The Janus kinase/signal transducers and activators of transcription (JAK/STAT) pathway contributes to the defense against bacterial infection in the pea aphid

The Janus kinase/signal transducers and activators of transcription (JAK/STAT) signaling is activated by infections of bacteria, fungi, viruses and parasites and mediated cellular and humoral immune responses. In the pea aphid Acyrthosiphon pisum little is known about the function of JAK/STAT signaling in its immune system. In this study, we first showed that expression of genes in the JAK/STAT signaling, including the receptors Domeless1/2, Janus kinase (JAK) and transcriptional factor Stat92E, is up-regulated upon bacteria Escherichia coli and Staphylococcus aureus and fungus Beauveria bassiana infections. After knockdown of expression of these genes by means of dsRNA injection, the aphids harbored more bacteria and suffered more death after infected with E. coli and S. aureus, but showed no significant change after B. bassiana infection. Our study suggests the JAK/STAT signaling contributes to the defense against bacterial infection in the pea aphid.

Evaluating climate change scenarios on global pea aphid habitat suitability using species distribution models

The global threat of invasive alien species (IAS) being introduced into new habitats is concerning, particularly in agricultural crops as invasive insect species are continuing to expand their distribution through anthropogenic activities and climate changes. Pea aphids (Acyrthosiphon pisum Harris) are an economic threat to numerous legume crops as they can reproduce parthenogenetically, damage crops directly, and vector over 30 plant viruses as the insect's distribution continues to spread. There are no existing pea aphid-specific risk maps that identify the habitat suitability of pea aphids at either a regional or global scale. Here, we used Species Distribution Models (SDMs) to evaluate which climatic variables influence pea aphid distribution, identify regions of potential distribution, and analyze the global distribution of pea aphids under current and future climate change scenarios (SSP 126, 245, and 370) by utilizing presence-only SDMs based on Maximum Entropy (MaxEnt). The modeling results indicate suitable conditions are relevant for pea aphid establishment in six out of seven continents, with significant range expansion in western Canada, the United States of America, and across Europe. We identified human influence to be the most prominent predictor in determining the distribution of pea aphids, supporting the fact that invasive species distributions are heavily impacted by human activities.

Transcriptomic basis of sex loss in the pea aphid

BackgroundTransitions from sexual to asexual reproduction are common in eukaryotes, but the underlying mechanisms remain poorly known. The pea aphid—Acyrthosiphon pisum—exhibits reproductive polymorphism, with cyclical parthenogenetic and obligate parthenogenetic lineages, offering an opportunity to decipher the genetic basis of sex loss. Previous work on this species identified a single 840 kb region controlling reproductive polymorphism and carrying 32 genes. With the aim of identifying the gene(s) responsible for sex loss and the resulting consequences on the genetic programs controlling sexual or asexual embryogenesis, we compared the transcriptomic response to photoperiod shortening—the main sex-inducing cue—of a sexual and an obligate asexual lineage of the pea aphid, focusing on heads (where the photoperiodic cue is detected) and embryos (the final target of the cue).ResultsOur analyses revealed that four genes (one expressed in the head, and three in the embryos) of the region responded differently to photoperiod in the two lineages. We also found that the downstream genetic programs expressed during embryonic development of a future sexual female encompass ∼1600 genes, among which miRNAs, piRNAs and histone modification pathways are overrepresented. These genes mainly co-localize in two genomic regions enriched in transposable elements (TEs).ConclusionsOur results suggest that the causal polymorphism(s) in the 840 kb region somehow impair downstream epigenetic and post-transcriptional regulations in obligate asexual lineages, thereby sustaining asexual reproduction.

Genomic insights into Spiroplasma endosymbionts that induce male-killing and protective phenotypes in the pea aphid.

The endosymbiotic bacteria Spiroplasma (Mollicutes) infect diverse plants and arthropods, and some of which induce male killing, where male hosts are killed during development. Male-killing Spiroplasma strains belong to either the phylogenetically distant Citri-Poulsonii or Ixodetis groups. In Drosophila flies, Spiroplasma poulsonii induces male killing via the Spaid toxin. While Spiroplasma ixodetis infects a wide range of insects and arachnids, little is known about the genetic basis of S. ixodetis-induced male killing. Here, we analyzed the genome of S. ixodetis strains in the pea aphid Acyrthosiphon pisum (Aphididae, Hemiptera). Genome sequencing constructed a complete genome of a male-killing strain, sAp269, consisting of a 1.5 Mb circular chromosome and an 80 Kb plasmid. sAp269 encoded putative virulence factors containing either ankyrin repeat, ovarian tumor-like deubiquitinase, or ribosome inactivating protein domains, but lacked the Spaid toxin. Further comparative genomics of Spiroplasma strains in A. pisum biotypes adapted to different host plants revealed their phylogenetic associations and the diversity of putative virulence factors. Although the mechanisms of S. ixodetis-induced male killing in pea aphids remain elusive, this study underlines the dynamic genome evolution of S. ixodetis and proposes independent acquisition events of male-killing mechanisms in insects.

Transgenerational plasticity in aphids reared in a poor-resource environment

The changing environmental conditions can affect insect biology over multiple generations and phenotypic plasticity is important for coping with these changes. Transgenerational plasticity occurs when the environment in which the parents developed influences the plastic response of the offspring phenotype. In the present study, the plastic effects of resource limitation on important life history traits such as body size, fecundity, survival, and resistance to starvation of the pea aphid Acyrthosiphon pisum were investigated over two generations. This study focused on understanding how resource limitation can determine an adaptive expression of maternal effects and transgenerational plasticity in fitness-related traits. Aphids showed phenotypic plasticity for the life history traits investigated, as they performed better when grown in an optimal environment than in a resource-poor one. Also, aphids had a poorer performance if their mothers were raised in a resource-poor environment. The effects of transgenerational plasticity were observed only in response to resistance to starvation, through increased survival in the offspring of the mother reared in a resource-poor environment, suggesting an evolutionary bet-hedging strategy. The results of this study showed that the effects of adaptive transgenerational plasticity may be partially masked in stressful environments, where developmental problems instead predominate. More information on the transgenerational response to resource limitation across generations can contribute to a better understanding of aphid biology.

Experimental evidence reveals that vector host preference and performance across host plants is not altered by vector-borne plant viruses

Many plant pathogens are transmitted by generalist insects that move between host species, and vector preference and performance on different hosts affects pathogen transmission. Most studies of host–vector–pathogen interactions focus on a single host, however, and it is relatively unknown if viruses affect the host breadth of vectors. For example, although pea aphids (Acyrthosiphon pisum) often form tight associations with specific hosts, viruses transmitted by aphids can break down host defenses, potentially weakening these associations and increasing the host breadth of their vectors. Here, we assessed vector host preference and performance in an experiment with five pea aphid colonies with distinct host associations, five host species, and two viruses (Pea enation mosaic virus [PEMV] and Bean leafroll virus [BLRV]). Our prediction was that pea aphids would have higher performance on their inferior hosts and be less discriminating among hosts when those are infected, compared with noninfected hosts. Our results show that aphid performance, measured as the rate of increase on hosts, differed among hosts based on their host association, and relative performance among hosts was indeed altered by virus infection. Aphid preferences, measured using a choice bioassay in which aphids could settle on any of the five host species, also differed among aphid colonies but was not affected by virus infection status. We also did not find that infection status of hosts altered the performance of aphids on their preferred host. Furthermore, virus infection did not disrupt preference performance relationships for the aphids. Overall, our study suggests that host preference and performance are deeply ingrained in pea aphids, and that host associations may be robust to the presence of host-manipulating plant pathogens.

Population Dynamics of Insect Pests of Pea

The seasonal incidence of major insect pests on pea Pisum sativum at different growth stages has been assessed in this study. Major pests observed include gram pod borer Helicoverpa armigera (Hubner), blue butterfly Lampedes boeticus (L.), pea leaf miner Chromatomyia horticola Goureau and pea aphid Acyrthosiphon pisum (Harris). These results revealed that peak incidence of H. armigera (7.80 larvae/ plant) and L. boeticus (8.60 larvae/ plant) was during the 8th and 9th SMW, respectively; while for C. horticola (5.50 larvae/ leaf) and A. pisum (94.1 aphids/ plant) it was 5th SMW. Helicoverpa armigera incidence revealed a significant negative correlation with minimum temperature (r= -0.628) while for L. boeticus it was positive one with maximum temperature (r= 0.604). The incidence of C. horticola was observed to be significantly negatively correlated with minimum temperature and rainfall (r= -0.826 and r= -0.584, respectively); while A. pisum revealed a significant negative correlation with minimum temperature (r= -0.806).

Use of double-stranded RNA targeting β2 divergent nicotinic acetylcholine receptor subunit to control pea aphid Acyrthosiphon pisum at larval and adult stages.

In recent years, the use of the RNA interference technology (RNAi) has emerged as one of the new strategies for species-specific control of insect pests. Its specificity depends on the distinctiveness of the target gene sequence for a given species. In this work, we assessed in the pea aphid Acyrthosiphon pisum (A. pisum) the use of a double-stranded RNA (dsRNA) that targets the β2 divergent nicotinic acetylcholine receptor (nAChR) subunit (dsRNA-β2), which shares low sequence identity with other subunits, to control populations of this pest at different developmental stages. Because nAChRs are targeted by neonicotinoid insecticides such as imidacloprid, we also assessed the effect of dsRNA-β2 coupled to this insecticide on aphid survival. Finally, because the effect of a control agent on beneficial insect must be considered before any use of new pest management strategies, the acute toxicity of dsRNA-β2 combined with imidacloprid was evaluated on honeybee Apis mellifera. In this work, we demonstrated that dsRNA-β2 alone has an insecticidal effect on aphid larvae and adults. Moreover, dsRNA-β2 and imidacloprid effects on aphid larvae and adults were additive, meaning that dsRNA-β2 did not alter the efficacy of imidacloprid on these two developmental stages. Also, no obvious acute toxicity on Apis mellifera was reported. Using RNAi that targets β2 divergent nAChR subunit is effective alone or combined with imidacloprid to control A. pisum at larval and adult stages. Because no obvious Apis mellifera mortality has been reported, this RNAi-based pest management strategy should be considered to control insect pest. © 2023 The Authors. Exposure to heat stress leads to striking clone-specific nymph deformity in pea aphid.

Climatic changes, such as heatwaves, pose unprecedented challenges for insects, as escalated temperatures above the thermal optimum alter insect reproductive strategies and energy metabolism. While thermal stress responses have been reported in different insect species, thermo-induced developmental abnormalities in phloem-feeding pests are largely unknown. In this laboratory study, we raised two groups of first instar nymphs belonging to two clones of the pea aphid Acyrthosiphon pisum, on fava beans Vicia faba. The instars developed and then asexually reproduced under constant exposure to a sub-lethal heatwave (27°C) for 14 days. Most mothers survived but their progenies showed abnormalities, as stillbirths and appendageless or weak nymphs with folded appendages were delivered. Clone N116 produced more deceased and appendageless embryos, contrary to N127, which produced fewer dead and more malformed premature embryos. Interestingly, the expression of the HSP70 and HSP83 genes differed in mothers between the clones. Moreover, noticeable changes in metabolism, e.g., lipids, were also detected and that differed in response to stress. Deformed offspring production after heat exposure may be due to heat injury and differential HSP gene expression, but may also be indicative of a conflict between maternal and offspring fitness. Reproductive altruism might have occurred to ensure some of the genetically identical daughters survive. This is because maintaining homeostasis and complete embryogenesis could not be simultaneously fulfilled due to the high costs of stress. Our findings shine new light on pea aphid responses to heatwaves and merit further examination across different lineages and species.

Mobile elements create strain-level variation in the services conferred by an aphid symbiont.

Heritable, facultative symbionts are common in arthropods, often functioning in host defence. Despite moderately reduced genomes, facultative symbionts retain evolutionary potential through mobile genetic elements (MGEs). MGEs form the primary basis of strain-level variation in genome content and architecture, and often correlate with variability in symbiont-mediated phenotypes. In pea aphids (Acyrthosiphon pisum), strain-level variation in the type of toxin-encoding bacteriophages (APSEs) carried by the bacterium Hamiltonella defensa correlates with strength of defence against parasitoids. However, co-inheritance creates difficulties for partitioning their relative contributions to aphid defence. Here we identified isolates of H. defensa that were nearly identical except for APSE type. When holding H. defensa genotype constant, protection levels corresponded to APSE virulence module type. Results further indicated that APSEs move repeatedly within some H. defensa clades providing a mechanism for rapid evolution in anti-parasitoid defences. Strain variation in H. defensa also correlates with the presence of a second symbiont Fukatsuia symbiotica. Predictions that nutritional interactions structured this coinfection were not supported by comparative genomics, but bacteriocin-containing plasmids unique to co-infecting strains may contribute to their common pairing. In conclusion, strain diversity, and joint capacities for horizontal transfer of MGEs and symbionts, are emergent players in the rapid evolution of arthropods.

Contrasting Evolutionary Patterns Between Sexual and Asexual Lineages in a Genomic Region Linked to Reproductive Mode Variation in the pea aphid.

Although asexual lineages evolved from sexual lineages in many different taxa, the genetics of sex loss remains poorly understood. We addressed this issue in the pea aphid Acyrthosiphon pisum, whose natural populations encompass lineages performing cyclical parthenogenesis (CP) and producing one sexual generation per year, as well as obligate parthenogenetic (OP) lineages that can no longer produce sexual females but can still produce males. An SNP-based, whole-genome scan of CP and OP populations sequenced in pools (103 individuals from 6 populations) revealed that an X-linked region is associated with the variation in reproductive mode. This 840-kb region is highly divergent between CP and OP populations (FST = 34.9%), with >2,000 SNPs or short Indels showing a high degree of association with the phenotypic trait. In OP populations specifically, this region also shows reduced diversity and Tajima's D, consistent with the OP phenotype being a derived trait in aphids. Interestingly, the low genetic differentiation between CP and OP populations at the rest of the genome (FST = 2.5%) suggests gene flow between them. Males from OP lineages thus likely transmit their op allele to new genomic backgrounds. These genetic exchanges, combined with the selection of the OP and CP reproductive modes under different climates, probably contribute to the long-term persistence of the cp and op alleles.

A snapshot of the transcriptome of Medicago truncatula (Fabales: Fabaceae) shoots and roots in response to an arbuscular mycorrhizal fungus and the pea aphid (Acyrthosiphon pisum) (Hemiptera: Aphididae).

Plants simultaneously interact with belowground symbionts such as arbuscular mycorrhizal (AM) fungi and aboveground antagonists such as aphids. Generally, plants gain access to valuable resources including nutrients and water through the AM symbiosis and are more resistant to pests. Nevertheless, aphids' performance improves on mycorrhizal plants, and it remains unclear whether a more nutritious food source and/or attenuated defenses are the contributing factors. This study examined the shoot and root transcriptome of barrel medic (Medicago truncatula Gaertn.) plants highly colonized by the AM fungus Rhizophagus irregularis (Blaszk., Wubet, Renker, and Buscot) C. Walker and A. Schüßler (Glomerales: Glomeraceae) and exposed to 7 days of mixed age pea aphid (Acyrthosiphon pisum (Harris)) herbivory. The RNA-seq samples chosen for this study showed that aphids were heavier when fed mycorrhizal plants compared to nonmycorrhizal plants. We hypothesized that (i) insect-related plant defense pathways will be downregulated in shoots of mycorrhizal plants with aphids compared to nonmycorrhizal plants with aphids; (ii) pathways involved in nutrient acquisition, carbohydrate-related and amino acid transport will be upregulated in shoots of mycorrhizal plants with aphids compared to nonmycorrhizal plants with aphids; and (iii) roots of mycorrhizal plants with aphids will exhibit mycorrhiza-induced resistance. The transcriptome data revealed that the gene repertoire related to defenses, nutrient transport, and carbohydrates differs between nonmycorrhizal and mycorrhizal plants with aphids, which could explain the weight gain in aphids. We also identified novel candidate genes that are differentially expressed in nonmycorrhizal plants with aphids, thus setting the stage for future functional studies.

The Influence of Lead and Acyrthosiphon pisum (Harris) on Generation of Pisum sativum Defense Signaling Molecules and Expression of Genes Involved in Their Biosynthesis.

The main aim of this study was to understand the regulation of the biosynthesis of phytohormones as signaling molecules in the defense mechanisms of pea seedlings during the application of abiotic and biotic stress factors. It was important to identify this regulation at the molecular level in Pisum sativum L. seedlings under the influence of various concentrations of lead-i.e., a low concentration increasing plant metabolism, causing a hormetic effect, and a high dose causing a sublethal effect-and during feeding of a phytophagous insect with a piercing-sucking mouthpart-i.e., pea aphid (Acyrthosiphon pisum (Harris)). The aim of the study was to determine the expression level of genes encoding enzymes of the biosynthesis of signaling molecules such as phytohormones-i.e., jasmonates (JA/MeJA), ethylene (ET) and abscisic acid (ABA). Real-time qPCR was applied to analyze the expression of genes encoding enzymes involved in the regulation of the biosynthesis of JA/MeJA (lipoxygenase 1 (LOX1), lipoxygenase 2 (LOX2), 12-oxophytodienoate reductase 1 (OPR1) and jasmonic acid-amido synthetase (JAR1)), ET (1-aminocyclopropane-1-carboxylate synthase 3 (ACS3)) and ABA (9-cis-epoxycarotenoid dioxygenase (NCED) and aldehyde oxidase 1 (AO1)). In response to the abovementioned stress factors-i.e., abiotic and biotic stressors acting independently or simultaneously-the expression of the LOX1, LOX2, OPR1, JAR1, ACS3, NCED and AO1 genes at both sublethal and hormetic doses increased. Particularly high levels of the relative expression of the tested genes in pea seedlings growing at sublethal doses of lead and colonized by A. pisum compared to the control were noticeable. A hormetic dose of lead induced high expression levels of the JAR1, OPR1 and ACS3 genes, especially in leaves. Moreover, an increase in the concentration of phytohormones such as jasmonates (JA and MeJA) and aminococyclopropane-1-carboxylic acid (ACC)-ethylene (ET) precursor was observed. The results of this study indicate that the response of pea seedlings to lead and A. pisum aphid infestation differed greatly at both the gene expression and metabolic levels. The intensity of these defense responses depended on the organ, the metal dose and direct contact of the stress factor with the organ.

Effects of different host plants on population fitness of pea aphid (Acyrthosiphon pisum)

BackgroundHost plants not only provide and living places and energy materials for insects, but also influence insect population parameters and population fitness. MethodsThis study examined the influence of various host plant species on the fitness of pea aphid (Acyrthosiphon pisum). The biological parameters and population parameters of pea aphid on 6 different host plants (Vicia fabae, Pisum sativum, Medicago sativa, Trifolium pratense, Onobrychis viciaefolia and Melilotus officinalis) were observed and counted by ecological experiments, which were carried out in a control chamber. ResultsThe results showed that the developmental duration of 1st and 2nd instar nymphs of pea aphids on T. pratense and P. sativum was significantly prolonged, whereas that of 3rd and 4th instar nymphs on O. viciaefolia and M. officinalis was significantly shortened. Compared with the pea aphid on the V. faba, the longevity of adults on M. officinalis and P. sativum was significantly prolonged, but only the generation time on P. sativum was significantly prolonged. Moreover, the survival rate of nymphs was significantly lower on O. viciaefolia and M. sativa than on others. Net reproductive rate and mean generation time on V. faba were significantly higher than in other host plants. The intrinsic rate of increase (rm) and finite rate of increase (λ) of pea aphid feeding of A. pisum on P. sativum and O. viciaefolia decreased. However, those on the double population time on P. sativum and O. viciaefolia were significantly higher than the others. ConclusionThe findings will clarify the population fitness of pea aphids on different hosts and guide the rational distribution of different host plants, and provide new references for aphid control strategies.

Identifying environmental factors affecting the production of pea aphid dispersal morphs in field populations

Abstract Many species of insects show phenotypically plastic dispersal traits in response to suboptimal environmental conditions. These polyphenetic traits can have large impacts on the population dynamics of the dispersing insects. Therefore, it is important to understand what environmental factors affect the development of dispersal traits and how these factors affect population dynamics via immigration/emigration. Pea aphids (Acyrthosiphon pisum) exhibit a polyphenetic wing dimorphism in which individuals are either winged (alate) or not (apterous). The production of alate offspring is determined by environmental cues experienced by the mother. Environmental factors associated with these cues have been intensively studied, with crowding, host plant quality, natural enemies, fungal infection, and viral infection being shown to affect the production of pea aphid alates under controlled laboratory conditions. Nonetheless, environmental factors affecting alate production have rarely been studied in field populations of pea aphids. Using data from a three‐year study of alate production in 5–9 alfalfa (lucerne) fields, we examined the effects of (i) pea aphid abundance, (ii) host plant maturity, (iii) temperature, (iv) predator and herbivore abundance, (v) parasitism, and (vi) entomopathic fungal incidence. In twice‐weekly samples taken over the growing season, alate production ranged from 0% to 83% of the nymphal population. Pea aphid abundance, temperature, and alfalfa maturity together explained 67% of the variation in alate production. The other factors we investigated explained little variation. These results suggest there is a limited number of key environmental factors that consistently predict changes in alate production in field populations, while many factors identified in lab studies may be unimportant. Our results highlight the value of investigating factors affecting the expression of plastic traits in insect species at a broad spatiotemporal scale and under natural conditions.

A diversity of endosymbionts across Australian aphids and their persistence in aphid cultures.

There is increasing interest in the use of endosymbionts in pest control, which will benefit from the identification of endosymbionts from potential donor species for transfer to pest species. Here, we screened for endosymbionts in 123 Australian aphid samples across 32 species using 16S DNA metabarcoding. We then developed a qPCR method to validate the metabarcoding data set and to monitor endosymbiont persistence in aphid cultures. Pea aphids (Acyrthosiphon pisum) were frequently coinfected with Rickettsiella and Serratia, and glasshouse potato aphids (Aulacorthum solani) were coinfected with Regiella and Spiroplasma; other secondary endosymbionts detected in samples occurred by themselves. Hamiltonella, Rickettsia and Wolbachia were restricted to a single aphid species, whereas Regiella was found in multiple species. Rickettsiella, Hamiltonella and Serratia were stably maintained in laboratory cultures, although others were lost rapidly. The overall incidence of secondary endosymbionts in Australian samples tended to be lower than recorded from aphids overseas. These results indicate that aphid endosymbionts probably exhibit different levels of infectivity and vertical transmission efficiency across hosts, which may contribute to natural infection patterns. The rapid loss of some endosymbionts in cultures raises questions about factors that maintain them under field conditions, while endosymbionts that persisted in laboratory culture provide candidates for interspecific transfers.

Aphid facultative symbionts confer no protection against the fungal entomopathogen Batkoa apiculata.

Fungi in the family Entomophthoraceae are prevalent pathogens of aphids. Facultative symbiotic bacteria harbored by aphids, including Spiroplasma sp. and Regiella insecticola, have been shown to make their hosts more resistant to infection with the fungal pathogen Pandora neoaphidis. How far this protection extends against other species of fungi in the family Entomophthoraceae is unknown. Here we isolated a strain of the fungal pathogen Batkoa apiculata infecting a natural population of pea aphids (Acyrthosiphon pisum) and confirmed its identity by sequencing the 28S rRNA gene. We then infected a panel of aphids each harboring a different species or strain of endosymbiotic bacteria to test whether aphid symbionts protect against B. apiculata. We found no evidence of symbiont-mediated protection against this pathogen, and our data suggest that some symbionts make aphids more susceptible to infection. This finding is relevant to our understanding of this important model of host-microbe interactions, and we discuss our results in the context of aphid-microbe ecological and evolutionary dynamics.

Pea Aphid (Acyrthosiphon pisum) Host Races Reduce Heat-Induced Forisome Dispersion in Vicia faba and Trifolium pratense.

Although phloem-feeding insects such as aphids can cause significant damage to plants, relatively little is known about early plant defenses against these insects. As a first line of defense, legumes can stop the phloem mass flow through a conformational change in phloem proteins known as forisomes in response to Ca2+ influx. However, specialized phloem-feeding insects might be able to suppress the conformational change of forisomes and thereby prevent sieve element occlusion. To investigate this possibility, we triggered forisome dispersion through application of a local heat stimulus to the leaf tips of pea (Pisum sativum), clover (Trifolium pratense) and broad bean (Vicia faba) plants infested with different pea aphid (Acyrthosiphon pisum) host races and monitored forisome responses. Pea aphids were able to suppress forisome dispersion, but this depended on the infesting aphid host race, the plant species, and the age of the plant. Differences in the ability of aphids to suppress forisome dispersion may be explained by differences in the composition and quantity of the aphid saliva injected into the plant. Various mechanisms of how pea aphids might suppress forisome dispersion are discussed.

Evolutionary consequences of loss of sexual reproduction on male-related traits in parthenogenetic lineages of the pea aphid.

Transition from sexual reproduction to parthenogenesis constitutes a major life-history change with deep evolutionary consequences for sex-related traits, which are expected to decay. The pea aphid Acyrthosiphon pisum shows intraspecific reproductive polymorphism, with cold-resistant cyclically parthenogenetic (CP) lineages that alternate sexual and asexual generations and cold-sensitive obligately parthenogenetic (OP) lineages that produce only asexual females but still males. Here, the genotyping of 219 pea aphid lineages collected in cold-winter and mild-winter regions revealed contrasting population structures. Samples from cold-winter regions consisted mostly of distinct multilocus genotypes (MLGs) usually represented by a single sample (101 different MLGs for 111 samples) and were all phenotyped as CP. In contrast, fewer MLGs were found in mild-winter regions (28 MLGs for 108 samples), all but one being OP. Since the males produced by OP lineages are unlikely to pass on their genes (sexual females being rare in mild-winter regions), we tested the hypothesis that their traits could degenerate due to lack of selection by comparing male production and male reproductive success between OP and CP lineages. Male production was indeed reduced in OP lineages, but a less clear pattern was observed for male reproductive success: females mated with OP males laid fewer eggs (fertilized or not) but OP and CP males fertilized the same proportion of eggs. These differences may stem from the type of selective forces: male production may be counter-selected whereas male performances may evolve under the slower process of relaxed selection. The overall effective reproductive capacity of OP males could result from recent sex loss in OP lineages or underestimated reproductive opportunities.

Dopamine mediates the pea aphid wing plasticity.

Many organisms exhibit phenotypic plasticity, in which developmental processes result in different phenotypes depending on their environmental context. Here we focus on the molecular mechanisms underlying that environmental response. Pea aphids (Acyrthosiphon pisum) exhibit a wing dimorphism, in which pea aphid mothers produce winged or wingless daughters when exposed to a crowded or low-density environment, respectively. We investigated the role of dopamine in mediating this wing plasticity, motivated by a previous study that found higher dopamine titres in wingless- versus winged-producing aphid mothers. In this study, we found that manipulating dopamine levels in aphid mothers affected the numbers of winged offspring they produced. Specifically, asexual female adults injected with a dopamine agonist produced a lower percentage of winged offspring, while asexual females injected with a dopamine antagonist produced a higher percentage of winged offspring, matching expectations based on the titre difference. We also found that genes involved in dopamine synthesis, degradation and signalling were not differentially expressed between wingless- and winged-producing aphids. This result indicates that titre regulation possibly happens in a non-transcriptional manner or that sampling of additional timepoints or tissues is necessary. Overall, our work emphasizes that dopamine is an important component of how organisms process information about their environments.
