Effective gmp-compliant point of care manufacturing of anticd19 chimeric antigen receptor t cells for non hodgkin lymphoma patients using the clinimacs prodigy®

Background & Aim Chimeric Antigen Receptor T-Cells (CAR Ts) have shown promising results in clinical studies to treat lymphomas, however clinical scale GMP-compliant manufacturing of CAR T cells can be challenging and unreproducible. We successfully manufactured autologous lentiviral anti-CD19 Chimeric Antigen Receptor T Cells for 18 relapsed or refractory patients in an investigator initiated Phase I clinical trial using the CliniMACS Prodigy®. Methods, Results & Conclusion T-Cells were isolated, transduced (LV-anti-CD19 CAR, Lentigen Technology, Inc.), and expanded in culture for 8 or 12 days. All 18 cultures met cell dose and safety requirements. Fifteen cultures were expanded for 12 days, then the culture time was reduced to 8 days to facilitate treatment to patients in an expedited manner. The 12 day cultures had a mean transduction efficiency of 48± 0.1% (range 29-62), a mean of 42-fold expansion ± 12 (range 30-78), and the mean number of CAR T+ cells at the end of culture was 2 × 109± 0.8 × 109. The 8 day cultures had a mean transduction efficiency of 42%, a mean of 12 fold expansion, and the mean CAR T+ cells at the end of culture was .6 × 109 cells. There were no manufacturing failures. For all patients, a predominance of CD8 cells was observed in the leukapheresis and post enrichment product. However, the phenotype of the infused products had a 2 fold increase of CD4 CAR T+ cells: CD8 CAR T+ cells. CAR T products were infused fresh for 16 patients and 2 received cryopreserved products. While the frequency of CAR T+ cells was not affected by freeze thaw, a decrease in T-effector memory cells was observed after thaw. All cultures were transduced with an MOI of 20 and had a mean vector copy number of .9 copies/cell. Patients monitored post infusion for vector persistence through proviral qPCR and flow cytometry show peak expression of CAR T+ cells between 6 days and 21 days post infusion, with most patients peaking around 14 days post infusion. To date, detectable vector has persisted for up to 12 months. Lastly, our CAR T products were tested for potency and showed a dose-dependent cytotoxicity towards CD19+ cancer cell lines in vitro. In conclusion, GMP-compliant anti-CD19 Chimeric Antigen Receptor T Cells can efficiently be manufactured using the CliniMACS Prodigy in as few as 8 days, resulting in a potent cell product and an expedited treatment for patients.

Negative Outcomes Associated with Reoperative Sternotomy and LVAD Explantation at the Time of Cardiac Transplantation

The number of heart transplants that require a reoperative sternotomy has increased dramatically with the growing utilization of LVADs. A reoperative surgical field complicates the recipient cardiectomy. The objective of this study was to evaluate the effect of reoperative sternotomy and LVAD explantation on the duration of surgery, the physiologic impact to the patient, and transfusion requirements. This study is a single institution, retrospective review of all adult heart transplants between January 2013 and August 2018. Patients were excluded if they underwent a multi-organ transplant. Eligible patients were divided into three groups: non-reoperative, non-LVAD reoperative sternotomy, and LVAD explantation. The physiologic effect on the patient was estimated by the peak postoperative lactate level. During the study period, 174 adult heart transplants were performed. Twenty-two patients met exclusion criteria and the final cohort included 152 patients. The average age was 54 ± 12 years. The non-LVAD reoperative sternotomy group included 32 patients (21.0%) patients and 39 (25.7%) were in the LVAD explantation group. The results are summarized in Table 1. Patient outcomes were negatively affected by reoperative sternotomy in regards to duration of operative times, intra- and post operative blood product requirements, and physiologic stress as indicated by lactate levels. When compared to other patients undergoing a reopeartive sternotomy, those with an LVAD had longer operative times and larger intraoperative blood product requirements. This study illustrates a negative impact of reoperative sternotomy and LVAD explanation on the heart transplant procedure itself and the consequences for the patient. The growing use of LVADs as a bridge to transplant will continue to increase the number of patients undergoing reoperative sternotomies. This study supports the development of less invasive devices for use as a bridge to transplant.

Identification of Persistent Sulfidogenic Bacteria in Shale Gas Produced Waters.

Produced waters from hydraulically fractured shale formations give insight into the microbial ecology and biogeochemical conditions down-well. This study explores the potential for sulfide production by persistent microorganisms recovered from produced water samples collected from the Marcellus shale formation. Hydrogen sulfide is highly toxic and corrosive, and can lead to the formation of “sour gas” which is costly to refine. Furthermore, microbial colonization of hydraulically fractured shale could result in formation plugging and a reduction in well productivity. It is vital to assess the potential for sulfide production in persistent microbial taxa, especially when considering the trend of reusing produced waters as input fluids, potentially enriching for problematic microorganisms. Using most probable number (MPN) counts and 16S rRNA gene sequencing, multiple viable strains of bacteria were identified from stored produced waters, mostly belonging to the Genus Halanaerobium, that were capable of growth via fermentation, and produced sulfide when supplied with thiosulfate. No sulfate-reducing bacteria (SRB) were detected through culturing, despite the detection of relatively low numbers of sulfate-reducing lineages by high-throughput 16S rRNA gene sequencing. These results demonstrate that sulfidogenic produced water populations remain viable for years post production and, if left unchecked, have the potential to lead to natural gas souring during shale gas extraction.

Application of VR virtual reality in film and television post-production

In recent years, VR virtual reality has gradually become a hot topic in society. As a medium integrating computer technology, imaging technology and human-computer interaction technology, VR virtual reality brings both a new interactive narrative method in the evolution of media and New means of communication in the era of text change. The purpose of this article is to analyze the current situation of the application of VR virtual reality in film and television post-production, and analyze the predicament of VR virtual reality in film and television post-production in order to help film and television post-production. In this paper, through a questionnaire survey and random selection of 50 students for digital image synthesis majors who have participated in digital film and television special effects production in a university, the survey results are collated and analyzed. 94.69% of students believe that VR virtual reality is in film and television post-production. The application makes the work more colorful. The application of VR virtual reality technology in the late stage of film and television has promoted the development of diversified creation and the development of film and television post production. At the same time, the application of VR virtual reality technology has improved the efficiency of film and television production, reduced the cost of physical setting, and saved resources.

Dilution effect of different combustion residuals on laminar burning velocities and burned gas Markstein lengths of premixed methane/air mixtures at elevated temperature

Most of the experimental examinations on laminar flame characteristics of a diluted combustible mixture have simulated combustion residuals with one of the main exhaust gases (N2, H2O, and CO2) or a mixture of two. However, flue gases have quite different thermodynamic properties and chemical reactivities. Therefore, simulating the post combustion products used for dilution with only one or two of the main exhaust gases may yield erroneous laminar flame characteristic data. In the present study, the laminar burning velocities and burned gas Markstein lengths of diluted methane/air mixtures at 1 bar and 473 K were measured with spherically expanding flames under constant pressure in an optically accessible constant volume combustion chamber. The mixtures were diluted with N2, H2O, and CO2 individually as well as with a mixture of 71.49% N2 + 19.01% H2O + 9.50% CO2 by volume, which represents the main product concentrations from stoichiometric methane/air combustion. Experimental results show that the laminar flame speed values of methane/air mixtures diluted with actual combustion residuals are between those of N2 and H2O dilution, whereas the laminar burning velocities of methane flames diluted with CO2 are considerably slower. The effects of different combustion residuals on the burned gas Markstein length were not found to be significant. At the experimentally-investigated initial conditions, CHEMKIN analyses were performed with the GRI-Mech 3.0, San Diego, and USC Mech II mechanisms to inquire about the performances of the kinetic schemes. The GRI-Mech 3.0 results were the closest to the experimental data. This mechanism was also utilized to numerically quantify the dilution, thermal diffusion, and chemical effects of combustion residuals. The dilution effect was the leading effect in decreasing the laminar burning velocity, while the thermal diffusion effect had the smallest contribution. Nevertheless, the thermal diffusion effect changes the thermal and mass diffusivities, and, as a result, the Lewis number, making it a vital parameter for flame stability and stretch. As the thermodynamic properties and chemical reactivities of the combustion residuals vary with temperature, pressure, equivalence and dilution ratios, real combustion residuals cannot be accurately represented with only one or two of the main exhaust gases, as shown in the current study.

Thermochemical, kinetic and ash characteristics behaviour of Thar Lignite, agricultural residues and synthetic polymer waste (EVA)

The study aimed to exploit the energy potential of naturally occurring fuels like Thar Lignite (TL), Sugarcane Bagasse (SB), Cotton Stalks (CS) and synthetic copolymer shoe manufacturing waste Ethylene Vinyl Acetate (EVA). Proximate, ultimate, chemical analysis along with Fourier transform infrared (FTIR) were carried out to analyze the compositional and structural differences between the studied fuel samples. Thermochemical decomposition of fuel samples was also evaluated using thermogravimetric analysis (TGA) under dry air conditions. Derivative thermogravimetry (DTG) obtained from TG curves were used to determine the ignition, peak, burnout temperatures along with combustion performance indices. The order of both average and peak rate of conversion (% s−1) was found to be; EVA > SB > CS > TL. Kinetic analysis of the samples was performed using Coats Redfern model. The apparent activation energy ‘Ea’ (kJ mole−1) was in the order of SB = 45.7 > EVA = 36.2 > CS = 36.1 > TL = 23.1. X-ray fluorescence (XRF) and ash fusion temperature (AFT) profile analysis of the ash samples was also performed to investigate the possible deposit formation propensity of post combustion residue products, alkali and slagging indices. SB showed the highest propensity towards slagging deposit formations with the order SB > CS > TL > EVA.

Mixture Design Study of Fiber‐Reinforced Self‐Compacting Concrete for Prefabricated Street Light Post Structures

In recent years, there has been an increasing demand to produce strong precast street light posts that are aesthetically pleasing. This study presents experimental results of a considerable number of mixture designs for fabricating precast street light posts where fiber‐reinforced self‐compacting concrete (FRSCC) was employed. The performance of many FRSCC mixtures was evaluated in terms of their structural properties and aesthetic characteristics. A trial‐and‐error procedure was performed for a series of FRSCC mixtures where silica fume, fly ash, and fibers were used. Slump flow and air content tests were conducted to determine the fresh FRSCC properties, and specimens were cast to evaluate their aesthetic. Three‐day and seven‐day compression tests were performed to examine the FRSCC hardened properties. The amount of cement in all batches was kept constant, whereas the distributions of fine and coarse aggregates, water, and other admixtures were adjusted. The largest slump flow of 73.7 cm (29 in) was recorded, and the maximum three‐day compressive strength was 43 MPa (6209 psi). Further refinement of the mixtures, which displayed the best strength and aesthetic attributes, was performed. Test results of the selected FRSCC mixtures indicated an excellent slump flow, air content, and compression values while achieving advantageous aesthetic qualities. Seven‐day compressive strength of 39 MPa (5686 psi) with the air content of 4.8 percent and the slump flow of 66 cm (26 in) was recorded. The study results and the developed FRSCC mixes can be used for mass production of precast concrete street light posts in precast plants.

This paper investigates the impulses, motives and conditions that are positively associated and problems, hurdles and concerns that are the obstacles in the resurgence of 21st century Pakistani film industry. It also gauges the priorities and concerns of young moviegoers and compares with the priorities and concerns of our young generation of filmmakers. To achieve this end we employed both qualitative and quantitative research methodologies and came up with following results that Pakistan’s screen to film production ratio is one of the best in the world. It is vehemently in search of its distinctive identity and plays, sometimes immaturely, with different genres are evidence to it. We see a visible drift from conventional Indo Pak Masala genre to social drama, physical reality, war movies and Wuxia. Production facilities of Pakistani film industry have been significantly improved and in terms of equipment Pakistani film industry is no more far behind Bollywood and Hollywood as it used to be even in the times of Khuda Kay Leay (KKL). Different production processes are gradually becoming worthier; cinematography, sound design and post production effects have been admired whereas viewers are critical about the standard of music, choreography, production budget, code switching, acting, script writing and screen playwright. Researchers found that most of the filmmakers complain about dearth and substandard script and screen play. Moreover, film viewers also complain about lack of originality in the script. Young filmmakers also complain about the dearth of trained human resources, who may use the modern equipment to its optimum level of output. Young viewers more or less face a paradox, they claim that they aspire to see their local culture and local social issues in Pakistani movies, they yearn to see local culture in Pakistani movies but when they are exposed to the choice to select either Pakistani film or Indian, they choose Indian movie hoping to get better entertainment value in terms of visual pleasure; a fact that requires separate in depth research about the psychology of young Pakistani film spectatorship.

Physical and thermal properties of various ajwain (Trachyspermum ammi L.) seed varieties as a function of moisture content

AbstractAjwain seeds are used as spice and have high medicinal values. The ajwain seed varieties selected were set at 8, 12, 16, and 20% moisture content on dry basis (db) for this study. The geometrical dimensions showed linear increment with the increase in moisture content of ajwain seeds. The true density and porosity increased from 1,069 to 1,697 kg m−3 and 15.84–42.88%, respectively, across the three varieties of ajwain seeds with the increasing moisture content. The bulk density showed a decreasing trend of 1049.81–857.14 kg m−3 with the increasing moisture content. The coefficient of friction was also found to increase for all the varieties, but the maximum friction was found for the cardboard surface and the minimum friction was observed in case of aluminium sheet. The thermal diffusivity of seeds decreased from 4.09 to 2.46 m2 s−1 with the increasing moisture content. Thermal conductivity and specific heat showed a marked increase with increasing moisture content. Varietal differences among the ajwain seeds of variety AA‐1, AA‐2, and AA‐93 were also studied at 5% significance level. Significant differences were observed between the three ajwain varieties, namely AA‐1, AA‐2, and AA‐93 in context to their physical and thermal properties.Practical applicationsThe physical and thermal properties of ajwain seeds will be useful in designing of processes and equipment required for its processing. These properties give detailed information about the quality of the seeds, its acceptability to consumers and the behavior of the ajwain post production, during storage and consumption and post consumption. The knowledge on physical properties such as seed size, shape, volume, surface area, mass, density, porosity helps in designing the separating, storing, handling, and drying systems. The grading and sizing machineries will be based on the size and shape of the ajwian seeds. The density and porosity of the ajwain seeds will help in sizing the hoppers and storage containers. The density of seeds will also play role in separation based on density or specific gravities. The frictional properties like coefficient of friction and angle of repose will help in designing the storage bins, chutes, hoppers, screw conveyers, threshers, and forage harvesters. Thermal properties of ajwain seeds will be useful in analyzing and designing of equipment involving heat transfer, such as in case of cooking, sterilizing and drying.

Doubling farmers' income : Scenario, challenges, opportunities and the way forward

The country's farm sector, the lifeline of economy, has been of late in the throes of a very paradoxical situation namely its shrinking share in country's GDP amidst dependence of close to 60% of country's population on it fortheir livelihood worsening the agrarian distress and the associated high farmer suicides every year in the face of spectacular performance on the production and productivity front in the form of 'green', 'blue' 'yellow' 'white' revolutions. The average income ofmore than 60% offarm households in the country with less than one hectare land is hardly a fourth of the current national average per capita income. While no doubt, the farm sector today offers a plethora of avenues for stepping up aggregate production as well as productivity of agri-horti, animal, fish, bird per unit area, input, time and effort, banishing current agrarian distress coupled with doubling farmers' incomes may however, continue to be a vision in spite of all such isolated and fragmented efforts. The paper therefore, calls for an integrated multi-pronged, multi-dimensional initiativesinvolving production as well as post production chain for translating not only PM's vision into reality at ground level but liberate the much distressed sections of the farm sector, i.e. the millions ofsmall and marginal farmers of the society fromtheir chronic vulnerabilities to weather and market risks, poverty, indebtedness, structural inefficiencies, adverse terms of trade, anti-farmer farm policies, rampant supplies of spurious and substandard inputs and thru it transform their lives and economies.

Biogas generation from insects breeding post production wastes

Abstract Insect breeding generates waste: insect excrements, often mixed with the remains of the feed. Insect waste is usually sold as a plant fertilizer, however, there is one more method of its use – methane production via the anaerobic digestion. To the best of the authors’ knowledge, this topic is very poorly studied. The aim of this work was the evaluation of the suitability of the waste derived from insects breeding (Hermetia illucens, Tenebrio molitor and Gryllus spp.) for methane production. The mesophilic anaerobic digestion process was performed in 500 ml bioreactors. The temperature of the process was 37 °C ± 1 °C and initial pH was 7.0 ± 0.2. The substrate loading comprised 3.5 g of total solids and the inoculum-to-substrate ratio was 2:1. The biomethane potential of investigated wastes was ∼177 ml g−1 TS for H. illucens, ∼212 ml g−1 TS for Tenebrio molitor to ∼225 ml g−1 TS for Gryllus spp.. The obtained biomethane potentials are similar to more commonly used substrates for anaerobic digestion like: cattle manure, mink manure, poultry manure, fruit and vegetables waste, ryegrass, switchgrass, wheat, and sewage sludge, which points to the reasonability of their use. Anaerobic digestion can be a new method for valorization of insect post-production wastes.

Promoting Students’ Motivation in Learning English Vocabulary through a Collaborative Video Project

The main objective of this study was to motivate EFL (English Foreign Language) students in learning English vocabulary by using collaborative video Project. This study followed a case study methodology to describe how video project experience can engage students to learn English and provide them an opportunity to participate in tasks as well as enrich their vocabulary. Twenty-five EFL students in the Vocabulary Building Course (VBC) participated in this study. This study used three phases for evaluations: the pre-production phase, production phase and post production phase. Data were collected from classroom observations, the video Project process and document analyses. A summary of the findings related to the video theme and narrative analysis of students’ videos are presented in this paper. Data analysis showed that students responded differently to their video project assignments and produced different types of collaborative videos with the help of a camcorder and computer application. Then, a survey was conducted to collect feedback from participants to learn their opinions and attitudes regarding the use of collaborative video project, students’ learning and motivation. Participants in this study expressed positive attitudes and opinions toward their video-project experiences. This study demonstrates that video Project can be a great tool for promoting students’ motivation and participation in learning English, enriching their vocabulary and can be an effective and powerful tool to create fun, interactive, and collaborative learning environments.

Violence Rating Prediction from Movie Scripts

Violent content in movies can influence viewers’ perception of the society. For example, frequent depictions of certain demographics as perpetrators or victims of abuse can shape stereotyped attitudes. In this work, we propose to characterize aspects of violent content in movies solely from the language used in the scripts. This makes our method applicable to a movie in the earlier stages of content creation even before it is produced. This is complementary to previous works which rely on audio or video post production. Our approach is based on a broad range of features designed to capture lexical, semantic, sentiment and abusive language characteristics. We use these features to learn a vector representation for (1) complete movie, and (2) for an act in the movie. The former representation is used to train a movie-level classification model, and the latter, to train deep-learning sequence classifiers that make use of context. We tested our models on a dataset of 732 Hollywood scripts annotated by experts for violent content. Our performance evaluation suggests that linguistic features are a good indicator for violent content. Furthermore, our ablation studies show that semantic and sentiment features are the most important predictors of violence in this data. To date, we are the first to show the language used in movie scripts is a strong indicator of violent content. This offers novel computational tools to assist in creating awareness of storytelling.

Deep Learning Application to Ensemble Learning—The Simple, but Effective, Approach to Sentiment Classifying

Sentiment analysis is an active research area in natural language processing. The task aims at identifying, extracting, and classifying sentiments from user texts in post blogs, product reviews, or social networks. In this paper, the ensemble learning model of sentiment classification is presented, also known as CEM (classifier ensemble model). The model contains various data feature types, including language features, sentiment shifting, and statistical techniques. A deep learning model is adopted with word embedding representation to address explicit, implicit, and abstract sentiment factors in textual data. The experiments conducted based on different real datasets found that our sentiment classification system is better than traditional machine learning techniques, such as Support Vector Machines and other ensemble learning systems, as well as the deep learning model, Long Short-Term Memory network, which has shown state-of-the-art results for sentiment analysis in almost corpuses. Our model’s distinguishing point consists in its effective application to different languages and different domains.

Mastodon Content Warnings: Inappropriate Contents in a Microblogging Platform

Our social communications and the expression of our beliefs and thoughts are becoming increasingly mediated and diffused by online social media. Beyond countless other advantages, this democratization and freedom of expression is also entailing the transfer of unpleasant offline behaviors to the online life, such as cyberbullying, sexting, hate speech and, in general, any behavior not suitable for the online community people belong to. To mitigate or even remove these threats from their platforms, most of the social media providers are implementing solutions for the automatic detection and filtering of such inappropriate contents. However, the data they use to train their tools are not publicly available.In this context, we release a dataset gathered from Mastodon, a distribute online social network which is formed by communities that impose the rules of publication, and which allows its users to mark their posts inappropriate if they perceived them not suitable for the community they belong to. The dataset consists of all the posts with public visibility published by users hosted on servers which support the English language. These data have been collected by implementing an ad-hoc tool for downloading the public timelines of the servers, namely instances, that form the Mastodon platform, along with the meta-data associated to them. The overall corpus contains over 5 million posts, spanning the entire life of Mastodon. We associate to each post a label indicating whether or not its content is inappropriate, as perceived by the user who wrote it. Moreover, we also provide the full description of each instance. Finally, we present some basic statistics about the production of inappropriate posts and the characteristics of their associated textual content.

Mechanical Assessment of Fuel Line Hoses under Variable Pressure

The automotive industry is one of the largest consumer of flexible hoses for fluids such as fuel or cooling. In order to limit the post production costs, every aspect of a vehicle must be tested in advance, including the lifespan, durability or failure of hoses. Throughout this paper summary results are presented for 4 types of fuel line hoses tested under controlled displacement loads at various pressures in order to validate the Finite Element Method simulations.
