An efficient protocol for inducing pseudopregnancy using estradiol dipropionate and follicular development associated with changes in reproductive hormones after prostaglandin F2alpha treatment in pseudopregnant sows

BackgroundUtilization of estrus synchronization program in livestock industry would provide greater options for reproductive management in herd. To develop a convenient method for estrus synchronization in pigs, we determined the effective protocol using estradiol dipropionate (EDP) for the establishment of pseudopregnancy and investigated follicular development during the estrus synchronization with prostaglandin F2alpha (PGF2alpha) in association with reproductive hormone profiles in pseudopregnant sows.MethodsIn Experiment 1, the effective dose (0, 10, 20, or 30 mg) and timing (5, 8, 11 or 13 days after ovulation) of a single administration of EDP in cyclic pigs for the induction of pseudopregnancy were investigated. In Experiment 2, four pseudopregnant sows were treated with PGF2alpha twice at a 24-h interval between 24 and 28 days after EDP treatment. The changes in plasma concentrations of reproductive hormones were analyzed by time-resolved fluoroimmunoassay. Follicular development and ovulation following PGF2alpha administration were monitored by transrectal ultrasonography.ResultsHigh efficiency (greater than 80%) of pseudopregnancy was achieved with a single treatment with 20 mg of EDP at 8 and 11 days after ovulation (equivalent to 9-13 days after the onset of estrus). Plasma estradiol-17beta concentrations in pseudopregnant sows were significantly higher between 12 h and 7 days than before EDP treatment. Total inhibin concentrations significantly decreased following EDP treatment and remained low for 14 days. The number of small follicles was increased from 6.3 +/- 2.6 at PGF2alpha treatment to 22.8 +/- 4.8 at 3 days later; this was associated with increased plasma concentrations of inhibin. Onset of estrus was detectable in all sows on 5.3 +/- 0.3 days after PGF2alpha treatment and the number of ovulated follicles was 15.5 +/- 1.4 detected at 7.6 +/- 0.2 days after the treatment.ConclusionsThis study has defined the effective dose and timing of EDP treatment for inducing pseudopregnancy in cyclic pigs. Our results also indicated that EDP caused a lowering of inhibin concentrations during pseudopregnancy and small numbers of follicles from 20 to 28 days after EDP. In contrast, EDP-induced pseudopregnancy appears to have no adverse effect on follicular development and subsequent ovulation following PGF2alpha administration.

Dynamic Nature of Alterations in the Endocrine System of Fathead Minnows Exposed to the Fungicide Prochloraz

The vertebrate hypothalamic-pituitary-gonadal (HPG) axis is controlled through various feedback mechanisms that maintain a dynamic homeostasis in the face of changing environmental conditions, including exposure to chemicals. We assessed the effects of prochloraz on HPG axis function in adult fathead minnows (Pimephales promelas) at multiple sampling times during 8-day exposure and 8-day depuration/recovery phases. Consistent with one mechanism of action of prochloraz, inhibition of cytochrome P450 (CYP) 19 aromatase activity, the fungicide depressed ex vivo ovarian production and plasma concentrations of 17beta-estradiol (E2) in female fish. At a prochloraz water concentration of 30 microg/l, inhibitory effects on E2 production were transitory and did not persist during the 8-day exposure phase. At 300 microg/l prochloraz, inhibition of E2 production was evident throughout the 8-day exposure but steroid titers recovered within 1 day of cessation of exposure. Compensation or recovery of steroid production in prochloraz-exposed females was accompanied by upregulation of several ovarian genes associated with steroidogenesis, including cyp19a1a, cyp17 (hydroxylase/lyase), cyp11a (cholesterol side-chain cleavage), and follicle-stimulating hormone receptor. In male fathead minnows, the 8-day prochloraz exposure decreased testosterone (T) production, possibly through inhibition of CYP17. However, as for E2 in females, ex vivo testicular production and plasma concentrations of T recovered within 1 day of stopping exposure. Steroidogenic genes upregulated in testis included cyp17 and cyp11a. These studies demonstrate the adaptability of the HPG axis to chemical stress and highlight the need to consider the dynamic nature of the system when developing approaches to assess potential risks of endocrine-active chemicals.

Serum levels of FSH, LH and estradiol-17 beta in female rats around the time of puberty onset.

Groups of female rats were autopsied at daily intervals from 27 days of age through the first vaginal cycle and the serum FSH and LH and plasma estradiol-17 beta (E2) concentrations were estimated. Fluctuating hormone concentrations were recorded between days 27 and 31. Both gonadotropins and E2 dropped to a very low level on days 32 and 33 and showed then a progressive increase that commenced on day 34 and terminated for LH and E2 on day 39, i.e. in proestrus, whereas FSH declined from day 37 to day 39. The circulating gonadotropin level was lower during the first postpubertal diestrus than during most of the prepubertal phase. The findings suggest that a high sensitivity to the gonadotropin -inhibiting effect of estrogen prevented completion of an ovarian cycle up to day 33. After that, desensitization to the negative estrogen feedback as indicated by the simultaneous rise of E2 and gonadotropins makes final maturation of ovarian follicles and the first ovulation possible. In part of the rats aged 27-38 days cell nuclear volumes of medial preoptic neurons were evaluated, because recent results suggest that estrogen induces the desensitization process by an inhibitory action on the medial preoptic area. In accordance with this assumption, an inverse relationship between nuclear size and the E2 level in the blood could be revealed.

Plasma and Fecal Sex Steroid Hormone Profiles During the Estrous Cycle in a Japanese Serow (Capricornis crispus)

We aimed to determine plasma progesterone and estradiol-17beta concentrations, as well as fecal progesterone concentrations during the estrous cycle in a female 8-year-old Japanese serow (Capricornis crispus). The step frequencies during the night were recorded by a pedometer attached to the serow's hind leg from October to the following June. Estrous behavior was also monitored during the day. Blood samples were taken once a day from 8 February to 10 March from an indwelling catheter placed in the jugular vein. Fecal samples were taken once or twice a day from 1 January to 29 April. Plasma and fecal progesterone and plasma estradiol concentrations were determined using time-resolved fluoroimmunoassays. The estrus behavior observed lasted for 2-3 days. Peak step frequencies were recorded between November and April, at intervals of 17-19 days. Plasma progesterone concentrations remained elevated (1-7.7 ng/ml) for 12 days during non-estrus, and plasma estradiol concentrations were highest when the peak step frequency and estrous behavior were observed. Step frequency increased around the times fecal progesterone levels fell to basal levels. Progesterone concentrations in feces were significantly correlated with those in sera. Thus, measurement of fecal progesterone concentrations might be useful for monitoring the reproductive status of the Japanese serow.

Effects of catecholamines on ovary morphology, blood concentrations of estradiol-17β, progesterone, zinc, triglycerides and rate of ovulation in domestic hens

The present study is an attempt to shed more light on the role of epinephrine (EP) and norepinephrine (NE) in regulating ovarian follicular development, folliculogenesis and ovulation in laying hens. Sixty Egyptian local cross females (Mandarah), 50 weeks old, were individually housed and equally divided into three treatments: control (saline, 0.9% NaCl), EP (0.15 mg epinephrine/hen/day) and NE (0.75 mg norepinephrine/hen/day) (n=20). Animals were injected intramuscularly once a day for 15 successive days. At the end of the experimental period, 10 females from each treatment were randomly chosen, weighed and killed by decapitation. Ovaries and oviducts and ovarian follicles were examined. Plasma concentrations of estradiol-17beta, progesterone, zinc and triglyceride were determined. Results indicated that the ovaries of NE- and EP-treated hens were more developed than those of control hens being heavier and containing more yellow yolk-filled follicles. EP or NE significantly increased the ovulation rate and plasma concentrations of estradiol-17beta, progesterone, zinc and triglyceride compared with control treatment. It could be concluded that catecholamines may have a part in promoting ovarian follicular development and in stimulating ovulation in laying hens at the end of their reproductive lives.

Expression of Nerve Growth Factor and Its Receptors NTRK1 and TNFRSF1B Is Regulated by Estrogen and Progesterone in the Uteri of Golden Hamsters1

Experiments were conducted using female golden hamsters to identify the presence of nerve growth factor (NGF) and its receptors NTRK1 and TNFRSF1B in the uteri of female animals and regulation on their expression by estrogen and progesterone. NGF and its receptor NTRK1 were immunolocalized to luminal epithelial cells, glandular cells, and stromal cells. TNFRSF1B was immunolocalized in luminal epithelial and glandular cells, with no staining found in stromal cells of the uterine horns of normal cyclic golden hamsters. Strong immunostaining of NGF and its receptors NTRK1 and TNFRSF1B was observed in uteri on the day of proestrus as compared to the other stages of the estrous cycle. Results of immunoblot analysis of NGF revealed that there was a positive correlation between uterine NGF expression and plasma concentrations of estradiol-17beta. To clarify the effects of estrogen and progesterone on NGF, NTRK1, and TNFRSF1B expression, adult female golden hamsters were ovariectomized and treated with estradiol-17beta and/or progesterone. Immunoblot analysis and immunohistochemistry indicated that estradiol-17beta stimulated expression of NGF and its two receptors in the uterus. Treatment with progesterone also increased NGF and NTRK1 expression in the uterus. However, no additive effect of these steroids on expression of NGF and its receptors was observed. Changes in uterine weights induced by estradiol-17beta and/or progesterone showed the same profile with that of NGF, suggesting that a proliferative act of NGF may be involved in uterine growth. These results suggest that NGF may play important roles in action of steroids on uterine function.

Relations between Plasma IGF-I Concentrations during Treatment with CIDR-based or Ovsynch Protocol for Timed AI and Conception in Early Postpartum Japanese Black Beef Cows

We examined the relations between plasma insulin-like growth factor (IGF) -I concentrations during treatment with CIDR-based or Ovsynch protocol for timed AI and conception and plasma steroid concentrations in early postpartum Japanese Black beef cows. Cows in the control group (Ovsynch; n = 21) underwent Ovsynch protocol (GnRH analogue on Day 0, PGF(2alpha) analogue on Day 7, and GnRH analogue on Day 9), with AI on Day 10, approximately 20 h after the second GnRH treatment. Cows in the Ovsynch+CIDR group (n = 22) received Ovsynch protocol plus a CIDR for 7 days (starting on Day 0). Cows in the further treatment group (EB+CIDR+GnRH; n = 22) received 2 mg of estradiol benzoate (EB) on Day 0 in lieu of the first GnRH treatment, followed by the same treatment as in the Ovsynch+CIDR protocol. Plasma IGF-I concentrations were determined on Days -7, 0, 7, 9 and 17. Conception rates were improved in the CIDR-combined groups (both CIDR-treated groups were combined) relative to Ovsynch group (P < 0.05) for cows with low IGF-I concentrations (<1,000 ng/ml) on Days -7, 0, and 7, but improved conception rate produced by the CIDR-based protocols did not occur in cows with a high IGF-I concentration (> or =1,000 ng/ml). Plasma estradiol-17beta concentrations increased from Day 0 to 7 (P < 0.05) and were unchanged from Day 7 to 9 in the Ovsynch group with low IGF-I concentrations on Day 0, while they were unchanged from Day 0 to 7 and increased from Day 7 to 9 (P < 0.05) in the Ovsynch group with high IGF-I concentrations on Day 0 and in the CIDR-combined group. Plasma progesterone concentrations in the Ovsynch group with low IGF-I concentrations on Day 0 were higher on Day 14 than in the Ovsynch group with high IGF-I concentrations on Day 0 and in the CIDR-combined group (P < 0.05). In conclusion, CIDR-based protocols may improve conception relative to Ovsynch in early postpartum beef cows with lower plasma IGF-I concentrations at the start of the protocols. This improvement is probably due to prevention of premature increases of estradiol-17beta and progesterone concentrations, which occurred in cows with low IGF-I concentrations treated with Ovsynch, by the CIDR treatment.

Elevated atopy in healthy obese women

Allergic disorders, including asthma, have increased dramatically in the United States in the past 20 y. Epidemiologic studies have found body mass index (body weight in kg/height squared in m) to be a positive independent correlate of atopy in women but not in men. We investigated the prevalence of atopy among healthy obese and nonobese women and its relation to fat mass (FM), insulin resistance, and plasma concentrations of 17beta-estradiol, interleukin 4 (IL-4), and leptin. A cross-sectional study of 21 obese (> or = 30% body fat) and 22 nonobese (< 30% body fat) women (18-41 y of age) was performed. The following measurements were taken: FM by plethysmography, total and specific immunoglobulin E (IgE) by automated immunosorbent analysis, and blood glucose, insulin, C-peptide, 17beta-estradiol, sex hormone-binding globulin, and IL-4. Insulin sensitivity was determined on the basis of the fasting insulin resistance index and with an oral-glucose-tolerance test. The frequency of specific IgE in the obese group was almost 3 times that in the nonobese group (P = 0.008). The total IgE concentration was not significantly different between groups. Plasma concentrations of 17beta-estradiol, the ratio of 17beta-estradiol to sex hormone-binding globulin, the fasting insulin resistance index, and C-peptide and leptin concentrations were higher in the obese than in the nonobese group (P < 0.05) after adjustment for oral contraceptive use. All factors correlated positively with FM. Logistic regression showed FM to be the only positive predictor of specific IgE (P = 0.01). The findings confirm a direct relation between obesity and a T helper 2 cell immune response in women.

Assessment of variability of renal blood flow Doppler parameters during the menstrual cycle in women

To estimate variability of renal flow Doppler parameters during the menstrual cycle in young women, and to explore whether the parameters oscillate substantially throughout the cycle. The peak systolic, mean, and end-diastolic velocities, pulsatility (PI) and resistance (RI) indices, acceleration time (AT) and index (AI) of the right renal artery were measured nine times during the cycle with duplex sonography in 14 healthy women (age range, 23-25 years), and correlated with plasma concentrations of 17beta-estradiol and progesterone. Coefficient of variation (CV), interclass correlation (rho), and repeatability coefficients (RC) were used to measure the variability. The measures of the variability for AI and AT were: CV = 45.4% and 37.3%; rho = 0.25 and 0.15; RC = 10.1 and 99.5, respectively. The CV for flow velocities varied from 24.1% to 25.5%, rho from 0.49 to 0.50, RC from 22.7 for end-diastolic to 63.3 for peak systolic velocity. The respective figures for PI and RI were: CV = 17% and 8.3%; rho = 0.21 and 0.29; RC = 0.53 and 0.15. Fluctuations of the flow parameters, hematocrit, hemoglobin, heart rate, and systolic blood pressure during the menstrual cycle were insignificant, while the diastolic blood pressure decreased by about 7 mmHg (P < 0.01) in the luteal phase. The variability of renal flow Doppler parameters during the menstrual cycle in young healthy women is substantial. However, fluctuations of the values of the parameters throughout the cycle were non-significant.

Immunization against endogenous inhibin increases normal oocyte/embryo production in adult mice.

The objectives of the present study were to induce super-ovulation in adult mice by immunoneutralization of endogenous inhibin and to investigate embryo development in vivo and in vitro. Adult female mice of the ddY strain at 3 mo of age were superovulated with a single ip injection of inhibin antiserum (inhibin-AS; 50, 100, or 200 micro L) at 12:00 h on metestrus of the 4-d estrous cycle. The control group was treated with 100 micro L normal goat serum (control serum). Five mice were sacrificed every 6 h and blood samples were collected for hormonal assay. Individual female mice were bred in the afternoon of proestrus with individual fertile males. Embryos were collected at 08.00 h on d 2 post-coitus and cultured in KSOM solution. Treatment with inhibin-AS significantly increased the concentrations of plasma FSH. Furthermore, there was a significant elevation in plasma concentrations of estradiol-17beta and progesterone in the inhibin-immunized groups in comparison with controls. The superovulated oocytes that were fertilized normally in vivo were able to develop normally to blastocysts in vitro. The number of oocytes and blastocysts developed in animals treated with inhibin-AS was significantly higher than that for mice treated with control serum. Moreover, the rate of fertilization and the rate of blastocyst development were similar in inhibin-AS treated and control groups. These results indicate that immunoneutralization of endogenous inhibin induces superovulation in adult mice without additional treatment with human chorionic gonadotropin (hCG). In addition, the superovulated oocytes obtained by administration of inhibin-AS have normal embryonic developmental competence.

Cervical dilatation related to uterine electromyographic activity and endocrinological changes during prostaglandin F(2alpha)-induced parturition in cows.

The temporal relationship between changes in cervical dilatation, uterine electromyographic (EMG) activity, and maternal plasma concentrations of estradiol 17beta (E(2)), progesterone (P(4)), and 13,14-dihydro-15-keto-prostaglandin-F(2alpha) (PGFM), was investigated in six parturient cows. Calving was induced with a single injection of a synthetic analogue of prostaglandin F(2alpha) (PG) on Day 274 of gestation. Cervical dilatation was measured continuously by measuring the transit time between two implanted ultrasound crystals while at the same time uterine EMG activity was measured through two silver electrodes sutured on the myometrial surface until the expulsive stage of calving had been reached. In blood samples collected at 4-h intervals, starting at the moment of PG injection, the mean plasma E(2) concentration gradually increased and was significantly elevated at 28 h after PG injection. At 4 h after PG treatment, the mean P(4) concentration had dropped significantly and continued to decrease until a value of around 1 ng/ml was reached, where it stayed until the onset of expulsion. Mean plasma PGFM concentrations increased steadily after PG injection, reaching significantly elevated concentrations at 20 h after treatment. In the five cows that delivered calves in anterior positions, uterine EMG activity, expressed as root mean square (RMS in microV), started to increase at a mean interval (+/- SD) of 13.1 +/- 3.7 h following PG treatment. The increase in EMG activity was significantly correlated with changes in plasma PGFM concentrations. In these cows, dilatation of the caudal cervix started after a mean (+/- SD) interval of 28.5 +/- 1.5 h following PG treatment and dilatation progressed at a mean (+/- SD) rate of 2.25 +/- 0.24 cm/h. In one cow with a calf in the posterior position, uterine EMG activity and dilatation started at 15.8 h and 31.8 h, respectively, after induction of calving. We conclude that a predictable sequence of physiological changes occurs around induction of calving, which allows specific timing of future studies on cellular and biochemical changes within the cervix during parturition.

Effects of pulp and paper mill effluents on reproductive success of largemouth bass

This study evaluated the effects of bleached and unbleached kraft mill effluent on reproductive success of largemouth bass (Micropterus salmoides). Bass were exposed to effluent concentrations (0, 10, 20, 40, or 80%) for 28 and 56 d. Parameters measured included hepatosomatic index (HSI) and gonadosomatic index (GSI) and plasma concentrations of 17beta-estradiol (E2), 11-ketotestosterone (11-KT), and vitellogenin (VTG). At the end of the 56-d period, bass were moved to hatchery ponds to evaluate spawning success. Spawning mats with eggs either were brought indoors for evaluation of fecundities, hatchabilities, and egg and fry size (measured at age 3 d), or were left in ponds and fry number and size recorded (average age of 14 d). Effluent exposure was verified by measuring resin acids (isopimaric, abietic. and dehydroabietic acids) in bile. Compared to controls, exposed bass had greater concentrations of resin acids in bile. In general, exposed females had lower concentrations of E2 and VTG (> or = 20% effluent), whereas males had lower concentrations of 11-KT (> or = 20% effluent) and increased E2 (> or = 20% effluent). The HSI values increased in females (> or = 10% effluent), and GSI values decreased in both sexes (> or = 40% effluent). Fecundity, egg size, and hatchability did not differ across treatments, but an increase in the frequency of fry abnormalities and a decrease in fry weights was observed at effluent exposures of 40% and higher. However, results from the pond study, revealed a significant reduction in fry growth and survival (> or = 10%). This decline may have been caused by an increased frequency of deformities, in conjunction with alterations of growth. These changes could have resulted from alterations in egg quality because of failure of parental reproductive systems, from acute embryo toxicity after translocation of contaminants from the mother to the developing embryo, or from both.

Pfaffia paniculata-induced changes in plasma estradiol-17beta, progesterone and testosterone levels in mice.

The present study undertook chemical analysis of components of Pfaffia paniculata roots. In addition, an animal experiment was conducted in which mice had ad libitum access to water enriched with powdered P. paniculata root for 30 days. Changes in plasma concentrations of estradiol-17beta and progesterone in female mice and of testosterone in male mice were ascertained. The results revealed that P. paniculata roots contain two types of phytosteroids, beta-sitosterol and stigmasterol, in addition to other compounds such as pfaffic acid, allantoin, saponins, beta-sitosteryl-beta-D-glucoside, and stigmasteryl-beta-D-glucoside. Regarding changes in plasma concentrations of hormones, levels of the sex hormones estradiol-17beta, progesterone and testosterone were clearly higher for mice that drank P. paniculata root-enriched water than for mice that drank plain water. Powdered P. paniculata root is easily dissolved in feed or water, and as no adverse reactions were seen in mice within 30 days of oral intake, consumption of P. paniculata for long periods of time appears safe.

Plasma steroid profiles following follicle-stimulating hormone or equine chorionic gonadotropin injection in cows chronically treated with gonadotropin-releasing hormone agonist.

Plasma steroid profiles following follicle-stimulating hormone (FSH) or equine chorionic gonadotropin (eCG) injection were studied in chronically gonadotropin releasing hormone agonist (GnRH-A)-treated cows. Follicular development and irINH secretion were stimulated by FSH or eCG injection. The plasma concentrations of estradiol-17 beta (E(2)) and testosterone (T) were markedly increased following eCG injection. However, significant increases of the plasma E(2) and T concentrations were not detected in FSH-treated cows. Ovulation of developed follicles were depended on the hCG injection in both groups. These results show: 1) Follicular response to an exogenous gonadotropin is still remained, 2) Ovulation of developed follicles is induced by hCG injection and 3) FSH and eCG cause disparate plasma steroid profiles, under the influence of repeated GnRH-A treatment.

Endocrine profiles during doe-litter separation and the subsequent pregnancy in rabbits

This study was carried out to determine the effects of a transient doe-litter separation on plasma prolactin, LH, FSH, estradiol-17beta and progesterone concentrations before artificial insemination and during the subsequent pregnancy. Control does (n=12) had free access to nursing, whereas separated does (n=12) were kept away from their litters for 48 hours before artificial insemination. Both groups were inseminated on day 11 after parturition. Teat stimulation by suckling caused a high increase in prolactin concentrations in separated does (p < 0.0001). Basal prolactin concentrations were observed in both groups on days 8 and 18 of pregnancy. No effect of the treatment was detected on LH and FSH concentrations during the sampling period. A rise of estradiol-17beta concentrations was observed 48 hours after doe-litter separation, compared to control does and to previous values (p < 0.003). Both groups showed low progesterone concentrations before artificial insemination. Pregnant rabbits in both groups showed increased progesterone concentrations on days 8 and 18 of pregnancy. Lower estradiol-17beta concentrations were observed in control does on day 18 of pregnancy compared with separated rabbits (p < 0.003). The results suggest that a transient separation of nursing does from their litters before artificial insemination may promote high follicular steroidogenesis activity leading to increased estradiol-17beta concentrations. This hormonal change could be a result of several stimulatory actions probably triggered by the absence of suckling episodes and may affect the luteotrophic function during the subsequent pregnancy.

Relationship between Testicular Transferrin and Plasma Estradiol-17.BETA. Concentrations of Dogs with Azoospermia and Dogs with Sertoli Cell Tumors.

Testicular Transferrin (Tf) and peripheral plasma estradiol-17beta (E2) concentrations were measured in 3 dogs with azoospermia (AZ dogs), 3 dogs with Sertoli cell tumors (SC dogs), and 5 normal male Beagles. The mean Tf concentrations in the testes of the AZ dogs and the affected testes of the SC dogs, and the plasma E2 concentrations in both these groups of dogs were significantly higher than the values in normal dogs (P<0.05, 0.01 and 0.01, respectively). Therefore, excessive E2 secretion by hyperfunctioning Sertoli cells is thought to have caused the azoospermia in the 3 dogs.

Use of a small dose of estradiol benzoate during diestrus to synchronize development of the ovulatory follicle in cattle.

We tested the hypothesis that a small dose of estradiol benzoate (EB) at the midstage of the estrous cycle in cattle would synchronize the subsequent pattern of ovarian follicular development, estrus, and ovulation. Nonlactating Friesian cows received either 1 mg of EB i.m. on d 13 of the estrous cycle (T; n = 12; estrus = d0) or served as untreated controls (C; n = 12). Their ovaries were examined daily with transrectal ultrasonography from d 7, and blood samples were collected 0, 2, 4, 8, 24, and 48 h after treatment on d 13. Plasma concentrations of estradiol-17beta were elevated to 12 pg/mL during the initial 24 h following treatment, compared with a baseline of 1 pg/mL in untreated controls (P < .001). Progesterone concentrations in cows of the T group declined between 24 and 48 h after treatment (-3.2 +/- .5 ng/mL) compared with little change in concentrations of progesterone in cows of the C group at this time (P < .01). This difference was coincident with an earlier time to regression of the corpus luteum in cows of the T group. Disregarding treatment groups, the second dominant follicle of the estrous cycle (DF2) emerged on d 10.6 +/- .3 and was 9.4 +/- .4 mm in diameter on d 13. Further growth of the DF2 was halted by EB treatment on d 13. Cessation of growth occurred irrespective of whether the DF2 was in the early or late growth phase, and a new follicular wave emerged 4.5 +/- .2 d later. The dominant follicle from this wave (DF3) ovulated 5 d after emergence in most cases. During the estrous cycle of every cow in the T group, there were three waves of follicular development (3-wave), whereas the ratio of 2:3 waves of follicular development in cows of the C group was 1:3. Consequently, the interval from emergence to ovulation of the ovulatory dominant follicle in cows of the C group ranged from 3 to 11 d. The dynamics of ovarian follicular wave development during the estrous cycle can be strategically manipulated by treating with a small dose of EB to synchronize proestrous development of the ovulatory follicle.

Azoospermia of dogs with apoptotic germ cells and Leydig cells.

Apoptotic cell death in the testes of 4 dogs with azoospermia was examined. Blood plasma luteinizing hormone (LH), testosterone (T), and estradiol-17beta (E2) concentrations, and testicular transferrin (Tf) concentration as a marker of Sertoli cell function were measured in the 4 azoospermic dogs and in 5 normal dogs. The spermatids in 2 of the 4 azoospermic dogs and the Leydig cells in 3 of them exhibited apoptotic cell death. Mean LH, E2, and Tf concentrations in the 4 azoospermic dogs were significantly higher than in the normal dogs (P<0.01). These findings suggested that the azoospermia in all 4 dogs might has been caused by abnormal functions of Sertoli cells as well as Leydig cells.

Ontogeny of luteinizing hormone and estradiol secretion in turkey hens exposed to different photoperiods from hatch to sixty weeks of age

Changes in concentrations of plasma luteinizing hormone (LH) and estradiol-17beta in turkey hens were measured during development utilizing stimulatory and nonstimulatory lighting treatments. All hens were maintained under 24 h light (L) from hatch to 3 wk of age, then 16L:8 dark (D) to 10 wk of age. From 10 wk of age, three lighting treatments were used: 1) 16L:8D from 10 to 60 wk of age (Treatment LD); 2) 6L:18D from 10 to 60 wk of age (Treatment SD); and 3) 16L:8D from 10 to 20 wk of age, 6L:18D from 20 to 30 wk of age, and then 16L:8D from 30 to 60 wk of age (Treatment SD-LD). Measurements of plasma LH and estradiol-17beta concentrations were based on weekly blood samples. Similar patterns of LH were observed in all treatments before 24 wk of age. The concentrations of LH were high at hatch then declined to low levels at 10 wk of age and rebounded to high levels at 18 wk of age before declining to low levels at about 24 wk of age. Low concentrations of LH were then maintained for hens on all treatments but were slightly higher in hens on Treatment SD from 35 to 60 wk of age. A transitory increase in LH, lasting about 3 wk, occurred for hens on Treatment SD-LD after photostimulation. The concentrations of estradiol-17beta were low and stable for hens on all treatments when they were not laying but were elevated about 2 wk prior to and during egg laying. All hens on Treatments LD and SD-LD laid eggs; no hens on Treatment SD laid eggs to 60 wk of age. It was concluded that 1) changes in plasma LH and estradiol-17beta were age related before about 24 wk of age, 2) turkey hens may not be photorefractory at hatch and can lay without juvenile exposure to SD photoperiods, and 3) LD photoperiods after about 20 wk of age are necessary for initiation of egg production in turkey hens before 60 wk of age.

Effects of body weight and feed allocation during sexual maturation in broiler breeder hens. 2. Ovarian morphology and plasma hormone profiles

The effects of broiler breeder BW and nutrient intake on ovary morphology and plasma reproductive hormone profiles were examined at photostimulation (PS) (21 wk) and at sexual maturity (SM) in standard (STD) and low (LOW), or high (HIGH) BW birds provided either restricted (RF) or ad libitum (AL) access to feed between PS and SM. At PS, 30 Shaver Starbro pullets at target BW were assigned to the STD treatment, and birds either 20% heavier (HIGH) or lighter (LOW) assigned accordingly. Ten birds of each size group were processed immediately for carcass analysis and 10 birds assigned to each size by feed interaction group. Blood samples were taken at 3-d intervals beginning at PS and profiles constructed for estradiol-17beta, luteinizing hormone (LH), and follicle-stimulating hormone (FSH) to examine the relationship between body size, feeding level, and reproduction. Birds were processed for assessment of reproductive traits following SM. The AL birds reached SM with 11.0 large yellow follicles (LYF) (> 10 mm diameter) compared to 7.1 in RF birds. Small follicle atresia (< 5 mm diameter) was low in AL birds (10.3) compared to RF birds (32.3). The extent of small follicle atresia in RF birds was found to be inversely proportional to LYF number by stepwise regression. Increased small follicle atresia was associated with a longer sexual maturation period in RF birds (r = 0.619; P = 0.0003). Plasma estradiol-17beta concentration was greater in HIGH than in STD or LOW birds at PS, suggesting more advanced ovary development in HIGH birds. Estradiol-17beta profiles were similar in shape in all treatments, with the primary difference being the length of time prior to a substantial estradiol-17beta increase. Following PS, plasma LH and FSH concentrations of AL birds increased to levels nearly double that of RF birds, indicating a role for nutrient intake with rate of reproductive development. Plasma LH and FSH concentrations remained elevated for a greater time period in RF birds, however, possibly relating to the development of processes limiting LYF recruitment. This experiment demonstrated a modulation of reproductive hormone concentrations during sexual maturation by feeding level in conjunction with a sensitivity of the ovary to nutritional effects.
