Screening of germplasm of peach and nectarines against Taphrina deformans

The leaf curl caused by Taphrina deformans (Berk.) Tul. is the most serious disease of stone fruits viz., peachs and nectarines which counts for loss in quantitative and qualitative yield. Germplasm of these stone fruits was screened against T. deformans and data in terms of disease incidence (%) and disease severity (%) were recorded. It was observed that in peach, cent percent disease incidence was recorded in Glo haven, Pratap, Shan-e-punjab and Florida Peach while, minimum in Androse (30%) whereas, maximum disease severity was observed in Red Haven (1.13%), Pratap (1.10%), Early Elberta (1.07%) and minimum in Early Grande (0.53%). In nectarine, cent per cent disease incidence was observed in Independence, Early Red Haven, Punjab Nectarine and Syria while, minimum disease incidence (33.33%) and severity (0.3%) was recorded in Silver of Rome whereas, maximum disease severity was observed in Red Gold (1.58%), Independence (1.2%) and Early Red Haven (1.2%).

Effective biocontrol of banana fusarium wilt tropical race 4 by a bacillus rhizobacteria strain with antagonistic secondary metabolites

Fusarium oxysporum f.sp. cubense Tropical Race 4 (FOC TR4), is one of the newly emerged dreaded pathogens of banana with an established global presence. Fungicides have not been effective in controlling the disease. There are very few effective antagonistic microbes reported for the biological management of this pathogen.In the present study, suppressive soil was selected for isolation of potential biocontrol plant growth-promoting bacteria. The bacterial strain CSR-D4 Bacillus licheniformis was identified with significant antagonistic attributes and plant growth-promoting properties. This strain exhibited a high inhibition percentage (77.59%) against the FOC TR4 pathogen in the dual plate assay. Further, the bioactive metabolite of the strain conferring biocontrol of FOC-TR4 was also identified through the Liquid Chromatography-Mass Spectrophotometry technique. Our analysis revealed the presence of many antifungal metabolites in the isolate CSR-D4, the majority of which were identified as iturin, fengycin, surfactin, and bacillomycin. These results indicated the involvement of metabolites in the antifungal activity of the strain CSR-D4 against FOC TR4. During sick-pots studies taken with susceptible Cavendish banana cultivar Grand Naine, application of B. licheniformis CSR-D4 showed a significant reduction in the disease incidence percentage (10%). B. licheniformis CSR-D4 treated plants also showed enhanced activity of defense-related enzymes such as β-1,3 glucanase, peroxidase, chitinase, polyphenol oxidase, and phenyl-alanine ammonium lyase compared to FOC TR4 alone treated plants that suggest the occurrence of induced host tolerance in Bacillus licheniformis CSR-D4 treated plants. Our results suggest the antagonistic potential of rhizobacterium Bacillus licheniformis CSR-D4 strain through secreted metabolites mediated the effective biocontrol of Fusarium oxysporum f.sp. cubense Tropical Race 4.

Evaluation of some botanicals and microbial bioformulations against grey blight disease of tea (Camellia sinensis)

Grey blight disease of tea (Camellia sinensis) is considered as a major threat to tea plantation owing to adverse affect on yield and quality parameters in tea. In the present study, an effort was made to explore some botanicals and microbial formulations for management of grey blight disease under field condition in the pretext of growing demand of organic tea. Seven botanicals at three concentrations (5.0, 10.0 and 15.0%) and four microbial bioformulations were screened in vitro against Pestalotiopsis theae, the grey blight pathogen. Of the botanicals, significant inhibition was recorded by water extract of Xanthium strumarium and Pongamia pinnata at 15% concentration. Biogreen 5 (Formulation of Trichoderma viride, Pseudomonas fluorescens, Bacillus thuringiensis, Beauveria bassiana and Metarhizium anisoplae) and Bioveer (T. viride) among the bioformulations recorded highest reduction in mycelial growth of pathogen. The two most effective botanicals and bioformulations screened in vitro were further evaluated individually and in combination under field condition for two consecutive seasons. Lowest percent disease incidence (5.23%) was recorded with combination of Biogreen 5 and Bioveer followed by Bioveer alone (7.79%) at 150 days of spray. Foliar spray of X. strumarium and P. pinnata in combination also showed satisfactory reduction in grey blight incidence with PDI of 17.97%. The study revealed that both bioagents and botanicals could be an ecofriendly and sustainable solution for grey blight disease management under organic tea production system.

Identification of new sources of resistance to dry root rot caused by Macrophomina phaseolina isolates from India and Myanmar in a mungbean mini-core collection

Dry root rot (DRR), caused by Macrophomina phaseolina, is a prevalent disease of mungbean in Myanmar, and an emerging problem in South Asia. The pathogen is a polyphagous necrotroph, survives in the soil for many years that results disease mitigation difficult. Managing DRR in mungbean through an integrated approach has been suggested, and the use of resistant varieties is one of the economical methods. The present study aimed to identify sources of resistance against DRR from a mungbean mini-core collection and to characterize the associated M. phaseolina isolates from India and Myanmar. Evaluation of the 296 mungbean mini-core accessions against the isolate MP1 by paper towel method identified 29 accessions with DRR resistance (disease scores: ≤ 3), and 18 of them with the consistent resistance in the repeated experiment. During the screening of 18 resistant accessions in the glasshouse, nine accessions were found DRR resistance in repeated sick pot experiments with ≤10% disease incidence. A subset of 30 accessions was selected from the mini-core collection based on their in vitro DRR reactions. These accessions were evaluated for DRR resistance in the field in Yezin, Myanmar in 2018 and 2019. Out of the 30 accessions, ten accessions were found DRR resistance with ≤10% disease incidence in both years of evaluations. Pooled analysis of percent disease incidence data of 15 accessions common in both glasshouse and field revealed the stability of accessions VI001509AG, VI001244AG, and VI001400AG for DRR resistance across years and locations. The three resistant accessions along with a susceptible check VC693088 were re-evaluated by paper towel method against nine additional M. phaseolina isolates from India (MP3-MP11). The accessions VI001509AG and VI001400AG were resistant to all nine isolates, while accession VI001244AG was resistant to MP5, MP6, and MP7 isolates. These accessions could be used in mungbean DRR resistance breeding programs.

Survey and incidence of rice sheath blight in major rice growing areas of eastern Uttar Pradesh

Rice is an important food grain and is a staple food for majority of the world’s population. Sheath blight (ShB), caused by the soilborne necrotrophic fungal pathogen Rhizoctonia solani Kuhn, is an economically important disease in rice. A purposive disease survey was conducted during Kharif-2019 to assess the occurrence and spread of rice sheath blight disease in five districts of eastern Uttar Pradesh. The percent disease incidence (PDI) ranged from 20 to 80%. Maximum PDI (80) was noticed at paddy field of Sadalpura block (80%), while minimum (20) was recorded at Narayanpur, and Maudli villages. The maximum severity (9 scale) was observed at Sadalpura, Satahara, Naini, Cholapur. The disease was recorded from panicle initiation to grain hardening stage.

Integrated management of vascular wilt of lentil caused by Fusarium oxysporum f. sp. Lentis

An extensive study was undertaken to manage the most vigorous, polyphagous pathogen Fusarium oxysporum causing wilt disease in lentil. A field experiment was conducted during 2017-18 cropping season to evaluate different management options (seed treatment with thirum, Trichoderma harzianum and soil drenching alone with propiconazole and in combination with organic manures) on wilt of lentil caused by Fusarium oxysporum f. sp. Lentis under artificial inoculation condition in pots and field. It was observed that the minimum 2.56% disease incidence was recorded in soil application of Propiconazole @ 500 ml/ha 15 DAS as compared to control (66.66%) in pot condition. At field level the lowest percent disease incidence (5.05%) was recorded in soil application of Propiconazole (@ 500 ml/ha after 15 DAS, whereas 7.07 percent disease incidence was observed in soil application of Neem cake @0.1 kg m-2 + Tricoderma harzianum @1 g m -2 and soil Application of Tricoderma harzianum @1 g m -2 + Press mud @ 2.5 kg m-2 compared to control (14.14%) in field condition.

Prevalence and Assessment of Disease Incidence and Severity in Citrus Orchards of Sub Zoba Hamelmalo

Citrus (Citrus spp. L.) production in Eritrea covers over 2409 ha of the land but the average potential of production is 91 q/ha to 150 -170 q/ha. This low yield might be due to lack of knowledge on management practices, adverse environmental factors and different diseases caused by various pathogens. Moreover, there is no proper recorded data on the identification of diseases on citrus and its intensity, hence, this survey: to identify the prevailed diseases; assessment of disease incidence, severity and calculate the ‘Area Under Disease Progress Curve’ (AUDPC). A survey was carried out in three orchards in each five citrus producing villages (Fredarb, Tsebabo, Hamelmalo, Wazntetand Genfelom) from sub-zoba Hamelmalo. The fungal diseases found on citrus were brown spot, sooty mold, gummosis, black spot, scab and melanose; the bacterial infection was canker; and viral diseases were leprosis and cholorotic dwarf. The highest percentage of disease incidence was recorded Fredarb with 77.16% and the lowest was found in Genfelom with 47.29 % in the same month. The highest percentage of diseases severity was found in Hamelmalo with 40.02%. On the other hand, the least percentage of disease severity was found in Genfelomwith 25.04%. The influence of epidemiological factors showed that they could be some of the reasons to develop and spread the mean disease incidence (65.6%) in February, 2018. The AUDPC was recorded highest (2829) in the orchard 3 of Hamelmalo, and lowest (1152) in orchard 3 of Tsebabo. A decreasing trend of temporal disease data from the first month to the third month was estimated in the orchard 2 and orchard 3 of Fredarb, orchard 2 of Genfelom, orchard 1 of Hamelmalo, all orchards of Tsebabo and in both orchard1 and 3 of Wazntet villages.

Association of causative agents of root rot complex of soybean in northern Karnataka

Examination of pure culture of the pathogens collected from the root samples of soybean plants showing typical wilting symptoms revealed that the major pathogens involved in causing root rot complex are Sclerotium rolfsii Sacc. Rhizoctonia bataticola and Fusarium sp. in northern Karnataka. The pathogenicity studies on susceptible cultivar JS 335 inoculated with all the pathogens revealed maximum percent disease incidence in case of dual inoculation of S. rolfsii + R. bataticola (83.33%) and mixed inoculation of S. rolfsii + R. bataticola + Fusarium sp. (88.50%) followed by dual inoculation of Fusarium sp. + R. bataticola (75%) and S. rolfsii + Fusarium sp. (72.34%).

Effect of pulsed magnetic field on seed borne pathogen and germination of tomato

Effect of Pulsed Magnetic Field (PMF) treatment on the control of Alternaria solani (Ellis and Martin) Jones and Grout causing Early blight of tomato (Solanum lycopersicum L.) cultivar PKM 1 was studied. The seeds pre-inoculated with A. solani were 1500nT PMF treated with frequency of 10 Hz for 5 h for varied durations of 0, 5, 10, 15, 20 and 25 days. PMF treatment significantly reduced pathogen incidence and increased both germination and plant growth in comparison with the control. Experimental results indicated that all the treatments were significantly different and the seeds treated for 20 and 25 days showed the minimum percent disease incidence (71%) with highest percent disease inhibition over control (29%). It also recorded the highest plant biometrics viz., germination (81%), speed of germination (7.74), root length (13.37 cm), shoot length (6.45 cm) and vigour index (1606). Control recorded the minimum plant biometrics viz., germination (61%), speed of germination (7.27), root length (12.92 cm), shoot length (6.17 cm) and vigour index (1164).

Identifikasi Cendawan Patogen Penyebab Penyakit pada Daun Jabon Merah (Anthocephalus macrophyllus (Roxb.) Havil)

Red Jabon (Anthocephalus macrophyllus (Roxb.) Havil) is a forest plant that can replace sengon (Falcataria moluccana) because it has advantages. The demand for red jabon wood is increasing, but the cultivation of red jabon is still constrained by the attack of pathogens that cause leaf disease. The attack of these pathogens can cause damage and death of seedlings so that it can be economically detrimental. The aim of this study was to identify the pathogens that cause disease in red Jabon. The method used is the Postulate Koch method, which starts from sampling red leaves with leaf disease symptoms, isolation, inoculation and analysis of disease incidence, re-isolation, and identification. Based on the identification results found as many as 2 pathogenic fungi that cause leaf spot disease, namely Pestalotia sp. and Rhizoctonia sp. In addition, 6 pathogenic fungi were found that cause blight, namely Lasiodiplodia theobromae, Fusarium sp., Colletrotrichum sp., Marssonina sp., Gloeosporium sp. 1, Gloeosporium sp. 2.

Comparative Management Studies on Banded Leaf and Sheath Blight of Maize Caused by <i>Rhizoctonia Solani F.</i><i>SP</i><i>. Sasakii</i>

Bioassay of biocontrol agents and fungicides against Maize Banded Leaf and Sheath Blight caused by Rhizoctonia solani showed inhibition. Among the biocontrol agents, Bacillus subtilis and among the fungicides Hexaconazole was found to be effective under field conditions with the percent disease incidence of 2.25% and 3.00% respectively over control 10.45%. Seed treatment (4g/kg) and soil application (2.5kg/ha) of Bacillus subtilis recorded 78.46% reduction in sheath blight incidence and highest grain yield of 6.7t/ha. Foliar spraying of Hexaconazole (0.2%) showed 71.30% reduction and grain yield of 6.4t/ha. Hence, seed treatment and soil application of Bacillus subtilis is considered to be effective against BLSB under field condition. Seed treatment and soil application of Bacillus subtilis, Foliar spraying of Hexaconazole may be recommended for Banded Leaf and Sheath Blight of Maize.

Evaluation of efficacy of chemical, botanicals and beejamrut in growth promotion and management of damping off disease in cauliflower at Udayapur, Nepal

Vegetable contributes 20.74% of the total Agricultural Gross Domestic Products (AGDP) of the country. Among the vegetables produced, cauliflower (Brassica oleracea L. var. botrytis) is one of the important vegetable crops in Nepal. Soil borne pathogens (soil inhabitants and soil transients) are one of the major factors contributing to lower yield in vegetables either through damage of whole crop, or by making them unmarketable. Damping off caused by Pythium, Fusarium, Rhizoctonia, Phytophthora is one of the major disease of vegetables crops. This disease attacks a crop in its two stages i.e. pre-emergence of seeds and in seedling stage. This study was conducted in a sick plot at Ghumne 5 of Belaka Municipality, Udayapur, Nepal with six treatments, each replicated four times. The pathogen causing damping off was identified as Rhizoctonia sp. Among the treatments Beejamrut was found to be superior to other treatments in terms of root length (5.97±0.62cm) and shoot length (13.25±1.16cm). In case of root weight and shoot weight Beejamrut (0.28±0.03g;1.85±0.50g), respectively, gave the similar results to Thiram (0.27±0.01g;1.89±0.4g), respectively. Lowest Percentage Disease Incidence (PDI) was found in Thiram (18.0±1) and Beejamrut (19.0±1) treated plots.

Diagnosis and control of the cucumber root rot causes using some chemical and biological agents: تشخيص ومكافحة مسببات مرض تعفن جذور نبات الخيار باستعمال بعض العوامل الكيميائية والأحيائية

This research was aimed to identify the causal agent of the root rot disease of cucumber in some provinces of Iraq, and evaluate its pathogenicity and control it using beneficial bacteria of Serratia odorifera (So) and Enterobacter cloacae (Ec) which isolated from the rhizosphere of healthy cucumber plants, and using silicon (Si) and copper (Cu). The fungus Fusarium solani was predominant, while its percentage of appearance was 80.0% with frequency of 46.2%, Isolate Bfs- 1 was most virulent which prevent germination of the cucumber's seed overall in vitro compared to the control. The bacterial isolates D6 and Q10 exhibited 100% percentage of inhibition against the pathogen in vitro. Results of bacterial isolates identification using Vitiq2 Compact System showed that they belong to S. odorifera and E. cloacae respectively. Under greenhouse conditions the quadrant treatment (So+Ec+Si+Cu) was superior in controlling the disease, while it exhibited 100% percentage of seed germination compared to the negative control (Fungus alone) which achieved 58.0% and the percentage of disease incidence and severity were 9.0%, 3.5% respectively compared to the negative control which were 90.0%, 66.5% respectively. And the quadrant treatment exhibited significant increase of plant growth criteria represented by the dry width of plant, while it achieved 2.69 gm/plant compared to negative and positive control which were 0.85, 2.16 gm/plant respectively.

Endophytic Trichoderma Species Isolated from Persea americana and Cinnamomum verum Roots Reduce Symptoms Caused by Phytophthora cinnamomi in Avocado.

Avocado root rot caused by the oomycete Phytophthora cinnamomi is a severe disease that affects avocado production in Mexico and worldwide. The use of biological control agents such as Trichoderma species isolated from places where the disease is always present, represents an efficient alternative to reduce losses. Thus, the objective of this research was to evaluate the biocontrol ability of 10 endophytic Trichoderma spp. strains against P. cinnamomi tested both in vitro and in the greenhouse. The endophytic Trichoderma spp. were recovered from Persea americana and Cinnamomum verum roots, isolated and purified on potato–dextrose–agar medium. Ten strains were identified by phylogenetic reconstruction of the internal transcribed spacer region of rDNA sequences as T. asperellum (T-AS1, T-AS2, T-AS6, and T-AS7), T. harzianum (T-H3, T-H4, and T-H5), T. hamatum (T-A12), T. koningiopsis (T-K8 and T-K11), and P. cinnamomi (CPO-PCU). In vitro dual-culture assay, the percentage of inhibition of radial growth (PIRG) between Trichoderma spp. and P. cinnamomi strains was measured according to the Bell’s scale. PIRG results indicated that T-AS2 reached the highest value of 78.32%, and T-H5 reached the lowest value of 38.66%. In the greenhouse, the infection was evaluated according to the percentage of disease incidence. Plants with the lowest incidence of dead by avocado root rot were those whose seedlings were inoculated with T-AS2 and T-AS7, resulting in only 5% death by root rot caused by P. cinnamomi. The disease incidence of seedlings with wilt symptoms and death decreased more than 50% in the presence of Trichoderma spp. Relying on the results, we conclude that T. asperellum and T. harzianum contribute to the biocontrol of soil-borne pathogenic oomycete P. cinnamomi.

Screening for identification of resistant sources of little millet varieties against banded blight disease incited by Rhizoctonia solani Kuhn

A field experiment was conducted during kharif, 2018 at the Agricultural Research Station, Vizianagaram, Acharya N. G. Ranga Agricultural University, to identify the resistant sources for Rhizoctonia solani which causes banded blight disease in little millet. Among them none of the variety could exhibit the immune reaction, in which one variety is found to be resistant and three varieties are found to be moderately resistant whereas RLM 223 (check) recorded as highly susceptible to banded blight. The percent disease incidence of Banded blight (Sheath blight) ranged from 4.4% (IIMRLM-8437-17) to 92.0% (JK 8 and TNPSu 202) where it was 95.3% in susceptible check RLM 223.

Grain discoloration in popular rice varieties (Oryza sativa L) in eastern India, associated mycoflora, quality losses and management using selected biocontrol agents

Production and productivity of rice is reduced due to the emerging disease grain discoloration. The disease is a complex symptom due to the association of a number of pathogenic and saprophytic fungi that infect the rice grains both in field and storage conditions. These pathogens not only reduce yield and quality, but also produce harmful mycotoxins which are detrimental to human and animal life. Looking into the severity of the disease, present investigation was undertaken to assess the disease loss among the popular rice varieties growing in eastern India, associated mycoflora and their effect on growth and quality parameters and disease management using selected biocontrol agents. The results indicated that, a significant loss in yield and quality of rice is observed in popular rice varieties (P < 0.0001). Maximum per cent disease incidence (PDI) was recorded in Swarna Sub-1 in two consecutive seasons (average PDI: 39.42%). Different types of discoloration were also noticed and significant reduction were observed in terms of test weight, germination percent, seedling vigor and activities of seed quality enzymes (P < 0.0001). Seed biopriming was opted for management of the disease in variety Nua Sughand Dhan-3, (P < 0.0001) and results indicated that, significant increase in germination (90.67%), seedling length (30.68 cm), seedling vigor (2781.96) and final crop establishment (74.09%) was recorded in treatment-4 (Bacillus subtilis-MH257586) that enhanced plant growth promotional activities. The same treatment also recorded increased defense enzyme activities such as catalase (7.42 Unit activity/Min/g of FW), peroxidase (3.12 δ absorbance/min/mg protein), super oxide dismutase (9.23 AU g FM min−1) and α amylase (8.34 units g-1 FW) when compared to untreated control. Our results clearly indicated that, grain discoloration can be efficiently managed using native bio-control agents which are well known for plant growth promotion and induced systemic resistance (ISR) activities.

Utilization of chemical fungicides in managing the wilt disease of chickpea caused by Fusarium oxysporum f. sp. ciceri

Seven fungicides (Captan, Thiram, Bavistin, Chlorothalonil, Mancozeb, Thiophenate methyl and Propiconazole) at eight different concentrations were evaluated for their effectiveness in managing Fusarium oxysporum f.sp. ciceri (FOC) responsible to cause wilt in chickpea. Among the selected eight fungicides, the best four fungicides (Bavistin, Captan, Thiram and Thiophenate methyl) were evaluated with three different doses as seed treatment and soil application against FOC under pot conditions for their efficacy in reducing wilt severity and promoting plant growth. Later, top three fungicides (Bavistin, Captan and Thiram) were further tested in field condition. Among the assessed fungicides, Bavistin gave the best results in restricting the pathogen’s colony growth in lab and in pots and field trials. Its seed treatment was more efficient in enhancing pods and nodule count per plant and yield of chickpea besides, improving chlorophyll and leghemoglobin contents by reducing the soil population of FOC and percent disease incidence and severity.

Ginger (Zingiber officinale Rosc.) is a type of rhizome plant that has high potential to be developed in Indonesia as a medicinal plant or spice. Inhibiting factors in ginger production is Ralstonia solanacearum which causes bacterial wilt disease. Rhizobacteria is a potential biological agent to control the disease. It is necessary to review from various study on rhizobacteria both the use of antagonistic bacteria singly or in a consortia. Aimed to analyze various sources of scientific literature regarding the potential of antagonistic bacteria singly or consortia compared to synthetic bactericides in controlling of R. solanacearum. The results showed that three recommended antagonist bacteria were found, namely Pseudomonas fluorescens, Bacillus subtilis, and Bacillus thuringiensis. which has the potential to control bacterial wilt disease. The rhizobacterial consortia are more effective as a sustainable, safe and environmentally friendly control on bacterial wilt disease with a percentage of disease incidence 10%. In addition, the rhizobacterial consortia are three and six times more effective than the single antagonistic bacteria and bactericides respectively.

Biological control of mustard blight caused by Alternaria brassicae using plant growth promoting bacteria

Indian mustard (Brassica juncea) is an important oilseed crop in India. Among various diseases infecting mustard crop, Alternaria leaf spot also known as Alternaria blight is incited by fungus Alternaria brassicae, which majorly affects the crop production leading to a yield loss of up to 70 %. The disease is controlled by application of different fungicides prior to winter rains but it is costly and also cause pollution in the environment. This investigation was designed to explore the potential of rhizosphere bacteria to use as bio control agents for suppression of the disease. Total of sixteen bacterial isolates were isolated from the rhizosphere of mustard, tomato and brinjal and they were screened for their antagonistic activity against A. brassicae in vitro and in vivo conditions. Out of sixteen isolates, two bacterial isolates (B3 and B12) showed maximum inhibition (35 % and 38.6 % respectively) against the pathogen and also displayed high degree of secondary metabolite formation. These two bacterial isolates molecularly characterized as Pseudomonas fluorescens were further tested in green house and field conditions through seed bacterization treatment for disease suppression. Mustard seeds were treated with the bacterial solution overnight and were sown in pots and fields the next day. It was found that the disease incidence percentage of B3 and B12 was 32 % and 35 % respectively compared to control with 62.8 %. The results of the above study were highly significant in evaluation of PGPR against A. brassicae.
