Fungal membrane determinants affecting sensitivity to antifungal cyclic lipopeptides from Bacillus spp.

Bacillus spp. produce numerous antimicrobial metabolites. Among these metabolites, cyclic lipopeptides (CLP) including fengycins, iturins, and surfactins are known to have varying antifungal activity against phytopathogenic fungi. The differential activities of CLP have been attributed to diverse mechanisms of action on fungal membranes. However, the precise biochemical determinants driving their antifungal modes of action have not been conclusively identified. In this study, three plant pathogenic fungi of varying lipopeptide sensitivities, Alternaria solani, Cladosporium cucumerinum, and Fusarium sambucinum, were studied to determine how their cell membrane lipid compositions may confer sensitivity and/or tolerance to fengycin, iturin, and surfactin. Results indicated that sensitivity to all three lipopeptides correlated with lower ergosterol content and elevated phospholipid fatty acid unsaturation. Fungal sensitivity to surfactin was also notably different than fengycin and iturin, as surfactin was influenced more by lower phosphatidylethanolamine amounts, higher levels of phosphatidylinositol, and less by phospholipid fatty acyl chain length. Results from this study provide insight into the fungal membrane composition of A. solani, F. sambucinum, and C. cucumerinum and the specific membrane characteristics influencing the antifungal effectiveness of fengycin, iturin, and surfactin. Understanding of these determinants should enable more accurate prediction of sensitivity-tolerance outcomes for other fungal species exposed to these important CLP.

Role of Lipid Composition in the Interaction and Activity of the Antimicrobial Compound Fengycin with Complex Membrane Models.

Fengycins are compounds produced by bacteria of the Bacillus genus with strong antifungal activity. In this work, lipids extracted from fungal and oomycetal molds were used to assess the ability of fengycin to bind and insert into complex membrane models prepared as Langmuir lipid monolayers. In addition, fengycin-induced leakage in liposomes prepared from these complex lipid extracts was also evaluated. Fengycin's ability to bind and incorporate into these membranes seemed to be mainly related to ergosterol content. Other membrane characteristics such as phospholipid fatty acyl chain length played a more peripheral role. A high ergosterol concentration appeared to allow other membrane characteristics generally associated with fengycin binding and/or insertion, such as higher proportion of phosphatidylcholine head groups or increased fatty acyl unsaturation, to be present without adversely affecting membrane integrity. Increased membrane leakage was also generally associated with the presence of low or no ergosterol. Leakage was also correlated with the previously reported biological activity of fengycin on these molds.

Detection of liposomal cholesterol and monophosphoryl lipid A by QS-21 saponin and Limulus polyphemus amebocyte lysate

Liposomes containing cholesterol (Chol) have long been used as an important membrane system for modeling the complex interactions of Chol with adjacent phospholipids or other lipids in a membrane environment. In this study we utilize a probe composed of QS-21, a saponin molecule that recognizes liposomal Chol and causes hemolysis of erythrocytes. The interaction of QS-21 with liposomal Chol results in a stable formulation which, after injection into the tissues of an animal, lacks toxic effects of QS-21 on neighboring cells that contain Chol, such as erythrocytes. Here we have used liposomes containing different saturated phospholipid fatty acyl groups and Chol, with or without monophosphoryl lipid A (MPLA), as model membranes. QS-21 is then employed as a probe to study the interactions of liposomal lipids on the visibility of membrane Chol. We demonstrate that changes either in the mole fraction of Chol in liposomes, or with different chain lengths of phospholipid fatty acyl groups, can have a substantial impact on the detection of Chol by the QS-21. We further show that liposomal MPLA can partially inhibit detection of the liposomal Chol by QS-21. The Limulus amebocyte lysate assay is used for binding to and detection of MPLA. Previous work has demonstrated that sequestration of MPLA into the liposomal lipid bilayer can block detection by the Limulus assay, but the binding site on the MPLA to which the Limulus protein binds is unknown. Changes in liposomal Chol concentration and phospholipid fatty acyl chain length influenced the detection of the liposome-embedded MPLA.

Self-association of model transmembrane alpha-helices is modulated by lipid structure.

We have developed a fluorescence quenching method using peptides containing 3,5-dibromotryrosine to measure oligomerization of model transmembrane alpha-helices in lipid bilayers. Peptides of the type Ac-LysLysGlyLeu(m)XLeu(n)LysLysAla-amide where X is tryptophan or 3,5-dibromotyrosine were found to form heterodimers in bilayers of phosphatidylcholine in the liquid-crystalline phase. The free energy of dimer formation changed little with increasing number of Leu residues from 16 to 22 but increased with increasing phospholipid fatty acyl chain length, with a slope of about 0.5 kJ mol(-1) per fatty acyl chain carbon. Peptides were excluded from lipid in the gel phase, resulting in increased levels of oligomerization. Addition of cholesterol to form the liquid-ordered state led to increased dimerization but without phase separation. The presence of phosphatidylethanolamine had little effect on dimerization.

Effects of phospholipid fatty acyl chain length on phosphorylation and dephosphorylation of the Ca2+-ATPase

The kinetics of the Ca(2+)-ATPase purified from sarcoplasmic reticulum have been studied after reconstitution into bilayers of dimyristoleoylphosphatidylcholine [di(C14:1)PC], dioleoylphosphatidylcholine[di(C18:1)PC] and dinervonylphosphatidylcholine [di(C24:1)PC]. In di(C24:1)PC the rate of phosphorylation of the ATPase by ATP was comparable with that in di(C18:1)PC (about 70 s-1), but in di(C14:1)PC the rate was much lower (21 s-1). Fluorescence responses of the ATPase suggest changes in the phosphoryl-transfer step rather than in the preceding conformational change E1Ca2ATP<-->E1'Ca2ATP. The rate of dephosphorylation of the phosphorylated ATPase was found to decrease in the order di(C24:1)PC < di(C14:1)PC < di(C18:1)PC. For the ATPase in di(C24:1)PC the rate of dephosphorylation (3.3 s-1) was slow enough to be the rate-limiting step for ATP hydrolysis; in di(C14:1)PC, it is suggested that both phosphorylation and dephosphorylation contribute to rate limitation. Phosphorylation of the ATPase in di(C24:1)PC by Pi was normal, but no phosphoenzyme could be detected in di(C14:1)PC. The rate of the Ca(2+)-transport step was normal in di(C24:1)PC, suggesting that the single Ca2+ ion bound to the ATPase in di(C24:1)PC could be transported.

Liposome—complement interactions in rat serum: implications for liposome survival studies

Serum complement opsonizes particles such as bacteria for clearance by the reticuloendothelial system. Complement has been reported to interact with liposomes and therefore may mediate the reticuloendothelial system clearance of liposomes. This study has used a rat serum model to define some of the characteristics of liposomes which modulate their ability to activate complement. Using functional hemolytic assays and C3/C3b crossed immunoelectrophoresis, we have demonstrated that liposomes activated rat complement in a dose-dependent manner with higher concentrations of liposomes activating higher levels of complement. The detection of complement activation required the inclusion of phospholipids bearing a net charge. Complement activation occurred via the classical pathway; no alternative pathway activation was detected. The presence of cholesterol contributed to complement activation in a dose-dependent manner. Phospholipid fatty acyl chain length did not influence complement activation while the introduction of unsaturated acyl chains markedly decreased levels of complement activation. Liposome size also influenced complement activation with 400 nm unilamellar vesicles more effectively activating complement than 50 nm vesicles for equivalent amounts of exposed lipid. These studies demonstrate that the composition of the liposome greatly affects the in vitro activation of rat serum complement and suggest that the biological half-life of liposomes in the circulation of rats may be altered by changing the liposome composition to reduce complement activation.

Nickel and Cadmium-related Changes in Growth, Plasma Membrane Lipid Composition, ATPase Hydrolytic Activity and Proton-pumping of Rice (Oryza sativaL. cv. Bahia) Shoots

Six-day-old rice plants (Oryza sativa L. cv. Bahia) were grown in the presence of 0-5 mol m-3 Ni or Cd for 5 or 10 d. Five days after treatment, some plants were transferred to a medium containing no heavy metal for 5 or 10 d. Plasmalemma vesicles from shoots of treated, transferred (recovery experiments) and control plants were isolated, ATPase activity and proton-pumping were assessed, and free sterols and phospholipids determined. The ATPase hydrolytic activity was increased by 37% and 34% in 5 and 10 d Cd-treated plants, respectively; and by 66% in 5 and 10 d Ni-treated plants. However, neither the initial rate of H + transport nor the proton-pumping rate at steady-state were significantly affected by the treatments. The relative proportion (%) of the plasmalemma sterols campesterol and d5-avenasterol decreased while sitosterol increased during heavy metal treatment. The overall plasmalemma phospholipid fatty acyl chain length and degree of unsaturation were also reduced. In experiments where plants recovered from Cd and Ni treatment, differences between treated and control plants were reverted, particularly in 10 d Ni recovered plants. The possible involvement of lipids in the regulation of the plasmalemma ATPase as well as the relationship between growth, ATPase and adaptations to stress conditions are discussed.

Fatty acyl chain composition in the determination of renal membrane order.

The relative roles of phospholipid fatty acyl chain length and phospholipid fatty acyl chain unsaturation in the determination of rat renal brush border membrane order were examined using multilamellar liposomes. Exposure of brush border membranes to sphingomyelinase resulted in a time- and concentration-dependent decrement in sphingomyelin content. Liposomes prepared from lipid extracts of these membranes were reconstituted to defined phosphatidylcholine (PC)/sphingomyelin (SPH) ratios with pure synthetic PCs of defined chain length and degrees of unsaturation. Mixed-acid PCs from bovine liver, egg, and the rat renal brush border membrane were also examined. The steady state fluorescence anisotropy of 1,6-diphenyl-1,3,5-hexatriene (DPH) at 37 degrees C was used to reflect acyl chain packing. The steady state anisotropy of DPH in liposomes isolated from the rat renal brush border membrane averaged 0.205 +/- 0.001, n = 8. When liposomes were reconstituted to PC/SPH ratios of 1.1, 1.6, and 2.4 with saturated PCs of acyl chain length 16 to 22, differences in anisotropy between groups were not observed. However, when PCs containing unsaturated or mixed-acid fatty acyl chains were introduced, anisotropy decreased in a concentration dependent fashion. These data suggest that phospholipid fatty acyl chain unsaturation, but not acyl chain length, has a powerful influence on renal brush border membrane order and the PC/SPH ratio is an important determinant of renal membrane order by virtue of the unsaturated fatty acids normally present with these phospholipids.

Interfacial properties of model membranes and plasma lipoproteins containing ether lipids.

The interfacial properties of synthetic ester and ether phosphatidylcholines (PCs) were investigated by using the polarity-sensitive fluorescent probes 6-propionyl-2-(dimethylamino)naphthalene (Prodan) and pyrene. The physical state of the phospholipid matrix was determined by fluorescence polarization of 1,6-diphenyl-1,3,5-hexatriene (DPH). Single-bilayer phospholipid vesicles formed by sonication and model high-density lipoproteins were studied. On the basis of a number of spectroscopic and thermodynamic criteria, the interfacial regions of PCs and their ether analogues are similar. The fluorescence properties of Prodan in model lipoproteins or single-bilayer vesicles were independent of the phospholipid fatty acyl chain length and polar head group, as well as the substitution of ether linkage for ester bonds in the phospholipid. The spectral shifts correlated mainly with the physical state of the phospholipid. The emission spectrum of Prodan appeared at shorter wavelengths upon transfer from water to liquid-crystalline phospholipid and blue shifted further when the lipid was cooled to its gel phase. The effect of cholesterol in model high-density lipoproteins on the emission spectrum of Prodan was dose dependent and, at 18 mol % cholesterol, the spectrum was similar to that observed in a pure gel-phase lipid and was independent of temperature. The quantum yield of Prodan fluorescence in an ether-PC matrix was similar to that observed in water, whereas in an ester-PC matrix it was enhanced by a factor of about 5. Phospholipid-water partition coefficients of Prodan were independent of the physical state of 1,2-dimyristoyl-sn-glycero-3-phosphocholine or 1,2-tetradecyl-sn-glycero-3-phosphocholine.(ABSTRACT TRUNCATED AT 250 WORDS)

Lipoprotein lipase-catalyzed hydrolysis of phospholipid monolayers: Effect of fatty acyl composition on enzyme activity

The phospholipase A 1 activity of lipoprotein lipase (LpL) was determined with monomolecular phospholipid films. Rates of phospholipid hydrolysis were dependent on apolipoprotein C-II (the activator protein for LpL) phospholipid fatty acyl composition, and lipid-packing density. In sphingomyelin: cholesterol (2:1, molar) monolayers containing 5 mol % disaturated phosphatidylcholines (PC) and at a surface pressure of 22 mNm −1, rates of LpL hydrolysis of diC 14:0PC, diC 16:0PC, and diC 18:0PC were 74, 207, and 65 nmol h −1 mg LpL −1, respectively. At 22 nNm −1, phospholipids containing unsaturated fatty acyl chains were hydrolyzed at rates 5–10 times greater than saturated lipids. At higher lipid packing densities, the difference in hydrolysis rates between saturated and unsaturated lipids was less apparent. Comparison of molecular areas indicate no simple dependency between the rate of LpL catalysis and phospholipid fatty acyl chain length and saturation/unsaturation.

Effect of monolayer lipid structure and composition on the lipoprotein lipase-catalyzed hydrolysis of triacylglycerol

The effect of lipid composition and structure on the lipoprotein lipase-catalyzed hydrolysis of triacylglycerols was determined in a monolayer system consisting of purified bovine milk lipoprotein lipase and fatty acid-free albumin. In a monolayer of dioleoylphosphatidylcholine containing 1–6 mol% of either tri[ 14C]oleoylglycerol or tri[ 14C]palmitoylglycerol, lipoprotein lipase catalyzed the hydrolysis of the unsaturated triacylglycerol at a higher rate than the saturated lipid and in either the presence or absence of apolipoprotein C-II, the activator protein for the enzyme. For example, with 3 mol% triacylglycerol and in the presence of apolipoprotein C-II, the rate of the lipoprotein lipase-catalyzed hydrolysis of tri[ 14C]oleoylglycerol was 27 μmol oleic acid produced/h per mg enzyme vs. 12 μmol for tri[ 14C]palmitoylglycerol. The effect of phospholipid fatty acyl chain length and unsaturation/saturation, polar head group and surface density on the lipoprotein lipase-catalyzed hydrolysis of tri[ 14C]oleoylglycerol was determined. The rate of enzyme hydrolysis of triacylglycerol was similar whether the phospholipid was a diester or diether lipid or the polar head group was ethanolamine or choline. In general, phospholipids with shorter and unsaturated fatty acyl chains gave higher rates of lipoprotein lipase hydrolysis of triacylglycerol than the corresponding longer and saturated lipids. However, with all phospholipids tested, the rate of enzyme hydrolysis decreased with increasing surface density. Lipoprotein lipase showed no activity toward triacylglycerol in a monolayer of sphingomyelin; addition of dioleoylphosphatidylcholine to the monolayer enhanced the rate of enzyme catalysis. Cholesterol (50 mol%) in a dipalmitoylphosphatidylcholine monolayer increased the rate of the lipoprotein lipase-catalyzed hydrolysis of tri[ 14C]oleoylglycerol, whereas cholesterol decreased the rate in a dioleoylphosphatidylcholine monolayer. The effect of phospholipid structure and surface density on lipoprotein lipase activity could not be accounted for by the amount of apolipoprotein C-II which was present at the interface. Based on these findings and other reports in the literature, we suggest that the catalytic activity of lipoprotein lipase toward tri[ 14C]oleylglycerol in various monolayers is dependent on the conformation or appropriate physical state of the triacylglycerol substrate at the lipid interface.

Influence of temperature on complement-dependent immune damage to liposomes

Maximal release of trapped liposomal glucose, in the presence of saturating amounts of liposomal antigen (galactocerebroside), antiserum (anti-galactocerebroside), and complement, was dependent on temperature. At lower temperatures (20–25°C), maximal glucose release was inversely related to liposomal phospholipid fatty acyl chain length (dimyristoyl phosphatidylcholine > dipalmitoyl phosphatidylcholine > distearoyl phosphatidylcholine > sphingomyelin). At higher temperatures (32–35°C) a limiting plateau of glucose release, at approx. 60%, was reached, or approached, by all preparations. Sphingomyelin liposomes still released less glucose than those prepared from other phospholipids, even at 35°C. The titers of antiserum and complement ( ABL 50/ ml and CL 50/ml) were dependent on temperature, and differences based on liposomal phospholipid fatty acyl chain length were observed. Analysis of antiserum and complement-dependence on temperature, and on phospholipid type, revealed that although antibody binding to galactocerebroside undoubtedly was subject to steric hindrance due to interference by surrounding phospholipids at 20–25°C, steric hindrance did not play a major role in blocking antibody binding above 32°C.

Activation of Human Complement by Liposomes: A Model for Membrane Activation of the Alternative Pathway

Liposomal model membranes were found to activate the alternative pathway of human complement. Activation was measured by C3 conversion and component consumption in serum that had been incubated with liposomes. C3 conversion did not require C1 or C2 of the classical pathway, since it was observed in serum from a C1r-deficient patient, serum from a C2-dificient patient, and normal serum in buffer containing EGTA and MgCl2. The incubation of liposomes with C2-deficient serum resulted in consumption of components C3 through C9 with no consumption of C1 or C4 in a profile typical of alternative pathwya activation. The reaction was further shown to require alternative pathway factor D, and to be independent of antibody. Activation of the alterative pathway was dependent on the membrane composition of the liposomes. A positive charge was required for liposomes to produce C3 conversion. Liposomal cholesterol concentration and phospholipid fatty acyl chain length and unsaturation all influenced activation, suggesting the importance of membrane fluidity. Positively charged liposomes containing dimyristoyl phosphatidylcholine and cholesterol required the presence of certain glycolipids for C3 conversion. The activation of the alternative complement pathway by liposomes of defined membrane composition may provide a suitable model for the study of alternative pathway activation by cellular membranes.

Interactions of C-Reactive Protein and Complement with Liposomes

Abstract We found previously that interaction of C-reactive protein (CRP) with liposomal model membranes resulted in complement(C)-dependent membrane damage. In the present study, we investigated the influence of membrane composition on the interactions of CRP and C with liposomes. Adsorption experiments showed that binding of CRP was greatest to strongly positive liposomes. A lesser, but still substantial, extent of CRP binding also was observed with negative liposomes, but negligible amounts of CRP bound to neutral or weakly positive liposomes. CRP-mediated consumption of hemolytic C, and C-dependent glucose release from liposomes both were strongly influenced by liposomal charge, positive being superior to negative. Glucose release and, to a lesser extent, consumption of hemolytic C were inversely related to phospholipid fatty acyl chain length. Phospholipid fatty acyl unsaturation and liposomal cholesterol concentration both had strong influences on C consumption and glucose release. The data suggest that CRP-mediated C consumption and membrane damage require an optimum membrane fluidity. Complement damage in the presence of CRP was enhanced by certain sphingolipids and also by digalactosyl diglyceride, but not by sphingomyelin. Our results thus demonstrate that CRP-mediated C consumption and C-dependent membrane damage both are influenced by the liposomal membrane composition.

Cholesterol-dependent tetanolysin damage to liposomes

Tetanolysin caused membrane damage, resulting in release of trapped glucose from liposomes containing cholesterol. Maximum glucose release occurred from liposomes that contained 50 mol% cholesterol. At higher or lower levels of cholesterol, glucose release was reduced and glucose release did not occur at all below 40 mol% cholesterol. The apparent activity of tetanolysin was not influenced by temperature (24°C compared to 32°C) or by liposomal phospholipid fatty acyl chain length. We conclude that tetanolysin caused cholesterol-dependent lysin-mediated damage to liposomes, possibly by means of a pore consisting of a complex of toxin and cholesterol.

Complement Activation by Liposome-CRP Complexes: Influence of Membrane Composition

Abstract Previous work has shown that under some conditions C-reactive protein (CRP) can interact with the phosphocholine components of phospholipids, and this results in complement (C) activation. Liposomes consisting of phospholipid, cholesterol (Chol), and either containing or lacking a charged lipid or galactosyl ceramide (Gal Cer), were assayed both for reactivity with CRP, and for C consumption and C-dependent membrane damage. Adsorption studies showed that CRP bound equally well to positively charged liposomes regardless of the phosphatidylcholine (PC) fatty acyl chain length (dimyristoyl vs distearoyl PC) or of the presence of Gal Cer. Negatively charged liposomes also bound CRP, but to a lesser extent. Despite the above binding studies, C-dependent membrane damage (resulting in glucose release) mediated by CRP was strongly dependent on charge, phospholipid fatty acyl chain length, Chol concentration, and Gal Cer. C damage resulting in glucose release occurred with liposomes containing a strong positive charge, but not with weakly positive, neutral, or negatively charged liposomes.
