Evaluation of Wound Healing Effect of Curcumin Loaded OPL Carbon Nanospheres Embedded Chitosan Membranes

Biowaste-derived carbon biomaterial scaffolds are being used for wound healing and are the focus of interest. Carbon nanospheres derived from oil palm leaves without any catalysts via pyrolysis were loaded with a traditional drug curcumin. The wound healing scaffolds were fabricated on the PP non-woven fabric support using chitosan as the biopolymer matrix. Prepared carbon nanospheres and the scaffolds were characterized using ATR-IR and FESEM techniques. The wettability of scaffolds was examined to ensure the feasible moisture absorption ability, in vitro drug release profile and in vitro antibacterial activity against two strains of bacteria. The in vivo wound healing feature of scaffolds was studied by excision wound model for MRSA infected wound. Measured wound contraction percentage and the bacterial count on wounds at regular time intervals proved that, the scaffold dressed with chitosan and curcumin loaded carbon nanospheres showed an efficient reconstruction of skin through histopathological investigations.Graphical abstract

Fabrication, In Vitro, and In Vivo Assessment of Eucalyptol-Loaded Nanoemulgel as a Novel Paradigm for Wound Healing.

Wounds are the most common causes of mortality all over the world. Topical drug delivery systems are more efficient in treating wounds as compared to oral delivery systems because they bypass the disadvantages of the oral route. The aim of the present study was to formulate and evaluate in vitro in vivo nanoemulgels loaded with eucalyptol for wound healing. Nanoemulsions were prepared using the solvent emulsification diffusion method by mixing an aqueous phase and an oil phase, and a nanoemulgel was then fabricated by mixing nanoemulsions with a gelling agent (Carbopol 940) in a 1:1 ratio. The nanoemulgels were evaluated regarding stability, homogeneity, pH, viscosity, Fourier-transform infrared spectroscopy (FTIR), droplet size, zeta potential, polydispersity index (PDI), spreadability, drug content, in vitro drug release, and in vivo study. The optimized formulation, F5, exhibited pH values between 5 and 6, with no significant variations at different temperatures, and acceptable homogeneity and spreadability. F5 had a droplet size of 139 ± 5.8 nm, with a low polydispersity index. FTIR studies showed the compatibility of the drug with the excipients. The drug content of F5 was 94.81%. The percentage of wound contraction of the experimental, standard, and control groups were 100% ± 0.015, 98.170% ± 0.749, and 70.846% ± 0.830, respectively. Statistically, the experimental group showed a significant difference (p < 0.03) from the other two groups. The results suggest that the formulated optimized dosage showed optimum stability, and it can be considered an effective wound healing alternative.

Effects of resveratrol topical ointment on wound healing of full-thickness cutaneous burns in albino rats.

In this study, the effects of resveratrol topical ointment on wound contraction and histopathology of full-thickness cutaneous burn wounds were evaluated. Adult albino rats were grouped into four equal-sized groups of 15 rats each, as follows: Group A-no wound, no treatment (control); Group B-1% silver sulphadiazine; Group C-5% resveratrol, and Group D-wound without treatment (control). A burn wound measuring 23.5mm was created on the skin at the dorsum of all rats in groups B-D after shaving. The percentage of wound contraction was measured using a digital Vernier Caliper on days 1, 3, 5, 7, 10, 14, 16, 18 and 21, post-wounding. From each group, five rats were then euthanised and tissue samples of the skin, liver and kidney were collected in 10% buffered formalin for histopathology. The percentage of wound contraction was significant (p<0.05) on 7, 14 and 18 days post treatment. Histopathologically, 5% resveratrol topical ointment application resulted in a thicker epidermis with neovascularisation and an increased collagen distribution. Resveratrol topical ointment ameliorated the extent of hepatocellular and nephrotubular injuries following burn-induced hepatocellular and acute kidney injuries. In this study, topical application of 5% resveratrol ointment appeared to enhance burn wound healing by increasing the rate of wound contraction through collagen fibre synthesis, granulation tissue formation and epithelial regeneration.

Sterile thermoresponsive formulations for emergency management of burns

Initial care and treatment of burn injuries significantly influence their healing and appearance. The apt treatment for a burn patient normally depends upon the severity of the burn. This study was intended for the development of thermo-responsive formulations for the emergency management of burns prior to hospitalization.Nano calcium sulfate was prepared by probe sonication and nano copper oxide by thermal decomposition method and subsequently evaluated for particle size and poly-dispersity index. Formulations were prepared with 100 ppm of nano calcium sulphate, nano copper oxide and thermoresponsive polymers such as Poloxamer-407/ Chitosan/chitosan + Sodium β -glycerophosphate in different ratios. The products were evaluated for pH, viscosity and gelation time and those formulations which exhibited optimum viscosity, pH and gelation time were sterilized and evaluated for elemental content, skin irritation potential and in vivo burn healing activity.Nano calcium sulfate and nano copper oxide exhibited an average particle size of 220 nm and 273 nm correspondingly. SEM-EDX of selected formulations showed intense peaks of calcium and copper in all formulations. Acute dermal irritation study carried out on rabbits showed that the formations were non-irritant in nature. The percentage of wound contraction was greater in burn wounds treated with the formulation containing poloxamer-407 when compared to commercial Silverex™ ionic gel and the control group. The healing actvity of the thermoresponsive formulations was supported by histopathological studies.Nano calcium sulfate and nano copper oxide incorporated thermo-responsive formulations with burn wound healing activity were developed successfully.

Extraction, Characterization, and Structure of a Novel Heteropolysaccharide from Lepidium sativum and Its Effects on Wound Healing in Diabetic Rats.

The present study undertakes the extraction of a novel polysaccharide from Lepidium sativum (PLS) and the determination of its physicochemical composition and antioxidant properties, as well as its potential wound healing activity in alloxan-induced diabetic rats. This polysaccharide presented a lighter natural color, whose luminosity (L∗), red-green intensity (a∗), and blue-yellow intensity (b∗) were recorded at 63.26, 5.87, and 27.28, respectively. The PLS was structurally characterized by Fourier transform infrared (FT-IR) spectroscopy, UV spectrum, high performance liquid chromatography (HPLC), gas chromatography (GC), nuclear resonance magnetic (NMR), and high-pressure gel filtration chromatography. The FT-IR and UV spectra showed the characteristic band of polysaccharides. According to HPLC, the crude PLS is a heteropolysaccharide composed of glucose, xylose, and galactose. Results obtained by 1H NMR indicated that PLS consisted of three monosaccharide residues with α and β anomers. This novel polysaccharide had an average molecular weight of 98.51 kDa and displayed potential antioxidant activities determined through three different assays: scavenging activity against 2,2′-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS), 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging assay, and reducing power. These results strongly support the beneficial effects of the PLS to accelerate wound healing in diabetic rats. Indeed, its application significantly increased wound contraction percentage (98 ± 1.11%) after 14 days of experiment. Furthermore, the histological assessment of the PLS-treated group demonstrated complete reepithelialized wounds by accelerating collagen synthesis. In general, the findings affirmed that PLS is efficient on wound closure in alloxan-induced diabetic rats.

Development of curcumin-loaded liposomes in lysine–collagen hydrogel for surgical wound healing

BackgroundA surgical wound is an incision made by a surgeon. Slow surgical wound healing may lead to chronic wounds which may be a potential health problem. The aim of this study is to formulate curcumin-loaded liposomes in lysine–collagen hydrogel for enhancing surgical wound healing. Curcumin-loaded liposomes were prepared using thin-film hydration method. The liposomal formulation was characterized by analysing its size, morphology, encapsulation efficiency, and in vitro release. The hydrogel base was prepared, and then, curcumin-loaded liposomes were infused to give formulation (F1). Curcumin-loaded liposomes were infused into the hydrogel base after which lysine and collagen were incorporated to give (F2), while (F3) comprised lysine and collagen incorporated in hydrogel base. All formulations were characterized by evaluation of the safety, stability, swelling index, pH, rheological properties, and in vivo wound healing assay. Histology and histomorphometry of tissue samples of wound area treated with formulations F1, F2, and F3 and the control, respectively, were examined.ResultsCurcumin-loaded liposomes were 5–10 µm in size, and the values for encapsulation efficiency and flux of the loaded liposomes are 99.934% and 51.229 µg/cm2/h, respectively. Formulations F1, F2, and F3 had a pH of 5.8. F1 had the highest viscosity, while F2 had the highest swelling index indications for efficient sustained release of drug from the formulation. The in vivo wound healing evaluation showed that curcumin-loaded liposomes in lysine–collagen hydrogel had the highest percentage wound contraction at 79.25% by day three post-surgical operation. Histological evaluation reflected a normal physiological structure of the layers of the epidermis and dermis after surgical wound healing in tissue samples from wound areas treated with formulations F1 and F2. The histomorphometrical values show highest percentage of collagen, lowest inflammatory rates, highest presence of microvessels, and re-epithelization rates at wound site in wounds treated with formulation F2 (curcumin-loaded liposomes in lysine–collagen hydrogel).ConclusionCurcumin-loaded liposomes in lysine–collagen hydrogel was found to be the most effective of the three formulations in promoting wound healing. Hence, this formulation can serve as a prototype for further development and has great potential as a smart wound dressing for the treatment of surgical wounds.

Globularia arabica Methanolic Leaf Extract Has Higher Efficacy on Burn Wound Healing in Diabetic Rats Compared to Malva slyvestries Methanolic Leaf Extract.

This study aimed to see how effective Globularia arabica and Malva slyvestries-based cream formulations were at healing scald burn wounds in rats. Depending on ointment, preparations of 1%, 5%, and 10% w/w were created. For comparison, an ointment base and a regular burn cream composed soframycine were utilized. Rats introduced a burn by solidifying equipment at 100°C on a 14-mm2 shaved dorsal region. A deep second-degree burn was created, and the percentage of wound contraction was measured over the next 15 days. The rats were euthanized on days 8 and 15, and histological slides were prepared using hematoxylin and eosin staining. Compared to the control group, there was a substantial increase in wound contraction and a significant decrease in the duration of epithelialization in the based ointment-treated groups. However, as paralleled to Globularia arabica, significant (P < .05) results were observed with 10% Globularia arabica cream, whereas Malva slyverstries indicate minimal healing. Soframycine causes a substantial increase in wound contraction (P < .05). Soframycine cream with 10% Globularia arabica therapy resulted in practically complete re-epithelialization and re-structuring of wound tissue on histological examination, whereas Malva slyversries treatment resulted in low epithelization during treatment days. The findings suggest that Globularia arabica-based cream has the wound-healing capability.

Investigation of wound healing potential of photo-active curcumin-ZnO-nanoconjugates in excisional wound model

Wound healing, being a dynamic process consisting of hemostasis, inflammation, proliferation, and remodeling, involves the complicated interplay of various growth mediators and the cells associated repair system. Current wound healing therapies usually fail to completely regain skin integrity and functionality. Traditionally, curcumin is considered a potent natural wound healing agent as it possesses antibacterial, antioxidant, and anti-inflammatory properties. It is also known that zinc oxide (ZnO) nanoparticles (NPs) have photocatalytic properties, including the generation of reactive oxygen species. ZnO nanoaprticles are also Food and Drug Administration (FDA) approved as safe substances. While ZnO oxide requires illumination with ultraviolet light to become photocatalytically active, dye-sensitized ZnO can be activated by illumination with visible light. In the present study, we explored the wound healing potential of ZnO nanoparticles sensitized with curcumin (Cu+ZnO Nps) and illuminated with visible (blue) light generated by an array of high power LEDs. We studied the antibacterial effect of our conjugates by percentage reduction in bacterial growth and biofilm formation. The wound healing potential was analyzed by percentage wound contraction, biochemical parameters, and histopathological analysis of the wounded site. Additionally, angiogenesis and wound associated cytokines was evaluated by immunohistochemistry of CD31 and gene expression analysis of IL-1β, TNF-α, and MMP-9 after 16 days of post-wound treatment, respectively. Our study suggests that the therapeutic effect of Cu+ZnO NPs with LED illumination increases its wound healing potential by producing an antibacterial and anti-inflammatory effect. Moreover, the treatment strategy of using a nano formulation in combination with LED illumination further increases its efficacy. It was concluded that the anti-inflammatory and bactericidal effects of the LED illuminated Cu+ZnO Np showed accelerated wound healing with increased wound contraction, collagen deposition, angiogenesis, and re-epithelialization.

Effect of Collagen/Ibuprofen Hydrogel in Wound Healing: An In Vivo Study

Background and Aim. Wound healing is a complicated physiological process to preserve skin integrity after injuries and includes the proliferative phase, hemostasis/inflammatory phase, and remodeling through extracellular, intercellular, and intracellular components synchronization. The study aimed to examine human placental collagen/ibuprofen’s effect on wound healing in an animal model. Materials and Methods. The cell viability test was performed by MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) assay. In this animal study, three circular excisions were made in the back of each of the 12 rats and injected with the following groups: collagen/ibuprofen, group 1; collagen, group 2; and unfilled as the control group, group 3 for two times. The healing procedures were explored via calculating wound contraction percentage after taking photographs on days 7 and 14. Also, histopathological assessments were conducted on days 7 and 14. Results. The MTT results showed no significant cytotoxicity, and macroscopic results demonstrated that the percentage of wound contraction was 85%, 79%, and 41% in the collagen/ibuprofen, collagen, and control groups on 14th day, respectively. Histopathological assessments showed that hair follicles number, fibroblasts content, and angiogenesis in the collagen/ibuprofen were remarkably more significant than collagen and control groups. Conclusion. The collagen/ibuprofen group had significant outcomes compared with collagen and the control groups in wound healing and wound contraction. Collagen/ibuprofen hydrogel can be a potential hydrogel in accelerating wound healing.

Antibacterial Effects of Commiphora gileadensis Methanolic Extract on Wound Healing.

Commiphora gileadensis (CG) is a small tree distributed throughout the Middle East. It was traditionally used in perfumes in countries in this area. In Saudi Arabia, it was used to treat wounds burns and as an antidote to scorpion stings. This study aimed to evaluate the antimicrobial activity and cutaneous wound healing efficiency of the CG extracts using microbiological tests, rate of wound contraction and histopathological changes. CG plant were extracted using the methanol extraction technique; then, the methanolic extract was characterized using liquid chromatography coupled with mass spectrometry (LC–MS). Afterwards, a six-millimetre (mm) excision wound was induced in 60 male Balb/c mice. Mice were classified into two classes; each class consisted of three groups of 10 mice. In the non-infected wound class, the group I was assigned as control and received normal saline. Group II received gentamicin treatment, and group III treated with CG-methanolic extract. In the Staphylococcus aureus-infected class, group IV received normal saline, and groups V and VI were treated with gentamicin and CG-methanolic extract, respectively. The colonization of infected wounds was determined using colony-forming units (CFUs), and the percentage of wound contraction was measured in all groups. Finally, the histopathologic semi-quantitative determination of wound healing was evaluated by inflammatory cell infiltration, the presence of collagen fibres and granulation tissue, and the grade of re-epithelization. Composition analysis of the methanolic extract confirmed the presence of a high amount of ceramide (69%) and, to a lesser extent, hexosylceramide (18%) and phosphatidylethanolamine (7%) of the total amount. Additionally, there was a statistically significant difference between the percentage of wound contraction in the CG-treated and control groups in both Staphylococcus aureus-infected and non-infected wounds (p < 0.01). The colonization of the infected wounds was lower in the group treated with CG than in the control group (p < 0.01). In both non-infected and infected wounds, the CG-treated group showed significant statistical differences in inflammatory cell infiltration, collagen fibres, re-epithelization and granulation tissue formation compared with the control group (p < 0.01). The CG extract possesses antibacterial and anti-inflammatory properties that induce wound healing.

The Effects of Javanese Turmeric (Curcuma xanthorriza Roxb) on Fibroblasts, Granulation, Blood Vessel Density, and Contraction in Wound Healing of STZ-Induced Diabetic Rats

Diabetic ulcers usually occur in the lower extremities of diabetic patients. One in 20 diabetic patients will develop an ulcer, and 10% of those cases lead to amputation. This study aimed to investigate the effects of Javanese turmeric (Curcuma xanthorriza Roxb.) extract on the number of fibroblasts, granulation, blood vessel, and the rate of wound contraction in a Wistar rat model of the diabetic ulcer. This was an experimental study with post-test observations only and randomized control group design. Rats were divided into five groups: (1) negative control (KN); (2) positive control (KP); (3) P1; (4) P2; and (5) P3. Every rat was assessed for fibroblast number, granulation, blood vessel density, and wound contraction percentage. Javanese turmeric extract had a significant effect on histological parameters (fibroblast, blood vessels and tissue granulation) (p &lt; 0.05) and wound contraction (p &lt; 0.05). The double linear test revealed a significant relationship between fibroblasts, granulation, blood vessel quantity, and wound contraction (p &lt; 0.05). Correlation and regression tests showed that Javanese turmeric extract explained 91% of the effects on fibroblasts, blood vessels, and granulation. Treatment with Javanese turmeric extract increased the number of fibroblasts, tissue granulation, blood vessel density and wound contraction in male diabetic Wistar rats.

Topical bilirubin-deferoxamine hastens excisional wound healing by modulating inflammation, oxidative stress, angiogenesis, and collagen deposition in diabetic rats

Aim of the studyThe study was performed to understand the detailed mechanism of diabetic wound healing by bilirubin-deferoxamine (DFO) combination on topical application. Materials and methodsThere were two study groups, control, and treatment. The granulation tissues collected on different days (3, 7, 14, and 19) were studied in detail for inflammatory mediators, angiogenesis markers, epithelialization, and oxidative stress parameters. ResultsA significant increase in wound contraction percentage was observed from day 7 in the bilirubin-DFO treatment group. The combinatorial treatment significantly reduced tumour necrosis factor-alpha (TNF-α) and interleukin-1 beta (IL-1β), and enhanced IL-10 levels. Upregulated mRNAs of vascular endothelial growth factor (VEGF) and hypoxia-inducible factor-1 alpha (HIF-1 α) along with CD31 immunohistochemistry showed the pro-angiogenesis potential of the combination. Hematoxylin and Eosin (H and E) staining and Masson's trichrome staining showed reduced inflammatory cell infiltration, enhanced fibroblast proliferation, well-organized collagen fibers, and the development of new blood vessels. Collagen deposition is further supported by immunohistochemistry studies and Masson's trichrome staining. Bilirubin-DFO combination also reduced lipid peroxidation and elevated antioxidative enzymes. ConclusionTopical application of bilirubin-DFO showed immense potential in augmenting skin wound regeneration in diabetes by upregulating the antioxidant status as well as increasing angiogenesis, collagen deposition, and modulating cytokines.

Topical Application of Premna integrifolia Linn on Skin Wound Injury in Rats Accelerates the Wound Healing Process: Evidence from In Vitro and In Vivo Experimental Models.

Background When the skin and tissues within the body are injured, the healing process begins. Medicinal herbs have been used to cure wounds since time immemorial. The antimicrobial and antioxidant activity possessed by P. integrifolia may accelerate wound healing. Objectives To assess the wound healing activity of Premna integrifolia extract (PIE) by employing in-vivo experimental animal models and an in-vitro migration scratch assay. Furthermore, to assess its cytotoxicity using the MTT assay. Methods Wistar albino rats were used for the in vivo wound healing models. The animals were divided into four groups at random: Group I was untreated. Group II was vehicle control (ointment base). Group III was PIE ointment (5% W/W). Group IV was standard (povidone-iodine ointment) (5% W/W). The ointments were applied directly to the wounds as described above until they healed completely. The wound contraction percentage and tensile strength were calculated. The MTT test was used to determine the viability of the test extract against the fibroblast cells. The scratch assay was used in vitro to determine the wound healing potential of the test drug. P ≤ 0.05 values were considered statistically significant. Results Premna integrifolia extract did not possess any noticeable cytotoxicity to the cell line and showed an IC50 of 185.98 μg/ml. The wound contraction potential of PIE ointment-treated animals was considerably greater (P ≤ 0.001) on days 4, 8, 12, 16, and 20 when compared to the control group. The percentage of wound contraction on day 20 was 99.92% in PIE-treated animals compared to 83.23% in untreated animals. Compared to the untreated group, the duration of full epithelization was significantly (P ≤ 0.01) shorter in the test group. When compared to the incision control group, the animals treated with PIE ointment had significantly higher (P ≤ 0.001) tensile strength. In addition, animals given the test drug had a significant (P ≤ 0.001) increase in total protein and hydroxyproline. In the in vitro scratch assay, test drug-treated cells demonstrated greater cell migration. Histology images confirmed that the test drug-treated group had epithelial tissue proliferation and keratinization. Conclusion The current study found that Premna integrifolia improved wound healing activity both in vitro and in vivo. These findings indicate that Premna integrifolia extract has wound-healing potential and could be a viable source of nutraceuticals with wound-healing properties.

The Effect of Eurycoma longifolia Jack Tongkat Ali Hydrogel on Wound Contraction and Re-Epithelialization in In Vivo Excisional Wound Model

BACKGROUND: Wound management is one of the significant health problems throughout the world. Medicinal plants have been used widely in wound management. Eurycoma longifolia Jack which is known as Tongkat Ali (TA) is a tropical medicinal plant in South East Asian countries. AIM: The aim of the study was to investigate the effect of (TA) hydrogel on wound contraction and re-epithelialization in excisional wound model in rats. METHODS: Twenty male Sprague Dawley rats were divided into four groups each group contained five rats (n = 5). Animal treatment groups are formed as: Untreated (−ve) control, Hydrocyn® aqua gel (+ve), vehicle hydrogel, and (TA) hydrogel. A full-thickness circular excisional wound was created on the dorsal back of each rat. The wounded area was measured and photographed on days 3, 6, 9, 12, 15, and 18 post wounding to determine the percentage of wound contraction and re-epithelialization. RESULTS: (TA) hydrogel showed significant increase in the percentage of wound contraction by 43.38% compared with the other groups (p = 0.032, p &lt; 0.050) during the first interval (inflammatory phase). Although in the later healing stages (proliferative and remodeling) and re-epithelialization, our test group (TA) hydrogel did not show statistically difference with the other groups yet it was comparable to medically certified wound healing agent. CONCLUSION: (TA) hydrogel significantly accelerated the wound healing process during the early stage, the inflammatory stage. Whereas during the later healing stages and re-epithelialization, it showed almost the same effect of Hydrocyn® aqua gel.

Efficacy of 10% Calendula officinalis extract gel for oral wound healing in rats

This study assessed the efficacy of 10% Calendula officinalis (C. officinalis) extract gel for oral wound healing in rats. Thirty-two male Wistar rats were randomized into test and control groups (n=16). The rats were anesthetized to create a wound in their buccal mucosa by a #3 puncher. Next, 10% C. officinalis gel and a placebo gel were applied over the wounds in the test and control groups, respectively. Each group was randomly divided into two subgroups for assessment at 7 and 14 days (duration of treatment). At each time point, a biopsy sample was obtained from the wound and type of inflammatory cells, severity of inflammation, type of connective tissue, and percentage of wound contraction were assessed by a pathologist. Data were analyzed by univariate ANOVA and Chi-square test. The mean percentage of wound contraction was 28.28% and 62.58% in the test and -15.98% and 7.08% in the control group at 7 and 14 days, respectively, indicating significantly greater wound contraction in the test group at both time points (p&lt;0.001). The type of connective tissue was significantly different between the two groups at 14 days (p&lt;0.05). The number of giant cells was significantly higher in the test group than the control group at 7 days (p&lt;0.05). No other significant differences were noted. Application of 10% C. officinalis gel improved wound contraction and enhanced healing but had no significant effect on the severity of inflammation in traumatic oral ulcers in rats.

810 Synthetic Platelet-Mimetics with Gentamicin: Outcomes in Deep Partial-Thickness Burns

IntroductionInfection and prolonged inflammation in deep partial-thickness burns can lead to inadequate healing. In addition to their role in hemostasis, activated platelets also contain granules with anti-inflammatory properties which may impact wound healing. However, portability and storage of platelets remain a challenge. Synthetic platelet-mimetics (SPM) avoid these difficulties, but like natural platelets, SPM have vesicles that can transport bioactive agents, such as antibiotics. We sought to evaluate wound healing outcomes in deep partial-thickness burns treated topically with SPM.MethodsA total of 30 circular, deep partial-thickness burns were created on the dorsum of 2 porcine models. Each wound measured 5cm in diameter and was standardized using a thermocoupled burn device. Sets of six wounds were randomized into five groups: SPM, gentamicin alone, SPM with gentamicin, a vehicle control (saline), or dry gauze (the standard of care, SOC). Additionally, two separate 5cm diameter circular areas were demarcated to serve as normal skin controls. Wounds were assessed at post-burn days 3, 7, 14, 21, 28, 60 and 90 with a variety of non-invasive imaging and punch biopsies. Treatments were re-applied at each assessment. The primary outcome was the amount of wound re-epithelialization, measured histologically, at post-burn day 28. Secondary outcomes consisted of the percentage of wound contraction, amount of superficial blood flow, and mean bacterial load.ResultsThe amount of wound re-epithelialization in wounds treated with SPM was 96% whereas those treated with the SOC measured only slightly less at 92% (p = 0.56). The percentage of wound contraction was 39% in the SPM with gentamicin group, while in the SOC group contraction was higher at 45% (p = 0.20). Moreover, superficial blood flow in the SPM group was measured to be 168% of normal skin controls, while wounds in the SPM with gentamicin group were slightly lower at 160%, and in wounds treated with the SOC blood flow was 110% of normal skin controls (p = 0.27). The mean bacterial load in each group was measured at post-burn day 3 and notably consisted of bacterial load being the lowest in wounds treated with gentamicin alone at 17/100, meanwhile in wounds treated with the SOC or those in the SPM with gentamicin group, mean bacterial loads were significantly higher at 43/100 and 47/100, respectively (p = 0.02).ConclusionsSPM applied to deep partial-thickness burns did not significantly improve measured outcomes over the SOC. Although numerical improvements were shown in all measured outcomes, no significant differences were noted. The theoretical and observed potential for SPM to improve wound healing deserves additional evaluation in larger preclinical studies.

Evaluation of wound healing activity of topical nicotine in an excision wound model in rats

Background: Wound can be defined as disruption of cellular, anatomical, or functional continuity of living tissues. Nicotine causes damage to the epithelial layer of blood vessel and delays wound healing. It plays an important pathogenic role in impaired wound healing. Although in the last millennium, topical use of nicotine has been reported. It promotes collagen synthesis and, in turn, promotes wound healing. The role of topical nicotine on wound healing is controversial. Therefore, it was planned to evaluate and compare wound healing activity of various doses topical nicotine in rats. Aim and Objectives: The objectives of this study were to evaluate the effect of topical nicotine on wound healing in an excision wound model in rats. Materials and Methods: For evaluation of the wound healing effects of the nicotine, excision wound model was used. Nicotine was applied topically in a dose of 1.5 g%, 3.0 g%, and 6.0 g% petroleum base. Petroleum jelly served as control for topical nicotine. Dressing done by applying topical nicotine until (20 days) complete wound healing was observed. Parameters evaluated were surface area of wound and percentage closure. Results: Findings of this show that, on day 4, nicotine 3.0 g% and 6 g% the wound surface area were more as compared to control. On day 12, nicotine 6.0 g% showed significantly more wound surface area than control (P &lt; 0.05). Percentage wound contraction with topical nicotine (6.0 g%) was less as compared to control on day 4, 8, and 12 (P &lt; 0.001). On day 16, percentage wound contracture with topical nicotine (6.0 g%) contraction was significantly less as compared to control (P &lt; 0.05). Although percentage wound contraction with topical nicotine (all preparations) and control was similar on day 20. Conclusion: Finding shows that topical nicotine impairs wound healing in a dose related pattern during initial stages of healing in an excision wound model. However, there is no delay in wound healing with any dose of topical nicotine.

Avaliação de células mesenquimais e dapsona para o tratamento de feridas dermonecróticas causadas pelo veneno de Loxosceles laeta em coelhos

Abstract This study aimed to evaluate the efficacy of mesenchymal stem cells (MSC), alone or associated with dapsone (DAP), in treating dermonecrotic wounds caused by Loxosceles laeta venom. Twenty-five male rabbits were distributed into five groups. Negative control received ultrapure water (C-), whilst all other groups were injected with 20 μg of L. laeta venom. After 4 hours, each group received one of the following treatments: PBS (C+), DAP, MSC, and DAP+MSC. Animals were evaluated daily and photographic records made for analysis of wound area. Twelve days after, animals were euthanized and skin samples removed for histological analysis. We observed that DAP showed the best percentage of wound contraction at day 3. In the treatments using MSCs, a negative value of wound contraction was observed for the isolated MSCs, as well as a lower contraction value for the association of the MSC + DAP when compared to PBS, probably, by the increase in initial infammation after the application of stem cells, due to the fact that MSCs secrete a broad spectrum of bioactive molecules such as cytokines and growth factors that favor regeneration. Histologically, it was observed that animals of C+ showed extensive areas of necrosis, ulcers, neutrophilic infiltrate, and mineralization. Collagen deposition showed increase in MSC+DAP treatment, however vascularization remained unchanged. This is the first report using MSC and MSC+DAP as a treatment for cutaneous loxoscelism and more studies are needed to determine its use as an alternative therapy for dermonecrotic lesions caused by Loxosceles spider.

Dermal toxicity and wound healing activity of Coriandrum sativum L. leaves extract and its formulated ointment in wistar rats

Wound is referred as any physical injury that breaks the skin layer. Wound healing is known as the regeneration of tissues whereby the new layer of skin is formed. Wound healing follows a cascade of events starting with homeostasis, inflammation, proliferation and remodeling of tissues. Although there were many studies have been conducted proving various traditional plants exhibiting wound healing properties but wound healing properties of coriander leaves were limited. Therefore, this research was conducted to determine the wound healing activity using the extract of Coriandrum Sativum L leaves (CSL) using formulated ointment in Wistar rats. CSL were extracted successively using methanol in a soxhlet extractor for 36 hours at 60°C. Skin toxicity was done on rats. For screening excision wound, animals were divided into four groups (n=6). Group-1 is treated with vehicle (control). Group-2 and 3 were treated with 10% and 20% of extract ointment respectively. Group-4 was treated with standard Betadine ointment. Methanolic extract of C. sativum L. (MECS) was applied topically on the excision wound of the rat for two times per day. The parameter observed were percentage of wound contraction. ANOVA test was used to analyze the results obtained for excision wound was statistically significant (P<0.05). The MECS extracts with 20% extract has shown a significant wound-healing activity that was supported by decrease in the period of epithelialization and also increase in rate of contraction of wound in experimental animals. The MECS extracts have proved to possess wound healing properties and a higher concentration of the extract has facilitated as a good choice of remedy for woundhealing.

Phytosomes of Naphthoquinone enriched Extract of Root bark of Onosma echioides exhibit wound healing activity in Rats

The present study is aimed at evaluation of phytosomal gel of the petroleum ether extract of root bark of Onosma echiodes for wound healing activity in rats. Extract of root bark of O. echioides was standardized by isolated naphthoquinone dimer using HPTLC. Phytosomes (equivalent to 2% w/w of naphthoquinones) of the standardized extract were prepared by thin film hydration technique. The wound healing efficacy of the formulation was evaluated in rats by inflicting excision and incision wounds followed by treatment of the wounds topically. The parameters evaluated for healing included determination of breaking strength and tensile strength of healed skin for incision model and percentage wound contraction, hydroxyproline content, granulation tissue free radicals and catalase in excision wound model. The formulation treated group showed a significant healing (p&lt;0.005) of both the excision and incision wounds with respect to wound contraction and tensile strength respectively, as compared to vehicle treated group. The oxidative stress of the granulation tissue was also found to be reduced as indicated by reduced lipid peroxidation and increase in catalase activity. The phytosomal gel of O. echioides effectively exhibited wound healing effect.
