Polyhydroxyalkanoate Copolymer Production by Recombinant Ralstonia eutropha Strain 1F2 from Fructose or Carbon Dioxide as Sole Carbon Source

Ralstonia eutropha strain H16 is a chemoautotrophic bacterium that oxidizes hydrogen and accumulates poly[(R)-3-hydroxybutyrate] [P(3HB)], a prominent polyhydroxyalkanoate (PHA), within its cell. R. eutropha utilizes fructose or CO2 as its sole carbon source for this process. A PHA-negative mutant of strain H16, known as R. eutropha strain PHB−4, cannot produce PHA. Strain 1F2, derived from strain PHB−4, is a leucine analog-resistant mutant. Remarkably, the recombinant 1F2 strain exhibits the capacity to synthesize 3HB-based PHA copolymers containing 3-hydroxyvalerate (3HV) and 3-hydroxy-4-methyvalerate (3H4MV) comonomer units from fructose or CO2. This ability is conferred by the expression of a broad substrate-specific PHA synthase and tolerance to feedback inhibition of branched amino acids. However, the total amount of comonomer units incorporated into PHA was up to around 5 mol%. In this study, strain 1F2 underwent genetic engineering to augment the comonomer supply incorporated into PHA. This enhancement involved several modifications, including the additional expression of the broad substrate-specific 3-ketothiolase gene (bktB), the heterologous expression of the 2-ketoacid decarboxylase gene (kivd), and the phenylacetaldehyde dehydrogenase gene (padA). Furthermore, the genome of strain 1F2 was altered through the deletion of the 3-hydroxyacyl-CoA dehydrogenase gene (hbdH). The introduction of bktB-kivd-padA resulted in increased 3HV incorporation, reaching 13.9 mol% from fructose and 6.4 mol% from CO2. Additionally, the hbdH deletion resulted in the production of PHA copolymers containing (S)-3-hydroxy-2-methylpropionate (3H2MP). Interestingly, hbdH deletion increased the weight-average molecular weight of the PHA to over 3.0 × 106 on fructose. Thus, it demonstrates the positive effects of hbdH deletion on the copolymer composition and molecular weight of PHA.

Tailored polyhydroxyalkanoate production from renewable non-fatty acid carbon sources using engineered Cupriavidus necator H16

As thermoplastic, nontoxic, and biocompatible polyesters, polyhydroxyalkanoates (PHAs) are considered promising biodegradable plastic candidates for diverse applications. Short-chain-length/medium-chain-length (SCL/MCL) PHA copolymers are flexible and versatile PHAs that are typically produced from fatty acids, which are expensive and toxic. Therefore, to achieve the sustainable biosynthesis of SCL/MCL-PHAs from renewable non-fatty acid carbon sources (e.g., sugar or CO2), we used the lithoautotrophic bacterium Cupriavidus necator H16 as a microbial platform. Specifically, we synthesized tailored PHA copolymers with varying MCL-3-hydroxyalkanoate (3HA) compositions (10–70 mol%) from fructose by rewiring the MCL-3HA biosynthetic pathways, including (i) the thioesterase-mediated free fatty acid biosynthetic pathway coupled with the beta-oxidation cycle and (ii) the hydroxyacyl transferase-mediated fatty acid de novo biosynthetic pathway. In addition to sugar-based feedstocks, engineered strains are also promising platforms for the lithoautotrophic production of SCL/MCL-PHAs from CO2. The set of engineered C. necator strains developed in this study provides greater opportunities to produce customized polymers with controllable monomer compositions from renewable resources.

Microbially synthesized poly(hydroxybutyrate-co-hydroxyhexanoate) with low to moderate hydroxyhexanoate content: Properties and applications

Plastic pollution is the biggest environmental concern of our time. Breakdown products like micro- and nano-plastics inevitably enter the food chain and pose unprecedented health risks. In this scenario, bio-based and biodegradable plastic alternatives have been given a momentum aiming to bridge a transition towards a more sustainable future. Polyhydroxyalkanoates (PHAs) are one of the few thermoplastic polymers synthesized 100 % via biotechnological routes which fully biodegrade in common natural environments. Poly(hydroxybutyrate-co-hydroxyhexanoate) [P(HB-co-HHx)] is a PHA copolymer with great potential for the commodity polymers industry, as its mechanical properties can be tailored through fine-tuning of its molar HHx content. We have recently developed a strategy that enables for reliable tailoring of the monomer content of P(HB-co-HHx).Nevertheless, there is often a lack of comprehensive investigation of the material properties of PHAs to evaluate whether they actually mimic the functionalities of conventional plastics. We present a detailed study of P(HB-co-HHx) copolymers with low to moderate hydroxyhexanoate content to understand how the HHx monomer content influences the thermal and mechanical properties and to link those to their abiotic degradation. By increasing the HHx fractions in the range of 2 – 14 mol%, we impart an extension of the processing window and application range as the melting temperature (Tm) and glass temperature (Tg) of the copolymers decrease from Tm 165 °C to 126 °C, Tg 4 °C to −5.9 °C, accompanied by reduced crystallinity from 54 % to 20 %. Elongation at break was increased from 5.7 % up to 703 % at 14 mol% HHx content, confirming that the range examined was sufficiently large to obtain ductile and brittle copolymers, while tensile strength was maintained throughout the studied range. Finally, accelerated abiotic degradation was shown to be slowed down with an increasing HHx fraction decreasing from 70 % to 55 % in 12 h.

Production, optimization, scale up and characterization of polyhydoxyalkanoates copolymers utilizing dairy processing waste

The microbial biotransformation using low-cost feedstock to produce biopolymers (degradable), an alternative to petrochemical-based synthesis plastics (non-degradable), can be a beneficial approach towards sustainable development. In this study, the dairy industry processes waste (whey) is used in polyhydroxyalkanoate (PHA) copolymer production. Initial screening suggested that Ralstonia eutropha produced higher PHA as compared to Bacillus megaterium. A central composite rotatable design-based optimization using two process variables (amino acid and tween-80) concentration remarkably influenced PHA co-polymer production under physiological conditions of pH (7), temperature (37 °C), and agitation rate of 150 rpm. High polyhydroxybutyrate (PHB) mass fraction yield of 69.3% was observed as compared to predicted yield of 62.8% from deproteinized whey as feed. The combination of tryptophan (50 mg L−1) and tween-80 (3 mL−1) enhanced R. eutropha mass gain to 6.80 g L−1 with PHB contents of 4.71 g L−1. Further, characterization of PHA and its copolymers was done by ESI–MS, FTIR, and TEM. On upscaling up to 3.0 L, the PHA contents and yields were noted as quite similar by R. eutropha. This study demonstrates that dairy waste processing waste can be potentially utilized as inexpensive feed for producing high content of biopolymers to develop a sustainable system of waste management.

Gas fermentation combined with water electrolysis for production of polyhydroxyalkanoate copolymer from carbon dioxide by engineered Ralstonia eutropha

A recycled-gas closed-circuit culture system was developed for safe autotrophic cultivation of a hydrogen-oxidizing, polyhydroxyalkanoate (PHA)-producing Ralstonia eutropha, using a non-combustible gas mixture with low-concentration of H2 supplied by water electrolysis. Automated feedback regulation of gas flow enabled input of H2, CO2, and O2 well balanced with the cellular demands, leading to constant gas composition throughout the cultivation. The engineered strain of R. eutropha produced 1.71 g/L of poly(3-hydroxybutyrate-co-12.5 mol% 3-hydroxyhexanoate) on a gas mixture of H2/CO2/O2/N2 = 4:12:7:77 vol% with a 69.2 wt% cellular content. Overexpression of can encoding cytosolic carbonic anhydrase increased the 3HHx fraction up to 19.6 mol%. The yields of biomass and PHA on input H2 were determined to be 72.9 % and 63.1 %, corresponding to 51.0 % and 44.2 % yield on electricity, respectively. The equivalent solar-to-biomass/PHA efficiencies were estimated to be 2.1–3.8 %, highlighting the high energy conversion capability of R. eutropha.

Rapid Estimation of Poly(3-hydroxybutyrate-co-3-hydroxyvalerate) Composition Using ATR-FTIR.

A great research effort is involved in polyhydroxyalkanoates (PHAs) production and characterization since they are an attractive degradable polyester family that potentially could substitute oil-based polymers. This is due to two main key factors: their production is sustainable, being that they are produced by microorganisms possibly fed by organic waste-derived products, and they are degradable. Moreover, PHAs' thermal and mechanical properties could be tuned by varying their monomeric composition through the proper selection of microorganism feedstock and bioreactor operative conditions. Hence, a rapid and facile determination of the PHA chemical structure by widely available instrumentation is useful. As an alternative to the standard gas-chromatographic method, a new procedure for the composition determination of poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (P3HBV), the most common PHA copolymer, by attenuated total reflection FTIR (ATR-FTIR) is presented. It is based on the linear dependence of selected and normalized absorption band intensity with the molar fraction of repeating units. To break free from the crystallinity variability, which affects the result reproducibility and data scattering, the polymer sample was rapidly quenched from the melt directly on the surface of the ATR internal reflection element and analyzed. The data obtained from 14 samples with a molar fraction of 3-hydroxybutyrate repeating units (X3HB) ranging from 0.15 to 1 were analyzed. According to preliminary analyses, the normalized intensity of two absorption bands was selected to develop a calibration method able to predict X3HB of unknown samples and to evaluate the related uncertainty through prediction intervals of inverse regression. The proposed method proves to be useful for an easy and rapid estimation of P3HBV composition.

(R/S)-lactate/2-hydroxybutyrate dehydrogenases in and biosynthesis of block copolyesters by Ralstonia eutropha

Bacterial polyhydroxyalkanoates (PHAs) are promising bio-based biodegradable polyesters. It was recently reported that novel PHA block copolymers composed of (R)-3-hydroxybutyrate (3HB) and (R)-2-hydroxybutyrate (2HB) were synthesized by Escherichia coli expressing PhaCAR, a chimeric enzyme of PHA synthases derived from Aeromonas caviae and Ralstonia eutropha. In this study, the sequence-regulating PhaCAR was applied in the natural PHA-producing bacterium, R. eutropha. During the investigation, (R/S)-2HB was found to exhibit strong growth inhibitory effects on the cells of R. eutropha. This was probably due to formation of excess 2-ketobutyrate (2KB) from (R/S)-2HB and the consequent l-valine depletion caused by dominant l-isoleucine synthesis attributed to the excess 2KB. Deletion analyses for genes of lactate dehydrogenase homologs identified cytochrome-dependent d-lactate dehydrogenase (Dld) and [Fe-S] protein-dependent l-lactate dehydrogenase as the enzymes responsible for sensitivity to (R)-2HB and (S)-2HB, respectively. The engineered R. eutropha strain (phaCAR+, ldhACd-hadACd+ encoding clostridial (R)-2-hydroxyisocaproate dehydrogenase and (R)-2-hydoroxyisocaproate CoA transferase, ∆dld) synthesized PHA containing 10 mol% of 2HB when cultivated on glucose with addition of sodium (RS)-2HB, and the 2HB composition in PHA increased up to 35 mol% by overexpression phaCAR. Polyhydroxyalkanoate biosynthesis and optimisation of thermophilic Geobacillus stearothermophilus strain K4E3_SPR_NPP.

Polyhydroxyalkanoates (PHA) can be used to combat the challenges associated with plastic because it is biodegradable and can be produced from renewable resources. Extremophiles are considered to be potential PHA producers. An initial screening for the PHA synthesizing ability of a thermophilic bacteria Geobacillus stearothermophilus strain K4E3_SPR_NPP was carried out using Sudan black B staining. Nile red viable colony staining was used to further verify that the isolates produced PHA. Crotonic acid assays were used to determine the concentrations of PHA. The bacteria showed 31% PHA accumulation per dry cell weight (PHA/DCW) when glucose was used as a carbon source for growth. The molecule was identified to be medium chain length PHA, A copolymer of PHA containing poly(3-hydroxybutyrate)-poly(3-hydroxyvalerate)-poly(3-hydroxyhexanoate) (PHB-PHV-PHHX) using 1H-NMR. Six carbon sources and four nitrogen sources were screened for the synthesis of maximum PHA content, of which lactose and ammonium nitrate showed45% and 53% PHA/DCW respectively. The important factors in the experiment are identified using the Plackett-Burman design, and optimization is performed using the response surface method. Response surface methodology was used to optimize the three important factors, and the maximum biomass and PHA productions were discovered. Optimal concentrations yielded a maximum of 0.48g/l biomass and 0.32g/l PHA, measuring 66.66% PHA accumulation. Dairy industry effluent was employed for the synthesis of PHA, yielding 0.73g/l biomass and 0.33g/l PHA, measuring 45% PHA accumulation. These findings add credibility to the possibility of adopting thermophilic isolates for PHA production using low-cost substrates.

Tailoring the HHx monomer content of P(HB-co-HHx) by flexible substrate compositions: scale-up from deep-well-plates to laboratory bioreactor cultivations.

The enhanced material properties exhibited by the microbially synthetized polyhydroxyalkanoate (PHA) copolymer poly(hydroxybutyrate-co-hydroxyhexanoate) [P(HB-co-HHx)] evidence that this naturally biodegrading biopolymer could replace various functionalities of established petrochemical plastics. In fact, the thermal processability, toughness and degradation rate of P(HB-co-HHx) can be tuned by modulating its HHx molar content enabling to manufacture polymers à-la-carte. We have developed a simple batch strategy to precisely control the HHx content of P(HB-co-HHx) to obtain tailor-made PHAs with defined properties. By adjusting the ratio of fructose to canola oil as substrates for the cultivation of recombinant Ralstonia eutropha Re2058/pCB113, the molar fraction of HHx in P(HB-co-HHx) could be adjusted within a range of 2-17mol% without compromising polymer yields. The chosen strategy proved to be robust from the mL-scale in deep-well-plates to 1-L batch bioreactor cultivations.

Production of polyhydroxyalkanoate (PHA) copolymer from food waste using mixed culture for carboxylate production and Pseudomonas putida for PHA synthesis

Production of polyhydroxyalkanoates (PHAs) with high concentration of carboxylate, that was accumulated from solid state fermentation (SSF) of food waste (FW), was tested using Pseudomonas putida strain KT2440. Mixed-culture SSF of FW supplied in a high concentration of carboxylate, which caused a high PHA production of 0.56 g PHA/g CDM under nutrients control. Interestingly, this high PHA fraction in CDM was almost constant at 0.55 g PHA/g CDM even under high nutrients concentration (25 mM NH4+), probably due to high reducing power maintained by high carboxylate concentration. PHA characterization indicated that the dominant PHA building block produced was 3-hydroxybutyrate, followed by 3-hydroxy-2-methylvalerate and 3-hydroxyhenxanoate. Carboxylate profiles before and after PHA production suggested that acetate, butyrate, and propionate were the main precursors to PHA via several metabolic pathways. Our result support that mixed culture SSF of FW for high concentration carboxylate and P. putida for PHA production enables sustainable production of PHA in cost-effective manners.

Cascading Beta-oxidation Intermediates for the Polyhydroxyalkanoate Copolymer Biosynthesis by Metabolic Flux using Co-substrates and Inhibitors

ABSTRACTPolyhydroxyalkanoates (PHAs) are biopolymers that are produced within the microbial cells in the presence of excess carbon and nutrient limitation. Different strategies have been studied to increase the quality and quantity of this biopolymer which in turn can be utilized as biodegradable polymers replacing conventional petrochemical plastics. In the present study, Bacillus endophyticus, a gram-positive PHA-producing bacterium, was cultivated in the presence of fatty acids along with beta-oxidation inhibitor acrylic acid. A novel approach for incorporating different hydroxyacyl groups provided using fatty acids as co-substrate and beta-oxidation inhibitors to direct the intermediates to co-polymer synthesis was experimented. It was observed that higher fatty acids and inhibitors had a greater influence on PHA production. The addition of acrylic acid along with propionic acid had a positive impact, giving 56.49% of PHA along with sucrose which was 1.2-fold more than the control devoid of fatty acids and inhibitors. Along with the copolymer production, the possible PHA pathway functional leading to the copolymer biosynthesis was hypothetically interpreted in this study. The obtained PHA was analyzed by FTIR and 1H NMR to confirm the copolymer production, which indicated the presence of poly3hydroxybutyrate-co-hydroxyvalerate (PHB-co-PHV), poly3hydroxybutyrate-co-hydroxyhexanoate (PHB-co-PHx).

Influence of PHA Substrate Surface Characteristics on the Functional State of Endothelial Cells

The needs of modern regenerative medicine for biodegradable polymers are wide and varied. Restoration of the viability of the vascular tree is one of the most important components of the preservation of the usefulness of organs and tissues. The creation of vascular implants compatible with blood is an important task of vascular bioengineering. The function of the endothelial layer of the vessel, being largely responsible for the development of thrombotic complications, is of great importance for hemocompatibility. The development of surfaces with specific characteristics of biomaterials that are used in vascular technologies is one of the solutions for their correct endothelialization. Linear polyhydroxyalkanoates (PHAs) are biodegradable structural polymeric materials suitable for obtaining various types of implants and tissue engineering, having a wide range of structural and physicomechanical properties. The use of PHA of various monomeric compositions in endothelial cultivation makes it possible to evaluate the influence of material properties, especially surface characteristics, on the functional state of cells. It has been established that PHA samples with the inclusion of 3-hydroxyhexanoate have optimal characteristics for the formation of a human umbilical vein endothelial cell, HUVEC, monolayer in terms of cell morphology as well as the levels of expression of vinculin and VE-cadherin. The obtained results provide a rationale for the use of PHA copolymers as materials for direct contact with the endothelium in vascular implants.

Toughening Poly(3-hydroxybutyrate) by Using Catalytic Carbonylative Terpolymerization of Epoxides

The development of biodegradable and recyclable polymers having the desired material properties required in practical use has provided a partial solution for realizing a sustainable plastics economy. Naturally produced commercial poly(3-hydroxybutyrate) (P3HB), one of the much studied biodegradable polyhydroxyalkanoates (PHAs), is a stereoregular thermoplastic material with a high degree of crystallinity, significantly restricting its application due to the stiff and brittle characteristics. Therefore, it is of great significance to overcome the inherent brittleness through decreasing the crystallinity of P3HB. In this contribution, we present a powerful strategy to toughen P3HB via catalytic carbonylative terpolymerization of enantiopure epoxides with different long alkyl side chains and configurations using a binary trimetallic CrIII complex/carbonyl cobalt catalyst system. The structure and stereochemistry of epoxides significantly impact the properties of the resultant PHA copolymers, which spans from viscous oil to tough/ductile and brittle plastics. This study represents a rather rare example of a method to toughen the extensively studied P3HB with epoxide and CO as raw materials.

Multitask Neural Network for Mapping the Glass Transition and Melting Temperature Space of Homo- and Co-Polyhydroxyalkanoates Using σProfiles Molecular Inputs

Polyhydroxyalkanoates (PHAs) are an emerging type of bioplastic that have the potential to replace petroleum-based plastics. They are biosynthetizable, biodegradable, and economically viable and have a range of tunable properties. Despite their great potential, the structure and properties of PHA remain unexplored due to their theoretically infinite chemical space. Therefore, computational approaches for accurate predictions of their various properties need to be developed to effectively explore this large chemical space. For this purpose, this work presents a multitask artificial neural network (ANN) capable of predicting the glass transition temperature (Tg) and melting temperature (Tm) of PHA homopolymers and copolymers. The ANN inputs included the σProfiles as molecular parameters describing the monomer chemistry and their composition. In contrast, the polymer molecular weight (M) and polydispersity index (PDI) were used to describe the polymer state. The results showed that after optimizing the hyperparameters, the selected ANN architecture was remarkable in predicting the Tg and Tm of PHA with R2 values of 0.979 and 0.986 and average absolute relative deviation (AARD) of 0.476% and 0.520%, respectively. The proposed model represents an initiative to promote the development of robust, open source, and user-friendly models capable of predicting the properties of polymers based solely on molecular parameters (σProfiles), thereby saving time and resources for researchers worldwide. The framework described in this work is flexible so that it can be applied to a larger chemical space and incorporate other properties of polymers.

Biosynthesis of versatile PHA copolymers by thermophilic members of the genus Aneurinibacillus

Thermophilic members of the genus Aneurinibacillus constitute a remarkable group of microorganisms that exhibit extraordinary flexibility in terms of polyhydroxyalkanoates (PHA) synthesis. In this study, we demonstrate that these Gram-positive bacteria are capable of the utilization of selected lactones, namely, γ-valerolactone (GVL), γ-hexalactone (GHL), and δ-valerolactone (DVL) as the structural precursors of related PHA monomers. In the presence of GVL, a PHA copolymer consisting of 3-hydroxybutyrate, 3-hydroxyvalerate, and also 4-hydroxyvalerate was synthesized, with a 4 HV fraction as high as 53.1 mol%. Similarly, the application of GHL resulted in the synthesis of PHA copolymer containing 4-hydroxyhexanaote (4HHx) (4HHx fraction reached up to 11.5 mol%) and DVL was incorporated into PHA in form of 5-hydroxyvalerate (5 HV) (maximal 5 HV content was 44.2 mol%). The produced materials were characterized by thermoanalytical and spectroscopic methods; the results confirmed extremely appealing material properties of produced copolymers. Further, due to their unique metabolic features and capability of incorporating various PHA monomers into the PHA chain, thermophilic Aneurinibacillus spp. can be considered not only promising chassis for PHA production but also potential donors of PHA-relevant genes to improve PHA production in other thermophiles by using approaches of synthetic biology.

Unsterile production of a polyhydroxyalkanoate copolymer by Halomonas cupida J9

Microbial production of bioplastics polyhydroxyalkanoates (PHA) has opened new avenues to resolve “white pollution” caused by petroleum-based plastics. PHAs consisting of short- and medium-chain-length monomers, designated as SCL-co-MCL PHAs, exhibit much better thermal and mechanical properties than PHA homopolymers. In this study, a halophilic bacterium Halomonas cupida J9 was isolated from highly saline wastewater and proven to produce SCL-co-MCL PHA consisting of 3-hydroxybutyrate (3HB) and 3-hydroxydodecanoate (3HDD) from glucose and glycerol. Whole-genome sequencing and functional annotation suggest that H. cupida J9 may possess three putative PHA biosynthesis pathways and a class I PHA synthase (PhaCJ9). Interestingly, the purified His6-tagged PhaCJ9 from E. coli BL21 (DE3) showed polymerizing activity towards 3HDD-CoA and a phaCJ9-deficient mutant was unable to produce PHA, which indicated that a low-substrate-specificity PhaCJ9 was exclusively responsible for PHA polymerization in H. cupida J9. Docking simulation demonstrated higher binding affinity between 3HB-CoA and PhaCJ9 and identified the key residues involved in hydrogen bonds formation between 3-hydroxyacyl-CoA and PhaCJ9. Furthermore, His489 was identified by site-specific mutagenesis as the key residue for the interaction of 3HDD-CoA with PhaCJ9. Finally, PHA was produced by H. cupida J9 from glucose and glycerol in shake flasks and a 5-L fermentor under unsterile conditions. The open fermentation mode makes this strain a promising candidate for low-cost production of SCL-co-MCL PHAs. Especially, the low-specificity PhaCJ9 has great potential to be engineered for an enlarged substrate range to synthesize tailor-made novel SCL-co-MCL PHAs.

Oxidation of methionine-derived 2-hydroxyalkanoate unit in biosynthesized polyhydroxyalkanoate copolymers

In this study, 2-hydroxy-4-methylthiobutyrate (2H4MTB)-containing polyhydroxyalkanoate (PHA) copolymers were biosynthesized using methionine as the 2H4MTB precursor. 2H4MTB is a novel monomer unit that contains a sulfur atom in its side chain. The 2H4MTB-containing PHA was biosynthesized by functionalizing the leucine degradation and PHA synthesis pathways in recombinant Escherichia coli. The 2H4MTB fraction in the PHA copolymer was increased up to 30.6 mol% by increasing the methionine concentration in the medium. Using purified polymers containing 10.9 mol% 2H4MTB unit, the sulfide group of the 2H4MTB side chain was oxidized with hydrogen peroxide or peracetic acid, which resulted in the conversion of sulfide to sulfoxide and sulfone groups. The oxidized polymer was relatively hydrophilic, as revealed by water contact angle measurements, and swelled slightly when soaked in water. These results suggest that the 2H4MTB unit can be used as an oxidation site to impart hydrophilicity to the PHA copolymers.

Characterization of tars from recycling of PHA bioplastic and synthetic plastics using fast pyrolysis

The aim of this study was to investigate the pyrolysis products of polyhydroxyalkanoates (PHAs), polyethylene terephthalate (PET), carbon fiber reinforced composite (CFRC), and block co-polymers (PS-b-P2VP and PS-b-P4VP). The studied PHA samples were produced at temperatures of 15 and 50 oC (PHA15 and PHA50), and commercially obtained from GlasPort Bio (PHAc). Initially, PHA samples were analyzed by nuclear magnetic resonance (NMR) spectroscopy and size exclusion chromatography (SEC) to determine the molecular weight, and structure of the polymers. Thermal techniques such as thermogravimetry (TG) and differential scanning calorimetry (DSC) analyses were performed for PHA, CFRC, and block co-polymers to investigate the degradation temperature range and thermal stability of samples. Fast pyrolysis (500 oC, ∼102 °C s−1) experiments were conducted for all samples in a wire mesh reactor to investigate tar products and char yields. The tar compositions were investigated by gas chromatography–mass spectrometry (GC–MS), and statistical modeling was performed. The char yields of block co-polymers and PHA samples (<2 wt. %) were unequivocally less than that of the PET sample (~10.7 wt. %). All PHA compounds contained a large fraction of ethyl cyclopropane carboxylate (~ 38–58 %), whereas PAH15 and PHA50 additionally showed a large quantity of 2-butenoic acid (~8–12 %). The PHAc sample indicated the presence of considerably high amount of methyl ester (~15 %), butyl citrate (~12.9 %), and tributyl ester (~17 %). The compositional analyses of the liquid fraction of the PET and block co-polymers have shown carcinogenic and toxic properties. Pyrolysis removed matrices in the CRFC composites which is an indication of potential recovery of the original fibers.

Effect of short- and medium-chain fatty acid mixture on polyhydroxyalkanoate production by Pseudomonas strains grown under different culture conditions.

Short- and medium-chain fatty acids (SMCFAs) derived from the acidogenic anaerobic mixed culture fermentation of acid whey obtained from a crude cheese production line and their synthetic mixture that simulates a real SMCFA-rich stream were evaluated for polyhydroxyalkanoate (PHA) production. Three individual Pseudomonas sp. strains showed different capabilities of growing and producing PHAs in the presence of a synthetic mixture of SMCFAs. Pseudomonas sp. GL06 exhibited the highest SMCFA tolerance and produced PHAs with the highest productivity (2.7 mg/L h). Based on these observations, this strain was selected for further investigations on PHA production in a fed-batch bioreactor with a SMCFA-rich stream extracted from the effluent. The results showed that PHA productivity reached up to 4.5 mg/L h at 24 h of fermentation together with the ammonium exhaustion in the growth medium. Moreover, the PHA monomeric composition varied with the bacterial strain and the type of the growth medium used. Furthermore, a differential scanning calorimetric and thermogravimetric analysis showed that a short- and medium-chain-length PHA copolymer made of 3-hydroxybutyric, -hexanoic, -octanoic, -decanoic, and -dodecanoic has promising properties. The ability of Pseudomonas sp. to produce tailored PHA copolymers together with the range of possible applications opens new perspectives in the development of PHA bioproduction as a part of an integrated valorization process of SMCFAs derived from waste streams.

Conversion of Short and Medium Chain Fatty Acids into Novel Polyhydroxyalkanoates Copolymers by Aeromonas sp. AC_01.

Polyhydroxyalkanoates (PHAs) production by Aeromonas sp. AC_01 was investigated using synthetic and waste derived short and medium chain fatty acids (SMCFAs). The obtained results revealed that the analyzed bacterial strain was able to grow and synthesize PHAs using SMCFAs. The highest PHA productivity was observed in the cultivation supplemented with a mixture of acetic acid and butyric acid (3.89 mg/L·h). Furthermore, SMCFAs-rich stream, derived from acidogenic mixed culture fermentation of acid whey, was found to be less beneficial for PHA productivity than its synthetic mixture, however the PHA production was favored by the nitrogen-limited condition. Importantly, Aeromonas sp. AC_01 was capable of synthesizing novel scl-mcl copolymers of 3-hydroxybutyrate (3HB), 3-hydroxyvalerate (3HV), 3-hydroxytridecanoate (3HtriD) and/or 3-hydroxytetradecaonate (3HTD) with high 3HB and 3HV fractions. They were identified with alterable monomers composition depending on the culture conditions used. Moreover, in-depth thermal analyses proved that they are highly resistant to thermal degradation regardless of their monomeric composition. The obtained results confirm that Aeromonas sp. AC_01 is a promising candidate for the biotechnological production of PHAs from SMCFAs with thermal properties that can be tuned together with their chemical composition by the corresponding adjustment of the cultivation process.
