Free heme regulates placenta growth factor through NRF2-antioxidant response signaling

Free heme activates erythroblasts to express and secrete Placenta Growth Factor (PlGF), an angiogenic peptide of the VEGF family. High circulating levels of PlGF have been associated in experimental animals and in patients with sickle cell disease with echocardiographic markers of pulmonary hypertension, a life-limiting complication associated with more intense hemolysis. We now show that the mechanism of heme regulation of PlGF requires the contribution of the key antioxidant response regulator NRF2. Mimicking the effect of heme, the NRF2 agonist sulforaphane stimulates the PlGF transcript level nearly 30-fold in cultured human erythroblastoid cells. Heme and sulforaphane also induce transcripts for NRF2 itself, its partners MAFF and MAFG, and its competitor BACH1. Furthermore, heme induction of the PlGF transcript is significantly diminished by the NRF2 inhibitor brusatol and by siRNA knockdown of the NRF2 and/or MAFG transcription factors. Chromatin immunoprecipitation experiments show that heme induces NRF2 to bind directly to the PlGF promoter region. In complementary in vivo experiments, mice injected with heme show a significant increase in their plasma PlGF protein as early as 3 h after treatment. Our results reveal an important mechanism of PlGF regulation, adding to the growing literature that supports the pivotal importance of the NRF2 axis in the pathobiology of sickle cell disease.

The Oxidant Response Transcription Factor NRF2 Mediates Heme Activation of Placenta Growth Factor Expression in Erythroid Cells, a Contributor to Pulmonary Hypertension in Sickle Cell Disease

Patients with sickle cell disease (SCD) have elevated plasma levels of placenta growth factor (PlGF) which promotes expression of the pulmonary vasoconstrictor endothelin-1 (ET-1) contributing to pulmonary hypertension, an important age-related and life-limiting complication of SCD. In SCD patients, markers of high iron burden are associated with the highest PlGF levels, leading us to hypothesize a mechanistic link between excessive iron and the induction of the PlGF protein. We have published evidence that heme-bound iron stimulates the PlGF promoter up to 400-fold in human K562 cells and in primary human erythroid cells. Gene transfer, knockout and reconstitution experiments have documented the requirement for erythroid Kruppel-like factor (EKLF; KLF1), confirmed by chromatin immunoprecipitation. We are currently investigating the role of other transcription factors known to sense intracellular iron.Using cultured human erythroid cells (K562 cells) treated with heme-bound iron (hemin) and quantitative real-time PCR, we have assessed the time course of heme regulation of the transcripts of several members of the Nrf2-small Maf family of transcription factors. The transcripts for the antioxidant NRF2, NFE2, MafF, MafG and BACH1 are dynamically regulated following exposure of the cells to heme. Gene expression knockdown and small molecule inhibitor and activator experiments have revealed a central role of NRF2 in activating the PlGF promoter in response to heme, further supported by chromatin immunoprecipitation experiments. Specifically, the PlGF promoter is robustly activated by the well characterized non-oxidative NRF2 agonist sulforaphane, and also activated by the nonspecific oxidant hydrogen peroxide. Furthermore, heme activation of the PlGF promoter is inhibited by the NRF2 inhibitor brusatol, by siRNA directed against NRF2, and by the nonspecific antioxidant N-acetyl cysteine. Although the small Maf proteins are often considered to be nonspecific, interchangeable heterodimeric partners of NRF2, our knockdown data suggest a specific role for MafG cooperating with NRF2 in transducing the heme signal on the PlGF promoter. Our knockdown and sulforaphane results also support a role for NRF2 in regulating the transcript levels of all the known NRF2 family members. We have found similarities and differences in the regulation of the PlGF promoter with another well described heme-responsive promoter, heme oxygenase-1. We are currently investigating these regulatory effects on heme on PlGF expression in vivo and in primary erythroid cells from Nrf2 knockout mice.Our results to date support a mechanism in which accelerated heme turnover in sickle cell disease promotes robust expression of PlGF in erythroblasts during erythroid differentiation, through a pathway that involves EKLF, NRF2 and MafG. Our prior published data in collaboration with the Malik lab has documented a causal role for PlGF in promoting expression of the potent vasoconstrictor endothelin-1 accompanied by the development of pulmonary hypertension, corroborated by data from humans with SCD. This mechanism helps to explain the clinical observation that heavily transfused, iron overloaded adults with SCD are more likely to develop pulmonary hypertension, as a potential consequence of excess heme trafficking from turnover of transfused red cells. PlGF mediates neutrophil elastase-induced airway epithelial cell apoptosis and emphysema.

BackgroundChronic pulmonary obstructive disease (COPD) has become the fourth leading cause of death worldwide. Cigarette smoking induces neutrophil elastase (NE) and contributes to COPD, but the detailed mechanisms involved are not fully established. In an animal model of pulmonary emphysema, there are increased expressions of placenta growth factor (PlGF) and lung epithelial (LE) cell apoptosis. This study hypothesized that excessive NE may up-regulate PlGF and that PlGF-induced LE apoptosis mediates the pathogenesis of pulmonary emphysema.MethodsHuman bronchial epithelial cells, BEAS-2B, and primary mouse type II alveolar epithelial cells were treated with NE. The PlGF promoter activity was examined by luciferase activity assay, while PlGF expression and secretion were evaluated by RT-PCR, Western blotting, and ELISA. Both cell lines were treated with PlGF to evaluate its effects and the downstream signaling pathways leading to LE cell apoptosis. PlGF knockout and wild-type mice were instilled with NE to determine the roles of PlGF and its downstream molecules in NE-promoted mice pulmonary apoptosis and emphysema phenotype.ResultsThe transcriptional factor, early growth response gene-1, was involved in the NE-promoted PlGF promoter activity, and the expression and secretion of PlGF mRNA and protein in LE cells. PlGF-induced LE cell apoptosis and NE-induced mice pulmonary apoptosis and emphysema were mediated by the downstream c-Jun N-terminal kinase (JNK) and protein kinase C (PKC)δ signaling pathways.ConclusionThe NE-PlGF-JNK/PKCδ pathway contributes to the pathogenesis of LE cell apoptosis and emphysema. Arecoline‐stimulated placenta growth factor production in gingival epithelial cells: modulation by curcumin

Placenta growth factor (PlGF) is associated with the progression and prognosis of oral cancer. This study used ELISA, quantitative polymerase chain reaction, and Western blotting to study the arecoline-stimulated (PlGF) protein or mRNA expression in human gingival epithelial S-G cells. Arecoline, a major areca nut alkaloid and an oral carcinogen, could stimulate PlGF protein synthesis in S-G cells in a dose- and time-dependent manner. The levels of PlGF protein secretion increased about 3.1- and 3.8-fold after 24-h exposure to 0.4 and 0.8 mM arecoline, respectively. Pretreatment with antioxidant N-acetyl-l-cysteine (NAC) and ERK inhibitor PD98059, but not NF-κB inhibitor Bay 11-7082, JNK inhibitor SP600125, p38 MAPK inhibitor SB203580, and PI3-K inhibitor LY294002, significantly reduced arecoline-induced PlGF protein synthesis. ELISA analyses demonstrated that NAC and PD98059 reduced about 43% and 38% of the arecoline-induced PlGF protein secretion, respectively. However, combined treatment with NAC and PD98059 did not show additive effect. Moreover, 10 μM curcumin and 4 mM NAC significantly inhibited arecoline-induced ERK activation. Furthermore, 10 μM curcumin completely blocked arecoline-induced PlGF mRNA expression. Arecoline-induced PlGF synthesis is probably mediated by reactive oxygen species/ERK pathways, and curcumin may be an useful agent in controlling oral carcinogenesis.

Evidence for a role of endothelial‐cell derived PLGF in paracrine communication

Placenta growth factor (PLGF) is a vascular endothelial growth factor (VEGF)‐family protein that mediates arteriogenesis (adaptive collateral artery remodeling) via a monocyte‐dependent mechanism. In a previous study of 10 different endothelial cell (EC) and vascular smooth muscle cell (SMC) lines, we found that EC express much higher levels of PLGF mRNA and protein than do SMC. The high level of PLGF expression by EC was surprising, as the related factor VEGF is expressed mainly in tissue surrounding EC, rather than in EC. PLGF has been shown not to be mitogenic for EC. Therefore, we hypothesized that the high level of PLGF expression in EC might be important for supporting other EC functions, such as survival, migration, and tube formation. To test this hypothesis, we knocked down PLGF expression in human coronary artery endothelial cells (HCAEC) using siRNA and performed functional assays. Knockdown of PLGF did not affect EC viability. Migration of EC towards VEGF and tube formation in GelTrex were likewise unaffected by PLGF knockdown. Survival of human coronary artery smooth muscle cells (HCASMC) was also unaffected by PLGF siRNA treatment. These results suggest that the high expression of PLGF by EC likely mediates paracrine communication between EC and non‐vascular cells, such as circulating monocytes. Prognostic impact of placenta growth factor and vascular endothelial growth factor A in patients with breast cancer

Placenta growth factor (PlGF) and vascular endothelial growth factor A (VEGF-A) are angiogenic growth factors interacting competitively with the same receptors. VEGF-A is essential in both normal and pathologic conditions, but the functions of PlGF seem to be restricted to pathologic conditions such as ischemic heart disease, arthritis and tumor growth. Angiogenesis is a complex process with several growth factors involved. Because PlGF modulates VEGF-A responses, we investigated their mutual relationship and impact on breast cancer prognosis. Quantitative PlGF and VEGF-A levels were measured in 229 tumor tissue specimen from primarily operated patients with unilateral breast cancer. Non-malignant breast tissue was also dissected near the tumor and quantitative measurements were available for 211 patients. PlGF and VEGF-A protein levels in homogenized tissue lysates were analyzed using the Luminex system. We found significantly higher median levels of PlGF and VEGF-A in tumor tissue compared to non-malignant tissue (PlGF: 69.8 vs. 31.4 pg/mg, p < 0.001 and VEGF-A: 1148.2 vs. 163.5 pg/mg, p < 0.001). PlGF and VEGF-A were correlated in both malignant tissue (r = 0.41, p < 0.001) and in non-malignant tissue (r = 0.69, p < 0.001). The proportion of node positive patients was higher with high PlGF expression (61.4%) than with low PlGF expression (45.6%) in tumor tissue, p = 0.024. High levels of PlGF and VEGF-A in tumor tissue were associated with significant shorter recurrence-free survival (RFS) in both univariate analysis (PlGF: p = 0.023; VEGF-A: p = 0.047) and in multivariate analysis (PlGF: p = 0.026; VEGF-A: p = 0.036). Neither PlGF nor VEGF-A expression in non-malignant tissue were predictors for RFS. In conclusion, our results support the mutual relationship between PlGF and VEGF-A and encourage further investigations as prognostic markers in breast cancer patients.

Expression of Placenta Growth Factor in Colorectal Carcinomas

PurposePlacenta growth factor (PlGF) is a member of the vascular endothelial growth factor (VEGF) family. PlGF is implicated in several pathologic processes, including the growth and spread of cancer and tumor angiogenesis. The aim of this study was to evaluate the expression and the clinical implications of PlGF in colorectal cancer.MethodsIn order to ascertain the clinical significance of PlGF expression in colorectal cancer, the researcher analyzed the expression pattern of PlGF by using an immunohistochemical method and attempted to establish if a relationship existed between PlGF expression and microvessel density (MVD), and subsequently between PlGF expression and the predicted prognosis. A total of 83 patients with colorectal cancer were included for immunohistochemical staining. Clinicopathological characteristics were defined according to the tumor-node-metastasis (TNM) criteria of the Union for International Cancer Control. Clinicopathologic factors, such as age, sex, histological types of tumors, tumor cell grade, TNM stage, lymphovascular invasion, and lymph-node metastasis, were reviewed.ResultsIn this study, the PlGF protein expression level was significantly correlated with MVD, patient survival, and clinicopathological factors such as lymph-node metastasis, TNM staging, lymphatic invasion and vascular invasion.ConclusionPlGF may be an important angiogenic factor in human colorectal cancer, and in this study, PlGF expression level was significantly correlated with positive lymph-node metastases, tumor stage, and patient survival. These findings suggest that PlGF expression correlates with disease progression and may be used as a prognostic marker for colorectal cancer.

Placenta growth factor and vascular endothelial growth factor B expression in the hypoxic lung

BackgroundChronic alveolar hypoxia, due to residence at high altitude or chronic obstructive lung diseases, leads to pulmonary hypertension, which may be further complicated by right heart failure, increasing morbidity and mortality. In the non-diseased lung, angiogenesis occurs in chronic hypoxia and may act in a protective, adaptive manner. To date, little is known about the behaviour of individual vascular endothelial growth factor (VEGF) family ligands in hypoxia-induced pulmonary angiogenesis. The aim of this study was to examine the expression of placenta growth factor (PlGF) and VEGFB during the development of hypoxic pulmonary angiogenesis and their functional effects on the pulmonary endothelium.MethodsMale Sprague Dawley rats were exposed to conditions of normoxia (21% O2) or hypoxia (10% O2) for 1-21 days. Stereological analysis of vascular structure, real-time PCR analysis of vascular endothelial growth factor A (VEGFA), VEGFB, placenta growth factor (PlGF), VEGF receptor 1 (VEGFR1) and VEGFR2, immunohistochemistry and western blots were completed. The effects of VEGF ligands on human pulmonary microvascular endothelial cells were determined using a wound-healing assay.ResultsTypical vascular remodelling and angiogenesis were observed in the hypoxic lung. PlGF and VEGFB mRNA expression were significantly increased in the hypoxic lung. Immunohistochemical analysis showed reduced expression of VEGFB protein in hypoxia although PlGF protein was unchanged. The expression of VEGFA mRNA and protein was unchanged. In vitro PlGF at high concentration mimicked the wound-healing actions of VEGFA on pulmonary microvascular endothelial monolayers. Low concentrations of PlGF potentiated the wound-healing actions of VEGFA while higher concentrations of PlGF were without this effect. VEGFB inhibited the wound-healing actions of VEGFA while VEGFB and PlGF together were mutually antagonistic.ConclusionsVEGFB and PlGF can either inhibit or potentiate the actions of VEGFA, depending on their relative concentrations, which change in the hypoxic lung. Thus their actions in vivo depend on their specific concentrations within the microenvironment of the alveolar wall during the course of adaptation to pulmonary hypoxia.

Placenta growth factor expression is regulated by hydrogen peroxide in vascular smooth muscle cells

When supply arteries become occluded, blood is diverted through preexisting collateral vessels. Shear stress arising from this increase in blood flow provides the initial physiological stimulus for expansion of the collateral circulation, a process termed arteriogenesis. Endothelial cells (EC) respond to increased shear stress by releasing a variety of mediators that can act on underlying smooth muscle cells (SMC). Placenta growth factor (PLGF) is known to mediate certain aspects of arteriogenesis, such as recruitment of monocytes to the vessel wall. Therefore, we tested whether SMC PLGF expression is influenced by mediators released by EC. We used A10 SMC cultured with medium that had been conditioned by EOMA EC for 4 days as a model. We found that EC-conditioned medium is able to upregulate PLGF gene expression in A10 SMC. Further experiments identified hydrogen peroxide (H(2)O(2)) as a key mediator of this response. We confirmed the physiological relevance of this mechanism in primary human coronary artery SMCs by demonstrating that exogenous H(2)O(2) specifically upregulates PLGF gene and protein expression. We also demonstrated that the physiological stimulus of shear stress raises endogenous H(2)O(2) levels in media into the range found to increase PLGF expression. In this study, we demonstrate that EC-released H(2)O(2) acts as a positive regulator of PLGF gene and protein expression in vascular SMC. To our knowledge, this is the first study to describe H(2)O(2) as a regulator of PLGF expression and therefore an upstream mediator of PLGF-driven arteriogenesis.

Role and prognostic value of placenta growth factor (PlGF) in colorectal carcinomas.

PlGF is implicated in several pathologic processes, including the growth, spread of cancer and tumor angiogenesis. The aim of this study was to evaluate expression and clinical implications of PlGF in colorectal cancer. Methods: In order to ascertain the clinical significance of PlGF expression in colorectal cancer, we analysed the expression pattern of PlGF using both immunohistochemical method and real time quantitative PCR and attempted to establish if a relationship existed between PlGF and microvessel density (MVD), and subsequently between PlGF and the predicted prognosis. We investigated 72 tumor/non-tumor pairs of fresh colorectal tissues for real time PCR and a total of 83 patients with colorectal cancer were included for immunohistochemical staining. Clinico-pathological characteristics were defined according to the TNM criteria of the UICC. Clinico-pathologic factors, such as age, sex, histological types of tumors, tumor cell grade, TNM stage, lymphovascular invasion, lymph node metastasis, were reviewed. Results: The mRNA expression levels for PlGF was significantly higher in tumor than in non- tumor tissues (p = 0.001). The ratio of PlGF expression in tumor to non-tumor in the advanced staged group was significantly higher than for the early staged group. PlGF mRNA was significantly higher in III-IV stage patients than in stage I-II patients (p = 0.011). The relationship between PlGF mRNA expression and sex, histological type, lymph node status was otherwise not statistically significant. Immunohistochemically, PlGF protein expression level was significantly correlated with MVD, patient survival, and clinicopathological factors such as lymph node metastasis, TNM staging, lymphatic invasion and vascular invasion in this study. Conclusions: PlGF may be an important angiogenic factor in human colorectal cancer, and PlGF expression level was significantly correlated with positive lymph node metastases, tumor stages, and patient survival. These findings suggest that PlGF expression correlates with disease progression and may be used as a tumor marker for colorectal cancer. No significant financial relationships to disclose.

Hypoxia Increases Placenta Growth Factor Expression in Human Myocardium and Cultured Neonatal Rat Cardiomyocytes

Placenta growth factor (PlGF) plays an important role in pathologic angiogenesis and is believed to be an independent biomarker in patients with coronary artery disease. However, little is known regarding the regulation of PlGF expression in heart tissue. We determined expression changes in PlGF and its receptor, VEGFR1, in normal and abnormal biopsies from human cardiac allografts and in cardiomyocytes cultured under hypoxia or cyclical stretch conditions. Human donor myocardium and biopsies from allografts without fibrin deposits expressed PlGF and VEGFR1 mRNA. Biopsies (n = 7) with myocardial fibrin, elevated serum cardiac troponin I titers (p < 0.03) and cellular infiltrates (p < 0.05) expressed 1.6-fold more PlGF mRNA than biopsies from allografts without fibrin (n = 11; p < 0.05). PlGF protein was localized in cardiomyocytes, extracellular matrix and some microvessels in areas with fibrin deposition. VEGFR1 mRNA expression was not different between groups. Cultured neonatal rat cardiomyocytes constitutively expressed PlGF/VEGFR1 under normoxia. PlGF expression was increased 3.88 +/- 0.62-fold after 12 hours (n = 6; p </= 0.05) and 3.64 +/- 0.41-fold after 24 hours of hypoxia (n = 6; p <or= 0.05). Shorter periods of hypoxia, conditioned media from hypoxic cells and cyclical stretch did not significantly alter PlGF or VEGFR1 expression. Cardiomyocyte PIGF expression is upregulated by hypoxia in vitro and its expression increases significantly in allografts with myocardial damage. Collectively, these results provide important temporal and spatial evidence that endogenous PlGF may facilitate cardiac healing after myocardial hypoxia/ischemia.

Metal Transcription Factor-1 Is Involved in Hypoxia-Dependent Regulation of Placenta Growth Factor in Trophoblast-Derived Cells

Placenta growth factor (PlGF) is a placental angiogenic factor. Metal-responsive transcription factor (MTF)-1 was reported to take part in the hypoxic induction of PlGF in RAS-transformed mouse fibroblasts. We contrarily showed that PlGF mRNA and protein levels decreased under hypoxia in a choriocarcinoma BeWo cell line derived from trophoblast. In this report, we examined whether hypoxia-dependent regulation of the PlGF gene in these cells also depends on MTF-1. We analyzed the effect of hypoxia on MTF-1 expression, and it was revealed to be decreased. Moreover, MTF-1 small interfering RNA treatment decreased PlGF mRNA level. To investigate the transcription of PlGF under hypoxia, we cloned promoter region of the human PlGF. Promoter deletion analysis suggested that triple repeats of metal-responsive element located between -511 and -468 bp in the promoter are important for the hypoxic regulation of PlGF. Treatment with MTF-1 small interfering RNA resulted in the significant decreased luciferase activity in PlGF reporter constructs. Chromatin immunoprecipitation showed the binding of the MTF-1 protein to the promoter region. We examined MTF-1 immunoreactivity in trophoblasts of term placental tissue from patients with normal pregnancies and preeclampsia, which represents a condition of placental hypoxia. Immunoreactivity of the MTF-1 protein was decreased in placentas from pregnant women with preeclampsia when compared with those from normal pregnant women. Taken together, these findings suggest that MTF-1 is involved in hypoxia-dependent regulation of PlGF in trophoblast-derived cells.

Placenta growth factor, PLGF, influences the motility of lung cancer cells, the role of Rho associated kinase, Rock1

Placenta growth factor (PlGF) is a member of the VEGF family and has been implicated in the aggressive capacity of solid tumours, partly via its impact on angiogenesis. The present study determined the direct biological function of endogenous PlGF in lung cancer cells. From the human non-small cell lung cancer cell line A549 which expressed good level of PlGF, we created sublines within which PlGF expression was knockdown by way of anti-PlGF ribozyme transgenes. Remarkable reductions of both PlGF mRNA and protein by the ribozyme transgenes were revealed in A549 transfectants (A549(DeltaPlGF)) using RT-PCR and Western blotting respectively. A549(DeltaPlGF) cells exhibited significantly reduced migration and adhesion compared with the wild-type (A549(WT)) and the empty plasmid control (A549(pEF/His)) cells. Immunocytochemistry and Western blotting further revealed that the expression of ROCK1, Rho associated kinase, was also reduced in A549(DeltaPlGF) cells, in comparison with wild-type and control cells. In addition, A549(DeltaPlGF) cells lost its response to a ROCK inhibitor, which otherwise strongly inhibited the motility of A549(WT) and A549(pEF/His) cells. These data indicate that PlGF directly regulates the motility of human lung cancer cells and that this regulation critically dependent on ROCK-1. The study further indicates that PlGF is a potential therapeutic target in lung cancer.

Microvasculature Change and Placenta Growth Factor Expression in the Early Stage of a Rat Remnant Kidney Model

Background/Aim: There is significant loss of microvasculature and impaired angiogenesis in rat remnant kidney (RK). Placenta growth factor (PlGF) is a potential angiogenic growth factor. In this study, we investigate the changes of microvasculature and expression of PlGF in the first 4 weeks of the early stage of a rat RK model. Method: RK was induced by right nephrectomy and ligation of two of the three branches of the left renal arteries (equivalent to 5/6 subtotal nephrectomy). Blood urea nitrogen (BUN), serum creatinine (Scr), and blood pressure (BP) were measured. Proliferation of endothelial cells was identified by double staining of two antibodies, anti-rat endothelial cell (RECA-1) and antiproliferating cell nuclear antigen (PCNA). RT-PCR and Western blot were used for PlGF analysis. Results: BUN, Scr and BP remained stable after rising within the first week. An angiogenic response occurred in RKs, with an increase in the proliferation of peritubular and glomerular endothelial cells. Both PlGF protein and mRNA expression were significantly upregulated 2- to 3-fold in RK at week 1 and week 2, compared to the sham-operated group (p < 0.05). Conclusion: The expression of PlGF is upregulated and coincident with an early angiogenic response in rat RK, suggesting that PlGF may be involved in angiogenesis in progressive renal injury.

Overexpression of placenta growth factor in human middle ear cholesteatoma

Conclusions. The expression and localization of placenta growth factor (PlGF) within cholesteatoma were defined. The authors propose that PlGF is an angiogenic growth factor in cholesteatoma, and participates in the neoangiogenesis of cholesteatoma. Objectives. Middle ear cholesteatoma is characterized by the presence of a keratinizing squamous epithelium with hyperproliferative features. Such growth can only be supported by abundant blood vessels. Because proliferating tissues require an enhanced blood supply, angiogenesis appears to be a prerequisite for the expansion of cholesteatoma. This study aimed to analyze the presence of PlGF as an angiogenic growth factor in human cholesteatoma. Materials and methods. Tissue samples from human cholesteatoma and normal auditory meatal skin were obtained from patients during surgery for cholesteatoma of the middle ear. PlGF mRNA expression was quantified by real-time reverse transcriptase-polymerase chain reaction (RT-PCR). PlGF was localized by immunohistochemical staining. Western blotting was used for detection of PlGF protein. Results. Expression of PlGF mRNA was significantly elevated in the epithelium of cholesteatoma compared with normal auditory meatal skin. PlGF was detected on cholesteatoma by Western blotting. PlGF was detected in the suprabasal layer of cholesteatoma using immunohistochemical study, but was not detected in normal auditory meatal skin.

Human retinal epithelium produces and responds to placenta growth factor

Placenta growth factor (PlGF) is an important co-factor in retinal neovascularization. To examine whether retinal pigment epithelial (RPE) cells may represent a source for PlGF during retinopathy, we determined whether human RPE cells in vitro produce and respond to PlGF. In addition, we determined whether the cells express receptors for PlGF, i.e. flt-1 and neuropilins. Cultured human RPE cells of passages 3-5 were used. The regulation of the PlGF gene and protein expression by growth factors and cytokines was evaluated by quantitative PCR and ELISA. Proliferation rates and chemotaxis were determined by a bromodeoxyuridine and a Boyden chamber assay. Human RPE cells express mRNAs for various members of the vascular endothelial growth factor family and for their receptors, including mRNAs for PlGF, flt-1, KDR, and neuropilins-1 and -2. The expression levels of the mRNAs for neuropilins-1 and -2 were significantly higher than those for flt-1 and KDR. Members of the transforming growth factor (TGF)-beta superfamily of growth factors (BMP-4, TGF-beta1, and beta2) were strong inducers of PlGF gene expression, and evoked secretion of PlGF-2 protein by RPE cells. Exogenous PlGF-2 induced chemotaxis in RPE cells and reduced slightly the cell proliferation at high concentrations. The findings that RPE cells produce and respond to PlGF indicate that the factor exerts an autocrine/paracrine action on these cells. It is suggested that increased expression of TGF-beta-related growth factors during diabetic retinopathy may cause PlGF secretion by RPE cells contributing to the stimulation of cell migration as a critical component of the progression of fibrovascular membranes.

Expression of Placenta growth factor (PlGF) in non-small cell lung cancer (NSCLC) and the clinical and prognostic significance.

BackgroundPlacenta growth factor (PlGF) is a member of the vascular endothelial growth factor (VEGF) family. Over-expression of PlGF is known to be associated with pathological angiogenesis. This study examined PlGF expression at protein and message levels in non-small cell lung cancer (NSCLC), in which no reports on the significance of PlGF expression is available to date.Patients and methodsWe used immunohistochemistry to assess the PlGF protein and correlated PlGF with microvessel density (MVD), as well as clinical outcome in patients with NSCLC tumours (n = 91). In addition, we applied a real time quantitative PCR assay using SYBR Green chemistry to measure PlGF mRNA in normal lung tissues and NSCLC tumours.ResultsPlGF was positively stained mainly in cytoplasm of lung cancer cells. High level staining of PlGF was found in 38.5% NSCLC patients. A high level of MVD in NSCLC was found in 42.9% of cases. Tumours with high level and low level PlGF staining had a significantly different MVD (26.69 vs. 20.79, respectively, p = 0.003). Using both univariate and multivariate analyses, PlGF was found to be an independent prognostic factor. Real time PCR analysis revealed that PlGF mRNA was higher in the cancer tissue than normal tissue (0.95 ± 0.19 vs. 0.57 ± 0.24; p < 0.005) and that PlGF mRNA was significant higher in III-IV stage patients than in I-II stage patients (1.03 ± 0.20 vs. 0.80 ± 0.17; p = 0.011).ConclusionPlGF expression is significantly more in NSCLC tumour tissues than in matched normal tissues. It has a significant positive association with MVD and is an independent factor for NSCLC patients. PlGF may have a pivotal role in NSCLC development and disease progression.

Expression of placenta growth factor is regulated by both VEGF and hyperglycaemia via VEGFR-2

Placenta growth factor (PlGF) has been implicated in both physiological and pathological angiogenesis; however, little is known about what regulates its expression. In this study, retinal microvascular endothelial cells and pericytes were exposed to varying concentrations of VEGF and glucose and PlGF expression measured by RT-PCR and Western blotting. Both PlGF mRNA and protein were observed in unstimulated microvascular endothelial cells with only weak expression in pericytes. In endothelial cells, VEGF (100 ng/ml) and glucose (15 mM) induced an increase in expression of PlGF at both the mRNA and protein level while no effect was observed for pericytes. The increase in PlGF expression could be totally abolished by blocking VEGFR-2, and in the case of glucose by neutralising VEGF. VEGF-stimulated PlGF expression was largely inhibited by PD 98059, an inhibitor of mitogen-activated protein kinase (MAPK) and partially by GF 109203X, an inhibitor of protein kinase C (PKC), indicating that VEGF up-regulates PlGF expression via the MAPK signalling pathway and partially through PKC. Taken together, our findings suggest that VEGF orchestrates the contribution of PlGF in angiogenesis via more than one intracellular pathway and that hyperglycaemia, as occurs in diabetes, is an important regulator of PlGF expression via VEGF up-regulation.

The significance of placenta growth factor in angiogenesis and clinical outcome of human gastric cancer

Placenta growth factor (PlGF) is a member of the vascular endothelial growth factor (VEGF) family of proangiogenic factors and its overexpression has been linked to pathological angiogenesis. We studied the relationship between the expression of PlGF and VEGF in human gastric cancer tissues and microvessel density (MVD), as well as clinical outcome in 79 patients with gastric cancer by using an enzyme immunoassay for PlGF and VEGF expression levels in gastric cancers and surrounding non-cancerous mucosa. PlGF protein levels were significantly higher in tumor than in the corresponding non-tumorous mucosa (median value 48.5 vs 9.8 pg/mg, P<0.001). In contrast, VEGF protein levels were not (66.7 vs 80.7 pg/mg, P=0.522). VEGF expression level was not significantly correlated with MVD, patient survival, and clinicopathological factors except Lauren classification in this study. PlGF may be an important angiogenic factor in human gastric cancer, and PlGF expression level was significantly correlated with serosal invasion, positive lymph node metastases, tumor stages, and patient survival.

Increased expression of vascular endothelial growth factor in placentas of p57Kip2 null embryos

Placentas of mice lacking p57Kip2 expression have trophoblastic hyperplasia. To elucidate the mechanism underlying this phenomenon, we studied expression of two angiogenic factors, vascular endothelial growth factor (VEGF) and placenta growth factor (PlGF). Immunohistochemical analysis with anti-VEGF antibodies indicated that VEGF expression was stronger and more clearly detectable in placentas of p57Kip2 null embryos compared to wild-type placentas. PlGF showed no significant differences between placentas of p57Kip2 null and wild-type embryos. In quantitative analysis, placentas of p57Kip2 null embryos showed higher VEGF messenger (m)RNA and protein levels than did wild-type placentas. PlGF mRNA and protein levels were not significantly different. These findings suggest that VEGF is involved in the hyperplasia that occurs in placentas of p57Kip2 null embryos.
