Near-Infrared Light and pH-Responsive Polypyrrole@Polyacrylic acid/Fluorescent Mesoporous Silica Nanoparticles for Imaging and Chemo-Photothermal Cancer Therapy.

We have rationally designed a new theranostic agent by coating near-infrared (NIR) light-absorbing polypyrrole (PPY) with poly(acrylic acid) (PAA), in which PAA acts as a nanoreactor and template, followed by growing small fluorescent silica nanoparticles (fSiO2 NPs) inside the PAA networks, resulting in the formation of polypyrrole@polyacrylic acid/fluorescent mesoporous silica (PPY@PAA/fmSiO2 ) core-shell NPs. Meanwhile, DOX-loaded PPY@PAA/fmSiO2 NPs as pH and NIR dual-sensitive drug delivery vehicles were employed for fluorescence imaging and chemo-photothermal synergetic therapy in vitro and in vivo. The results demonstrate that the PPY@PAA/fmSiO2 NPs show high in vivo tumor uptake by the enhanced permeability and retention (EPR) effect after intravenous injection as revealed by in vivo fluorescence imaging, which is very helpful for visualizing the location of the tumor. Moreover, the obtained NPs inhibit tumor growth (95.6 % of tumors were eliminated) because of the combination of chemo-photothermal therapy, which offers a synergistically improved therapeutic outcome compared with the use of either therapy alone. Therefore, the present study provides new insights into developing NIR and pH-stimuli responsive PPY-based multifunctional platform for cancer theranostics.

Dendrimer-entrapped gold nanoparticles modified with RGD peptide and alpha-tocopheryl succinate enable targeted theranostics of cancer cells

We report here the synthesis of multifunctional dendrimer-entrapped gold nanoparticles (Au DENPs) modified with alpha-tocopheryl succinate (α-TOS) and arginine–glycine–aspartic acid (RGD) peptide for targeted chemotherapy and computed tomography (CT) imaging of cancer cells. In this work, generation 5 poly(amidoamine) dendrimers pre-conjugated with fluorescein isothiocyanate (FI), RGD peptide via a polyethylene glycol (PEG) spacer, and PEG-linked α-TOS were used as templates to synthesize AuNPs. Followed by acetylation of the remaining dendrimer terminal amines, multifunctional Au DENPs with an Au core size of 4.0nm were generated. The formed multifunctional Au DENPs were characterized via different techniques. We show that the multifunctional Au DENPs are stable at different pH (5–8) and temperature (4–50°C) conditions and display enhanced efficacy in the generation of reactive oxygen species, which is associated with their increased ability to induce apoptosis. Thanks to the role played by RGD-mediated targeting, the multifunctional Au DENPs are able to target cancer cells overexpressing αvβ3 integrin and specifically inhibit the growth of the cancer cells. Likewise, the existence of AuNPs enabled the multifunctional Au DENPs to have a better X-ray attenuation property than clinically used iodinated CT contrast agents (e.g., Omnipaque) and the use of them as a nanoprobe for targeted CT imaging of cancer cells in vitro. The formed multifunctional Au DENPs may hold great promise to be used as a theranostic platform for cancer theranostics.

Near-infrared light-responsive nanomaterials for cancer theranostics.

Early diagnosis and effective cancer therapy are required, to properly treat cancer, which causes more than 8.2 million deaths in a year worldwide. Among various cancer treatments, nanoparticle-based cancer therapies and molecular imaging techniques have been widely exploited over the past decades to overcome current drawbacks of existing cancer treatments. In particular, gold nanoparticles (AuNPs), carbon nanotubes (CNTs), graphene oxide (GO), and upconversion nanocrystals (UNCs) have attracted tremendous attention from researchers due to their near-infrared (NIR) light-responsive behaviors. These nanomaterials are considered new multifunctional platforms for cancer theranostics. They would enable on-demand control of drug release or molecular imaging in response to a remote trigger by NIR light exposure. This approach allows the patient or physician to adjust therapy precisely to a target site, thus greatly improving the efficacy of cancer treatments, while reducing undesirable side effects. In this review, we have summarized the advantages of NIR light-responsive nanomaterials for in vivo cancer treatments, which includes NIR triggered photothermal therapy (PTT) and photodynamic therapy (PDT). Furthermore, recent developments, perspectives, and new challenges of NIR light-responsive nanomaterials are discussed for cancer theranostic applications.

Silver nanoparticle gated, mesoporous silica coated gold nanorods (AuNR@MS@AgNPs): low premature release and multifunctional cancer theranostic platform.

Multifunctional nanoparticles integrated with an imaging module and therapeutic drugs are promising candidates for future cancer diagnosis and therapy. Mesoporous silica coated gold nanorods (AuNR@MS) have emerged as a novel multifunctional cancer theranostic platform combining the large specific surface area of mesoporous silica, which guarantees a high drug payload, and the photothermal modality of AuNRs. However, premature release and side effects of exogenous stimulus still hinder the further application of AuNR@MS. To address these issues, herein, we proposed a glutathione (GSH)-responsive multifunctional AuNR@MS nanocarrier with in situ formed silver nanoparticles (AgNPs) as the capping agent. The inner AuNR core functions as a hyperthermia agent, while the outer mesoporous silica shell exhibits the potential to allow a high drug payload, thus posing itself as an effective drug carrier. With the incorporation of targeting aptamers, the constructed nanocarriers show drug release in accordance with an intracellular GSH level with maximum drug release into tumors and minimum systemic release in the blood. Meanwhile, the photothermal effect of the AuNRs upon application to near-infrared (NIR) light led to a rapid rise in the local temperature, resulting in an enhanced cell cytotoxicity. Such a versatile theranostic system as AuNR@MS@AgNPs is expected to have a wide biomedical application and may be particularly useful for cancer therapy.

A Smart Nanoassembly for Multistage Targeted Drug Delivery and Magnetic Resonance Imaging

Efficient delivery of DNA‐toxin anticancer drugs into nucleus of targeted tumor cells while simultaneously minimizing the side effects to normal tissue is a major challenge for cancer therapy. Herein, a multistage continuous targeting strategy based on magnetic mesoporous silica nanoparticles to overcome the challenge is demonstrated. At the initial‐stage, the magnetic nanoparticle is capable of efficiently accumulating in tumor tissue guided by magnet. Following by the magnetic targeting, the targeting ligand gets it right into the cancer cell by receptor‐mediated endocytosis. Accompanied by endocytosis into the lysosomes, the nanoparticle reverses its surface charge from negative to positive which leads to the separation of charge‐conversional polymer from the nanoparticle to re‐expose the nuclear‐targeting TAT peptide. Finally, TAT peptide facilitates the carriers to enter nucleus and the DNA‐toxin camptothecin can inhibit topoisomerase I to induce cell apoptosis. Furthermore, the nano‐drug delivery system can be simultaneously used as predominant contrast agents for magnetic resonance imaging. This proof of concept might open the door to a new generation of carrier materials in the fields of targeted drug transport platform for cancer theranostics.

Nanotechnologies for Noninvasive Measurement of Drug Release

A wide variety of chemotherapy and radiotherapy agents are available for treating cancer, but a critical challenge is to deliver these agents locally to cancer cells and tumors while minimizing side effects from systemic delivery. Nanomedicine uses nanoparticles with diameters in the range of ∼1-100 nm to encapsulate drugs and target them to tumors. The nanoparticle enhances local drug delivery efficiency to the tumors via entrapment in leaky tumor vasculature, molecular targeting to cells expressing cancer biomarkers, and/or magnetic targeting. In addition, the localization can be enhanced using triggered release in tumors via chemical, thermal, or optical signals. In order to optimize these nanoparticle drug delivery strategies, it is important to be able to image where the nanoparticles distribute and how rapidly they release their drug payloads. This Review aims to evaluate the current state of nanotechnology platforms for cancer theranostics (therapeutic and diagnostic particles) that are capable of noninvasive measurement of release kinetics.

Self-assembled, aptamer-tethered DNA nanotrains for targeted transport of molecular drugs in cancer theranostics

Nanotechnology has allowed the construction of various nanostructures for applications, including biomedicine. However, a simple target-specific, economical, and biocompatible drug delivery platform with high maximum tolerated doses is still in demand. Here, we report aptamer-tethered DNA nanotrains (aptNTrs) as carriers for targeted drug transport in cancer therapy. Long aptNTrs were self-assembled from only two short DNA upon initiation by modified aptamers, which worked like locomotives guiding nanotrains toward target cancer cells. Meanwhile, tandem "boxcars" served as carriers with high payload capacity of drugs that were transported to target cells and induced selective cytotoxicity. aptNTrs enhanced maximum tolerated dose in nontarget cells. Potent antitumor efficacy and reduced side effects of drugs delivered by biocompatible aptNTrs were demonstrated in a mouse xenograft tumor model. Moreover, fluorophores on nanotrains and drug fluorescence dequenching upon release allowed intracellular signaling of nanotrains and drugs. These results make aptNTrs a promising targeted drug transport platform for cancer theranostics.

Theranostic applications of nanomaterials in cancer: drug delivery, image-guided therapy, and multifunctional platforms.

Successful cancer management depends on accurate diagnostics along with specific treatment protocols. Current diagnostic techniques need to be improved to provide earlier detection capabilities, and traditional chemotherapy approaches to cancer treatment are limited by lack of specificity and systemic toxicity. This review highlights advances in nanotechnology that have allowed the development of multifunctional platforms for cancer detection, therapy, and monitoring. Nanomaterials can be used as MRI, optical imaging, and photoacoustic imaging contrast agents. When used as drug carriers, nanoformulations can increase tumor exposure to therapeutic agents and result in improved treatment effects by prolonging circulation times, protecting entrapped drugs from degradation, and enhancing tumor uptake through the enhanced permeability and retention effect as well as receptor-mediated endocytosis. Multiple therapeutic agents such as chemotherapy, antiangiogenic, or gene therapy agents can be simultaneously delivered by nanocarriers to tumor sites to enhance the effectiveness of therapy. Additionally, imaging and therapy agents can be co-delivered to provide seamless integration of diagnostics, therapy, and follow-up, and different therapeutic modalities such as chemotherapy and hyperthermia can be co-administered to take advantage of synergistic effects. Liposomes, metallic nanoparticles, polymeric nanoparticles, dendrimers, carbon nanotubes, and quantum dots are examples of nanoformulations that can be used as multifunctional platforms for cancer theranostics. Nanomedicine approaches in cancer have great potential for clinically translatable advances that can positively impact the overall diagnostic and therapeutic process and result in enhanced quality of life for cancer patients. However, a concerted scientific effort is still necessary to fully explore long-term risks, effects, and precautions for safe human use.

Multifunctional Magnetic Quantum Dots for Cancer Theranostics

The development of an innovative platform for cancer theranostics that will be capable of noninvasive imaging and treatment of cancerous tumors using biocompatible and multifunctional Fe3O4-ZnO core-shell magnetic quantum dots (M-QDs) is being explored. This multi-functional approach will facilitate deep tumor targeting using a combination of a specific cancer marker and an external magnetic field will simultaneously provide therapy that may evolve as a new paradigm in cancer theranostics.

Mesoporous silica nanoparticles functionalized with an oxygen-sensing probe for cell photodynamic therapy: potential cancer theranostics

Functionalization of mesoporous silica nanoparticles (MSNs) with Pd-porphyrins for cancer cell photodynamic therapy is reported. The composite platform, MSN–PdTPP, expands the role of Pd-porphyrins from their routine use as phosphorescence probes for oxygen sensing/imaging (diagnostics) to that of novel nano-photosensitizers for cancer cell phototherapy (therapeutics). The utility of MSN–PdTPP in the phototherapeutic treatment of MDA-MB-231 breast cancer cells is also evaluated, suggesting it is a promising cancer theranostic platform.
