A unique substrate specificity of PonAAS2, an aromatic aldehyde synthase, involved in a phytohormone auxin biosynthesis in a gall-inducing sawfly Euura sp. "Pontania".

The aromatic aldehyde synthase (AAS), PonAAS2, from the gall-inducing sawfly has been identified as a biosynthetic enzyme for indole-3-acetic acid (IAA), a key molecule of the plant hormone auxin, which is thought to play a role in gall induction. Unlike other insect AASs that convert Dopa, PonAAS2 uniquely converts L-tryptophan (Trp) into indole-3-acetaldehyde, a precursor of IAA. In this study, an examination of AAS enzymes from various insect species revealed that the ability to convert Trp has been acquired in only a very limited taxonomic group. Comparative analysis between PonAAS2 and DjAAS2 from a gall wasp showed that, despite having conserved substrate-recognition amino acids, they exhibit different substrate specificities. This difference likely arises from variations in how these enzymes' monomers interact during dimer formation, as demonstrated by amino acid substitution experiments and structural predictions.

Utilization of Indole-3-acetic acid–Secreting Bacteria in Algal Environment to Increase Biomass Accumulation of Ochromonas and Chlorella

Application of plant hormones is an effective strategy to further improve the accumulation of microalgae biomass. In this study, the IAA (indole-3-acetic acid) production capacity of symbiotic bacteria isolated from the culture system of Ochromonas and Chlorella was detected and compared, with the bacterial community structure revealed. The results demonstrated that Algoriphagus, Porphyrobacter, Roseococcus, and Rhizobium were the dominant bacterial genera both in Ochromonas and Chlorella system, distributed in Proteobacteria and Bacteroidetes. Sixteen of 40 strains classified into 15 genera isolated from the two genera of microalgae could produce IAA, with Agrobacterium and Rhizobium as the main representatives with highest IAA production level (22.53–34.18 mg/L in 30 h). And through determination of changes of chlorophyll content in the co-culture system of microalgae and the supernatant of IAA high-yielding bacteria suspension from medium with or without tryptophan (synthetic precursor of IAA), Rhizobium (C-3) and Agrobacterium (O-16, C-2) were proved to promote the growth of Ochromonas and Chlorella through secreting IAA. The biomass yield of Ochromonas and Chlorella could be respectively increased by 78.98% and 72.73% through the co-culture with Agrobacterium (C-2), by 68.15% and 69.93% for Rhizobium (C-3) and by 80.89% and 65.73% for Agrobacterium (O-16). Moreover, compared with the application of exogenous IAA to improve the microalgae biomass, it is more efficient and economical to construct algae bacteria symbiosis system using IAA secretory bacteria commonly existed in microalgae culture system. In addition, the growth promoting effect of IAA secretory bacteria on microalgae was different among the species of algae.

Indole-3-butyric acid priming reduced cadmium toxicity in barley root tip via NO generation and enhanced glutathione peroxidase activity.

Activation of GPX and enhanced NO level play a key role in IBA-mediated enhanced Cd tolerance in young barley roots. Application of exogenous indole-3-acetic acid (IAA) or an IAA precursor improves the tolerance of plants to heavy metals. However, the physiology of these tolerance mechanisms remains largely unknown. Therefore, we studied the priming effect of indole-3-butyric acid (IBA), an IAA precursor, on mild and severe cadmium (Cd) stress-induced responses in roots of young barley seedlings. IBA, similarly to mild Cd stress, significantly increased the glutathione peroxidase (GPX) activity in the apexes of barley roots, which remained elevated after the IBA pretreatment as well. IBA pretreatment-evoked high nitric oxide generation in roots effectively reduced the high superoxide level under the severe Cd stress, leading to less toxic peroxynitrite accumulation accompanied by markedly reduced Cd-induced cell death. On the other hand, the IBA-evoked changes in IAA homeostasis resulted in root growth reorientation from longitudinal elongation to radial swelling. However, the application of an IAA signaling inhibitor, following the activation of defense responses by IBA, was able to promote root growth even at high concentrations of Cd. Based on the results, it can be concluded that the application of IBA, as an effective activator of Cd tolerance mechanisms in young barley roots, and the subsequent use of an IAA signaling inhibitor for the inhibition of root morphogenic responses induced by altered auxin metabolism, results in a high degree of root Cd tolerance, helping it to withstand even the transient exposure to lethal Cd concentration without the absolute inhibition of root growth.

A role for the auxin precursor anthranilic acid in root gravitropism via regulation of PIN-FORMED protein polarity and relocalisation in Arabidopsis.

Auxin flow is directed by the subcellular polar distribution and dynamic relocalisation of auxin transporters such as the PIN-FORMED (PIN) efflux carriers, which can be influenced by the main natural plant auxin indole-3-acetic acid (IAA). Anthranilic acid (AA) is an important early precursor of IAA and previously published studies with AA analogues have suggested that AA may also regulate PIN localisation. Using Arabidopsis thaliana as a model species, we studied an AA-deficient mutant displaying agravitropic root growth, treated seedlings with AA and AA analogues and transformed lines to over-produce AA while inhibiting its conversion to downstream IAA precursors. We showed that AA rescues root gravitropic growth in the AA-deficient mutant at concentrations that do not rescue IAA levels. Overproduction of AA affects root gravitropism without affecting IAA levels. Treatments with, or deficiency in, AA result in defects in PIN polarity and gravistimulus-induced PIN relocalisation in root cells. Our results revealed a previously unknown role for AA in the regulation of PIN subcellular localisation and dynamics involved in root gravitropism, which is independent of its better known role in IAA biosynthesis.

Short-term salt stress in Brassica rapa seedlings causes alterations in auxin metabolism

Salinity is one of major abiotic stresses affecting Brassica crop production. Here we present investigations into the physiological, biochemical, and hormonal components of the short-term salinity stress response in Chinese cabbage seedlings, with particular emphasis on the biosynthesis and metabolism of auxin indole-3-acetic acid (IAA). Upon salinity treatments (50–200 mM NaCl) IAA level was elevated in a dose dependent manner reaching 1.6-fold increase at the most severe salt treatment in comparison to the control. IAA precursor profiling suggested that salinity activated the indole-3-acetamide and indole-3-acetaldoxime biosynthetic pathways while suppressing the indole-3-pyruvic acid pathway. Levels of the IAA catabolites 2-oxoindole-3-acetic acid and indole-3-acetic acid-aspartate increased 1.7- and 2.0-fold, respectively, under the most severe treatment, in parallel with those of IAA. Conversely, levels of the ester conjugate indole-3-acetyl-1-O-ß-d-glucose and its catabolite 2-oxoindole-3-acetyl-1-O-ß-d-glucose decreased 2.5- and 7.0-fold, respectively. The concentrations of stress hormones including jasmonic acid and jasmonoyl-isoleucine (JA and JA-Ile), salicylic acid (SA) and abscisic acid (ABA) confirmed the stress induced by salt treatment: levels of JA and JA-Ile increased strongly under the mildest treatment, ABA only increased under the most severe treatment, and SA levels decreased dose-dependently. These hormonal changes were related to the observed changes in biochemical stress markers upon salt treatments: reductions in seedling fresh weight and root growth, decreased photosynthesis rate, increased levels of reactive oxygen species, and elevated proline content and the Na+/K+ ratio. Correlations among auxin profile and biochemical stress markers were discussed based on Pearson's coefficients and principal component analysis (PCA).

Characterization of auxin production plant growth promotion by a bacterium isolated from button mushroom compost

An auxin-producing bacterial strain, designated 4-3, was isolated from waste button mushroom compost in Boryeong-si, Chungnam. The strain 4-3 was classified as a novel strain of Leucobacter tardus, based on chemotaxonomic and phylogenetic analyses. TLC and HPLC the isolated L. tardus strain 4-3 produced indole-3-acetic acid (IAA), the auxin. Maximum IAA productionof 94.3 mg L-1 was detected for bacteria cultured in R2A medium with 0.1% l-tryptophan, incubated for 24 h at 35oC. Negative correlationwas observed between IAA production and pH of the culture medium, indicating that the increase inIAA caused acidification ofthe medium. The effect of supplementation with varying concentrations of l-tryptophan, a known precursor of IAA, was also assessed. production was maximal at 0.1% l, but decreased at lconcentrations above 0.2%. To investigate the plant growth-promoting effects of the bacterium, L. tardus strain 4-3 culture broth was used to inoculate water cultures and seed pots of mung bean. We found thatadventitious root induction and root growth were 2.2-times higher in thethan in the non-inoculated plants.

Variation in Indole-3-Acetic Acid Production by Wild Saccharomyces cerevisiae and S. paradoxus Strains from Diverse Ecological Sources and Its Effect on Growth.

Phytohormone indole-3-acetic acid (IAA) is the most common naturally occurring and most thoroughly studied plant growth regulator. Microbial synthesis of IAA has long been known. Microbial IAA biosynthesis has been proposed as possibly occurring through multiple pathways, as has been proven in plants. However, the biosynthetic pathways of IAA and the ecological roles of IAA in yeast have not been widely studied. In this study, we investigated the variation in IAA production and its effect on the growth of Saccharomyces cerevisiae and its closest relative Saccharomyces paradoxus yeasts from diverse ecological sources. We found that almost all Saccharomyces yeasts produced IAA when cultured in medium supplemented with the primary precursor of IAA, L-tryptophan (L-Trp). However, when cultured in medium without L-Trp, IAA production was only detected in three strains. Furthermore, exogenous added IAA exerted stimulatory and inhibitory effects on yeast growth. Interestingly, a negative correlation was observed between the amount of IAA production in the yeast cultures and the IAA inhibition ratio of their growth.

Accumulation of Indole‐3‐Acetic Acid in Rice sl Mutant Leaves Infected with Bipolaris oryzae

AbstractRice leaves accumulate serotonin in response to infection by Bipolaris oryzae. The leaves of the sl mutant, which is deficient in the gene encoding tryptamine 5‐hydroxylase, accumulate tryptamine instead of serotonin upon infection by B. oryzae. Because tryptamine is a possible precursor of indole‐3‐acetic acid (IAA), we investigated the accumulation of IAA in sl leaves infected with B. oryzae. Liquid chromatography coupled with tandem mass spectrometry analysis indicated that IAA accumulated at approximately 1.5 μmol/gFW in the leaves of sl mutant. This accumulation was suppressed by 95% by the treatment with the tryptamine decarboxylase inhibitor, (S)‐α‐(fluoromethyl)tryptophan, at 100 μm, indicating that tryptamine served as the precursor of IAA. The accumulation of IAA was not reproduced by treatment with CuCl2 or by exogenous feeding of tryptamine. Furthermore, inoculation of Magnaporthe grisea induced only a lower level of IAA accumulation. On the other hand, B. oryzae produced IAA in culture media containing tryptamine. These findings strongly suggested that the metabolism of tryptamine by B. oryzae was responsible for IAA accumulation in the leaves of the sl mutant. Serotonin added to the culture media was also converted into 5‐hydroxyindole‐3‐acetic acid (5HIAA) at a rate similar to that of tryptamine. Considering that wild‐type rice leaves accumulate serotonin for defensive purposes, reducing the concentration of serotonin by conversion into 5HIAA may be significant as a detoxification process in the interaction between B. oryzae and rice.

Quantitative Auxin Metabolite Profiling Using Stable Isotope Dilution UHPLC-MS/MS.

Auxin, a plant hormone, is able to stimulate growth and to control developmental processes. To get a deeper level of understanding of how auxin activity is regulated in planta, we have developed a sensitive and selective mass spectrometry-based protocol for multiplex quantification of indole 3-acetic acid (IAA) as well as its precursors and degradation products. As part of the protocol development, we have also established a derivatization protocol for the IAA precursors IPyA and IAAld, compounds that are highly labile during sample extraction and purification. This article describes a standard isotope dilution method for quantitative profiling of the IAA metabolome in small amounts of plant tissues. © 2016 by John Wiley & Sons, Inc.

Many rhizobacteria can promote plant growth through various direct or indirect mechanisms, and their production of phytohormones such as indole-3-acetic acid (IAA) may have pronounced effects on growth and development of plants. Rhizobacterial strain isolated from rhizosphere of foxtail (Setaria viridis), Acinetobacter sp. SW5 produced 118.1 mg/L of IAA and 4.5 mg/L of gibberellin (GA3) in brain heart broth medium at 2 and 1 day of incubation, respectively. In a pot test the lengths of stem and root and fresh weight of the germinated tomato seedlings treated with Acinetobacter sp. SW5 significantly increased by 26.3, 33.3, and 105.3%, respectively compared to those of the uninoculated control in 12 weeks of cultivation. When the root exudate secreted from tomato seedlings was analyzed by HPLC, 3.75 ng mg tomato root -1 of tryptophan which is an IAA precursor was detected. Acinetobacter sp. SW5 could produce 4.06 μM of IAA from root exudate from 8 tomato seedlings. Together with the capability of growth of Acinetobacter sp. SW5 in the tomato root exudates, this IAA secreted by bacteria might contribute to enhance the growth of tomato plants.

The main auxin biosynthesis pathway in Arabidopsis

The phytohormone auxin plays critical roles in the regulation of plant growth and development. Indole-3-acetic acid (IAA) has been recognized as the major auxin for more than 70 y. Although several pathways have been proposed, how auxin is synthesized in plants is still unclear. Previous genetic and enzymatic studies demonstrated that both TRYPTOPHAN AMINOTRANSFERASE OF ARABIDOPSIS (TAA) and YUCCA (YUC) flavin monooxygenase-like proteins are required for biosynthesis of IAA during plant development, but these enzymes were placed in two independent pathways. In this article, we demonstrate that the TAA family produces indole-3-pyruvic acid (IPA) and the YUC family functions in the conversion of IPA to IAA in Arabidopsis (Arabidopsis thaliana) by a quantification method of IPA using liquid chromatography-electrospray ionization-tandem MS. We further show that YUC protein expressed in Escherichia coli directly converts IPA to IAA. Indole-3-acetaldehyde is probably not a precursor of IAA in the IPA pathway. Our results indicate that YUC proteins catalyze a rate-limiting step of the IPA pathway, which is the main IAA biosynthesis pathway in Arabidopsis.

Erratum to: Levels of endogenous abscisic acid and indole-3-acetic acid influence shoot organogenesis in callus cultures of rice subjected to osmotic stress

Osmotic stress and endogenous hormone levels may have a role in shoot organogenesis, but a systematic study has not yet to investigate the links. We evaluated the changes of the endogenous indole-3-acetic acid (IAA) and abscisic acid (ABA) levels in rice (Oryza sativa L. cv. Tainan 5) callus during shoot organogenesis induced by exogenous plant growth regulator treatments or under osmotic stress. Non-regenerable callus showed low levels of endogenous ABA and IAA, with no fluctuation in level during the period evaluated. The addition of 100 μM ABA or 2 mM anthranilic acid (IAA precursor) into Murashige and Skoog basal induction medium containing 10 μM 2,4-D enhanced the regeneration frequency slightly, to 5 and 35%, respectively, and their total cellular ABA or IAA levels were increased significantly, correspondingly to the treatments. However, the regeneration frequency was greatly increased to 80% after treatment with 0.6 M sorbitol or 100 μM ABA and 2 mM anthranilic acid combined. Both treatments produced high levels of total cellular ABA and IAA at the callus stage, which was quickly decreased on the first day after transfer to regeneration medium. Thus, osmotic stress-induced simultaneous accumulation of endogenous ABA and IAA is involved in shoot regeneration in rice callus.

Tryptophan Regulates Thaxtomin A and Indole-3-Acetic Acid Production in Streptomyces scabiei and Modifies Its Interactions with Radish Seedlings

The virulence of Streptomyces scabiei, the causal agent of common scab, depends mainly on the production of the toxin thaxtomin A. S. scabiei also produces indole-3-acetic acid (IAA) but the role of this hormone in the interaction between pathogenic streptomycetes and plants has not yet been elucidated. Tryptophan is a biosynthetic precursor of both IAA and thaxtomin A. In this study, the effect of tryptophan on thaxtomin A and IAA production as well as its effect on the transcription of the corresponding biosynthetic genes in S. scabiei has been analyzed. In vitro IAA production depended on the availability of tryptophan. However, addition of this amino acid to the culture medium inhibited the biosynthesis of thaxtomin A. Expression of thaxtomin A biosynthetic genes nos and txtA were strongly repressed in the presence of tryptophan; however, modulation of the expression was not observed for the IAA biosynthetic genes iaaM and iaaH. The effects of an exogenous tryptophan supply on S. scabiei virulence were assessed on radish seedlings. Addition of tryptophan reduced symptoms on inoculated radish roots compared with seedlings grown in the absence of the bacterium, by way of inhibition of thaxtomin A production and increase of IAA biosynthesis.

Production of indole-3-acetic acid by the cyanobacterium Arthrospira platensis strain MMG-9.

The filamentous cyanobacterium Arthrospira platensis strain MMG-9 was isolated from a rice field. The ability of this strain to synthesize the bioactive compound indole-3-acetic acid (IAA) was demonstrated. IAA was extracted from the culture A. platensis strain MMG-9 and its identity was confirmed by thin layer chromatography (TLC) as well as by high performance liquid chromatography (HPLC). The IAA precursor L-tryptophan was required for IAA biosynthesis. Released IAA increased with the increase of the initial concentration of L-tryptophan in the medium and with the incubation time. A. platensis strain MMG-9 accumulates more IAA than it released it into the medium. The bioactivity of the secreted IAA was shown by its effect on the formation of roots by Pisum sativum. There was a significant positive effect of the supernatant of cultures of A. platensis strain MMG-9 on the number of lateral roots of P. sativum while a negative effect on root length was observed.

TheArabidopsisPLEIOTROPIC DRUG RESISTANCE8/ABCG36 ATP Binding Cassette Transporter Modulates Sensitivity to the Auxin Precursor Indole-3-Butyric Acid

Plants have developed numerous mechanisms to store hormones in inactive but readily available states, enabling rapid responses to environmental changes. The phytohormone auxin has a number of storage precursors, including indole-3-butyric acid (IBA), which is apparently shortened to active indole-3-acetic acid (IAA) in peroxisomes by a process similar to fatty acid beta-oxidation. Whereas metabolism of auxin precursors is beginning to be understood, the biological significance of the various precursors is virtually unknown. We identified an Arabidopsis thaliana mutant that specifically restores IBA, but not IAA, responsiveness to auxin signaling mutants. This mutant is defective in PLEIOTROPIC DRUG RESISTANCE8 (PDR8)/PENETRATION3/ABCG36, a plasma membrane-localized ATP binding cassette transporter that has established roles in pathogen responses and cadmium transport. We found that pdr8 mutants display defects in efflux of the auxin precursor IBA and developmental defects in root hair and cotyledon expansion that reveal previously unknown roles for IBA-derived IAA in plant growth and development. Our results are consistent with the possibility that limiting accumulation of the IAA precursor IBA via PDR8-promoted efflux contributes to auxin homeostasis.

Arabidopsis IAR4 Modulates Auxin Response by Regulating Auxin Homeostasis

In a screen for enhancers of tir1-1 auxin resistance, we identified two novel alleles of the putative mitochondrial pyruvate dehydrogenase E1alpha-subunit, IAA-Alanine Resistant4 (IAR4). In addition to enhancing the auxin response defects of tir1-1, iar4 single mutants exhibit numerous auxin-related phenotypes including auxin-resistant root growth and reduced lateral root development, as well as defects in primary root growth, root hair initiation, and root hair elongation. Remarkably, all of these iar4 mutant phenotypes were rescued when endogenous indole-3-acetic acid (IAA) levels were increased by growth at high temperature or overexpression of the YUCCA1 IAA biosynthetic enzyme, suggesting that iar4 mutations may alter IAA homeostasis rather than auxin response. Consistent with this possibility, iar4 mutants exhibit increased Aux/IAA stability compared to wild type under basal conditions, but not in response to an auxin treatment. Measurements of free IAA levels detected no significant difference between iar4-3 and wild-type controls. However, we consistently observed significantly higher levels of IAA-amino acid conjugates in the iar4-3 mutant. Furthermore, using stable isotope-labeled IAA precursors, we observed a significant increase in the relative utilization of the Trp-independent IAA biosynthetic pathway in iar4-3. We therefore suggest that the auxin phenotypes of iar4 mutants are the result of altered IAA homeostasis.

A New Gene for Auxin Synthesis

There is much interest in understanding the pathways that trigger biosynthesis of the plant hormone auxin. In this issue, Stepanova et al. (2008) and Tao et al. (2008) reveal that a small family of tryptophan aminotransferases catalyze formation of indole-3-pyruvic acid (IPA) from L-tryptophan (L-Trp), the first step in a pathway for auxin biosynthesis.

Production of auxin and other indolic and phenolic compounds by Paenibacillus polymyxa strains isolated from different proximity to plant roots

We determined the actual (no tryptophan, Trp, addition) and potential (Trp 0.1 g l−1 added) production of indolic and phenolic compounds produced in different culture media by Paenibacillus polymyxa strains which had been isolated from different proximity to wheat roots. The production of indole-3-acetic acid (IAA) and related indolic metabolites by all strains tested was greatly enhanced upon Trp addition, suggesting that Trp is important as IAA precursor. A gradual decrease in potential IAA and an increase in potential indole-3-ethanol and indole-3-lactic acid production by strains isolated from non-rhizosphere soil (NRS) over rhizosphere soil (RS) to the rhizoplane (RP) were obtained. The NRS strain showed also a higher (potential) production of oxidized compounds such as indole-3-carboxylic and benzoic acid besides IAA. The determined metabolic differences indicate together with previous results on genetic and physiological parameters of P. polymyxa populations that plant roots select different P. polymyxa subpopulations. Our results suggest that the selected genotypes are differentially adapted to the decreasing oxygen pressure from NRS to RP. Possible effects of the probably microhabitat-specific metabolic profiles of the P. polymyxa isolates for plant growth are discussed.

IAR4, a gene required for auxin conjugate sensitivity in Arabidopsis, encodes a pyruvate dehydrogenase E1alpha homolog.

The formation and hydrolysis of indole-3-acetic acid (IAA) conjugates represent a potentially important means for plants to regulate IAA levels and thereby auxin responses. The identification and characterization of mutants defective in these processes is advancing the understanding of auxin regulation and response. Here we report the isolation and characterization of the Arabidopsis iar4 mutant, which has reduced sensitivity to several IAA-amino acid conjugates. iar4 is less sensitive to a synthetic auxin and low concentrations of an ethylene precursor but responds to free IAA and other hormones tested similarly to wild type. The gene defective in iar4 encodes a homolog of the E1alpha-subunit of mitochondrial pyruvate dehydrogenase, which converts pyruvate to acetyl-coenzyme A. We did not detect glycolysis or Krebs-cycle-related defects in the iar4 mutant, and a T-DNA insertion in the IAR4 coding sequence conferred similar phenotypes as the originally identified missense allele. In contrast, we found that disruption of the previously described mitochondrial pyruvate dehydrogenase E1alpha-subunit does not alter IAA-Ala responsiveness or confer any obvious phenotypes. It is possible that IAR4 acts in the conversion of indole-3-pyruvate to indole-3-acetyl-coenzyme A, which is a potential precursor of IAA and IAA conjugates.

Identification of indole-3-acetic acid producing freshwater wetland rhizosphere bacteria associated with Juncus effusus L.

Production of indole-3-acetic acid (IAA), a key physiological feature of culturable, O2-tolerant bacteria associated with the freshwater macrophyte Juncus effusus L., was examined over a period of 2 years. Up to 74% of rhizobacteria identified and tested produced IAA. The number of indoleacetic acid producers decreased in winter. IAA was produced even when L-tryptophan, a precursor of IAA, was not added to the medium. Most of the IAA-producing strains were dominated by strains that were not identifiable to species level on the basis of API testing. Based on 16S rRNA gene sequencing and fatty acid analysis, it was found that IAA-producing rhizosphere bacteria associated with the freshwater wetland plant Juncus effusus L. are representatives of several families, including the Enterobacteriaceae, Pseudomonadaceae, Aeromonadaceae, Burkholderiaceae, and Bacillaceae. This study identifies numerous potentially important bacterial physiological groups of freshwater wetlands. Additionally, the study provides a baseline for monitoring and assessing the mutualistic relationships of wetland plants with rhizosphere bacteria in freshwater wetlands.
