Chromium and Palmitic Acid Supplementation Modulate Adipose Tissue Insulin Sensitivity in Postpartum Dairy Cows

Periparturient dairy cows exhibit intense lipolysis driven by reduced dry matter intake, enhanced energy needs, and the loss of adipose tissue (AT) insulin sensitivity. Extended periods of low insulin sensitivity and negative energy balance induce lipolysis dysregulation, leading to increased disease susceptibility and poor lactation performance. Chromium (Cr) supplementation improves systemic insulin sensitivity, while palmitic acid (PA) increases energy availability for milk production. However, the effect of supplementing Cr and PA alone or in combination on insulin sensitivity in AT is unknown. Thirty-two multiparous cows were used in a randomized complete block design experiment and randomly assigned to one of 4 diets fed from 1 to 24 DIM. Diets included: control, no supplementation (CON, n = 8); Cr (Cr-propionate at 0.45 ppm Cr/kg DM, n = 8); PA (1.5% DM, n = 8); or Cr+PA (n = 8). Plasma samples were collected at -13 ± 5.1 d prepartum (PreP), and 14.4 ± 1.9d (PP1) and 21 ± 1.9d (PP2) after calving for albumin, BHB, BUN, calcium, cholesterol, glucose, nonesterified fatty acids (NEFA), total protein, iron, transferrin, triglycerides, and oxylipids quantification. Subcutaneous AT (SCAT) explants were collected at PreP, PP1 and PP2 and incubated in the presence of the lipolytic agent isoproterenol (ISO = 1 µM, BASAL = 0 µM) for 3 h. The antilipolytic effect of insulin (1µL/L) on SCAT explants was evaluated during ISO stimulation (IN+ISO). Lipolysis was quantified by glycerol release in the media (nmol glycerol/mg AT). Macrophage infiltration and adipocyte size were measured using hematoxylin and eosin-stained AT sections and immunohistochemistry. Cr tended to reduce postpartum NEFA concentrations when compared with CON, PA, and Cr+PA. Likewise, Cr increased the percentage of large adipocytes (>9000 µm2) postpartum compared with other diets. In line with higher lipid content, Cr-fed cows had higher ex-vivo BASAL lipolysis at PP2 when compared with PA and Cr+PA. ISO induced higher lipolysis at PP1 and PP2 but it was not affected by Cr and PA. IN+ISO reduced lipolysis by 29.91 ± 11% in Cr compared with ISO. In contrast, IN+ISO did not affect ISO lipolysis in CON, PA, and Cr+PA. Plasma transferrin was reduced by Cr. At PP2, PA cows had 3.3-fold higher macrophage infiltration in SCAT when compared with CON and Cr. Plasma 9-HODE and 9-oxoODE were increased by Cr+PA. PA increased plasma 13-oxoODE and Cr increased the ratio of 13-HODE:13-oxoODE. PA increased 5-iso Prostaglandin F2α-VI. Our results demonstrate that supplementing Cr during the immediate postpartum enhances SCAT insulin sensitivity and lipid accumulation. Further studies should determine the effects and mechanisms of action of Cr and PA on AT lipogenesis, adipogenesis, and their impact on lactation performance.

Transition cow nutrition and management strategies of dairy herds in the northeastern United States: Part III—Associations of management and dietary factors with analytes, health, milk yield, and reproduction

The objective was to evaluate relationships between putative periparturient management and dietary factors at the pen and herd levels with metabolic- and inflammation-related analytes, health disorders, milk yield, and reproductive performance. Multiparous and primiparous cows from 72 farms in the northeastern United States were enrolled in a prospective cohort study. Farms were visited 3 times during the prepartum and postpartum periods: during the far-off dry, close-up dry, and fresh periods. Pen measurements were taken at each visit for the pens where cows sampled were housed, and particle size was determined for the total mixed ration for the pen. A survey was used to acquire data on herd-level management variables. Blood samples were collected from the same 11 to 24 cows per farm during the close-up and fresh period visits. Whole blood was analyzed for postpartum β-hydroxybutyrate (BHB) concentrations, and plasma was analyzed for prepartum and postpartum nonesterified fatty acids (NEFA) and postpartum haptoglobin (Hp) concentrations. Health event, milk yield, and reproductive records were acquired through the records management software program used on the farm. For the pen-level analysis, primiparous and multiparous cows were analyzed separately. For the pen- and herd-level analysis, a simple linear regression was conducted on all possible explanatory variables. Variables were included in the full multivariable general linear model if P < 0.20, and a manual backward stepwise elimination process ensued until all variables had P < 0.10. Our results indicate that pen- and herd-level management factors are associated with blood biomarkers, health, milk yield, and reproductive performance. For the prepartum period, our results support increasing the proportion of particles on the 19-mm sieve of the Penn State Particle Separator, optimizing bunk space, and not overfeeding metabolizable energy (ME), to decrease the prevalence of elevated postpartum NEFA, BHB, and Hp concentrations, decrease disorder incidence, maximize milk yield, and improve pregnancy risk to first service. For the fresh period, our results generally support optimizing bunk space, avoiding commingling, increasing feeding frequency, avoiding high physically effective undigested neutral detergent fiber (NDF) after 240 h of in vitro fermentation and high total fermentable carbohydrate diets while optimizing the inclusion of forage NDF, and ensuring adequate diet ME and metabolizable protein to reduce the prevalence of elevated postpartum NEFA, BHB, and Hp concentrations, minimize disorder incidence, maximize milk yield, and improve pregnancy risk to first service. At the herd level, our results generally support not vaccinating in the calving pen, minimizing the number of prepartum and postpartum pen moves, and avoiding long stays in the calving pen after parturition to reduce the prevalence of elevated biomarker concentrations, decrease disorder incidence, increase milk yield, and improve reproductive performance.

Transition cow nutrition and management strategies of dairy herds in the northeastern United States: Part II—Associations of metabolic- and inflammation-related analytes with health, milk yield, and reproduction

The objectives were as follows: (1) establish cow-level thresholds for prepartum nonesterified fatty acids (NEFA) and postpartum NEFA, β-hydroxybutyrate (BHB), and haptoglobin (Hp) concentrations associated with negative health events; (2) evaluate cow-level associations between biomarkers and 305-d mature equivalent milk at the fourth test day (ME305) and reproductive performance; and (3) identify herd-alarm levels (proportion of cows sampled above the critical threshold) for biomarkers that are associated with herd-level changes in disorder incidence (displaced abomasum and clinical ketosis), reproductive performance, and ME305. In a prospective cohort study, 1,473 cows from 72 farms were enrolled from the northeastern United States. Blood samples were collected from the same 11 to 24 cows per herd during the late-prepartum and early-postpartum periods. Whole blood was analyzed for postpartum BHB concentrations; plasma was analyzed for prepartum and postpartum NEFA and postpartum Hp concentrations. Critical thresholds for the biomarkers associated with health events for all cows were established using a receiver operating characteristic curve analysis. Poisson, linear mixed effects, and Cox proportional hazards models investigated the association of the biomarkers with health and performance. The prepartum NEFA and Hp threshold associated with culling was ≥0.17 mmol/L and 0.45 g/L, respectively. The postpartum NEFA and BHB thresholds associated with diagnosis of metritis, displaced abomasum, or clinical ketosis were ≥0.46 mmol/L and ≥0.9 mmol/L, respectively. Multiparous cows with prepartum NEFA concentration ≥0.17 mmol/L produced 479 kg less ME305. Multiparous and primiparous cows with postpartum NEFA concentration ≥0.46 mmol/L produced 280 kg less and 446 kg more ME305, respectively. Cows with BHB concentration ≥0.9 and ≥1.1 mmol/L produced 552 kg more ME305 and had a 20% decreased risk of pregnancy within 150 d in milk, respectively; however, multiparous cows with BHB concentration ≥1.5 mmol/L produced 376 kg less ME305. Cows with Hp concentration ≥0.45 g/L produced 492 kg less ME305 and had 28% decreased risk of pregnancy within 150 DIM. Cows with Hp concentration ≥0.45 g/L had 19% decreased pregnancy risk to first service (PRFS). Herds above the herd-alarm levels for prepartum NEFA had a 6.0-percentage unit increase in disorder incidence and a 6.0-percentage unit decrease in 21-d pregnancy rate (PR) for multiparous cows, a 3.9-percentage unit increase in PR and a 5.8-percentage unit increase in the probability of pregnancy for primiparous cows. Herds above the herd-alarm levels for postpartum NEFA had a 5.8- and 4.2-percentage unit increase in disorder incidence for multiparous and primiparous cows, respectively, a 789 kg decrease in ME305 for multiparous cows, and a 6.8- and 6.3-percentage unit decrease and increase in PR and PRFS for multiparous cows, respectively. Herds above the herd-alarm levels for BHB had an 8.5-percentage unit increase in disorder incidence, a 332 and 229 kg increase in ME305 for primiparous and multiparous cows, respectively, and a 3.2-, 5.2-, and 7.0-percentage unit decrease in PR, probability of pregnancy, and PRFS, respectively. Herds above the herd-alarm levels for postpartum Hp had a 5.3-percentage unit increase in disorder incidence. At the cow level and herd level, elevated biomarker concentrations were associated with an increased disorder risk and varied performance responses.

Phenotyping metabolic status of dairy cows using clustering of time profiles of energy balance peripartum

Due to a combination of a relatively low energy intake and a high demand of energy required for milk production, dairy cows experience a negative energy balance (EB) at the start of lactation. This energy deficit causes body weight reduction and an increased risk for metabolic diseases. Severity and length of negative EB can differ among cows. Peripartum time profiles of EB for dairy cows are not described yet in the literature. Creating EB-derived time profiles with corresponding metabolic status and disease treatments could improve understanding the relationship between EB and metabolic status, as well as enhance identification of cows at risk for compromised metabolic status. In this research we propose a novel method to cluster EB time series and examine associated metabolic status and disease treatments of dairy cows in the peripartum period. In this study, data of 3 earlier experiments were merged and examined. Four dairy cow clusters for time profiles of EB from wk -3 until +7 relative to calving were generated by the global alignment kernel algorithm. For each cluster, mean of body weight prepartum was distinguishable, indicating this might be a possible on-farm biomarker for the peripartum EB profile. Moreover, cows with severe EB drop postpartum were more treated for milk fever and had high plasma nonesterified fatty acids and β-hydroxybutyrate concentration, and low IGF-1, insulin, and glucose concentration in the first 7 wk of lactation. Overall, this study demonstrated that cows can be clustered based on EB time profiles and that characteristics such as prepartum body weight, and postpartum nonesterified fatty acids and glucose concentration are promising biomarkers to identify the time profile of EB and potentially the risk for metabolic diseases.

Effects of prepartum stocking density and a blind on physiological biomarkers, health, and hygiene of transition Holstein dairy cows

Many dairy cows succumb to disease after calving. Disease risk may be affected by the cows' social environment and ability to perform maternal behaviors. In nature, cattle isolate from others and find seclusion to give birth; these behaviors may be limited in indoor group pens and could potentially affect the cows' ability to cope. The aim was to determine whether stocking density and a physical blind in prepartum bedded-pack group pens affected physiological biomarkers, disease risk, and hygiene of peripartum dairy cows. A randomized complete block designed with a 2 × 2 factorial arrangement of treatments was used. Approximately 3 wk before calving, 374 cows (primiparous = 127; multiparous = 247) were assigned randomly to 1 of 4 treatment pens with the following 2 factors: (1) high versus low stocking density (7.7 to 12.9 m2 vs. 15.4 to 25.8 m2 lying space per cow), and (2) presence or absence of a blind. The blind was created using plastic road barriers and plywood, a steel gate, and shade cloth. After calving, cows were moved into a freestall pen and milked 3 times per day. Blood was sampled on the day of enrollment (baseline; -24 d) and approximately -14, -7, 3, 7, 10, and 14 d relative to calving, to measure inflammatory (haptoglobin) and metabolic (nonesterified fatty acids and β-hydroxybutyrate) biomarkers. β-Hydroxybutyrate (≥1.2 mmol/L) was used to diagnose subclinical ketosis. Vaginal discharge was scored 3, 7, 10, and 14 d after calving, to diagnose metritis (none, mild, or severe). Hygiene was scored on -24, -14, and -7 d before calving. Before calving, haptoglobin was lower in pens with a blind. After calving, cows in pens with low stocking density before calving tended to be at greater odds of being diagnosed with metritis. Cows were more likely to have poorer hygiene scores in high stocking density pens. No treatment effects were detected for pre- and postpartum nonesterified fatty acids, β-hydroxybutyrate, postpartum haptoglobin, or subclinical ketosis. Results suggest that the provision of a blind and lower stocking density may be beneficial for reducing inflammation before calving. However, low prepartum stocking density might increase the odds of metritis after calving. Although the reason for this paradox is unclear, the effects of prepartum stocking density may require further exploration.

Relationships between body condition, body condition loss, and serum metabolites during the transition period in primiparous and multiparous cows

In the transition period from late gestation to early lactation, dairy cows undergo tremendous metabolic changes. Insulin is a relevant antilipolytic factor. Decreasing serum concentrations of insulin and glucose, increasing serum concentrations of nonesterified fatty acids (NEFA) and β-hydroxybutyrate (BHB), and changes in body condition score (BCS) reflect the negative energy balance around calving. This study investigated peripartum metabolic adaptation in 359 primiparous and 235 multiparous German Holstein cows from a commercial dairy herd under field conditions. Body condition score was recorded and blood samples were taken 10 to 1 d prepartum, 2 to 4 d postpartum, and 12 to 20 d postpartum. Generalized mixed models and generalized estimation equations were applied to assess associations between prepartum BCS; BCS changes during the transition period; insulin, glucose, NEFA, and BHB serum concentrations; and milk yield, which was taken from an electronic milk meter from d 6 of lactation. Serum insulin concentrations of multiparous postpartum cows were lower compared with prepartum, and compared with primiparous cows. In general, primiparous cows had lower postpartum NEFA and BHB concentrations than multiparous cows. In primiparous cows, we identified a positive association between prepartum BCS and prepartum serum insulin concentration. Prepartum obese multiparous cows, but not primiparous cows, were characterized by higher postpartum serum NEFA and BHB concentrations and lower milk yield than other cows in the same parity class. Primiparous cows with a smaller degree of BCS loss during the transition period had higher postpartum insulin and lower NEFA concentrations and lower milk yield than other primiparous cows. In conclusion, primiparous cows had less lipolysis and lower milk yield than multiparous cows, associated with higher insulin concentrations. Avoiding high body condition loss during the transition period is a main factor in preventing peripartal metabolic imbalances of glucose and fat metabolism.

Effects of Boron Supplementation on Peripartum Dairy Cows’ Health

Although many different dietary studies on the prevention of negative energy balance related diseases are often encountered, this is the first study investigating the effects of boron supplementation on peripartum dairy cows' health in the light of an omics approach. Twenty-eight healthy cows (1 control and 3 experimental groups) were enrolled from 2months before predicted calving until 2months after calving. Experimental groups were assigned to receive boron at increasing doses as an oral bolus. Production parameters, biochemical profile, Nuclear Magnetic Resonance based metabolomics profile, and mRNA abundance of gluconeogenic enzymes and lipid oxidation genes were determined. Pivotal knowledge was obtained on boron distribution in the body. Production parameters and mRNA abundance of the genes were not affected by the treatments. Postpartum nonesterified fatty acids, β-hydroxybutyrate, and triglyceride concentrations were significantly decreased in experimentals. The primary differences among groups were in lipid-soluble metabolites. There were significant differences in metabolites including postpartum valine, β-hydroxybutyrate, polyunsaturated fatty acid and citrate, propionate, isobutyrate, choline metabolites (betaine, phosphatidylcholine, and sphingomyelin), and some types of fatty acids and cholesterol in experimentals. Boron appears to be effective in minimizing negative energy balance and improving health of postpartum dairy cows.

Short communication: Concentrations of nonesterified fatty acids and β-hydroxybutyrate in dairy cows are not well correlated during the transition period

The objective was to use longitudinal data of blood nonesterified fatty acids (NEFA) and β-hydroxybutyrate (BHBA) concentrations to describe the relationship between NEFA and BHBA in dairy cows during the periparturient period. Blood NEFA and BHBA concentration data collected from d 21 prepartum to 21 postpartum for 269 multiparous Holstein cows were selected from 4 different studies carried out within our research groups. Overall, NEFA concentrations were increased beginning near parturition with a relatively steady elevation of NEFA through d 9, after which concentrations gradually decreased. Prepartum BHBA concentrations began to increase beginning several days before parturition, continued to increase during the first week after parturition, and remained elevated through d 21 postpartum. Of the 269 cows included in the data set, 117 cows (43.5%) had at least one postpartum hyperketonemic event (BHBA ≥1.2mmol/L), and 202 cows (75.1%) had at least one event of elevated postpartum NEFA concentrations (≥0.70mmol/L) between 3 and 21 d in milk. Area under the curve (AUC) was used to investigate relationships between metabolites over time. Overall, the correlations between transition period NEFA and BHBA AUC were weak. We detected a negative correlation between prepartum BHBA AUC and postpartum NEFA AUC (r=−0.26). A positive correlation existed between postpartum NEFA AUC and postpartum BHBA AUC; however, the correlation coefficient was low (r=0.26). Large variation was found between the day of maximum NEFA concentration within the first 21 d in milk and day of maximum BHBA concentration for the same period. The mean and median times of maximum NEFA concentration were 6.8 and 6 d, respectively, whereas the mean and median times of maximum BHBA were 9.6 and 8 d, respectively; however, the range in days for both the mean and median day of maximum concentrations was very large. Overall, our data set indicates a weak relationship between blood concentrations of NEFA and BHBA during the periparturient period of dairy cows, suggesting that elevated concentrations of one should not be extrapolated to suggest elevated concentrations of the other metabolite.

Concentrations of nonesterified fatty acids and glucose in blood of periparturient dairy cows are indicative of pregnancy success at first insemination

Greater blood concentrations of nonesterified fatty acids (NEFA) and lesser blood concentrations of glucose are indicative of the normal process of nutrient partitioning that occurs in early postpartum dairy cows. The objective was to determine the relationship between blood NEFA and glucose concentrations and subsequent conception at first insemination in postpartum dairy cows. Holstein (n=148) and Guernsey (n=8) dairy cows were blood sampled at approximately d 10, 7, and 3 prepartum, on the day of calving and 3, 7, 14, and 21d postpartum for measurement of NEFA and glucose concentrations. Serum and plasma were harvested and used for measurement of NEFA and glucose concentrations, respectively. Cows were given a presynchronization treatment (2 injections of PGF2α 14d apart) with the second PGF2α injection occurring 14d before the initiation of the timed AI (TAI) protocol. Blood for determination of progesterone concentrations was collected at each presynchronization injection and at the initiation of the TAI protocol that was used for first insemination (74±7d postpartum). Cows were considered noncycling if serum progesterone concentrations at the 2 presynchronization PGF2α injections (d 37 and 51±7 postpartum) and at the initiation of the TAI protocol (d 65±7 postpartum) were ≤1ng/mL, and there was no indication of ovulation or presence of a corpus luteum by ultrasound examination at the initiation of the TAI protocol. Pregnancy was determined at 33d and again at 61d after first insemination by using ultrasound. Across all days, serum NEFA and plasma glucose concentrations were not different between cows that ovulated before the initiation of the TAI program (cycling) compared with those that did not ovulate (noncycling). Serum NEFA concentrations, however, were less and plasma glucose concentrations were greater during the early postpartum period for cows that subsequently became pregnant at first insemination compared with those that failed to become pregnant. Logistic regressions were used to predict the probability of pregnancy based on NEFA and glucose concentrations from individual days. The prediction with the greatest likelihood ratio was for d 3 postpartum NEFA and glucose concentrations. Nutritional status during the early postpartum period (within 1wk after calving), as indicated by blood NEFA and glucose concentrations, may affect subsequent fertility by a mechanism that is independent from interval to first ovulation.

High NEFA concentrations around parturition are associated with delayed ovulations in grazing dairy cows

The objectives of this study were to assess indicators of metabolic status of grazing dairy cows around parturition, and the relationship between these indicators with the resumption of ovulations postpartum (ROP). Holstein multiparous cows (N=20) grazing on improved pastures and supplemented with concentrates were body condition scored and tail bled weekly from wk −2 through +9 related to parturition. Plasma samples were analyzed for non-esterified fatty acids (NEFA), leptin, insulin-like growth factor-1 (IGF-1) and progesterone (P4). Data were analyzed with mixed models, logistic regression, with receiver operator characteristic (ROC), and Cox regression analysis. Cows having Delayed Ovulation ([DO], ROP on week ≥5) had lower BCS, and higher NEFAs than cows having a normal ROP around parturition (BCS: 2.73±0.08 vs. 2.94±0.05, P<0.05, and NEFA: 0.43±0.04 vs. 0.35±0.02mM, P<0.10, respectively). Also, DO cows had lower BCS than normal herdmates (2.59±0.10 vs. 2.99±0.06, P<0.01) around time of ROP, but they had similar NEFA, leptin and IGF-1. The risk for DO increased as NEFA increased (0.4% and 0.5% per every increasing mM of NEFA in prepartum and postpartum, respectively). The ROC curve showed that NEFA (prepartum and postpartum) had areas of 0.85 and 0.80, and cut-off values of 0.39 and 0.47mM. Finally, hazard for ROP increased as prepartum IGF-1 increased, and it decreased as postpartum NEFA increased. In conclusion, cows with lower BCS and higher prepartum and postpartum NEFA had higher odds for getting DO than herdmates with greater BCS and lower NEFA concentrations.

Effect of fat supplementation during transition period on plasma leptin and non-esterified fatty acid concentrations in Holstein cows

The objective of this experiment was to investigate the effect of fat supplementation during the transition period on pre and postpartum body weight (BW), body condition score (BCS), non-esterified fatty acids (NEFA), glucose and leptin concentrations in Holstein cows. Holstein cows (n = 15) received a low fat diet (LF; 1.61 Mcal net energy for lactation (NEL)/kg of dry matter [DM]), moderate fat diet (MF; 1.68 Mcal NEL/kg DM) or a high fat diet (HF; 1.74 Mcal NEL/kg DM) for 4 weeks prior to calving. All cows were fed similar lactation diets ad libitum (1.74 Mcal NEL/kg DM) for 30 days after calving. Increasing diet energy density during transition period had no effect on prepartum DMI, BCS, BW, glucose and NEFA concentrations (P > 0.05); but leptin concentrations and energy balance (EB) were affected by treatments (P < 0.05). Animals fed HF had less plasma leptin prepartum. After parturition, BW, milk production, milk fat, protein, urea nitrogen and plasma glucose concentrations were affected by prepartum diets (P < 0.05). Fat supplementation prepartum did not affect postpartum NEFA. In conclusion, prepartum fat supplementation decreased leptin concentration prepartum.

Associations of elevated nonesterified fatty acids and β-hydroxybutyrate concentrations with early lactation reproductive performance and milk production in transition dairy cattle in the northeastern United States

The objectives were to evaluate the effects of elevated pre- and postpartum nonesterified fatty acids (NEFA) and β-hydroxybutyrate (BHBA) concentrations during the transition period on reproductive performance and milk production in dairy cattle. In a prospective cohort study of 91 freestall, total mixed ration-fed herds in the northeastern United States, blood samples were collected from approximately 15 prepartum and 15 different postpartum transition animals in each herd. All samples were stratified based on pre- or postpartum status at the time of sample collection, and 2,259 and 2,290 animals were used to evaluate reproductive and milk production performance, respectively. Reproductive performance was assessed by time to conception within 70 d post-voluntary waiting period (VWP) and milk production was assessed using mature-equivalent 305-d (ME305) milk yield estimated at 120 d in milk. While controlling for body condition score (BCS), calving season, median ME305 milk production, and parity, NEFA and BHBA concentrations were evaluated with time to event analysis to investigate reproductive performance. These same predictor variables were used to determine the effects of elevated NEFA and BHBA concentrations on ME305 milk yield with herd as a random effect. Heifers and cows were grouped in the final analyses if the results between groups were similar. In all animals sampled prepartum, the risk of pregnancy within 70 d post-VWP was reduced by 19% when NEFA concentrations were ≥0.27mEq/L. In all animals sampled postpartum, those with NEFA concentrations ≥0.72mEq/L had a 16% decrease in risk of pregnancy and those with BHBA concentrations ≥10mg/dL had a 13% decrease in risk. In cows and heifers, ME305 milk yield was decreased by 683kg when prepartum NEFA concentrations were ≥0.33mEq/L. In heifers sampled postpartum, ME305 milk yield was increased by 488kg when NEFA concentrations were ≥0.57mEq/L and increased by 403kg when BHBA concentrations were ≥9mg/dL. In cows sampled postpartum, ME305 milk yield was decreased by 647kg when NEFA concentrations were ≥0.72mEq/L and decreased by 393kg when BHBA concentrations were ≥10mg/dL. With the exception of milk production in heifers, this study indicates that increased concentrations of serum NEFA and BHBA had a detrimental effect on reproductive performance and milk production.

Evaluation of Non-Esterified Fatty Acids (NEFA) through the Transition Period as a Predictor of Clinical Disease

During the transition period, most dairy cows will transiently visit a state of negative energy balance and hence have elevated non-esterified fatty acids (NEFA) levels; however, only a subset of cows and farms develop levels of clinical illness past acceptable limits. Previous studies have demonstrated an association between NEFA levels above a certain cut-point and displaced abomasums. The objective of this study was to evaluate pre- and post-partum NEFA levels and their association with clinical disease in northeast US dairy herds.

Metabolism of Dairy Cows as Affected by Prepartum Dietary Carbohydrate Source and Supplementation with Chromium Throughout the Periparturient Period

Holstein cows (n=72) entering second or later lactation were used to determine whether metabolic indices and hepatic capacities for oxidation and gluconeogenesis from propionate are affected by source of carbohydrate in the prepartum diet and chromium-l-methionine (Cr-Met) supplementation throughout the periparturient period. Cows were fed prepartum diets as total mixed rations with the concentrate portion based either on starch-based cereals [high nonfiber carbohydrate (NFC); 1.59 Mcal/kg of net energy for lactation (NEL), 14.4% crude protein (CP), 40.3% NFC] or nonforage fiber sources (low NFC; 1.54 Mcal/kg of NEL, 14.5% CP, 33.6% NFC) from 21 d before expected parturition until parturition. After parturition all cows were fed a common lactation total mixed ration (1.74 Mcal/kg of NEL, 16.5% CP, 40.0% NFC). The Cr-Met was supplemented once daily via gelatin capsule at dosages of 0, 0.03, or 0.06mg of Cr/kg of BW0.75. Thus, treatments were in a 2 (carbohydrate source)×3 (Cr-Met) factorial arrangement. There was no effect of prepartum carbohydrate source on pre- and postpartum plasma concentrations of glucose, nonesterified fatty acids (NEFA), β-hydroxybutyrate (BHBA), insulin, glucagon, or insulin to glucagon ratio. However, cows fed the low NFC diet during the prepartum period tended to have greater plasma NEFA and lower BHBA concentrations postpartum. Liver glycogen concentrations tended to be greater on d 1 postpartum for cows fed low NFC prepartum. Supplementing 0.03 mg/kg of BW0.75 of Cr as Cr-Met increased prepartum plasma glucose and glucagon concentrations and tended to decrease prepartum plasma NEFA concentrations compared with either 0 or 0.06mg of Cr/kg of BW0.75. Postpartum plasma glucose concentrations decreased linearly and glucagon concentrations were increased quadratically by administering increasing amounts of Cr-Met. Supplementing Cr-Met did not affect prepartum plasma concentrations of insulin or BHBA, postpartum NEFA or BHBA, or liver composition. There was an interaction of prepartum carbohydrate source and Cr-Met supplementation such that in vitro hepatic conversion of [1-14C]propionate to both CO2 and glucose was similar or increased when Cr-Met was supplemented to cows fed the low NFC diet but decreased when Cr-Met was supplemented to cows fed the high NFC diet. Insulin addition in vitro did not affect hepatic metabolism of propionate on d 1 postpartum. Overall, both the NFC content of the prepartum diet and Cr-Met had only modest effects on metabolic indices in this experiment.

Role of Peripartum Dietary Propylene Glycol or Protected Fats on Metabolism and Early Postpartum Ovarian Follicles

Eighty multiparous cows were used to test the effects of feeding a supplement containing 55% dry propylene glycol (PGLY), prilled fat (PrFA) containing a low proportion of unsaturated fatty acids (FA), or calcium soaps of long-chain FA (CaLFA) containing a high proportion of unsaturated FA on energy balance (EB), blood metabolites, and early postpartum (PP) ovarian follicles. Dry cows (256 d pregnant) were divided into 6 groups and began the following dietary treatments: 1) control group, fed a dry cow diet and fed a lactating cow diet PP; 2) PGLY group, diet supplemented with 500 g/d per cow of dry PGLY prepartum through 21 d in milk; 3) PrFA:control group, diet supplemented with 230 g/d per cow of PrFA prepartum and fed the control diet PP; 4) PrFA:PrFA group, diet supplemented with 230 g/d per cow of PrFA prepartum through 21 d in milk; 5) CaLFA:control, supplemented with 215 g/d per cow of CaLFA prepartum and fed the control diet PP; 6) CaLFA:CaLFA, supplemented with 215 g/d per cow of CaLFA prepartum through 21 d in milk. Follicular fluid was aspirated from follicles ≥6mm on d 12 PP. The daily average calculated EB during the first 21 d in milk was lower in the PrFA:PrFA (−4.16 Mcal/d) and CaLFA:CaLFA (−3.64 Mcal/d) groups than in the control (−1.71 Mcal/d) and PGLY (−2.19 Mcal/d) groups. Postpartum plasma β-hydroxybutyrate was higher, and insulin concentrations were lower in the PrFA:PrFA (6.2 mg/dL and 126.1 pg/mL, respectively) and CaLFA:CaLFA (7.0 mg/dL and 130.7 pg/mL) groups than in the control (4.5 mg/dL and 274.5 pg/mL) and PGLY (4.3 mg/dL and 272.6 pg/mL) groups, whereas nonesterified FA concentrations were higher only than the control group. Postpartum nonesterified FA were 21% lower and insulin plasma concentrations were 86% higher in the CaLFA:control group as compared with the PrFA:control group. The progesterone concentrations in the follicular fluid of estradiol-active follicles were higher in the CaLFA:CaLFA (200.7 ng/mL) group than in all other groups (57.3 to 92.4 ng/mL), and androstenedione and estradiol concentrations were higher (54.2 and 1,049.1 ng/mL, respectively) than in the PGLY (15.5 and 440.1 ng/mL), PrFA:control (22.6 and 314.1 ng/mL), and CaLFA:control (17.5 and 451.9 ng/mL) groups. In conclusion, supplementation of protected fat during the peripartum period negatively affected the EB status of the cows. Neither fat supplementation nor PGLY influenced the development of ovarian follicles during the early PP period, but feeding fat containing a high ratio of unsaturated FA (CaLFA) increased progesterone concentrations in estradiol-active follicles that were aspirated at 12 d in milk.

Effects of Sucrose Supplementation on Dry Matter Intake, Milk Yield, and Blood Metabolites of Periparturient Holstein Dairy Cows

Thirty-four multiparous Holstein dairy cows (780±17.2kg body weight; 3.4±0.08 body condition score) were used in a completely randomized design to evaluate the effects of fermentable carbohydrate source on dry matter (DM) intake, milk production, and blood metabolites of transition cows. Treatments were initiated 30 d before expected calving date. After calving, all cows went onto a similar lactating cow diet. Dry matter intake was measured daily, and milk production and composition were measured weekly for 56 d after calving The control diet consisted of 11.5% ground corn, whereas the treatment diet consisted of sucrose replacing 2.7% of ground corn on a DM basis. Prepartum plasma glucose concentrations were higher (66.3 vs. 69.3±1.13 mg/dl) for cows fed the diet containing sucrose. Glucose concentrations were not different postpartum. Prepartum and postpartum nonesterified fatty acids, blood urea N, and insulin did not differ between treatments. Substitution of sucrose as a readily available carbohydrate source for ground corn did not affect prepartum or postpartum DM intake. Milk yield, 3.5% fat-corrected milk yield, and milk components did not differ between treatments. Results from this research demonstrated that partially replacing ground corn with sucrose did not enhance prepartum or postpartum intake or performance.
