Purification, characterization of a plant esterase from red kidney bean and its feasibility for pesticide residue analysis in foods

Plant-derived esterase is a promising alternative enzyme source for pesticide residue analysis. The present study aimed to obtain purified red kidney bean esterase (RKBE), investigate its biochemical properties and application potential in pesticide residue assay. RKBE was extracted and purified by simple aqueous two-phase extraction combined with dextran gel filtration chromatography, with specific activity of 11.845 U/mg and purification fold of 18.952. SDS-PAGE revealed that RKBE has a molecular weight of 33 kDa. Preliminary structural characterization showed the amino acid sequence of RKBE was similar to that of α/β-hydrolase (ESW18998) derived from Phaseolus vulgaris with 19.5% sequence coverage, and the relative contents of β-sheet, β-turn, α-helix, and random coil were 22.69%, 24.19%, 30.12%, and 23.00%, respectively. The optimal pH and temperature of RKBE were 6.5 and 40 °C. Ca2+, Mg2+, Zn2+, and low concentrations of methanol and ethanol stimulated the enzyme activity. The half-life was 138.6 days at −20 °C. Kinetic studies showed that RKBE exhibited a high affinity and catalytic activity towards 1-naphthyl acetate, with a Km of 0.277 mM and a Kcat/Km of 0.455 s−1 mM−1. Enzyme-specific inhibition assay indicated RKBE to be a carboxylesterase. Finally, pesticide susceptibility experiments confirmed that organophosphorus and carbamate pesticides exerted inhibitory effects on RKBE catalytic activity. For methamidophos, methomyl, dichlorvos, and carbaryl, the limit of detection (IC10) values in cabbage were 0.0031, 0.0045, 0.001, and 0.0065 mg/kg, respectively, suggesting that RKBE can serve as a new, widely available and cost-effective enzyme source for pesticide residual assay in foods.

Evaluation of the Results of Pesticide Residue Analysis in Food Sampled between 2017 and 2021

As mandated by the EU and the national risk management duties, pesticide residues were determined by four specialized laboratories in 9924 samples taken from 119 crops of economic importance in Hungary and imported foodstuffs during 2017–2021. The screening method applied covered 622 pesticide residues as defined for enforcement purposes. The limit of detection ranged between 0.002 and 0.008 mg/kg. The 1.0% violation rate concerning all commodities was lower than in the European Union. No residue was detectable in 45.9% of the samples. For detailed analyses, six commodities (apple, cherry, grape, nectarine/peach, sweet peppers, and strawberry) were selected as they were analyzed in over 195 samples and most frequently contained residues. Besides testing their conformity with national MRLs, applying 0.3 MRL action limits for pre-export control, we found that 73% of the sampled lots would be compliant with ≥90% probability based on a second independent sampling. Multiple residues (2–23) in one sample were detected in 36–50% of the tested lots. Considering the provisions of integrated pest management, and the major pests and diseases of selected crops, normally three to four and exceptionally, seven to nine active ingredients with different modes of action should suffice for their effective and economic protection within four weeks before harvest.

Optimisation and validation of a modified QuEChERS method for the determination of 222 pesticides in edible oils using GC-MS/MS: a case study on corn oil

ABSTRACT Recently, various pesticides have been employed at different stages of cultivation to protect the oil crop from insect, disease, and weed. Those pesticides, especially lipophilic ones, bio-accumulate in the oilseeds and get co-extracted into edible oils during the refining process. Hence, this study aimed to optimise an extraction method for routine pesticide multi-residue analysis in edible vegetable oil samples using gas chromatography coupled to tandem mass spectrometry (GC – MS/MS). Two parameters were optimised using a factorial design of experiment (solvent type and solvent/sample ratio). In general, the number of pesticides that conformed to our criteria (mean recovery 70–120%) increased as the solvent/sample ratio increased. Each of the three tested solvents had an advantage over the other, ethyl acetate showed the highest capability for non-polar pesticides extraction, acetonitrile had the highest sensitivity, and the ethyl acetate/acetonitrile mixture showed the highest number of pesticides with acceptable criteria. The three solvents were tested using two different cleanup sorbents (EMR, and Z-Sep), the best solvent/cleanup combination was found to be ethyl acetate/acetonitrile mixture in addition to Z-Sep. The final method was validated in accordance with the European guidelines for Analytical Quality Control and Method Validation Procedures for Pesticide Residues Analysis in Food and Feed (SANTE/11312/2021). The precision and trueness of the method were determined from recovery experiments on six replicates of spiked blank corn oil samples at 0.01, 0.025, and 0.05 mg/kg. The number of pesticides that showed accepted criteria were 130, 166, and 182 out of 222 for each spiking level respectively.

Meeting nutritional adequacy in the Brazilian population increases pesticide intake without exceeding chronic safe levels

Achieving nutritional adequacy requires an increase in fresh foods consumption, which may increase pesticide intakes. This study aimed to identify required dietary modifications to achieve nutritional adequacy without exceeding the acceptable daily intake (ADI) for pesticides. Data from the National Dietary Survey 2017–2018 were linked to the pesticide database from the Program on Pesticide Residue Analysis in Food. We performed linear programming models to design nutritionally adequate diets constrained by food preferences for different constraints on pesticide intake at the least cost increment. Nutritional adequacy led to an increase in pesticide intakes without exceeding their ADI. Modifications in diets varied according to the model, but, in general, consisted in an increase in fruits and vegetables, dairy, and seafood, and a reduction in rice, red meat, and sugar-sweetened beverages quantities. In conclusion, meeting nutritional adequacy increases pesticide intake compared to the observed diets, without representing a health concern to consumers.

A review of analytical methods for the determination of herbicide residues from the group of nitrodiphenyl ethers in the environment and agricultural products, developed and validated a method for oxyfluorfen determination in soybean grain and oil method GC/ECD, which meets regulatory requirements and solves the problem of toxicant control in the above matrices. The developed method uses modern methods of sample preparation, identification and quantification of oxyfluorfen (selective extraction, capillary gas-liquid chromatography (GC/ECD GC/MS). The method is validated according to EU requirements) (SANTE/12682/2019 quality control and analytical validation procedures for pesticide residues analysis in food and feed). Selective multiresidue determination of highly polar anionic pesticides in plant-based milk, wine and beer using hydrophilic interaction liquid chromatography combined with tandem mass spectrometry

In this work, an easy and fast procedure for the selective multiresidue determination of 14 highly polar pesticides (including glyphosate, glufosinate, ethephon and fosetyl) and metabolites in beverages is presented. After an initial sample dilution (1:1, v/v), the extract is shaken and centrifuged, further diluted and then injected directly into the LC-MS/MS system, using hydrophilic interaction liquid chromatography (HILIC) and tandem mass spectrometry. No clean-up procedure was needed. The method was validated according to the current European guidelines for pesticide residue analysis in food and feed and linearity, limits of detection and quantification, matrix effects, trueness and precision were assessed. For plant-based milk, wine and beer samples, 10, 11 and 12 analytes, respectively, out of 14 were fully validated at 10 µg kg−1, the lowest spike level tested. The matrix effect was negative in most of the cases, showing for some compounds, such as HEPA, up to 80% suppression when compared to the response from standards in solvent. The use of isotopically labelled internal standards is required for the optimal quantification, as it compensates for high and varying matrix effects and also for recovery losses during extraction.

Analysis of highly polar pesticides and their main metabolites in animal origin matrices by hydrophilic interaction liquid chromatography and mass spectrometry

In this work, a method for the analysis of 14 highly polar pesticides in animal origin matrices by hydrophilic interaction liquid chromatography and mass spectrometry has been described. Three methanol-based extraction procedures with slight modifications, depending on the matrix, have been used. The method has been validated according to the performance criteria from the SANTE guideline for pesticide residues analysis in food and feed. Most of the compounds showed a limit of quantitation at the lowest spike levels tested (0.01–0.02 mg kg−1) in the matrices evaluated. A SPE clean-up step with mixed mode cation exchange sorbent has been used with some matrices, like chicken meat, eggs, liver and kidney, in order to improve the limits of quantification. Glufosinate, glyphosate and HEPA were the compounds which presented more difficulties during the study, due to the lack of sensitivity of the qualifier ions and/or because of the interference from the matrices.

Validation and analysis of thiram, a dithiocarbamate, as CS2 from soybean (Glycine max) samples on GC–MS

A method was validated for the estimation of dithiocarbamates in soybean leaf, pod, seed, oil and in soil. Method involves acid hydrolysis and reduction of dithiocarbamates to carbon disulfide in the presence of stannous chloride and hydrochloric acid. Thiram was spiked in the samples at the levels of 0.5, 2.5 and 5.0 µg g−1 and estimated as carbon disulfide on gas chromatograph coupled to single quadrupole (GC–MS) by monitoring m/z 76 (quantitation ion). Retention time of carbon disulfide was 2.119. All method validation parameters like linearity, accuracy and precision were within the acceptable range as per SANTE (in European Union Guidance document on analytical quality control and method validation procedures for pesticide residues analysis in food and feed. Document No. SANTE/11945/2015, 2015). The linearity, expressed as correlation coefficient, was 0.997. Accuracy was expressed as percent recovery. The recovery ranged from 87 to 102% in soybean leaf, 81–98% in soybean pod, 83–113% in soybean seed, 85–109% in soybean oil and 75–109% in soil at different fortification levels. Precision, expressed as relative standard deviation, ranged from 0.1 to 0.11 in various soybean samples. Limit of quantitation (LOQ) was 0.32 µg g−1 for soybean leaf, 0.18 µg g−1 for pod, 0.19 µg g−1 for seed and 0.1 µg g−1 for oil and soil. To our knowledge, this is the first report describing the extraction and analysis of thiram, a dithiocarbamate (DTCs) from soybean (oilseeds and oil). The method described here can be extended for the estimation of DTCs in other oilseeds and oils.

Food contaminated by pesticides: a Brazilian situation overview

Consumers are becoming increasingly demanding about products quality, including new decision variables at their purchases time. Information on nutritional aspects and the use of pesticides has changed consumer preferences. The new information strategies emerged in response to new consumer’s behavioral characteristics and new competition characteristics of the food market. These strategies privilege the use of quality signals, such as labeling, traceability, certification, brand names and adoption of minimum quality standards. The main feature of Brazilian markets is the lack of minimum standards and strong information asymmetries. The market for fresh food is affected by the indiscriminate use of pesticides. The information analysis gathered by the Program for Pesticide Residue Analysis in Food (PARA) found a very worrying situation in Brazil. In this sense, with this paper, our aim is to bring more information about the pesticides contamination in the fresh food markets in Brazil. Transaction Cost Economics is adopted as a theoretical framework to examine the coordination among the agents on the chain and seek sustainable alternatives to reduce the excessive use of pesticides in food production.

Further improvements in pesticide residue analysis in food by applying gas chromatography triple quadrupole mass spectrometry (GC-QqQ-MS/MS) technologies.

Nowadays, the control of pesticide residues in food is well established. The capacity of triple quadrupole technology to satisfy the current food regulations has been demonstrated. However, the permanent high demand of consumers for more sensitive and faster testing is driving the development of improved analytical methodologies that increase the performances of sensitivity and robustness and reduce the analysis time. In this work, the feasibility of decreasing the run time to 12.4min by modifying the oven temperature program, for a multiresidue method covering 203 pesticides, was evaluated. Satisfactory sensitivity results were achieved by reaching a limit of quantitation of 2μgkg-1 for a great variety of fruits and vegetables. The validated method based on updated GC-QqQ-MS/MS has confirmed the abovementioned challenges with adequate robustness by its application to routine analyses for 69 real samples. The proposed method can represent great benefit for laboratories as it allows increasing samples throughput. It is also very useful for risk assessment studies, where the needs of low reporting limits and very wide analytical scope are necessary.

Evaluation of nanoflow liquid chromatography high resolution mass spectrometry for pesticide residue analysis in food

This article reports on the evaluation of nanoflow liquid chromatography-mass spectrometry (LC–MS) for pesticide residue analysis in food. The approach is based on the use of reversed-phase C18nano columns with an integrated emitter, so that separation, ionization and detection are performed minimizing dead volumes. The use of nanoflow not only increases ionization efficiency and minimizes ionization suppression but also boost sensitivity compared to analytical-scale LC–MS methods. The nanoflow LC system was combined with full-scan high resolution mass spectrometry using a Q-Exactive Orbitrap instrument. The analytical performance was assessed for over 60 representative pesticides in five representative commodities (tomato, baby food, orange, fruit-based jam and olive oil). The sensitivity achieved with this configuration enables the implementation of high dilution factors (eg. 1:20, 1:50 or beyond) in pesticide residue workflows without compromising sensitivity, featuring limits of quantitation in the low ng kg−1 range. Using this dilution factors, signal suppression was found negligible in most cases (<10% in most cases, especially with 1:50 dilution), so that matrix-matched standards may be skipped, thus simplifying laboratory workflows. The robustness of the nanoflow LC system and its capability to withstand long analytical runs was also evaluated. Appropriate precision in terms of peak area and retention time was obtained at different concentration levels for over 125 injections without any instrument servicing. The main benefits of the nanoflow liquid chromatography approach are the high sensitivity gain and the outstanding reduction in matrix effects thanks to the high sample dilution factors that can be implemented, along with the substantial reduction in solvent usage.

Targeted approach for qualitative screening of pesticides in salmon feed by liquid chromatography coupled to traveling-wave ion mobility/quadrupole time-of-flight mass spectrometry

The current work reports the development and validation of a qualitative screening method for 156 pesticides in Atlantic salmon feeds based on the use of ultrahigh-performance liquid chromatography (UHPLC) coupled to traveling-wave ion mobility spectrometry (TWIMS)/quadrupole time-of-flight mass spectrometry (QTOFMS). This novel hyphenation allows a straightforward estimation of the collision cross section (CCS), a physicochemical property that recently has been proposed as an additional identification point for pesticides.In order to increase the overall efficiency of the screening workflow, data processing parameters and tolerances involved in the automated analyte detection were first optimized. The use of the CCS as an additional identification point greatly improved the method selectivity, decreasing the rate of false-positive detections in blank feed samples. Qualitative validation was then performed in accordance with the EU document SANTE/11945/2015 on analytical quality control and validation procedures for pesticide residues analysis in food and feed. Feed samples were spiked with 156 pesticides at three levels (0.01, 0.05 and 0.20 mg/kg) and analyzed by the proposed method. Under the optimized conditions, screening detection limits (SDLs) ≤0.05 mg/kg were achieved for 82% of the investigated pesticides.The applicability of this new approach for the screening of pesticides was finally proven by analyzing several commercial Atlantic salmon feed samples as well as various plant-based feed ingredients. Pirimiphos-methyl and ethoxyquin were among the most frequently detected analytes in the investigated samples.

Recent achievements and applications of solid – phase microextraction in pesticide residues analysis in food

Abstract The current trend in sample preparation methods is devoted to minimizing or eliminating the volume of extractive solvent. In this review, the focus on solid phase microextraction (SPME) as a solvent free sample preparation method for the isolation of pesticides residues in different food matrices is given. To achieve satisfactory extraction efficiency, selection of the fiber coating is an important step in the method development. Here, recent trends in new supporting materials and new fiber coatings development are discussed. Finally, applicability of SPME for the pesticide residues analysis in various food matrices using mainly chromatographic methods is also reviewed.

Recent Trends in Sample Preparation and Liquid Chromatography/Mass Spectrometry for Pesticide Residue Analysis in Food and Related Matrixes.

A rising interest by consumers and various governmental organizations towards the quality of food has been continuously observed. Pesticide residue analysis has a significant role in assessing food safety and quality. This article reviews the new analytical approaches for efficient extraction and reliable identification and quantification of pesticides in foodstuffs and related matrixes. Emphasis is given to the new materials used for effective extract purification. We discuss the potential and pitfalls of the different LC/MS approaches, including application of high resolution mass spectrometry in the area of pesticide residue analysis. Untargeted and retrospective screening is outlined, highlighting prospects and achievements as well as its major drawbacks.

Pesticide residues analysis in wine by liquid chromatography–tandem mass spectrometry and using ethyl acetate extraction method: validation and pilot survey in real samples

This paper presents a cost-effective and validated method for the determination of 172 pesticide residues monitored during a survey study on 27 commercially available Cypriot wines (red/white, dry/sweet). Sample preparation involved ethyl acetate extraction of pesticides from wines. For the validation of the method, blank samples were spiked with a solution of 172 pesticides (such as insecticides, fungicides, herbicides) at three levels such as 0.01, 0.05 and 0.1 mg/kg. The samples were analyzed by LC-MS/MS (liquid chromatography tandem mass spectrometry). The validation study was in accordance with the DG SANCO (Directorate-General for Health and Consumers) guidelines [Document No. SANCO/12495/2012. Method validation and quality control procedures for pesticide residues analysis in food and feed]. The scope of validation included recovery, linearity, limits of quantification and precision. Recoveries of the majority of compounds were found to be in the range of 70–120% and were characterized by precision lower than 20%. Fifteen out of the 27 real samples analyzed were positive with pesticide residues and all observed values were much lower than the MRLs.

Alternative QuEChERS-based modular approach for pesticide residue analysis in food of animal origin.

The approach for pesticide residue analysis in food of animal origin differs strongly from the one established for food of plant origin, as laboratories mainly focus on conventional methods for the analysis of non-polar pesticides known to accumulate in fatty tissues. However, these group-specific methods are very laborious and cost intensive and typically require extraction of fat components followed by extensive clean-up steps to remove matrix constituents. This work highlights the development and validation of a straightforward QuEChERS-derived clean-up procedure enabling facile, precise, and reliable identification and quantitation of pesticide residues in food of animal origin, which can be extended to various other commodities with moderate fat content and applied to replace traditional group-specific methods. Two additional methods for lean and highly fatty commodities complete this well-established "simplified modular system". The proposed method was in-house validated in terms of accuracy and precision, recovery and linearity as well as specificity and sensitivity on two representative commodities for food of animal origin (egg and meat). For the majority of the more than 80 pesticides investigated, satisfactory results were obtained. Procedural matrix calibration was applied for screening purposes in conjunction with GC-MS/MS or LC-MS/MS as integral part of the approach. Apparent recoveries for most of the compounds ranged from 70 to 120 % (RSD < 20%) at spiking levels of 0.01 (LOQ level) and 0.05 mg/kg. Accuracy of the method has been demonstrated by application to reference material from previous EU proficiency tests. Only in case of positive screening results a standard addition approach was applied for precise quantitation.
