Exploring the three-dimensional vasculature of dermal hard tissues in thyestiid osteostracans using synchrotron radiation microtomography

ABSTRACT Osteostracans are stem-gnathostomes with bony tissues that represent an important link between living jawed and jawless vertebrates, the latter of which lack skeletal hard tissues. In this study, a number of skeletal microremains from a diverse group of osteostracans called thyestiids have been investigated using propagation phase-contrast X-ray synchrotron microtomography. This enabled detailed reconstructions of their three-dimensional vasculature. The dermal skeleton of thyestiids is divided into three layers that vary in proportion between taxa, similar to gnathostomes and other osteostracans. The basal layer is confirmed as an acellular bony tissue composed of fiber bundles that contains narrow, vertical canals and/or large cavities. Bundles bend around smaller canals but form jagged edges around larger cavities, which suggests delayed mineralization. The presumed vascular mesh canals of the deep middle layer are radially arranged in some taxa, but more irregular or even tree-like in others, suggesting a relation to how the elements developed and grew. A number of mesh canals ascend toward the superficial layer where they supply the subepidermal vascular plexus, or create pore fields that open to the surface in some taxa. These ascending mesh canals most likely expanded and formed a network of canals underneath more extensive pore fields. Aestiaspis viitaensis has a simple upper canal system above these fields, while Tremataspis mammillata and Tr. milleri scales have a polygonal network of upper mesh canals above extensive perforated septa. These systems are most likely homologous, but their relation to intra- and inter-areal canals described in other osteostracans remains ambiguous.

Dental cementum virtual histology of Neanderthal teeth from Krapina (Croatia, 130-120 kyr): an informed estimate of age, sex and adult stressors.

The evolution of modern human reproductive scheduling is an aspect of our life history that remains vastly uncomprehended. The present work aims to address this gap by validating a non-destructive cutting-edge methodology to infer adult life-history events on modern teeth with known life history and then applying it to fossil specimens. We use phase-contrast synchrotron X-ray microtomography to visualize the dental cementum of 21 specimens: nine contemporary humans; 10 Neanderthals from Krapina (Croatia, 130-120 kyr); one Neolithic Homo sapiens from Ajmana (Serbia); and one Mesolithic H. sapiens from Vlasac (Serbia). We were able to correctly detect and time (root mean square error = 2.1 years; R2 = 0.98) all reproductive (menarche, parturition, menopause) and other physiologically impactful events in the modern sample. Nonetheless, we could not distinguish between the causes of the events detected. For the fossil specimens, we estimated age at death and age at occurrence of biologically significant events. Finally, we performed an exploratory analysis regarding possible sexual dimorphism in dental cementum microstructure, which allowed us to correctly infer the sex of the Neolithic specimen, for which the true value was known via DNA analysis.

Paleohistology of the Cretaceous resin-producing conifer Geinitzia reichenbachii using X-ray synchrotron microtomography.

The conifer Geinitzia reichenbachii was a common member of the Cretaceous Laurasian floras. However, the histology of G. reichenbachii leafy axes was never described in detail, and our knowledge of its paleoecology remains very limited. Using new and exquisitely preserved silicified material from the Upper Cretaceous of western France, we describe G. reichenbachii from the gross morphology to the cellular scale, then discuss paleoecological and taphonomical implications. We examined specimens from two localities in western France (Claix and Moragne) using propagation phase-contrast X-ray synchrotron microtomography. The cuticle and the inner tissues of leafy axes are preserved in three dimensions. Epidermis, hypodermis, palisade parenchyma, spongy parenchyma, transfusion tracheids, and most of tissues of the vascular cylinder are clearly discernible. The numerous resin ducts are sometimes filled by persisting resin. Additionally, surfaces of some leaves preserved drops and flows of resin. Depositional environmental context combined with histological features of G. reichenbachii suggest that this conifer was adapted to a range of marginal-littoral ecosystems including those open to the sea paleoenvironments and innermost ones influenced by strong continental inputs. Geinitzia reichenbachii was adapted to withstand intense sunlight, hot temperatures coupled with salty sea wind, and dry conditions. The frequent amber-bearing beds in the Cretaceous from western France with Geinitzia as a main component of the associated floras raises the possibility of a role for Geinitzia in the production of the "Charentese amber".

A parasitic coevolution since the Miocene revealed by phase-contrast synchrotron X-ray microtomography and the study of natural history collections

The discovery of a new fossil species of the Caribbeo-Mexican genus Proptomaphaginus (Coleoptera, Leiodidae, Cholevinae) from Dominican amber, associated with a new fossil parasitic fungus in the genus Columnomyces (Ascomycota, Laboulbeniales), triggered an investigation of extant species of Proptomaphaginus and revealed the long-enduring parasitic association between these two genera. This effort resulted in the description of the fossil species †Proptomaphaginus alleni sp. nov., and one fossil and two extant species of Columnomyces, selectively associated with species of Proptomaphaginus: †Columnomyces electri sp. nov. associated with the fossil †Proptomaphaginus alleni in Dominican amber, Columnomyces hispaniolensis sp. nov. with the extant Proptomaphaginus hispaniolensis (endemic of Hispaniola), and Columnomyces peckii sp. nov. with the extant Proptomaphaginus puertoricensis (endemic of Puerto Rico). Based on biogeography, our current understanding is that the Caribbean species of Proptomaphaginus and their parasitic species of Columnomyces have coevolved since the Miocene. This is the first occurrence of such a coevolution between a genus of parasitic fungus and a genus of Coleoptera. The phylogenetic relations among Proptomaphaginus species are also addressed based on a parsimony analysis. Fossil specimens were observed by propagation phase-contrast synchrotron X-ray microtomography (PPC-SRμCT) and extant specimens were obtained through the study of preserved dried, pinned insects, attesting for the importance of (i) technological advancement and (ii) natural history collections in the study of microparasitic relationships.

The spatiotemporal spread of cervical spinal cord contusion injury pathology revealed by 3D in-line phase contrast synchrotron X-ray microtomography

Extensive structural changes occur within the spinal cord following traumatic injury. Acute tissue debris and necrotic tissue are broken down, proliferating local glia and infiltrating leukocytes remodel tissue biochemical and biophysical properties, and a chronic cavity surrounded by a scar forms at the injury epicentre. Serial-section 2D histology has traditionally assessed these features in experimental models of spinal cord injury (SCI) to measure the extent of tissue pathology and evaluate efficacy of novel therapies. However, this 2D snapshot approach overlooks slice intervening features, with accurate representation of tissue compromised by mechanical processing artefacts. 3D imaging avoids these caveats and allows full exploration of the injured tissue volume to characterise whole tissue pathology. Amongst 3D imaging modalities, Synchrotron Radiation X-ray microtomography (SRμCT) is advantageous for its speed, ability to cover large tissue volumes at high resolution, and need for minimal sample processing. Here we demonstrate how extended lengths of formalin-fixed, paraffin-embedded (FFPE) rat spinal cord can be completely imaged by SRμCT with micron resolution. Label-free contrast derived from X-ray phase interactions with low-density soft tissues, reveals spinal cord white matter, gray matter, tissue damage and vasculature, with tissue still viable for targeted 2D-histology after 3D imaging. We used SRμCT to quantify tissue pathology after a midline, cervical level (C6), 225 kDyne contusion injury over acute-to-chronic (24 h to 5 weeks) post injury time points. Quantification revealed acute tissue swelling prior to chronic atrophy across the whole imaged region (spanning 2 spinal segments above and below injury), along with rostro-caudal asymmetries in white and gray matter volume loss. 3D volumes revealed satellite damage in tissue far removed from the epicentre, and extensive rostro-caudal spread of damage through the base of the dorsal columns at 24 h post injury. This damage overlapped regions of vasogenic oedema, confirmed with subsequent histology. Tissue damage at later time points in border regions was most prominent in the dorsal columns, where it overlapped sites of damaged venous vasculature. Elaborating rostro-caudal and spatiotemporal asymmetries in reduced traumatic injury models centred on these regions may inform future treatments that seek to limit the spread of tissue pathology to these ‘at-risk’ regions.

Synchrotron “virtual archaeozoology” reveals how Ancient Egyptians prepared a decaying crocodile cadaver for mummification

Although Ancient Egyptians mummified millions of animals over the course of one millennium, many details of these mummification protocols remain unknown. Multi-scale propagation phase-contrast X-ray synchrotron microtomography was used to visualise an ancient Egyptian crocodile mummy housed at the Musée des Confluences (Lyon, France). This state-of-the-art non-destructive imaging technique revealed the complete interior anatomy of the mummy in three dimensions. Here, we present detailed insight into the complex post-mortem treatment of a decaying crocodile cadaver in preparation for mummification. Except for the head and the extremities of the limbs, everything beneath the skin of the crocodile (i.e. organs, muscles, and even most of the skeleton) was removed to cease further putrefaction. This unexpected finding demonstrates that earlier knowledge obtained from textual and other archaeological sources does not sufficiently reflect the diversity of mummification protocols implemented by Ancient Egyptians.

Exceptional soft tissues preservation in a mummified frog-eating Eocene salamander.

Fossils are almost always represented by hard tissues but we present here the exceptional case of a three-dimensionally preserved specimen that was ‘mummified’ (likely between 40 and 34 million years ago) in a terrestrial karstic environment. This fossil is the incomplete body of a salamander, Phosphotriton sigei, whose skeleton and external morphology are well preserved, as revealed by phase-contrast synchrotron X-ray microtomography. In addition, internal structures composed of soft tissues preserved in three dimensions are now identified: a lung, the spinal cord, a lumbosacral plexus, the digestive tract, muscles and urogenital organs that may be cloacal glands. These are among the oldest known cases of three-dimensional preservation of these organs in vertebrates and shed light on the ecology of this salamander. Indeed, the digestive tract contains remains of a frog, which represents the only known case of an extinct salamander that fed on a frog, an extremely rare type of predation in extant salamanders. These new data improve our scarce knowledge on soft tissue anatomy of early urodeles and should prove useful for future biologists and palaeontologists working on urodele evolutionary biology. We also suggest that the presence of bat guano and carcasses represented a close source of phosphorus, favouring preservation of soft tissues. Bone microanatomy indicates that P. sigei was likely amphibious or terrestrial, and was probably not neotenic.

Virtual histological assessment of the prenatal life history and age at death of the Upper Paleolithic fetus from Ostuni (Italy)

The fetal remains from the Ostuni 1 burial (Italy, ca 27 ka) represent a unique opportunity to explore the prenatal biological parameters, and to reconstruct the possible patho-biography, of a fetus (and its mother) in an Upper Paleolithic context. Phase-contrast synchrotron X-ray microtomography imaging of two deciduous tooth crowns and microfocus CT measurements of the right hemimandible of the Ostuni 1b fetus were performed at the SYRMEP beamline and at the TomoLab station of the Elettra - Sincrotrone laboratory (Trieste, Italy) in order to refine age at death and to report the enamel developmental history and dental tissue volumes for this fetal individual. The virtual histology allowed to estimate the age at death of the fetus at 31–33 gestational weeks. Three severe physiological stress episodes were also identified in the prenatal enamel. These stress episodes occurred during the last two months and half of pregnancy and may relate to the death of both individuals. Compared with modern prenatal standards, Os1b’s skeletal development was advanced. This cautions against the use of modern skeletal and dental references for archaeological finds and emphasizes the need for more studies on prenatal archaeological skeletal samples.

100-million-year-old conifer tissues from the mid-Cretaceous amber of Charente (western France) revealed by synchrotron microtomography.

Terrestrial plant remains in fossilized tree resin are relatively common. However, histology and preservation of plants entombed in Cretaceous ambers remain poorly known. We report an exquisitely preserved conifer leafy axis from 100-million-year-old opaque amber of western France that is assignable to Glenrosa carentonensis Moreau, Néraudeau, Tafforeau & Dépré. The purpose of this paper is to discuss the taphonomy and the use of microtomography for studies of palaeobotanical remains in amber. A leafy axis was examined using propagation phase-contrast X-ray synchrotron microtomography with voxel sizes of 14·9, 1·4, and 0·7 μm. The conifer leafy axis described is preserved in three dimensions. Despite desiccation of the specimen within the surrounding amber, the cuticle, as well as most of inner tissues, is preserved in three dimensions down to the cellular level. Epidermis, palisade parenchyma, spongy parenchyma, transfusion tracheids and vascular bundles are clearly distinguished. Gross morphology and histology of the specimen were revealed using synchrotron microtomography, allowing an unprecedented resolution for the study of soft-bodied plants entombed in amber. The study reveals a peculiar combination of authigenic and duripartic preservation as well as permineralization, and highlights the complexity of taphonomic processes that can occur in amber inclusions. This fossil demonstrates the difficulty of studying amber-preserved plant remains under certain conditions.

Three-dimensional paleohistology of the scale and median fin spine of Lophosteus superbus (Pander 1856).

Lophosteus superbus is one of only a handful of probable stem-group osteichthyans known from the fossil record. First collected and described in the late 19th century from the upper Silurian Saaremaa Cliff locality in Estonia, it is known from a wealth of disarticulated scales, fin spines, and bone fragments. In this study we provide the first description of the morphology and paleohistology of a fin spine and scale from Lophosteus using virtual thin sections and 3D reconstructions that were segmented using phase-contrast synchrotron X-ray microtomography. These data reveal that both structures have fully or partially buried odontodes, which retain fine morphological details in older generations, including sharp nodes and serrated ridgelets. The vascular architecture of the fin spine tip, which is composed of several layers of longitudinally directed bone vascular canals, is much more complex compared to the bulbous horizontal canals within the scale, but they both have distinctive networks of ascending canals within each individual odontode. Other histological characteristics that can be observed from the data are cell spaces and Sharpey’s fibers that, when combined with the vascularization, could help to provide insights into the growth of the structure. The 3D data of the scales from Lophosteus superbus is similar to comparable data from other fossil osteichthyans, and the morphology of the reconstructed buried odontodes from this species is identical to scale material of Lophosteus ohesaarensis, casting doubt on the validity of that species. The 3D data presented in this paper is the first for fossil fin spines and so comparable data is not yet available. However, the overall morphology and histology seems to be similar to the structure of placoderm dermal plates. The 3D datasets presented here provide show that microtomography is a powerful tool for investigating the three-dimensional microstructure of fossils, which is difficult to study using traditional histological methods. These results also increase the utility of fin spines and scales suggest that these data are a potentially rich source of morphological data that could be used for studying questions relating to early vertebrate growth and evolution.

Palaeobiology of tanaidaceans (Crustacea: Peracarida) from Cretaceous ambers: extending the scarce fossil record of a diverse peracarid group

Diverse assemblages of tanaidacean peracarid crustaceans from western Tethyan continental deposits suggest that the group was relatively common in or around ancient resin-producing forests. Here we report the results of an examination of 13 tanaidacean specimens from three Cretaceous (Albian to Turonian) French amber deposits. Two new species of the fossil family Alavatanaidae are placed in the previously described Early Cretaceous genus Eurotanais: Eurotanais pyrenaensis sp. nov. from Cenomanian Pyrenean amber (Fourtou, Aude) and Eurotanais seilacheri sp. nov. from Turonian Vendean amber (La Garnache, Vendee). The remaining specimens are placed in three newly erected genera and species (but family incertae sedis): Arcantitanais turpis gen. et sp. nov. from Albian–Cenomanian Charentese amber (Archingeay, Charente-Maritime), and Tytthotanais tenvis gen. et sp. nov. and Armadillopsis rara gen. et sp. nov. from Pyrenean amber. These are the first formally described fossils that might be related to the paratanaoidean families Nototanaidae and Paratanaidae, sharing with these some putatively derived features and providing possible evidence for the antiquity and morphological stability of these families and the suborder Tanaidomorpha. The distinctive features and character combinations of these fossil taxa are discussed in connection with possible relationships to the living lineages of tanaidaceans. Propagation phase-contrast X-ray synchrotron microtomography was used to obtain high-quality 3D images for some fossils. A discussion is provided on the putative palaeobiology of tanaidaceans and the French resiniferous forest ecosystem. The discovery of these new tanaidaceans extends the palaeogeographical distribution and stratigraphical range of the family Alavatanaidae and sheds new light on the palaeoecology and diversity of tanaidaceans in pre-angiospermous woodlands.

A novel protocol for resolving feldspar crystals in synchrotron X-ray microtomographic images of crystallized natural magmas and synthetic analogs

X-ray computed microtomography is a non-destructive imaging technique recognized in the geosciences as a powerful tool to investigate rock textures directly in three dimensions (3D) at the micrometer and sub-micrometer scale. The quantitative morphological and textural analysis of images requires segmentation and characterization of phases in the reconstructed volume based upon their gray levels (related to their relative X-ray attenuation) and/or morphological aspects. Often the differences in X-ray attenuation of some phases are so small that no contrast is observed in the reconstructed slices or, although the human eye can discern the differences between these phases, it is difficult, or sometimes impossible, to reliably segment and separately analyze these phases. Facing this challenge, we propose an experimental and computational procedure that allows the segmentation of phases with small density variations in geomaterials. By using an experimental protocol based on phase-contrast synchrotron X-ray microtomography combined with a customized 3D image processing procedure, we successfully segmented feldspar from the glassy matrix in both a natural volcanic sample and a synthetic analog. Our results demonstrate that crystallized natural volcanic rocks and synthetic analogs can be characterized by synchrotron X-ray phase-contrast microtomography and that phase-retrieval processing is an invaluable tool for the reconstruction of 3D multiphase textures.

Synchrotron imaging of dentition provides insights into the biology of Hesperornis and Ichthyornis, the "last" toothed birds.

BackgroundThe dentitions of extinct organisms can provide pivotal information regarding their phylogenetic position, as well as paleobiology, diet, development, and growth. Extant birds are edentulous (toothless), but their closest relatives among stem birds, the Cretaceous Hesperornithiformes and Ichthyornithiformes, retained teeth. Despite their significant phylogenetic position immediately outside the avian crown group, the dentitions of these taxa have never been studied in detail. To obtain new insight into the biology of these ‘last’ toothed birds, we use cutting-edge visualisation techniques to describe their dentitions at unprecedented levels of detail, in particular propagation phase contrast x-ray synchrotron microtomography at high-resolution.ResultsAmong other characteristics of tooth shape, growth, attachment, implantation, replacement, and dental tissue microstructures, revealed by these analyses, we find that tooth morphology and ornamentation differ greatly between the Hesperornithiformes and Ichthyornithiformes. We also highlight the first Old World, and youngest record of the major Mesozoic clade Ichthyornithiformes. Both taxa exhibit extremely thin and simple enamel. The extension rate of Hesperornis tooth dentine appears relatively high compared to non-avian dinosaurs. Root attachment is found for the first time to be fully thecodont via gomphosis in both taxa, but in Hesperornis secondary evolution led to teeth implantation in a groove, at least locally without a periodontal ligament. Dental replacement is shown to be lingual via a resorption pit in the root, in both taxa.ConclusionsOur results allow comparison with other archosaurs and also mammals, with implications regarding dental character evolution across amniotes. Some dental features of the ‘last’ toothed birds can be interpreted as functional adaptations related to diet and mode of predation, while others appear to be products of their peculiar phylogenetic heritage. The autapomorphic Hesperornis groove might have favoured firmer root attachment. These observations highlight complexity in the evolutionary history of tooth reduction in the avian lineage and also clarify alleged avian dental characteristics in the frame of a long-standing debate on bird origins. Finally, new hypotheses emerge that will possibly be tested by further analyses of avian teeth, for instance regarding dental replacement rates, or simplification and thinning of enamel throughout the course of early avian evolution.Electronic supplementary materialThe online version of this article (doi:10.1186/s12862-016-0753-6) contains supplementary material, which is available to authorized users.

Osteogenic tumour in Australopithecus sediba: Earliest hominin evidence for neoplastic disease

We describe the earliest evidence for neoplastic disease in the hominin lineage. This is reported from the type specimen of the extinct hominin Australopithecus sediba from Malapa, South Africa, dated to 1.98 million years ago. The affected individual was male and developmentally equivalent to a human child of 12 to 13 years of age. A penetrating lytic lesion affected the sixth thoracic vertebra. The lesion was macroscopically evaluated and internally imaged through phase-contrast X-ray synchrotron microtomography. A comprehensive differential diagnosis was undertaken based on gross- and micro-morphology of the lesion, leading to a probable diagnosis of osteoid osteoma. These neoplasms are solitary, benign, osteoid and bone-forming tumours, formed from well-vascularised connective tissue within which there is active production of osteoid and woven bone. Tumours of any kind are rare in archaeological populations, and are all but unknown in the hominin record, highlighting the importance of this discovery. The presence of this disease at Malapa predates the earliest evidence of malignant neoplasia in the hominin fossil record by perhaps 200 000 years.

Lidryopinae, new byrrhid subfamily from the Lower Cretaceous of France (Coleoptera, Byrrhidae)

The byrrhid subfamily Lidryopinae subfam. nov. is proposed for Lidryops occultus gen. et sp. nov. described from Lower Cretaceous “opaque” amber originated from Archingeay-Les Nouillers, Charente-Maritime, southwest France which was studied using propagation phase contrast X-ray synchrotron microtomography at the European Synchrotron Radiation Facility (Grenoble). This byrrhid subfamily differs from members of the closely related groups (Chelonariidae, Limnichidae, Dryopidae, Elmidae, Lutrochidae) in the set of characters including the moderately convex dorsum, the subpectinate antennae, the five-segmented tarsi with strongly lobed tarsomeres 1–4, subsecuriform ultimate maxillary and labial palpomeres. An assumption on probable lifestyle of Lidryops occultus gen. et sp. nov. is proposed on the basis of structural peculiarities. A short review of the byrrhid fossil record and its analysis are made.

The origin of novel features by changes in developmental mechanisms: ontogeny and three-dimensional microanatomy of polyodontode scales of two early osteichthyans.

Recent advances in synchrotron imaging allow us to study the three-dimensional (3D) histology of vertebrate fossils, including microfossils (e.g. teeth and scales) of early jawed vertebrates. These microfossils can often be scanned at submicron resolution (<1 µm) because of their small size. The resulting voxel (3D pixel) stacks can be processed into virtual thin sections revealing almost every internal detail of the samples, comparable to traditional thin sections. In addition, 3D models of the internal microanatomical structures, such as embedded odontodes and vasculature, can be assembled and examined in situ. Scales of two early osteichthyans, Psarolepis romeri from the Early Devonian of China and Andreolepis hedei from the Late Silurian of Sweden, were scanned using propagation phase-contrast synchrotron X-ray microtomography (PPC-SRµCT), and 3D models of internal canal systems and buried odontodes were created from the scans. Based on these new data, we review the evolutionary origin of cosmine and its associated pore-canal system, which has been long recognized as a synapomorphy of sarcopterygians. The first odontode that appeared during growth shows almost identical morphology in the two scales, but the second odontode of the Psarolepis scale shows a distinctive morphology with several pores on the surface. It is suggested that a shift from ridge-like odontode to pore-bearing odontode was the key step in the origin of cosmine, which was then elaborated further in more-derived sarcopterygians. We perform a detailed comparison between the two scales and propose a primary homology framework to generate microanatomical characters, which can be used in the phylogenetic analysis of early osteichthyans when more 3D data become available. Our results highlight the importance of 3D data for the study of histology and ontogeny of the dermal skeleton of early jawed vertebrates, especially scales of the polyodontode type. The traditional microvertebrate collection is not only useful for biostratigraphic studies, but also preserves invaluable biological information about the growth of vertebrate hard tissues. Today, we are only beginning to understand the biological meaning of the new 3D data. The increasing availability of such data will enable, and indeed require, a complete revision of traditional palaeohistological studies on early vertebrates.

New species from Late Cretaceous New Jersey amber and stasis in subfamily Attageninae (Insecta: Coleoptera: Dermestidae)

AbstractA new fossil species of Dermestidae (Insecta: Coleoptera), preserved in Late Cretaceous (Turonian) amber from New Jersey, is described asAttagenus(Aethriostoma)turonianensisn. sp. The specimen is fossilized in translucent amber, but 3D imaging using propagation phase-contrast X-ray synchrotron microtomography allowed detailed classification and description. This species is the oldest representative of the subfamily Attageninae and the third fossil species described in the family from the entire Mesozoic. Dermestidae comprise beetle species that typically feed on carcasses, although some Recent species ofAttagenusLatreille, 1802 are known to feed on plant debris, which is highly abundant in amber deposit sediments. This new species is evidence for diversification in the family during the Early Cretaceous as well as long morphological conservation of diagnostic features of the genusAttagenusfrom the Late Cretaceous. Analyzing the taxa from Mesozoic ambers that show stasis, the small size of the specimens together with a specific ecology could explain the stability of these lineages.

Study of the Histology of Leafy Axes and Male Cones of Glenrosa carentonensis sp. nov. (Cenomanian Flints of Charente-Maritime, France) Using Synchrotron Microtomography Linked with Palaeoecology

We report exceptionally well-preserved plant remains ascribed to the extinct conifer Glenrosa J. Watson et H.L. Fisher emend. V. Srinivasan inside silica-rich nodules from the Cenomanian of the Font-de-Benon quarry, Charente-Maritime, western France. Remains are preserved in three dimensions and mainly consist of fragmented leafy axes. Pollen cones of this conifer are for the first time reported and in some cases remain connected to leafy stems. Histology of Glenrosa has not previously been observed; here, most of internal tissues and cells are well-preserved and allow us to describe a new species, Glenrosa carentonensis sp. nov., using propagation phase-contrast X-ray synchrotron microtomography, a non-destructive technique. Leafy axes consist of characteristic helically arranged leaves bearing stomatal crypts. Glenrosa carentonensis sp. nov. differs from the other described species in developing a phyllotaxy 8/21, claw-shaped leaves, a thicker cuticle, a higher number of papillae and stomata per crypt. Pollen cones consist of peltate, helically arranged microsporophylls, each of them bearing 6–7 pollen sacs. The new high resolution tomographic approach tested here allows virtual palaeohistology on plants included inside a dense rock to be made. Most tissues of Glenrosa carentonensis sp. nov. are described. Lithological and palaeontological data combined with xerophytic features of Glenrosa carentonensis sp. nov. suggest that this conifer has been adapted to survive in harsh and instable environments such as coastal area exposed to hot, dry conditions.

On the possibilities of polychromatic synchrotron radiation microtomography for visualization of internal structures of Rhodnius prolixus

The recent years advancements in microtomography have increased the achievable resolution and contrast, making this relatively inexpensive and a widely available technology, potentially useful for studies of insect’s internal morphology. Phase Contrast X-Ray Synchrotron Microtomography (SR-PhC-µCT) is a non-destructive technique that allows the microanatomical investigations of Rhodnius prolixus, one of the most important insect vectors of Trypanosoma cruzi, the etiologic agent of Chagas’ disease. In Latin America, vector control is the most useful method to prevent Chagas’ disease, and a detailed knowledge of R. prolixus’ interior structures is crucial for a better understanding of their function and evolution. Traditionally, in both biological morphology and anatomy, the internal structures of whole organisms or parts of them are accessed by dissecting or histological serial sectioning; so studying the internal structures of R. prolixus’ head using SR-PhC-µCT is of great importance in researches on vector control. In this work, volume-rendered SR-PhC-µCT images of the heads of selected R. prolixus were obtained using the new set-up available at the SYRMEP beamline of ELETTRA (Trieste, Italy). In this new set-up, the outcoming beam from the ring is restrained before the monochromator and in a devoted end-station, absorption and phase contrast radiography and tomography set-up are available. The images obtained with polychromatic X-ray beam in phase contrast regimen and 2 µm resolution, showed details and organs of R. prolixus never seen before with SR-PhC-µCT.

Three-dimensional virtual histology of Silurian osteostracan scales revealed by synchrotron radiation microtomography.

We used propagation phase contrast X-ray synchrotron microtomography to study the three-dimensional (3D) histology of scales of two osteostracans, Tremataspis and Oeselaspis, members of a jawless vertebrate group often cited as the sister group of jawed vertebrates. 3D-models of the canal systems and other internal structures are assembled based on the virtual thin section datasets and compared with previous models based on real thin sections. The primary homology framework of the canal systems in the two taxa is revised and new histological details are revealed based on the results of this work. There is no separation of vascular canals and lower mesh canals in the Tremataspis scale, contrary to previous results. The secondary upper mesh canals have a limited distribution to the anterior region of the Tremataspis scale. The upper and lower mesh canal systems of Tremataspis have different geometries, inferred to reflect different developmental origins: we interpret the upper system as a probable epithelial invagination, the lower system as entirely vascular. Oeselaspis has no equivalent of the upper mesh canal system. The upper mesh canal system of Tremataspis may have been sensory in function. In Oeselaspis, numerous polyp-shaped structures opening from the canal system onto the surface of the scale resemble the innervation tracts for neuromast organs. The growth of the Oeselaspis scale proceeds by addition of small odontodes containing unmineralized lacunae, which may further mineralize and become more compact. Our results highlight that 3D-histological investigation on scales and other dermal skeletons of osteostracans is necessary to fully appreciate the diversity of skeletal histologies in the group. Traditional 3D-models based on thin sections alone are not reliable and should no longer be used as the basis for homology assessments or functional hypotheses.
