ChemInform Abstract: Synthesis of Some Benzimidazole Derivatives as Potential Anthelmintics.

AbstractThe title compounds (I) are prepared following common procedures.

Development and validation of an in vitro Trichostrongylus colubriformis motility assay

Development and validation of an in vitro Trichostrongylus colubriformis motility assay. International Journal for Parasitology 17: 1441–1444. An in vitro Trichostrongylus colubriformis motility assay involving the use of a micromotility meter has been developed and validated. Four commercially available ruminant anthelmintics (albendazole, ivermectin, levamisole hydrochloride, and coumaphos) and an investigational hydrazone compound ( p-toluoyl chloride phenylhydrazone) were evaluated in this assay at four concentrations each. At 100 μg ml -1, all five treatments significantly ( P ⩽ 0.05) reduced the motility of ensheathed L-3 T. colubriformis larvae, thereby indicating anthelmintic activity. At this concentration, coumaphos was significantly less active than any of the other four treatments. At 10 μg ml -1 albendazole, ivermectin, levamisole hydrochloride and the hydrazone compound were active, but coumaphos was not. At 1 μg ml -1 albendazole, ivermectin and levamisole hydrochloride remained significantly active, but neither coumaphos nor the hydrazone compound showed significant activity. At all three of the higher concentrations (1,10 and 100 μg ml -1), levamisole hydrochloride indicated greater activity than any of the other treatments. This difference was statistically significant at the 1 and 10 μg ml -1 concentrations. None of the five treatments showed significant activity at the lowest concentration (0.1 μg ml -1). The in vitro T. colubriformis motility assay proved to be sensitive, accurate, rapid, and repeatable. This assay system should be another valuable addition to the tests used to identify potential anthelmintics, monitor helminth resistance to drugs, and define the kinetics and mode of action of drugs.

Studies in potential filaricides. 18. Synthesis of 2,2'-disubstituted 5,5'-dibenzimidazolyl ketones and related compounds as potential anthelmintics.

A series of 2,2'-disubstituted 5,5'-dibenzimidazolyl ketones and related compounds have been synthesized of which 2,2'-bis(carbomethoxyamino)-5,5'-dibenzimidazolyl ketone exhibited a broad spectrum of anthelmintic activity in experimental animals. At doses of 10-50 mg/kg given intraperitoneally, 5 killed 100% of the adult worms of Litomosoides carinii, Dipetalonema viteae, and Brugia malayi. By the oral route the macrofilaricidal efficacy of 5 was 97-100% at 100-200 mg/kg X 5 days. The treated animals showed gradual disappearance of microfilariae and before autopsy they became amicrofilariaemic. Some of the compounds also showed 100% efficacy against the human hookworms and tapeworm, Ancylostoma ceylanicum in hamsters, and Hymenolepis nana in rats at a single oral dose of 50-250 mg/kg. Compound 5 was also effective against Syphacia obvelata in mice at a single oral dose of 100 mg/kg and was found to be well tolerated by mice up to an oral dose of 2500 mg/kg.

Synthesis of substituted benzimidazoles as potential anthelmintics.

AbstractThe substituted biphenyls 8–13, 5(6)‐phenyl‐2‐substituted benzimidazoles 7, 14–18, 9‐phenylbenzimidazo[1,2‐c]quinazolin‐6‐one and ‐thione 20 as well as the 1‐aroyl‐2‐mercapto‐5‐phenylbenzimidazoles 21, 24, 25 have been synthesized. Their anthelminthic activities are reported.

Synthesis of 6-N-Aryl and Heteroaryl Benzthiazoles as Potential Anthelmintics

Abstract A number of 6-substituted arylaminobenzthiazoles (5-8 and 16-18), 6-(substituted -1-benzimidazolyl)benzthiazoles (9-15) and 6-(3-quinazolyl)benzthiazoles (19) have been synthesized starting from corresponding 6-aminobenzthiazoles. None of the compounds showed significant anthelmintic activity.

The rabbit — Trichostrongylus axei: T. colubriformis system as a preliminary screen for testing potential anthelmintics

Rabbits infected with the ruminant parasites Trichostrongylus axei and T. colubriformis were administered eight drugs (phenothiazine, thiabendazole, haloxon, cambendazole, mebendazole, levamisole, fenbendazole, and oxibendazole) at dose rates known to be highly effective as anthelmintics in ruminants. All but phenothiazine and thiabendazole were highly active against adult T. colubriformis; mebendazole, levamisole, and fenbendazole were highly active against T. axei. When the dose rates of thiabendazole, haloxon, cambendazole, and oxibendazole were doubled, all but haloxon were highly active against T. axei adults. Efficacy against 3 and 10-day old worms was generally poor; oxibendazole showed best overall activity.

A technic for initial evaluation of potential anthelmintics

1. 1. An in vitro method utilizing Ascaridia galli for the preliminary screening of substances for anthelmintic activity is described. 2. 2. Nicotine causes a paralyzing effect with a contraction of the worm, which has been called the “nicotine effect”. Atropine also causes paralysis but it is accompanied by a relaxation of the worm. The latter has been called the “atropine effect”. 3. 3. There appears to be correlation between the “nicotine effect” and anthelmintic activity.
