Phenotypic Characterization of Citron Watermelon (Citrullus lanatus var. citroides) Genotypes for Bioenergy Production

Citron watermelon (Citrullus lanatus var. citroides) is an important climate smart crop, characterized by high oil content in the seeds and it is a popular fruit in southern Africa. To determine best performing genotypes eight phenotypic traits were assessed to identify cultivars with potential for biodiesel production. A field experiment was conducted at Sebele Agricultural Research Station, in Southern Botswana, during 2019 to 2020 cropping season. Four (4) melon genotypes were planted in a Randomized Complete Block Design replicated three times and eight (8) characters were assessed. Except seed oil content (26.6%) there was significant (P < 0.05) difference among all the traits, an indication of higher genetic diversity in the selected citron watermelon. SC1-Sesoswane, exhibited highest seed yield (1540 kg/ha) and oil yield (435.03 kg/ha) while MMB-280-Lerotse recorded lowest seed yield and oil yield of 548.1 kg/ha and 144 kg/ha respectively. There was a significant correlation of (0.9) between oil yield (kg/ha), number of fruits produced, and seed yield (kg/ha), an indication that increasing the seed yield will positively increase the amount of oil produced. This study revealed the potential bioenergy production of the four cultivars. However, SC1-Sesoswane appears a more promising alternative cultivar for feedstock for biodiesel production.

Screening of efficient microalgae strains isolated from Tunisian ecosystems: Assessment of algal growth rate and added-value bioproducts for biorefinery applications

Microalgae are highly diverse organisms with significant applications in the production of high-value molecules and biodiesel from microalgae rich in oils. In this study, our main objectives were to evaluate the potential applications of four isolated microalgae strains, namely Scenedesmus sp., Nannochloris sp., Oscillatoria sp., and Amphora sp., which were selected from the natural environment. These strains were batch cultured, and their growth was monitored. We determined the content of chlorophyll a, carotenoids, and lipids, and analyzed the fatty acid profile of each cultivated strain to assess their lipid potential for biodiesel production. Our findings revealed that these strains exhibited high biomass production, ranging from 3000 mg. L−1 for Amphora sp. to 3800 mg. L−1 for Oscillatoria sp. Notably, Oscillatoria sp. and Scenedesmus sp. were found to be the richest in saturated fatty acids, with 10.68% and 9.36% of dry cell weight, respectively, suggesting their potential use in the production of biodiesel from microalgal oils with possibility of blending with other oils feedstock to improve biodiesel final quality. Furthermore, our results showed that the Nannochloris sp. strain could be cultivated to extract pigments for various applications, as it exhibited high levels of chlorophyll a (38.43 mg. L−1) and carotenoids (4.153 μg. mL−1).

Synthesis of HNTs-Ca/Zn catalyst for biodiesel production from acidulated palm oil: Optimized by GA-BP

The development of heterogeneous base catalyst with acid resistance has become a major topic in the current research on the biodiesel production process. Halloysite nanotubes (HNTs) is supported with bimetallic of calcium and zinc for modification and labeled as HNTs-Ca/Zn to show both the acid and base characteristics. Further, this supported catalyst is used to produce biodiesel from the acidulated palm oil through catalyzing transesterification in one-step in this study. The HNTs-Ca/Zn catalyst, synthesized by the impregnation method, is characterized by TG-DTG, XRD, XPS, TEM, SEM-EDX, nitrogen adsorption-desorption, CO2/NH3-TPD, FTIR and Raman. The capability of the HNTs-Ca/Zn catalyst in catalyzing transesterification is investigated, where the maximum yield of 95.22% is achieved with the molar ratio of Zn to Ca of 1.5. The advantage of HNTs-Ca/Zn1.5 catalyst is prominent when the transesterification source oil is acidulated, where the biodiesel yield of 92.66% is obtained even at the acid value of 26.02 mg KOH g−1. Besides, reusability of the catalyst is acceptable, where the yield of 77.91% is maintained after the fourth reused cycle. Further, the Genetic Algorithm-Back Propagation (GA-BP) neural network is used to train and predict the transesterification parameters of the reaction temperature, catalyst amount and methanol to oil molar ratio. The prediction of the biodiesel yield is reasonable. At the predicted reaction parameters of 158 °C, catalyst amount of 7.1 wt.% and methanol to oil molar ratio of 15.5, the predicted biodiesel yield is 98.71%, where the experimental verification value is 98.60%. Therefore, the HNTs-Ca/Zn catalyst is considered to demonstrate exceptional catalytic performance and recyclability, offering the potential for biodiesel production.

Nutrient and salinity stress induced biodiesel production from a green alga, Monoraphidium neglectum

The potential of a green alga as a versatile and sustainable feedstock for biodiesel production was assessed. Molecular identification revealed the alga to be Monoraphidium neglectum. Under diverse growth conditions, including nutrient deficiency and salinity stress, the microalga showed robust adaptability. Under nitrate deficiency (N-, the lipid content peaked (30.67 %), with a notable increase in monounsaturated fatty acids (MUFA), reaching 50.72 % in the stressed sample. Additionally, the highest polyunsaturated fatty acid (PUFA) content (24.55 %) was observed at a salinity level of 0.12 M. At 0.01 M salinity, the highest total carbohydrate content was observed. A significant fatty acid methyl ester (FAME) yield of 85.08 % was obtained under nitrate deficiency (N-) emphasizing its biodiesel production potential. The as-obtained biodiesel exhibited low viscosity, superior low-temperature performance and enhanced oxidation stability rendering the alga as an ideal candidate for eco-friendly, sustainable biodiesel production.

Lipid Yields and Fatty Acid Composition if Indigenous Microalgae, Isolated from the Beira, and Diyawanna Urban Lakes, for Sustainable Biofuel Production in Sri Lanka


 Biofuels are carbon-neutral combustibles produced from renewable biomass. Third generation biofuels, derived from microalgal biomass, are distinctly superior to their preceding generations, due to higher productivity and resource use efficiency of microalgae compared to conventional energy crops. It is a viable source of sustainable energy for our country due to the availability of water, ambient temperatures and year-round sunlight needed for microalgae cultivation, and most importantly, the abundance of diverse microalgae species. However, these native microalgae are relatively unexplored for their productivities and thereby, greatly underutilized. This research was aimed to investigate the lipid yields and fatty acid compositions of local freshwater microalgae, and to identify their potential for biodiesel production. This was carried out by (1) microalgae isolation and morphological identification (2) microalgae cultivation (3) biomass harvesting, lipid extraction, gravimetric measurement of extracted lipids and (4) acid-catalysed transesterification, followed by GCMS analysis of the biodiesel product. Chlorella sp., Chlorococcum sp. and Chlamydomonas sp. were isolated from Beira/ Diyawanna Lakes using streak plate technique in solid standard BBM media. They were next cultivated under axenic conditions in liquid standard BBM media under 16: 8 (light: dark), at 25°C for 20 days in duplicates. The biomass was then harvested using alum, dried to reach a constant weight and the Bligh and Dyer lipid extraction was done in triplicate. The extracted lipids were trans-esterified with methanol, using sulfuric acid catalyst at 90° C for 60 minutes. The separated product was analysed by GCMS. Chlorella sp., Chlorococcum sp. and Chlamydomonas sp. cultivated under standard conditions gave average lipid yields of 11.34±1.01%, 15.00±1.40% and 2.10±0.50% respectively. Average lipid yield of Chlamydomonas sp. was significantly lower than the other two species at 95% confidence level (Two-sample T test on SPSS Version 21). GCMS analysis of the biodiesel product from Chlorella sp. indicated significant levels of saturated fatty acids including palmitic and stearic acids, monounsaturated fatty acids like cis-11- eicosenoic acid and polyunsaturated fatty acids including linoleic and linolenic acids. The fatty acid ratios were used to calculate the saturation value, cetane number, density, iodine value and pour point of the produced biodiesel. These quality parameters were in accordance with the ASTM D6751- 20a standards. Therefore, with a substantial yield and fatty acid composition of intracellular lipids, this local species of Chlorella can be a prospective candidate for sustainable bioenergy production in Sri Lanka.

Potential Application of Bacterial Lipase from Bacillus subtilis in the Production of Biodisel

This work aims to evaluate the potential of bacterial lipase of Bacillus subtilis in biodiesel production. Assays were performed to obtain, purify, and immobilize the enzyme and compare chemical and enzymatic transesterification tests. The Tryptic Soy Broth (TSB) culture medium without supplementation showed the highest enzymatic activity and purification yields when using 60% of (NH4)2SO4. Citral was the most efficient agent for lipase immobilization. Even though the free enzymes showed a higher activity rate, the immobilized enzymes are associated with a better-quality product. In conclusion, the lipase enzyme showed positive potential in biodiesel production.

Chemical Profiling and Industrial Viability of Neem Seed Oil: A Comprehensive Study for Sustainable Biodiesel Production

The study explores the chemical composition of neem seed oil, highlighting its potential for commercial biodiesel production and various industrial applications. The oil, with a liquid state, pale greenish-yellow color, and pleasant odor, yielded 32.1%, surpassing industrially extracted oils like soybean and cottonseed. Factors influencing its composition include seasonal variations, geographical origin, genetic disparities, growth stages, plant segments, and postharvest processing. Despite these, neem seeds meet the required oil content percentage for large-scale industrial biodiesel production. However, challenges like high free fatty acid content and elevated acid values necessitate acid esterification before alkaline transesterification, potentially increasing production costs. The study also highlights the importance of appropriate seed processing practices to optimize oil yield and quality.

Biodiesel production potential of Eichhornia crassipes (Mart.) Solms: comparison of collection sites and different alcohol transesterifications

Renewable resources have stood out as raw materials in producing biofuels. This study aimed to evaluate the parameters of alcohol transesterification (ethanol and methanol) and localization of collection of aquatic macrophyte Eichhornia crassipes (Mart.) Solms in the production of biodiesel by in situ transesterification. E. crassipes was collected in Dourados and Corumbá (Brazil) municipalities. The fatty acid ester composition of the biodiesel was characterized and quantified by gas chromatography. The biodiesel properties were estimated using the BiodieselAnalyzer© program prediction. The ethyl transesterification resulted in higher yields, but the localization of collection was the most relevant parameter in biodiesel production according to the Permutation Multivariate Analysis of Variance. The simulation and comparison of the physical–chemical properties of E. crassipes biodiesel and BD 100 (commercial biodiesel) were promising for commercial application.

Biodiesel production in India: Prospects, challenges, and sustainable directions.

Biodiesel has the potential to serve as a feasible substitute for fossil fuels in many sectors, including transportation and internal combustion engines, without requiring extensive modifications. UsinHemg alternative energy sources, including biodiesel, is necessary to effectively tackle the growing demand predominantly observed in the transportation sector. This review is aimed to examine the technological progress, potential benefits, obstacles, and prospects associated with using biodiesel in India. India exhibits a significant potential for biodiesel production due to the abundance of various biofuel crops within its territory. The information supplied includes recent information from official government reports and literature. Collaborative efforts among all stakeholders in the energy industry can achieve the realization of reducing imports of petroleum-based fuel. However, it is essential to consider several significant elements specific to the Indian context when considering the utilization of biodiesel. The reported findings in this research are expected to shed light on the current and prospects of biodiesel deployment in India.

Biodiesel Production by Biocatalysis using Lipids Extracted from Microalgae Oil of Chlorella vulgaris and Aurantiochytrium sp.

Microalgae are lipid-rich microscopic eukaryotic algae that can be used aiming for more sustainable biodiesel production by employing environmentally sound processes. The present work evaluates biodiesel production using a biocatalyst and two microalgae species as oil feedstock (Chlorella vulgaris and Aurantiochytrium sp.). Lipid extraction was performed using different techniques, namely, Soxhlet extractions (8 h — both species) with different solvents (hexane; hexane:ethanol (1:1 v/v); and chloroform) and room temperature hexane extraction (72 h — Aurantiochytrium sp.). Transesterification occurred for 24 h (150 rpm), using 30 % lipase loading. The results showed that high extraction temperatures cause microalgae oil degradation, focused on unsaturated fatty acids, leading to a lower biodiesel conversion yield. Using Aurantiochytrium sp. oil, it was possible to obtain around 55 %wt. of biodiesel conversion yield using oil extracted at room temperature (6:1 methanol:oil molar ratio), whereas for the oil extracted in the Soxhlet apparatus, the biodiesel conversion yield was around 30 %wt.. The low lipid content (1.0 %wt.) and biodiesel conversion yield (up to 25 %wt.) obtained using C. vulgaris show that the biomass used in the current study has low potential for biodiesel production. However, enzymatic biodiesel production from microalgae represents a promising avenue for sustainable energy generation, offering a renewable and environmentally responsible solution to the world’s energy needs. For that purpose, further studies, such as the optimisation of the extraction and transesterification of Aurantiochytrium sp. oil, should be carried out.

Indigenous microalgae strains characterization for a sustainable biodiesel production.

Microalgae have been widely recognized as a promising feedstock for sustainable biofuels production to tackle global warming and pollution issues related to fossil fuels uses. This study identified and analyzed indigenous microalgae strains for biodiesel production, specifically Chlorella vulgaris and Coelastrella thermophila var. globulina, from two distinct locations in Algeria. Molecular identification confirmed their identity, and the microalgae exhibited notable growth characteristics. Local Chlorella vulgaris and Coelastrella thermophila var. globulina showed good growth and high biomass yield, compared to Chlorella vulgaris CCAP211/11B reaching a weight of 1.48 gL-1 , 1.95 gL-1 , and 2.10 gL-1 , respectively. Lipids content of local Chlorella vulgaris, Coelastrella thermophila var. globulina, and Chlorella vulgaris CCAP211/11B, were found to be 31.39±3.3%, 17±2.26%, and 19±0.64%, respectively. Chlorella vulgaris stood out as a candidate for biodiesel production due to its equilibrium between SFA and PUFA (43.24% and 45.27%). FAs are predominated by SFA and MUFA for Coelastrella thermophila var. globulina with value of 81.49% (SFA+MUFA). Predicted biodiesel qualities comply with ASTM6751 and EN14214 standards. Studied microalgae have therefore a promising potential for biodiesel production. However, optimising cultivation conditions is necessary to enhance biomass and lipids yield at a large scale.

Cloning and characterization of an acidic lipase from a lipolytic bacterium in tempeh

BackgroundLipases have emerged as essential biocatalysts, having the ability to contribute to a wide range of industrial applications. Microbial lipases have garnered significant industrial attention due to their stability, selectivity, and broad substrate specificity. In the previous study, a unique lipolytic bacterium (Micrococcus luteus EMP48-D) was isolated from tempeh. It turns out the bacteria produce an acidic lipase, which is important in biodiesel production. Our main objectives were to clone the acidic lipase and investigate its potential in biodiesel production. ResultIn this study, the gene encoding a lipase from M. luteus EMP48-D was cloned and expressed heterologously in Escherichia coli. To our knowledge, this is the first attempt at the cloning and expression of the lipase gene from Micrococcus luteus. The amino acid sequence was deduced from the nucleotide sequence (1356 bp) corresponded to a protein of 451 amino acid residues with a molecular weight of about 40 kDa. The presence of a signal peptide suggested that the protein was extracellular. A sequence analysis revealed that the protein had a lipase-specific Gly-X-Ser-X-Gly motif. The enzyme was identified as an acidic lipase with a pH preference of 5.0. Fatty acid preferences for enzyme activities were C8 and C12 (p-nitrophenyl esters), with optimum temperatures at 30–40 °C and still remaining active at 80°C. The enzyme was also shown to convert up to 70% of the substrate into fatty acid methyl ester. ConclusionThe enzyme was a novel acidic lipase that demonstrated both hydrolytic and transesterification reactions. It appeared particularly promising for the synthesis of biodiesel as this enzyme’s catalytic reaction was optimum at low temperatures and was still active at high temperatures.

Exploring the biodiesel production potential of marine microalgae Nodosilinea nodulosa SNMVBTAR001 from Kerala coastal region, India

Microalgae-derived biodiesel has emerged as a promising solution to meet the increasing global demand for carbon-free and environmentally sustainable energy. This study explores the diversity of microalgae across seasons in Kerala, India, to assess their potential for biodiesel production. A total of Ninety marine microalgal isolates were analyzed, revealing Cyanophyceae sp. as the dominant group (67.64%), followed by Bacillariophyceae (22.54%), Chlorophyceae (5.88%), Xanthophyceae (1.96%), and Trebouxiophyceae (1.96%). Microalgae species distribution varied across regions, with Munnakal region exhibiting the highest diversity. Seasonal fluctuations played a significant role, with diversity peaking during pre-monsoon, followed by post-monsoon, summer, and winter. Five isolates with the highest growth (OD values) were identified, and SNMVBTAR007 (285 ± 1.5 mg/L) and SNMVBTAR001 (294 ± 2.0 mg/L) displayed the highest biomass concentration. Lipid analysis revealed SNMVBTAR001 (59%) and SNMVBTAR044 (55%) as having the highest lipid concentrations. Molecular characterization confirmed their identification as Nodosilinea nodulosa. Transesterification of extracted microalgal lipids yielded biodiesel meeting ASTM and EN standards. Fatty acid analysis indicated the presence of desirable fatty acids, particularly oleic acid. Qualitative assessments, including density, acid value, flashpoint, and soap content, aligned with biodiesel standards. Nodosilinea nodulosa SNMVBTAR001, a potential candidate for biodiesel production, with maximum efficiency of 23.10 km/L in an electrical generator. This study underscores the promise of Nodosilinea nodulosa microalgae for sustainable biodiesel production, offering eco-friendly energy solutions and reduced carbon emissions. Further research is needed to optimize cultivation and production techniques for large-scale biodiesel production, contributing to a greener and more sustainable energy future.

Biomimetic mineralization of lipase@MOF biocatalyst for ease of biodiesel synthesis: Structural insights into the catalytic behavior

A hybrid lipase@ZIF-8 biocatalyst was fabricated via a facile one-pot approach and coordination-driven self-assembly strategy. Entrapment immobilization of lipase in metal–organic frameworks (MOFs) was meticulously validated. The reduced Km value and apparent activation energy confirmed that the lipase@ZIF-8 registered a superior enzyme-substrate affinity than free ones. A concomitant conformational rearrangement occurred as lipase was encapsulated into the porous zeolite-like topologies, wherein lipase architectures presented more flexibility in character. An approximate 75% biodiesel yield was achieved when the biocomposites-catalyzed transesterification was conducted at given conditions. Notably, the lipase@ZIF-8 exhibited certain substrate-selectivity by deciphering the discrepancy in fatty acid profiles of the resulting biodiesel. Besides, the lipase@ZIF-8 biocomposites showed reasonable reusability and remained around 80% of initial activity after five cycles of transesterification. Consequently, the encapsulation of lipase into MOFs is a viable and sustainable avenue to address free lipase's intrinsic limitation, and underscores the immense potential in biodiesel production.

A betaine-modified whole-cell biocatalyst with surface display of Thermomyces lanuginosus lipase for biodiesel production from Jatropha curcas oil

Yeast surface displaying lipases are promising biocatalysts for biodiesel synthesis. Thermomyces lanuginosus lipase (TLL) was displayed on Pichia pastoris cell surface by screening proper anchor system, resulting in a whole-cell biocatalyst (termed D-TLL). The lipase activity of D-TLL is the highest activity to date for surface-displayed TLL, reaching 2274.7 U/g dry cell weight (DCW). The obtained biocatalyst was efficient in converting Jatropha curcas oil into biodiesel. The biodiesel yield peaked 92.6% after 12 h adopting only 3.3% (w/w) of D-TLL in the solvent-free system. As compared to free TLL, the whole-cell biocatalyst showed a dramatic reduction of 48 h in reaction time. The further treatment with betaine enabled enhanced thermostability, hydrophobicity and methanol tolerance of biocatalyst (named D-TLL-betaine), and thereby increasing the reusability of the whole-cell biocatalyst. These results exhibited that the new biocatalyst, D-TLL-betaine, has great potential for biodiesel production.

Exploring the Potential of Biomass Pyrolysis for Renewable and Sustainable Energy Production: A Comparative Study of Corn Cob, Vine Rod, and Sunflower

The paper investigates the potential of biomass pyrolysis as a sustainable and renewable energy solution. The study focuses on three biomass types: corn cob, vine rod, and sunflower, which are abundant agricultural residues with potential for biofuel production. The pyrolytic gas, oil, and char produced during pyrolysis at a heating rate of 10 °C/min were analyzed. At the pyrolysis temperature of 500 °C, the corn cob showed the smallest final residual mass of 24%, while the vine rod exhibited the largest mass loss of 40%. Gas analysis revealed the concentrations of CO2, CO, H2, and CH4 in the pyrolytic gas, indicating its energy potential. Sunflower presented the largest calorific value of the produced biogas, while corn cob was the lowest. The chemical composition of the bio-oils was determined, with aliphatic acids identified as the dominant compounds, suggesting their potential for biodiesel production. Fourier Transform–Infrared Spectroscopy (FT-IR) analysis of raw biomass and char products demonstrated varying extents of decomposition among the biomass samples. A multicriteria assessment approach was employed to evaluate the differences between the selected three biomass feedstock and determined that sunflower biomass ranked the highest among the three, although the overall difference was small, confirming the suitability of all three biomass samples for pyrolysis conversion to higher-value-added fuels.

Determination of Selected Engineering Properties of Jathropha Seed

Jatropha (Jatropha curcas L.) is a prominent renewable energy plant with great potential for biodiesel production from its seeds. This research project aimed to investigate the engineering properties of Jatropha seeds to evaluate their suitability for biodiesel production and various industrial applications. By reducing reliance on fossil diesel, Jatropha biodiesel offers the prospect of enhanced energy security and reduced environmental pollution resulting from fossil fuel combustion (Werby and Mousa, 2016). The study focused on understanding the physical, mechanical, and aerodynamic properties of Jatropha seeds. Knowledge of these properties is crucial for the design of efficient machines and equipment used in processing, handling, cleaning, transporting, and storage of agricultural products like Jatropha seeds. Specifically, the research investigated the physical characteristics of the seeds, including size, shape, color, and texture. Moisture content was analyzed using standard drying methods, while bulk density and true density were measured through displacement techniques. Results indicated that Jatropha seeds possess favorable physical properties for handling and processing. In conclusion, this research contributes valuable insights into the engineering properties of Jatropha seeds, making them a promising bioresource for biodiesel production and diverse industrial applications. Addressing the identified challenges can further enhance their potential utilization and contribute to sustainable energy practices.

Production, optimization, and physicochemical characterization of biodiesel from seed oil of indigenously grown Jatropha curcas

With the growing demand for vegetable oils, alternative non-edible feedstocks like Jatropha curcas seed oil have gained interest for biodiesel production. The study aimed to comprehensively evaluate the physicochemical properties and biodiesel production potential of locally produced J. curcas seeds in Pakistan. Two different approaches were applied: a chemical synthesis approach involving acidic pretreatment and alkaline transesterification, and a biosynthetic approach using a lipase-producing strain of the Bacillus subtilis Q5 strain. The microbial biosynthesized biodiesel was further optimized using the Plackett–Burman design. The physicochemical properties of the J. curcas methyl esters were analyzed to assess their suitability as biodiesel fuel. Initially, the raw oil had a high free fatty acid content of 13.11%, which was significantly reduced to 1.2% using sulfuric acid pretreatment, keeping the oil to methanol molar ratio to be 1:12. Afterward, alkaline transesterification of purified acid-pretreated seed oil resulted in 96% biodiesel yield at an oil to methanol molar ratio of 1:6, agitation of 600 revolutions per minute (RPM), temperature 60°C, and time 2 h. Moreover, alkaline transesterification yielded ∼98% biodiesel at the following optimized conditions: oil to methanol molar ratio 1:6, KOH 1%, time 90 min, and temperature 60°C. Similarly, the Bacillus subtilis Q5 strain yielded ∼98% biodiesel at the following optimized conditions: oil: methanol ratio of 1:9, agitation 150 RPM, inoculum size 10%, temperature 37°C, and n-hexane 10%. The fuel properties of J. curcas seed biodiesel are closely related to standard values specified by the American Society for Testing and Materials (ASTM D6751–20a), indicating its potential as a viable biodiesel fuel source.

Screening of wastewater Oedogonium oblangum algae for hyper-oil transformation into biodiesel by response surface methodology

The algae have gained more significance as a renewable third-generation feedstock for being easily available and a hyper-production source for lipids and biodiesel. Sustainable and cost-effective production of algal biodiesel requires screening and optimization of various culture conditions. Screening of algae on the basis of its lipid content is crucial for exploring its potential for production of biodiesel. In the current study, the screening of six indigenous algal strains was performed based on lipid contents, out of which Oedogonium oblangum (O. oblangum) exhibited hyper-lipid content (15.76%). Bold basal media and wastewater media were compared as culture media to evaluate the growth of O. oblangum; the respective strain showed better growth on wastewater media, therefore, this media was used for further experimentation. A laboratory-based integrated approach was used to enhance production of algal lipids by applying the effects of temperature, pH, and wastewater dilutions. The oil contents were interestingly improved under the optimal culture conditions of temperature (25°C), pH (6.5), wastewater dilution (45%) with oil contents of 18.35%, 17.15%, and 17.95%, respectively. The extracted oil was transformed into biodiesel by alkali-mediated transesterification reactions using four factors (Oil/methanol ratio, temperature, time, and catalyst) by response surface methodology (RSM). The RSM successfully optimized the transesterification reactions with 86.73% of biodiesel under optimal conditions of oil/alcohol ratio (1:7.6 v/v), temperature (60.5°C), time (59 ​min), and catalyst (0.53 ​wt. %). Physical properties of biodiesel (kinematic viscosity; 5.17; acid value; 0.63; flash point; 152; specific gravity; 0.74; calorific value; 42.11; and water contents; 0.03) comply with the ASTM standards and lead to enhance the quality of produced biodiesel. The optimized culture conditions, transesterification reactions, and wastewater medium can be employed for the economically production of biodiesel at commercial scale. Overall, the results showed that biodiesel derived from an indigenous novel algal strain, O. oblangum, has the potential to compete and replace commercially available petro-diesel using wastewater media. It might contribute to resolve the energy challenges being faced by Pakistan and the entire world.
