Mapping electric field components of superchiral field with photo-induced force.

Circular dichroism (CD) of materials, difference in absorbance of left- and right-circularly polarized light, is a standard measure of chirality. Detection of the chirality for individual molecules is a frontier in analytical chemistry and optical science. The usage of a superchiral electromagnetic field near metallic structure is one promising way because it boosts the molecular far-field CD signal. However, it is still elusive as to how such a field actually interacts with the molecules. The cause is that the distribution of the electric field vector is unclear in the vicinity of the metal surface. In particular, it is difficult to directly measure the localized field, e.g., using aperture-type scanning near-field optical microscope. Here, we calculate the three-dimensional (3D) electric field vector, including the longitudinal field, and reveal the whole figure of the near-field CD on a two-dimensional (2D) plane just above the metal surface. Moreover, we propose a method to measure the near-field CD of the whole superchiral field by photo-induced force microscopy (PiFM), where the optical force distribution is mapped in a scanning 2D plane. We numerically demonstrate that, although the presence of the metallic probe tip affects the 3D electric field distribution, the PiFM is sufficiently capable to evaluate the superchiral field. Unveiling the whole figure of near-field is significantly beneficial in obtaining rich information of single molecules with multiple orientations and in analyzing the boosted far-field CD signals.

Photoinduced force microscopy as a novel method for the study of microbial nanostructures.

A detailed comparison of the capabilities of electron microscopy and nano-infrared (IR) microscopy for imaging microbial nanostructures has been carried out for the first time. The surface sensitivity, chemical specificity, and non-destructive nature of spectroscopic mapping is shown to offer significant advantages over transmission electron microscopy (TEM) for the study of biological samples. As well as yielding important topographical information, the distribution of amides, lipids, and carbohydrates across cross-sections of bacterial (Escherichia coli, Staphylococcus aureus) and fungal (Candida albicans) cells was demonstrated using PiFM. The unique information derived from this new mode of spectroscopic mapping of the surface chemistry and biology of microbial cell walls and membranes, may provide new insights into fungal/bacterial cell function as well as having potential use in determining mechanisms of antimicrobial resistance, especially those targeting the cell wall.

Optical Imaging of a Single Molecule with Subnanometer Resolution by Photoinduced Force Microscopy.

Visualizing the optical response of individual molecules is a long-standing goal in catalysis, molecular nanotechnology, and biotechnology. The molecular response is dominated not only by the electronic states in their isolated environment but also by neighboring molecules and the substrate. Information about the transfer of energy and charge in real environments is essential for the design of the desired molecular functions. However, visualizing these factors with spatial resolution beyond the molecular scale has been challenging. Here, by combining photoinduced force microscopy and Kelvin probe force microscopy, we have mapped the photoinduced force in a pentacene bilayer with a spatial resolution of 0.6 nm and observed its "multipole excitation". We identified the excitation as the result of energy and charge transfer between the molecules and to the Ag substrate. These findings can be achieved only by combining microscopy techniques to simultaneously visualize the optical response of the molecules and the charge transfer between the neighboring environments. Our approach and findings provide insights into designing molecular functions by considering the optical response at each step of layering molecules.

Sub-10 nm Nanoparticle Detection Using Multi-Technique-Based Micro-Raman Spectroscopy.

Microplastic pollution is a growing public concern as these particles are ubiquitous in various environments and can fragment into smaller nanoplastics. Another environmental concern arises from widely used engineered nanoparticles. Despite the increasing abundance of these nano-sized pollutants and the possibility of interactions with organisms at the sub cellular level, with many risks still being unknown, there are only a few publications on this topic due to the lack of reliable techniques for nanoparticle characterization. We propose a multi-technique approach for the characterization of nanoparticles down to the 10 nm level using standard micro-Raman spectroscopy combined with standard atomic force microscopy. We successfully obtained single-particle spectra from 25 nm sized polystyrene and 9 nm sized TiO2 nanoparticles with corresponding mass limits of detection of 8.6 ag (attogram) and 1.6 ag, respectively, thus demonstrating the possibility of achieving an unambiguous Raman signal from a single, small nanoparticle with a resolution comparable to more complex and time-consuming technologies such as Tip-Enhanced Raman Spectroscopy and Photo-Induced Force Microscopy.

Remote-Controllable Interfacial Electron Tunneling at Heterogeneous Molecular Junctions via Tip-Induced Optoelectrical Engineering.

Molecular electronics enables functional electronic behavior via single molecules or molecular self-assembled monolayers, providing versatile opportunities for hybrid molecular-scale electronic devices. Although various molecular junction structures are constructed to investigate charge transfer dynamics, significant challenges remain in terms of interfacial charging effects and far-field background signals, which dominantly block the optoelectrical observation of interfacial charge transfer dynamics. Here, tip-induced optoelectrical engineering is presented that synergistically correlates photo-induced force microscopy and Kelvin probe force microscopyto remotely control and probe the interfacial charge transfer dynamics with sub-10nm spatial resolution. Based on this approach, the optoelectrical origin of metal-molecule interfaces is clearly revealed by the nanoscale heterogeneity of the tip-sample interaction and optoelectrical reactivity, which theoretically aligned with density functional theory calculations. For a practical device-scale demonstration of tip-induced optoelectrical engineering, interfacial tunneling is remotely controlled at a 4-inch wafer-scale metal-insulator-metal capacitor, facilitating a 5.211-fold current amplification with the tip-induced electrical field. In conclusion, tip-induced optoelectrical engineering provides a novel strategy to comprehensively understand interfacial charge transfer dynamics and a non-destructive tunneling control platform that enables real-time and real-space investigation of ultrathin hybrid molecular systems.

Visualizing the Structure, Composition and Activity of Single Catalyst Particles for Olefin Polymerization and Polyolefin Decomposition.

The structural and morphological characterization of individual catalyst particles for olefin polymerization, as well as for the reverse process of polyolefin decomposition, can provide an improved understanding for how these catalyst materials operate under relevant reaction conditions. In this review, we discuss an emerging analytical toolbox of 2D and 3D chemical imaging techniques that is suitable for investigating the chemistry and reactivity of related catalyst systems. While synchrotron-based X-ray microscopy still provides unparalleled spatial resolutions in 2D and 3D, a number of laboratory-based techniques, most notably focused ion beam-scanning electron microscopy, confocal fluorescence microscopy, infrared photoinduced force microscopy and laboratory-based X-ray nano-computed tomography, have helped to significantly expand the arsenal of analytical tools available to scientists in heterogeneous catalysis and polymer science. In terms of future research, the review outlines the role and impact of in situ and operando (spectro-)microscopy experiments, involving sophisticated reactors as well as online reactant and product analysis, to obtain real-time information on the formation, decomposition, and mobility of polymer phases within single catalyst particles. Furthermore, the potential of fluorescence microscopy, X-ray microscopy and optical microscopy is highlighted for the high-throughput characterization of olefin polymerization and polyolefin decomposition catalysts. By combining these chemical imaging techniques with, for example, chemical staining methodologies, selective probe molecules as well as particle sorting approaches, representative structure-activity relationships can be derived at the level of single catalyst particles.

Nanoscale chemical characterization of secondary protein structure of F-Actin using mid-infrared photoinduced force microscopy (PiF-IR)

The recently developed photoinduced force microscopy for mid-infrared (PiF-IR) offers high spectral resolution in combination with surface sensitivity and a spatial resolution in the range of a few nanometers. Although PiF-IR has primarily been applied to polymer materials, this technology presents significant potential for the chemical characterization of cellular structures approaching single-molecule sensitivity. We applied PiF-IR to differently polymerized F-Actin samples finding general agreement with FTIR spectra from the same samples. Single PiF-IR spectra of F-Actin show variations in the amide I band spectral region, which is related to secondary protein structure. Local variations are also seen in PiF-IR hyperspectra in this region. Such high sensitivity is a necessary requirement for discriminating Actin organization into bundles and other networks in cells and tissue. We applied PiF-IR to mouse liver tissue ex vivo. Single-frequency PiF-IR scans at three different IR frequencies show significant variations in local contrast. However, the presence of other proteins and the unique spatial resolution of PiF-IR pose a challenge to interpreting and validating such data. Careful design of model systems and further theoretical understanding of PiF-IR data far from bulk averages are needed to fully unfold the potential of PiF-IR for high-resolution chemical investigation in the Life Sciences.

Bionanocomposites with Enhanced Physical Properties from Curli Amyloid Assemblies and Cellulose Nanofibrils.

Proteinaceous amyloid fibrils are one of the stiffest biopolymers due to their extensive cross-β-sheet quaternary structure, whereas cellulose nanofibrils (CNFs) exhibit interesting properties associated with their nanoscale size, morphology, large surface area, and biodegradability. Herein, CNFs were supplemented with amyloid fibrils assembled from the Curli-specific gene A (CsgA) protein, the main component of bacterial biofilms. The resulting composites showed superior mechanical properties, up to a 7-fold increase compared to unmodified CNF films. Wettability and thermogravimetric analyses demonstrated high surface hydrophobicity and robust thermal tolerance. Bulk spectroscopic characterization of CNF-CsgA films revealed key insights into the molecular organization within the bionanocomposites. Atomic force microscopy and photoinduced force microscopy revealed the high-resolution location of curli assemblies into the CNF films. This novel sustainable and cost-effective CNF-based bionanocomposites supplemented with intertwined bacterial amyloid fibrils opens novel directions for environmentally friendly applications demanding high mechanical, water-repelling properties, and thermal resistance.

Three-dimensional photoinduced force microscopy reveals artifacts from photothermal tip vibrations

Photoinduced force microscopy (PiFM) is a technique for nanoscale characterization of optically active materials. For correct data analysis, it is important to understand and recognize potential artifacts. In this study, we provide a Fourier-based framework for the different origins of the photoinduced force, including the photothermal motion of the atomic force microscopy (AFM) tip in PiFM measurements. We use individual gold nanoparticles as a model system to explore such tip-related phenomena and to demonstrate the benefits of three-dimensional analysis of the photoinduced force. Comparison of the in-phase heterodyne PiFM signal and the derivative of the tip–sample interaction, ∂⟨kts⟩/∂zc, directly reveals a photothermal origin for the PiFM signal in our experiments. Furthermore, we observe a lateral photothermal vibration of the tip apex induced by tip asymmetry. This lateral vibration emphasizes the importance of considering the motion of the AFM tip during data analysis. The improved lateral resolution conferred by ∂⟨kts⟩/∂zc can lead to misinterpretation. Our findings highlight that meticulous data analysis is required to ensure the correct interpretation of PiFM data.

Spatial Tuning of Light-Matter Interaction via Strain-Gradient-Induced Polarization in Freestanding Wrinkled 2D Materials.

To date, controlled deformation of two-dimensional (2D) materials has been extensively demonstrated with substrate-supported structures. However, interfacial effects arising from these supporting materials may suppress or alter the unique behavior of the deformed 2D materials. To address interfacial effects, we report, for the first time, the formation of a micrometer-scale freestanding wrinkled structure of 2D material without any encapsulation layers where we observed the enhanced light-matter interactions with a spatial modulation. Freestanding wrinkled monolayer WSe2 exhibited about a 330% enhancement relative to supported wrinkled WSe2 quantified through photoinduced force microscopy. Spatial modulation and enhancement of light interaction in the freestanding wrinkled structures are attributed to the enhanced strain-gradient effect (i.e., out-of-plane polarization) enabled by removing the constraining support and proximate dielectrics. Our findings offer an additional degree of freedom to modulate the out-of-plane polarization and enhance the out-of-plane light-matter interaction in 2D materials.

A diagnostic electrochemical aptasensor development for sCD80 protein detection in human serum

Elevating soluble CD80 (sCD80) in human serum is a natural response to autoimmune diseases such as rheumatoid arthritis (RA). The level of sCD80 is associated with RA development and prognosis; therefore, it is potentially used as a biomarker. sCD80 is commonly measured in human serum using immunoassays (e.g., ELISA) with multiple drawbacks, mainly cross-reactivity. Aptamer-based biosensors (aptasensors) development for quantifying and detecting different biological molecules demonstrates applicability in next-generation medicine and biomarker detection. Herein, we selected a specific aptamer for sCD80 by conventional in-vitro selection process (SELEX) with the high-affinity aptamer (Kd = 47.69 nM). A sensitive aptasensor, for the first time, was developed on a screen-printed gold electrode (AuSPE) platform compatible with easy-to-use label-free electrochemical impedance spectroscopy. The immobilization of the aptamer on the gold surface and the presence of sCD80 in a complex with the aptamer were characterized by photo-induced force microscopy, which revealed the uniform assembly of the aptamer monolayer and the distribution of sCD80 on the electrode surface. The developed aptasensor showed a linear performance (0.025–10.0 nM of protein) with a detection limit of 8.0 pM. Furthermore, the aptasensor was tested in a biological matrix, where a linear signal was observed for the increased amount of spiked sCD80 (R2 = 0.9887). The recovery of the spiked amounts ranged from 105 to 125% with coefficient of variation (CV%) <7%, which supported the applicability of this sensor in detecting sCD80 for diagnosis.

Heterostructure particles enable omnidispersible in water and oil towards organic dye recycle

Dispersion of colloidal particles in water or oil is extensively desired for industrial and environmental applications. However, it often strongly depends on indispensable assistance of chemical surfactants or introduction of nanoprotrusions onto the particle surface. Here we demonstrate the omnidispersity of hydrophilic-hydrophobic heterostructure particles (HL-HBPs), synthesized by a surface heterogeneous nanostructuring strategy. Photo-induced force microscopy (PiFM) and adhesion force images both indicate the heterogeneous distribution of hydrophilic domains and hydrophobic domains on the particle surface. These alternating domains allow HL-HBPs to be dispersed in various solvents with different polarity and boiling point. The HL-HBPs can efficiently adsorb organic dyes from water and release them into organic solvents within several seconds. The surface heterogeneous nanostructuring strategy provides an unconventional approach to achieve omnidispersion of colloidal particles beyond surface modification, and the omnidispersible HL-HBPs demonstrate superior capability for dye recycle merely by solvent exchange. These omnidispersible HL-HBPs show great potentials in industrial process and environmental protection.

Coupled harmonic oscillators model with two connected point masses for application in photo-induced force microscopy

Abstract To comprehensively describe the operation of photo-induced force microscopy (PiFM), we have developed a model based on coupled harmonic oscillators. This model features two point masses connected by massless elastic wires, offering greater intuitiveness compared to existing PiFM models. It simplifies these models into a unified theoretical framework. By solving the equations of motion using adjusted oscillator parameters, we have successfully replicated all dynamic features from previous theories. These features include resonance frequencies and shapes of eigenmodes, as well as the responses to various external forces in the two PiFM modes: direct coupling and sideband coupling. Furthermore, by integrating our model with a recently developed photo-induced thermal expansion force model, which covers both tip-enhanced and global expansions, we have managed to uncover the underlying physical mechanism responsible for the unique signal behaviors observed in sideband coupling mode, where the signal plot, as a function of sample thickness, unexpectedly exhibits a peak followed by a valley, rather than a proportionally increasing signal. Our study has the potential to enhance the comprehension of various other physical phenomena associated with PiFM in the future.

Chemical Identification of Sub-20 nm Defects and Sub-Monolayer Residues with Nano IR PiFM

With the advent of EUV lithography and other emerging processes such as area selective deposition, and SOIC hybrid bonding, the need to identify the chemical composition of defects is of paramount importance. The defects of concern range from 10nm to 500nm in lateral size and as thin as ~ 1 nm in thickness, which the current batch of molecular analytical tools cannot address adequately since the indications are that many of the defects and residues detected are organic in nature. In this paper, a relatively new nanoscale technique called infrared photo-induced force microscopy (IR PiFM), which combines non-contact atomic force microscopy (AFM) and infrared (IR) spectroscopy with ~ 5 nm spatial resolution, is introduced. The paper will show how the nanoscale hyperspectral IR PiFM data can provide unambiguous and speedy feedback to process engineers engaged in EUV lithography and other advanced semiconductor processes.

Enhanced efficiency of launching hyperbolic phonon polaritons in stacked α-MoO3 flakes.

In this work, we reported a systemic study on the enhanced efficiency of launching hyperbolic phonon polaritons (PhPs) in stacked α-phase molybdenum trioxide (α-MoO3) flakes. By using the infrared photo-induced force microscopy (PiFM), real-space near-field images (PiFM images) of mechanically exfoliated α-MoO3 thin flakes were recorded within three different Reststrahlen bands (RBs). As referred with PiFM fringes of the single flake, PiFM fringes of the stacked α-MoO3 sample within the RB 2 and RB 3 are greatly improved with the enhancement factor (EF) up to 170%. By performing numerical simulations, it reveals that the general improvement in near-field PiFM fringes arises from the existence of a nanoscale thin dielectric spacer in the middle part between two stacked α-MoO3 flakes. The nanogap acts as a nanoresonator for prompting the near-field coupling of hyperbolic PhPs supported by each flake in the stacked sample, contributing to the increase of polaritonic fields, and verifying the experimental observations Our findings could offer fundamental physical investigations into the effective excitation of PhPs and will be helpful for developing functional nanophotonic devices and circuits.

Postsynthetic Modification of Core-Shell Magnetic Covalent Organic Frameworks for the Selective Removal of Mercury.

Core-shell magnetic covalent organic framework (COF) materials were prepared, followed by shell material functionalization with different organic ligands, including thiosemicarbazide, through a postsynthetic modification approach. The structures of the prepared samples were characterized with various techniques, including powder X-ray diffraction (PXRD), Brunauer-Emmett-Teller (BET) method, thermogravimetric analysis (TGA), photoinduced force microscopy (PiFM), transmission electron microscopy (TEM), scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), and solid 13C NMR. PXRD and BET studies revealed that the crystalline and porous nature of the functionalized COFs was well maintained after three steps of postsynthetic modification. On the other hand, solid 13C NMR, TGA, and PiFM analyses confirmed the successful functionalization of COF materials with good covalent linkage connectivity. The use of the resulting functionalized magnetic COF for selective and ultrafast adsorption of Hg(II) has been investigated. The observations displayed rapid kinetics with adsorption dynamics conforming to the quasi-second-order kinetic model and the Langmuir adsorption model. Furthermore, this prepared crystalline magnetic material demonstrated a high Langmuir Hg(II) uptake capacity, reaching equilibrium in only 5 min. Thermodynamic calculations proved that the adsorption process is endothermic and spontaneous.

PiFM and XPS Studies of Porous TiO2 Films for the Photocatalytic Decomposition of Polystyrene

The rate of photocatalytic oxidation of polystyrene over 0.1 wt% copper-doped TiO2 catalyst spin coated onto a flat substrate is investigated as a function of the catalyst deposition spin speed and, hence, film thickness. Photoinduced force microscopy and photoelectron spectroscopy show no evidence of any products of the photocatalytic oxidation remaining on the surface after reaction. The oxidation reaction shows no discernible dependence on spin speed; it is close to zero order in polystyrene concentration and exhibits a ½ life almost 50 times greater than the equivalent photocatalytic oxidation of stearic acid on the same catalysts. No difference between the rate of decay of the aliphatic and aromatic hydrogens of the polymer was observed, suggesting that once a polymer chain begins oxidising, subsequent steps are relatively rapid. This is consistent with the rate-determining step being dependent on the extent of coordination to the surface, which is much more favourable with stearic acid because of its carboxylic group.

Development of low-temperature and ultrahigh-vacuum photoinduced force microscopy.

In this paper, we develop optical and electronic systems for photoinduced force microscopy (PiFM) that can measure photoinduced forces under low temperature and ultrahigh vacuum (LT-UHV) without artifacts. For our LT-UHV PiFM, light is irradiated from the side on the tip-sample junction, which can be adjusted through the combination of an objective lens inside the vacuum chamber and a 90° mirror outside the vacuum chamber. We measured photoinduced forces due to the electric field enhancement between the tip and the Ag surface, and confirmed that photoinduced force mapping and measurement of photoinduced force curves were possible using the PiFM that we developed. The Ag surface was used to measure the photoinduced force with high sensitivity, and it is effective in enhancing the electric field using the plasmon gap mode between the metal tip and the metal surface. Additionally, we confirmed the necessity of Kelvin feedback during the measurement of photoinduced forces, to avoid artifacts due to electrostatic forces, by measuring photoinduced forces on organic thin films. The PiFM, operating under low temperature and ultrahigh vacuum developed here, is a promising tool to investigate the optical properties of various materials with very high spatial resolution.

Near-field circular dichroism of single molecules.

Near-field images of molecules provide information about their excited orbitals, giving rise to photonic and chemical functions. Such information is crucial to the elucidation of the full potential of molecules as components in functional materials and devices at the nanoscale. However, direct imaging inside single molecules with a complex structure in the near-field is still challenging because it requires in situ observation at a higher resolution than the molecular scale. Here, using a proven theoretical method that has demonstrated sub-nanoscale resolution based on photoinduced force microscopy (PiFM) experiment [Nat. Commun.12, 3865 (2021)10.1038/s41467-021-24136-2], we propose an approach to obtaining the near-field imaging with spatial patterns of electronic transitions of single molecules. We use an extended discrete dipole approximation method that incorporates microscopic nonlocal optical response of molecules and demonstrate that PiFM can visualize circular-dichroism signal patterns at sub-nanometer scale for both optically allowed and forbidden transitions. The result will open the possibility for the direct observation of complex spatial patterns of electronic transitions in a single molecule, providing insight into the optical function of single molecules and helping realize new functional materials and devices.

Structure sensitivity in gas sorption and conversion on metal-organic frameworks

Many catalytic processes depend on the sorption and conversion of gaseous molecules on the surface of (porous) functional materials. These events often preferentially occur on specific, undercoordinated, external surface sites. Here we show the combination of in situ Photo-induced Force Microscopy (PiFM) with Density Functional Theory (DFT) calculations to study the site-specific sorption and conversion of formaldehyde on the external surfaces of well-defined faceted ZIF-8 microcrystals with nanoscale resolution. We observed preferential adsorption of formaldehyde on high index planes. Moreover, in situ PiFM allowed us to visualize unsaturated nanodomains within extended external crystal planes, showing enhanced sorption behavior on the nanoscale. Additionally, on defective ZIF-8 crystals, structure sensitive conversion of formaldehyde through a methoxy- and a formate mechanism mediated by Lewis acidity was found. Strikingly, sorption and conversion were influenced more by the external surface termination than by the concentration of defects. DFT calculations showed that this is due to the presence of specific atomic arrangements on high-index crystal surfaces. With this research, we showcase the high potential of in situ PiFM for structure sensitivity studies on porous functional materials.
