Dissecting LuxS/AI-2 quorum sensing system-mediated phenyllactic acid production mechanisms of Lactiplantibacillus plantarum L3

The phenyllactic acid (PLA) produced by lactic acid bacteria (LAB) inhibits fungi and facilitates the quality control of fermented milk. A strain of Lactiplantibacillus plantarum L3 (L. plantarum L3) with high PLA production was screened in the pre-laboratory, but the mechanism of its PLA formation is unclear. The amount of autoinducer-2 (AI-2) increased with increasing culture time, as did cell density and PLA. The results in this study suggest that PLA production in L. plantarum L3 may be regulated by the LuxS/AI-2 Quorum Sensing (QS) system. Tandem mass tag (TMT) quantitative proteomics analysis showed that a total of 1291 differentially expressed proteins (DEPs) were quantified in the incubated for 24 h compared with the incubated for 2 h, of which 516 DEPs were up-regulated and 775 DEPs were down-regulated. Among them, S-ribosomal homocysteine lyase (luxS), aminotransferase (araT), and lactate dehydrogenase (ldh) are the key proteins for PLA formation. The DEPs were mainly involved in the QS pathway and the core pathway of PLA synthesis. Furanone effectively inhibited the production of L. plantarum L3 PLA. In addition, Western blot analysis demonstrated that luxS, araT, and ldh were the key proteins regulating PLA production. This study reveals the regulatory mechanism of PLA based on the LuxS/AI-2 QS system, which provides a theoretical basis for the efficient and large-scale production of PLA in industries in the future.

Coenzyme self-sufficiency system-recent advances in microbial production of high-value chemical phenyllactic acid.

Phenyllactic acid (PLA), a natural antimicrobial substance, has many potential applications in the food, animal feed, pharmaceutical and cosmetic industries. However, its production is limited by the complex reaction steps involved in its chemical synthesis. Through advances in metabolic engineering and synthetic biology strategies, enzymatic or whole-cell catalysis was developed as an alternative method for PLA production. Herein, we review recent developments in metabolic engineering and synthetic biology strategies that promote the microbial production of high-value PLA. Specially, the advantages and disadvantages of the using of the three kinds of substrates, which includes phenylpyruvate, phenylalanine and glucose as starting materials by natural or engineered microbes is summarized. Notably, the bio-conversion of PLA often requires the consumption of expensive coenzyme NADH. To overcome the issues of NADH regeneration, efficiently internal cofactor regeneration systems constructed by co-expressing different enzyme combinations composed of lactate dehydrogenase with others for enhancing the PLA production, as well as their possible improvements, are discussed. In particular, the construction of fusion proteins with different linkers can achieve higher PLA yield and more efficient cofactor regeneration than that of multi-enzyme co-expression. Overall, this review provides a comprehensive overview of PLA biosynthesis pathways and strategies for increasing PLA yield through biotechnology, providing future directions for the large-scale commercial production of PLA and the expansion of downstream applications.

Impact of the Inoculation Method of Geotrichum candidum, Used as Biocontrol Agent, on T-2 Toxin Produced by Fusarium sporotrichioides and F. langsethiae during the Malting Process.

In malt production, steeping and germination steps offer favorable environmental conditions for fungal proliferation when barley is already contaminated by Fusarium species, T-2 toxin producers. However, the use of G. candidum as a biocontrol agent can prevent this proliferation. Indeed, in previous work, a correlation between phenyllactic acid (PLA) production by G. candidum and the reduction in Fusarium sporotrichioides and F. langsethiae growth and T-2 toxin concentration was demonstrated. In the present study, to improve the efficiency of G. candidum, the effects of the inoculum concentration and the inoculation method of G. candidum on PLA and T-2 toxin concentrations were evaluated. First, co-culture experiments with Fusarium species and G. candidum were conducted in a liquid synthetic medium. The results showed that inoculation of G. candidum in the freeze-dried form at 0.4 g/L allowed the production of PLA from the second day of incubation associated with a reduction in T-2 toxin concentration of 82% and 69% produced by F. sporotrichioides and F. langsethiae, respectively. Moreover, the activated form of G. candidum at 0.4 g/L enhanced PLA concentration leading to better T-2 toxin reduction. Second, experiments were conducted on artificially infected barley kernels with both Fusarium species under conditions mimicking the malting step. As for co-culture experiments, the use of the activated form of G. candidum was established as the best condition for T-2 toxin concentration reduction for a 3 day malting period.

Preparation and characterization of semi-hydrophobic cryogels for culture of Lactobacillus strains and bioconversion towards phenyllactic acid bioproduction

Semi-hydrophobic poly(2-hydroxyethyl methacrylate-butyl methacrylate) cryogels were prepared via cryo-copolymerization and characterized with a capillary-based model. These cryogels were used as carriers for culture of Lactobacillus cells to prepare cell-loaded cryogels, which were employed as novel biocatalysts for production of phenyllactic acid. Lactobacillus cells were grown successfully within cryogels and the cell production was enhanced. The maximum cell concentrations of 32.7, 38.7 and 30.8 g/L were achieved with cryogels as cell carriers in the cultivation and co-culture of L. casei and L. paracasei, which were 18.2%, 37.5% and 22.0% higher than those without cryogels. The maximum yield from phenylalanine to phenyllactic acid with the cell-loaded cryogels by co-culture of two strains was 5.4 and 4.2 times of those with the cell-loaded cryogels by culture of single strain of L. casei and L. paracasei, respectively. The results indicated that semi-hydrophobic cryogels could be interesting carriers for phenyllactic acid biosynthesis with Lactobacillus strains directly from the cheap precursor, phenylalanine.

Production of Phenyllactic Acid from Porphyra Residues by Lactic Acid Bacterial Fermentation

The concept of algae biorefinery is attracting attention because of the abundant valuable compounds within algal biomass. Phenyllactic acid (PhLA), a broad-spectrum antimicrobial organic acid that can be produced by lactic acid bacteria (LAB), is considered a potential biopreservative. In this study, a cascading biorefinery approach was developed to harvest PhLA from Porphyra residues by LAB fermentation. LAB strains were cultivated in de Man, Rogosa and Sharpe (MRS) broth to screen their ability to produce PhLA. As the strains of Lactobacillus plantarum KP3 and L. plantarum KP4 produced higher concentrations of PhLA at 0.09 mg/mL, these two strains were employed for fermentation. After phycobiliprotein extraction, the Porphyra residues, ultrafiltration eluate, phenylalanine (Phe) and yeast extract with a volume of 20 mL were inoculated with LAB strain KP3 and fermented at 37 °C for 120 h. The PhLA content of the fermented broth was 1.86 mg. To optimize the biorefinery process, the ultrafiltration eluate was replaced by commercial cellulase. Up to 4.58 mg of PhLA, which was 2.5 times greater than that produced from KP3 cultured in MRS broth, could be harvested. Taken together, the findings provide an optimized process for LAB fermentation, which is shown to be a feasible algae biorefinery approach to obtaining PhLA from Porphyra residues.

Optimization and Multiomic Basis of Phenyllactic Acid Overproduction by Lactobacillus plantarum.

The goal of this study was to explore the regulatory mechanisms of phenyllactic acid (PLA) overaccumulation in Lactobacillus plantarum. The dynamics of PLA production revealed that 24 h was a suitable fermentation time, at which one of the largest differences in PLA content between strains S1 and YM-4-3 was 22.42 mg/L. Additionally, an optimization experiment showed that PLA production under the optimal condition (sample YM-4-3y) was up to 400.74 mg/L, 7.61-13.26 times as those of YM-4-3 and S1. Subsequently, an integrated analysis of genomic, transcriptomic and metabolomic data revealed that, YM-4-3 and YM-4-3y, compared with S1, although lacking a complete de novo biosynthetic pathway, increased PLA production by strengthening the core pathway and central carbon metabolism, and weakening the biosynthesis pathway of amino acids and their derivatives. These changes can provide sufficient precursors and compensate for or balance the energy consumed by the reinforced core pathway.

Effect of lactic acid bacteria on phenyllactic acid production in kimchi

Phenyllactic acid (2-hydroxy-3-phenyl propionic acid; PLA) is a natural antibacterial compound derived from phenylalanine catabolism. PLA shares a metabolic pathway with lactic acid (LA) and is metabolized by the glycolytic enzyme, lactate dehydrogenase, during fermentation. In this study, we identified PLA in fermented kimchi and investigated its relationship with kimchi-derived lactic acid bacteria (LAB) during kimchi fermentation using liquid chromatography-mass spectrometry. PLA was detected in four kimchi samples to different extents depending on the initial glucose concentration and fermentation stage, indicating a link between bacterial growth and PLA production. PLA content in the kimchi was 12.0–21.1 μg/ml at the early stage and decreased to 4.8–9.5 μg/ml at 3 or 4 weeks. PLA production in LAB was associated with the presence of lactate dehydrogenase genes, and differed from lactic acid production. Bacteria lacking the d-lactate dehydrogenase gene (2-Hacid dh) did not produce PLA. Addition of Lactobacillus plantarum enhanced PLA production 1.7-fold, reaching a maximum at 14 days. Lactobacillus brevis and Leuconostoc lactis caused increases of approximately 1.5-fold compared to the control. These results suggested that addition of specific LAB to kimchi as starter cultures can increase PLA content, and has great potential to enhance safety of the food product.

Role of Glucose and 2-Oxoglutarate/Malate Translocator (OMT1) in the Production of Phenyllactic Acid and p-Hydroxyphenyllactic Acid, Two Food-Borne Pathogen Inhibitors.

This work aims to uncover how glucose affected the production of phenyllactic acid (PLA) and p-hydroxyphenyllactic acid ( p-OH-PLA). The highest yields of PLA (68.53 mg/L) and p-OH-PLA (50.39 mg/L) were observed after Lactobacillus plantarum strain YM-4-3 fermentation in media containing 30 and 10 g/L glucose, respectively. Additionally, the antimicrobial activity of YM-4-3 against food-borne pathogens and the NADH/NAD+ ratio were positively correlated with the production of PLA and p-OH-PLA, respectively. In addition, a 2-oxoglutarate/malate translocator coding gene ( Omt1) was selected based on the qPCR results, and its knockout mutant, compared with the wild-type strain YM-4-3, showed that the PLA and p-OH-PLA production was decreased by 1.37-6.99 and 1.53-1.59 times, respectively. This result indicated that OMT1 was involved in the biosynthesis of PLA and p-OH-PLA. To conclude, this study suggests that glucose, NADH/NAD+ ratio and/or the Omt1 gene, PLA, and p-OH-PLA production, and antimicrobial activity contribute to a cause-and-effect relationship.

Combination of multi-enzyme expression fine-tuning and co-substrates addition improves phenyllactic acid production with an Escherichia coli whole-cell biocatalyst

The aim of this study was to develop an environmentally safe and efficient method for phenyllactic acid (PLA) production using whole-cell cascade catalysis with l-amino acid deaminase (l-AAD), lactate dehydrogenase (LDH), and formate dehydrogenase (FDH). The PPA titer was low due to relatively low expression of LDH, intermediate accumulation, and lack of cofactors. To address this issue, ribosome binding site regulation, gene duplication, and induction optimization were performed to increased the PLA titer to 43.8 g/L. Then co-substrates (glucose, yeast extract, and glycerol) were used to increase NADH concentration and cell stability, resulting that the PLA titer was increased to 54.0 g/L, which is the highest reported production by biocatalyst. Finally, glucose was replaced with wheat straw hydrolysate as co-substrate to decrease the cost. Notably, the strategies reported herein may be generally applicable to other whole-cell cascade biocatalysts.

Enhanced Phenyllactic Acid Production in Escherichia coli Via Oxygen Limitation and Shikimate Pathway Gene Expression.

3-Phenyllactic acid (PhLA) is useful as a start-up material in the pharmaceutical and biorefinery industries. To enhance the production of PhLA from glucose using recombinant Escherichia coli, the effects of glucose concentration and oxygen limitation on PhLA production are assessed in a fed-batch system using dissolved oxygen (DO)-stat method. The highest titer of PhLA (7.3 g L-1 ) is observed with a high concentration of glucose and under oxygen-limited conditions (DO = 0 ppm). Under oxygen limitation, cell growth and the formation of acetate and l-phenylalanine (Phe) by-products after 72 h of cultivation are reduced by 30%, 70%, and 81%, respectively, as compared to that under high DO conditions (DO = 5 ppm). Gene expression levels are compared between low and high DO conditions by quantitative polymerase chain reaction (qPCR) analysis. Several genes in the glycolysis (gapA and pykA), pentose phosphate (tktA), and early shikimate pathways for PhLA biosynthesis (aroF, aroG, and aroH) are upregulated under oxygen limitation. The results suggest that oxygen limitation affects metabolism in the shikimate pathway at both metabolic and transcriptional levels and that controlling the DO level is critical for enhanced production of a variety of aromatic compounds through the shikimate pathway.

A new approach for efficient synthesis of phenyllactic acid from L-phenylalanine: Pathway design and cofactor engineering

Phenyllactic acid (PLA), a novel biological antiseptic agent with broad and effective antimicrobial activity, finds wide applications in the food processing field. In this work, a new approach for biocatalytic synthesis of PLA from l-phenylalanine (Phe) was constructed. First, selection of l-lactate dehydrogenase (LDH) was done to achieve the efficient synthesis of PLA from phenylpyruvic acid (PPA); Second, l-amino acid deaminase (l-AAD) and LDH were co-expressed to achieve the synthesis of PLA from Phe; Finally, formate dehydrogenase (FDH) was overexpressed to accelerate the regeneration of cofactor NADH from NAD, which is necessary for the biocatalysis of LDH. By this biocatalytic system, 30 g·L−1 Phe was transformed into PLA with 100% conversion ratio in 12 hr. This work developed a new and efficient biocatalytic approach for PLA production with Phe as a cheap substrate, which provides an alternative for PLA production in a green manner. Practical applications PLA is a novel antimicrobial compound with broad inhibition activity and has less side-effect on the health of human being. It is an ideal antiseptic that has received much attention in the food industry, for the control of microbial contaminants. In addition, PLA can be used as feed additives to replace antibiotics in livestock feeds. The newly established biosynthetic system in this work may be a promising technique for the industrial production of PLA due to high production and high efficiency.

Enhanced biosynthesis of chiral phenyllactic acid from L-phenylalanine through a new whole-cell biocatalyst.

Phenyllactic acid (PLA) is a high-value compound, which was usually produced by lactic acid bacteria (LAB) as biocatalysts and glucose or phenylpyruvic acid (PPA) as starting materials for PLA synthesis in previous studies. However, the PLA produced using LAB is a racemic mixture. Besides, both glucose and PPA were unsatisfactory substrates, as the former could not produce high concentrations of PLA while the latter is not a renewable and green substrate. To overcome these drawbacks, in this study, a new biotransformation process was developed for chiral PLA production from L-phenylalanine via the intermediate PPA using recombinant Escherichia coli co-expressing L-amino acid deaminase, NAD-dependent L-lactate dehydrogenase or NAD-dependent D-lactate dehydrogenase, and formate dehydrogenase. After optimization, the recombinant E. coli produced L- and D-PLA at concentrations of 59.9 and 60.3mM in 6h, respectively. Hence, this process provides an effective and promising alternative method for chiral PLA production.

Optimization of phenyllactic acid production by Pediococcus acidilactici CRL 1753. Application of the formulated bio-preserver culture in bread

Phenyl lactic acid (PLA) is an antimicrobial agent with a broad antimicrobial spectrum produced mainly by lactic acid bacteria. This study focused in the optimization of a low-cost medium to obtain biomass and PLA production by Pediococcus (Ped.) acidilactici CRL 1753 strain and propose its use as a liquid bio-preserver in bread making to increase the shelf life of wheat bread. First, the activity of CRL 1753 strain against filamentous fungi and yeasts was study and a wide antifungal inhibitory spectrum was observed. Using a factorial design 29-5 an optimized and low-cost medium (10 g/L meat peptone, 5 g/L yeast extract, 10 g/L glucose, 0.0038 g/L MnSO4, 6 g/L K2HPO4 and initial pH = 7.0) that could replace MRS was obtained. A significant increase in shelf life was observed when calcium propionate (CP) and CRL 1753 strain were combined into the bread formulation. In fact, at 18 days of storage none moulds were observed when the liquid bio-preserver was added while 70% of breads were spoilage with CP alone. In this work we report for the first time the obtaining of an optimized low-cost culture medium for biomass and PLA production by Ped. acidilactici strain. The results obtained could be very useful as a basis to design new strategies that allow improve the PLA production at a low cost.

Chromatographic separation of phenyllactic acid from crude broth using cryogels with dual functional groups

Phenyllactic acid (PLA) is an important organic acid with wide antimicrobial activities against gram-positive and gram-negative bacteria and some fungi. This interesting compound can be synthesized by the microbial fermentation or the bioconversion using phenylpyruvic acid (PPA) as the key substrate and microorganisms as the whole-cell biocatalysts. However, the isolation of high-purity PLA with a high recovery from the crude fermentation or conversion broth is a challenging task. In this work, the separation of PLA from the crude conversion broth prepared by employing Lactobacillus buchneri cells as the whole-cell catalysts was achieved by the chromatography using the poly(hydroxyethyl methacrylate) (pHEMA)-based cryogel with a combination of anion-exchange and hydrophobic benzyl groups. The static adsorption behaviors of PLA under different salt concentrations and the adsorption capacities of PLA on the cryogel were measured experimentally. The chromatographic performance of PLA from the crude conversion broth was compared with that from the clarified broth. The results showed that the pHEMA-based cryogel has a high capacity of PLA, i.e., 14.64 mg mL−1 cryogel, and the adsorption of PLA was influenced by the salt concentration. By using deionized water as running buffer, PLA with a high purity of 97.6% was obtained with one step elution using 0.3 M NaCl as the elution solution with the recovery at the range of 80.2–90.8% from crude feedstock without any pretreatment at various flow velocities. These values were close to those obtained for the clarified broth, i.e., the purity of 98.4% and the recovery of 92.3% under the same chromatography conditions at 1 cm min−1. The cryogel was then applied to separate PLA from clarified feedstock, high purity (>96.7%) and recovery (>91.4%) of PLA were found with 20 cycles, which verified the selectivity and robustness of prepared pHEMA-VBTAC cryogel. Therefore, the chromatography using pHEMA-based cryogel with the dual functional groups is an effective approach for the isolation of PLA directly from the crude bioconversion broth and thus could be interesting in the separation and production of high-purity PLA in industry.

Optimizational production of phenyllactic acid by a Lactobacillus buchneri strain via uniform design with overlay sampling methodology

Abstract A Lactobacillus buchneri GBS3 strain isolated from the traditional Chinese pickles was used for the production of 3-phenyllactic acid (PLA), an important compound with antimicrobial activities against a wide species of gram-positive and gram-negative bacteria and some fungi. The growth performance of this strain in the de Man, Rogosa and Sharpe (MRS) medium, the production of metabolites of valuable organic acids, and the biosynthesis of PLA using this strain as the whole-cell biocatalyst and phenylpyruvic acid (PPA) as the precursor, were investigated experimentally. The uniform design method with overlay sampling was developed for the optimization of the biotransformation conditions. The results showed that although it produced naturally lactic acid with the maximum concentration of 1.84 g·L− 1 and PLA with the concentration of 0.015 g·L− 1 after 66 to 72 h cultivation in MRS broth by fermentation, the present strain displayed an effective utilization ability by transforming PPA to PLA. By the uniform design method with overlay sampling for the design and optimization of transformation conditions, a maximum yield of 10.93 g·L− 1 PLA with the mole conversion ratio of 83.07% from PPA to PLA was achieved under the optimized condition, i.e., 20 g·L− 1 glucose, 270 g·L− 1 cells, 13 g·L− 1 PPA, pH 8.0 and the reaction time of 15 h, indicating that Lactobacillus buchneri GBS3 was an interesting strain for the biosynthesis of PLA via the microbial transformation. The prediction of PLA yield under different conditions was achieved successfully based on the limited information of only a small number of experiments by the uniform design with overlay sampling. Therefore, the present methodology is effective and helpful for the optimization of the biosynthesis processes of PLA.

Fundamental study on the improvement of the antifungal activity of Lactobacillus reuteri R29 through increased production of phenyllactic acid and reuterin

Lactic acid bacteria have shown great potential as bio-preservative agents to maintain high food quality and safety. The strain Lactobacillus reuteri R29 is reported to have a broad spectrum of antifungal activity and suitability for application in food systems. Its antifungal activity is predominantly based on phenyllactic acid (PLA). Furthermore, it is potentially a producer of reuterin, a potent antimicrobial agent. This study focused on increasing the antifungal activity in vitro by supplementation of the growth medium with phenylalanine and glycerol to increase the yield of PLA and reuterin, respectively. For PLA, the addition of 1.5% phenylalanine (w/v) to MRS, resulted in significantly increased accumulation of PLA and antifungal performance against Fusarium culmorum. Supplementation of MRS with 500mM glycerol combined with a reduced glucose content (1.5%) showed the highest reuterin accumulation combined with fungal inhibition. To investigate the antifungal performance in situ, these cell-free supernatants (cfs) were applied in a bread system. The application of PLA-enriched cfs resulted in significantly extended (4 days) microbial shelf life compared to the control. The reuterin-enriched medium did not lead to significant shelf life extension. In conclusion, Lactobacillus reuteri R29 and its PLA-enriched cfs were found to be very promising alternatives for food bio-preservation.

Novel strategy for phenyllactic acid biosynthesis from phenylalanine by whole cell recombinant Escherichia coli coexpressing L-phenylalanine oxidase and L-lactate dehydrogenase.

To enhance the efficiency of phenyllactic acid (PLA) production from L-phenylalanine (L-Phe) by introducing a novel artificial pathway into Escherichia coli RESULTS: The production of PLA from L-Phe by recombinant E. coli (ldh-lpox) coexpressing L-phenylalanine oxidase and L-lactate dehydrogenase was studied. The new PLA synthesis pathway was confirmed to be efficient in recombinant E. coli. Subsequently, two different biocatalyst processes were carried out and optimized for PLA production. In the whole cell biosynthesis process at high cell density using collected recombinant cells as catalyst, at optimal conditions (L-Phe 6g/l, pH 7.5, 35°C, CDW 24.5g/l and 200rpm), the recombinant E. coli (ldh-lpox) produced 1.62g PLA/l with a conversion of 28% from L-Phe. Similarly, during the two-temperature-stage fermentation process in flasks using IPTG-induced cells, the temperature in the second stage was increased to 35°C to benefit the biocatalyst process, and comparable phenyllactic acid production of 1.47g/l was obtained from 12g L-Phe/l. Recombinant E. coli (ldh-lpox) was efficient in PLA production realizing a high titer of several folds compared with studies using L-Phe as substrate.

Two-step biocatalytic reaction using recombinant Escherichia coli cells for efficient production of phenyllactic acid from l-phenylalanine

Abstract 3-Phenyllactic acid (PLA) is a natural product with broad and effective antimicrobial activity that is often used as a preservative in the food industry and as feed additive to replace antibiotics in livestock feeds. In this study, a two-step biocatalytic process for production of PLA from l-phenylalanine was developed and investigated. In the first step, l-phenylalanine was converted to phenylpyruvic acid (PPA) by recombinant Escherichia coli cells expressing membrane-bound l-amino acid deaminase from Proteus vulgaris. In the second step, unpurified PPA was directly converted to PLA by recombinant E. coli cells co-expressing d-2-hydroxyisocaproate dehydrogenase from Lactobacillus delbrueckii subsp. bulgaricus and formate dehydrogenase from Mycobacterium vaccae N10. Through two-step cascade reactions, 121 mM PLA was produced from 150 mM l-phenylalanine with a yield of 81.3%. Thus, this process showed great production efficiency and is a promising method for industrial production of PLA.

Lactobacillus plantarum with broad antifungal activity: A promising approach to increase safety and shelf-life of cereal-based products

Cereal-based fermented products are worldwide diffused staple food resources and cereal-based beverages represent a promising innovative field in the food market. Contamination and development of spoilage filamentous fungi can result in loss of cereal-based food products and it is a critical safety concern due to their potential ability to produce mycotoxins. Lactic Acid Bacteria (LAB) have been proposed as green strategy for the control of the moulds in the food industry due to their ability to produce antifungal metabolites. In this work, eighty-eight Lactobacillus plantarum strains were screened for their antifungal activity against Aspergillus niger, Aspergillus flavus, Fusarium culmorum, Penicillium roqueforti, Penicillium expansum, Penicillium chrysogenum, and Cladosporium spp. The overlayed method was used for a preliminary discrimination of the strains as no, mild and strong inhibitors. L. plantarum isolates that displayed broad antifungal spectrum activity were further screened based on the antifungal properties of their cell-free supernatant (CFS). CFSs from L. plantarum UFG 108 and L. plantarum UFG 121, in reason of their antifungal potential, were characterized and analyzed by HPLC. Results indicated that lactic acid was produced at high concentration during the growth phase, suggesting that this metabolic aptitude, associated with the low pH, contributed to explain the highlighted antifungal phenotype. Production of phenyllactic acid was also observed. Finally, a new oat-based beverage was obtained by fermentation with the strongest antifungal strain L. plantarum UFG 121. This product was submitted or not to a thermal stabilization and artificially contaminated with F. culmorum. Samples containing L. plantarum UFG 121 showed the best biopreservative effects, since that no differences were observed in terms of some qualitative features between not or contaminated samples with F. culmorum. Here we demonstrate, for the first time, the suitability of LAB strains for the fermentation and antifungal biopreservation of oat-based products.
