Cellular cohesiveness in benign and malignant thyroid follicular tumours varies significantly, but the difference is not useful in diagnosis of individual cases

The aim of our study was to search for new, readily available and statistically reliable cytological markers for differentiating benign and malignant follicular thyroid neoplasms pre-operatively. Cohesiveness of tumour cells in cytology slides from a series of 58 follicular tumours diagnosed between 1998 and 2004 inclusive was studied, including 48 follicular adenomas, and eight minimally invasive and two widely invasive follicular carcinomas. Photomicrographs of the cytology slides were taken and the digital images were analysed using computer image analysis software. We evaluated the relative proportions of cells arranged in groups of various sizes. The cohesiveness of the cells in cytological smears was then correlated with the immunohistochemical expression of E-cadherin in corresponding histological slides. Cases from 15 men (26%) and 43 women (74%) with a mean age of 50 years (range, 19-79) were analysed. In follicular adenomas and carcinomas, respectively, isolated cells were seen in 16.8% and 24.7% (P = 0.028), groups of two to five cells in 9.7% and 11.5% (P = 0.145) and groups of more than five cells in 73.5% and 63.8% (P = 0.041). The mean cell count in groups with more than five cells was 46.5 and 27.0 in adenomas and carcinomas, respectively (P < 0.001). Cell cohesiveness, either as percentage of cells in groups of more than five (R(2) = 0.026) or as mean cell count per group of more than five (R(2) = 0.005), was not found to be dependent on the expression of E-cadherin. Using a threshold of 13% isolated tumour cells in cytological smears, follicular adenomas and carcinomas could be distinguished with 90% sensitivity and 41% specificity. Although we demonstrated a statistically significant difference in cell cohesion between follicular adenomas and carcinomas, these could not be distinguished in the clinical setting by evaluation of the percentage of isolated cells in cytological smears because the specificity was too low. The absence of correlation of cellular cohesiveness with E-cadherin expression indicates that other factors are probably responsible for the loss of cohesiveness observed in follicular thyroid malignancy.

Regional variations in certain cellular characteristics in human lumbar intervertebral discs, including the presence of α-smooth muscle actin

An evaluation of the regional variation of certain cellular features in the human intervertebral disc (IVD) could lead to a better understanding of site-specific properties relative to degradation, response to injury, and healing processes. The objective of this study was to determine how cell density, cell morphology, cell grouping, and expression of a specific actin isoform varied with location and degeneration in the human disc. A total of 41 human L4-L5 and L5-S1 discs removed postmortem from 21 individuals were analyzed. The discs were graded for degeneration based on the Thompson scale and processed for evaluation. Microtomed sections from paraffin-embedded specimens were stained with hematoxylin and eosin or a monoclonal antibody to α-smooth muscle actin (α-SMA), an actin isoform often associated with contraction. A significant regional dependence was found for most of the measured parameters. A fourfold increase in cell density was found in proceeding from the nucleus pulposus (NP) to the outer annulus (OA) of the IVD. Approximately 30% of the cells in the NP were present in groups. Virtually all of the cells in the NP and 40% of those in the OA were round. Moreover, notable percentages (12–15%) of the cells in the NP and inner annulus (IA) contained α-SMA. Only pair density was found to be correlated with Thompson grade, with more degenerated specimens having higher values. A greater effect was also observed on the percentage of cells in groups. These findings provide the basis for future work to investigate the importance of cells in groups, the role of α-SMA in the disc, and the changes in these cellular characteristics in pathological disc conditions.

Diversity of cell lengths in terminal portions of roots: implications to cell proliferation

Terminal meristems are responsible for all primary growth of roots. It has been asserted that all cells of root meristems are actively dividing (no cells cycle slowly or arrest in the cycle) and stem cell populations expand exponentially. Because cells do not slide relative to each other in roots, relative cell lengths may be used to determine relative cell cycle durations and/or proportions of cells actively dividing in root tissues. If all cells are cycling, no interphase cells should be longer than critical length (length of longest mitotic cell in the meristem) and cells should exhibit an exponential cell–age distribution. Lengths of all cells were obtained radially across entire median longitudinal root sections at 0.5, 1.0, 1.5, 2.0, 2.5 and 3.0 mm from the founder cell/root cap boundary for five plant species to estimate percentages of cells longer than critical length. For example, up to 15 and 90% of all interphase cells were longer than critical length in 0.5 and 2.0 mm tissues, respectively, indicating that slow-cycling and/or non-proliferative cells are present in such tissues. In order to determine if the distribution of cell lengths in 0.5 mm segments approximated an exponential cell–age distribution, lengths of interphase cells less than critical length were determined. Such interphase cells were placed into ten groups according to cell length and percentages of cells in each group were compared with percentages of cells in groups calculated from an exponential cell–age distribution. Percentages of cells were significantly different from predicted percentages of between 6 and 9 out of ten groups — cell lengths were not distributed exponentially. Because there are significant numbers of interphase cells longer than critical length and since lengths of interphase cells shorter than critical length do not resemble an exponential cell–age distribution, it must be concluded that not all cells in root segments from 0.5 to 3.0 mm root segments are actively dividing. Heretofore, no databases of cell lengths have been used to test these assertions.
