Expression of biologically active middle T antigen of polyoma virus from recombinant baculoviruses.

Two different recombinant baculoviruses have been generated for expressing the middle T antigen (MT) of polyoma virus in insect (Sf9) cells. One (pAcI-PyMT) produces moderate levels of MT and the other (pVL-PyMT) high levels. Indirect immunofluorescence and cellular fractionation studies with pAcI-PyMT infected Sf9 cells give results similar to those observed with wild type polyoma virus infected mouse cells, and show MT to be mainly associated with cytoplasmic membranes in the insect cell. In the latter, a sub-population of MT is phosphorylated in in vitro protein kinase assays. The yields of MT from pVL-PyMT infected cells are high enough to suggest that this protein can now be produced by this method in sufficient amounts for definitive biochemical and crystallographic analyses.

Characterization of middle T antigen expressed by using an adenovirus expression system

The adenovirus Ad5(pymT) has been used to express middle T antigen at very high levels in 293 cells. The middle T antigen produced was localized to membranes and was modified in the same way as that expressed in polyoma virus-infected mouse cells. It was phosphorylated in vivo on serine residues and in vitro on tyrosine residues. The in vivo phosphorylations occurred between residues 223 and 275. The middle T antigen encoded by A d5(pymT) was phosphorylated in vitro in a complex with human pp60c-src. Interestingly, the extreme overexpression of middle T antigen did not cause a parallel increase in the amount of complex; most of the pp60c-src remained unassociated. Immunoaffinity purification resulted in approximately 100 micrograms of middle T antigen from a 100-mm tissue culture dish. Several cell proteins copurified with the Ad5(pymT)-derived middle T antigen. Two of these, the 74- and 63-kilodalton species, are of particular interest because they were also purified from mouse tumors expressing middle T antigen.

Duplication of functional polyadenylation signals in polyomavirus DNA does not alter efficiency of polyadenylation or transcription termination.

We constructed viable insertion mutants of polyomavirus that contain duplications of the nucleotide sequences surrounding the polyadenylation sites for both E- and L-strand RNAs. Our results showed that formation of poly(A)+ 3'termini of L-strand mRNAs requires sequence elements located between 12 and 87 nucleotides downstream of AAUAAA. No more than 19 nucleotides upstream and 44 nucleotides downstream of AAUAAA are required for polyadenylation of E-strand mRNAs. Our results and those of others suggest that there are three distinct sequence elements required for mRNA 3' end formation: AAUAAA and two downstream elements. An insertion mutant containing two adjacent functional polyadenylation signals produced E-strand and L-strand mRNAs with 3' ends at both sites. However, the overall level of polyadenylation of L-strand RNAs was not increased over the low (10 to 25%) levels seen with wild-type virus. Neither was the efficiency of termination of L-strand transcription increased in mutant virus-infected cells. We conclude that factors required for both polyadenylation and transcription termination are limiting in polyomavirus-infected mouse cells.

The expression and properties of polyoma virus middle-T antigen in simian cells

SV40 late replacement vectors containing the polyoma middle-T coding sequences have been constructed. Mixed hybrid virus stocks have been obtained through complementation with a defective SV40 helper genome (dl 1055) following DNA transfection into CV-1 cells. Middle-T antigen is expressed in the infected simian cells at about 5–10 fold higher levels than in polyoma virus-infected mouse cells and has the pp60 c-sec-associated tyrosine-specific protein kinase activity in vitro. However, the ‘specific activity’ of the kinase in extracts of the infected CV-1 cells is lower than that observed in polyoma infected 3T6 cell extracts. The half-life of middle-T antigen in the CV-1 cells is about 4 h but the in vitro kinase activity associated with middle-T has a half-life of at least 8 h and hence appears to be stabilized. The in vivo phosphorylated species of middle-T has been shown by sucrose gradient analyses to be largely distinct from the middle-T with associated protein kinase activity in vitro.

Three species of polyoma virus tumor antigens share common peptides probably near the amino termini of the proteins

Detergent extracts of polyoma virus-infected mouse cells contain three major proteins of approximately 100,000–108,000 (100K), 55,000 (55K) and 21,500 (22K) daltons, which react with sera obtained from rats carrying tumors induced by the virus. A comparison of the 35S-methionine-, 3H-leucine- and 3H-proline-labeled tryptic peptides of each of these proteins by cation-exchange chromatography followed by descending paper chromatography has shown that: at least five peptides are shared by all three T-reactive proteins; at least three peptides are shared by the 55K and 22K proteins, but not by the 100K protein; at least three peptides are found only in the 22K protein; at least six peptides are found only in the 55K protein; and at least sixteen peptides are found only in the 100K protein. The results are consistent with the hypothesis that the polypeptide chains of the 100K, 55K and 22K dalton tumor antigens of polyoma virus share a common virus-coded amino terminal region. The data also suggest that there is a portion of the polypeptide chains (probably immediately adjacent to the common amino terminal region of the molecules) that is shared by the 55K and 22K proteins, but not by the 100K protein (perhaps because this portion of the genetic information is spliced out of the messenger RNA coding for the 100K protein). The facts that all the peptides common to the 100K and 55K proteins are also found in the 22K protein and are thus assigned to the common amino terminal region of the molecules, and that there are several peptides unique to the 100K protein, as well as several peptides unique to the 55K protein, suggest that the presumed carboxy terminal portion of the polypeptide chain of the 100K protein is considerably, if not entirely, different from that of the 55K protein.

Polyoma virus giant RNAs contain tandem repeats of the nucleotide sequence of the entire viral genome.

The bulk of late virus-specific RNA synthesized in polyoma virus-infected mouse cells is larger than a single strand of poloma DNA. The arrangement of viral nucleotide sequences in these giant polyoma RNAs was studied by electron microscopy of hybrids between purified high molecular weight viral RNA and the HindII-1 fragment of polyoma DNA, which contains 91% of the viral genome. Hybrid molecules containing a short single-stranded gap (corresponding to the 9% of viral sequences not present in HindII-1), flanked by double-stranded regions, were photographed and measured. The majority of hybrid molecules contained no single-stranded loops or branches, showing that all viral sequences are transcribed contiguously and that no nonviral sequences are present in the RNA. Hybrid molecules, containing RNA up to 3.5 times the genome length, had a repeating structure of single-stranded gaps 8% of genome length interspersed with double-stranded regions 89% of genome length, showing that giant polyoma RNAs contain tandem repeats of the nucleotide sequence of the entire viral DNA. A small proportion of hybrid molecules contained single-stranded branches or deletion loops in characteristic positions, indicating that RNA "splicing" may occur on high molecular weight nuclear polyoma RNA.

Nuclear Localisation of DNA Polymerase α and DNA Synthesis in Polyoma Virus Infected Mouse Cells

Studies using inhibitors of DNA synthesis have shown that DNA polymerase alpha is located in nuclei of polyoma virus infected mouse cells to the same degree as these nuclei are engaged in DNA replication. These results indicate that either the enzyme is actively transported into nuclei concomitant with the onset of DNA synthesis, or that it is bound much more strongly in nuclei during DNA replication. In any case, these observations support the hypothesis that DNA polymerase alpha is involved in the replication of cellular and viral DNA.

DNA-Binding Proteins in Mouse Cells Infected with Polyoma Virus

DNA-binding proteins from uninfected or polyoma virus-infected mouse cells (3T6) were isolated and compared by DNA-cellulose chromatography and SDS-PAGE. During the early phase of infection, few but significant and reproducible differences were observed. In the 0.4 M NaCl eluate, four major new polypeptides were detected in infected cells, with approximate molecular weights of 160 K, 130 K, 86 K and 81 K, the last two being compatible with previous estimates for polyoma T antigen molecular weight.

Induction of DNA polymerase in polyoma virus-infected mouse cells requires transcription and translation

The induction of DNA polymerase activity and of DNA synthesis in polyoma virus-infected mouse kidney cells is inhibited by actinomycin D and cycloheximide, even in cells that are already T-antigen positive. This indicates that the induction of the DNA-synthesizing apparatus requires transcription and translation. The increase in cellular DNA polymerase activity and in the DNA synthesizing capacity follows the appearance of T-antigen only after a characteristic lag period of several hours.

Immunochemical procedure for partial purification of newly synthesized proteins in polyoma virus-infected mouse cells.

A broad-spectrum antiserum has been obtained against nuclear proteins from normal mouse fibroblasts. Solid phase immunoadsorbants prepared from the serum specifically bind the corresponding antigens. The removal in this way of a significant fraction of host proteins leads to partial purification of viral and virus-induced polypeptides from polyoma virus-infected cell extracts. Four main peptides can be selected, with respective molecular weights of 90,000, 70,000, 46,000, and 41,000.
