Utilization of hydrophobic bacterium Rhodococcus opacus B-4 as whole-cell catalyst in anhydrous organic solvents

Rhodococcus opacus strain B-4, which has recently been isolated as an organic solvent-tolerant bacterium, has a high hydrophobicity and exhibits a high affinity for hydrocarbons. This bacterium was able to survive for at least 5 days in organic solvents, including n-tetradecane, oleyl alcohol, and bis(2-ethylhexyl) phthalate (BEHP), which contained water less than 1% (w/v). The biocatalytic ability of R. opacus B-4 was demonstrated in the essentially nonaqueous BEHP using indigo production from indole as a model conversion. By the catabolism of oleic acid for NADH regeneration, indigo production increased up to 71.6 microg ml(-1) by 24 h.

A Newly Isolated Organic Solvent Tolerant Staphylococcus saprophyticus M36 Produced Organic Solvent-Stable Lipase

Thirty-eight high lipase activity strains were isolated from soil, seawater, and Brassica napus. Among them, a novel organic solvent tolerant bacterium (strain M36) was isolated from the seawater in Jiangsu, China. Isolate M36 was able to grow at high concentration of benzene or toluene up to 40% (vol/vol), and later identified as Staphylococcus saprophyticus by biochemical test and 16s ribosomal DNA sequence. No work on Staphylococcus producing lipase with organic solvent tolerance has been reported so far. The lipase of strain M36 whose activity in liquid medium was 42 U mL(-1) at 24-h incubation time was stable in the presence of 25% (vol/vol) p-xylene, benzene, toluene, and hexane.

Nutritional factors affecting organic solvent-tolerant alkaline protease production by a newBacillus cereus strain 146

An organic solvent-tolerant bacterium designated as 146 capable of producing an organic solvent-stable alkaline protease was isolated from contaminated soil of a wood factory. The strain was a Gram-positive, spore-forming, nitrate-positive, rod-shaped organism capable of hydrolysing gelatine, starch, skim milk and identified asBacillus cereus. Activity of the protease was drastically increased in the presence of 1–decanol, isooctane, n-dodecane and n-tetradecane, but reduced in the presence of ethyl acetate, benzene, toluene, 1-heptanol, ethylbenzene and hexane. The bacterium was shown to require lactose as a carbon source and peptone as a nitrogen source. The optimum fermentation condition for the production of alkaline protease was in the presence of beef and yeast extract. Optimum pH was determined to be at 10.0 at incubation temperature of 37 °C for 48 h. Results from the studies suggest that 146 is a new strain of Bacillus cereus capable of producing organic solvent-tolerant alkaline protease with potential use in industries.

Structural elucidation of the core-lipid A backbone from the lipopolysaccharide of Acinetobacter radioresistens S13, an organic solvent tolerant Gram-negative bacterium

The structure of the core oligosaccharide of the lipopolysaccharide from an organic solvent tolerant Gram-negative bacterium, Acinetobacter radioresistens S13, was investigated by chemical analysis, NMR spectroscopy and MALDI-TOF mass spectrometry. All the experiments were performed on the oligosaccharides obtained either by alkaline degradation or mild acid hydrolysis. The data showed the presence of two novel oligosaccharide molecules containing a trisaccharide of 3-deoxy- d- manno-octulopyranosonic acid in the inner core region and a glucose rich outer core whose structure is the following: ▪ R = H in the main oligosaccharide and β-Glc in the minor product. The bacterium was grown on aromatic (phenol and benzoic acid) and nonaromatic carbon sources and the core oligosaccharide resulted to occur always with this novel structure.

Isolation and characterization of novel organic solvent-tolerant bacteria

Some organic solvents are extremely toxic to living organisms by virtue of their ability to partition into and disrupt the normal functioning of biological membranes. In recent years, several bacteria have been discovered that are more tolerant to these toxic solvents than most microorganisms. Using enrichment procedures, we have isolated new organic solvent-tolerant bacteria from both hyrdocarbon-contaminated and pristine soil samples. These organisms were characterized by several different experimental procedures including description of their cellular physiology, 16S rDNA homology, organic solvent tolerance range, and survival after solvent exposure. The results indicate that gram-positive bacteria can be isolated from the environment that are as tolerant to toxic organic solvents, if not more so, than the most organic solvent-tolerant gram-negative bacteria.

An organic solvent-tolerant protease from Pseudomonas aeruginosa strain K : Nutritional factors affecting protease production

An organic solvent-tolerant bacterium producing an organic solvent-stable protease was isolated from soil and identified as Pseudomonas aeruginosa strain K. Nutritional requirements for optimized protease production by this strain were investigated. Maximum protease activity was achieved with sorbitol as the sole carbon source, followed by starch and lactose at pH 7.0 and 37 °C. Dextrose, sucrose and glycerol greatly reduced the protease production. The best organic nitrogen source was casamino acid. Tryptone, soytone and yeast extract supported protease production while corn steep liquor and beef extract inhibited the protease activity. Significant protease production was observed with sodium nitrate as a sole nitrogen source however, ammonium nitrate completely inhibit it. More than 62% drop in production occurred in the presence of amino acids. Addition of metal ions such as K +, Mg 2+ and Ca 2+ maximized the enzyme production.

Industrial potential of organic solvent tolerant bacteria.

Most bacteria and their enzymes are destroyed or inactivated in the presence of organic solvents. Organic solvent tolerant bacteria are a relatively novel group of extremophilic microorganisms that combat these destructive effects and thrive in the presence of high concentrations of organic solvents as a result of various adaptations. These bacteria are being explored for their potential in industrial and environmental biotechnology, since their enzymes retain activity in the presence of toxic solvents. This property could be exploited to carry out bioremediation and biocatalysis in the presence of an organic phase. Because a large number of substrates used in industrial chemistry, such as steroids, are water-insoluble, their bioconversion rates are affected by poor dissolution in water. This problem can be overcome by carrying out the process in a biphasic organic-aqueous fermentation system, wherein the substrate is dissolved in the organic phase and provided to cells present in the aqueous phase. In bioprocessing of fine chemicals such as cis-diols and epoxides using such cultures, organic solvents can be used to extract a toxic product from the aqueous phase, thereby improving the efficiency of the process. Bacterial strains reported to grow on and utilize saturated concentrations of organic solvents such as toluene can revolutionize the removal of such pollutants. It is now known that enzymes display striking new properties in the presence of organic solvents. The role of solvent-stable enzymes in nonaqueous biocatalysis needs to be explored and could result in novel applications.

Isolation of an organic-solvent-tolerant cholesterol-transforming Bacillus species, BC1, from coastal sediment.

Steroid transformation is of great importance in the pharmaceutical industry. The major limiting factor in this process is the extremely poor solubility of steroids in aqueous media, which lowers their transformation rate and increases costs. This problem can be overcome by using organic-solvent-tolerant bacteria (OSTB), which can carry out the desired bioconversions in an organic-solvent-saturated system. OSTB are a relatively novel group of extremophilic microbes that have developed various adaptations to withstand solvent toxicity. The aim of this study was to isolate marine bacteria producing organic-solvent-stable cholesterol-transforming enzymes. A Bacillus species, BC1, isolated from Arabian Sea sediment was found to degrade cholesterol and exhibit excellent solvent tolerance particularly to chloroform. OSTB have tremendous potential in industrial processes involving nonaqueous biocatalysis and transformation in the presence of an organic phase.

Stability studies of protease from Bacillus cereus BG1

An organic solvent-tolerant bacterium producing organic solvent-stable protease was isolated from fishing industry wastewater and identified as B. cereus. The enzyme retained more than 95% of its initial activity after pre-incubation 24 h at 30 °C in the presence of 25% methanol, DMSO, acetonitrile and DMF. Above 37 °C, Ca 2+ was required for enzyme activity. The optimum temperature for the protease activity was at 60 °C in the presence of 2 mM Ca 2+ and 50 °C in the absence of Ca 2+. At 60 °C, Ca 2+ (2 mM) stimulated the protease activity by 500%. Other bivalent metal ions such as Mg 2+ and Mn 2+ also increased activity by 285 and 157%, respectively, while Zn 2+ and Cu 2+ had a strong inhibitory effect. Thermostability of the enzyme was enhanced by Ca 2+ at temperature values above 40 °C. In the presence of 10 mM Ca 2+, the enzyme retained 100, 93 and 26% of its initial activity after heating for 15 min at 55, 60 and 70 °C, respectively. However, the enzyme was completely inactivated when incubated at 55 °C for 15 min in the absence of calcium. The pH optimum was 8.0 and the enzyme was quite stable in various pH buffers between pH 6.0 and 9.0 when incubated at 50 °C for 1 and 3 h. Enzyme activity was inhibited by EDTA, suggesting that the preparation contains metalloprotease(s).

Simple and rapid cell growth assay using tetrazolium violet coloring method for screening of organic solvent tolerant bacteria

Tolerance to organic solvents is an important characteristic for selecting bacteria that are useful for producing commodity chemicals. An assay method for tolerance should be easy to perform and differences in tolerance should be reproducible. We propose a cell growth assay using tetrazolium violet and image analysis for assessing tolerance. When five microorganisms including Escherichia coli were tested, the organic solvent tolerance (OST) of microorganisms used was rapidly and qualitatively assessed and the degrees of OST coincided with those from the conventional method using solvent overlaid-solid medium. Moreover, the proposed method was applicable to the estimation of OST for cyclobexane resistant E. coli OST3410 using various organic solvent mixtures.

Lipase Production in Two-Step Fed-Batch Culture of Organic Solvent-Tolerant Pseudomonas aeruginosa LST-03.

Efficient lipase production by two-step fed-batch culture of an organic solvent-tolerant bacterium, Pseudomonas aeruginosa LST-03, was investigated. When FB synthetic medium was used in flask culture, no lipase activity was detected, whereas lipase was produced at 2.3 I.U./ml in C2 complex medium. However, lipase production was induced in FB medium when a fatty acid was added to the culture broth in the stationary phase. Among fatty acids tested, long chain saturated fatty acids, such as C18 (stearic acid) and C20 (arachidic acid), were found to function as effective inducers for the production of lipase, giving an activity level almost the same as that obtained in C2 medium in flask culture. Two-step lipase production, comprised of a growth phase in fed-batch mode and a production phase in which lipase was induced by the addition of 5% (v/v) stearic acid, was carried out in a jar-fermentor. In the growth phase, the maximum cell concentration at 16 h was only 20 in terms of the optical density at 660 nm (OD660), and a low level of lipase production (8 I.U./ml) was obtained after 167 h. This was considered to be due to the exhaustion of several medium components brought about by the use of an unsuitable medium or feeding solution. After analyzing the contents of the compounds in the culture broth by inductively coupled plasma spectrometry for metal ions and HPLC for anions, a modified FB medium was designed. When this modified FB medium was used in two-step fed-batch culture, the maximum cell concentration reached an OD660 of 55 (30.2 g-dry cells/l) at 16.5 h, and lipase was produced at 96 I.U./ml after 35 h, which is approximately 40 times higher than the production level obtained in flask culture using C2 medium.

Organic solvent-tolerant bacterium which secretes an organic solvent-stable proteolytic enzyme

A bacterial strain which can be grown in a medium containing organic solvents and can secrete a proteolytic enzyme was isolated and identified as Pseudomonas aeruginosa. The strain was derived by the following two-step procedures: high proteolytic enzyme producers were first isolated by the usual method, and then the organic solvent-tolerant microorganism was selected from these high-rate proteolytic enzyme producers. The proteolytic activity of the supernatant of the culture was stable in the presence of various organic solvents. The stability of the enzyme in the presence of organic solvents, of which the values of the logarithm of the partition coefficient (log P) were equal to or more than 3.2, was almost the same as that in the absence of organic solvents. It is expected that both the solvent-tolerant microorganism and the solvent-stable enzyme produced by this strain can be used as catalysts for reactions in the presence of organic solvents.

Degradation of polyaromatic hydrocarbons by organic solvent-tolerant bacteria from deep sea.

We isolated three organic solvent (OS)-tolerant bacterial strains DS-1051, DS-1902, and DS-313 from a depth of 1,168 m in Sagami Bay, Japan. These isolates were tolerant to various kinds of toxic OSs such as benzene, toluene, and p-xylene. They also could degrade polyaromatic hydrocarbons, naphthalene or biphenyl, in a medium-OS (9:1) two-liquid-phase system. Percentage degradation of polyaromatic hydrocarbons in OS by these strains were higher than those obtained from cultures in which substrates were in the medium without OS.
