Efficient performance of InP and InP/ZnS quantum dots for photocatalytic degradation of toxic aquatic pollutants.

In recent years, the growing concern over the presence of toxic aquatic pollutants has prompted intensive research into effective and environmentally friendly remediation methods. Photocatalysis using semiconductor quantum dots (QDs) has developed as a promising technology for pollutant degradation. Among various QD materials, indium phosphide (InP) and its hybrid with zinc sulfide (ZnS) have gained considerable attention due to their unique optical and photocatalytic properties. Herein, InP and InP/ZnS QDs were employed for the removal of dyes (crystal violet, and congo red), polyaromatic hydrocarbons (pyrene, naphthalene, and phenanthrene), and pesticides (deltamethrin) in the presence of visible light. The degradation efficiencies of crystal violet (CV) and congo red (CR) were 74.54% and 88.12% with InP, and 84.53% and 91.78% with InP/ZnS, respectively, within 50min of reaction. The InP/ZnS showed efficient performance for the removal of polyaromatic hydrocarbons (PAHs). For example, the removal percentage for naphthalene, phenanthrene, and pyrene was 99.8%, 99.6%, and 88.97% after the photocatalytic reaction. However, the removal percentage of InP/ZnS for pesticide deltamethrin was 90.2% after 90 min light irradiation. Additionally, advanced characterization techniques including UV-visible spectrophotometer (UV-Vis), photoluminescence (PL), X-ray diffractometer (XRD), energy-dispersive spectrometer (EDS) elemental mapping, transmission electron microscopy (TEM), and thermogravimetric analysis (TGA) were used to analyze the crystal structure, morphology, and purity of the fabricated materials in detail. The particle size results obtained from TEM are in the range of 2.28-4.60nm. Both materials (InP and InP/ZnS) exhibited a spherical morphology, displaying distinct lattice fringes. XRD results of InP depicted lattice planes (111), (220), and (311) in good agreement with cubic geometry. Furthermore, the addition of dopants was discovered to enhance the thermal stability of the fabricated material. In addition, QDs exhibited efficacy in the breakdown of PAHs. The analysis of their fragmentation suggests that the primary mechanism for PAHs degradation is the phthalic acid pathway.

Bacterial crude oil and polyaromatic hydrocarbon degraders from Kazakh oil fields as barley growth support

Bacterial strains of the genera Arthrobacter, Bacillus, Dietzia, Kocuria, and Micrococcus were isolated from oil-contaminated soils of the Balgimbaev, Dossor, and Zaburunye oil fields in Kazakhstan. They were selected from 1376 isolated strains based on their unique ability to use crude oil and polyaromatic hydrocarbons (PAHs) as sole source of carbon and energy in growth experiments. The isolated strains degraded a wide range of aliphatic and aromatic components from crude oil to generate a total of 170 acid metabolites. Eight metabolites were detected during the degradation of anthracene and of phenanthrene, two of which led to the description of a new degradation pathway. The selected bacterial strains Arthrobacter bussei/agilis SBUG 2290, Bacillus atrophaeus SBUG 2291, Bacillus subtilis SBUG 2285, Dietzia kunjamensis SBUG 2289, Kocuria rosea SBUG 2287, Kocuria polaris SBUG 2288, and Micrococcus luteus SBUG 2286 promoted the growth of barley shoots and roots in oil-contaminated soil, demonstrating the enormous potential of isolatable and cultivable soil bacteria in soil remediation.Key points• Special powerful bacterial strains as potential crude oil and PAH degraders.• Growth on crude oil or PAHs as sole source of carbon and energy.• Bacterial support of barley growth as resource for soil remediation.

Deciphering the photocatalytic degradation of polyaromatic hydrocarbons (PAHs) using hausmannite (Mn3O4) nanoparticles and their efficacy against bacterial biofilm

Polyaromatic hydrocarbons (PAHs) are life-threatening organic pollutants that severely threaten ecosystems worldwide due to their poisonous qualities, cancer-causing properties, and mutation-causing qualities. Water and soil together form a critical component of the ecosystem that supports all life. Due to the pollutants that are being disposed of in them, their characteristics have changed, and their toxicity has increased. The goal of this study was to investigate the ability of hausmannite nanoparticles to degrade fluorene from soil and water. Using the chemical method, hausmannite nanoparticles were synthesized and further characterization was performed using UV–Vis, FTIR, DLS, XRD, and SEM-EDAX. Hausmannite significantly degraded fluorene using the batch adsorption method. The degradation was also confirmed by performing reactive kinetics using Freundlich's isotherm model and Langmuir's pseudo-second-order model of soil and water. In addition to the degradation efficacy, hausmannite was also proved to inhibit biofilm formation by Pseudomonas aeruginosa. The findings of the experiments confirmed the presence of hausmannite nanoparticles, as well as their physical properties, chemical properties, degradation properties, and parameters of the kinetic study. As a result, synthesized nanoparticles have been extensively utilized as a low-cost option for removing pollutants and microbial biofilm.

Phylloremediation of pyrene and anthracene by endophytic fungi inhabiting tea leaves (Camellia sinensis (L.) Kuntze) in Sri Lanka

ABSTRACT Polyaromatic hydrocarbons (PAHs) released from vehicular emissions and oil refineries deposit on the phyllosphere, compromising the quality of leaf-based food products by posing many health issues. Nevertheless, the tea phyllosphere harbours a variety of endophytes that are highly effective at degrading polyaromatic hydrocarbons, anthracene, and pyrene. The present study attempts to analyse the pyrene and anthracene degrading capability of phyllosphere endophytic fungi that inhabit Camellia sinensis (L.) Kuntze leaves. The frequency of occurrence of endophytic fungi in different leaf tissue layers was examined using light and scanning electron microscopy (SEM). The best pyrene and anthracene degrading strains were selected based on the High-Performance Liquid Chromatography (HPLC) results, and further kinetic assays. Light microscopy and SEM observations highlighted a heterogeneous endophytic fungal distribution among leaf tissue layers; the upper epidermis had the highest fungal distribution compared to other leaf layers. HPLC results revealed that Phyllosticta capitalensis, Colletotrichum gloeosporioides, Colletotrichum siamense, Pseudopestalotiopsis chinensis, and Daldinia eschscholtzii, have higher pyrene and anthracene degradation respectively and their PAH degradation kinetics follow the first-order kinetic model. The best anthracene and pyrene degrader, P. capitalensis showed the lowest half-life. The present investigation highlights the potential of P. capitalensis, the best pyrene and anthracene degrader that can remediate PAHs deposited on the phyllosphere of tea leaves.

Assessment of Vinca rosea (Apocynaceae) Potentiality for Remediation of Crude Petroleum Oil Pollution of Soil

Petroleum oil pollution is a worldwide problem that results from the continuous exploration, production, and consumption of oil and its products. Petroleum hydrocarbons are produced as a result of natural or anthropogenic practices, and their common source is anthropogenic activities, which impose adverse effects on the ecosystem’s nonliving and living components including humans. Phytoremediation of petroleum hydrocarbon-polluted soils is an evolving, low-cost, and effective alternative technology to most traditional remediation methods. The objective of this study is to evaluate the phytoremediation potentiality of Vinca rosea for crude oil-contaminated soil by understanding its properties and involvement in the enhanced degradation of crude oil. The remediation potentiality was determined by evaluating the total petroleum hydrocarbon degradation percentage (TPH%) and changes in the molecular type composition of saturated and aromatic hydrocarbon fractions. TPH% was estimated gravimetrically, and changes in the molecular type composition of saturated and aromatic fractions were measured using gas chromatography and high-performance liquid chromatography, respectively. Sulfur concentration was measured using X-ray fluorescence. Cadmium and lead quantification was measured using Inductively Coupled Plasma-Atomic Emission Spectrometry (ICP-AES). The results revealed that V. rosea enhanced total petroleum hydrocarbon (TPH) degradation and altered the molecular composition of the crude oil. The saturated hydrocarbons increased and the aromatic hydrocarbons decreased. The saturated hydrocarbon fraction in the crude oil showed a wider spectrum of n-paraffin peaks than the oil extracted from unplanted and V. rosea-planted soils. Polyaromatic hydrocarbon degradation was enhanced in the presence of V. rosea, which was reflected in the increase of monoaromatic and diaromatic constituents. This was parallel to the increased sulfur levels in planted soil. The determination of sulfur and heavy metal content in plant organs indicated that V. rosea can extract and accumulate high amounts from polluted soils. The ability of V. rosea to degrade TPH and alter the composition of crude petroleum oil by decreasing the toxicity of polyaromatic hydrocarbons in soil, as well as its capability to absorb and accumulate sulfur and heavy metals, supports the use of plant species for the phytoremediation of crude oil-polluted sites.

A novel oxygen-deficient core–shell ZA@ZnO1-X nanocomposite for enhanced degradation of multiple polyaromatic hydrocarbons

Persistent organic pollutants such as polyaromatic hydrocarbons (PAHs) are one of the emerging micropollutants present in several industrial effluents. Being trace refractory organics, its treatment by conventional method is very challenging. Thus, we report a novel core–shell zeolite A@oxygen-deficient ZnO (ZA@ZnO1-X) nanocomposite for degradation of PAHs mixture containing fluorene, phenanthrene, and anthracene. The hierarchical structure helps to reduce the recombination of photogenerated electron and holes pair, thereby availing more active species for photodegradation. The enhanced photodegradation efficacy is due to the synergistic effect of oxygen-defect sites in ZA@ZnO1-X and photoactive species. Core-shell ZA@ZnO1-X shows improved catalytic properties with a band gap value of 2.65 eV lower than the sole ZnO nanosheet (3.03 eV). The porous shell of the ZnO1-X structure provides an enhanced adsorption site and fully utilizes photon energy (visible light) with no aggregation because of ZA support. The maximum photocatalytic degradation is achieved by optimizing reaction parameters of 96%, 95.1%, and 93% of fluorene (0.02436 min−1), phenanthrene (0.02421 min−1), and anthracene (0.02102 min−1), respectively in 2 h at neutral pH and catalyst load 1 g/l. The primary active species responsible for PAHs degradation is h+, followed by HO•, O2•- radicals analyzed by quenching experiment. The degradation intermediates, and degraded products analysis by GC–MS gives a plausible general reaction pathway of PAHs and reveals primary intermediate is a phthalate derivative. Finally, the regeneration of active catalytic sites in the ZA@ZnO1-X photocatalyst shows its stability and reusability over five consecutive cycles.

Biodegradation of asphaltene by lipopeptide-biosurfactant producing hydrocarbonoclastic, crude oil degrading Bacillus spp.

Asphaltene is the most recalcitrant compound in crude oil. Bacteria were isolated from crude oil contaminated soil and their efficiency for hydrocarbon degradation was determined using GC–MS and isolates were screened for biosurfactant production using FT-IR. Two Bacillus spp. having hydrocarbonoclastic and lipo-peptide biosurfactant-producing abilities were experimented for their asphaltene removal potential through oil removal efficiency (ORE%) and asphaltene degradation efficiency (ADE%). B. thuringeinsis SSL1 and B. cereus SSL3 could degrade 76.4% and 67.4% of asphaltene (20 g L−1), in vitro, respectively, which is much higher than previous reports. B. thuringiensis SSL1 is recommended for effective breakdown of asphaltene, total petroleum hydrocarbon, and polyaromatic hydrocarbon degradation, aided by its biosurfactants, which is useful for crude oil cleanup. Biosurfactants are important for enhancing the availability of hydrophobic hydrocarbons to bacteria, which is beneficial for efficient crude oil remediation. These findings could lead to more effective strategies for complete clean-up of crude oil pollution.

Polyaromatic hydrocarbons (PAHs) in the water environment: A review on toxicity, microbial biodegradation, systematic biological advancements, and environmental fate

Polycyclic aromatic hydrocarbons (PAHs) are considered a major class of organic contaminants or pollutants, which are poisonous, mutagenic, genotoxic, and/or carcinogenic. Due to their ubiquitous occurrence and recalcitrance, PAHs-related pollution possesses significant public health and environmental concerns. Increasing the understanding of PAHs' negative impacts on ecosystems and human health has encouraged more researchers to focus on eliminating these pollutants from the environment. Nutrients available in the aqueous phase, the amount and type of microbes in the culture, and the PAHs' nature and molecular characteristics are the common factors influencing the microbial breakdown of PAHs. In recent decades, microbial community analyses, biochemical pathways, enzyme systems, gene organization, and genetic regulation related to PAH degradation have been intensively researched. Although xenobiotic-degrading microbes have a lot of potential for restoring the damaged ecosystems in a cost-effective and efficient manner, their role and strength to eliminate the refractory PAH compounds using innovative technologies are still to be explored. Recent analytical biochemistry and genetically engineered technologies have aided in improving the effectiveness of PAHs' breakdown by microorganisms, creating and developing advanced bioremediation techniques. Optimizing the key characteristics like the adsorption, bioavailability, and mass transfer of PAH boosts the microorganisms' bioremediation performance, especially in the natural aquatic water bodies. This review's primary goal is to provide an understanding of recent information about how PAHs are degraded and/or transformed in the aquatic environment by halophilic archaea, bacteria, algae, and fungi. Furthermore, the removal mechanisms of PAH in the marine/aquatic environment are discussed in terms of the recent systemic advancements in microbial degradation methodologies. The review outputs would assist in facilitating the development of new insights into PAH bioremediation.

Comparative Roles of HBB5 Biosurfactant and Poultry Wastes in Polyaromatic Hydrocarbon Biodegradiation of Crude Oil-contaminated Sediment

The comparative study of poultry wastes- and HBB5 biosurfactant-mediated polyaromatic hydrocarbon biodegradation in sediment polluted with crude oil were investigated. The experiments were carried out for a period of 28 days by monitoring pH, nitrate, phosphate, polycyclic aromatic hydrocarbon and microbiological parameters using standard procedures. The pH values obtained ranged between 6.21 and 6.93 in days 1 and 28 for the most effective treatment recipes. Generally, there was depletion in the concentrations of nitrate and phosphate for all set ups, but the most effective recipe witnessed highest reduction. For the polycyclic aromatic hydrocarbons, the recipe with highest limiting nutrients depletion also recorded the most hydrocarbon loss, and yet highest increase in density of hydrocarbonoclastic bacteria and fungi. The sample containing polluted sediment + poultry wastes + HBB5 biosurfactant recorded PAH values of 1932.6472ppm on day 1 and 481.2272ppm on day 28. Total hydrocarbon-utilizing bacterial counts ranged from 1.48×104 cfu/g to 9.70×106 cfu/g, while hydrocarbon-utilizing fungal counts ranged between 2.30×103 cfu/g and 3.90×105 cfu/g. From the results obtained, poultry wastes combined with HBB5 biosurfactant recorded the highest efficiency in the biodegradation of polycyclic aromatic hydrocarbons in sediments, and HBB5 biosurfactant in isolation recorded higher degradation efficiency for polyaromatic hydrocarbons than the degradation effect mediated by poultry wastes alone. It is therefore recommended that a combination of surface-active agent, nutrient amendment source and viable microbial biomass be adopted and employed as potent recipe for the degradation of polyaromatic hydrocarbons in crude oil-contaminated sediments.

Bacterial degradation of mixed-PAHs and expression of PAH-catabolic genes.

Polyaromatic hydrocarbons (PAHs) are hazardous organic compounds with established toxicity, carcinogenicity, and mutagenicity, ubiquitous distribution, and persistence in different environmental matrices. In the present study, degradation of the mixture of PAHs (phenanthrene, anthracene, fluorene, and pyrene) by Kocuria flava and Rhodococcus pyridinivorans was investigated. The individual strains and consortium of both degraded 55.6%, 59.5%, and 59.1% of 10mgL-1 of mixed PAHs, respectively, within 15days. The participation of catabolic enzymes [catechol 2,3-dioxygenase (C23O), dehydrogenase (DH), and peroxidase (POD)] was confirmed during catalytic oxidation through meta-cleavage of mixed PAHs in this study. The catabolic gene expression of naphthalene dioxygenase (NAH) and catechol 2,3-dioxygenase (C23O) during degradation was confirmed using RT-qPCR in the present study. This is the first study that shows significant gene expression of the catabolic genes during degradation of mixed PAHs by selected bacterial strains. The C23O gene showed a 6.02 log fold higher expression in Kocuria flava in comparison to Rhodococcus pyridinivorans whereas NAH gene exhibited a 7.9 log fold higher expression in Rhodococcus pyridinivorans in comparison to Kocuria flava. Hence it is likely to conclude that combination of Kocuria flava and Rhodococcus pyridinivorans can effectively remove hazardous mixture of PAHs from the contaminated environmental matrix.

Depolymerization of polyaromatic hydrocarbons by Penicillium spp. inhabit the phyllosphere of urban ornamental plants

AbstractA variety of anthropogenic sources release hazardous polyaromatic hydrocarbons (PAHs) into the phyllosphere which is an excellent niche for diverse fungi, and some of them have PAHs degradation capabilities. Therefore, this research attempted to determine the PAHs (phenanthrene, anthracene, naphthalene, and pyrene) degradation capability of phyllosphere inhabited Penicillium species. The leaf samples were collected from highly polluted urban areas (Panchikawatta, Pettah, Orugodawatta, Maradana, Sapugaskanda, and Colombo Fort) in Sri Lanka to isolate fungal species inhabiting the phyllosphere. Furthermore, their distribution patterns among the leaf tissue layers were studied using bright‐field microscopic observations. Moreover, the best PAH degraders were screened out using plate assays and confirmed through High Performance Liquid Chromatography (HPLC) analysis. Further, their enzymatic activities during the PAHs degradation were analyzed. As per the microscopic observations, the highest fungal distribution was in the upper epidermis of the leaves followed by the fungal distribution in the interspaces of palisade mesophyll layers. Out of isolated fungal species, two Penicillium spp. (Penicillium citrinum P23B‐91 and Penicillium griseofulvum P9B ‐ 30) showed the highest PAHs (phenanthrene, anthracene, naphthalene, and pyrene) degradation capabilities. Manganese peroxidase (MnP) enzyme dominated phenanthrene degradation in P. griseofulvum P9B ‐ 30, which showed the highest phenanthrene degradation ability (61%). In addition, P. citrinum P23B‐91 was good at degrading anthracene (88%) and also displayed a higher MnP activity during the anthracene degradation than laccase and lignin peroxidase activities. The discoveries from the toxicity assay during the PAHs degradation processes revealed that the produced byproducts had no toxic effects on the fungal growth cycle and the phyllosphere. Therefore this phyllosphere Penicillium spp. are ideal for the bioremediation of polluted air in urbanized areas.

Mixed polyaromatic hydrocarbon degradation by halotolerant bacterial strains from marine environment and its metabolic pathway

Accidents involving diesel oil spills are prevalent in sea- and coastal regions. Polycyclic aromatic hydrocarbons (PAHs) can be adsorbed in soil and constitute a persistent contaminant due to their poor water solubility and complex breakdown. PAHs pollution is a pervasive environmental concern that poses serious risks to human life and ecosystems. Thus, it is the need of the hour to degrade and decontaminate the toxic pollutant to save the environment. Among all the available techniques, microbial degradation of the PAHs is proving to be greatly beneficial and effective. Bioremediation overcomes the drawbacks of most physicochemical procedures by eliminating numerous organic pollutants at a lower cost in ambient circumstances and has therefore become a prominent remedial option for pollutant removal, including PAHs. In the present study, we have studied the degradation of Low molecular Weight and High Molecular Weight PAH in combination by bacterial strains isolated from a marine environment. Optimum pH, temperature, carbon, and nitrogen sources, NaCl concentrations were found for efficient degradation using the isolated bacterial strains. At 250 mg/L concentration of the PAH mixture an 89.5% degradation was observed. Vibrio algiolytcus strains were found to be potent halotolerant bacteria to degrade complex PAH into less toxic simple molecules. GC-MS and FTIR data were used to probe the pathway of degradation of PAH.

Polyaromatic Hydrocarbons (PAHs) Degradation Ability of Penicillium spp. Isolated from Phyllosphere of Urban Areas in Sri Lanka

Polyaromatic hydrocarbons (PAHs), like highly toxic pollutants, are released into the air by various anthropogenic sources, and then they get settled on ground level surfaces as phyllosphere. Phyllosphere is an excellent niche to diverse fungi, and some of them are with PAH degradation capabilities. Out of them, Penicillium spp., which are highly abundant in different natural and contaminated atmospheres, have been leading for many research due to their high degree of physiological and genetic adaptation to degrade broad range of polyaromatic compounds. Therefore, this research attempted to determine the PAH (phenanthrene, anthracene, naphthalene and pyrene) degradation capability of phyllosphere inhabited Penicillium species. Fungal isolations were done from leaf samples collected from Panchikawatta, Orugodawatta, Pettah, Maradana, Colombo Fort and Sapugaskanda oil refinery sites in Sri Lanka. Isolated Penicillium spp. were identified up to genus level through colony morphology and microscopic observations. PAH degradation ability of isolated Penicillium spp. was screened using plate assay and confirmed through High Performance Liquid Chromatography (HPLC) analysis. Further, toxicity assays were performed. HPLC analysis results revealed that Penicillium sp. 1 showed the efficient degradation on phenanthrene (79%) and naphthalene (78%) while Penicillium sp. 2 showed the most efficient degradation on Anthracene (80%) and pyrene (66%) Toxicity assays results revealed that the metabolites of these PAHs degradation mechanism were not toxic for the growth of Penicillum spp, and also confirmed that those by-products were not harmful to the phyllosphere. All in all, Penicillium spp. showed efficient phenanthrene, anthracene, naphthalene, and pyrene like PAH degradation ability, with more than 54% of PAH degradation. Therefore this phyllosphere Penicillium spp. can be used in bioremediation of polluted air in the urbanized areas.

Impact of biosurfactant and iron nanoparticles on biodegradation of polyaromatic hydrocarbons (PAHs)

Polycyclic aromatic hydrocarbons (PAHs) are hazardous toxic contaminants and considered as primary pollutants due to their persistent nature and most of them are carcinogenic and mutagenic. The key challenge in PAHs degradation is their hydrophobic nature, which makes them one of the most complex materials and inaccessible by a broad range of microorganisms. This bioavailability can be increased by using a biosurfactant. In the present study mixed PAHs were degraded using the biosurfactant producing bacterial strains. In addition, iron nanoparticles were synthesized and the impact of iron nanoparticles on the growth of the mixed bacterial strains (Pseudomonas stutzeri NA3 and Acinetobacter baumannii MN3) was optimized. The mixed PAHs (anthracene, pyrene, and benzo(a)pyrene) degradation was enhanced by addition of biosurfactant (produced by Bacillus subtilis A1) and iron nanoparticles, resulting in 85% of degradation efficiency. The addition of the biosurfactant increased the bioavailability of the PAHs in the aqueous environment, which might help bacterial cells for the initial settlement and development. The addition of iron nanoparticles increased both bacterial biomass and PAHs adsorption over their surface. These overall interactions assisted in the utilization of PAHs by the mixed bacterial consortia. This study illustrates that this integrated approach can be elaborated for the removal of the complex PAHs pollutants from soil and aqueous environments.

Metagenomic analysis for taxonomic and functional potential of Polyaromatic hydrocarbons (PAHs) and Polychlorinated biphenyl (PCB) degrading bacterial communities in steel industrial soil.

Iron and steel industries are the major contributors to persistent organic pollutants (POPs). The microbial community present at such sites has the potential to remediate these contaminants. The present study highlights the metabolic potential of the resident bacterial community of PAHs and PCB contaminated soil nearby Bhilai steel plant, Chhattisgarh (India). The GC-MS/MS analysis of soil samples MGB-2 (sludge) and MGB-3 (dry soil) resulted in identification of different classes of POPs including PAHs {benzo[a]anthracene (nd; 17.69%), fluorene (15.89%, nd), pyrene (nd; 18.7%), benzo(b)fluoranthene (3.03%, nd), benzo(k)fluoranthene (11.29%; nd), perylene (5.23%; nd)} and PCBs (PCB-15, PCB-95, and PCB-136). Whole-genome metagenomic analysis by Oxford Nanopore GridION Technology revealed predominance of domain bacteria (97.4%; 97.5%) followed by eukaryote (1.4%; 1.5%), archaea (1.2%; 0.9%) and virus (0.02%; 0.04%) in MGB-2 and MGB-3 respectively. Proteobacteria (44.3%; 50.0%) to be the prominent phylum followed by Actinobacteria (22.1%; 19.5%) in MBG-2 and MBG-3, respectively. However, Eukaryota microbial communities showed a predominance of phylum Ascomycota (20.5%; 23.6%), Streptophyta (18.5%, 17.0%) and unclassified (derived from Eukaryota) (12.1%; 12.2%) in MGB-2 and MGB-3. The sample MGB-3 was richer in macronutrients (C, N, P), supporting high microbial diversity than MGB-2. The presence of reads for biphenyl degradation, dioxin degradation, PAH degradation pathways can be further correlated with the presence of PCB and PAH as detected in the MGB-2 and MGB-3 samples. Further, taxonomic vis-à-vis functional analysis identified Burkholderia, Bradyrhizobium, Mycobacterium, and Rhodopseudomonas as the keystone degrader of PAH and PCB. Overall, our results revealed the importance of metagenomic and physicochemical analysis of the contaminated site, which improves the understanding of metabolic potential and adaptation of bacteria growing under POP contaminated environments.

Bio-polysaccharide composites mediated degradation of polyaromatic hydrocarbons in a sandy soil using free and immobilized consortium of Kocuria rosea and Aspergillus sydowii.

Based on our previous study in minimal medium, Kocuria rosea and Aspergillus sydowii were identified as the best microbes for degradation of mixture of polyaromatic hydrocarbons (PAHs). The present study reports PAH degradation potential of these microbes in free and immobilized form. PAHs were extracted using QuEChERS-mediated process followed by quantification by high performance liquid chromatography. The microbial consortium of Kocuria rosea + Aspergillus sydowii was formulated in three bio-formulations, namely (i) bentonite-alginate composite beads; (ii) water dispersible granule composite using guar gum-nanobentonite; and (iii) composites of carboxymethyl cellulose-bentonite and were applied in PAH fortified (100µgg-1) sandy loam soil. Results suggested that degradation data fitted well to first order kinetics as in most of the cases, the values of correlation coefficient (r) were > 0.95. The half-life (t1/2) values for PAHs in the uninoculated control soil were: naphthalene (10.43 d), fluorene (22.43 d), phenanthrene (24.64 d), anthracene (38.47 d), and pyrene (34.34 d). Inoculation of soil with free culture microbial consortium (without or with nutrient) and bio-formulation of degrading cultures enhanced degradation of all PAHs and half-life values were significantly reduced for each PAH: naphthalene (1.76-2.00 d), fluorene (2.52-6.65 d), phenanthrene (4.61-6.37 d), anthracene (9.01-12.22 d), and pyrene (10.98-15.55 d). Among different bio-formulations, guar gum-nanobentonite-based composite exhibited better efficacy for degradation of naphthalene, fluorene, phenanthrene, anthracene, and pyrene. The addition of microbial consortium in PAH fortified soil increased 16S rRNA gene copies of Alphaproteobacteria and Bacteroidetes, compared to the uninoculated, PAH-fortified control. The microbial functional gene assays showed that the gene copies of amoA, nirK, nirS, and anammox increased, suggesting nitrogen regulation in the PAH-fortified soil.

Pragmatic Treatment Strategies for Polyaromatic Hydrocarbon Remediation and Anti-biofouling from Surfaces Using Nano-enzymes: a Review.

In this review, two important environmental pollutants have been considered for its potential remediation using microbial-derived nano-enzymes. Firstly, polyaromatic hydrocarbons (PAHs) are one of the major industrial contaminants in the environment due to their ubiquitous occurrence, toxicity, and proclivity for bioaccumulation. Secondly, biofouling due to biofilm-forming organisms that impact tremendous economic and environmental consequences in many industries, especially marine vessels where it causes an increase in hydrodynamic drag, which results in a loss of ship speed at constant power or a power increase to maintain the same speed with higher fuel consumption and emissions into the atmosphere, particularly Green House Gases (GHGs).Among the remediation strategies, biological routes are found to be promising, efficient, and sustainable. Natural ligninolytic enzymes such as MnP, LiP, laccase, peroxidases, and polysaccharide and protein degradative enzymes are found to be highly efficient for PAH degradation and antifouling respectively. However, large-scale usage of these enzymes is difficult due to various reasons like their poor stability, adaptation, and high-cost production of these enzymes. In recent years, the use of nanoparticles, particularly nano-enzymes, is found to be an innovative and synergistic approach to detoxify contaminated areas with concomitant maintenance of enzyme stability.

Identification of bacteria by poly-aromatic hydrocarbon biosensors.

Human health is consistently threatened by different species of pathogenic bacteria. To fight the spread of diseases, it is important to develop rapid methods for bacterial identification. Over the years, different kinds of biosensors were developed for this cause. Another environmental risk is poly-aromatic hydrocarbons (PAHs) that may be emitted from industrial facilities and pollute environmental water and soil. One of the methods for their purification is conducted by the addition of bacteria that can degrade the PAHs, while the bacteria can be filtrated at the end of the process. Although many studies reported monitoring of the PAHs degradation by fluorescence, not much attention was dedicated to studying the influence of the PAHs on the intrinsic fluorescence of the degrading bacteria. In this work, we apply synchronous fluorescence (SF) measurements to study the ability of the 5 PAHs: 9-Antracene carboxylic acid (9ACA), Pyrene, Perylene, Pentacene, and Chrysene to interact with bacteria and change its fluorescence spectra. We show that upon incubation of each PAH with the bacterium E. coli, only the 2 PAHs 9ACA and Perylene cause an intensity decrease in the emission at λ = 300-375nm, which derives from the emission of tyrosine and tryptophan (TT). Also, we show that upon incubation of 9ACA and Perylene with 5 different pathogenic bacteria, the intensity increase or decrease in the TT emission is unique to each bacterial species. Based on this observation, we suggest that the PAHs 9ACA and Perylene can be utilized as biosensors for bacterial identification.

Unravelling the genetic and functional diversity of dominant bacterial communities involved in manure co-composting bioremediation of complex crude oil waste sludge

The present study aimed to characterize the bacterial community and functional diversity in co-composting microcosms of crude oil waste sludge amended with different animal manures, and to evaluate the scope for biostimulation based in situ bioremediation. Gas chromatography–mass spectrometry (GC–MS) analyses revealed enhanced attenuation (>90%) of the total polyaromatic hydrocarbons (PAHs); the manure amendments significantly enhancing (up to 30%) the degradation of high molecular weight (HMW) PAHs. Microbial community analysis showed the dominance (>99% of total sequences) of sequences affiliated to phyla Proteobacteria, Firmicutes, Actinobacteria and Bacteroidetes. The core genera enriched were related to hydrocarbon metabolism (Pseudomonas, Delftia, Methylobacterium, Dietzia, Bacillus, Propionibacterium, Bradyrhizobium, Streptomyces, Achromobacter, Microbacterium and Sphingomonas). However, manure-treated samples exhibited high number and heterogeneity of unique operational taxonomic units (OTUs) with enrichment of additional hydrocarbon-degrading bacterial taxa (Proteiniphilum, unclassified Micrococcales, unclassified Lachnospiraceae, Sphingobium and Stenotrophomonas). Thirty-three culturable hydrocarbon-degrading microbes were isolated from the co-composting microcosms and mainly classified into Burkholderia, Sanguibacter, Pseudomonas, Bacillus, Rhodococcus, Lysinibacillus, Microbacterium, Brevibacterium, Geobacillus, Micrococcus, Arthrobacter, Cellulimicrobacterium, Streptomyces Dietzia,etc,. that was additionally affirmed with the presence of catechol 2,3-dioxygenase gene. Finally, enhanced in situ degradation of total (49%), LMW (>75%) and HMW PAHs (>35%) was achieved with an enriched bacterial consortium of these microbes. Overall, these findings suggests that co-composting treatment of crude oil sludge with animal manures selects for intrinsically diverse bacterial community, that could be a driving force behind accelerated bioremediation, and can be exploited for engineered remediation processes.
