Some properties of chromosomal non histone proteins (NHP) have been studied: The biosynthesis of several NHP is modified in regenerating liver after hepatectomy, it is completely disturbed in Zajdela hepatoma cells. In cultured fibroblasts (coll. with Dr. Y. Courtois), a rapid cell growth is accompagnied by an increase in the synthesis of some NHP, a deprivation of lysine is followed by an increased synthesis of another group of NHP. Liver NHP contain an histone kinase which is also active with phosphoproteins present in NHP. Histones are able to bind to RNA. The phosphorylation of histones diminishes the affinity of these proteins toward RNA. The addition of NHP also diminishes the ability of histones to bind to RNA. NHP reduce the inhibition of transcription by histones throug 2 mechanisms : (a) a binding of phosphoproteins to histones, (b) the enzymatic phosphorylation of histones and of phosphoproteins since the more these phosphoproteins are phosphorylated the more they stimulate nucleohistone templated RNA synthesis. Two kinases have been purified by chromatography : kinase I is active with casein, kinase II is active with casein and with phosvitin. Three active peaks have been found in kinase II by gel electrophoresis. Les protéines non histones (NHP) ont été étudiées sous plusieurs angles : La biosynthèse de certaines de ces protéines dans le foie est modifiée au cours de la régénération après hépatectomie, la biosynthèse est complètement perturbée dans les hépatomes de Zajdela. Dans les fibroblastes en culture (coll. avec Dr. Y. Courtois), la croissance cellulaire est accompagnée de la synthèse d'un groupe de NHP, la carence en lysine est accompagnée de la synthèse d'un autre groupe de NHP. Les NHP contiennent des histones kinases actives sur des phosphoprotéines également présentes dans les NHP. Les histones sont capables de se combiner aux RNA. La phosphorylation des histones empêche cette association. L'addition de NHP diminue également l'affinité des histones vis-à-vis des RNA. Les NHP lèvent en partie l'inhibition de la transcription par les histones. Ceci se fait par 2 mécanismes (a) Par une liaison des phosphoprotéines aux histones (b) Par une phosphorylation des histones et également des phosphoprotéines car plus ces dernières sont phosphorylées plus elles activent la transcription. La chromatographie a permis de purifier à partir du foie une kinase I active sur la caséine et une kinase II active sur la caséine et la phosvitine. Cette dernière est fractionnée en 3 enzymes par electrophorèse.
