Benzyl Isothiocyanate Loaded Gelatin Nanoparticles Display Unique in Vitro Antioxidant Prospects

This present investigation focuses on the fact that nanoformulation of phytochemicals could enhance the therapeutic capacity in different physiological systems by enhancing hydrophilicity and bioavailability. In this study gelatin nano-formulation of benzyl isothiocyanate (BITC) was prepared and characterized by dynamic light scattering and UV-Visible spectrometry. Then antioxidant activity of BITC and BITC-gelatin NPs was determined in different concentrations through measuring 2,2- diphenyl-1- picrylhydrazyl (DPPH), superoxide, hydroxyl radical, nitric oxide scavenging and lipid peroxidation inhibition activities. DLS and UV-Vis study revealed the production of uniform nanosized particles and effective encapsulation of BITC respectively. The results of antioxidant assays suggested that BITC-gelatin NPs more effectively scavenged free radicals and inhibited lipid peroxidation compared to free BITC. The findings proposed that gelatin formulated BITC nanoparticles could be effective against oxidative stress related disorders.

The Effect of ZnCl2 Concentrations and Heating Methods on the Chlorophyll, Phenolic, Andrographolide Content and Antioxidant Activity of Sambiloto (Andrographis paniculata) Simplicia Powder

The drying process in Sambiloto simplicia production causes the degradation of chlorophyll as the major compound. However, the stability of chlorophyll can be enhanced by forming a metallochlorophyll complex with zinc (Zn) metal. This research aims to produce Zn-rich Sambiloto simplicia powder to produce high stability of chlorophyll. Sambiloto simplicia powder was prepared through sorting, grinding, sieving, and mixing with ZnCl2 solution at concentrations of 0, 200, 300, 400, and 500 ppm, followed by heating (using either oven or an autoclave) and drying. The analysis included determination of total chlorophyll, phenolic, flavonoid, zinc, and andrographolide content. DPPH and FTC assays were employed to evaluate antioxidant activity. The results showed that increasing ZnCl2 concentration up to 300 ppm enhances chlorophyll, phenolic, flavonoid, zinc, and andrographolide content. Higher concentrations of ZnCl2 in the oven resulted in decreased lipid peroxidation inhibition (LPI) of Sambiloto simplicia powder, whereas using an autoclave led to increased LPI. Furthermore, higher ZnCl2 concentrations, up to 400 ppm, corresponded to increased DPPH radical scavenging activity. Lipid peroxidation inhibition activity correlated with andrographolide, zinc, and total phenolic contents. Conversely, DPPH radical scavenging activity strongly correlated with total phenolic, total flavonoid, Zn, chlorophyll, andrographolide content.

Comparison of some biological activities of propolis and bee bread samples obtained from Apis mellifera Anatoliaca and its Muğla and Efe ecotypes

Honey and other bee products have been extensively researched in recent years due to the demand for natural products with nutritional and therapeutic qualities, as they contain high levels of biologically active compounds and valuable nutrients. In this study, antioxidant, anti-inflammatory and antimicrobial activities of bee bread and propolis samples formed after colony development by bees of different races and ecotypes accepted to core colonies in the same location were evaluated. Total phenolic, flavonoid and antioxidant content values were the the highest in propolis samples, especially in propolis from Apis mellifera anatoliaca. Values of this type gave the most effective results in almost all of the other tests performed, such as DPPH free radical scavenging, lipid peroxidation inhibition, anti-inflammatory and antimicrobial activity. In addition, the amount of phenolics detected in the phenolic component analysis of this species is higher. It was surprising that propolis samples obtained from Efe and Muğla ecotypes showed higher values, respectively, in the test for chelating activity with Fe. The fact that the values calculated for the bee breads were lower in all cases compared to the propolis samples, the highest values were calculated for the bee bread harvested from the efe ecotype, in contrast to the propolis ecotypes. None of the tested bee breads showed antimicrobial activity on selected pathogenic bacteria. Thus, with this study, it was evaluated how the differences arising from both race and ecotype and the type of bee product reflected on biological activities.

Determination of antioxidant and anti-inflammatory properties of Salvia aethiopis/sclarea and synthesized silver nanoparticles

Aim: In the study, the phenolic compounds of Salvia aethiopis/sclarea will be determined and the antioxidant and anti-inflammatory activities of the extracts of these plants and the synthesized silver nanoparticles (AgNP) will be examined in vitro. Materials and Methods: Extracts obtained from Salvia spp. (SE) collected from the Kyrenia region of Cyprus were used in the study. Methanol extracts were prepared and total phenolic, flavonoid and alkaloid contents were determined. The amount of phenolic compounds was then quantified and identified using HPLC. The antioxidant properties of Salvia aethiopis/sclarea were analyzed using various markers, including lipid peroxidation inhibition activity, 1,1-Diphenyl-2-picrylhydrazyl (DPPH), and ferric reducing antioxidant power (FRAP) scavenging capacity. Additionally, their anti-inflammatory effects were investigated by proteinase inhibitor activity, heat-induced hemolysis and albumin denaturation analyses. Results: S. aethiopis and S. sclarea were contained high levels of the phenolic compounds isoquercetin and caffeic acid or naringenin and caffeic acid, respectively. After green synthesis of silver nanoparticles, the antioxidant and anti-inflammatory properties of plant extracts increased significantly (p<0.05). Conclusion: Our study found that silver nanoparticles synthesized using S. aethiopis/sclarea extract increased the antioxidant and anti-inflammatory effects of plant extracts and suggested that AgNP/SE could be used as a potential therapeutic agent in medicine.

Antioxidant, Antistress, Nootropic activity and its Correlation studies of aqueous extract of Punica granatum fruit estimated by Noninvasive biomarkers and Y-maze test in rodents

The memory loss associated with increased oxidative stress due to increased level of free radicals. Oxidative stress has been involved in several diseases includes cancer, atherosclerosis and neurodegenerative diseases.This study aimed to estimate the correlation of antioxidant and antistress activity with nootropic activity of aqueous extract of pomegranate (Punica granatum) by in-vivo noninvasive studies using rats. The in-vitro antioxidant activity was determined based on the ability of the Punica granatum to scavenge free radicals and lipid peroxidation inhibiting activity. The antistress effect of the aqueous extract of Punica granatum for 24h treatment (100 and 300mg/kg, p.o.) was evaluated by using the forced swim stress test in rats. The 24h urinary excretion of vanillylmandellic acid (VMA), 5-hydroxyindoleacetic acid (5-HIAA), homovanillic acid (HVA) and ascorbic acid (AA) was determined in all groups under normal and stressed conditions by HPLC and spectrophotometric methods in all groups, were selected as noninvasive biomarkers. Nootropic activity activities of Punica granatum fruit extract were estimated as locomotor and working memory in rats in a Y-maze apparatus. Administration of aqueous extract of Punica granatum at a dose of 100 and 300mg/kg reduced the urinary metabolite levels. Punica granatum treatment showed significant dose dependent variation in non-invasive biomarker levels in urine samples of rats taken after 24h. The treatment with Punica granatum extracts improved the percentage entry of rats into safer zone, it means acquisition (learning) retention and retrieval (memory) were improved in rats compared to stress controls. Cognition determined by working memory and locomotor activity results were shown to be dose-dependent. The results of this study strongly suggestedantioxidant, antistress and nootropic activity effect of Punica granatumin rodents were correlated. The data obtained were analyzed by one-way ANOVA followed bytukey test. p<0.05 was considered to be significant. There is substantial evidence that flavonoids play an active role in providing antioxidant, antistress and nootropic activities of Punica granatum extracts. The findings of the present investigations indicate that the Punica granatum has significant antistress activity, which may be due to the immunostimulating property and increased resistance, nonspecifically, against all experimental stress conditions. The study provided scientific evidence for their utility as nootropic agents and to advocate their use in foods.

Identification of bolegrevilol B and C as novel antioxidant compounds in Suillus grevillei mushroom.

Suillus grevillei is a popular species of mushroom available worldwide. In this study, we isolated compounds, bolegrevilol B and bolegrevilol C, from the mushroom and observed their potent lipid peroxidation-inhibiting activity. The structures of bolegrevilol B and bolegrevilol C were elucidated as 3-geranylgeranyl-1,2,4-trihydroxybenzene and 3-geranylgeranyl-1,2-dihydroxy-4-methoxybenzene, respectively, through high-resolution electrospray ionization mass spectrometry (-) and 1D and 2D nuclear magnetic resonance analyses. Bolegrevilol B and C inhibited lipid peroxidation and exhibited IC50 values of 2.0±0.29 µm and 1.0±0.13 µm, respectively. Furthermore, bolegrevilol B and C demonstrated potent neuroprotective activities in neuronal hybridoma N18-RE-105 cells against L-glutamate toxicity (EC50 of 1.8±1.7n m and 7.2±6.9n m, respectively). Bolegrevilol B was found in nature for the first time, and, to the best of our knowledge, this is the first study to report the antioxidant activities of bolegrevilol B and C.

Non-Enzymatic and Enzymatic Antioxidant Responses of Hypericum perforatum L. Hairy Roots upon Photooxidative Stress

The aim of this study was to evaluate the non-enzymatic and enzymatic antioxidant response of fifteen Hypericum perforatum L. dark-grown (HR1 A-HR1 O) and photoperiod-exposed (HR2 A-HR2 O) hairy root clones. Dark-grown HR1 clones were characterized with high biomass accumulation and up-regulated phenylpropanoid metabolism through phenylalanine ammonia lyase (PAL)-mediated production of flavonoids, flavan-3-ols and anthocyanins. These groups of phenolics showed superior hydrogen-donating capability and significantly contributed to the antioxidant capacity of dark-grown HR1 clones. Photoperiod-exposed HR2 clones showed green coloration with shoot regenerative potential and reduced biomass accumulation. Photoperiod exposition improved the production of hypericins, as well as ferrous chelating properties and lipid peroxidation inhibition activity in HR2 cultures. Furthermore, HR2 clones were represented with enhanced activity of antioxidant enzymes (guaiacol peroxidase, catalase and superoxide dismutase) that resulted in suppression of oxidative stress markers (hydrogen peroxide, superoxide anion and malondialdehyde). These observations revealed the involvement of an efficient antioxidant defense system in the adaptive response of HR to photooxidative stress. Altogether, photoperiod-exposed H. perforatum HR2 clones were considered as a promising alternative for further scale-up production of naphthodianthrones that could be used in the pharmaceutical industry.

Evaluation of Semecarpus kurzii Engler from Bay Islands for oxidative DNA damage protective activity and in vitro antioxidant potential

Introduction and Aim: Harnessing traditional knowledge of medicinal plants is important for the betterment of mankind. We must safeguard traditional knowledge from misuse by miscreants living in modern societies and knowledge must be available in the public domain for use in drug designing in the healthcare system. Since the beginning of recorded history, Indians have used plants as a source of medicine, and their cultural past is incredibly rich. This study was conducted to evaluate the oxidative stress-induced DNA damage prevention and antioxidant potential of Semecarpus kurzii found in the Bay Islands. Materials and Methods: The scavenging of superoxide, hydroxyl radicals, nitric oxide (NO), 2,2-diphenyl-2-picrylhydrazyl hydrate (DPPH) radicals, ferric reducing power (FRAP), and lipid peroxidation inhibition activity were tested to determine antioxidant activity. DNA damage inhibition test was used to evaluate the protection against oxidative DNA damage. Results: The S. kurzii extract showed dose-dependent scavenging of DPPH, superoxide anion, nitric oxide, hydroxyl radical and reducing power evaluated by comparing with standard antioxidants (ascorbic acid, alpha-tocopherol and BHT). However, the nitric oxide scavenging (IC50 =186.47) and superoxide scavenging (IC50 = 678.32) was more than that of DPPH (IC50 =28.03) and Hydroxyl radical (IC50 =89.10) scavenging capacity. In addition, S. kurzii extract and ascorbic acid showed lipid peroxidation inhibition activity. The extract also exhibited DNA protection which was confirmed by the DNA damage inhibition assay. Conclusion: Our results corroborated that, the methanolic extract of S. kurzii bark has substantial antioxidant activity and DNA damage inhibition. The potential antioxidant and oxidative DNA damage preventive activity could be due to the existence of polyphenolic compounds in the S. kurzii bark.

Selection of bioactive chemical markers for anti-aging skin assessment of Dimocarpus longan Lour. leaf extract

Hydroethanolic longan (Dimocarpus longan Lour.) leaf extract has reported remarkable biological activities related to the aging process for use in cosmeceuticals. Finding the bioactive chemical marker (BCM) could play a crucial role for cosmeceutical manufacturers in guaranteeing the quality of the extract and its efficacy. This study aimed to select the most potential BCM established and predicted for the anti-aging skin of hydroethanolic longan leaf extract. The assessment of anti-aging of the skin was determined by evaluating in vitro antioxidant activities, including 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging and inhibiting lipid peroxidation assay. Furthermore, anti-hyaluronidase and anti-collagenase activity were also determined. The extract underwent bioassay-guided fractionation using normal phase preparative-thin layer chromatography. Ethyl gallate in the fractionated extract possessed biological activities as a high proportion part. The strong positive linear relationship between DPPH scavenging, lipid peroxidation inhibition, anti-collagenase, and anti-hyaluronidase activity (r = 0.997, 0.972, 0.998, and 0.998, respectively) under the deterioration of the ethyl gallate was shown. The molecular docking of ethyl gallate to collagenase and hyaluronidase resulted in a good and acceptable binding affinity. Therefore, ethyl gallate could be potentially used as a bioactive chemical marker for hydroethanolic longan leaf extract in pharmaceutical and cosmeceutical applications.

Alterations in the phytochemical composition and antioxidant activity of Ligustrum robustum according to continuous wet‐ and dry‐heat treatment

Small-leaved Kuding tea (SLKDT) obtained from Ligustrum robustum is a traditional tea substitute in southern China and has a range of physiological effects. However, the changes in its phytochemical composition after various heat treatments are not reported yet. Thus, the phytochemical composition and antioxidant activities of fresh leaves of SLKDT (LrF1) and SLKDT after high-temperature wet-heat treatment (LrF2) and wet- and dry-heat treatments (LrF3) were assessed using liquid chromatography-mass spectrometry, and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonate (ABTS) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities and lipid peroxidation inhibition activity of LrF1 and LrF3 were investigated. The results indicated that the phytochemical composition of LrF1, LrF2, and LrF3 was significantly different. Overall, 258 and 83 differential constituents, respectively, were obtained in LrF1 versus LrF2 and LrF2 versus LrF3. The differential constituents mainly included amino acids and their derivatives, nucleosides, flavonoids, terpenoids, simple phenylpropanoids, and coumarins. After heat treatment, SLKDT exhibited obvious changes in sensory characteristics and physiological properties, which may be related to the changes in the levels of amino acids, linalool, beta-geraniol, myricetin, naringin, fraxetin, and isoacteoside. Moreover, the antioxidant activities significantly changed after heat treatment of SLKDT. Overall, our study demonstrated that heat treatment can alter the phytochemical composition of SLKDT, thus affecting its sensory properties and physiological properties. PRACTICAL APPLICATION: This study preliminarily assessed the changes in the composition of small-leaved Kuding tea (SLKDT) after various heat treatments and revealed that the composition of SLKDT tea can be adjusted by various heat and temperature treatments.

Hepatoprotective Effect of Albumin Peptide Fractions from Corn Germ Meal against Alcohol-Induced Acute Liver Injury in Mice.

Acute alcoholic liver disease can cause serious liver damage. This study reports on the hepatoprotective effect of albumin peptide fractions from corn germ meal (MW < 1 kDa) (APF4) on acute alcohol hepatic damage in mice. In the mice model, the results indicated that APF4 at a dose of 800 mg/kg/bw could markedly boost alcohol metabolism, which was shown in the reduced duration of the loss of the righting reflex; the reduced level of blood alcohol concentration (BAC), cytochrome P450 2E1 (CYP2E1), alanine aminotransferase (ALT), aminotransferase (AST), triglycerides (TG), and malondialdehyde (MDA) (p < 0.01); the enhanced activity of aldehyde dehydrogenase (ALDH); and the superoxide dismutase (SOD) and glutathione (GSH) levels being increased by up to 84.02% and 193.22% (p < 0.01) compared to the control group. The antioxidant capability and lipid peroxidation inhibition activity of APF4 may be responsible for its protective effect against liver damage induced by alcohol. The findings suggested that APF4 had the hepatoprotective property against liver damage induced by alcohol.

Protective effects of sugarcane phenolic extract against lipid oxidative damages in raw ground pork and beef during refrigerated storage conditions

The study evaluated the bioactive components and antioxidant properties of sugarcane phenolic extracts (SCPE) against lipid oxidation in raw ground pork and beef during refrigerated storage conditions. Results showed that SCPE has a high total phenolic content of 35.9 g GAE/100 g sample majority of which are vanillic (799.77 mg/100 g), sinapic (434.38 mg/100 g), and coumaric (342.37 mg/100 g) acids. These phenolic compounds exhibited strong radical scavenging activities against DPPH (191.00 mg TE/g) and ABTS (359.80 mg TE/g) radical, ferric reducing capacity (97.80 mg TE/g), and lipid peroxidation inhibition (120.20 mg TE/g) activities which are comparable to BHT. Supplementation of SCPE at 400 mg/kg fresh weight of ground pork and ground beef improved the antioxidant activities of meat samples, leading to the delay in TBARS formation, lower reduction in heme iron content, and minimal increase in metmyoglobin content during 14-day refrigerated storage as compared to BHT-treated and control. Moreover, the redness of meat was preserved in SCPE- and BHT-treated samples as measured by a* color value. Correlation heat maps showed that TBARS, metmyoglobin, and ΔE are positively correlated with each other, and at the same time showed negatively correlated with heme iron and a*. This study revealed that antioxidant extracts from sugarcane presented a positive effect on the oxidative stability of raw ground pork and beef during refrigerated storage.

Screening for acetylcholinesterase inhibition, lipid peroxidation inhibition and antioxidant activity of medicinal plants from Morocco

Acetylcholinesterase (AChE), hydrolyzes acetylcholine to choline and acetate, thereby terminating this neurotransmitter effect at cholinergic synapses. Therefore, AChE inhibition is used for counterbalance the cholinergic deficit in Alzheimer’s disease (AD) patients. In the present work, in order to find new plant acetylcholinesterase inhibitors, the hydroalcoholic extracts from seventeen medicinal plant species were screened for their acetylcholinesterase inhibition activity, as well as total phenolic (TPC) and flavonoids contents (TFC) and antioxidant activity using ORAC (Oxygen Radical Absorbance Capacity) assay, and their ability to inhibit lipid peroxidation. The results revealed that Rumex acetosa, Taraxacum officinaleand Hypericum perforatumextracts possessing the highest TPC and TFC, were the most effective in terms of ORAC antioxidant activity, and acetylcholinesterase inhibition, in addition to their ability to inhibit liposomes peroxidation, suggesting that those plant species may provide a substantial source of secondary metabolites, which act as natural antioxidants and acetylcholinesterase inhibitors, and may be beneficial in the treatment of AD.

HEPATOTOXICITY EFFECTS OF PARACETAMOL-DENIK CLEANSER® CO-ADMINISTRATION

Aim and Objective: The concurrent use of herbal products with orthodox medicine is on the rise with the risk of herb-drug interaction that could be beneficial or harmful to the body. Paracetamol (acetaminophen) is an anti-pyretic and analgesic drug metabolized by CYP2E1 to give the major hepatotoxin, N-acetyl-p-benzoquinone imine (NAPQI). Denik cleanserâ is an oral herbal preparation from plants part of Occimum grattissimum, Colocynthis citrullus, Khaya ivorensi. The aim of this study was to evaluate the hepatotoxic effects triggered by Denikâ and paracetamol co-administration in rats. The study sought to mimic conventional usage of Denik cleanserâ followed by ingestion of paracetamol, a likely scenario given the popularity of both compounds. Method: Twenty animals were randomly assigned to four groups, the first group (control) received 0.3 mL distilled water, 2nd group received paracetamol 100 mg/kg, 3rd group received Denik cleanserâ2 mL/kg while the 4th group received both paracetamol and Denik cleanserâ at 100 mg/kg and 2 mL/kg daily for 3 days after which biochemical and histological analysis were carried out. Results: From histological analysis revealed that rats that received Denikâ-only and Denikâ/paracetamol (concomitantly) showed markedly distorted liver architecture compared to control indicating toxicity. Similarly, the biochemical analysis results showed a significant (p&lt;0.05) increase in AST and ALT for the Denikâ/paracetamol group compared to the control group indicating a hepatotoxic event. A non-significant increase in ALP and GGT were also observed in the Denik cleanserâ + paracetamol group. Enhanced metabolism of paracetamol by Denik cleanserâ to NAPQI (hepatotoxic metabolite) is indicated as possible mechanism. Conclusion: The results of this study demonstrated the in vivo potential for a herb/drug interaction involving paracetamol and Denik cleanserâ resulting in liver injury.Therefore, caution is strongly advised against its casual, non-medically supervised usage. Extraction, Profiling, and Characterization of Phytosterols and Triterpenoids from Pili (Canarium ovatum Engl.) Pulp Oil Exhibiting Antioxidant and Antibacterial Properties.

Pili (Canarium ovatum Engl.), an indigenous tree found in the Philippines, is highly regarded for its fruit due to its high economic value. During processing, the pulp is often discarded as waste but contains considerable amounts of oil and bioactive minor lipid components. The present study explored the antioxidant and antibacterial properties of saponified diethyl ether extract of pili pulp oil and related this activity to the nature of compounds present in the extract through GCMS. The extract indicated the elution of 18 major compounds which are mostly cyclic triterpenic (α-and β-amyrin, lupenone, and β-amyrone) and phytosterol (β-sitosterol, brassicasterol, and stigmasterol) class of compounds. Characterization of the bioactivity of the extract showed high antioxidant activities measured by DPPH radical scavenging (EC50: 74.45 ± 1.29 μg/mL) and lipid peroxidation inhibition (EC50: 3.02 ± 0.06 μg/mL) activities that were comparable with that of α-tocopherol. Moreover, an observed bactericidal activity was demonstrated by the extract against E. coli and S. typhi with MIC values of 40 and 35 μg/mL, respectively. The observed bioactivity of the pili pulp oil extract can be attributed to these compounds which may provide desirable health benefits.

Efficient green photocatalyst of silver-based palladium nanoparticles for methyle orange photodegradation, investigation of lipid peroxidation inhibition, antimicrobial, and antioxidant activity.

Nanotechnology is an interdisciplinary study that has been developing worldwide in recent years and has a serious impact on human life. The fact that the nanoparticles of plant origin are clean, non-toxic, and biocompatible has enabled new fields of study. The Hibiscus sabdariffa (H. sabdariffa) plant has been attracted by scientists because of its impact on health and many other areas. The lipid peroxidation inhibiting activity, antioxidant properties, and antimicrobial properties of H. sabdariffa plant with Ag-Pd metal was ditermined. For the total phenolic component, gallic acid was used as the standard and quarcetin was used for the total flavonoid. The lipid peroxidation inhibition activity of Ag-Pd NPs in ethanol extract was found to be very well compared to the positive control (BHA). The lowest and highest concentrations of DPPH radical scavenging activity were 82.178-97.357%, whereas for BHA these values were found to be 84.142-94.142%. The highest concentration of Ag-Pd NPs at 200μg/mL the DPPH radical quenching activity was higher than BHA. Ag-Pd NPs showed a good antimicrobial activity against certain pathogenic microorganisms such as Bacillus subtilis, Enterococcus faecalis, Escherichia coli, Pseudomonas aeruginosa, Candida albicans, which are the causative agents of various diseases in humans. The photodegradation activity of Ag-Pd NPs also investigated against Methyl orange dye (MO) under sunlight irradiation for 120min and was found to be as 67.88.

Nutrients and antioxidant properties of enzymatically hydrolyzed anchovy (Engraulis japonicus) paste

ABSTRACT Anchovy, a low-value small marine fish, is mainly processed into fish meal, whose high-value functional components are not fully exploited. In this study, the enzymatically hydrolyzed anchovy (Engraulis japonicus) paste (EHAP) was prepared by hydrolyzing anchovy using Alcalase2.4 L, and the nutrients and antioxidant properties of EHAP were assessed to increase its added value. Protein, total amino acids and oligopeptide of the nutrients in EHAP reached 73.9 ± 0.7%, 56.1 ± 0.8 g/100 g and 50.4 ± 1.6%, respectively. The molecular weight distribution of EHAP displayed a high dipeptides and tripeptides proportion of 60.1 ± 0.4%. Furthermore, EHAP showed the highest antioxidant activities of ferrous ion chelating, lipid peroxidation inhibition and DPPH scavenging activity, reached about 34%, 40% and 49%, respectively. Collectively, the EHAP belonged to a product with strong antioxidant properties, and was rich in nutrients, which could serve as a valuable bioactive marine resource applied in the aquatic feed and functional food industry.

Phytochemical, Proximate and Mineral Composition, Antioxidant and Antidiabetic Properties Evaluation and Comparison of Mistletoe Leaves from Moringa and Kolanut Trees

This study evaluates the chemical properties of mistletoe leaves powder from moringa trees (MLPM) and mistletoe leaves powder from kola nut trees (MLPK). The MLPM and MLPK were subjected to phytochemical analysis, vitamin A determination, lipid peroxidation inhibition activity, 2, 2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) analysis, proximate and mineral composition analysis, alpha-amylase inhibitory activity and alpha-glucosidase inhibitory activity. Total phenol, steroids, and tannins concentrations were higher in MLPK than in MLPM; steroids was higher in MLPK than in MLPM; whereas total flavonoids and saponins concentrations, were equivalents. MLPK had greater vitamin A concentrations and DPPH than MLPM. The lipid peroxidation inhibition of MLPM and MLPK did not differ significantly. MLPM had higher moisture, crude fibre, and ash content than MLPK. Furthermore, MLPK had larger fat, crude protein, and nitrogen-free extract contents than MLPM. The MLPM and MLPK have similar Zn (0.9 vs. 1.3 mg/kg), Ca (20.1 vs. 13.1 mg/kg), Mg (66.1 vs. 70.2 mg/kg), and P (185.8 vs. 183.5 mg/kg) contents. The alpha-amylase inhibitory action of MLPM (40.40%) and MLPK (35.07%) were similar. MLPK (44.37%), on the other hand, has a stronger alpha-glucose inhibitory action than MLPM (32.47%). Finally, phenol, steroids, tannins, and steroids varied between MLPM and MLPK, with phenol, steroids, and tannins being greater in MLPK. The MLPM had higher vitamin A concentrations and antioxidant activity than MLPK. The MLPM and MLPK inhibited alpha-amylase and alpha-glucosidase activity.

Antioxidant, Anti-Tumour, and Anticoagulant Activities of Polysaccharide from Calocybe indica (APK2).

The initial structural features and in vitro biological study of crude polysaccharides from Calocybe indica (CICP) extracted by hot water followed by ethanol precipitation was investigated. High-performance gel permeation chromatography, HPLC-DAD, UV, IR and NMR spectroscopy, X-ray diffraction, scanning electron microscopy, and Congo red methods were used to determine structural features. The results revealed that CICP is a hetero-polysaccharide with a molecular weight of 9.371 × 104 Da and 2.457 × 103 Da which is composed of xylose, mannose, fucose, rhamnose, arabinose, galactose, and glucose. The antioxidant activity of CICP was evaluated using radical scavenging activity (three methods), reducing ability (three methods), metal chelating activity, and lipid peroxidation inhibition activity (two methods). It was found that the antioxidant capacity is concentration-dependent and EC50 values were found to be 1.99–3.82 mg/mL (radical scavenging activities), 0.78–2.78 mg/mL (reducing ability), 4.11 mg/mL (metal chelating activity), and 0.56–4.18 mg/mL (lipid peroxidation inhibition activity). In vitro anticoagulant assay revealed that CICP could prolong activated partial thromboplastin time (APTT), thrombin time (TT), but not prothrombin time (PT). CICP exhibited antiproliferative activity on HeLa, PC3, HT29, HepG2, and Jurkat cell lines with IC50 (μg/mL) values of 148.40, 143.60,151.00, 168.30, and 156.30, respectively. The above findings suggested that CICP could be considered a natural antioxidant and cancer preventative.

Antioxidant, cytotoxic and apoptotic activities of the rhizome of Zingiber zerumbet Linn. in Ehrlich ascites carcinoma bearing Swiss albino mice

Due to having a long history of traditional uses as a functional food, Zingiber zerumbet was selected here to explore the inherent antioxidant and antineoplastic activities of methanolic extract of its rhizome (MEZZR) against Ehrlich ascites carcinoma (EAC) cells. The rich polyphenol containing MEZZR showed a marked DPPH, ABTS, nitric oxide radicals and lipid peroxidation inhibition activity with an IC50 of 3.43 ± 1.25, 11.38 ± 1.39, 23.12 ± 3.39 and 16.47 ± 1.47 µg/ml, respectively, when compared to the standard catechin. In vivo, MEZZR significantly inhibited EAC cell growth, decreased body weight gain, increased life span and restored the altered hematological characteristics of EAC-bearing mice. Moreover, MEZZR induced nuclear condensation and fragmentation, which are notable features of apoptosis as observed by fluorescence microscopy after staining EAC cells of MEZZR-treated mice with Hoechst 33342. Additionally, in vitro, the cell growth inhibition caused by the MEZZR in MTT assay, was remarkably decreased in the presence of caspase-3, -8 and -9 inhibitors. This study thus suggests that MEZZR may possess promising antiproliferative efficacy against EAC cells by inducing cell apoptosis.
