Supplementation of vitamin E as an addition to a commercial renal diet does not prolong survival of cats with chronic kidney disease

BackgroundThe aim of this double-blind, placebo-controlled study was to investigate the effect of vitamin E supplementation as an addition to a commercial renal diet on survival time of cats with different stages of chronic kidney disease (CKD). In addition, we were interested whether vitamin E supplementation affects selected oxidative stress and clinical parameters. Thirty-four cats with CKD and 38 healthy cats were included in the study. Cats with CKD were classified according to the IRIS Guidelines; seven in IRIS stage 1, 15 in IRIS stage 2, five in IRIS stage 3 and seven in IRIS stage 4. Cats with CKD were treated according to IRIS Guidelines. Cats with CKD were randomly assigned to receive vitamin E (100 IU/cat/day) or placebo (mineral oil) for 24 weeks in addition to standard therapy. Plasma malondialdehyde (MDA) and protein carbonyl (PC) concentrations, DNA damage of peripheral lymphocytes and plasma vitamin E concentrations were measured at baseline and four, eight, 16 and 24 weeks thereafter. Routine laboratory analyses and assessment of clinical signs were performed at each visit.ResultsVitamin E supplementation had no effect on the survival time and did not reduce the severity of clinical signs. Before vitamin E supplementation, no significant differences in vitamin E, MDA and PC concentrations were found between healthy and CKD cats. However, plasma MDA concentration was statistically significantly higher (p = 0.043) in cats with early CKD (IRIS stages 1 and 2) than in cats with advanced CKD (IRIS stages 3 and 4). Additionally, DNA damage was statistically significantly higher in healthy cats (p ≤ 0.001) than in CKD cats. Plasma vitamin E concentrations increased statistically significantly in the vitamin E group compared to the placebo group four (p = 0.013) and eight (p = 0.017) weeks after the start of vitamin E supplementation. During the study and after 24 weeks of vitamin E supplementation, plasma MDA and PC concentrations and DNA damage remained similar to pre-supplementation levels in both the placebo and vitamin E groups.ConclusionsVitamin E supplementation as an addition to standard therapy does not prolong survival in feline CKD.

Efficacy of Zingiber Officinale (ZO) in The Treatment of Lead Acetate-induced Hepatopathy in Rabbits: An Ultrastructural Assessment of Sub-acute Trial.

Lead acetate (Pb) is a hazardous heavy metal that is well-known to impair the functions of bodily systems and organs. This present study identifies the histopathological alterations that Pb toxicity causes in the liver of rabbits as well as the efficacy of ginger (Zingiber Officinale, ZO) in mitigating its negative effects. As ZO is a rich source of several antioxidants, this present study examines its ability to decrease Pb toxicity in test animals. A total of 30 rabbits were randomly divided into three groups (n = 10). The Control group was only fed distilled water, the Negative group was only fed a 2% Pb solution, while the Treated group was fed a 2% Pb solution as well as 100 mg of ZO extract per kilogram of body weight daily. Lead acetate (Pb) was found to significantly alter the histopathology of rabbit livers by increasing inflammation, fibrosis, vacuolation, and degeneration; significantly decreasing plasma superoxide dismutase (SOD) and catalase (CAT) enzymatic activities; and increasing plasma malondialdehyde (MDA) concentrations. Furthermore, ZO extract not only significantly decreases the negative hepatic impacts of Pb but also prevents Pb-induced hepatopathy.

Effects of fermented and unfermented aging corn on ruminal fermentation, bacterial communities, lactation performance and plasma metabolites in Holstein cows

Aging reduces the nutritional value of corn grain, which may be improved by fermentation prior to use. This study investigated the effects of replacing conventional corn (CC) with aging corn (AC) and fermented aging corn (FAC) in the diets of lactating Holstein cows. Six healthy third-parity Holstein cows were used in a replicated 3 × 3 Latin square experiment with 21-day periods. Cows were fed twice and milked twice daily. The cows were randomly divided into three treatment groups: (1) CC diet: a diet with 23.6% starch in diet DM containing 24.0% conventional corn; (2) AC diet: a diet with 23.5% starch in diet DM containing 24.0% aging corn replacing conventional corn; and (3) FAC diet: a diet with 23.2% starch in diet DM containing 24.6% fermented aging corn replacing conventional corn. The diets were formulated to be isonitrogenous and contained identical roughage. The FAC treatment increased the starch digestibility compared with AC. Feeding FAC increased the concentrations of total volatile fatty acid compared with CC and AC and decreased the molar proportion of acetate compared with AC. At the family level, the relative abundance of Prevotellaceae was higher on FAC than CC. The relative abundance of Succinivibrionaceae was lower on FAC than on CC and AC. Besides, at the genera level, the relative abundance of Succinivibrionaceae_UCG_002 in the rumen was higher on AC than CC and FAC. The relative abundance of Prevotella and Ruminococcus was higher on FAC than CC and AC. The relative abundance of Succinivibrionaceae_UCG_001 was lower on FAC than CC. The Simpson index was lower on FAC than CC and AC. The FAC treatment increased the milk yield (34.0, 33.7, and 35.2 kg/d for CC, AC, and FAC group, respectively) and protein yield, and thus, energy-corrected milk production was increased, and at the same time, decreased the somatic cell score compared with CC and AC. The AC treatment increased the malondialdehyde concentration in plasma compared with CC and FAC. The concentrations in plasma of triglyceride and malondialdehyde were lower on FAC than AC. The immunoglobulin G concentration in plasma was higher on FAC than CC and AC. Overall, feeding AC resulted in decreased plasma antioxidant capacity compared with CC, whereas feeding FAC altered the relative abundance of bacteria in the rumen and improved starch digestibility, ruminal bacterial diversity, lactation performance, plasma antioxidant capacity and immune competence compared with AC in dairy cows.

Levels of melatonin and some indicators of angiogenesis, antioxidant system and lipid peroxidation in blood plasma in women with uterine leiomyoma

Uterine leiomyoma is one of the most common gynaecological diseases, which often leads to loss of fertility. It is known that in this pathology, damage to the tissues of the uterus is accompanied by oxidative stress, and the mechanisms of its compensation play a decisive role in the process of myometrial regeneration, especially when performing organ-preserving operations. The aim of the study was to determine the levels of melatonin, reproductive hormones and state of angiogenesis, antioxidant system and lipid peroxidation in women of reproductive age who were diagnosed with uterine leiomyoma. Material and methods. 60 women of reproductive age with uterine leiomyoma (study group) were examined. The control group consisted of 20 healthy women of the same age group. Melatonin levels in women's blood were determined once, on an empty stomach, at 8 a.m.; the concentrations of estradiol and progesterone in the follicular phase of the menstrual cycle were also measured. The state of the angiogenesis system was studied by examining the levels of vascular endothelial growth factor (VEGF) and the content of the final metabolites of nitric oxide NO in blood plasma. The activity of the antioxidant system was assessed by blood plasma concentrations of reduced glutathione, glutathione peroxidase, and glutathione transferase. Indicators of lipid peroxidation were investigated by the content of malondialdehyde (MDA) in blood plasma and erythrocytes. Results. It was found that in the patients of the study group with leiomyoma, there was a significantly lower level of melatonin in blood plasma (111.1 ± 18.5 ng/mL) compared to the control group (153.5 ± 8.5 ng/mL), while the concentration of estradiol was almost three times higher (107.4 ± 25.3 pg/mL) compared to the control group (36.2 ± 3.2 pg/mL), and the concentration of progesterone was 1.9 times higher (2.1 ± 0.4 ng/mL compared to 1.1 ± 0.5 ng/mL in the control group). The level of VEGF in blood plasma in women with uterine leiomyoma was also higher (90.4 ± 23.6 pg/mL) compared to the control group (35.1 ± 8.3 pg/mL), as well as the concentration of final metabolites of nitric oxide, which reached 25.3 ± 5.9 pg/mL compared to 9.9 ± 3.9 pg/mL in the control group. The reduced glutathione level in the blood plasma of women with uterine leiomyoma was significantly lower (0.77 ± 0.13 µmol/L) compared to healthy women (1.02 ± 0.14 µmol/L) in the control group, while the concentrations of glutathione-S-transferase and glutathione peroxidase enzymes were higher (161.3 ± 22.3 ng/mL and 235.7 ± 35.9 ng/mL, respectively), whereas in the control group these indicators were 118.9 ± 18.0 ng/mL and 105.3 ± 41.2 ng/mL, respectively. The MDA content in women of the study group was higher, measuring 5.2 ± 0.8 nmol/L in plasma and 10.8 ± 1.1 nmol/L in erythrocytes compared to 2.3 ± 0.8 nmol/L and 5.3 ± 0.8 nmol/L in the control group, respectively. Conclusions. The levels of melatonin and reduced glutathione in the blood plasma of women with uterine leiomyoma were significantly lower, while the concentrations of estradiol, progesterone, glutathione-S-transferase, glutathione peroxidase, VEGF, final metabolites of nitric oxide, as well as the average MDA content in plasma and erythrocytes, were significantly higher compared to healthy women. In the study group, the relationship between the level of progesterone in blood plasma and the content of malondialdehyde in erythrocytes was described using a linear regression equation, which, as we suggest, indicates an activating effect of progesterone on oxidative stress mechanisms in the myometrium. In our opinion, oxidative stress in uterine leiomyoma occurs due to inadequate activity of the antioxidant system, an integral part of which is melatonin, leading to enhanced angiogenesis against the background of estrogen-induced myometrial proliferation.

FRESH AND CRYOPRESERVED SEMEN CHARACTERISTICS OF BUFFALO BULLS AS INFLUENCED BY MELATONIN IMPLANTATION

This study explored the effect of melatonin implantation on the fresh and cryopreserved semen quality of Iraqi buffalo bulls (Bubalus bubalis). Nine adult bulls were used and divided equally into three groups. The first group was left without treatment and regarded as control (T1), whereas the second (T2) and third (T3) groups were subcutaneously implanted with 72 and 90 mg of melatonin respectively on the left ear base and repeated one month post the first implantation. Semen was collected for 12 weeks and evaluated weekly for fresh, cooling, and post-cryopreserved (PC) protocols. The T3 group exhibited a higher (P≤0.05) sperm cell individual motility percentage, whereas, the T1 and T2 groups recorded better (P≤0.05) sperm acrosome integrity percentage for fresh semen. A lesser (P≤0.05) total sperm abnormality (TSA) percentage in the T2 and T3 groups and a greater (P≤0.05) sperm acrosome integrity (SAI) percentage in the T3 group were noticed during the cooling preservation. Moreover, the T3 group exhibited a lesser (P≤0.05) TSA percentage whereas, the T2 and T3 groups revealed a leaser (P≤0.05) SAI percentage during the PC period. Concomitantly, lower (P≤0.05) PC malondialdehyde concentration in seminal plasma was observed in the T2 and T3 groups than control group. In conclusion, the melatonin implantation (72 and 90 mg) ameliorated some fresh and PC semen attributes of the Iraqi buffalo bulls.

Early-age thermal manipulation and supplemental antioxidants on physiological, biochemical and productive performance of broiler chickens in hot-tropical environments

Heat stress has been ranked as a critical environmental issue confronting chicken farmers worldwide because of its detrimental effect on the growth, performance and health of the birds. This study evaluated the effects of early-age thermal manipulation (EATC) and supplemental antioxidants on the physiological responses of broilers in a hot tropical environment. A total of 300 day-old Ross broiler chicks were allocated to five thermal and dietary treatments, having 5 replicates of twelve birds each. The treatments were: chicks reared using the conventional method (CC), chicks exposed to early thermal manipulation with a temperature of 38 °C at day 5 with no antioxidant supplementation (TC), TC plus vitamin E at 250 mg/kg of feed (TV), TC plus selenium at 0.5 mg/kg of feed (TS) and the combination of TS and TV(TVS). The experiment was laid out in a Completely Randomized Design and data collected were analyzed using SAS (2008). The results showed that TVS broilers had significantly higher (P < 0.05) body weights at the finisher phase than the other treatment groups. The feed conversion ratio of TVS broilers was comparable to the TV group but lower (P < 0.05) than the other treatments. Reduced levels (P < 0.05) of heterophil, lymphocytes and hetrophil and lymphocyte ratio were recorded in the TVS compared to TV, TS and TC broilers. On day 42, the rectal temperature was significantly higher in CC than those in other treatment groups, which were comparable. TVS birds had higher (P < 0.05) weights of spleen, liver and lower abdominal fat than other treatments. The lowest concentration of plasma malondialdehyde and the highest activity of superoxide dismutase and glutathione peroxidase were recorded in TV and TVS birds. The study concluded that the growth performance and oxidative status in broilers were improved by the combination of EATC with supplemental Se and vitamin E (TVS).

Effects of Dietary Supplementation of Stimbiotics to Sows on Lactation Performance, Immune Function, and Anti-Inflammatory and Antioxidant Capacities during Late Gestation and Lactation.

Stimbiotic supplementation may provide an innovative feed additive solution to accelerate the proliferation of beneficial fiber-degrading bacteria in the distal intestine and the utilization of dietary fiber. Optimal utilization of dietary fiber has multiple benefits for gut health and nutrient utilization. This study was conducted to evaluate the late gestation and lactation performance, the plasma, colostrum, and milk immunoglobulin (IgA, IgG, and IgM) concentrations, and the anti-inflammatory and antioxidant biomarkers in plasma of sows fed with or without a stimbiotic during the late gestation and lactation phase. A total of 40 sows were allocated to two treatment groups: control (CT) with no supplementation or 100 mg/kg stimbiotic (VP), with 20 sows per treatment. Sows were fed the treatment diets from d 85 of gestation to d 28 of lactation. In the results, the average daily weight gain of piglets during lactation was greater from sows fed in the VP group compared to that in the CT group (p < 0.05). The plasma concentrations of IgM at farrowing and IgG at weaning of the sows fed the diet with the stimbiotic supplementation were much higher than those in the CT sows (p < 0.05), respectively. In addition, the dietary stimbiotic increased the concentrations of IgM in the colostrum and of IgA and IgM in the milk at d 14 of lactation (p < 0.05). Plasma concentrations of malondialdehyde (MDA) on d 0 and d 28 of lactation tended to be lower in sows fed the VP diets compared with those of the sows fed the CT diets. Thus, our study indicated that stimbiotic supplementation could improve the daily weight gain of piglets and the immune function of sows in lactation.

AMELIORATING POST-THAWED SEMEN OF BUFFALO BULLS USING A MILK-BASED EXTENDER SUPPLEMENTED WITH RESVERATROL

The objective of the current study was to investigate the influence of adding resveratrol (Res) to a milk-based extender on some post-thawed semen quality of Iraqi buffalo bulls for different preservation periods (cooling at 5˚C, 48 h, 1, and 2 months post-cryopreservation; PC). Four bulls, 3.5 to 4 years old, were used from 11th November 2020 to 1st June 2021. Semen samples were collected, pooled, and divided into four groups using different Res concentrations (0, 100, 150, and 200 μM) added to the milk extender. The Res-150 μM group exhibited higher (P≤0.01) plasma membrane, acrosome integrity percentages, and total antioxidants concentration (TAC) compared to the other groups. Sperm abnormalities and DNA damage percentages, as well as malondialdehyde (MDA) concentration, were lesser (P≤0.01) in the Res-150 μM group. Finally, Res-150 μM added to a milk-based extender increased the percentage of post-thawed live sperm and TAC in seminal plasma and minimized abnormal sperm percentage, DNA damage percentage, and MDA concentration in the seminal plasma of Iraqi buffalo bulls.

PSVI-2 Dietary Supplementation of Phyllanthus Urinaria Ameliorates Fatty Liver Hemorrhagic Syndrome in Hens

Abstract The purpose of the present study was to investigate the effect of Phyllanthus urinaria supplement on fatty liver hemorrhagic syndrome (FLHS) in hens. Forty 7-week-old ISA hens were randomly divided into four groups (n = 10), including control diet group (CON), high cholesterol and low choline diet group (CLC), and CLC with 1% P. urinaria powder group (PU). CLC diet and the supplementation of P. urinaria for six weeks exhibited no impact on feed intake and body weight (P &amp;gt; 0.05). The triglyceride (TG) and the total cholesterol (TC) in plasma was increased at week 6 (TG: CON = 18.86 ± 0.97 mg/dL and CLC = 66.45 ± 7.17 mg/dL; TC: CON = 130.41 ± 5.87 mg/dL and CLC = 308.11 ± 27.97 mg/dL); nevertheless, P. urinaria decreased the triglyceride in plasma (PU = 41.19 ± 4.88 mg/dl, P &amp;lt; 0.05), but not total cholesterol (PU = 259.11 ± 20.88 mg/dL, P &amp;gt; 0.05). The malondialdehyde (MDA) concentration in plasma was decreased with 1% P. urinaria powder group (CLC = 64.81 ± 3.71 mg/dL and PU = 49.13 ± 4.20 mg/dL, P &amp;lt; 0.05). Under liver macroscopic analysis, PU demonstrated the least percentage of severe in hepatic appearance (CLC = 22% and PU = 0%). Similarly, the liver microscopic examination (H&amp;E) suggested the PU supplementation reduced steatosis score (CLC = 2.89 ± 0.11 and PU = 2.11 ± 0.16) and blooning (CLC = 1.63 ± 0.22 and PU = 0.88 ± 0.31) in the hepatocyte. To sum up, this study revealed the hepatoprotective effect of Phyllanthus urinaria and provided a potential strategy for preventing FLHS to improve the welfare in hens.

Analyzing and Examining Smoking Effects and Oxidative Stress Markers in Seminal Plasma in Iraqi Infertility Men

The oxidative stress (OS) and sperm DNA fragmentation have been associated with sub-infertility in males and infertility due to a lack of means of protection and defense (antioxidants) for spermatozoa and an increase in reactive oxygen spices (ROS) related to wrong lifestyle (cigarettes and hookah) or what also known as Shisha. The study included (90) men between the ages of (23-40) years, (60), infertile men who smoked cigarettes and hookahs. They were divided into two groups, the first oligozoospermia and the second asthenozoospermia, according to the guidelines of the World Health Organization for seminal fluid analysis (SFA), 5 th edition, WHO(2010). Each group included (30) smoking infertile males, with a control group included (30) non-smoking fertile males. The study aimed to estimate the concentration level of Ascorbic Acid (AA) in the seminal plasma of two groups of infertile men smoking cigarettes and hookahs using enzyme-linked immunosorbent assay (ELISA) method, with calculating the concentration of Malondialdehyde (MDA) as well as estimating the percentage of sperm DNA fragmentation (SDF) using sperm chromatin dispersion (SCD), Halo view. Compared with healthy fertile men of non-smokers of cigarettes and hookahs as one of the biomarkers of oxidative stress in infertile men in Nineveh Governorate-Iraq. The results indicated that there were significant differences at the probability level (p&lt;0.01) for the studied parameters (AA, MDA, SDF) compared to the control group (fertile men). The results showed an increase in the percentage of sperm DNA fragmentation with increasing the concentration of MDA in seminal plasma and decreasing the concentration of AA. Cigarette and hookah smoking increased the generation of ROS with a decrease in the water-soluble non-enzymatic antioxidant Ascorbic acid (AA) and thus increased the oxidative processes of the plasma membranes of spermatozoa and the occurrence of oxidative stress and impaired fertility and caused infertility. Lipid metabolism and oxidative stress in patients with Alzheimer's disease and amnestic mild cognitive impairment.

Lipid metabolism and oxidative stress are key mechanisms in Alzheimer's disease (AD). The link between plasma lipid metabolites and oxidative stress in AD patients is poorly understood. This study was to identify markers that distinguish AD and amnestic mild cognitive impairment (aMCI) from NC, and to reveal potential links between lipid metabolites and oxidative stress. We performed non-targeted lipid metabolism analysis of plasma from patients with AD, aMCI, and NC using LC-MS/MS. The plasma malondialdehyde (MDA), glutathione peroxidase (GSH-Px), and superoxide dismutase (SOD) levels were assessed. We found significant differences in lipid metabolism between patients with AD and aMCI compared to those in NC. AD severity is associated with lipid metabolites, especially TG (18:0_16:0_18:0) + NH4, TG (18:0_16:0_16:0) + NH4, LPC(16:1e)-CH3, and PE (20:0_20:4)-H. SPH (d16:0) + H, SPH (d18:1) + H, and SPH (d18:0) + H were high-performance markers to distinguish AD and aMCI from NC. The AUC of three SPHs combined to predict AD was 0.990, with specificity and sensitivity as 0.949 and 1, respectively; the AUC of three SPHs combined to predict aMCI was 0.934, with specificity and sensitivity as 0.900, 0.981, respectively. Plasma MDA concentrations were higher in the AD group than in the NC group (p = 0.003), whereas plasma SOD levels were lower in the AD (p < 0.001) and aMCI (p = 0.045) groups than in NC, and GSH-Px activity were higher in the AD group than in the aMCI group (p = 0.007). In addition, lipid metabolites and oxidative stress are widely associated. In conclusion, this study distinguished serum lipid metabolism in AD, aMCI, and NC subjects, highlighting that the three SPHs can distinguish AD and aMCI from NC. Additionally, AD patients showed elevated oxidative stress, and there are complex interactions between lipid metabolites and oxidative stress.

Effect of Electroejaculation Protocols on Semen Quality and Concentrations of Testosterone, Cortisol, Malondialdehyde, and Creatine Kinase in Captive Bengal Tigers

The Bengal tiger (Panthera tigris tigris) is critically endangered, so assisted reproductive technologies, including artificial insemination, are important conservation tools. For wild and domestic felids, electroejaculation (EE) is the most common semen collection method, with protocols optimized to obtain sufficient amounts of viable sperm for artificial insemination. However, less attention has been paid to ensuring animal wellbeing during the process. This study examined the effects of three EE protocols (Low, 2–5 volts; Medium, 3–6 volts; High, 4–7 volts) on semen quality, testicular size, serum testosterone, creatine kinase (CK), and malondialdehyde (MDA) concentrations, and serum cortisol as a proxy for stress. Blood samples were collected before, during, and after each EE series. Seminal plasma pH, and sperm motility, viability, and morphology were evaluated after each procedure. Seminal plasma and sperm pellet MDA concentrations were also determined. Primary sperm abnormalities and seminal plasma MDA were higher in the Low compared to Medium and High voltage groups (p &lt; 0.05). Serum CK in the High voltage group increased during the EE series (p &lt; 0.05), suggesting the potential for muscle damage. However, no significant changes were observed for serum cortisol, testosterone, or MDA concentrations. Results suggest the Medium voltage protocol produced good quality samples at lower voltages than the High protocol with no negative effect on muscle function, which might be better for animal welfare.

Hydrolysed yeast from Kluyveromyces fragilis improves plasma antioxidant efficiency and immunoglobulin concentration, and faecal microbiota of weaned piglets

The aim of this study was to evaluate the effect of hydrolysed yeast from Kluyveromyces fragilis (HK) on growth performance, diarrhoea incidence, plasma antioxidant efficiency and immune status, and faecal microbiota of weaned piglets. A total of 100 weaned piglets with BW (7.03 ± 0.14 kg) and age (25 ± 1 days) were randomly allotted to 4 groups with 5 replicates in a 21-day experiment. Piglets were fed with basal diet (NC), NC + 2 g/kg zinc oxide (PC), NC + 7.5 g/kg HK (HK1), or NC + 10 g/kg HK (HK2). Blood and faecal samples were collected on day 21. Significant differences were pointed out in the PC and HK2 piglets compared to the NC group as to the diarrhoea incidence from day 0 to 21 (p < 0.001; p = 0.032), the activity of plasma superoxide dismutase (p = 0.019; p = 0.003) and the concentration of plasma malondialdehyde (p = 0.042; p = 0.010). Moreover, significant differences were pointed out in the HK2 piglets compared to the NC group as to the content of plasma immunoglobulin A (p = 0.005), the Ace index and Chao1 index (p = 0.023; p = 0.018,) and the relative abundance of Campylobacterota and Escherichia-Shigella (p = 0.023; p = 0.032). In conclusion, dietary HK at 10 g/kg alleviated diarrhoea incidence of piglets that might be attribute to the improved plasma antioxidant efficiency and immune status and the regulated faecal microbial community, and could be alternative to the high dose ZnO. HIGHLIGHTS Dietary HK improved plasma antioxidant and immune status and reduced the diarrhoea incidence in weaned piglets. The dosage of 10 g/kg HK showed better effect than that of 7.5 g/kg. The supplementation of 10 g/kg HK to post-weaning diet may be an alternative to the high-dose ZnO.

8-Hydroxy-2'-deoxyguanosine and malondialdehyde in plasma and their association with disease severity in 20 cats with chronic kidney disease.

Oxidative stress is associated with the development and progression of chronic kidney disease (CKD) in humans. The aim of this study was to evaluate the concentrations of oxidative stress markers, 8-hydroxy-2'-deoxyguanosine (8-OHdG) and malondialdehyde (MDA), in the plasma and urine of cats with different stages of CKD. Plasma and urine samples were collected from cats with CKD that were referred to the Veterinary Medical Center of the University of Tokyo between April 2019 and October 2022. Plasma and urine samples were collected from healthy cats (n = 6 at most), cats with stage 2 CKD (n = 8) and stage 3-4 CKD (n = 12), and cats with idiopathic cystitis (disease control, n = 5). Plasma and urine concentrations of 8-OHdG and MDA were measured using ELISA and thiobarbituric acid reactive substances assay kits, respectively. The median plasma 8-OHdG concentrations were 0.156 ng/ml (<0.125-0.210 ng/ml) in the healthy group, <0.125 ng/ml (range <0.125 ng/ml) in the idiopathic cystitis group, 0.246 ng/ml (range 0.170-0.403 ng/ml) in cats with stage 2 CKD and 0.433 ng/ml (range 0.209-1.052 ng/ml) in cats with stage 3-4 CKD. Concentrations in stage 3-4 CKD were significantly higher than those in the healthy and disease control groups. Plasma MDA concentrations were low in the healthy and disease control groups and significantly higher in cats with stage 3-4 CKD. In every cat with CKD, plasma 8-OHdG and MDA concentrations were positively correlated with plasma creatinine concentrations (8-OHdG, rs = 0.68; MDA, rs = 0.67). Urinary 8-OHdG/urinary creatinine (u-CRE) and urinary MDA/u-CRE levels did not differ significantly between the groups; however, it was difficult to evaluate them because of the small sample size. This report shows that plasma 8-OHdG and MDA concentrations increase with the severity of feline CKD. These markers may be useful for assessing oxidative stress in cats with CKD.

Changes in the cortisol level under the influence of physical exertion in patients with hypertension and in practically healthy individuals

Background. Today, there are difficulties with the prevention and diagnosis of early stages of hypertension, especially in active and able-bodied young people. Among the factors that potentiate an increase in blood pressure, one of the components is chronic stress, which a person often faces in modern society. The purpose of study is to evaluate and compare blood cortisol and plasma malondialdehyde content in healthy individuals and in patients with stage II hypertension before and after dosed exercises. Materials and methods. Thirty patients with stage II hypertension and 10 practically healthy persons were examined. Subjects performed two-stage physical exercises on a bicycle ergometer with an intensity corresponding to 50 and 75% of the appropriate maximum oxygen consumption of the body. The duration of the load was equal to 5 minutes at each stage with a three-minute rest between them. Blood was taken from the ulnar vein before cycling ergometry and 5 minutes after it. Blood cortisol and plasma malondialdehyde content were determined in patients before and after exercises. Results. A significant increase in cortisol content after exercises was noted in healthy individuals against the background of some decrease in patients with hypertension. It has been suggested that since people with high blood pressure already have an elevated initial level of cortisol, their threshold of stress sensitivity to physical exertion is increased. Submaximal bicycle ergometric load does not lead to an increase in blood cortisol levels in hypertensive patients. Conclusions. The background higher level of cortisol in individuals with hypertension does not increase under the influence of submaximal dosed exercises. An increase in the level of malondialdehyde can be considered a marker of the degree of stress for assessing the power of physical exertion. In view of the above, it can be stated that the activation of the sympathoadrenal system under the influence of emotional stress leads to an increase in blood pressure. There is sufficient evidence that psycho-emotional stress is a cause of white-coat hypertension and hypertension in the workplace.

&lt;i&gt;NOS2&lt;/i&gt;, &lt;i&gt;NOS3&lt;/i&gt;, &lt;i&gt;SONE&lt;/i&gt; gene transcripts levels in peripheral blood leukocytes and their relationship with markers of endothelial dysfunction in hypertension

Objective. The aim of the study was to evaluate the level of expression of the NOS2, NOS3, SONE genes in peripheral blood leukocytes (PBL) of patients with hypertension (HTN) and to study the relationship between the level of transcripts of these genes and the content of nitric oxide metabolites and markers of endothelial dysfunction.Design and methods. The study included healthy people (25 people) and patients with HTN (stages I–II) before prescribing antihypertensive drugs (15 people) and taking cardioselective β-adrenergic receptor blockers for more than a year (metoprolol (25 mg per day) or bisoprolol (5–10 mg per day)) (20 people). The level of gene transcripts was assessed by real-time polymerase chain reaction (PCR). The level of nitric oxide metabolites was determined by the colorimetric method using the Griess reagent. The content of asymmetric dimethylarginine (ADMA), soluble forms of vascular cell adhesion molecule (sVCAM), and intercellular adhesion molecule (sICAM) in blood plasma was determined by ELISA. The content of malondialdehyde (MDA) in blood plasma was determined spectrophotometrically by color reaction with thiobarbituric acid. Statistical processing of the results was carried out using the Statgraphics Centurion XVI software package (version 16.1.11).Results. The level of nitric oxide metabolites in the blood plasma of HTN patients without antihypertensive therapy was 2,1 times higher than in healthy individuals (p = 0,001) and 1,7 times higher than in patients with HTN taking metoprolol or bisoprolol (p = 0,002). The relative content of mRNA of the NOS3 gene in PBL of individuals included in the study did not differ (p &gt; 0,05). The level of NOS2 gene transcripts in PBL of HTN patients before the prescription of antihypertensive drugs exceeded that in healthy individuals (p = 0,0009) and in HTN patients taking metoprolol or bisoprolol (p = 0,0002). The number of SONE transcripts in the PBL of HTN patients was higher than in people with normal blood pressure (p &lt; 0,00001 when comparing patients before the prescription of antihypertensive therapy and individuals from the control group; p = 0,04 when comparing patients with HTN taking antihypertensive drugs and normotensive subjects). The content of MDA, ADMA, sVCAM was higher in the plasma of HTN patients without antihypertensive therapy compared with people from the control group (p = 0,005, 0,003, 0,039, respectively) and patients taking metoprolol or bisoprolol (p = 0,0006, 0,019, 0,016, respectively). The content of nitric oxide metabolites positively correlated with NOS2, SONE, VCAM1 mRNA level in PBL, the content of MDA and ADMA in blood plasma (p &lt; 0,05). A positive correlation was found between the concentration of MDA and ADMA in plasma (p = 0,03).Conclusions. An increase in the level of nitric oxide metabolites in HTN is associated with an increase in the transcriptional activity of the NOS2 gene, a disturbance of the redox balance of the body, and the development of endothelial dysfunction. The SONE gene is probably involved in the modulation of nitric oxide levels in HTN not only as an antisense transcript that destabilizes the mRNA of the NOS3 gene in vascular endothelial cells, but also indirectly, namely, through the regulation of homeostasis of immune system cells through autophagy.

Effect of Copper and Zinc Ions on Biochemical and Molecular Characteristics of Calcium Oxalate Renal Stones: a Controlled Clinical Study.

Contradictory results are existed in the literature regarding the impact of trace elements on the pathogenesis of calcium oxalate (CaOx) stone patients. Therefore, the aim of our study was to investigate the effect of Cu and Zn on biochemical and molecular characteristics of CaOx stones. Plasma and urine concentrations of Cu and Zn in 30 CaOx stones patients and 20 controls were determined by flame atomic absorption spectrometry (FAAS). Urinary levels of citric acid and oxalate were measured by commercial spectrophotometric kits. Blood levels of glutathione reduced (GSH) and catalase (CAT) were determined as markers of antioxidant activity, while blood malondialdehyde (MDA) and urine level of nitric oxide (NO) were used to assess oxidative stress. Gene expression of MAPk pathway (ERK, P38, and JNK) were estimated. The plasma and urine levels of Cu were significantly increased in the patient group compared to those of controls, while the levels of Zn were decreased. Excessive urinary excretion of citric acid and oxalate were found among CaOx stone patients. The GSH and CAT concentration were significantly reduced in CaOx stones patients compared to healthy group. The plasma MDA and urine NO concentration were significantly increased in CaOx stones patients compared to control group. The expressions of the studied genes were significantly increased in CaOx stones patients. These findings suggest that alteration in Cu and Zn might contribute to pathogenesis of CaOx patients through oxidative stress and MAPK pathway genes (ERK, P38 and JNK).

55 Impacts of dietary Protandim Nrf2 Synergizer on antioxidant status of mature, sedentary horses

Reactive oxygen species are normal by-products of cellular metabolism but may have detrimental effects on cellular matrices when overproduced. Dietary antioxidants may neutralize excess free radicals and prevent the imbalance of oxidants to antioxidants, referred to as oxidative stress. To test the hypothesis that supplementation of an herbal extract combination (Protandim Nrf2 Synergizer, LifeVantage Corporation) would improve the body's enzymatic defense system, 40 mature, sedentary horses (32 mares, 8 geldings, mean ± SD; 15.7 ± 4.9 y, 519 ± 46 kg) were stratified by age, sex, and body weight (BW) and randomly assigned to one of 4 dietary treatment groups for 56 d: 1) 0 mg (CON); 2) 675 mg (Pro1); 3) 2,025 mg (Pro3); or 4) 4,050 mg (Pro6) Protandim Nrf2 Synergizer per day (n = 10 per group). Horses were group housed and received a basal diet of coastal bermudagrass hay ad libitum and a custom-formulated concentrate grain fed individually at 0.5% BW/d (dry matter basis) split evenly between 2 meals per d. The supplement was top-dressed on the morning grain meal. Blood samples were collected at d 0, 28, and 56 of supplementation before the morning feeding and analyzed for hydrogen peroxide (H2O2) production and concentration, superoxide dismutase(SOD) and glutathione peroxidase (GPx) activities, and malondialdehyde (MDA) concentration. Data were analyzed using linear models in SAS v9.4; sex, treatment, time, time × treatment were fixed effects and time was a repeated effect with horse(treatment) as the subject. Day 0 was included in all models as a covariate. No measure was impacted by dietary treatment. Regardless of treatment, whole blood SOD activity, plasma MDA, and serum H2O2 concentration decreased from d 0 to 28 (P ≤ 0.0001) and remained suppressed at d 56 (P ≤ 0.0007). Conversely, whole blood GPx activity increased from d 0 to 28 (P = 0.01) and decreased from d 28 to 56 (P = 0.02) to be similar to d 0 at d 56. Serum H2O2 production was greater in geldings than mares (P = 0.04) but no other variable was impacted by sex. Dietary supplementation of up to 4,050 mg/d Protandim Nrf2 Synergizer did not impact antioxidant status in mature, sedentary horses. Effects of supplementation on antioxidant status should be investigated in horses subjected to elevated oxidative insult, such as exercise.

ОЦЕНКА ИНТЕНСИВНОСТИ СВОБОДНО-РАДИКАЛЬНЫХ ПРОЦЕССОВ ПО ИЗМЕНЕНИЮ КОНЦЕНТРАЦИИ МАЛОНОВОГО ДИАЛЬДЕГИДА ПРИ РАЗЛИЧНЫХ СПОСОБАХ ПАРЕНТЕРАЛЬНОГО ВВЕДЕНИЯ ЛЮМИНОЛА

Assessment of the intensity of free-radical processes that change their activity in response to exogenous and endogenous intoxication is carried out by biochemical methods of determining the state of the antioxidant system of the body (AOS). This is a complex of methods, one indicator of which is the level of lipid peroxidation by accumulation of malondialdehyde (MDA). Revealing certain features of the mutually regulating influence of the processes of lipid oxidation and antioxidant system of the body may serve as a criterion for evaluation and prognosis of the course of the pathological process in endogenous toxicity and correction of therapeutic tactics. Peroxidation in the elderly and old age is of particular importance, since there is still no definitive opinion on the dynamics of the physiological course of lipid peroxidation in this age group, the criteria for the transformation of peroxidation reactions from normal to pathogenic. In this work we evaluated the antioxidant activity and duration of the preservation effect on the antioxidant protection of the organism of the drug "Tameron" (sodium luminol) with different methods of parenteral administration in the model of age-related lower primates. At intramuscular and intravenous injection of the drug "Tameron" in experimental animals a marked decrease in the concentration of MDA in blood plasma was observed. This suggests a decrease in the activity of peroxidation processes and activation of antioxidant protection of the body, which is especially important in age-matched animals. It should be noted the preservation of low indicators of oxidative stress indicator in the representatives of the experimental group for a long time period (about a year) and suggest a pronounced antioxidant prolonged effect of the studied preparation, accompanied by alignment of MDA concentration throughout the group to statistically similar values.

Responses of growth performance, antioxidant function, small intestinal morphology and mRNA expression of jejunal tight junction protein to dietary iron in yellow-feathered broilers

This study aimed to investigate the dose-effect of iron on growth performance, antioxidant function, intestinal morphology, and mRNA expression of jejunal tight junction protein in 1- to 21-d-old yellow-feathered broilers. A total of 720 1-d-old yellow-feathered male broilers were allocated to 9 treatments with 8 replicate cages of 10 birds per cage. The dietary treatments were consisted of a basal diet (contained 79.6 mg Fe kg–1) supplemented with 0, 20, 40, 60, 80, 160, 320, 640, and 1,280 mg Fe kg–1 in the form of FeSO4·7H2O. Compared with the birds in the control group, birds supplemented with 20 mg Fe kg–1 had higher average daily gain (ADG) (P<0.0001). Adding 640 and 1,280 mg Fe kg–1 significantly decreased ADG (P<0.0001) and average daily feed intake (ADFI) (P<0.0001) compared with supplementation of 20 mg Fe kg–1. Malondialdehyde (MDA) concentration in plasma and duodenum increased linearly (P<0.0001), but MDA concentration in liver and jejunum increased linearly (P<0.05) or quadratically (P<0.05) with increased dietary Fe concentration. The villus height (VH) in duodenum and jejunum, and the ratio of villus height to crypt depth (V/C) in duodenum decreased linearly (P˂0.05) as dietary Fe increased. As dietary Fe increased, the jejunal relative mRNA abundance of claudin-1 decreased linearly (P=0.001), but the jejunal relative mRNA abundance of zona occludens-1 (ZO-1) and occludin decreased linearly (P˂0.05) or quadratically (P˂0.05). Compared with the supplementation of 20 mg Fe kg–1, the supplementation of 640 mg Fe kg–1 or higher increased (P˂0.05) MDA concentrations in plasma, duodenum, and jejunum, decreased VH in the duodenum and jejunum, and the addition of 1,280 mg Fe kg–1 reduced (P˂0.05) the jejunal tight junction protein (claudin-1, ZO-1, occludin) mRNA abundance. In summary, 640 mg of supplemental Fe kg–1 or greater was associated with decreased growth performance, increased oxidative stress, disrupted intestinal morphology, and reduced mRNA expression of jejunal tight junction protein.
