripolyphosphate (0.2 wt.%) ultrasonically for 45 min. FT-IR spectroscopy and TEM images were used to characterize and validate CSNP production. Cellulosic materials with different concentrations of CSNPs have better antibacterial and colouring characteristics. The treated cellulosic fabrics were analyzed by FT-IR spectroscopy, SEM, and thermogravimetric analysis. Colourimetric data measurements expressed in K/S values were used to evaluate the impact of CSNPs on the dyeing affinity of cellulosic materials. In addition, antibacterial activity against bacteria and fungi was tested on the treated cellulosic fabrics. According to the K/S values, cellulosic textiles treated with CSNPs (0.3 wt.%) had a better affinity for acid dyeing. These textiles also offer better antibacterial properties and are more resistant to washing, light, and rubbing. A cytotoxicity study found that CSNPs give cellulosic materials antibacterial and acid dyeing properties, which is good for the environment.

In vitro bioactivity and gene silencing effect of shRNA-VEGF loaded chitosan nanoparticles

Purpose: In this study, it is aimed to prepare chitosan nanoparticles containing shRNA-VEGF and evaluate their bioactivity by in vitro cell culture studies and to perform mechanical characterization of nanoparticles.&#x0D; Material and Methods: Ionic chelation method was used to prepare nanoparticles. The XTT assay was used to assess the cytotoxic activity of shRNA-VEGF and shRNA-VEGF loaded NP on the HeLa and NIH 3T3 cells.&#x0D; Results: According to the results IC50 values of shRNA-VEGF and NP including shRNA-VEGF were calculated. IC50 values of shRNA-VEGF and NP including shRNA-VEGF were 0.89±0.010 µg/mL and 0.52±0.004 µg/mL on HeLa cell line. Bax quantities of control, shRNA-VEGF, and shRNA-VEGF loaded NP was measured as 23.70±0.27 ng/mg protein, 34.64±0.36 ng/mg protein, and 39.46±0.54 ng/mg protein, respectively. According to the results, cleaved caspase 3 quantities of control, shRNA-VEGF, and shRNA-VEGF loaded NP was measured as 711.70±4.40 pg/mg protein, 767.23±3.82 pg/mg protein, and 825.32±5.06 pg/mg protein, respectively.&#x0D; Conclusion: shRNA-VEGF and shRNA-VEGF loaded NP significantly reduced HeLa cell reproduction in a concentration-dependent manner while generating no cytotoxicity in NIH 3T3 cells. The expression of pro-apoptotic Bax and cleaved caspase 3 proteins was significantly increased by shRNA-VEGF and shRNA-VEGF loaded NP.

Alginate-coated chitosan nanoparticles for pH-dependent release of tamoxifen citrate

Chitosan-based nano-sized particles increase the penetration of the drug through the narrow junction into the bloodstream and target the specific site. The objective of this study was to prepare chitosan nanoparticles to entrap a hydrophobic drug (tamoxifen citrate), followed by the alginate coating of the developed nanoparticles to decrease their degradation in the acidic pH. Drug-loaded chitosan nanoparticles were prepared by the ionic gelation method. Alginate coating was done by dissolving sodium alginate to buffer solution and drug-loaded chitosan nanoparticles drop-wise under mild agitation. The size of alginate coated chitosan nanoparticles, zeta potential, surface morphology, in-vitro drug release, and entrapment efficiency was measured. The optimised formulation of both uncoated (SH3) and coated (SH7) formulation showed the particle size, PDI, and zeta potential with values 221 & 338 nm, 0.161 & 0.230 and 36.5 & −20.7 mV, respectively. The resulted nanoparticle surface was non-porous. The percentage yield of the optimised formulation SH3 was 28% and SH6 was 33%. The entrapment efficiency of the optimised formulation SH3 (uncoated formulation) and SH6 (coated formulation) is 69.5 and 58.51%, respectively. Chitosan nanoparticles were successfully prepared to entrap tamoxifen citrate. The coating of chitosan nanoparticles decreased their degradation in the acidic pH.

Outstanding Approach to Enhance the Safety of Ready-to-Eat Rice and Extend the Refrigerated Preservation.

Rice is a broad-spectrum meal consumed annually in large amounts. Ready-to-eat rice is a member of dishes with a high risk of contamination. The present study aimed to increase the safety and shelflife of ready-to-eat rice during temporary storage. To prepare a mixture for extraction, three spices were chosen ginger: thyme:coriander (1:2:1). Two types of extract were prepared, aromatic and water extracts. The bioactive aromatic extract was preserved by encapsulation using chitosan nanoparticle preparation, while water extracts were prepared by warm diffusion. The aromatic extract possessed volatiles with antimicrobial features, including α-pinene, cymene, camphor, 1, 8 cineol, and limonene. The results expressed the extracts’ better antifungal and antibacterial effect, with a distinguishing aromatic one. Water extract was recorded as being rich in phenolic and flavonoids, like Salysilic, p-hydroxybenzoic acid, ferulic, Luteolin 7 glucoside, and quercitin. These molecules play functionality for microbial inhibition in the simulated media. Ready-to-eat rice shelflife was extended by applying the aromatic extract of the encapsulated mixture at the late stage of cooking and before packaging. It can preserve the samples for up to five days at room temperature and up to eight days of refrigerator storage (8 °C). However, water extract had lower activity as antibacterial and antifungal than the aromatic one. Again, water extract activity reduces fungal citrinin secretion by low efficiency more than the aromatic extract. These results recommended the addition of aromatic extract to the ready-to-eat rice meals as a final additive just before packaging.
