Optimization of Bacterial Cellulose Production by Komagataeibacter rhaeticus K23

The use of bacterial cellulose (BC), having high purity, a high degree of crystallinity, water-holding capacity, tensile strength and adaptability on a broad scale is limited because of the low yield. In this study, the optimal conditions for bio-cellulose production by Komagataeibacter rhaeticus K23 were investigated. Optimal values for temperature, pH, inoculum concentration and incubation time were determined via Taguchi design. The maximum BC production, 9.1 ± 0.66 g·L−1 (dry weight), was obtained from 32 °C, pH 5.5, 8 log CFU·mL−1 and 14 days of incubation. The inoculum concentration was the most significant factor affecting BC yield. A value of 8 log CFU·mL−1 and 14 days of incubation led to significantly higher levels of BC yield than other concentrations (8.5, 9, 9.5, 10 and 10.5 log CFU·mL−1) (p < 0.002) and days (15, 16, 17, 21 and 28) (p < 0.001). The studied features, namely absorption peaks (Fourier transform infrared spectroscopy), pattern and the crystallinity index (X-ray diffraction analysis) of the BC obtained in this study were all in parallel with the characteristics of cellulose I. The study demonstrates that optimized parameters were effective in producing BC with high water-holding capacity, tensile strength, elongation and Young’s modulus (mechanical tests) by K. rhaeticus K23.

Optimization of bacterial cellulose production by Komagataeibacter sucrofermentans in synthetic media and agrifood side streams supplemented with organic acids and vitamins

The effect of thiamine (TA), ascorbic acid (AA), citric acid, and gallic acid (GA) on bacterial cellulose (BC) production by Komagataeibacter sucrofermentans, in synthetic (Hestrin and Schramm, HS) and natural substrates (industrial raisins finishing side stream extract, FSSE; orange juice, OJ; green tea extract, GTE), was investigated. The Response Surface Methodology was found reliable for BC yield prediction and optimization. Higher yields were achieved in the FSSE substrates, especially those supplemented with AA, TA, and GA (up to 19.4 g BC/L). The yield in the non-fortified substrates was 1.1–5.4 and 11.6–15.7 g/L, in HS and FSSE, respectively. The best yield in the natural non-fortified substrate FSSE-OJ-GTE (50–20-30 %), was 5.9 g/L. The porosity, crystallinity, and antioxidant properties of the produced BC films were affected by both the substrate and the drying method (freeze- or oven-drying). The natural substrates and the process wastewaters can be further exploited towards added value and sustainability. Overproduction of bacterial cellulose from Acetobacter xylinum BPR2001 using food industries wastes.

In this study, a cost-effective complex culture media containing molasses and corn steep liquor (CSL) was developed for the high production of bacterial cellulose (BC) by investigating the effect of four effective factors on BC production at three levels using Taguchi and combined methods. The predicted and actual values of BC production in optimal conditions by Taguchi and combined methods were 8.41 and 14.52g/L, respectively. These results showed that the combined method was more suitable for predicting the optimal conditions in the optimization of BC production, the cost of developed culture medium was around 94% cost of HS medium preparation, molasses was the most effective factor in both experimental design methods, and initial pH adjustment had little impact on BC production. Then, the effect of inoculation conditions containing three factors of inoculation age, ethanol addition time, and agitation rate on the increase of BC production at three levels was investigated using the response surface methodology with the Box-Behnken design algorithm. Under the optimal conditions including inoculum age of 3 days, ethanol addition time of 10 days, and stirring speed of 100rpm, the predicted and experimental results of BC production were 21.61 and 20.21g/L, respectively. This is among the highest ever reported for BC production, which was achieved with a more cost-effective culture medium containing molasses and CSL.

Optimization of bacterial cellulose production from prickly pear peels and its ex situ impregnation with fruit byproducts for antimicrobial and strawberry packaging applications

Bacterial cellulose (BC) is currently among the most promising natural polymers. However, the production costs and biological inactivity are still challenges. The current study exploited the enzymatically hydrolyzed prickly pear peels (PPP) for BC production, which supported about 2.94 g/L as the sole production medium. The BC production was further optimized through a central composite design, where the maximum BC production was 6.01 g/L at 68 % PPPE at pH 4 after 11 days of incubation at 20 °C. The produced BC was characterized by FT-IR spectroscopy, XRD, and SEM analysis, and the results showed that PPPE is a promising carbon source for pure BC production. The BC membrane was separately loaded with several fruit byproduct extracts to enhance its biological activity for multiple applications. BC loaded with pomegranate peel extract (BC/PPE) revealed significant broad-spectrum antimicrobial activity, followed by BC loaded with pomegranate molasses (BC/PM). The BC/PPE membrane enhanced the shelf-life storage of strawberry fruits by about 5 days, with a reduction in the fruits' weight loss of 15 % compared to the uncovered group. The current study revealed the successful application of PPE for sustainable BC production with its packaging potential for enhancing strawberry shelf-life when loaded with PPE or PM.

Optimization of Bacterial Cellulose Production from Wastewater of Noodle Processing by Komagataeibacter sp. PAP1 and Bio-Cellulose Paper Production

Bacterial Cellulose (BC) production from Fermented Rice Noodle Wastewater (FRNW) was fermented by Komagataeibacter sp. PAP1. In order to increase the production of BC, the FRNW-based medium was prepared and optimized for the cultivation of this bacterium. The optimized FRNW-based medium was composed of 5 % (w/v) mannitol, 0.1 % (w/v) beef extract, 0.5 % (v/v) ethanol, 1 % (v/v) acetic acid, pH 7.0 which were incubated at 30 °C for 10 days. Under these conditions, BC yielded 11.76 ± 0.34 g/L higher (4.40 fold) than the standard Hestrin-Schramm (HS) medium. The study on growth and BC production by Komagataeibacter sp. PAP1 in optimized culture condition showed that BC production by Komagataeibacter sp. PAP1 was growth-associated. The bacterial cell of Komagataeibacter sp. PAP1 increased exponentially from the 3rd to 5th day. The BC paper sheets produced using the obtained BC pellicle from an optimized FRNW-based medium gave higher mechanical strength than those from standard HS medium. This study reveals the use of FRNW as a substrate for BC production. The results indicated that FRNW, which is more environmentally friendly, can be used as an alternative low-cost substrate for BC production.

The optimization of bacterial cellulose production and its applications: a review

Bacterial cellulose (BC) is a promising biomaterial due to its specific and excellent properties such as high cellulose purity, mechanical strength, high crystallinity and biodegradability. BC has potential application in electronics, cosmetics, medicine and food and its related products due to its properties. Various chemical compounds have been added to the BC production process to analyse the effect on the BC yield and enhance BC’s properties. In this review, we discuss in-situ and ex-situ modifications and biotechnological approach as alternative optimisation methods in BC production. Given that BC has excellent structural characteristics, which lead to better physical and mechanical properties compared with plant cellulose, the properties and applications of BC are also discussed in this paper.

Optimization of bacterial cellulose production by Komagataeibacter xylinus PTCC 1734 in a low-cost medium using optimal combined design.

This study was aimed to optimize the production of bacterial cellulose (BC) by Komagataeibacter xylinus PTCC 1734 using mixture of date syrup and cheese whey as carbon sources as well as ascorbic acid as a supplementary agent and to characterize the properties of produced BC. The results showed the highest BC production on the 10th day. The 50:50 ratio of date syrup and cheese whey lead to the highest BC production. Three samples were selected in optimal cultivation conditions until the 10th day, with different ascorbic acid concentrations (0, 0.1 and 0.4%). SEM results showed no difference in the morphology of BC product in the optimal samples, where the average diameter of cellulose nanofibers produced was in the range of nanometer. The FTIR test results showed no difference in the chemical structure of cellulose product in different ascorbic acid concentrations. According to XRD and TGA analyses, the highest degree of BC crystallinity and thermal resistance was obtained at maximum ascorbic acid concentration (0.04%). Consequently, the 50:50 ratio of date syrup and cheese whey and 10th day of fermentation time were selected as the best conditions for BC production. Though ascorbic acid reduced production efficiency, it improved the physical properties of the BC product.

Statistical optimization of bacterial cellulose production by Leifsonia soli and its physico-chemical characterization

Bacterial cellulose has multiple applications in various industries such as food, biomedical, textile due to its uniqueness of being a better bio-compatible coating agent, binding material, etc. In this study, optimization of the culture medium for producing BC from Leifsonia soli was carried out by selecting different parameters. Five significant factors such as maltose, pH, incubation days, soy whey and calcium chloride were estimated through ANOVA based response surface methodology. Maximum cellulose production (5.97 g/L) was obtained where maltose 1 % (w/v) supplemented with 0.8 % (v/v) soy whey and calcium chloride 0.8 % (w/v) at pH 6.5 for 7 days of incubation. In addition, assurance of cellulose production from bacteria was done by using High-performance liquid chromatography analysis. Further, the structure and purity of obtained cellulose were examined by SEM and elemental analysis where it was observed that the sample holds the value of carbon 44.1 ± 0.20 % and hydrogen 6.2 ± 0.3 %, respectively. This study concludes that the addition of maltose and soy whey could be used as carbon, nitrogen sources and calcium chloride was used as an additive for the bacterial cellulose production compared to the Hestrin Schramm medium. In addition, the calculated water holding capacity of the sample was found to be 73 %.

Optimization of Bacterial Cellulose Production from Pineapple Waste using Different Fermentation Method

Bacterial cellulose or Nata is produced using pineapple waste as substrate for the fermentation process by Acetobacter xylinum. This study was aimed to optimize the production of bacterial cellulose in static and agitated condition. Production of bacterial cellulose in agitated condition has been performed at different rpm, with and without adding micro particles. The fermentation conditions were set at pH 5.0 and 28 °C. The highest bacterial cellulose formation is obtained from agitated condition with additional micro particles at 120 rpm whereas the bacterial cellulose weight is increased by 70.23 % compared to the static condition. The addition of microparticles helped in increasing the bacterial cellulose formation by 15.19 %. From the optimization using Design Expert®, yeast extracts and potassium dihydrogen phosphate (KH2PO4) are the most significant factors affecting the bacterial cellulose production. From the results, the optimum formulation for bacterial cellulose production from pineapple waste as medium were yeast extract 6 g, sucrose 20 g, bactopepton 1.49 g and KH2PO4 1.08 g for 176.47 g (wet weight) of bacterial cellulose production.

Isolation and Identification of Bacteria-producing Cellulose from Tropical Fruit for Halal Capsule Application

Bacterial cellulose (BC) is pure cellulose synthesized by various species of bacteria. Raising demands on bacterial cellulose is due to its pure and simpler structure. It has plenty of applications in various industries such as food, medical and cosmetics, rendering it a choice in halal industry application as a substitute for non-halal gelatine. However, challenges arise during the BC production such as high production cost and low volumetric yield. In this study, BC is studied to overcome the barriers to BC production. Isolation and identification of cellulose producing bacteria were carried out on eight different tropical fruit sources using Hestrin-Schramm media in static culture condition. Morphological and molecular identification by microscopic observation, gram staining, and 16S rRNA analysis were conducted to identify the characteristics and strain of the new isolates. Next, the selected colonies were challenged to grow in agitating condition using modified HS media. The effects of carbon concentration and agitation speed on the production of cellulose were investigated using on central composite design (CCD). Three new cellulose producing bacteria were successfully isolated and identified to be similar to Enterobacter sp. SJZ-5, Bacterium sp NLAE-zl-H356, and Bulkhoderia sp. RD_DACAR_02 through morphological and molecular analysis. The most potent strain which is similar to Enterobacter sp. SJZ-5 (named as Enterobacter sp.M003) has been chosen for BC optimization study for high BC production using modified HS media. Optimization of bacterial cellulose production using response surface methodology (RSM) with 13 runs indicated that the optimal production parameters were 17.5 g/L for carbon concentration at 277 rpm for agitation speed gave 1.7g/L cellulose. It is expected that the newly isolated bacteria will be able to provide an alternative to gelatine for halal capsule production, thus minimizing and replacing non-halal gelatine usage.

Biochemical and cellular properties of Gluconacetobacter xylinus cultures exposed to different modes of rotating magnetic field

Abstract The aim of the present study was to evaluate the impact of a rotating magnetic field (RMF) on cellular and biochemical properties of Gluconacetobacter xylinus during the process of cellulose synthesis by these bacteria. The application of the RMF during bacterial cellulose (BC) production intensified the biochemical processes in G. xylinus as compared to the RMF-unexposed cultures. Moreover, the RMF had a positive impact on the growth of cellulose-producing bacteria. Furthermore, the application of RMF did not increase the number of mutants unable to produce cellulose. In terms of BC production efficacy, the most favorable properties were found in the setting where RMF generator was switched off for the first 72 h of cultivation and switched on for the further 72 h. The results obtained can be used in subsequent studies concerning the optimization of BC production using different types of magnetic fields including RMF, especially.

The statistical optimization of bacterial cellulose production via semi-continuous operation mode

Bacterial cellulose (BC) is highly pure and has a higher crystallinity and molecular weight than plant cellulose. Therefore, BC can be used in many different areas such as biotechnology, pharmaceutical, cosmetics. Because of the price of BC, the productivity of BC is an important parameter for industrial scale applications. In this study, BC was produced in static culture using a semi-continuous operation mode; the conditions were optimized using response surface methodology (RSM). The collected BC was characterized by Fourier-transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM), and tensile strength. Optimization parameters were selected as glucose concentration, surface area/volume ratio, surface area and incubation day intervals. The optimum values for incubation day intervals, volume changing ratios, glucose concentrations and surface area/volume ratios were 7 days, 66%, 50g/L and 1.22cm−1, respectively. BC productivity reached 0.284g/L/day under optimal conditions, while the model equation proposed 0.289g/L/day. RSM is essential for determining the optimum values of parameters for BC production compared with the one-variable-at-a-time method. The semi-continuous operation mode is alternative and a good candidate for the industrial scale production of BC.

Optimization of bacterial cellulose production by Gluconacetobacter xylinus using carob and haricot bean

Bacterial cellulose (BC) can be used in medical, biomedical, electronic, food, and paper industries because of its unique properties distinguishing it from plant cellulose. BC production was statistically optimized by Gluconacetobacter xylinus strain using carob and haricot bean (CHb) medium. Eight parameters were evaluated by Plackett–Burman Design and significant three parameters were optimized by Central Composite Design. Optimal conditions for production of BC in static culture were found as: 2.5g/L carbon source, 2.75g/L protein source, 9.3% inoculum ratio, 1.15g/L citric acid, 2.7g/L Na2HPO4, 30°C incubation temperature, 5.5 initial pH, and 9days of incubation. This study reveals that BC production can be carried out using carob and haricot bean extracts as carbon and nitrogen sources, and CHb medium has higher buffering capacity compared to Hestrin and Schramm media. Model obtained from this study is used to predict and optimize BC production yield using CHb medium.

Statistical optimization of culture conditions for enhanced bacterial cellulose production by Gluconoacetobacter hansenii NCIM 2529

In this study we carried out optimization of bacterial cellulose (BC) production from Gluconoacetobacter hansenii NCIM 2529 using a statistical model. Optimization of medium components and physical parameters used for bacterial cellulose production in G. hansenii NCIM 2529 was performed by two-step optimization. Eight culture parameters were screened by the Plackett–Burman design and significant parameters were optimized by the response surface methodology using a central composite design. Through these two-phase experiments, optimization of BC production was carried out for high production. The maximum average productivity of BC with optimized medium was 0.71 g/100 mL upon fermentation in a shake flask. The average BC production was observed to increase by about 44 % using these statistical techniques of media optimization.

Production of Microbial Cellulose by a Bacterium Isolated from Fruit

This study presents the production of bacterial cellulose (BC) by a bacterium isolated from a rotten fruit and its process optimization. Here, isolation and screening of potent cellulose producers were carried out from different natural sources, viz., soil, rotten fruits, and vegetables and vinegar. A total of 200 bacterial isolates were obtained, which were screened for cellulose production using Hestrin-Schramm medium. A novel and potent cellulose-producing bacterium was newly isolated from a rotten fruit and identified as Gluconacetobacter sp. F6 through 16S ribosomal DNA sequencing and morphological, cultural, and biochemical characteristics. After optimization of culture conditions, including pH, temperature, agitation, carbon/nitrogen sources, and inducers, the BC production was greatly increased from 0.52 to 4.5 g/l (8.65-fold increase). The optimal culture medium contained 1% (w/v) glucose, 1.5% (w/v) yeast extract, 0.5% (w/v) peptone, 0.27% (w/v) disodium hydrogen phosphate, 0.115% (w/v) citric acid, and 0.4% (w/v) ethanol. BC produced was analyzed for the presence of cellulose fibrils by epiflourescent microscopy using Calcofluor white stain and scanning electron microscopy and confirmed by NMR. There are very scanty reports about the optimization of BC production by bacteria isolated from rotten fruits.
