Influence of medicinal plants’ extracts of the Altai region on hematological and biochemical profiles of laboratory mice

Purpose: to study the effect of oregano and marsh cinquefoil extracts on the hematological and biochemical profiles of laboratory mice.Materials and methods. The article presents a study of the effect of MC and O extracts on laboratory mice. The raw materials for the production of extracts were rhizomes of MC and above-ground parts of O collected in the Altai Krai. The effect of the extracts was studied on 54 two-month-old ICR mice of different sexes. Three experimental groups were formed: control, experimental with O extract (20 mg/kg) and experimental with MC extract (20 mg/kg). The following hematological and biochemical parameters were studied: the content of erythrocytes and hemoglobin, hematocrit, average volume of erythrocytes, the content of leukocytes and lymphocytes, the content of calcium, phosphorus, cholesterol, urea, glucose, triglycerides, total protein and albumin.Results. As a result of the study, it was revealed that the studied extracts are capable of having a significant effect on such metabolic parameters of laboratory mice as the content of glucose, cholesterol and total protein. The most significant effect was observed in female mice consuming the MC extract. Based on the results obtained, it was concluded that it is advisable to further study MC and O extracts as components for the development of hyperglycemic, hypolipidemic and restorative drugs based on medicinal plants.

Antioxidant activity of extracts of oregano and marsh cinquefoil, growing in the Altai region, in connection with their content of favonoids

Drugs based on medicinal plants have such advantages over synthetic analogues as low toxicity, better tolerability and reduced risk of addiction. Antioxidant agents in medicinal plants include substances from the flavonoid group, due to their ability to inactivate free radical processes, due to the mechanism of electron transfer between reaction components to neutralize free radicals. Among the medicinal plants of Western Siberia and the Altai Territory, oregano and marsh cinquefoil have a high content of flavonoids. The purpose of the study was to evaluate the antioxidant activity of alcohol-free extracts of oregano and marsh cinquefoil growing in the Altai region in connection with the content of flavonoids (flavonols and proanthocyanidins). To conduct the study, alcohol-free extracts of the studied plants were prepared using patented technology. The content of flavonols and proanthocyanidins was studied by arbitration techniques using spectrophotometry. The antioxidant activity of the extracts was measured using cathodic voltammetry method. The results showed that the average flavonol content in oregano extract was 866.5 μg/ml, while only traces were found in the extract of cinquefoil (60.0 μg/ ml). The average content of proanthocyanidins in the extract of cinquefoil was 4.05%, in the extract of oregano content of proanthocyanidins was 0.43%. The average coefficient of antioxidant activity for cinquefoil extract was 11.63 mmol/l×min and 3.81 mmol/l×min for oregano extract. From the results obtained, we can conclude that it is advisable to use extracts of marsh cinquefoil and oregano, growing in the Altai Territory, in the composition of biologically active antioxidant additives.

The genome sequence of the marsh cinquefoil, Comarum palustre L., also known as Potentilla palustris (L.) Scop. (Rosaceae)

We present a genome assembly from an individual Comarum palustre (the marsh cinquefoil; Streptophyta; Magnoliopsida; Rosales; Rosaceae). The genome sequence has a total length of 528.90 megabases. Most of the assembly is scaffolded into 21 chromosomal pseudomolecules suggesting the individual is an allohexaploid (2n = 6x = 42). The mitochondrial and plastid genome assemblies have lengths of 362.32 kilobases and 154.29 kilobases, respectively. Gene annotation of this assembly on Ensembl identified 37,459 protein-coding genes.

Complex influence of structural characteristics of low-methoxyl pectins on rheological properties of semi-solid Ca-gels

The semi-solid Ca-gels were prepared from low-methoxyl pectins (methoxylation degree DM 4–36 %) of Oberna behen, Potamogeton natans, Tanacetum vulgare, Comarum palustre, Bergenia crassifolia, and Heracleum sosnowskyi. Pectins are characterized by Mw of 77–556 kDa and different content of HG and RG-I. The gels were prepared by addition of Ca2+ ions at varying stoichiometric ratios, R = 2 × [Ca2+]/[COO−], of 0.2–0.6, and a fixed pectin concentration of 10 g/L. It was shown that the presence of certain sets of structural characteristics in low-methoxyl pectins from different sources leads to the formation of gels with similar rheological properties. Pectins with Mw 77–98 kDa, DM 6–14 % form stiffer Ca-gels than pectins with Mw 218–253 kDa, DM 24–36 % at the same R. Pectins with Mw 346–556 kDa form the strongest Ca-gels. Their high Mw compensates for the influence of DM on the properties of Ca-gels. The stability of gels in PBS (pH 7.4) is determined mainly by proportion of 1,4-linked non-methoxylated GalA in pectins. Pectins with DM 4–14 % form more stable Ca-gels in PBS than pectins with DM 21–36 %.

CONTENT OF CATECHINS IN LEAVES AND ROOTS OF COMARUM SALESOVIANUM AND COMARUM PALUSTRE (ROSACEAE)

The search for additional sources of catechins among the local flora is relevant today. The method for determining the content of catechins by the spectrophotometric method in the leaves of the subshrub Zalesov's cinquefoil (Comarum salesovianum (Steph.) Asch. et Graebn.) from the Rosaceae Juss. family was validated. The content of catechins in terms of (±)-catechin in the leaves and rhizomes of two species of the genus Comarum ‒ C. salesovianum and C. palustre L. (marsh cinquefoil) was studied. The highest content of catechins was found in the roots of C. palustre (4%) and leaves of C. salesovianum (2.58%). A method for determining the composition and content of individual catechins in the leaves and roots of two representatives of the genus Comarum by high-performance liquid chromatography has been proposed. Three catechins [(±)-catechin, epigallocatechin gallate and (-)-epicatechin] were found in leaves and roots. (±)-catechin and (-)-epicatechin are predominant in the leaves and roots of C. palustre and C. salesovianum. The content of epigallocatechin gallate does not exceed 0.1 mg/g. The concentration of (±)-catechin is highest in the roots (1.21 mg/g) of C. palustre, and (-)-epicatechin is highest in the leaves (1.35 mg/g) of C. salesovianum.

Evaluation of the antimicrobial ac- tivity of the extracts of oregano and marsh cinquefoil against gram-positive and gram-negative bacteria

One of the most critical problems in animal husbandry and public health is antibiotic resistance, associated with the widespread use of antibiotic drugs. As a result, pathogenic microorganisms develop antibiotic resistance, which creates the need to either increase their dosage or replace them with other medications. Phytobiotics are a promising alternative to antibiotic drugs. Phytobiotics are active substances of medicinal plants that exhibit antimicrobial properties against pathogenic microorganisms. The primary sources of antimicrobial activity of phytobiotics are phenolic and polyphenolic compounds. The research aimed to study the antimicrobial activity of oregano (O) extracts and marsh cinquefoil (MC). Alcohol extracts (AE) were prepared from dry raw materials by water-alcohol extraction, and then alcohol-free extracts (AFE) were obtained using vacuum distillation. The content of polyphenolic compounds was studied in the obtained extracts, and the hole method against pathogens such as Staphylococcus aureus, Escherichia coli and Salmonella enterica evaluated their antimicrobial properties. The results showed that in O AFE, the content of polyphenolic compounds was 1.487 mg/ml, in O AE 1.166 mg/ml, in MC AFE 1.349 mg/ml, and MC AE 1.074 mg/ml. All studied extracts showed antimicrobial activity against Staphylococcus aureus. The most significant zones of suppression (1.86 cm) were formed around the wells with O AFE, which were significantly (P<0.01) larger than in the samples with O AE. About gram-negative cultures, zones of suppression were found only in alcoholic extracts, which is most likely a demonstration of the antibacterial activity of ethanol.

Rare, protected, and understudied vascular plant species of the pinewood complex of the Mozh River valley (Kharkiv Region, Ukraine)

The composition of the biota is dynamic and changes for many reasons, making biodiversity research relevant. Studies of rare species that are most vulnerable to anthropogenic pressures are of particular importance. Although the Mozh River flows through the central part of the Kharkiv Region, its valley remains understudied, especially the pine forests and their rare plants. The results of the current study include data from our field research, revision of the Herbarium of V.N. Karazin Kharkiv National University (CWU), and literature sources from the second half of the 20th century to the present. This paper contains an annotated list of 35 species of vascular plants. Each species is provided with the data on its geographical range, distribution in Ukraine, population status and its study history in the Kharkiv Region, conservation status, and the state of its population in the studied forests. We found the localities of two species included in Appendix I of the Bern Convention (Salvinia natans (L.) All. and Jurinea cyanoides (L.) Rchb. and six species included in the Red Data Book of Ukraine (Botrychium lunaria (L.) Sw., Pulsatilla pratensis (L.) Mill., Iris pineticola Klok., Neottia nidus-avis (L.) Rich., Neottia ovata (L.) Hartm., Tulipa quercetorum Klok. & Zoz). We also found 19 species rare in the Kharkiv Region (Lycopodium clavatum L., Athyrium filix-femina (L.) Roth, Ophioglossum vulgatum L., Campanula persicifolia L., Chimaphila umbellata (L.) W.P.C. Barton, Comarum palustre L., Hottonia palustris L., Rubus saxatilis L., Carex pseudocyperus L., etc.). Nine regionally rare plant species have the boreal range type; they occur at the southern limit of their distribution range which indicates the specificity of ecological and climatic conditions formed in the pine forests in the Mozh River valley. In addition, we found five species without conservation status, but they are rare in the region (Caltha palustris L., Catolobus pendulus (L.) Al-Shehbaz, Gratiola officinalis L., Lythrum hyssopifolia L., Rubus polonicus Barr. ex Weston), and, therefore, they are recommended to be included in the next edition of the Official List of the Regionally Rare Plants of the Kharkiv Region. Three species (Buglossoides czernjajevii (Klokov & Des.-Shost.) Czerep., Jacobaea andrzejowskyi (Tzvelev) B.Nord. & Greuter, Sedum album L.) were identified as understudied in the Kharkiv region due to the lack of or insufficient information on their distribution in the study area.

Mass Spectrometric Fingerprint Mapping Reveals Species-Specific Differences in Plant Polyphenols and Related Bioactivities.

Plant species show large variation in the composition and content of their tannins and other polyphenols. These large metabolites are not easy to measure accurately, but they are important factors for species bioactivity and chemotaxonomy. Here, we used an automated group-specific UHPLC-DAD-MS/MS tool to detect and quantify eight most common polyphenol groups in 31 chemically diverse plant species representing many types of growth forms and evolutionary ages. Ten replicate plants were used for each species and two polyphenol-related bioactivities, i.e., protein precipitation capacity and oxidative activity were measured in all samples as well. By the help of a novel 2D fingerprint mapping tool we were able to visualize the qualitative and quantitative differences between the species in hydrolysable tannins (galloyl and hexahydroxydiphenoyl derivatives), proanthocyanidins (procyanidins and prodelphinidins), flavonols (kaempferol, quercetin and myricetin derivatives) and quinic acid derivatives together with the two bioactivities. The highest oxidative activities were found with species containing ellagitannins (e.g., Quercus robur, Geranium sylvaticum, Lythrum salicaria and Chamaenerion angustifolium) or prodelphinidin-rich proanthocyanidins (e.g., Ribes alpinum, Salix phylicifolia and Lysimachia vulgaris). The best species with high protein precipitation capacity were rich in gallotannins (Acer platanoides and Paeonia lactiflora) or oligomeric ellagitannins (e.g., Comarum palustre, Lythrum salicaria and Chamaenerion angustifolium). These types of tools could prove their use in many types of screening experiments and might reveal even unusually active polyphenol types directly from the crude plant extracts.

INFLUENCE OF ADDITIVES OF COMMON ORIGANUM AND MARSH CINQUEFOIL ON BIOINDICATION INDICATORS OF LARVAE GALLERIA MELLОNELLA L.

Galleria mellоnella L. is one of the main pests in beekeeping. Therefore, scientists are studying Galleria mellоnella L. to find methods to combat it and as a model object for physiological and biochemical studies. In addition, some medicinal plants contain active substances that affect the physiological parameters of insects and even exhibit insecticidal properties. In the study, the authors evaluated the effect of additives from dry powders of common origanum and marsh cinquefoil on the dynamics of weight change, survival and uniformity of the metamorphic composition in the Galleria mellоnella L. Larvae of the third age were selected for the experiment. Additives were added to the insect food substrate at 10, 50 and 100 mg per 3 g of the standard diet. For 16 days, the mass of insects and the stage of their development were recorded in the experimental and control groups. The results showed that the supplements studied had a stimulating effect on larval growth at low doses. In the experimental groups, a trend towards faster development of larvae was found. The average daily weight gain per larva in all experimental groups was significantly (P<0.01) more significant than in the control group. At a dosage of 100 mg of joint origanum supplement, the survival rate of insects was minimal and amounted to 80%. At dosages of 50 and 100 mg of additions of common origanum and marsh cinquefoil, a violation of the homogeneity of the age composition was observed. The data obtained allow us to conclude that it is promising to study further the impact of marsh cinquefoil and common oreganum additives on insect pests.

Plant phenology and species-specific traits control plant CH4 emissions in a northern boreal fen.

Aerenchymatic transport is an important mechanism through which plants affect methane (CH4 ) emissions from peatlands. Controlling environmental factors and the effects of plant phenology remain, however, uncertain. We identified factors controlling seasonal CH4 flux rate and investigated transport efficiency (flux rate per unit of rhizospheric porewater CH4 concentration). We measured CH4 fluxes through individual shoots of Carex rostrata, Menyanthes trifoliata, Betula nana and Salix lapponum throughout growing seasons in 2020 and 2021 and Equisetum fluviatile and Comarum palustre in high summer 2021 along with water-table level, peat temperature and porewater CH4 concentration. CH4 flux rate of C. rostrata was related to plant phenology and peat temperature. Flux rates of M. trifoliata and shrubs B. nana and S. lapponum were insensitive to the investigated environmental variables. In high summer, flux rate and efficiency were highest for C. rostrata (6.86 mg m-2 h-1 and 0.36 mg m-2 h-1 (μmol l-1 )-1 , respectively). Menyanthes trifoliata showed a high flux rate, but limited efficiency. Low flux rates and efficiency were detected for the remaining species. Knowledge of the species-specific CH4 flux rate and their different responses to plant phenology and environmental factors can significantly improve the estimation of ecosystem-scale CH4 dynamics in boreal peatlands.

The Influence of Shallow Peatland Water Quality on Characteristics of the Occurrence of Selected Herb Species in the Peatlands of Eastern Poland

The aim of the analysis was to compare the physicochemical variables of the quality of shallow groundwater in the peatlands of Eastern Poland in the context of the occurrence of selected herb species with similar habitat requirements: bogbean (Menyanthes trifoliata), small cranberry (Oxycoccus palustris), and purple marshlocks (Comarum palustre). The analysis of the quality variables of the shallow groundwater included the following physicochemical variables: reaction (pH), electrolytic conductivity (EC), dissolved organic carbon (DOC), total nitrogen (Ntot.), ammonium nitrogen (N-NH4), nitrite nitrogen (N-NO2), nitrate nitrogen (N-NO3), total phosphorus (Ptot.), phosphates (P-PO4), sulphates (SO2), sodium (Na), potassium (K), calcium (Ca), and magnesium (Mg). Internal metabolism was shown to influence the hydro-chemical status of peatland water, free of substantial human impact. The variables tested were within the range of the habitat preferences of the herb species and indicated that they have a wide ecological tolerance. However, their identical habitat preferences were not reflected in identical values for the physicochemical variables of the water essential for building populations of these species. The occurrence of these plant species was also shown to be determined by the hydro-chemical characteristics of the habitat, but the characteristics of their occurrence did not indicate the hydro-chemical aspect of the habitat.

The results of the field accounting of medicinal plants in the forest and forest-steppe zones of Central Russia

To assess the potential of the raw material base of medicinal plants and search for new plant sources of biologically active substances, it is necessary to monitor and record the biological diversity of plants, their number and abundance in natural plant communities. Plants are counted by the transect method. In the course of field research in 2022, the biodiversity, abundance, and abundance of medicinal plants in three regions of Central Russia were assessed. In the Lipetsk Region (the lower reaches of the Chichera River, the valley of the Sukhaya Lubna River), the most notable species of the Ranunculaceae families were identified: Adonis vernalis L. (spring adonis); Rosaceae: Agrimonia eupatoria L. (common agrimony); Filipendula vulgaris Moench (six-petalled meadowsweet). In the Samara region (floodplain of the Sok River) - Rosaceae: Sanguisorba officinalis L. (burnet officinalis); Filipendula vulgaris Moench; Ranunculaceae: Adonis vernalis L. In the Moscow, Ryazan region (Meshchera) species of families - Rosaceae: Potentilla argentea L. (silver cinquefoil) (Rosaceae), Fragaria vesca L. (wild strawberry), Potentilla erecta L. (upright cinquefoil), Comarum palustre L. (marsh cinquefoil); Onagraceae: Chamaenerion angustifolium (L.) Scop. (fireweed narrow-leaved); Asteraceae: Helichrysum arenarium (L.) Moench.; Caprifoliaceae: Valeriana officinalis L. (valerian officinalis); Hypericaceae: Hypericum maculatum L. (St. John’s wort); Equisetaceae: Equisetum arvense L. (horsetail); Lamiaceae: Stachys palustris L. (marsh weed); Primulaceae: Lysimachia vulgaris L. (common loosestrife). A total of 65 species of medicinal plants have been identified. Species with a noticeable abundance are characterized as a source of a certain group of chemical compounds. EFFICACY OF BOLUSES OF RHIZOMES WITH ROOTS OF MARSH CINQUEFOIL AGAINST STRONGYLATOSES OF GASTROINTESTINAL TRACT OF SHEEP

The paper presents data on the study of the effect of boluses of rhizomes with roots of marsh cinquefoil on the body of sheep invaded by Strongylata of the gastrointestinal tract. In accordance with the data obtained, it follows that the use of boluses of rhizomes with roots of marsh cinquefoil in therapeutic doses, as an anthelminthic agent, had no negative effect on the body of animals in experimental groups, and there was a decrease in the infestation rate in sheep with strongylatoses of the gastrointestinal tract.

Isolation of Individual Compounds from the Aerial Part of <i>Comarum palustre </i>L. and Their Structure Elucidation Using Spectroscopic Methods

Introduction. Medicinal plants are a rich, almost inexhaustible source of medicinal substances, and due to their large chemical diversity of metabolites available for isolation their research is always an important task. One of the promising medicinal plants for research is marsh cinquefoil (Comarum palustre L.), widely used in folk medicine for the treatment of diseases of the musculoskeletal system.Aim. Isolation of individual secondary metabolites from the aerial part of C. palustre and their subsequent structural elucidation by NMR experiments.Materials and methods. The aerial parts of the marsh cinquefoil, were collected next to the Saint Petersburg State Chemical-Pharmaceutical University Nursery Garden of Medicinal Plants (Leningrad region, Vsevolozhsky district, Priozerskoe highway, 38 km) in July 2021. Fraction analysis was performed through analytical high-performance liquid chromatography (HPLC) using a LC-20 Prominence (Shimadzu corp., Japan) equipped with a SPD-M20A diode-array detector. The isolation of compounds was carried out by open column chromatography using sorbents with different selectivity, as well as by preparative HPLC using a Smartline system (Knauer, Germany) equipped with a spectrophotometric detector. The structures of the isolated compounds were established by 1D NMR experiments (Bruker Avance III 400 MHz, Germany).Results and discussion. Seven individual compounds (1–7) were isolated and their structures elucidated. Two compounds (1 and 2) are derivatives of ellagic acid, namely 4-O-α-L-arabinofuranoside ellagic acid (1) and 4-O-β-D-glucopyranoside ellagic acid (2), while the other five compounds are derivatives of flavonoids: kaempferol-3-O-β-D-glucuronide (3), quercetin-3-O-β-D-glucuronide (4), quercetin-3-O-β-D-(6’’-β-D-glucopyranosyl)-glucopyranoside (5), quercetin-3-O-β-D-(2’’-galloyl)-glucopyranoside (6) and (+)-catechin (7).Conclusion. As a result of the current research, seven individual compounds were isolated from the aerial part of the marsh cinquefoil and their structure were elucidated. Compounds 1, 2, 4, 5 and 6 were found and isolated from the aerial part of C. palustre L. for the first time.

Barium, Lithium and Titanium Content in Herbs of Mid-Field Wet Depressions in East-Central Poland

This paper presents the results of research on the Ba, Li and Ti content in six species of herbs sampled from mid-field wet depressions and from the soil. These temporary flooded depressions were surrounded by arable crops, permanent grassland and shrubby vegetation. The research area was located in the eastern part of the Mazovian Voivodeship, east-central Poland. The following plants were used in the experiment: corn mint (Mentha arvensis L.), purple marshlocks (Comarum palustre L.), silverweed (Potentilla anserina L.), yarrow (Achillea millefolium L.), yellow loosestrife (Lysimachia vulgaris L.) and gypsy-wort (Lycopus europaeus L.). The Li, Ba and Ti content of plants, bottom sediment and soil was determined by the ICP-AES method after previous dry mineralization. Of the six herb species, Mentha arvensis L. was with the greatest accumulation potential of the chemical elements. However, no excessive Ba, Li and Ti content was found in herbs growing at different distances from arable fields, permanent grassland and shrubby vegetation. The highest Ba content was found in periodically flooded soil (zone II), while the highest amounts of Li and Ti were recorded in non-flooded soil (zone III).

Introduction. Understanding the mechanisms of accumulation of individual groups of biologically active substances in promising types of plant raw materials and the possibility of predicting them is important for solving fundamental and applied problems of pharmaceuticals. To date, differences have been revealed in the qualitative and quantitative composition of secondary metabolites in the aboveground and underground of Comarum palustre L., however, the issue remains unstudied.Aim. Comparative metabolomic study of the composition of the primary metabolites of the aboveground and underground parts of Comarum palustre L.Materials and methods. The object of the study was the aboveground and underground parts of Comarum palustre L., harvested in the vicinity of the nursery of medicinal plants of the St. Petersburg State Chemical and Pharmaceutical University (Leningrad Region, Vsevolozhsky District, Priozerskoe Highway, 38 km) in 2019 and dried. Metabolomic studies based on GC-MS method was perfomed. A statistical analysis based on the MetaboAnalyst 5.0 platform was used.Results and discussion. Analysis of the chromatograms obtained using the GC-MS method revealed the content of 933 primary metabolites in the aboveground and underground parts of Comarum palustre L., 120 of which were identified. Using a number of statistical methods, 10 metabolites from monosaccharides, acids and alcohols, making the greatest contribution to the manifestation of differences between the studied samples, were identified.Conclusion. The study revealed the relationship between the composition of primary and secondary metabolites in medicinal plant raw materials.
