Integrated omics approach reveals the molecular pathways activated in tomato by Kocuria rhizophila, a soil plant growth-promoting bacterium

Plant microbial biostimulants application has become a promising and eco-friendly agricultural strategy to improve crop yields, reducing chemical inputs for more sustainable cropping systems. The soil dwelling bacterium Kocuria rhizophila was previously characterized as Plant Growth Promoting Bacteria (PGPB) for its multiple PGP traits, such as indole-3-acetic acid production, phosphate solubilization capability and salt and drought stress tolerance. Here, we evaluated by a multi-omics approach, the PGP activity of K. rhizophila on tomato, revealing the molecular pathways by which it promotes plant growth. Transcriptomic analysis showed several up-regulated genes mainly related to amino acid metabolism, cell wall organization, lipid and secondary metabolism, together with a modulation in the DNA methylation profile, after PGPB inoculation. In agreement, proteins involved in photosynthesis, cell division, and plant growth were highly accumulated by K. rhizophila. Furthermore, “amino acid and peptides”, “monosaccharides”, and “TCA” classes of metabolites resulted the most affected by PGPB treatment, as well as dopamine, a catecholamine neurotransmitter mediating plant growth through S-adenosylmethionine decarboxylase (SAMDC), a gene enhancing the vegetative growth, up-regulated in tomato by K. rhizophila treatment. Interestingly, eight gene modules well correlated with differentially accumulated proteins (DAPs) and metabolites (DAMs), among which two modules showed the highest correlation with nine proteins, including a nucleoside diphosphate kinase, and cytosolic ascorbate peroxidase, as well as with several amino acids and metabolites involved in TCA cycle. Overall, our findings highlighted that sugars and amino acids, energy regulators, involved in tomato plant growth, were strongly modulated by the K. rhizophila-plant interaction.

The effects of plant growth-promoting bacteria on seed germination and seedling growth in Crassocephalum rubens

Crassocephalum rubens, a herb with potential anticancer properties, encounters challenges in production efficiency due to small seed size and elevated temperatures in cultivation areas. Our study investigates the effects of seed pelleting and plant growth-promoting bacteria (PGPB) on germination and seedling growth across temperature variations in Taiwan. Pelleted seeds demonstrate superior germination percentages linked to enhanced seed quality. Elevated temperature, particularly at 30/25 °C, enhanced germination performance, with the highest final germination percentage observed in pelleted seeds treated with water. Unexpectedly, PGPB strains— Klebsiella sp. KsGRB10 and Bacillus sp. BsGFB04—exhibited limited impact on germination rates, showing a marginal increase in seedling growth under 25/20 °C and 30/25 °C, respectively. Air temperature fluctuations influenced seedling vigor, leaf color, and physiological parameters. Remarkably, inoculation with BsGFB04 and KsGRB10 enhanced C. rubens’ tolerance to high-temperature stress conditions. Diurnal measurements in week 4, under 25/20 °C, reveal that PGPB inoculation decreased stomatal conductance and transpiration rate while maintaining the quantum yield of PSII, indicating potentially improved water-use efficiency. This study provides crucial insights into the interplay among PGPB, environmental stress, and the physiology of a wild species, paving the way for further research in the domestication of C. rubens for medicinal herb mass production.

Effect of Mineral Fertilization and Microbial Inoculation on Cabbage Yield and Nutrition: A Field Experiment

Cabbage serves as an important food and nutrition source for numerous communities in the world, yet its production requires substantial quantities of chemical fertilizers. In this study, we assessed the impact of both increasing nitrogen and phosphorus mineral (NP) fertilization, along with the application of plant growth-promoting bacteria (PGPB) on the N and P uptake, quality, and yield of cabbage. To this end, we conducted two consecutive field experiments following a randomized block design with four replicates and two factors: NP doses and PGPB inoculation. PGPB inoculation used a bacterial consortium comprising Azospirillum brasilense D7, Herbaspirillum sp. AP21, and Rhizobium leguminosarum T88. Our results showed a significant influence of both biofertilization and NP fertilization across both crop cycles; however, no interaction between these factors was observed. In the first crop cycle, 75% of NP mineral fertilization (equivalent to 93.6 kg ha−1 of N and 82.1 kg ha−1 of P) positively impacted yield and N uptake. Also, microbial inoculation significantly influenced crop yield, resulting in a 9-ton increase in crop yield per hectare due to biofertilization. In the second crop cycle, we observed a significant positive effect of mineral fertilization on cabbage yield and nutritional quality. The relative agronomic effectiveness (RAE) index showed that combining biological fertilization with 50% and 75% of the NP fertilization, respectively, increased yield by 66% and 48% compared to the commercial NP dosage without PGPB. Collectively, our results demonstrated that within our experimental setup, NP fertilization dosage can be reduced without any detrimental impact on yield. Moreover, biofertilization could enhance cabbage quality and yield in field conditions.

Unravelling the Combined Use of Soil and Microbial Technologies to Optimize Cultivation of Halophyte Limonium algarvense (Plumbaginaceae) Using Saline Soils and Water

Salt-affected soils have detrimental effects on agriculture and ecosystems. However, these soils can still be used for halophyte (salt-tolerant plants) cultivation using brackish and/or saline water. In this study, we employed soil technologies and mutualistic microorganisms as a sustainable strategy to improve the growth and reproduction of the halophyte Limonium algarvense Erben’s growth and reproduction under saline conditions. A microcosm assay was conducted under controlled greenhouse conditions to cultivate L. algarvense using a saline Fluvisol (FLU) amended—or not—with a Technosol (TEC). Plants were inoculated with the arbuscular mycorrhizal fungus (AMF) Rhizoglomus irregulare and/or a consortium of plant growth-promoting bacteria (PGPB), and they were irrigated with estuarine water. Soil enzyme analysis and physicochemical characterisation of the soils, collected at the beginning and at the end of the assay, were carried out. The physiological status of non-inoculated and inoculated plants was monitored during the assay for 4 months, and AMF root colonisation was evaluated. In FLU, only plants inoculated with the AMF survived. These plants had lower number of leaves, and shoot and root dry biomass than the ones grown in the TEC by the end of the assay. In the TEC, PGPB inoculation led to higher NDVI and PRI values, and AMF inoculation promoted higher reproductive development but not pollen fertility. The findings show that the combined use of soil and microbial technologies can be successfully applied to cultivate L. algarvense, suggesting their generalized use for other Limonium species with economic interest, while contributing to the sustainable use of marginal lands.

Improving Salicornia ramosissima photochemical and biochemical resilience to extreme heatwaves through rhizosphere engineering with Plant Growth-Promoting Bacteria

The anticipated rise in the length, frequency, and intensity of heatwaves (HW) in the Mediterranean region poses a danger to the crops, as these brief but high-intensity thermal stress events halt plant productivity. This arises the need to develop new eco-friendly sustainable strategies to overcome food demand. Halophytes such as Salicornia ramosissima appear as cash crop candidates, alongside with new biofertilization approaches using Plant Growth Promoting Bacteria (PGPB). In the present work, S. ramosissima plants exposed to heatwave (HW) treatments with and without marine PGPB inoculation is studied to evaluate the physiological responses behind eventual thermal adaptation conditions. Plants exposed to HW inoculated with ACC deaminase and IAA-producing PGPB showed a 50% reduction in the photochemical energy dissipation, when compared to their non-inoculated counterparts, indicating higher light-use efficiency. The observed concomitant increase (76–234%) in several pigments indicates improved inoculated HW-exposed individuals' light harvesting and photoprotection under stressful conditions. This reduction of the physiological stress levels in inoculated plants was also evident by the significant reduction of several antioxidant enzymes as well as of membrane lipid peroxidation products. Additionally, improved membrane stability could also be observed, through the regulation of fatty acid unsaturation levels, decreasing the excessive fluidity imposed by HW treatment. All these improved physiological traits associated with specific PGP traits highlight a key potential of the use of these PGPB consortiums as biofertilizers for S. ramosissima cash crop production in the Mediterranean, where increasing frequency in HW-events is a major drawback to plant production, even to warm-climate plants.

Genome‐wide association analysis of hyperspectral reflectance data to dissect the genetic architecture of growth‐related traits in maize under plant growth‐promoting bacteria inoculation

Plant growth-promoting bacteria (PGPB) may be of use for increasing crop yield and plant resilience to biotic and abiotic stressors. Using hyperspectral reflectance data to assess growth-related traits may shed light on the underlying genetics as such data can help assess biochemical and physiological traits. This study aimed to integrate hyperspectral reflectance data with genome-wide association analyses to examine maize growth-related traits under PGPB inoculation. A total of 360 inbred maize lines with 13,826 single nucleotide polymorphisms (SNPs) were evaluated with and without PGPB inoculation; 150 hyperspectral wavelength reflectances at 386-1021 nm and 131 hyperspectral indices were used in the analysis. Plant height, stalk diameter, and shoot dry mass were measured manually. Overall, hyperspectral signatures produced similar or higher genomic heritability estimates than those of manually measured phenotypes, and they were genetically correlated with manually measured phenotypes. Furthermore, several hyperspectral reflectance values and spectral indices were identified by genome-wide association analysis as potential markers for growth-related traits under PGPB inoculation. Eight SNPs were detected, which were commonly associated with manually measured and hyperspectral phenotypes. Different genomic regions were found for plant growth and hyperspectral phenotypes between with and without PGPB inoculation. Moreover, the hyperspectral phenotypes were associated with genes previously reported as candidates for nitrogen uptake efficiency, tolerance to abiotic stressors, and kernel size. In addition, a Shiny web application was developed to explore multiphenotype genome-wide association results interactively. Taken together, our results demonstrate the usefulness of hyperspectral-based phenotyping for studying maize growth-related traits in response to PGPB inoculation.

Seed priming with plant growth-promoting bacteria (PGPB) improves growth and water stress tolerance of Secalemontanum

Abiotic and biotic stresses are major global threats to food security in the 21st century. Application of plant growth-promoting bacteria (PGPB) in rangeland plants is the only possible alternative that supports plant growth and development to combat environmental stress and successfully restoring rangelands. PGPBs were also found to be a potential substitute for chemical fertilizers and pesticides. The challenge is to determine which biofertilizers can be used for Secale montanum in normal and under water stress conditions. We sought to determine the benefits of PGPB for S. montanum under water stress conditions in terms of seedling growth traits, growth indicators, and nutrient uptake in the research greenhouse. Therefore, a completely randomized factorial design was conducted with two treatments of PGPB inoculation, including the control (no PGPB inoculation), PGPBs Bacillus cereus, Pseudomonas aeruginosa, Azospirillum lipoferm, and Azotobacter chroococcum, and water stress in the research greenhouse. Overall, the results of the current study showed that water stress greatly reduced the above-ground fresh weight of above-ground plant parts and the nitrogen and potassium content of S. montanum. The present study confirms the positive effects of PGPB on fresh and dry weights of above- and below-ground parts and seedling, vigor index, quality index, and nitrogen and potassium content of S. montanum, except for below-ground parts length, compared with the controls, which shows that PGPB usually improves some indicators of plant growth and development. We suggest that restoration of S. montanum seed inoculation with PGPB should be supported in degraded rangelands and marginal drylands in low rainfall years, which may cause water scarcity and consequently water stress in arid and semi-arid regions.

Effective drought mitigation by rhizobacteria consortium in wheat field trials

To evaluate the drought mitigation on the yield and morpho-physiological traits of wheat by plant growth-promoting bacteria (PGPB), a two-year factorial split-plot field experiment was conducted based on a randomized complete block design. The factors were three levels of normal, moderate, and severe stress, and two levels of control and inoculated bacteria (Azotobacter, Azospirillum, Microbacterium, and Pseudomonas). The results indicated that drought stress negatively affected the morpho-physiological traits of wheat. An increase in oxidative stress and lipid peroxidation reduced wheat chlorophyll content and grain yield (GY) by 18% and 49%, respectively. Moreover, wheat root system traits, including root biomass, volume, surface area, and length density decreased significantly under drought stress. The inoculation of PGPB significantly increased the biomass, volume, and surface area of wheat roots by 48%, 31%, and 18.5%, respectively. An improved root system in the inoculated treatment with an increase in the relative leaf water content and a decrease in oxidative stress could increase leaf chlorophyll content. The PGPB inoculation significantly elevated wheat GY under normal and stress conditions, with the highest yield increase at the moderate stress level (12%). At the high stress level, however, bacterial inoculation did not significantly influence wheat yield. Altogether, the inoculation of bacteria with various growth-stimulating traits improved the morphological, physiological, and metabolic traits of wheat under drought stress, ultimately increasing wheat GY in real field conditions.

Compost and plant growth-promoting bacteria enhanced steviol glycoside synthesis in stevia (Stevia rebaudiana Bertoni) plants by improving soil quality and regulating nitrogen uptake

Stevia ( Stevia rebaudiana Bertoni) is an important industrial crop, and its leaves contain steviol glycosides (SGs), which are valuable compounds because they are characterized by a high sweetness and low caloric content. SGs yield depends on nitrogen (N) nutrition; however, the role of N uptake in SGs synthesis needs clarification, especially in organic farming systems that depend on organic and biofertilizers. Plant growth-promoting bacteria (PGPB; i.e., Azospirillum brasilense and Bacillus circulans ) and compost were used to study the effect of N nutrition on SGs yield. The experiment consisted of five N treatments with or without PGPB inoculation. The fertilization treatments were: C = without N, T 1 = urea, T 2 = compost, T 3 = urea and compost (75% of N from urea and the rest from compost) and T 4 = urea and compost (50% of N from urea and the rest 50% from compost). Compost and PGPB increased the soil quality indicators and enhanced the synthesis of chlorophyll and nutrient uptake by stevia plants. Mixtures of compost and urea under inoculation with PGPB enhanced soil microbes and enzymatic activity. The highest significant value of nitrogen use efficiency (NUE) resulted from urea alone, while the maximum phosphorus (P) and potassium (K) uptake came from compost. Although urea led to an increase in stevia N uptake, it led to a clear decrease in SGs yield. Urea decreased the total soluble sugar and leaf C/N ratio by 29% and 31%, respectively, compared to those of C. T 4 increased the SGs concentration by 74% and 46%, respectively, compared to that of urea alone in the presence and absence of PGPB. PGPB enhanced the soil quality and promoted nutrient uptake, resulting in significant increases in SGs yield. Regulating N uptake enhanced synthesis of soluble sugars and increased the C/N ratio, which caused an increase in SGs production. The use of compost and plant growth-promoting bacteria is a suitable strategy to produce stevia plants with low rates of inorganic nitrogen without any yield loss. • Compost and plant growth-promoting bacteria (PGPB) enhances stevia growth. • Compost and PGPB reregulate N-uptake in stevia plants. • Urea decreased the total soluble sugar and leaf C/N ratio. • Regulating N-uptake increased the synthesis steviol glycosides (SGs). • The findings might be an aid for sustainable N management in stevia production.

Optimistic contributions of plant growth-promoting bacteria for sustainable agriculture and climate stress alleviation

Global climate change is the major cause of abiotic and biotic stresses that have adverse effects on agricultural productivity to an irreversible level, thus threatening to limit gains in production and imperil sustainable agriculture. These climate change-induced abiotic stresses, especially saline, drought, extreme temperature, and so on affect plant morphological, physiological, biochemical, and metabolic characteristics through various pathways and mechanisms, ultimately hindering plant growth, development, and productivity. However, overuse and other inappropriate uses of agrochemicals are not conducive to the protection of natural resources and the environment, thus hampering sustainable agricultural development. With the vigorous development of modern agriculture, the application of plant growth-promoting bacteria (PGPB) can better ensure sustainable agriculture, due to their ability to improve soil properties and confer stress tolerance in plants. This review deciphered the underlying mechanisms of PGPB involved in enhancing plant stress tolerance and performance under various abiotic and biotic stresses. Moreover, the recent advancements in PGPB inoculation techniques, the commercialization of PGPB-based technology and the current applications of PGPB in sustainable agriculture were extensively discussed. Finally, an outlook on the future directions of microbe-aided agriculture was pointed out. Providing insights into plant-PGPB interactions under biotic and abiotic stresses and offering evidence and strategies for PGPB better commercialization and implementation can inspire the development of innovative solutions exploiting PGPB under climatological conditions.

Mechanistic Insights of Plant Growth Promoting Bacteria Mediated Drought and Salt Stress Tolerance in Plants for Sustainable Agriculture.

Climate change has devastating effects on plant growth and yield. During ontogenesis, plants are subjected to a variety of abiotic stresses, including drought and salinity, affecting the crop loss (20–50%) and making them vulnerable in terms of survival. These stresses lead to the excessive production of reactive oxygen species (ROS) that damage nucleic acid, proteins, and lipids. Plant growth-promoting bacteria (PGPB) have remarkable capabilities in combating drought and salinity stress and improving plant growth, which enhances the crop productivity and contributes to food security. PGPB inoculation under abiotic stresses promotes plant growth through several modes of actions, such as the production of phytohormones, 1-aminocyclopropane-1-carboxylic acid deaminase, exopolysaccharide, siderophore, hydrogen cyanide, extracellular polymeric substances, volatile organic compounds, modulate antioxidants defense machinery, and abscisic acid, thereby preventing oxidative stress. These bacteria also provide osmotic balance; maintain ion homeostasis; and induce drought and salt-responsive genes, metabolic reprogramming, provide transcriptional changes in ion transporter genes, etc. Therefore, in this review, we summarize the effects of PGPB on drought and salinity stress to mitigate its detrimental effects. Furthermore, we also discuss the mechanistic insights of PGPB towards drought and salinity stress tolerance for sustainable agriculture.

Plant growth promoting bacteria improve growth and phytostabilization potential of Zea mays under chromium and drought stress by altering photosynthetic and antioxidant responses

Drought in heavy metal polluted arid and semiarid regions severely inhibits the plant growth and phytoremediation potential by affecting photosynthesis, antioxidant defense mechanism, and other biochemical processes. In the present study, we explored the role of plant growth-promoting bacteria (PGPB) on Zea mays growth and phytoremediation efficiency in Chromium (Cr) contaminated soils under drought stress by assessing plant stress tolerance, photosynthetic gas exchange activities, chlorophyll fluorescence, and Cr accumulation. Two efficient Cr and drought resistant PGPB with the potential to reduce Cr(VI) to Cr(III) and produce plant growth-promoting metabolites even under Cr, drought, or Cr+drought stress conditions were isolated and identified as Providencia sp. (TCR05) and Proteus mirabilis (TCR20). In pot experiments, the inoculation of TCR05 and TCR20 increased the plant growth, pigments, protein, phenolics, and relative water content and decreased the lipid peroxidation, proline, and superoxide dismutase activity under Cr, drought, or Cr+drought conditions. Irrespective of stress treatment, TCR05 and TCR20 also improved plant photosynthetic efficiency by increasing the CO 2 assimilation rate, stomatal conductance to water vapor, transpiration rate, maximum quantum efficiency of PSII, actual quantum efficiency of PSII, electron transport rate, photochemical quenching, reducing the internal CO 2 concentration and non-photochemical quenching. Besides, the PGPB decreased the translocation of Cr through immobilization of Cr in root. These results denoted that strains TCR05 and TCR20 could be a capable bio-inoculant for improving plant growth and phytostabilization practices in Cr contaminated sites even under water-limited conditions. • Potential Cr(VI) reducing MST-PGPB strains TCR05 and TCR20 were isolated. • MST-PGPB enhanced stress tolerance of Z. mays under combined stress conditions. • PGPB inoculation improved the PSII performance of Z. mays under combined stresses. • PGPB inoculation improved Cr phytostabilization under combined stress conditions.

Application of PGPB Combined with Variable N Doses Affects Growth, Yield-Related Traits, N-Fertilizer Efficiency and Nutritional Status of Lettuce Grown under Controlled Condition

Nitrogen (N) fertilization is a crucial agricultural practice for boosting production traits in vegetables. However, N synthetic fertilizers—commonly adopted by farmers—have several counterproductive effects on the environment and on humans. The research was performed to assess the combined influence of plant growth promoting bacteria (PGPB) (Azospirillum brasilense DSM 1690, A. brasilense DSM 2298 and Pseudomonas sp. DSM 25356) and various N fertilization doses (0, 30, 60 or 120 kg ha−1) on growth, yield, quality and nitrogen indices of lettuce in protected cultivation. Plant height, root collar diameter, number of leaves and fresh weight were enhanced by A. brasilense DSM 2298 inoculation and N at 30 or 60 kg ha−1. Overall, soluble solids content (SSC), ascorbic acid, total phenolics, carotenoids, total chlorophyll and total sugars were augmented by the combined effect of A. brasilense strains and 30, 60 or 120 kg N ha−1. Furthermore, PGBP inoculation improved potassium (K) and magnesium (Mg) concentrations in leaf tissues. PGPB inoculation increased N leaf concentration; however, it hastened N indices. These results suggest that the PGPB tested can be considered an eco-friendly tool to improve lettuce yield, particularly when combined with N at 30 or 60 kg ha−1.

A low‐cost greenhouse‐based high‐throughput phenotyping platform for genetic studies: A case study in maize under inoculation with plant growth‐promoting bacteria

AbstractGreenhouse‐based high‐throughput phenotyping (HTP) presents a useful approach for studying novel plant growth‐promoting bacteria (PGPB). Despite the potential of this approach to leverage genetic variability for breeding new maize (Zea Mays L.) cultivars exhibiting highly stable symbiosis with PGPB, greenhouse‐based HTP platforms are not yet widely used because they are highly expensive; hence, it is challenging to perform HTP studies under a limited budget. In this study, we built a low‐cost greenhouse‐based HTP platform to collect growth‐related image‐derived phenotypes. We assessed 360 inbred maize lines with or without PGPB inoculation under nitrogen‐limited conditions. Plant height, canopy coverage, and canopy volume obtained from photogrammetry were evaluated five times during early maize development. A plant biomass index was constructed as a function of plant height and canopy coverage. Inoculation with PGPB promoted plant growth in early developmental stages. Phenotypic correlations between the image‐derived phenotypes and manual measurements were at least 0.47 in the later stages of plant development. The genomic heritability estimates of the image‐derived phenotypes ranged from 0.23 to 0.54. Moderate‐to‐strong genomic correlations between the plant biomass index and shoot dry mass (0.24–0.47) and between HTP‐based plant height and manually measured plant height (0.55–0.68) across the developmental stages showed the utility of our HTP platform. Collectively, our results demonstrate the usefulness of the low‐cost HTP platform for large‐scale genetic and management studies to capture plant growth.

Endophytic PGPB Improves Plant Growth and Quality, and Modulates the Bacterial Community of an Intercropping System

The intercropping of ryegrass and red clover constitutes a sustainable alternative to mitigate the adverse effects of intensive livestock production on grassland degradation by increasing forage yield and quality. The implementation of biofertilization technologies has been widely used to improve soil nutritional properties, and therefore has the potential to ensure the success of this multicrop system. To determine the impact of bioaugmentation on forage growth and quality, as well as the associate changes in the rhizosphere bacterial community, we evaluated the inoculation with two plant growth-promoting bacteria (PGPB) under reduced nitrogen usage. Overall, Herbaspirillum sp. AP21 had a larger effect than Azospirillum brasilense D7 on plant growth. Inoculation with Herbaspirillum sp. AP21 together with 50% of the required nitrogen rate increased shoot dry weight, crude protein, and shoot nitrogen content, and decreased the amount of neutral detergent fiber. PGPB inoculation changed the rhizosphere bacterial community structure, which associated with forage growth and quality. We conclude that PGPB inoculation has the potential to improve the growth of the ryegrass-red clover system, decreasing the requirements for nitrogen fertilization.

Photosynthetic responses of durum wheat to chemical/microbiological fertilization management under salt and drought stresses

The current research was carried out to evaluate the stress tolerance potential of durum wheat plants, in response to the inoculation of native plant growth-promoting bacteria (PGPB), through assessing PSII photochemistry and photosynthetic traits, as well as grain yield and plant height, and to investigate the possibility of using PGPB as a sustainable alternative or in combination with traditional fertilization plans. A greenhouse experiment included chemical/microbiological fertilization and stress (salinity and drought) treatments. The results indicated that the application of bacterial consortium of four PGPB markedly augmented some biochemical and functional traits in photosystem II, such as effective quantum yield of PSII photochemistry (Y(II)), electron transport rate of PSII (ETR), photosynthesis capacity, transpiration rate and stomatal conductance in unstressed plants, and prevented severe changes in the mentioned traits under drought and salinity conditions. The application of PGPB contributed to enhanced grain yield, too. Furthermore, a better performance of the PGPB inoculation was found in combination with half-dose of the recommended chemical fertilizers. In conclusion, PGPB inoculants maintain or improve the photosynthesis efficiency of durum wheat, grain yield and plant height, particularly under stress conditions, and can help to minimize the consumption of chemical fertilizers.

The adaptive metabolomic profile and functional activity of tomato rhizosphere are revealed upon PGPB inoculation under saline stress

Salinization of agricultural lands is of increasing concern at global scale due to its negative impacts on several physiological and developmental processes of most crops, that in turn can lead to threatening food security. Salt-tolerant plant growth-promoting bacteria (PGPB) represent an emerging strategy to protect plants and enhance the productivity of agroecosystems suffering from salinization. This study aimed at investigating the individual effect of two PGPB in alleviating salt-stressed tomato plants, as well as the rhizosphere functioning and metabolomic profiles under soil salinity and PGPB inoculation treatments. The Biolog Eco-Plate functional analysis and untargeted metabolomics approaches were used to this aim. Under saline conditions, plants inoculated with Pseudomonas 16S showed higher biomass (p < 0.05) than both uninoculated and Enterobacter 15S-inoculated plants, indicating that the former strain was efficient in alleviate the saline stress. Moreover, independently from PGPB inoculation, the microbial metabolism in tomato’s rhizosphere was significantly increased by salinity, especially related to the catabolism of amines, carbohydrates, and polymers. The UHPLC/QTOF-MS approach allowed the identification of signature metabolomic patterns for the rhizosphere of tomato, with inflected accumulation of compounds imposed by either Pseudomonas 16S, Enterobacter 15S or saline stress. Crucial compounds from primary and secondary metabolism related to salinity stress or PGPB inoculation were detected, including low molecular weight osmolytes (e.g., amino acids), as well as anthocyanins, hydroxycinnamic acids, coumarins and saponins. An increase in the content of ROS-scavenging and antioxidant compounds in addition to the facilitation of Fe acquisition induced by Pseudomonas 16S, are suggested as mechanisms responsible for the higher biomass accumulation by tomato plants grown under saline stress.

A Multifactorial Approach to Untangle Graphene Oxide (GO) Nanosheets Effects on Plants: Plant Growth-Promoting Bacteria Inoculation, Bacterial Survival, and Drought.

Drought is a limiting factor for agricultural productivity. Climate change threatens to expand the areas of the globe subjected to drought, as well as to increase the severity and duration of water shortage. Plant growth-promoting bacteria (PGPB) are widely studied and applied as biostimulants to increase plant production and to enhance tolerance to abiotic and biotic constraints. Besides PGPB, studies on the potential of nanoparticles to be used as biostimulants are also thriving. However, many studies report toxicity of tested nanoparticles in bacteria and plants in laboratory conditions, but few studies have reported effects of nanoparticles towards bacterial cells and communities in the soil. The combined application of nanoparticles and PGPB as biostimulant formulations are poorly explored and it is important to unravel the potentialities of their combined application as a way to potentiate food production. In this study, Rhizobium sp. E20-8 and graphene oxide (GO) nanosheets were applied on container-grown maize seedlings in watered and drought conditions. Bacterial survival, seedling growth (dry weight), and biochemical endpoints (photosynthetic pigments, soluble and insoluble carbohydrates, proline, lipid peroxidation, protein, electron transport system, and superoxide dismutase) were evaluated. Results showed that the simultaneous exposure to GO and Rhizobium sp. E20-8 was able to alleviate the stress induced by drought on maize seedlings through osmotic and antioxidant protection by GO and mitigation of GO effects on the plant’s biochemistry by Rhizobium sp. E20-8. These results constitute a new lead on the development of biostimulant formulations to improve plant performance and increase food production in water-limited conditions.

Sugarcane pre-sprouted seedlings produced with beneficial bacteria and arbuscular mycorrhizal fungi

ABSTRACT The demand for the increased productivity of sugarcane crops has required changes in its production chain, such as the use of pre-sprouted seedlings (PSS) of sugarcane into the chain. In this system, the inoculation of beneficial microorganisms, such as plant growth-promoting bacteria (PGPB) and arbuscular mycorrhizal fungi (AMF), can improve seedling development. The objective was to evaluate the beneficial effect of PGPB and AMF inoculation on sugarcane PSS production system. Experiments were carried out in greenhouse using a commercial substrate with different levels of fertilization to evaluate the plant biomass and nutritional status. The bacteria strains are able to produce indole acetic acid and to amplify the nifH gene. The coinoculation of strains IAC-BeCa-095 with AMF (Glomus macrocarpum and Glomus etunicatum) improved the plant shoot biomass (25%) on the fertilized substrate. The strains IAC-BeCa-088 (Burkholderia caribensis), IAC-RBca5 (Pseudomonas sp.) and IAC-RBca10 (Bacillus sp.) without AMF and fertilization improved the shoot biomass by up to 35%. Coinoculation with strain IAC-BeCa-095 (Kosakonia radicincitans) and AMF improved the shoot and root biomass by up to 27 and 75%, respectively, in the conventionally fertilized substrate, demonstrating a synergistic effect of these microorganism consortia. The use of beneficial microorganisms may be a viable practice in the production of PSS sugarcane. Moreover, this study is the first to demonstrate the synergistic effect of endophytic bacteria (K. radicincitans) or rhizobacteria (Bacillus sp.) with AMF and Pseudomonas sp. or B. caribensis, without AMF inoculum on the production of sugarcane PSS to improve plant growth and plant nutrition.
