Anti-Muellerian hormone, inhibin A, gonadotropins, and gonadotropin receptors in bull calves after partial scrotal resection, orchidectomy, and Burdizzo castration

Eight-week-old calves were either castrated by partial scrotal resection (SR) without removing the testes (n = 10), Burdizzo (BZ) clamp (n = 10), orchidectomy (OR; n = 10), or were left gonad intact as controls (CO; n = 10). Concentrations of anti-Muellerian hormone (AMH), inhibin A, luteinizing hormone (LH), and follicle-stimulating hormone (FSH) in plasma were determined from 16 to 48 weeks of age. At 18 months, testes of SR, BZ, and CO bulls were obtained and the immunolocalization of LH and FSH receptors and AMH analyzed. Concentration of AMH in plasma of CO and SR bulls decreased with increasing age (P < 0.001). A similar AMH profile in CO and SR indicates that SR did not induce a true cryptorchid state. In groups OR and BZ, AMH was undetectable. Plasma inhibin concentration was higher in groups CO and SR than BZ and OR (P < 0.001). Plasma LH and FSH concentrations decreased over time (P < 0.001) and were higher in groups BZ and OR than SR and CO (P < 0.001). In the testes, immunolabeling for AMH existed in Sertoli cells of CO and SR but not BZ bulls. FSH receptors were localized in Sertoli cells, Leydig cells, spermatocytes, and the epididymis of CO and SR animals, whereas LH receptors were restricted to Leydig cells. In BZ animals, FSH and LH receptors and AMH were absent, indicating complete testicular degeneration. In conclusion, AMH is a more reliable marker for the presence of testicular tissue in bulls than inhibin. Scrotal resection did not induce a true inguinal cryptorchid state but affected testicular responsiveness to gonadotropic stimulation.

Inhibin B and Antimüllerian Hormone as Markers of Gonadal Function after Treatment for Medulloblastoma or Posterior Fossa Ependymoma during Childhood

To evaluate the roles of hypothalamic-pituitary and spinal irradiations and chemotherapy in gonadal deficiency after treatment for medulloblastoma or posterior fossa ependymoma by measuring levels of plasma inhibin B and antimüllerian hormone (AMH). A total of 34 boys and 22 girls were classified as having normal levels of plasma follicle-stimulating hormone (FSH; <9 IU/L), or abnormal levels of FSH (>9 IU/L) and luteinizing hormone (LH; <5 or >5 IUL). Two boys had partial gonadotropin deficiency, combined with testicular deficiency in one boy. Six boys had increased levels of FSH, indicating tubular deficiency, combined with Leydig cell deficiency in 5 boys. The 7 boys with inhibin B levels <100 ng/mL included the one with combined deficiencies and the 6 with testicular deficiency. Puberty did not progress in 7 girls; 3 had gonadotropin deficiency, combined with ovarian deficiency in one, and 4 had increased FSH levels, indicating ovarian deficiency. Inhibin B and AMH levels were low in the girl with combined deficiencies, in the 4 girls with ovarian deficiency, and in 4 girls with normal clinical-biological ovarian function, including 2 who underwent ovarian transposition before irradiation. The plasma concentrations of inhibin B and AMH are useful means of detecting primary gonad deficiency in patients with no increase in their plasma gonadotropin levels because of radiation-induced gonadotropin deficiency.

Immunodetection of inhibin in the human testis and epididymis during normal development and in non-tumoural testicular lesions

Plasma concentrations of inhibin are correlated with spermatogenetic function. Inhibin is secreted mainly by the Sertoli and Leydig cells of the testis. In the human epididymis, the location and function of inhibin are contentious. Thus, the aim of the present study was to determine the location of inhibin in the human epididymis. Investigations were performed in samples with normal testicular function at different stages of development, as well as in samples in which testicular function or the testicular-epididymal connection were altered. In fetal, newborn and infant testes, Sertoli and Leydig cells stained positive for inhibin, whereas no such staining was detected in the epididymides. Inhibin was located in both the Sertoli and Leydig cells, as well as in the epididymis, in the apical pole of mainly secretory cells in the efferent ducts. This staining pattern was not correlated with the staining pattern for macrophages. The main duct of the epididymis was negative for inhibin staining. In ischaemic atrophic testes, the few tubules in which Sertoli cells were present stained positive for inhibin, whereas the epididymides stained negative. In paediatric cryptorchidism, Sertoli and Leydig cells stained positive for inhibin, whereas the epididymides were negative. In adult cryptorchidism, Sertoli and Leydig cells stained positive for inhibin, even in tubules containing Sertoli cells only. Interestingly, inhibin was absent from the efferent ducts. In three cases undergoing hormonal treatment prior to subsequent gender change, Sertoli and Leydig cells stained positive for inhibin. In contrast, the efferent ducts were negative or only faintly positive in cases of shorter hormonal treatment. In all cases studied, the presence of inhibin in the efferent ducts was associated with its production in the testis, suggesting that the epididymis is not responsible for the production of inhibin in men. The pattern of inhibin staining does not correlate with that of macrophages, suggesting that inhibin is not degraded in the human epididymis. The data suggest that, in humans, inhibin is secreted by Sertoli cells into the seminiferous tubules and then travels towards the efferent ducts, where it is reabsorbed into the bloodstream.

Peripheral concentrations of inhibin A, ovarian steroids, and gonadotropins associated with follicular development throughout the estrous cycle of the sow

We investigated changes in peripheral concentrations of inhibin A, total inhibin, steroids, and gonadotropins throughout the intact estrous cycle of the sow in relation to ovarian changes determined by daily transrectal ultrasonography. All visible follicles of 3 mm or more in diameter were classified as small (> or =3 and <6 mm) or large (> or =6 mm). Follicular recruitment was identified in two periods of the cycle: one from the late luteal to the follicular phase, characterized by an increase in the number of small follicles followed by the appearance of large follicles; and another during the early luteal phase, consisting only of increased numbers of small follicles. Plasma concentrations of inhibin A increased (P<0.05), coinciding with the two periods of follicle emergence. Estradiol (E(2)) levels increased (P<0.05) during the follicular phase, but not during the early luteal phase. An inverse relationship (P<0.01) between the patterns of inhibin and FSH concentrations was noted around the two periods of follicle emergence, but there was no relationship (P> or =0.1) between the patterns of plasma E(2) and FSH during the early luteal phase. In conclusion, measurement of plasma inhibin A levels combined with ultrasonographic examination of the ovaries revealed two periods of synchronous follicular growth during the sow's estrous cycle. The results strongly suggest that inhibin A functions as a negative feedback regulator of FSH secretion throughout the estrous cycle, whereas E(2) appears to influence FSH secretion only during the follicular phase.

Effect of administration of a single dose of testosterone oenanthate on human serum and seminal plasma inhibin concentration.

The effect of a single administration of testosterone oenanthate (250 mg, intra-muscularly) on serum and seminal plasma inhibin concentrations were studied in four adult human volunteers. A significant decrease in serum FSH and LH levels were observed by day 6 when the serum testosterone levels were high. These were followed by a sharp increase in serum and seminal plasma inhibin concentration by day 8 and day 12 respectively. The present investigations also suggest the existence of a positive relationship between the serum and seminal plasma inhibin concentrations.

Relationship between peripheral plasma inhibin and FSH concentrations in Sahiwal cows (Bos indicus) and Murrah buffaloes (Bubalus bubalis) during estrous cycle

The present investigation was undertaken to study the relationship between circulating inhibin and FSH concentrations during the estrous cycle in buffaloes and Sahiwal cattle. The pattern of inhibin concentrations was similar, with peak concentrations on Day -2 (Day 0 = day of estrus) and minimum concentrations on Days 12 and 11 in buffaloes and cattle, respectively. Circulating FSH concentrations were the highest on Day 0 and lowest on Days 8 and 13 in buffaloes and cattle, respectively. Peripheral plasma inhibin concentrations were negatively correlated to FSH concentrations in buffaloes (r = -0.27, P < 0.01) and cattle (r = -0.35, P < 0.01) indicating that inhibin is involved in negative feedback regulation of FSH in both these species.

Effects of Experimental Cryptorchidism on Sperm Motility and Testicular Endocrinology in Adult Male Rats

The effect of induced cryptorchidism on testicular function and sperm motility was investigated. Bilateral cryptorchidism was created surgically in adult male rats (treated group), and sham-operated rats were used as a control group. Five rats from each group were sacrificed on days 1, 3, 5, and 7 after surgery. The percentage of motile spermatozoa began to decrease 1 day after the operation, followed by an abrupt decline 3 and 5 days later in cryptorchid rats. Furthermore, there were significant decreases in the other sperm motility parameters 5 days after inducement of cryptorchidism. In cryptorchid rats, plasma concentrations of LH, FSH, testosterone, and inhibin B were significantly lower than in the control group 1 day after the operation. Thereafter, plasma concentrations of LH, FSH, and testosterone gradually increased in the cryptorchid rats. On the other hand, plasma concentrations of inhibin B showed a further decline from day 3 after the operation onward. Concentrations of immunoreactive (ir)-inhibin, but not testosterone, in testicular interstitial fluid were remarkably increased until 3 days after surgery in the cryptorchid rats, and declined thereafter. Testicular response to human chorionic gonadotropin (hCG) for testosterone release was decreased in the cryptorchid rats compared with the control rats, indicating that heat stress to testes resulted in a reduction of the activity of Leydig cells and Sertoli cells. These results clearly indicate that heat stress to the testes resulted in a significant reduction of sperm activity within 3 days, and this was followed by changes in testicular endocrine function.

Plasma concentrations of inhibin A in cattle with follicular cysts: relationships with turnover of follicular waves and plasma levels of gonadotropins and steroid hormones

We investigated the profiles of circulating levels of inhibin A and total inhibin in beef cows with follicular cysts in relation to the patterns of follicular development and circulating gonadotropins and steroid hormones. Turnover of follicular waves was monitored in five cows every 2 days for 70 days from 10 days after detection of estrus without ovulation. The mean interwave intervals were 19.6 ± 1.0 days ( n = 18 waves with cysts from the five cows). Circulating levels of inhibin A were approximately 170 pg/ml before emergence of follicular waves with cysts and increased ( P < 0.05) concomitantly with follicle emergence. High concentrations of inhibin A (greater than 300 pg/ml) were noted for 7 days during the growth phase of cystic follicles, but inhibin A levels decreased gradually when development of the cysts reached a plateau. This profile of inhibin A was similar to those of total inhibin and estradiol, but was inversely related to the changes in plasma FSH concentrations. LH pulse frequency and mean concentrations of LH in cows with cysts were higher than those observed in the luteal phase of normal cyclic cows. These results indicate that the capacity to secrete inhibin, as well as estradiol, is maintained in cystic follicles, the growth of which is extended by LH secretion at levels greater than those seen in the normal luteal phase. Inhibin A plays an important role in the extension of interwave intervals by suppressing recruitment of a new cohort of follicles.

Changes in circulating concentrations of inhibins A and pro-alpha C during first trimester medical termination of pregnancy.

Both inhibin A and inhibin pro-alpha C are detectable in the circulation in increasing amounts during establishment of pregnancy. However, their origins and functions remain to be elucidated. We have studied levels of inhibin A and inhibin pro-alpha C in serum samples collected at various stages during medical termination of pregnancy with consecutive use of mifepristone and misoprostol. Samples were collected from three groups of patients at different weeks of gestation (group A: 6-7 weeks, n = 6; group B: 7-8 weeks, n = 6; group C: 8-9 weeks, n = 6) at the time of administration of oral mifepristone, 48 h later just before administration of vaginal misoprostol and again soon after expulsion of the products of conception. Plasma concentrations of inhibin A and pro-alpha C were assayed using specific and sensitive enzyme-linked immunosorbent assays. Results were correlated with concentrations of hCG and progesterone. We observed a significant fall in plasma concentration of inhibin pro-alpha C following administration of mifepristone, which continued after administration of misoprostol. In contrast mifepristone had no effect on plasma levels of inhibin A, which fell steeply only after administration of misoprostol. These results suggest dissociation between major sources of inhibin A and inhibin pro-alpha C in early pregnancy. Treatment with mifepristone, a competitive antagonist of the progesterone receptor, resulted in a significant and rapid fall in concentrations of inhibin pro-alpha C, identifying a link between production of pro-alpha C and luteal steroidogenesis. In contrast, concentrations of inhibin A did not fall after mifepristone, identifying a predominantly feto-placental origin in early human pregnancy.

Plasma concentrations of inhibin a and follicle-stimulating hormone differ between cows with two or three waves of ovarian follicular development in a single estrous cycle.

Patterns of ovarian follicle development were monitored daily in Holstein-Friesian cows that had two (n = 4) or three (n = 4) waves of ovarian follicle development during a single estrous cycle. The plasma from daily blood samples was used in assays for inhibin A, FSH, progesterone, and estradiol-17beta. Mean cycle lengths for cows with two and three waves were 21.8 and 25.3 days, respectively (P < 0.02). Although the average number of follicles >3-mm diameter on each pair of ovaries was similar for two- and three-wave cows on Days 2, 3, and 4 (Day 0 = day of ovulation; 8.6 vs. 9.6 follicles), there were more follicles >6-mm diameter on the ovaries of cows with two waves on Days 3 and 4. This difference was associated with a shorter interval from wave emergence to peak concentrations of inhibin A during the first wave in two-wave cows (2.0 vs. 3.8 days; P = 0.03) and with higher peak concentrations (474 vs. 332 pg/ml; P = 0.03). Differences in peak FSH concentrations were not significant (1.7 vs. 1.3 ng/ml; P = 0.10) and were inversely related to inhibin A concentrations. The peak concentrations of inhibin A and FSH in the second nonovulatory wave in the three-wave cows were similar to the low concentrations measured in the first wave (292 vs. 332 pg/ml of inhibin A, 1.3 vs. 1.3 ng/ml of FSH; P > 0.20). Average peak concentrations of inhibin A and FSH were similar during the ovulatory wave for cows with either two or three waves in a cycle (432 vs. 464 pg/ml of inhibin A, 2.3 vs. 2.1 ng/ml of FSH; P > 0.3). The lower concentrations of FSH during the emergence of the first follicular wave in cows with three-wave cycles may have reduced the rate of development of some of the follicles and reduced the concentrations of inhibin A. This pattern of lower concentrations of FSH and inhibin A was repeated in the second nonovulatory wave but not in the ovulatory wave. Subtle differences in the concentrations of these two hormones may underlie the mechanism that influences the number of waves of ovarian follicle development that occur during the bovine estrous cycle.

Inhibin secretion in the golden hamster (Mesocricetus auratus) testis during active and inactive states of spermatogenesis induced by the restriction of photoperiod.

Gonadal function in the male golden hamster (Mesocricetus auratus) was investigated during exposure to a short photoperiod condition. Within 3 weeks of exposure to the short photoperiod condition, FSH and testosterone in the plasma significantly decreased, and subsequently immunoreactive (ir)-inhibin significantly decreased. Testicular contents of ir-inhibin and testosterone, and pituitary contents of LH and FSH also significantly decreased by 3 weeks with regression of weight of testes, epididymis and seminal vesicles and sperm head count. Circulating LH varied but not significantly. Thereafter, all reproductive parameters and secretion of LH, FSH, ir-inhibin and testosterone gradually recovered after 17 weeks of exposure even though animals continued to be subjected to the short photoperiod condition. Plasma concentrations of inhibin B and inhibin pro-alphaC were detectable and were significantly decreased after 15 weeks of exposure to the short photoperiod, but their levels were still detectable. Immunopositive reaction of inhibin alpha and betaB subunits was found in Sertoli cells and Leydig cells in the regressed testes of animals subjected to short photoperiod as was also seen in animals before exposure to the short photoperiod. Although the spermatogenic cycle was suppressed like prepubertal animals, the present study showed that the testicular recovery, so-called refractoriness, is functionally different from the developing stage of immature animals, especially with regard to inhibin secretion. The present results showed that changes in FSH preceded changes in inhibin during the regression and recovery phases, indicating that FSH is a major regulatory factor of inhibin secretion in male golden hamsters. The present study also demonstrated that regressed testes still secrete a small amount of bioactive inhibin during exposure to a short-photoperiod condition.

Relationship between Peripheral Plasma Inhibin and Progesterone Concentrations in Sahiwal Cattle (Bos Indicus) and Murrah Buffaloes (Bubalus bubalis)

The present study investigated peripheral plasma immunoreactive inhibin (ir-inhibin) concentrations in relation to the stage of oestrous cycle and progesterone concentrations in cycling Sahiwal cattle (Bos indicus) and Murrah buffaloes (Bubalus bubalis). Blood samples were collected once daily for thirty-two consecutive days from cattle and buffaloes (5 each) during winter months of January and February. Mean (±S.E.M.) plasma ir-inhibin concentrations ranged from 0.40±0.01 to 0.59±0.03 ng/ml in cattle and from 0.29±0.03 to 0.52±0.05 ng/ml in buffaloes. In cattle, ir-inhibin concentrations increased from 0.47±0.07 ng/ml on day -4 (day 0=day of oestrus) to reach a maximum value of 0.59±0.03 ng/ml on day -2. Thereafter, ir-inhibin concentrations showed a decline to reach a low of 0.40±0.01 ng/ml on day 11 of the oestrous cycle. In buffaloes, ir-inhibin concentrations increased from 0.38±0.04 ng/ml on day -4 to reach a maximum concentration of 0.52±0.05 ng/ml on day -2. Ir-inhibin concentrations then declined to reach a low of 0.29±0.03 ng/ml on day 9 of the cycle. In both cattle and buffaloes, ir-inhibin concentrations which were lowest (0.43±0.02 and 0.34±0.02 ng/ml, respectively) during the mid-luteal phase of the oestrous cycle increased (p<0.05) to 0.52±0.03 and 0.44±0.04 ng/ml, respectively, during the late luteal phase, and then further to the highest value of 0.53±0.02 and 0.49±0.04 ng/ml, respectively, during the perioestrus phase, following which these declined to 0.50±0.02 and 0.39±0.03 ng/ml, respectively, during the early luteal phase. The variations in peripheral plasma ir-inhibin profile in both the species appear to be related to the changes in characteristics of follicular populations during the oestrous cycle. Peripheral plasma ir-inhibin concentrations were negatively correlated with progesterone concentrations in cattle (r=-0.51, p<0.01) and buffaloes (r=-0.30, p<0.01) indicating that the corpus luteum is not a source of peripheral ir-inhibin in these species. Dynamics of messenger RNAs encoding inhibin/activin subunits and follistatin in the ovary during the rat estrous cycle.

Quantitative changes in ovarian inhibin/activin subunit and follistatin mRNAs during the rat estrous cycle were examined by ribonuclease protection assay using digoxygenin-labeled RNA probes. Levels of ovarian inhibin alpha subunit mRNA remained low throughout estrus, metestrus, and diestrus; abruptly increased on the morning of proestrus; then rapidly decreased when the primary gonadotropin surge occurred. A similar changing pattern was observed in inhibin/activin beta(A) subunit mRNA. On the other hand, inhibin/activin beta(B) subunit mRNA showed a different changing pattern. Levels of beta(B) subunit mRNA remained constant during metestrus and diestrus, abruptly decreased on the afternoon of proestrus, then quickly recovered from the nadir by 1100 h on estrus. Throughout the rat estrous cycle, especially during the periovulatory period, alpha subunit mRNA levels were considerably higher than beta(A) and beta(B) subunit mRNA levels. In addition, changes in plasma concentrations of inhibin A and inhibin B were very similar to that in ovarian beta(A) and beta(B) subunit mRNA levels, respectively, with several-hour delays. These results suggest that levels of beta subunit mRNAs restrict secretion of dimeric inhibins. Levels of follistatin mRNA remained low from the midnight of metestrus to the midnight of diestrus, then increased until initiation of the primary gonadotropin surge. Thereafter, follistatin mRNA decreased, reached the nadir at 0200 h on estrus, then increased abruptly at 1100 h on estrus. Afterward, follistatin mRNA levels remained high until the morning of metestrus. The changing pattern of ovarian follistatin mRNA was similar to, and preceded, the changes in plasma concentrations of progesterone, suggesting that ovarian follistatin may modulate progesterone secretion during the rat estrous cycle.

Secretory pattern of inhibin A, inhibin B and inhibin pro-alpha C during induced follicular atresia and subsequent follicular development in the golden hamster (Mesocricetus auratus)

The changes in plasma concentrations of inhibins A, B and pro-alpha C were determined in the cyclic golden hamster during follicular atresia induced with antiserum against luteinizing hormone releasing hormone (LHRH-AS) at 1100 h on day 4 (day 1=day of ovulation). Follicular status in the ovary was also studied by determining the number of follicles ovulating in response to human chorionic gonadotrophin (hCG) injection. The time-courses of changes in plasma concentrations of inhibins A, B and pro-alpha C were different from each other during induced follicular atresia and subsequent follicular development. Plasma concentrations of inhibin A decreased to 58.6% of initial values by 24 h after LHRH-AS treatment, and then remained relatively low until at least 60 h later. Plasma concentrations of inhibin B decreased to 64.2% of the initial values by 18 h after LHRH-AS treatment and remained at basal values for 36 h, but increased abruptly to greater than initial values at 42 h after the treatment. Plasma concentrations of inhibin pro-alpha C increased at 6 and 12 h, decreased suddenly to 21.9% of the initial values by 24 h after LHRH-AS treatment, and then gradually increased until 60 h after LHRH-AS. The number of follicles responding to hCG decreased gradually between 0 and 30 h after LHRH-AS, when no ovulations were observed, and then gradually increased until 60 h. The changes in follicular ovulatory responses to hCG correlated with the plasma profile of inhibin A throughout the experiment. These results suggest that inhibin A is mainly secreted by large antral follicles. In contrast, during the subsequent follicular development, the plasma concentration of inhibin B increased earlier than that of inhibin A. These results suggest that inhibin B is secreted by small and large antral follicles. Plasma concentrations of inhibin pro-alpha C were high at a time when plasma concentrations of oestradiol-17 beta had already decreased, indicating that inhibin pro-alpha C is secreted not only from healthy follicles but also from early atretic antral follicles.

Circulating Inhibin A and Inhibin B in Normal Menstrual Cycle during Breeding Seasons of Japanese Monkeys.

In order to clarify the endocrinological profiles of inhibins during normal menstrual cycles in Japanese monkeys, plasma concentrations of inhibin A and inhibin B were measured by enzyme-linked immunosorbent assay. In addition, circulating gonadotropins and steroid hormones were measured. Blood samples were taken from four Japanese monkeys showing normal menstrual cycles. Plasma inhibin A remained low during the early follicular phase but was high during the luteal phase. Plasma inhibin B was high in the early follicular phase, but fell during the late follicular phase, and remained low during the luteal phase. A significant inverse correlation was observed between plasma concentrations of inhibin B and FSH during the mid-follicular phase, but not between inhibin A and FSH. Plasma inhibin A showed a significant positive correlation with progesterone in the normal menstrual cycle. These results suggest that inhibin B during mid-follicular phase is mainly involved in the suppression of FSH secretion from the pituitary gland, and also that the main source of inhibin A during the luteal phase is corpora lutea in normal menstrual cycle of Japanese monkeys.

Plasma Inhibin Levels in Relation to Steroids and Gonadotrophins during Oestrous Cycle in Buffalo

The study was conducted on six Murrah buffalo synchronized and induced to oestrus. An indwelling catheter was placed in the jugular vein of each buffalo 4 days before the expected onset of the oestrus following the induced oestrus and blood samples were collected at 8 h intervals from each animal throughout the oestrous cycle. Plasma immunoreactive inhibin, follicle-stimulating hormone (FSH), luteinizing hormone (LH), estradiol-17 beta and progesterone were estimated by radioimmunoassay to study the variations in the peripheral levels of these hormones and their inter-relationships in order to elucidate the feedback systems controlling them during the oestrous cycle of buffalo. Plasma inhibin levels ranged between 391.25 and 631.97 pg/ml during various phases of the oestrous cycle and were found to be higher than reported in cows. Peak LH and FSH levels during oestrus were 38.40 +/- 9.21 and 24.04 +/- 4.75 ng/ml, respectively and estradiol-17 beta and progesterone were 19.50 +/- 5.51 pg/ml and 0.61 +/- 0.25 ng/ml, respectively. The mean plasma inhibin concentration on the day of oestrus was 562.5 +/- 18.9 pg/ml. Levels of FSH in the plasma showed three mid-cycle elevations which corresponded to comparatively lower inhibin and elevated estradiol-17 beta levels during the same period. From this observation it was deduced that both inhibin and estradiol-17 beta have a feed-back regulatory effect on FSH secretion in buffalo.

Effect of number and diameter of follicles on plasma concentrations of inhibin and FSH in mares

The role of the number of follicles and circulating immunoreactive inhibin in the decrease in plasma FSH concentrations that occurs during development of a follicular wave was studied in mares. All follicles > or = 6 mm in diameter were ablated by ultrasound-guided transvaginal aspiration of follicular fluid on day 10 after ovulation. During the subsequent wave, all follicles, the three largest follicles (three follicle group), the largest follicle (single follicle group) or no follicles were retained and the remaining follicles were ablated before they reached > 10 mm in diameter (n = 10-11 mares per group). Ablation of new follicles was continued until the day on which the largest follicle of the new wave reached 25 mm in diameter (day 18 after ovulation in the 'no follicle' group). Diameters of retained follicles were measured once a day by transrectal ultrasonography. Plasma samples were taken once a day and analysed by radioimmunoassay for concentrations of FSH and immunoreactive inhibin (includes dimeric inhibin as well as free alpha-subunit forms). Data were normalized to the day of the expected start of the decrease in plasma FSH concentrations (day 0: largest follicle 13 mm in diameter in the follicle-retained groups). A simultaneous increase in circulating concentrations of FSH (P < 0.05) and immunoreactive inhibin (P < 0.05) occurred before the largest follicle reached 13 mm in diameter, which indicates that immunoreactive inhibin produced by follicles < 13 mm in diameter did not suppress FSH. Plasma concentrations of FSH decreased (P < 0.05) and immunoreactive inhibin concentrations increased (P < 0.05) after day 0 in the follicle-retained groups. A slower decrease in FSH concentrations was associated temporally with a delay in the increase in immunoreactive inhibin concentrations in the 'single follicle' group relative to the 'three follicle' and 'all follicle' groups. All follicle-retained groups had similar plasma concentrations of FSH and immunoreactive inhibin after the expected beginning of deviation in growth rates between the two largest follicles (largest follicle 22-23 mm in diameter). These results indicated that the decrease in plasma FSH concentrations from the start of the decrease until the expected day of deviation was a function of multiple follicles of a wave and was attributable to the secretion of inhibin. Thereafter, the largest follicle alone accounted for the continued FSH suppression.
