The ventrolateral hypothalamic area and the parvafox nucleus: Role in the expression of (positive) emotions?

The lateral hypothalamus has been long suspected of triggering the expression of positive emotions, because stimulations of its tuberal portion provoke bursts of laughter. Electrophysiological studies in various species have indeed confirmed that the lateral hypothalamus contributes to reward mechanisms. However, only the rudiments of the neural circuit underlying the expression of positive emotions are known. The prefrontal cortex, the lateral hypothalamus, and the periaqueductal gray matter (PAG) are involved in these circuits; so, too, are the brainstem nuclei that control the laryngeal muscles and subserve mimicry, as well as the cardiovascular and respiratory systems. The implicated populations of hypothalamic neurons have not been defined either anatomically or molecularly. One promising candidate is the novel parvafox nucleus, which we recently described, in the murine medial forebrain bundle (mfb), which specifically expresses parvalbumin and Foxb1. With the molecularly defined parvafox nucleus as a centerpiece, the inputs from the prefrontal cortex and the projections to the PAG and brainstem can be studied with precision. By drawing on genetic approaches, it will be possible to manipulate the circuitry selectively with spatial and temporal exactitude and to evaluate the concomitant autonomic changes. These data will serve as a basis for imaging studies in humans using various paradigms to provoke the expression of positive emotions. In conclusion, studies of the hypothalamic parvafox nucleus will reveal whether this entity represents the fulcrum for positive emotions, as is the amygdala for fear and the insula for disgust.

Understanding how discrete populations of hypothalamic neurons orchestrate complicated behavioral states.

A major question in systems neuroscience is how a single population of neurons can interact with the rest of the brain to orchestrate complex behavioral states. The hypothalamus contains many such discrete neuronal populations that individually regulate arousal, feeding, and drinking. For example, hypothalamic neurons that express hypocretin (Hcrt) neuropeptides can sense homeostatic and metabolic factors affecting wakefulness and orchestrate organismal arousal. Neurons that express agouti-related protein (AgRP) can sense the metabolic needs of the body and orchestrate a state of hunger. The organum vasculosum of the lamina terminalis (OVLT) can detect the hypertonicity of blood and orchestrate a state of thirst. Each hypothalamic population is sufficient to generate complicated behavioral states through the combined efforts of distinct efferent projections. The principal challenge to understanding these brain systems is therefore to determine the individual roles of each downstream projection for each behavioral state. In recent years, the development and application of temporally precise, genetically encoded tools has greatly improved our understanding of the structure and function of these neural systems. This review will survey recent advances in our understanding of how these individual hypothalamic populations can orchestrate complicated behavioral states due to the combined efforts of individual downstream projections.

Fertility-regulating Kiss1 neurons arise from hypothalamic POMC-expressing progenitors.

Hypothalamic neuronal populations are central regulators of energy homeostasis and reproductive function. However, the ontogeny of these critical hypothalamic neuronal populations is largely unknown. We developed a novel approach to examine the developmental pathways that link specific subtypes of neurons by combining embryonic and adult ribosome-tagging strategies in mice. This new method shows that Pomc-expressing precursors not only differentiate into discrete neuronal populations that mediate energy balance (POMC and AgRP neurons), but also into neurons critical for puberty onset and the regulation of reproductive function (Kiss1 neurons). These results demonstrate a developmental link between nutrient-sensing and reproductive neuropeptide synthesizing neuronal populations and suggest a potential pathway that could link maternal nutrition to reproductive development in the offspring.

TRAP-seq defines markers for novel populations of hypothalamic and brainstem LepRb neurons.

ObjectiveLeptin acts via its receptor (LepRb) on multiple subpopulations of LepRb neurons in the brain, each of which controls specific aspects of energy balance. Despite the importance of LepRb-containing neurons, the transcriptome and molecular identity of many LepRb subpopulations remain undefined due to the difficulty of studying the small fraction of total cells represented by LepRb neurons in heterogeneous brain regions. Here we sought to examine the transcriptome of LepRb neurons directly and identify markers for functionally relevant LepRb subsets.MethodsWe isolated mRNA from mouse hypothalamic and brainstem LepRb cells by Translating Ribosome Affinity Purification (TRAP) and analyzed it by RNA-seq (TRAP-seq).ResultsTRAP mRNA from LepRb cells was enriched for markers of peptidergic neurons, while TRAP-depleted mRNA from non-LepRb cells was enriched for markers of glial and immune cells. Genes encoding secreted proteins that were enriched in hypothalamic and brainstem TRAP mRNA revealed subpopulations of LepRb neurons that contained neuropeptide-encoding genes (including prodynorphin, Pdyn) not previously used as functional markers for LepRb neurons. Furthermore, Pdyncre-mediated ablation of Leprflox in Pdyn-expressing neurons (LepRbPdynKO mice) blunted energy expenditure to promote obesity during high-fat feeding.ConclusionsTRAP-seq of CNS LepRb neurons defines the LepRb neuron transcriptome and reveals novel markers for previously unrecognized subpopulations of LepRb neurons.

Brain insulin receptors link stress and metabolism.

In this issue of Molecular Metabolism, Chong et al. [1] observed altered functioning of the hypothalamic–pituitary–adrenal (HPA) axis in mice lacking insulin receptors (IR) in selected regions of the brain. The rationale was to push beyond the original report of altered HPA activity in mice lacking IR in all brain neurons (NIRKO mice) [2] by assessing animals lacking IR in more targeted neuronal types. Specifically, male mice lacking IR broadly in forebrain areas, including most of the hypothalamus (IRNkx2.1 KO mice), had enhanced anxiety-like behavior on several tasks as well as elevated basal plasma arginine vasopressin (AVP) and increased secretion of corticosterone (CORT) in response to physical restraint. To determine the impact of IR knockout in specific populations of hypothalamic neurons, HPA axis function was also assessed in IRSim1 KO and IRAgrp KO male mice. In IRSim1 KO, the IR should be absent in the paraventricular and supraoptic nuclei (as well as a few extrahypothalamic areas) [3]. These mice manifested no changes of HPA activity or related behavior, implying that neurons located within the PVN (and/or SON) that express IR are unlikely to be the source of the stress phenotypes observed in IRNkx2.1 KO mice. There was a phenotype involving the HPA axis in male IRAgrp KO mice that appeared superficially to be somewhat opposite to that observed in IRNkx2.1 KO mice, including reduced basal AVP and reduced CORT in response to restraint. From these data, Chong et al. concluded that signaling through IR on AgRP-expressing neurons increases responsivity of the HPA and AVP systems.

Preserved Energy Balance in Mice Lacking FoxO1 in Neurons of Nkx2.1 Lineage Reveals Functional Heterogeneity of FoxO1 Signaling Within the Hypothalamus

Transcription factor forkhead box O1 (FoxO1) regulates energy expenditure (EE), food intake, and hepatic glucose production. These activities have been mapped to specific hypothalamic neuronal populations using cell type–specific knockout experiments in mice. To parse out the integrated output of FoxO1-dependent transcription from different neuronal populations and multiple hypothalamic regions, we used transgenic mice expressing Cre recombinase from the Nkx2.1 promoter to ablate loxP-flanked Foxo1 alleles from a majority of hypothalamic neurons (Foxo1KONkx2.1 mice). This strategy resulted in the expected inhibition of FoxO1 expression, but only produced a transient reduction of body weight as well as a decreased body length. The transient decrease of body weight in male mice was accompanied by decreased fat mass. Male Foxo1KONkx2.1 mice show food intake similar to that in wild-type controls, and, although female knockout mice eat less, they do so in proportion to a reduced body size. EE is unaffected in Foxo1KONkx2.1 mice, although small increases in body temperature are present. Unlike other neuron-specific Foxo1 knockout mice, Foxo1KONkx2.1 mice are not protected from diet-induced obesity. These studies indicate that, unlike the metabolic effects of highly restricted neuronal subsets (proopiomelanocortin, neuropeptide Y/agouti-related peptide, and steroidogenic factor 1), those of neurons derived from the Nkx2.1 lineage either occur in a FoxO1-independent fashion or are compensated for through developmental plasticity.

Modeling Interindividual Differences in Spontaneous Internal Desynchrony Patterns

A physiologically based mathematical model of a putative sleep-wake regulatory network is used to investigate the transition from typical human sleep patterns to spontaneous internal desynchrony behavior observed under temporal isolation conditions. The model sleep-wake regulatory network describes the neurotransmitter-mediated interactions among brainstem and hypothalamic neuronal populations that participate in the transitions between wake, rapid eye movement (REM) sleep, and non-REM (NREM) sleep. Physiologically based interactions among these sleep-wake centers and the suprachiasmatic nucleus (SCN), whose activity is driven by an established circadian oscillator model, mediate circadian modulation of sleep-wake behavior. When the sleep-wake and circadian rhythms are synchronized, the model simulates stereotypically normal human sleep-wake behavior within the limits of individual variation, including typical NREM-REM cycling across the night. When effects of temporal isolation are simulated by increasing the period of the sleep-wake cycle, the model replicates spontaneous internal desynchrony with the appropriate dependence of multiple features of REM sleep on circadian phase. In temporal isolation experiments, subjects have exhibited different desynchronized sleep-wake behaviors. Our model can generate similar ranges of desynchronized behaviors by variations in the period of the sleep-wake cycle and the strength of interactions between the SCN and the sleep-wake centers. Analysis of the model suggests that similar mechanisms underlie several different desynchronized behaviors and that the phenomenon of phase trapping may be dependent on SCN modulation of REM sleep-promoting centers. These results provide predictions for physiologically plausible mechanisms underlying interindividual variations in sleep-wake behavior observed during temporal isolation experiments.

Relationships between dietary macronutrients and adult neurogenesis in the regulation of energy metabolism – CORRIGENDUM

Of the environmental factors which have an impact on body weight, nutrients are most influential. Within normal limits, hypothalamic and related neuronal populations correct perturbations in energy metabolism, to return the body to its nutritional set-point, either through direct response to nutrients or indirectly via peripheral appetite signals. Excessive intake of certain macronutrients, such as simple carbohydrates and SFA, can lead to obesity and attendant metabolic dysfunction, also reflected in alterations in structural plasticity, and, intriguingly, neurogenesis, in some of these brain regions. Neurogenesis, previously thought to occur only in the embryo, is now known to take place in the adult brain, dependent on numerous stimulating and inhibiting factors, including dietary components. Because of classic associations between neurogenesis and the hippocampus, in learning and cognition, this brain region has also been the focus of attention in the study of links between diet and neurogenesis. Recently, however, a more complete picture of this relationship has been building: not only has the hypothalamus been shown to satisfy the criteria for a neurogenic niche, but appetite-related mediators, including circulating hormones, such as leptin and ghrelin, pro-inflammatory cytokines and the endocannabinoid intracellular messengers, are also being examined for their potential role in mediating neurogenic responses to macronutrients. The present review draws together these observations and investigates whether n-3 PUFA may exert their attenuating effects on body weight through the stimulation of adult neurogenesis. Exploration of the effects of nutraceuticals on neurogenic brain regions may encourage the development of new rational therapies in the fight against obesity.

Synchronous activation of gonadotropin-releasing hormone gene transcription and secretion by pulsatile kisspeptin stimulation

Pulsatile release of hypothalamic gonadotropin-releasing hormone (GnRH) is essential for pituitary gonadotrope function. Although the importance of pulsatile GnRH secretion has been recognized for several decades, the mechanisms underlying GnRH pulse generation in hypothalamic neural networks remain elusive. Here, we demonstrate the ultradian rhythm of GnRH gene transcription in single GnRH neurons using cultured hypothalamic slices prepared from transgenic mice expressing a GnRH promoter-driven destabilized luciferase reporter. Although GnRH promoter activity in each GnRH neuron exhibited an ultradian pattern of oscillations with a period of ∼10 h, GnRH neuronal cultures exhibited partially synchronized bursts of GnRH transcriptional activity at ∼2-h intervals. Surprisingly, pulsatile administration of kisspeptin, a potent GnRH secretagogue, evoked dramatic synchronous activation of GnRH gene transcription with robust stimulation of pulsatile GnRH secretion. We also addressed the issue of hierarchical interaction between the circadian and ultradian rhythms by using Bmal1-deficient mice with defective circadian clocks. The circadian molecular oscillator barely affected basal ultradian oscillation of GnRH transcription but was heavily involved in kisspeptin-evoked responses of GnRH neurons. In conclusion, we have clearly shown synchronous bursts of GnRH gene transcription in the hypothalamic GnRH neuronal population in association with episodic neurohormone secretion, thereby providing insight into GnRH pulse generation.

Contrasting Existence and Robustness of REM/Non-REM Cycling in Physiologically Based Models of REM Sleep Regulatory Networks

Typical human sleep throughout the night consists of alternating periods of rapid eye movement (REM) sleep and non-REM (NREM) sleep. This ultradian rhythm of NREM/REM cycling is thought to be produced by the state-dependent activity of “REM-on” and “REM-off” brainstem and hypothalamic neuronal populations that, respectively, promote or suppress REM sleep. Synaptic interactions among these populations define REM sleep regulatory networks; however, the identity of the key neuronal populations in these networks and the dynamics of interactions among them are disputed and cannot be addressed comprehensively with current experimental techniques. The purpose of this study is to use physiologically based mathematical models to explore the dynamic implications associated with competing hypotheses for network-based REM sleep regulation. Generally, putative REM sleep regulatory networks fall into two categories: a reciprocal interaction network consisting of an excitatory REM-on population interacting with an inhibitory REM-off population, and a mutual inhibition network where both REM-on and REM-off populations are inhibitory. We focus on the generation of regular periodic cycling solutions, which would generate the NREM/REM ultradian rhythm, in these networks. By applying our understanding of network mechanisms, we develop efficient numerical criteria for the existence of stable cycling solutions in each network structure. To investigate the robustness of cycling, we systematically analyze the response of model dynamics to manipulation of the components governing network interactions. By establishing the implications of network structure for the mechanisms and dynamics of NREM/REM state transitions, this comparative analysis identifies key targets for future experimental work to distinguish the structure of the proposed physiological REM sleep regulatory network.

A population of hypothalamic thermosensitive neurons regulating cardiovascular functions

Thyroid hormone is known for its profound direct effects on cardiovascular function and metabolism; however, recent studies demonstrated that the hormone also regulates these systems indirectly through the autonomic nervous system. While some of the mechanisms underlying the hormone's central effects on metabolism have been identified, its actions in the central cardiovascular control have remained enigmatic. Here we describe a novel population of neurons in the hypothalamus that require thyroid hormone receptor signaling for their development. Stereotaxic ablation of these cells in the mouse using a conditionally neurotoxic virus results in hypertension and temperature dependent tachycardia, indicating a role in the central autonomic control of blood pressure and heart rate. Moreover, the neurons exhibit intrinsic temperature sensitivity in patch clamping experiments, thus providing a new connection between cardiovascular function and core temperature. Taken together, we have discovered a previously uncharacterized hypothalamic cell population potentially important for understanding hypertension, and identified developmental hypothyroidism as a novel epigenetic risk factor for cardiovascular disorders.

Developmental changes in embryonic hypothalamic neurons during prenatal fat exposure

Maternal consumption of a fat-rich diet during pregnancy, which causes later overeating and weight gain in offspring, has been shown to stimulate neurogenesis and increase hypothalamic expression of orexigenic neuropeptides in these postnatal offspring. The studies here, using an in vitro model that mimics in vivo characteristics after prenatal high-fat diet (HFD) exposure, investigate whether these same peptide changes occur in embryos and if they are specific to neurons. Isolated hypothalamic neurons were compared with whole hypothalamus from embryonic day 19 (E19) embryos that were prenatally exposed to HFD and were both found to show similar increases in mRNA expression of enkephalin (ENK) and neuropeptide Y (NPY) compared with that of chow-exposed embryos, with no change in melanin-concentrating hormone, orexin, or galanin. Further examination using immunofluorescence cytochemistry revealed an increase in the number of cells expressing ENK and NPY. By plotting the fluorescence intensity of each cell as a probability density function, three different populations of neurons with low, medium, or high levels of ENK or NPY were found in both HFD and chow groups. The prenatal HFD shifted the density of neurons from the population containing low peptide levels to the population containing high peptide levels. This study indicates that neuronal culture is a useful in vitro system for studying diet effects on neuronal development and shows that prenatal HFD exposure alters the population of hypothalamic neurons containing ENK and NPY in the embryo. These changes may contribute to the increase in HFD intake and body weight observed in offspring.

Differential sensitivity of specific neuronal populations of the rat hypothalamus to prolactin action

Prolactin stimulates dopamine release from neuroendocrine dopaminergic (NEDA) neurons in the hypothalamic arcuate nucleus (ARC) to maintain low levels of serum prolactin. Elevated prolactin levels during pregnancy and lactation may mediate actions in other hypothalamic regions such as the paraventricular nucleus (PVN) and rostral preoptic area (rPOA). We predicted that NEDA neurons would be more sensitive prolactin targets than neurons in other regions because they are required to regulate basal prolactin secretion. Moreover, differences in the accessibility of the ARC to prolactin in blood may influence the responsiveness of this population. Therefore, we compared prolactin-induced signaling in different hypothalamic neuronal populations following either systemic or intracerebroventricular (icv) prolactin administration. Phosphorylation of the signal transduction factor, STAT5 (pSTAT5), was used to identify prolactin-responsive neurons. In response to systemic prolactin, pSTAT5-labeled cells were widely observed in the ARC but absent from the rPOA and PVN. Many of these responsive cells in the ARC were identified as NEDA neurons. The lowest icv prolactin dose (10 ng) induced pSTAT5 in the ARC, but with higher doses (>500 ng) pSTAT5 was detected in numerous regions, including the rPOA and PVN. NEDA neurons were maximally labeled with nuclear pSTAT5 in response to 500 ng prolactin and appeared to be more sensitive than dopaminergic neurons in the rPOA. Subpopulations of oxytocin neurons in the hypothalamus were also found to be differentially sensitive to prolactin. These data suggest that differences in the accessibility of the arcuate nucleus to prolactin, together with intrinsic differences in the NEDA neurons, may facilitate homeostatic feedback regulation of prolactin release.

Introduction to special topic—estrogenic control of hypothalamic GnRH neurons

In female mammals, sexual maturation is initiated by a change in the release pattern of pituitary gonadotropins, luteinizing hormone (LH) and follicle-stimulating hormone (FSH), and a consequential increase in the secretion of ovarian sex-steroids, estradiol and progesterone. Orchestrating these endocrine changes is a diffuse population of hypothalamic neurons that produce the neuropeptide, gonadotropin-releasing hormone (GnRH). Although the development and functional integration of GnRH neurons with the rest of the central nervous system is still poorly understood, recent progress has been made on several fronts. For example, there is now evidence that some mammalian species express more than one molecular form of GnRH (GnRH-I and GnRH-II), and that the two corresponding GnRH neuronal sub-populations may play different roles in the regulation of reproductive function and behavior. Moreover, through the use of transgenic animal models, neuronal fiber tracing, gene expression profiling, and electrophysiologcal recordings, new insights have been gained into the mechanisms that regulate GnRH release. This volume brings together 10 articles that reflect current thinking about the role of GnRH in mediating estrogenic feedback to the neuroendocrine reproductive axis. The main focus is on the negative and positive actions that estrogens exert within the hypothalamus of mammals, especially around the time of the preovulatory LH surge. However, the articles also present findings from other vertebrate classes, and so provide an intersting comparative perspective. Taken together, the knowledge presented in this volume represents a foundation upon which to develop more effective therapies for pubertal disorders, infertility, and menopause. The first article (Sower and Baron, 2011) demonstrates the expression of estrogen receptors within the lamprey hypothalamus, suggesting potential feedback interactions between esatradiol and GnRH neurons. Phylogenetically, lampreys are positioned as basal vertebrates, and like many other non-mammalian vertebrates they express multiple forms of GnRH. Consequently, studies in lampreys provide us with important insights into the molecular evolution of estrogenic-GnRH interactions. A cluster of three articles then focus on hypothalamic neuropeptides that exert a major influences on the secretion of GnRH, and which are thought to mediate the feedback effects of sex-steroids on the reproductive axis. For example, studies in rodents have clearly shown the importance of kisspeptin to both pulsatile and surge modes of GnRH release (Navarro, 2012), while human studies have observed sexual dimorphism in kisspeptin neurons during aging (Hrabovszky et al., 2011). Again, an interesting evolutionary perspective is provided by comparing the steroid sensitivity of kisspeptin neurons in various non-mammalian vertebrates (Kanda and Oka, 2012). Although the exact mechanism by which estrogens interact with the reproductive neuroendocrine axis is unclear, our general understanding of this issue is neatly summarized by a series of three reviews (Micevych and Sinchak, 2011; Kenealy and Terasawa, 2012; Radovick et al., 2012). Of particular interest is the finding that sex-steroids exert their feedback not only through classic estrogen receptors located in the nucleus, but also more rapidly through receptors located in the cell membrane. Various physiological functions show adaptations for life in an environment that undergoes marked changes across the 24-h day. For example, in many mammals the timing of the preovulatory LH surge occurs at a very specific time of day. Two articles discuss the interactions between the circadian timing system and estrogen-sensitive neural circuits that regulate GnRH secretion and the preovulatory surge (Sun et al., 2012; Williams and Kriegsfeld, 2012). Finally, a novel hypothesis is proposed to explain the conundrum of how estrogens appear to exert both negative and positive feedback actions on the reproductive axis (Urbanski, 2012). Data is presented from non-human primate studies to suggest that negative and positive estrogen feedback are two completely separate mechanisms, which are mediated, respectively, by two distinct GnRH neuronal populations. This hypothesis challenges the common assumption that has dominated the field for the past four decades, namely that individual GnRH neurons are relatively homogenous and can mediate both negative and positive estrogen feedback.

Estrogens and Obesity: Is It All in Our Heads?

Estrogens have preventative effects on weight gain and associated comorbidities, but the tissue-specific targets remain unknown. Here, Xu et al. (2011) demonstrate that ablation of estrogen signaling in two populations of hypothalamic neurons leads to weight gain and subsequent metabolic dysregulation and could be important target sites of estrogen actions.

Paradoxical or REM sleep, characterized by cortical activation combined with muscle atonia and rapid eye movements, was discovered at the end of the 1950s by Michel Jouvet and William C. Dement. Studies over the next twenty years suggested that the onset and maintenance of paradoxical sleep was due to a reciprocal inhibitory interaction between monoaminergic neurons inhibiting PS and cholinergic neurons generating PS located in a small part of the pontine reticular formation called the sublaterodorsal tegmental nucleus. Our recent studies rather indicate that these neurons are respectively GABAergic and glutamatergic. Further, they suggest that three populations of GABAergic neurons and population of hypothalamic neurons expressing melanin concentrating hormone, a peptide, play a important role in PS control.

Context fear learning specifically activates distinct populations of neurons in amygdala and hypothalamus

The identity and distribution of neurons that are involved in any learning or memory event is not known. In previous studies, we identified a discrete population of neurons in the lateral amygdala that show learning-specific activation of a c-fos-regulated transgene following context fear conditioning. Here, we have extended these studies to look throughout the amygdala for learning-specific activation. We identified two further neuronal populations, in the amygdalo-striatal transition area and medial amygdala, that show learning-specific activation. We also identified a population of hypothalamic neurons that show strong learning-specific activation. In addition, we asked whether these neurons are activated following recall of fear-conditioning memory. None of the populations of neurons we identified showed significant memory-recall-related activation. These findings suggest that a series of discrete populations of neurons are involved in fear learning in amygdala and hypothalamus. The lack of reactivation during memory recall suggests that these neurons either do not undergo substantial change following recall, or that c-fos is not involved in any such activation and change.
