Is Biological Aging a Treatable Disease? A Consideration Based on Proposed Mechanisms of Aging

In view of the current claim by many researchers that biological aging is a treatable disease, the possibility is discussed whether the claim is realistic, based on several proposed mechanisms of aging. The definition of biological aging is stated referring to physiological aging and pathological aging, since biological aging must be defined for the discussion of whether it can be cured. Aging in animal model is compared with that in humans in terms of common age-associated phenotypes. Major proposed mechanisms of aging are next examined including Genome Instability Theory of aging, Free Radical or Oxidative Stress Theory of Aging, Mitochondrial Theory of Aging, Error Catastrophe Theory of Aging/Translational Error Theory of Aging, Altered Protein Theory of Aging/Proteostasis Theory of Aging, and Epigenetic Theory of Aging. Finally, we discuss whether treatment of aging as a disease is realistic in comparison with possible lifespan extension by retardation of biological aging.

The Evolution and Ecology of Oxidative and Antioxidant Status: A Comparative Approach in African Mole-Rats.

The naked mole-rat of the family Bathyergidae has been the showpiece for ageing research as they contradict the traditional understanding of the oxidative stress theory of ageing. Some other bathyergids also possess increased lifespans, but there has been a remarkable lack of comparison between species within the family Bathyergidae. This study set out to investigate how plasma oxidative markers (total oxidant status (TOS), total antioxidant capacity (TAC), and the oxidative stress index (OSI)) differ between five species and three subspecies of bathyergids, differing in their maximum lifespan potential (MLSP), resting metabolic rate, aridity index (AI), and sociality. We also investigated how oxidative markers may differ between captive and wild-caught mole-rats. Our results reveal that increased TOS, TAC, and OSI are associated with increased MLSP. This pattern is more prevalent in the social-living species than the solitary-living species. We also found that oxidative variables decreased with an increasing AI and that wild-caught individuals typically have higher antioxidants. We speculate that the correlation between higher oxidative markers and MLSP is due to the hypoxia-tolerance of the mole-rats investigated. Hormesis (the biphasic response to oxidative stress promoting protection) is a likely mechanism behind the increased oxidative markers observed and promotes longevity in some members of the Bathyergidae family.

Oxidation of biological molecules with age and induced oxidative stress in different growth phases of Saccharomyces cerevisiae.

One of the mechanistic approaches for explaining ageing is the oxidative stress theory of ageing. Saccharomyces cerevisiae has been used as a model to study ageing due to many factors. We have attempted to investigate if the differential ability to withstand oxidative stress can be correlated with their lifespans. In all the four strains studied (AP22, 699, 8C, and SP4), there was no age-associated increases in lipid peroxidation levels measured as thiobarbituric acid reactive substances (TBARS). Under induced oxidative stress conditions, there was an increased TBARS level in both the ages assessed with a quantum-fold increase in the stationary phase cells of AP22.In contrast, the late stationary phase cells of 8C exhibited the least susceptibility to induced oxidative stress. The level of TBARS in both exponential and late stationary phase cells of 699 was overall more than that in the other three strains. Protein carbonylation increased with age in 8C and 699. Induced stress increased carbonylation in the exponential cells of SP4 and 699 and the stationary phase cells of all four strains. Protein carbonylation data indicate that the AP22 cells exhibit decreased protein carbonylation vis-à-vis the other strains. Induced stress data showed that while the exponential cells of 699 are susceptible, the late stationary phase cells of 699 are most resistant. Western blotting analysis using anti-HNE antibodies showed two proteins of molecular mass ~ 56 and ~ 84kDa that were selectively modified with age in all the strains. Under induced stress conditions, an additional protein of ~ 69kDa was oxidized. Our investigation shows that the difference in lifespan between the four strains of S. cerevisiae may be regulated by oxidative stress. Knowledge of the identity of the oxidized proteins will significantly facilitate a better understanding of the effect of oxidative stress conditions on the cells of S. cerevisiae.

Investigating the Role of Salicylic Acid in Determining the Lifespan of Short-Lived and Long-Lived Flax Species and Lines

Introduction: Reactive oxygen species (ROS) accumulation has been related to aging in both plant and human physiology. Leaked electrons from the electron transport system (ETS) react with molecular oxygen producing ROS, such as hydrogen peroxide (H2O2) and superoxide anions. There are numerous antioxidative mechanisms present in plants that aid in redox balance, one of which is the mild uncoupling of mitochondria. Uncoupling agents, such as salicylic acid (SA), can abolish the connection between oxidation and phosphorylation allowing electron transport to take place without the need for parallel ATP synthesis. “Uncoupling to Survive” Hypothesis states that uncoupling activity should be greater in long-lived species since uncoupled mitochondria result in reduced ROS production thereby increasing lifespan. Since ROS accumulation has been linked to lifespan in the Oxidative Stress Theory of Aging, the idea that long-lived species should exhibit greater antioxidative capacity than short-lived species is the foundation upon which this hypothesis is established. We tested this hypothesis in various flax species and lines differing in lifespan. Methods: Spectrophotometry was used to determine SA levels in plant tissue while respiration rates were measured using a dissolving oxygen electrode. Results: The “Uncoupling to Survive” Hypothesis does not seem to hold for flax since long-lived and short-lived species did not exhibit significant differences in oxygen consumption rates that would indicate differences in the uncoupling activity of their mitochondria. Discussion: However, early flowering lines exhibited lower levels of SA suggesting that flax plants do not require elevated SA expression to display the early flowering phenotype as seen in Arabidopsis species. In addition, SA significantly increased the oxygen consumption rates in wild-type and early flowering plants which further provides evidence for its role as an uncoupling agent. Conclusion: Our findings add to our understanding of the many roles that SA plays in plant physiology. Advantages of SA are well established in the cosmetic and medical communities and investigating its effects on plants can be beneficial. We also looked at the "Uncoupling to Survive Hypothesis" in plants, which helped us get a better grasp of the antioxidant mechanisms that may eventually reduce oxidative stress and lead to senescence.

A Comprehensive Overview of the Complex Role of Oxidative Stress in Aging, The Contributing Environmental Stressors and Emerging Antioxidant Therapeutic Interventions.

IntroductionAging is a normal, inevitable, irreversible, and progressive process which is driven by internal and external factors. Oxidative stress, that is the imbalance between prooxidant and antioxidant molecules favoring the first, plays a key role in the pathophysiology of aging and comprises one of the molecular mechanisms underlying age-related diseases. However, the oxidative stress theory of aging has not been successfully proven in all animal models studying lifespan, meaning that altering oxidative stress/antioxidant defense systems did not always lead to a prolonged lifespan, as expected. On the other hand, animal models of age-related pathological phenotypes showed a well-correlated relationship with the levels of prooxidant molecules. Therefore, it seems that oxidative stress plays a more complicated role than the one once believed and this role might be affected by the environment of each organism. Environmental factors such as UV radiation, air pollution, and an unbalanced diet, have also been implicated in the pathophysiology of aging and seem to initiate this process more rapidly and even at younger ages.AimThe purpose of this review is to elucidate the role of oxidative stress in the physiology of aging and the effect of certain environmental factors in initiating and sustaining this process. Understanding the pathophysiology of aging will contribute to the development of strategies to postpone this phenomenon. In addition, recent studies investigating ways to alter the antioxidant defense mechanisms in order to prevent aging will be presented.ConclusionsCareful exposure to harmful environmental factors and the use of antioxidant supplements could potentially affect the biological processes driving aging and slow down the development of age-related diseases. Maybe a prolonged lifespan could not be achieved by this strategy alone, but a longer healthspan could also be a favorable target.

Urinary Biomarkers of Oxidative Stress in Aging: Implications for Prediction of Accelerated Biological Age in Prospective Cohort Studies.

Background Aging is a major risk factor for a range of chronic diseases. Oxidative stress theory of aging has been previously proposed as one of the mechanisms responsible for the age-related decline in organ/tissue function and the development of age-related diseases. Urine contains rich biological information on the health status of every major organ system and can be an important noninvasive source for biomarkers of systemic oxidative stress in aging. Aims The objective of this cross-sectional study was to validate a novel panel of urinary oxidative stress biomarkers. Methods Nucleic acid oxidation adducts and oxidative damage markers of lipids and proteins were assessed in urine samples from nondiabetic and currently nonsmoking subjects (n = 198) across different ages (20 to 89 years old). Urinary parameters and chronological age were correlated then the biological age of enrolled individuals was determined from the urinary oxidative stress markers using the algorithm of Klemera and Doubal. Results Our findings showed that 8-oxo-7,8-deoxyguanosine (8-oxoG), 8-oxo-7,8-dihydroguanosine (8-OHdG), and dityrosine (DTyr) positively correlated with chronological age, while the level of an F2-isoprostane (iPF2α-VI) correlated negatively with age. We found that 8-oxoG, DTyr, and iPF2α-VI were significantly higher among accelerated agers compared to nonaccelerated agers and that a decision tree model could successfully identify accelerated agers with an accuracy of >92%. Discussion. Our results indicate that 8-oxoG and iPF2α-VI levels in the urine reveal biological aging. Conclusion Assessing urinary biomarkers of oxidative stress may be an important approach for the evaluation of biological age by identifying individuals at accelerated risk for the development of age-related diseases.

Supercomplex Organization of the Electron Transfer System in Marine Bivalves, a Model of Extreme Longevity

The mitochondrial oxidative stress theory of aging suggests that the organelle's decay contributes to the aging phenotype via exacerbated oxidative stress, loss of organ coordination and energetics, cellular integrity, and activity of the mitochondrial electron transfer system (ETS). Recent advances in understanding the structure of the ETS show that the enzymatic complexes responsible for oxidative phosphorylation are arranged in supramolecular structures called supercomplexes that lose organization during aging. Their exact role and universality among organisms are still under debate. Here, we take advantage of marine bivalves as an aging model to compare the structure of the ETS among species ranging from 28 to 507 years in maximal life span. Our results show that regardless of life span, the bivalve ETS is arrayed as a set of supercomplexes. However, bivalve species display varying degrees of ETS supramolecular organization with the highest supercomplex structures found in Arctica islandica, the longest-lived of the bivalve species under study. We discuss this comparative model in light of differences in the nature and stoichiometry of these complexes and highlight the potential link between the complexity of these superstructures and longer life spans.

The Role of Oxidative Stress in Cardiovascular Aging and Cardiovascular Diseases

Aging can be seen as process characterized by accumulation of oxidative stress induced damage. Oxidative stress derives from different endogenous and exogenous processes, all of which ultimately lead to progressive loss in tissue and organ structure and functions. The oxidative stress theory of aging expresses itself in age-related diseases. Aging is in fact a primary risk factor for many diseases and in particular for cardiovascular diseases and its derived morbidity and mortality. Here we highlight the role of oxidative stress in age-related cardiovascular aging and diseases. We take into consideration the molecular mechanisms, the structural and functional alterations, and the diseases accompanied to the cardiovascular aging process.

Divergences in the Control of Mitochondrial Respiration Are Associated With Life-Span Variation in Marine Bivalves.

The role played by mitochondrial function in the aging process has been a subject of intense debate in the past few decades, as part of the efforts to understand the mechanistic basis of longevity. The mitochondrial oxidative stress theory of aging suggests that a progressive decay of this organelle's function leads to an exacerbation of oxidative stress, with a deleterious impact on mitochondrial structure and DNA, ultimately promoting aging. Among the traits suspected to be associated with longevity is the variation in the regulation of oxidative phosphorylation, potentially affecting the management of oxidative stress. Longitudinal studies using the framework of metabolic control analysis have shown age-related differences in the flux control of respiration, but this approach has seldom been taken on a comparative scale. Using 4 species of marine bivalves exhibiting a large range of maximum life span (from 28 years to 507 years), we report life-span-related differences in flux control at different steps of the electron transfer system. Increased longevity was characterized by a lower control by NADH (complex I-linked) and Succinate (complex II-linked) pathways, while respiration was strongly controlled by complex IV when compared to shorter-lived species. Complex III exerted strong control over respiration in all species. Furthermore, high longevity was associated with higher citrate synthase activity and lower ATP synthase activity. Relieving the control exerted by the electron entry pathways could be advantageous for reaching higher longevity, leading to increased control by complex IV, the final electron acceptor in the electron transfer system.

Oxidative stress in aging: stayin’ alive?

There is still ongoing controversy about the oxidative stress theory of aging, particularly in mammals, after a significant number of studies have been conducted to test this theory. Results generated from the studies strongly indicate that accumulation of oxidative damage alone does not play a significant role as an underlying mechanism of aging, which calls into question that significant modifications to the theory are required to understand the relationship between oxidative stress and aging. To examine the exact role of oxidative stress in aging and age-related diseases, our laboratory has been conducting studies with unique animal models: 1) mice overexpressing or down-regulating thioredoxin (Trx) in the cytosol (Trx1) or in mitochondria (Trx2); and 2) rats overexpressing Cu/Zn superoxide dismutase (SOD). Results generated from these studies strongly indicate that: 1) changes in oxidative stress and redox state could play more important roles in age-related pathological changes, e.g., cancer and metabolic disorders; 2) redox regulation of signaling pathways could play more important roles in aging than accumulation of oxidative damages; 3) the potential benefit of changes in oxidative stress and redox state could be organ/tissue specific; 4) the roles of oxidative stress could vary in different stages of life (i.e., young versus old); and 5) synergetic effects of changes in oxidative stress in multiple cellular compartments may be required to have a significant impact on aging. Therefore, the studies with more careful approach would uncover the exact roles and pathophysiological consequences of oxidative stress during aging. Lipophilic Antioxidants and Cognitive Function in the Elderly

Objective:To determine the relationship between blood serum lipophilic antioxidant levels and cognitive function (CF) in older adults aged 60 and above guided by the oxidative stress theory of aging.Methods:Cross-sectional data from the National Health and Nutrition Examination Survey (n = 291) for older adults aged 60 and above were examined using Pearson correlation coefficient and multiple linear regression to determine whether blood serum antioxidant status predicted CF while controlling for age, sex, race, hypertension, smoking status, and body mass index.Results:Alpha-tocopherol, retinyl palmitate, trans-lycopene, and retinyl stearate were all significantly correlated with CF. After controlling for covariates, α-tocopherol and retinyl palmitate were associated with CF. Age, sex, and current smoking status were significant predictors of CF.Conclusions:The benefits of antioxidants in CF may be a part of nutritional recommendations which include α-tocopherol and retinyl palmitate for delay of CI, and subsequently a better quality of life.

Why Naked Mole-Rats Have High Oxidative Damage but Live a Long Life: A Simple Explanation Based on the Oxidative Stress Theory of Aging

Compared to mice with the same body mass, naked mole-rats have a lower metabolic rate, higher homeostasis-maintaining activities, higher oxidative damage levels, but a longer lifespan. These observations have raised serious challenges to the widely accepted oxidative stress theory of aging, which suggests a negative correlation between damage levels and lifespan. Here, we introduce a simple theoretical model based on energy conservation and the oxidative stress theory. Employing the model and the physiological parameters of mice and naked mole-rats, we explain why naked mole-rats have higher damage levels despite their higher somatic maintenance efforts; why damage levels in naked mole-rats seem not to change over age; and how these factors concertedly result in a longer lifespan in naked mole-rats. Our results highlight the energy tradeoff between biosynthesis and somatic maintenance, and suggest that the rate of damage accumulation over age and the existence of a threshold of damage for death are the keys to solve the paradox raised by naked mole-rats.

Mitochondrial Traits Previously Associated With Species Maximum Lifespan Do Not Correlate With Longevity Across Populations of the Bivalve Arctica islandica.

The mitochondrial oxidative stress theory of aging posits that membrane susceptibility to peroxidation and the organization of the electron transport system (ETS) linked with reactive oxygen species (ROS) generation are two main drivers of lifespan. While a clear correlation has been established from species comparative studies, the significance of these characteristics as potential modulators of lifespan divergences among populations of individual species is still to be tested. The bivalve Arctica islandica, the longest-lived non-colonial animal with a record lifespan of 507 years, possesses a lower mitochondrial peroxidation index (PI) and reduced H2O2 efflux linked to complexes I and III activities than related species. Taking advantage of the wide variation in maximum reported longevities (MRL) among 6 European populations (36–507 years), we examined whether these two mitochondrial properties could explain differences in longevity. We report no relationship between membrane PI and MRL in populations of A. islandica, as well as a lack of intraspecific relationship between ETS complex activities and MRL. Individuals from brackish sites characterized by wide temperature and salinity windows had, however, markedly lower ETS enzyme activities relative to citrate synthase activity. Our results highlight environment-dependent remodeling of mitochondrial phenotypes.

The exceptional longevity of the naked mole-rat may be explained by mitochondrial antioxidant defenses.

Naked mole‐rats (NMRs) are mouse‐sized mammals that exhibit an exceptionally long lifespan (>30 vs. <4 years for mice), and resist aging‐related pathologies such as cardiovascular and pulmonary diseases, cancer, and neurodegeneration. However, the mechanisms underlying this exceptional longevity and disease resistance remain poorly understood. The oxidative stress theory of aging posits that (a) senescence results from the accumulation of oxidative damage inflicted by reactive oxygen species (ROS) of mitochondrial origin, and (b) mitochondria of long‐lived species produce less ROS than do mitochondria of short‐lived species. However, comparative studies over the past 28 years have produced equivocal results supporting this latter prediction. We hypothesized that, rather than differences in ROS generation, the capacity of mitochondria to consume ROS might distinguish long‐lived species from short‐lived species. To test this hypothesis, we compared mitochondrial production and consumption of hydrogen peroxide (H2O2; as a proxy of overall ROS metabolism) between NMR and mouse skeletal muscle and heart. We found that the two species had comparable rates of mitochondrial H2O2 generation in both tissues; however, the capacity of mitochondria to consume ROS was markedly greater in NMRs. Specifically, maximal observed consumption rates were approximately two and fivefold greater in NMRs than in mice, for skeletal muscle and heart, respectively. Our results indicate that differences in matrix ROS detoxification capacity between species may contribute to their divergence in lifespan.

The Naked Mole Rat: A Unique Example of Positive Oxidative Stress.

The oxidative stress theory of aging, linking reactive oxygen species (ROS) to aging, has been accepted for more than 60 years, and numerous studies have associated ROS with various age-related diseases. A more precise version of the theory specifies that mitochondrial oxidative stress is a direct cause of aging. The naked mole rat, a unique animal with exceptional longevity (32 years in captivity), appears to be an ideal model to study successful aging and the role of ROS in this process. Several studies in the naked mole rat have shown that these animals exhibit a remarkable resistance to oxidative stress. At low concentrations, ROS serve as second messengers, and these important intracellular signalling functions are crucial for the regulation of cellular processes. In this review, we examine the literature on ROS and their functions as signal transducers. We focus specifically on the longest-lived rodent, the naked mole rat, which is a perfect example of the paradox of living an exceptionally long life with slow aging despite high levels of oxidative damage from a young age.

Long-lived birds suffer less from oxidative stress

BackgroundOxidative stress, caused by an imbalance between reactive oxygen species and antioxidants, is thought to be an important intrinsic mechanism for aging. Ecologists have tested this hypothesis in birds, although the evidence supporting the link between oxidative stress and lifespan has so far been ambiguous. Two previous studies based on a wide range of different free-living bird species provided contradictory findings: antioxidants were negatively associated with survival rate in one study, but positively associated with longevity in another.MethodsIn this study, we identified possible shortcomings in previous research, and then used the comparative methods to test whether long-lived birds experience less oxidative stress reflected by four blood redox state markers (total antioxidant status, uric acid, total glutathione, malondialdehyde) based on data for 78 free-living species.ResultsRelatively long-lived bird species had high levels of antioxidants (total antioxidant status, total glutathione) and low levels of reactive oxygen species (malondialdehyde). These associations were independent of statistical control for any effects of body mass, sampling effort and similarity among taxa due to common phylogenetic descent.ConclusionsThe direction of these associations is consistent with the oxidative stress theory of aging.

Longevity and life history coevolve with oxidative stress in birds.

1. The mechanisms that underpin the evolution of ageing and life histories remain elusive. Oxidative stress, which results in accumulated cellular damages, is one of the mechanisms suggested to play a role. 2. In this paper, we set out to test the "oxidative stress theory of ageing" and the "oxidative stress hypothesis of life histories" using a comprehensive phylogenetic comparison based on an unprecedented dataset of oxidative physiology in 88 free-living bird species. 3. We show for the first time that bird species with longer lifespan have higher non-enzymatic antioxidant capacity and suffer less oxidative damage to their lipids. We also found that bird species featuring a faster pace-of-life either have lower non-enzymatic antioxidant capacity or are exposed to higher levels of oxidative damage, while adult annual mortality does not relate to oxidative state. 4. These results reinforce the role of oxidative stress in the evolution of lifespan and also corroborate the role of oxidative state in the evolution of life histories among free-living birds.

Oxidative stress, aging, and diseases.

Reactive oxygen and nitrogen species (RONS) are produced by several endogenous and exogenous processes, and their negative effects are neutralized by antioxidant defenses. Oxidative stress occurs from the imbalance between RONS production and these antioxidant defenses. Aging is a process characterized by the progressive loss of tissue and organ function. The oxidative stress theory of aging is based on the hypothesis that age-associated functional losses are due to the accumulation of RONS-induced damages. At the same time, oxidative stress is involved in several age-related conditions (ie, cardiovascular diseases [CVDs], chronic obstructive pulmonary disease, chronic kidney disease, neurodegenerative diseases, and cancer), including sarcopenia and frailty. Different types of oxidative stress biomarkers have been identified and may provide important information about the efficacy of the treatment, guiding the selection of the most effective drugs/dose regimens for patients and, if particularly relevant from a pathophysiological point of view, acting on a specific therapeutic target. Given the important role of oxidative stress in the pathogenesis of many clinical conditions and aging, antioxidant therapy could positively affect the natural history of several diseases, but further investigation is needed to evaluate the real efficacy of these therapeutic interventions. The purpose of this paper is to provide a review of literature on this complex topic of ever increasing interest.

Idh2 deficiency accelerates renal dysfunction in aged mice

The free radical or oxidative stress theory of aging postulates that senescence is due to an accumulation of cellular oxidative damage, caused largely by reactive oxygen species (ROS) that are produced as by-products of normal metabolic processes in mitochondria. The oxidative stress may arise as a result of either increased ROS production or decreased ability to detoxify ROS. The availability of the mitochondrial NADPH pool is critical for the maintenance of the mitochondrial antioxidant system. The major enzyme responsible for generating mitochondrial NADPH is mitochondrial NADP+-dependent isocitrate dehydrogenase (IDH2). Depletion of IDH2 in mice (idh2-/-) shortens life span and accelerates the degeneration of multiple age-sensitive traits, such as hair grayness, skin pathology, and eye pathology. Among the various internal organs tested in this study, IDH2 depletion-induced acceleration of senescence was uniquely observed in the kidney. Renal function and structure were greatly deteriorated in 24-month-old idh2-/- mice compared with wild-type. In addition, disruption of redox status, which promotes oxidative damage and apoptosis, was more pronounced in idh2-/- mice. These data support a significant role for increased oxidative stress as a result of compromised mitochondrial antioxidant defenses in modulating life span in mice, and thus support the oxidative stress theory of aging.
