Hydrogen Peroxide Mediates Pharmacological Ascorbate Induced Radio-Sensitization of Pancreatic Cancer Cells by Enhancing G2-accumulation and Reducing Cyclin B1 Protein Levels.

Pharmacological ascorbate (P-AscH-, high dose, intravenous vitamin C) preferentially sensitizes human pancreas ductal adenocarcinoma (PDAC) cells to radiation-induced toxicity compared to non-tumorigenic epithelial cells. Radiation-induced G2-checkpoint activation contributes to the resistance of cancer cells to DNA damage induced toxicity. We hypothesized that P-AscH- induced radio-sensitization of PDAC cells is mediated by perturbations in the radiation induced activation of the G2-checkpoint pathway. Both non-tumorigenic pancreatic ductal epithelial and PDAC cells display decreased clonogenic survival and increased doubling times after radiation treatment. In contrast, the addition of P-AscH- to radiation increases clonogenic survival and decreases the doubling time of non-tumorigenic epithelial cells but decreasing clonogenic survival and increasing the doubling time of PDAC cells. Results from the mitotic index and propidium iodide assays showed that while the P-AscH- treatments did not affect radiation-induced G2-checkpoint activation, it enhanced G2-accumulation. The addition of catalase reverses the increases in G2-accumulation, indicating a peroxide-mediated mechanism. In addition, P-AscH- treatment of PDAC cells suppresses radiation-induced accumulation of cyclin B1 protein levels. Both translational and post-translational pathways appear to regulate cyclin B1 protein levels after the combination treatment of PDAC cells with P-AscH- and radiation. The protein changes seen are reversed by the addition of catalase suggesting that hydrogen peroxide mediates P-AscH- induced radiation sensitization of PDAC cells by enhancing G2-accumulation and reducing cyclin B1 protein levels.

Biogenic fabrication of nanomaterials from flower-based chemical compounds, characterization and their various applications: A review.

Nanotechnology is evolving as a significant discipline of research with various applications. It includes the materials and their applications having one dimension in the range of 1–100 nm. Many chemical and physical protocol have been utilized for the nanoparticles (NPs) fabrication. These protocols are costly, hazardous and consumes high energy. Thus, researchers are inclined towards biological synthesis of NPs using plant and or herbal extract as these methods are simple, sustainable, ecofriendly and cost-effective. Flower is an important part of plants, and contained several phytochemicals such as flavonoids, terpenoids, coumarins, sterol and xanthones which acts as an important precursor for NPs synthesis. These compounds acted as reducing as well as stablishing agent during fabrication processes. They have been thoroughly characterized by various techniques. The fabricated NPs have shown potential antimicrobial activity against bacterial and fungal infections. They have been also used as potential therapeutic agent for human breast cancer, gastric adenocarcinoma cell, colorectal adenocarcinoma cell and pancreas ductal adenocarcinoma cells. Overall, the aim of this review article to facilitates the recent understanding of flower-mediated NPs fabrication (a sustainable and ecofriendly resource), their application in different disciplines and challenges.

Diallyl disulphide-loaded spherical gold nanoparticles and acorn-like silver nanoparticles synthesised using onion extract: catalytic activity and cytotoxicity

Onion (Allium cepa) extract was used for the green synthesis of gold and silver nanoparticles. Each colloidal solution exhibited surface plasmon resonance, with a peak at 532 nm for gold nanoparticles and 391 nm for silver nanoparticles. Microscopic results confirmed the presence of spherical shapes. The X-ray diffraction pattern demonstrated a face-centered cubic structure. Both nanoparticles had negative zeta potentials and retained colloidal stability in cell culture medium. Catalytic applications were evaluated for 4-nitrophenol reduction and methyl orange degradation reactions by monitoring with UV-visible spectrophotometry. Furthermore, the nanoparticles demonstrated no significant cytotoxicity against human pancreas ductal adenocarcinoma cells (PANC-1) and human colorectal adenocarcinoma cells (HT-29). PEGylation and diallyl disulphide loading of the gold and silver nanoparticles meaningfully reduced the cell viability of both cell lines. Furthermore, diallyl disulphide loading resulted in more cytotoxicity against PANC-1 cells than against HT-29 cells. Additionally, the gold nanoparticles were more cytotoxic than the silver nanoparticles upon diallyl disulphide loading. Interestingly, after PEGylation and diallyl disulphide loading, the silver nanoparticles exhibited acorn-like shapes, while the gold nanoparticles retained spherical shapes. This result suggested that nanoparticles green-synthesised by onion extract have possibilities as nanocatalysts and drug delivery nanocarriers for catalytic and nanomedicine applications.

Forkhead box B2 inhibits the malignant characteristics of the pancreatic cancer cell line Panc-1 in vitro.

Forkhead box (FOX) proteins constitute a family of transcription factors that are evolutionarily conserved in various species ranging from yeast to humans. These proteins have functions during development as well as in adulthood. To date, many reports have described the functions of FOX family genes in cancer cells, but the role of FOXB2 is not well understood. In one of the pancreas ductal adenocarcinoma cell lines, Panc-1 cells, we showed here that FOXB2 expression is barely detectable and that CpG islands in the 5' regions of the FOXB2 are highly methylated. These findings led us to hypothesize that FOXB2 acts as a tumor suppressor. To clarify our hypotheses, we investigated the effects of FOXB2 over-expression in Panc-1 cells. We obtained FOXB2 stable transfectants, and these clones exhibited reduced spheroid formation ability. Expression of β-catenin, which is reported to be over-expressed in various cancer cells, was highly suppressed in FOXB2 stable transfectants. Moreover, side population (SP) cell fractions, which have a high tumorigenesis and metastatic potential, as well as anchorage-independent growth ability, were reduced. These results suggest that FOXB2 has the ability to inhibit the malignant characteristics of Panc-1 in vitro.

Differentially Expressed microRNAs in MIA PaCa-2 and PANC-1 Pancreas Ductal Adenocarcinoma Cell Lines are Involved in Cancer Stem Cell Regulation.

Pancreatic ductal adenocarcinoma (PDAC) is one of the most lethal malignancies, and thus better understanding of its molecular pathology is crucial for us to devise more effective treatment of this deadly disease. As cancer cell line remains a convenient starting point for discovery and proof-of-concept studies, here we report the miRNA expression characteristics of two cell lines, MIA PaCa-2 and PANC-1, and discovered three miRNAs (miR-7-5p, let-7d, and miR-135b-5p) that are involved in cancer stem cells (CSCs) suppression. After transfection of each miRNA’s mimic into PANC-1 cells which exhibits higher stemness feature than MIA-PaCa-2 cells, partial reduction of CSC surface markers and inhibition of tumor sphere formation were observed. These results enlighten us to consider miRNAs as potential therapeutic agents for pancreatic cancer patients via specific and effective inhibition of CSCs.

Sesquiterpenoids from Tussilago farfara Flower Bud Extract for the Eco-Friendly Synthesis of Silver and Gold Nanoparticles Possessing Antibacterial and Anticancer Activities.

Sesquiterpenoids from the flower bud extract of Tussilago farfara were effectively utilized as a reducing agent for eco-friendly synthesis of silver and gold nanoparticles. The silver and gold nanoparticles had a characteristic surface plasmon resonance at 416 nm and 538 nm, respectively. Microscopic images revealed that both nanoparticles were spherical, and their size was measured to be 13.57 ± 3.26 nm for the silver nanoparticles and 18.20 ± 4.11 nm for the gold nanoparticles. The crystal structure was determined to be face-centered cubic by X-ray diffraction. Colloidal stability of the nanoparticle solution was retained in a full medium, which was used in the cell culture experiment. The antibacterial activity result demonstrated that the silver nanoparticles showed better activity (two- to four-fold enhancement) than the extract alone on both Gram-positive and Gram-negative bacteria. Interestingly, the highest antibacterial activity was obtained against vancomycin-resistant Enterococci Van-A type Enterococcus faecium. Cytotoxicity on cancer cell lines confirmed that gold nanoparticles were more cytotoxic than silver nanoparticles. The highest cytotoxicity was observed on human pancreas ductal adenocarcinoma cells. Therefore, both nanoparticles synthesized with the sesquiterpenoids from T. farfara flower bud extract can be applicable as drug delivery vehicles of anticancer or antibacterial agents for future nanomedicine applications.

SNHG14 enhances gemcitabine resistance by sponging miR-101 to stimulate cell autophagy in pancreatic cancer

BackgroundDue to the poor prognosis and high mortality (over 90%), Pancreas ductal adenocarcinoma (PDAC) is listed as the 7th leading cause of cancer-related death in the world, while gemcitabine sensitivity is key important in PDAC therapy. SNHG14 is thought to be an oncogene in cancer progression. However, the possible role of SNHG14 underlying the progress of the PDAC cell, specifically in gemcitabine resistance remains to be determined. MethodsWe analyzed the PDAC-related data collected from TCGA. PDAC cell line (SW1990) was used as in vitro model. RT-qPCR and western blot were used to detect the autophagy-related gene expression level. MTT and flow cytometry approaches were used to determine cell viability and apoptosis rate. The luciferase reporter assay was used to confirm the direct interaction between SNHG14 and miR-101. The wound healing assay and transwell assay were used to detect the migration and invasion abilities of PDAC cells. ResultsThe expression of SNHG14 was significantly higher in the PDAC tissues than in the normal tissues, while miR-101 was significantly downregulated in the PDAC tissues. Moreover, the correlation analysis showed that SNHG14 was negatively correlated with miR-101. The in vitro experiments furthermore confirmed their impacts on PDAC cells. Overexpression of SNHG14 and miR-101 inhibitor significantly enhanced cell proliferation, migration, and invasion rate of PDAC cell line. Moreover, SNHG14 knockdown and miR-101 mimics both led to attenuation of gemcitabine resistance-PDAC cell viability and promoted cell apoptosis rate, as well as the reduction of autophagy-related proteins (such as RAB5A and ATG4D). Overexpression of SNHG14 enhanced PDAC cell progression and inhibited cell apoptosis in gemcitabine treatment, as well as the increase of autophagy-related proteins, thus enhanced the chemoresistance of PDAC cells to gemcitabine. ConclusionsCollectively, we first time revealed that SNHG14 could sponge miR-101 to enhance PDAC cell progression and find the specific axis of SNHG14/miR-101/autophagy underlying the chemoresistance in PDAC cells to gemcitabine, which could promote the progress of PDAC therapy.

The impact of acute inflammation on progression and metastasis in pancreatic cancer animal model

BackgroundThe impact of acute inflammation on cancer is unclear. Therefore, we evaluated the impact of acute inflammation on cancer progression in an animal model concerning morphological change as well as molecular features. Material and methodsMurine pancreas ductal adenocarcinoma cell line (Panc-02) and zymosan were used for the induction of cancer and acute inflammation respectively, in C57/BL6 mice. In the C group (n = 10), 2 × 107Panc02 cells were injected into the tail of the pancreas. In the Z1 (n = 10) and Z2 (n = 10), 3 mg of zymosan was injected intraperitoneally once and twice respectively, after tumor cell injection. All of the mice were sacrificed 4 weeks after tumor cell injection. ResultsThe degree of inflammation was more severe in the Z2 group than in the other two groups. The tumor volume of the Z2 group was larger than that of the C group (P = 0.032) and the presence of liver metastasis was significantly more common in the Z2 group than in the C and Z1 groups (P = 0.025). Confocal microscopy analysis revealed significantly more expression of Snail, EpCAM, Muc1, NLRP3 and miR-155 in the liver and pancreas in the Z2 group than in the other two groups. EpCAM and Muc1 were detected in blood samples in the Z2 group by flow cytometry, but not in the other two groups. In the Z2, expression of E-Cadherin was weaker and vimentin, snail1 were stronger compared to the C and Z1 group in the PCR and western blot. ConclusionPresent results suggest that acute inflammation accompanied with cancer promotes cancer progression by the epithelial-to-mesenchymal transition (EMT) and circulating tumor cells (CTC) process.

Clinically approved PEGylated nanoparticles are covered by a protein corona that boosts the uptake by cancer cells.

Today, liposomes are an advanced technology of drug carriers with a dozen drugs in clinical practice and many more in clinical trials. A bottleneck associated with the clinical translation of liposomes has long been 'opsonization', i.e. the adsorption of plasma proteins at the liposome surface resulting in their rapid clearance from circulation. For decades, the most popular way to avoid opsonization has been grafting polyethylene glycol (PEG) onto the liposome surface. Recent studies have clarified that grafting PEG onto the liposome surface reduces, but does not completely prevent protein binding. In this work, we employed dynamic light scattering, zeta-potential analysis, one-dimensional sodium dodecyl sulfate polyacrylamide gel electrophoresis (1D-SDS-PAGE), semi-quantitative densitometry and cell imaging to explore the bio-nano-interactions between human plasma (HP) and Onivyde, a PEGylated liposomal drug that has recently been approved by the Food and Drug Administration (FDA) for the treatment of metastatic pancreatic ductal adenocarcinoma (PDAC). To properly evaluate the role of PEGylation, an unPEGylated variant of Onivyde was used as a reference. Collectively, our findings suggest that: (i) although PEGylated, Onivyde is not "stealth" in HP; (ii) surface chemistry is more important than PEGylation in controlling the bio-nano-interactions between Onivyde and plasma components. Of note is that the PC was found to boost the cellular uptake of Onivyde in the pancreas ductal adenocarcinoma cell line (PANC-1) thus suggesting its prominent role in its indication for PDAC treatment. Relevant implications for drug delivery and drug design are discussed.

MIA PaCa-2 and PANC-1 - pancreas ductal adenocarcinoma cell lines with neuroendocrine differentiation and somatostatin receptors.

Studies using cell lines should always characterize these cells to ensure that the results are not distorted by unexpected morphological or genetic changes possibly due to culture time or passage number. Thus, the aim of this study was to describe those MIA PaCa-2 and PANC-1 cell line phenotype and genotype characteristics that may play a crucial role in pancreatic cancer therapeutic assays, namely neuroendocrine chemotherapy and peptide receptor radionuclide therapy. Epithelial, mesenchymal, endocrine and stem cell marker characterization was performed by immunohistochemistry and flow cytometry, and genotyping by PCR, gene sequencing and capillary electrophoresis. MIA PaCa-2 (polymorphism) expresses CK5.6, AE1/AE3, E-cadherin, vimentin, chromogranin A, synaptophysin, SSTR2 and NTR1 but not CD56. PANC-1 (pleomorphism) expresses CK5.6, MNF-116, vimentin, chromogranin A, CD56 and SSTR2 but not E-cadherin, synaptophysin or NTR1. MIA PaCA-1 is CD24−, CD44+/++, CD326−/+ and CD133/1−, while PANC-1 is CD24−/+, CD44+, CD326−/+ and CD133/1−. Both cell lines have KRAS and TP53 mutations and homozygous deletions including the first 3 exons of CDKN2A/p16INK4A, but no SMAD4/DPC4 mutations or microsatellite instability. Both have neuroendocrine differentiation and SSTR2 receptors, precisely the features making them suitable for the therapies we propose to assay in future studies.

Genomic alterations link Rho family of GTPases to the highly invasive phenotype of pancreas cancer.

Pancreas ductal adenocarcinoma (PDAC) is a highly lethal cancer that typically presents as advanced, unresectable disease. This invasive tendency, coupled with intrinsic resistance to standard therapies and genome instability, are major contributors to poor long-term survival. The genetic elements governing the invasive propensity of PDAC have not been well elucidated. Here, in the course of validating resident genes in highly recurrent and focal amplifications in PDAC, we have identified Rio Kinase 3 (RIOK3) as an amplified gene that alters cytoskeletal architecture as well as promotes pancreatic ductal cell migration and invasion. We determined that RIOK3 promotes its invasive activities through activation of the small G protein, Rac. This genomic and functional link to Rac signaling prompted a genome wide survey of other components of the Rho family network, revealing p21 Activated Kinase 4 (PAK4) as another amplified gene in PDAC tumors and cell lines. Like RIOK3, PAK4 promotes pancreas ductal cell motility and invasion. Together, the genomic and functional profiles establish the Rho family GTP-binding proteins as integral to the hallmark invasive nature of this lethal disease.
