The sky is not the limit: Successful foliar application of Steinernema spp. entomopathogenic nematodes to control Lepidopteran caterpillars

Entomopathogenic nematodes (EPNs) are ubiquitous soil-thriving organisms that use chemical cues to seek and infect soil-dwelling arthropods, yielding various levels of biological control. Going beyond soil application, scientists and practitioners started exploring the option of applying EPNs onto the foliage of crops in attempts to manage leaf-dwelling insect pests as well. Despite some success, particularly with protective formulations, it remains uncertain whether EPNs could indeed survive the phyllospheric environment, and successfully control foliar insect pests. In this context, we tested the potential of commercially produced Steinernema feltiae and S. carpocapsae, two of the most commonly used EPNs in the field of biological control, in controlling Lepidopteran foliar pests of economic importance, i.e. Tuta absoluta and Spodoptera spp. caterpillars as models. We first tested the survival and efficacy of both EPN species against the Lepidopteran caterpillars when applied onto tomato, sweet pepper and lettuce leaves, under controlled conditions and in commercial greenhouse conditions, respectively. Subsequently, we explored the behavioural responses of the EPNs to environmental cues typically encountered in the phyllosphere, and analysed plant volatile organic compounds (VOCs). Our results show that both S. feltiae and S. carpocapsae successfully survived and infected the foliar caterpillars, reaching similar level of control to a standard chemical pesticide in commercial practices. Remarkably, both EPN species survived and remained effective up to four days in the phyllosphere, and needed only a few hours to successfully penetrate the caterpillars. Interestingly, S. feltiae was attracted to VOCs from tomato plants, and tended to prefer those from caterpillar-induced plants, suggesting that the nematodes may actively forage toward its host, although it has never been exposed to leaf-borne volatiles during its evolution. The present study shows the high potential of steinernematids in managing major foliar pests in greenhouses and in becoming a key player in foliar biological control. In particular, the discovery that EPNs use foliar VOCs to locate caterpillar hosts opens up new opportunities in terms of application techniques and affordable effective doses.

Terpinen-4-ol from tea tree oil prevents Aspergillus flavus growth in postharvest wheat grain

Plant volatile organic compounds (PVOCs) have gained increasing attention for their role in preventing fungal spoilage and insect contamination in postharvest agro-products owing to their effectiveness and sustainability. In this study, the essential oil was extracted from fresh M. alternifolia (tea tree) leaves, and the fumigation vapor of tea tree oil (TTO) completely inhibited the growth of Aspergillus flavus on agar plates at a concentration of 1.714 μL/mL. Terpinen-4-ol was identified as the major component (40.76 %) of TTO volatiles analyzed using headspace gas chromatography–mass spectrometry. Terpinen-4-ol vapor completely inhibited the A. flavus growth on agar plates and 20 % moisture wheat grain at 0.556 and 1.579 μL/mL, respectively, indicating that terpinen-4-ol serves as the main antifungal constituent in TTO volatiles. The minimum inhibitory concentration of terpinen-4-ol in liquid-contact culture was 1.6 μL/mL. Terpinen-4-ol treatment caused depressed, wrinkled, and punctured mycelial morphology and destroyed the plasma membrane integrity of A. flavus. Metabolomics analysis identified significant alterations in 93 metabolites, with 79 upregulated and 14 downregulated in A. flavus mycelia exposed to 1.6 μL/mL terpinen-4-ol for 6 h, involved in multiple cellular processes including cell membrane permeability and integrity, the ABC transport system, pentose phosphate pathway, and the tricarboxylic acid cycle. Biochemical analysis and 2,7-dichlorofluorescein diacetate staining showed that terpinen-4-ol induced oxidative stress and mitochondrial dysfunction in A. flavus mycelia. This study provides new insights into the antifungal effects of the main TTO volatile compounds terpinen-4-ol on the growth of A. flavus.

Characteristic volatile components analysis of Camelina sativa (L.) Crantz by GC-MS and their antibacterial and antioxidant activities

Camelina sativa (L.) Crantz is an oilseed plant common in Europe and Asia. This study used the gas chromatography–mass spectrometry (GC-MS) to examine the differences in the aroma on the basis of extraction method such as water distillation extraction (CSPW), Solid-phase microextraction (CSPM) and subcritical extraction (CSPS). Antibacterial test was evaluated by the microdilution method against Salmonella typhimurium, Streptococcus pneumoniae, Escherichia coli, Strepococcus pyogenens, Staphylococcus aureus, and antioxidant activity was determined through DPPH free radical, hydroxyl free radical, and superoxide anion radical scavenging capacity activity. The result revealed that three extraction methods were distinct from each other based on their volatile compounds. Sixty-one volatiles of diverse chemical nature were identified and quantified. The volatile components contain thioether, aldehydes, alcohols, ketones, acids, esters, alkene, alkanes, amide, and furan compounds. The volatile components of Camelina sativa (L.) Crantz have good antibacterial and antioxidant activities. Furthermore, this work provides reference methods for detecting novel volatile organic compounds in plants and products.

Drought induces moderate, diverse changes in the odour of grassland species

Plants react to drought stress with numerous changes including altered emissions of volatile organic compounds (VOC) from leaves, which provide protection against oxidative tissue damage and mediate numerous biotic interactions. Despite the share of grasslands in the terrestrial biosphere, their importance as carbon sinks and their contribution to global biodiversity, little is known about the influence of drought on VOC profiles of grassland species. Using coupled gas chromatography-mass spectrometry, we analysed the odorants emitted by 22 European grassland species exposed to an eight-week-lasting drought treatment (DT; 30% water holding capacity, WHC). We focused on the odorants emitted during the light phase from whole plant shoots in their vegetative stage. Emission rates were standardised to the dry weight of each shoot. Well-watered (WW) plants (70% WHC) served as control. Drought-induced significant changes included an increase in total emission rates of plant VOC in six and a decrease in three species. Diverging effects on the number of emitted VOC (chemical richness) or on the Shannon diversity of the VOC profiles were detected in 13 species. Biosynthetic pathways-targeted analyses revealed 13 species showing drought-induced higher emission rates of VOC from one, two, three, or four major biosynthetic pathways (lipoxygenase, shikimate, mevalonate and methylerythritol phosphate pathway), while six species exhibited reduced emission rates from one or two of these pathways. Similarity trees of odorant profiles and their drought-induced changes based on a biosynthetically informed distance metric did not match species phylogeny. However, a phylogenetic signal was detected for the amount of terpenoids released by the studied species under WW and DT conditions. A comparative analysis of emission rates of single compounds released by WW and DT plants revealed significant VOC profile dissimilarities in four species only. The moderate drought-induced changes in the odorant emissions of grassland species are discussed with respect to their impact on trophic interactions across the food web. (294 words)

1-nonene plays an important role in the response of maize-aphid-ladybird tritrophic interactions to nitrogen.

Plant volatile organic compounds (VOCs) are the key distress signals involved in tritrophic interactions, by which plants recruit predators to protect themselves from herbivores. However, the effect of nitrogen fertilization on VOCs that mediate tritrophic interactions remains largely unidentified. In this study, a maize (Zea mays)-aphid (Rhopalosiphum padi)-ladybird (Harmonia axyridis) tritrophic interaction model was constructed under high-nitrogen (HN) and low-nitrogen (LN) regimens. H. axyridis had a stronger tendency to be attracted by aphid-infested maize under HN conditions. Then, volatiles were collected and identified from maize leaves on which aphids had fed. All of the HN-induced volatiles (HNIVs) elicited an electroantennogram (EAG) response from H. axyridis. Of these HNIVs, 1-nonene was attractive to H. axyridis under simulated natural volatilization. Furthermore, our regression showed that the release of 1-nonene was positively correlated with H. axyridis visitation rates. Supplying 1-nonene to maize on which aphids had fed under LN enhanced attractiveness to H. axyridis. These results supported the conclusion that 1-nonene was the active compound that mediated the response to nitrogen in the tritrophic interaction. In addition, the 1-nonene synthesis pathway was hypothesized, and we found that the release of 1-nonene might be related to the presence of salicylic acid (SA) and abscisic acid (ABA). This research contributes to the development of novel environmentally friendly strategies to optimize nitrogen fertilizer application and to improve pest control in maize crops.

Limited effects of plant-beneficial fungi on plant volatile composition and host-choice behavior of Nesidiocoris tenuis.

Biological control using plant-beneficial fungi has gained considerable interest as a sustainable method for pest management, by priming the plant for enhanced defense against pathogens and insect herbivores. However, despite promising outcomes, little is known about how different fungal strains mediate these beneficial effects. In this study, we evaluated whether inoculation of tomato seeds with the plant-beneficial fungi Beauveria bassiana ARSEF 3097, Metarhizium brunneum ARSEF 1095 and Trichoderma harzianum T22 affected the plant's volatile organic compound (VOC) profile and the host-choice behavior of Nesidiocoris tenuis, an emerging pest species in NW-European tomato cultivation, and the related zoophytophagous biocontrol agent Macrolophus pygmaeus. Results indicated that fungal inoculation did not significantly alter the VOC composition of tomato plants. However, in a two-choice cage assay where female insects were given the option to select between control plants and fungus-inoculated plants, N. tenuis preferred control plants over M. brunneum-inoculated plants. Nearly 72% of all N. tenuis individuals tested chose the control treatment. In all other combinations tested, no significant differences were found for none of the insects. We conclude that inoculation of tomato with plant-beneficial fungi had limited effects on plant volatile composition and host-choice behavior of insects. However, the observation that N. tenuis was deterred from the crop when inoculated with M. brunneum and attracted to non-inoculated plants may provide new opportunities for future biocontrol based on a push-pull strategy.

Plant-derived monoterpene S-linalool and β-ocimene generated by CsLIS and CsOCS-SCZ are key chemical cues for attracting parasitoid wasps for suppressing Ectropis obliqua infestation in Camellia sinensis L.

Insect-induced plant volatile organic compounds (VOCs) may function as either direct defence molecules to deter insects or indirect defence signals to attract the natural enemies of the invading insects. Tea (Camellia sinensis L.), an important leaf-based beverage crop, is mainly infested by Ectropis obliqua which causes the most serious damage. Here, we report a mechanistic investigation of tea plant-derived VOCs in an indirect defence mechanism against E. obliqua. Parasitoid wasp Parapanteles hyposidrae, a natural enemy of E. obliqua, showed strong electrophysiological response and selection behaviour towards S-linalool and β-ocimene, two monoterpenes with elevated emission from E. obliqua-damaged tea plants. Larvae frass of E. obliqua, which also released S-linalool and β-ocimene, was found to attract both mated female or male Pa. hyposidrae according to gas chromatography-electroantennogram detection and Y-tube olfactometer assays. In a field setting, both S-linalool and β-ocimene were effective in recruiting both female and male Pa. hyposidrae wasps. To understand the molecular mechanism of monoterpenes-mediated indirect defence in tea plants, two novel monoterpene synthase genes, CsLIS and CsOCS-SCZ, involved in the biosynthesis of S-linalool or β-ocimene, respectively, were identified and biochemically characterised. When the expression of these two genes in tea plants was inhibited by antisense oligodeoxynucleotide, both volatile emission and attraction of wasps were reduced. Furthermore, gene expression analysis suggested that the expression of CsLIS and CsOCS-SCZ is regulated by the jasmonic acid signalling pathway in the tea plant.

Identification of Volatiles in Tomato Fruit Using Headspace Solid-Phase-Micro-Extraction (HS-SPME) Coupled with Gas Chromatography-Mass Spectrometry (GC-MS).

Plant volatile organic compounds (VOCs) are organic chemicals that plants release as part of their natural biological processes. Various plant tissues produce VOCs, including leaves, stems, flowers, and roots. VOCs are essential in plant communication, defense against pests and pathogens, aroma and flavor, and attracting pollinators. The study of plant volatiles has become an increasingly important area of research in recent years, as scientists have recognized these compounds' important roles in plant physiology. As a result, there has been a growing interest in developing methods for collecting and analyzing plant VOCs. HS-SPME-GC-MS (headspace solid-phase microextraction-gas chromatography-mass spectrometry) is commonly used for plant volatile analysis due to its high sensitivity and selectivity. This chapter describes an efficient method for extracting and identifying volatile compounds by HS-SPME coupled with GC-MS in tomato fruits.

Individual and Combined Effects of Predatory Bug Engytatus nicotianae and Trichoderma atroviride in Suppressing the Tomato Potato Psyllid Bactericera cockerelli in Greenhouse Grown Tomatoes

The tomato potato psyllid (TPP) Bactericera cockerelli is a serious pest of the Solanaceae family. The management of this pest using synthetic pesticides is problematic because of the development of pesticide resistance and environmental concerns including impacts on non-target organisms. The predatory bug Engytatus nicotianae has recently been identified as a useful biocontrol agent for TPP in greenhouses. The soil fungus Trichoderma Pers. is commonly used as a plant growth enhancer and biocontrol agent against phytopathogenic fungi. Therefore, there could be advantages associated with the combined use of these biocontrol agents. Some reports in other systems suggest that Trichoderma inoculation may alter the behaviour of pests and their natural enemies by modifying plant defence metabolites such as volatile organic compounds (VOCs). For this reason, this study aimed to investigate the individual and combined efficacy of these biocontrol agents (i.e., Trichoderma atroviride and E. nicotianae) against TPP in greenhouse grown tomatoes (Solanum lycopersicum cv. Merlice). To this end, we compared the effect of each biocontrol agent and their combination on TPP abundance across different developmental stages (egg, nymphs, adults) and the number of infested leaves. We also investigated plant VOC emissions under the different treatments. Across all measured TPP stages, the treatments tested (E. nicotianae alone, T. atrovirdae alone, and T. atrovirdae + E. nicotianae) significantly reduced mean TPP counts relative to the control, and no significant differences were observed in VOC emissions among treatments. Overall, T. atrovirdae alone was less effective than E. nicotianae alone and its combination with T. atrovirdae in suppressing TPP populations. However, the combined use of Trichoderma + E. nicotianae did not show significant advantages over the use of E. nicotianae alone in controlling TPP. Therefore, their combined use needs to be further assessed in light of other advantages of Trichoderma to the crop (e.g., growth promotion or pathogen defence).

Antifungal effects of carvacrol, the main volatile compound in Origanum vulgare L. essential oil, against Aspergillus flavus in postharvest wheat

Plant volatile organic compounds (VOCs) with antimicrobial activity could potentially be extremely useful fumigants to prevent and control the fungal decay of agricultural products postharvest. In this study, antifungal effects of volatile compounds in essential oils extracted from Origanum vulgare L. against Aspergillus flavus growth were investigated using transcriptomic and biochemical analyses. Carvacrol was identified as the major volatile constituent of the Origanum vulgare L. essential oil, accounting for 66.01 % of the total content. The minimum inhibitory concentrations of carvacrol were 0.071 and 0.18 μL/mL in gas-phase fumigation and liquid contact, respectively. Fumigation with 0.60 μL/mL of carvacrol could completely inhibit A. flavus proliferation in wheat grains with 20 % moisture, showing its potential as a biofumigant. Scanning electron microscopy revealed that carvacrol treatment caused morphological deformation of A. flavus mycelia, and the resulting increased electrolyte leakage indicates damage to the plasma membrane. Confocal laser scanning microscopy confirmed that the carvacrol treatment caused a decrease in mitochondrial membrane potential, reactive oxygen species accumulation, and DNA damage. Transcriptome analysis revealed that differentially expressed genes were mainly associated with fatty acid degradation, autophagy, peroxisomes, the tricarboxylic acid cycle, oxidative phosphorylation, and DNA replication in A. flavus mycelia exposed to carvacrol. Biochemical analyses of hydrogen peroxide and superoxide anion content, and catalase, superoxide dismutase, and glutathione S-transferase activities showed that carvacrol induced oxidative stress in A. flavus, which agreed with the transcriptome results. In summary, this study provides an experimental basis for the use of carvacrol as a promising biofumigant for the prevention of A. flavus contamination during postharvest grain storage.

A tug-of-war to control plant emission of an airborne alarm signal

Aphids represent a major threat to crops. Hundreds of different viruses are aphid-borne. Upon aphid attack, plants release volatile organic compounds (VOCs) as airborne alarm signals to turn on the airborne defense (AD) of neighboring plants, thereby repelling aphids as well as reducing aphid fitness and virus transmission. This phenomenon provides a critical community-wide plant protection to fend off aphids, but the underlying molecular basis remains undetermined for a long time. In a recent article, Gong et al. established the NAC2-SAMT1 module as the core component regulating the emission of methyl-salicylate (MeSA), a major component of VOCs in aphid-attacked plants. Furthermore, they showed that SABP2 protein is critical for the perception of volatile MeSA signal by converting MeSA to Salicylic Acid (SA), which is the cue to elicit AD against aphids at the community level. Moreover, they showed that multiple viruses use a conserved glycine residue in the ATP-dependent helicase domain in viral proteins to shuttle NAC2 from the nucleus to the cytoplasm for degradation, leading to the attenuation of MeSA emission and AD. These findings illuminate the functional roles of key regulators in the complex MeSA-mediated airborne defense process and a counter-defense mechanism used by viruses, which has profound significance in advancing the knowledge of plant-pathogen interactions as well as providing potential targets for gene editing-based crop breeding.

Rational design, synthesis and binding mechanisms of novel benzyl geranate derivatives as potential eco-friendly aphid repellents.

The push-pull strategy is considered as a promising eco-friendly method for pest management. Plant volatile organic compounds (PVOCs) act as semiochemicals constitute the key factor in implementing this strategy. Benzyl alcohol and geraniol, as functional PVOCs, were reported to regulate insect behavior, showing the potential application in pest control. Using geraniol as lead, a geraniol derivative 5i with fine repellent activity was discovered in our previous work. In order to explore novel, eco-friendly aphid control agents, a series of benzyl geranate derivatives was designed and synthesized using 5i as the lead and benzyl alcohol as the active fragment. Benzyl alcohol was firstly evaluated to have repellent activity to Acyrthosiphon pisum. Based on this repellent fragment, a series of novel benzyl geranate derivatives was rationally designed and synthesized using a scaffold-hopping strategy. Among them, compound T9, with a binding affinity (Kd = 0.43 μm) and a substantial repellency of 64.7% against A. pisum, is the most promising compound. Molecule docking showed that hydrophobic and hydrogen-bonding interactions substantially influenced the binding affinity of compounds with ApisOBP9. Additionally, T9 exhibited low-toxicity to honeybees and ladybugs. Using a simple scaffold-hopping strategy combined with active fragment benzyl alcohol, a new derivative T9, with high aphid-repellency and low-toxicity to nontarget organisms, can be considered as a novel potential eco-friendly aphid control agent for sustainable agriculture. © 2023 Society of Chemical Industry.

Potential of the endophyte Penicillium commune in the control of olive anthracnose via induction of antifungal volatiles in host plant

Olive anthracnose, caused by several Colletotrichum species, is the most economically harmful fruit disease of the olive crop. This work aimed to evaluate the ability of the endophyte Penicillium commune CIMO 14FM009 to protect the olive tree against Colletotrichum nymphaeae via induction of plant volatile organic compounds (VOCs). Accordingly, olive tree branches were inoculated with the endophyte and one month later with the pathogen. After 0, 3, and 24 h of pathogen inoculation, the volatile composition of leaves and fruits was analyzed by HS-SPME-GC/MS, and compared with controls (branches inoculated with buffer, endophyte, or pathogen). The effect of plant-derived volatiles on C. nymphaeae was also evaluated. Penicillium commune induced the release of VOCs on the olive trees, with the capacity to reduce significantly the growth (up to 1.4-fold) and sporulation (up to 1.2-fold) of C. nymphaeae. This effect was most notorious on olives than on leaves, and occurred 3 h after pathogen-challenge, suggesting the need for a stressful stimulus for the production of antifungal VOCs. The observed inhibition was associated to a specific set of VOCs released from olives (mostly belonging to the alcohols and esters chemical classes) and leaves (mostly belonging to the alkenes). Curiously, a set of VOCs belonging to alkene, alkane and ester classes, were emitted exclusively in olive branches inoculated with C. nymphaeae. These findings provide new possibilities for controlling olive anthracnose using P. commune and/or volatiles, which efficacy should be tested in future works.

The low dose of L-lactic acid as loess soil amendment enhances wheat rhizosphere microecosystem

Loess covers ∼12% of the global continental area. However, soil salinization, due to chemical or physical weathering of rock, overgrazing and inappropriate agricultural practices, etc., threatens the quality of loess and the yield of crops. Exploring sustainable, accessible, and affordable soil amendments is consequently required to upgrade loess soil quality. Herein, L-lactic acid, as the main by-product in the process of biowaste fermentation, has been investigated due to its affordability and accessibility for this purpose. The interactions of L-lactic acid with loess soil and rhizosphere microecosystem were explored to unveil its capacity to ameliorate salinity-related concerns and rejuvenate the agricultural capabilities of the soil. The effects of L-lactic acid addition on soil rhizosphere microorganisms and growth of wheat seedlings were investigated at the L-lactic acid dosages of 0.07–2.99 mg-C/g-soil with determination of the physiological characteristics of wheat seedlings, bulk soil, and rhizosphere microorganisms. The low dose of L-lactic acid (0.07–0.75 mg-C/g-soil) was demonstrated to abate the salt ion content (Na+, K+, and Ca2+) in bulk soil and alleviate the effects of salt stress on plants. By contrast, the addition of a high dose of L-lactic acid (1.49–2.99 mg-C/g-soil) boosted the salt ion content (Na+, K+, and Ca2+) in bulk soil and exacerbated salt stress. In accordance with the K+:Na+ ratio and chlorophyll content indicators of wheat seedlings, a low dose of L-lactic acid enhanced plant photosynthesis and tolerance to salt stress. The L-lactic acid–induced rhizosphere microecosystem established an environment that supported microbial growth with elevated soluble phosphorus levels, which provided the basis for the plant–microbe–soil nutrient cycle and metabolism. Gas chromatography–high resolution Orbitrap mass spectrometry used plant volatile organic compounds (VOC) as the indicators of specific and sensitive signals to reveal that a low dose of L-lactic acid supported the enrichment of VOC in the aerial parts of wheat by heightening plant disease resistance and thereby increasing the aboveground biomass of plants. A high dose of L-lactic acid diminished the K+:Na+ ratio and chlorophyll content in wheat and reduced plant photosynthesis resistance to salt stress; furthermore, the number of disease-resistant VOC available to wheat was curtailed, inhibiting wheat growth. Our study suggests that L-lactic acid added at an appropriate dose can effectively benefit the rhizosphere microecosystem and plant growth, thereby enhancing crop defense and yield. Thus, a soil amendment derived from L-lactic acid demonstrates significant potential in the application for loess soil.

VOCs from fungi-infected apples attract and increase the oviposition of yellow peach moth Conogethes punctiferalis.

Plant volatile organic compounds (VOCs) modified by plant-associated microbes can attract or repel the oviposition of herbivores. Here, we explored the effects of three different fungi on apples' VOCs and the cascading impacts on the oviposition preference of yellow peach moth [YPM, Conogethes punctiferalis (Guenée)]. Among Penicillium crustosum-infected apples (PCA), Rhizopus oryzae-infected apples (ROA), Colletotrichum gloeosporioides-infected apples (CGA) and healthy apples (HA), mated YPM females preferred to oviposit eggs on ROA and CGA, and showed significant attractiveness to VOCs from PCA, ROA, and CGA under laboratory conditions. The VOCs analyses showed that there were significant differences between fungi-infected apples (ROA, CGA) and control treatments (mechanically damaged apples (MDA), HA) in terms of the relative contents of 13 VOCs. The relative contents of ethyl 2-methylbutyrate, ethyl caprylate, estragole, ethyl hexanoate in ROA and CGA were higher than those in MDA. The relative content of isopropyl 2-methylbutyrate in ROA was significantly higher than those in HA and CGA. The relative contents of 2-methylbutyl acetate, butyl 2-methylbutyrate, hexyl 2-methylbutyrate, amyl hexanoate, hexyl hexanoate, (E, E)-α-farnesene in ROA and CGA were lower than those in HA. The relative content of hexyl acetate in ROA and CGA was significantly higher than that in MDA, but lower than that in HA. Additionally, 10 fungi-induced VOCs were detected in ROA and/or CGA. When 20 VOCs from ROA and/or CGA were tested as individuals or mixed blends in Y-tube olfactometer assays, mated YPM females preferred amyl 2-methylbutyrate, isoamyl 2-methylbutyrate, isopropyl 2-methylbutyrate, hexyl propionate (common VOCs in ROA, CGA, and HA), and heptacosane (a fungi-induced VOC in ROA), but no significant preferences were observed between individual compounds and mixed blends, except for hexyl propionate. Different fungi infection increased the relative contents of common VOCs from healthy and fungi-infected apples, which ultimately resulted in the significant attractiveness for the oviposition of mated YPM females. This study clarified why fungi-infected apples were more attractive to YPMs than healthy apples and screened out the crucial VOCs for YPM oviposition. © 2023 Society of Chemical Industry.

Selection Behavior and OBP-Transcription Response of Western Flower Thrips, Frankliniella occidentalis, to Six Plant VOCs from Kidney Beans.

Plant volatile organic compounds (VOCs) are an important link that mediates chemical communication between plants and plants, plants and insects, and plants and natural enemies of insect pests. In this study, we tested the response in the selective behavior of western flower thrips, Frankliniella occidentalis, to the VOCs of kidney bean, Phaseolus vulgaris L., to explore their "attraction" or "repellent" effects regarding their application in integrated pest management (i.e., IPM). The results indicated that 12.7 μL/mL (E, E, E, E)-squalene, 3.2 μL/mL dioctyl phthalate, and 82.2 μL/mL ethyl benzene had a significantly attractive effect on the selective behavior of F. occidentalis, while 10.7 μL/mL and 21.4 μL/mL 2,6-ditert-butyl-4-methyl phenol had a significantly repulsive effect on the selective behavior of F. occidentalis, showing that F. occidentalis responds differently to specific concentrations of VOCs from P. vulgaris plant emissions. Interestingly, the three compounds with the specific above concentrations, after being mixed in pairs, significantly attracted F. occidentalis compared to the control treatment; however, the mixture with the three above compounds had no significant different effect on F. occidentalis compared to the control treatment. It can be seen that the effect with the mixtures of three kinds of VOCs had the same function and may not get better. Simultaneously, the reasons for this result from the transcription levels of odorant-binding protein genes (OBPs) were determined. There were differences in the types and transcription levels of OBPs, which played a major role in the host selection behavior of F. occidentalis under the mixed treatment of different VOCs. It is presumed that there are specific VOCs from P. vulgaris plants that have a good repellent or attracting effect on the selective behavior of F. occidentalis, which can be used for the development of plant-derived insect attractants and repellents to serve as IPM in fields. But attention should be paid to the antagonism between plant-derived preparations and VOCs produced by plants themselves after application.

Identification of an odorant receptor responding to sex pheromones in Spodoptera frugiperda extends the novel type-I PR lineage in moths.

In moths, pheromone receptors (PRs) are crucial for intraspecific sexual communication between males and females. Moth PRs are considered as an ideal model for studying the evolution of insect PRs, and a large number of PRs have been identified and functionally characterized in different moth species. Moth PRs were initially thought to fall into a single monophyletic clade in the odorant receptor (OR) family, but recent studies have shown that ORs in another lineage also bind type-I sex pheromones, which indicates that type-I PRs have multiple independent origins in the Lepidoptera. In this study, we investigated whether ORs of the pest moth Spodoptera frugiperda belonging to clades closely related to this novel PR lineage may also have the capacity to bind type-I pheromones and serve as male PRs. Among the 7 ORs tested, only 1 (SfruOR23) exhibited a male-biased expression pattern. Importantly, invitro functional characterization showed that SfruOR23 could bind several type-I sex pheromone compounds with Z-9-tetradecenal (Z9-14:Ald), a minor component found in female sex pheromone glands, as the optimal ligand. In addition, SfruOR23 also showed weak responses to plant volatile organic compounds. Altogether, we characterized an S.frugiperda PR positioned in a lineage closely related to the novel PR clade, indicating that the type-I PR lineage can be extended in moths.

Volatile compound-mediated plant-plant interactions under stress with the tea plant as a model.

Plants respond to environmental stimuli via the release of volatile organic compounds (VOCs), and neighboring plants constantly monitor and respond to these VOCs with great sensitivity and discrimination. This sensing can trigger increased plant fitness and reduce future plant damage through the priming of their own defenses. The defense mechanism in neighboring plants can either be induced by activation of the regulatory or transcriptional machinery, or it can be delayed by the absorption and storage of VOCs for the generation of an appropriate response later. Despite much research, many key questions remain on the role of VOCs in interplant communication and plant fitness. Here we review recent research on the VOCs induced by biotic (i.e. insects and pathogens) and abiotic (i.e. cold, drought, and salt) stresses, and elucidate the biosynthesis of stress-induced VOCs in tea plants. Our focus is on the role of stress-induced VOCs in complex ecological environments. Particularly, the roles of VOCs under abiotic stress are highlighted. Finally, we discuss pertinent questions and future research directions for advancing our understanding of plant interactions via VOCs.

Plant volatiles and priority effects interactively determined initial community assembly of arthropods on multiple willow species.

Plant traits, which are often species specific, can serve as environmental filtering for community assembly on plants. At the same time, the species identity of the initially colonizing arthropods would vary between plant individuals, which would subsequently influence colonizing arthropods and community development in the later stages. However, it remains unclear whether interindividual divergence due to priority effects is equally important as plant trait-specific environmental filtering in the initial stages. In this study, we propose that plant volatile organic compounds (PVOCs) may play a crucial role as an environmental filter in the initial stages of community assembly, which can prevent the community assembly process from being purely stochastic. To test this hypothesis, we conducted short term but highly frequent monitoring (19 observations over 9 days) of arthropod community assembly on intact individuals of six willow species in a common garden. PVOC compositions were analyzed before starting the experiment and compared with arthropod compositions occurring on Days 1-2 of the experiment (earliest colonizer community) and those occurring on Days 8-9 of the experiment (subsequent colonizer community). Unintentionally, deer herbivory also occurred at night of Day 2. Distance-based statistics demonstrated that PVOC compositions were plant species specific, but neither the earliest colonizer nor the subsequent colonizer community composition could be explained by plant species identity. Rather, Procrustes analysis showed that both the PVOC composition and that of the earliest colonizer community can be used to explain the subsequent colonizer community. In addition, the linkage between PVOCs and the subsequent colonizer community was stronger on individuals with deer herbivory. These findings indicate that PVOCs have widespread effects on initial community assembly, as well as priority effects brought on by stochastic immigration, and that plant species identity only has weak and indirect effects on the actual composition of the community.

Inter-laboratory comparison of plant volatile analyses in the light of intra-specific chemodiversity

IntroductionAssessing intraspecific variation in plant volatile organic compounds (VOCs) involves pitfalls that may bias biological interpretation, particularly when several laboratories collaborate on joint projects. Comparative, inter-laboratory ring trials can inform on the reproducibility of such analyses.ObjectivesIn a ring trial involving five laboratories, we investigated the reproducibility of VOC collections with polydimethylsiloxane (PDMS) and analyses by thermal desorption-gas chromatography-mass spectrometry (TD-GC-MS). As model plant we used Tanacetum vulgare, which shows a remarkable diversity in terpenoids, forming so-called chemotypes. We performed our ring-trial with two chemotypes to examine the sources of technical variation in plant VOC measurements during pre-analytical, analytical, and post-analytical steps.MethodsMonoclonal root cuttings were generated in one laboratory and distributed to five laboratories, in which plants were grown under laboratory-specific conditions. VOCs were collected on PDMS tubes from all plants before and after a jasmonic acid (JA) treatment. Thereafter, each laboratory (donors) sent a subset of tubes to four of the other laboratories (recipients), which performed TD-GC-MS with their own established procedures.ResultsChemotype-specific differences in VOC profiles were detected but with an overall high variation both across donor and recipient laboratories. JA-induced changes in VOC profiles were not reproducible. Laboratory-specific growth conditions led to phenotypic variation that affected the resulting VOC profiles.ConclusionOur ring trial shows that despite large efforts to standardise each VOC measurement step, the outcomes differed both qualitatively and quantitatively. Our results reveal sources of variation in plant VOC research and may help to avoid systematic errors in similar experiments.
