Light Energy Use Efficiency in Photosystem II of Tomato Is Related to Leaf Age and Light Intensity

The fundamental key to increase photosynthetic efficiency of crop plants lies in optimizing the light energy use efficiency. In our study, we used tomato to evaluate the allocation of absorbed light energy in young and mature leaves, and to estimate if the extent of photoinhibition and photoprotection can be affected by the leaf age. A reduced efficiency of the oxygen-evolving complex, in young leaves compared to mature ones, resulted in a donor-side photoinhibition, as judged from the significantly lower Fv/Fm ratio, in young leaves. The detected increased 1O2 production in young leaves was probably due to a donor-side photoinhibition. The effective quantum yield of photosystem II (PSII) photochemistry (ΦPSII), at low light intensity (LLI, 426 μmol photons m−2 s−1), was significantly lower in young compared to mature leaves. Moreover, the non-significant increase in non-photochemical energy loss in PSII (ΦNPQ) could not counteract the decreased ΦPSII, and as a result the non-regulated energy loss in PSII (ΦNO) increased in young leaves, compared to mature ones. The significantly lower ΦPSII in young leaves can be attributed to the increased reactive oxygen species (ROS) creation that diminished the efficiency of the open PSII reaction centers (Fv’/Fm’), but without having any impact on the fraction of the open reaction centers. The reduced excess excitation energy, in mature leaves compared to young ones, at LLI, also revealed an enhanced PSII efficiency of mature leaves. However, there was almost no difference in the light energy use efficiency between young and mature leaves at the high light intensity (HLI, 1000 μmol photons m−2 s−1). The ability of mature tomato leaves to constrain photoinhibition is possible related to an enhanced photosynthetic function and a better growth rate. We concluded that the light energy use efficiency in tomato leaves is influenced by both the leaf age and the light intensity. Furthermore, the degrees of photoinhibition and photoprotection are related to the leaf developmental stage.

Elevated CO2 Concentration Enhances Drought Tolerance by Mitigating Oxidative Stress and Enhancing Carbon Assimilation in Foxtail Millet (Setaria italica)

ABSTRACTElevated CO2 concentration (eCO2) can modulate the response of crop plants to drought stress (DS). This study aimed to investigate the response of leaf gas exchange, chlorophyll fluorescence, antioxidant activities, osmotic adjustment substance, phytohormone and signal transduction regulatory enzymes, as well as related genes in foxtail millet to DS (water stress for 10 days), ambient condition (aCO2, 400 μmol mol−1) and eCO2 (600 μmol mol−1). eCO2 significantly increased the net photosynthetic rate, maximum net photosynthetic rate, chlorophyll a content, transpiration rate and stomatal conductance, but did not affect leaf instantaneous water‐use efficiency under DS. eCO2 also significantly enhanced the quantum yield of Photosystem II (PSII), photosynthetic electron transport, and proportion of open PSII reaction centers under DS. Moreover, eCO2 significantly increased abscisic acid (ABA) content, proline content, and the activities of peroxidase, superoxide dismutase, and calcium‐dependent protein kinase under DS, leading to a significant reduction in malondialdehyde content. eCO2 significantly increased the expressions of gene encoding ABA‐, stress‐ and ripening‐induced proteins and ABA‐responsive element binding factor under DS. Our results clearly demonstrated the vital role of eCO2 in mitigating the drought‐induced damage over ambient CO2 grown foxtail millet.

Deciphering the Mechanism of Melatonin-Induced Enhancement of Photosystem II Function in Moderate Drought-Stressed Oregano Plants.

Melatonin (MT) is considered as an antistress molecule that plays a constructive role in the acclimation of plants to both biotic and abiotic stress conditions. In the present study, we assessed the impact of 10 and 100 μM MT foliar spray, on chlorophyll content, and photosystem II (PSII) function, under moderate drought stress, on oregano (Origanum vulgare L.) plants. Our aim was to elucidate the molecular mechanism of MT action on the photosynthetic electron transport process. Foliar spray with 100 μM MT was more effective in mitigating the negative impact of moderate drought stress on PSII function, compared to 10 μM MT. MT foliar spray significantly improved the reduced efficiency of the oxygen-evolving complex (OEC), and PSII photoinhibition (Fv/Fm), which were caused by drought stress. Under moderate drought stress, foliar spray with 100 μM MT, compared with the water sprayed (WA) leaves, increased the non-photochemical quenching (NPQ) by 31%, at the growth irradiance (GI, 205 μmol photons m-2 s-1), and by 13% at a high irradiance (HI, 1000 μmol photons m-2 s-1). However, the lower NPQ increase at HI was demonstrated to be more effective in decreasing the singlet-excited oxygen (1O2) production at HI (-38%), in drought-stressed oregano plants sprayed with 100 μM MT, than the corresponding decrease in 1O2 production at the GI (-20%), both compared with the respective WA-sprayed leaves under moderate drought. The reduced 1O2 production resulted in a significant increase in the quantum yield of PSII photochemistry (ΦPSII), and the electron transport rate (ETR), in moderate drought-stressed plants sprayed with 100 μM MT, compared with WA-sprayed plants, but only at the HI (+27%). Our results suggest that the enhancement of PSII functionality, with 100 μM MT under moderate drought stress, was initiated by the NPQ mechanism, which decreased the 1O2 production and increased the fraction of open PSII reaction centers (qp), resulting in an increased ETR.

Hormetic Response of Photosystem II Function Induced by Nontoxic Calcium Hydroxide Nanoparticles.

In recent years, inorganic nanoparticles, including calcium hydroxide nanoparticles [Ca Ca(OH)2 NPs], have attracted significant interest for their ability to impact plant photosynthesis and boost agricultural productivity. In this study, the effects of 15 and 30 mg L-1 oleylamine-coated calcium hydroxide nanoparticles [Ca(OH)2@OAm NPs] on photosystem II (PSII) photochemistry were investigated on tomato plants at their growth irradiance (GI) (580 μmol photons m-2 s-1) and at high irradiance (HI) (1000 μmol photons m-2 s-1). Ca(OH)2@OAm NPs synthesized via a microwave-assisted method revealed a crystallite size of 25 nm with 34% w/w of oleylamine coater, a hydrodynamic size of 145 nm, and a ζ-potential of 4 mV. Compared with the control plants (sprayed with distilled water), PSII efficiency in tomato plants sprayed with Ca(OH)2@OAm NPs declined as soon as 90 min after the spray, accompanied by a higher excess excitation energy at PSII. Nevertheless, after 72 h, the effective quantum yield of PSII electron transport (ΦPSII) in tomato plants sprayed with Ca(OH)2@OAm NPs enhanced due to both an increase in the fraction of open PSII reaction centers (qp) and to the enhancement in the excitation capture efficiency (Fv'/Fm') of these centers. However, the decrease at the same time in non-photochemical quenching (NPQ) resulted in an increased generation of reactive oxygen species (ROS). It can be concluded that Ca(OH)2@OAm NPs, by effectively regulating the non-photochemical quenching (NPQ) mechanism, enhanced the electron transport rate (ETR) and decreased the excess excitation energy in tomato leaves. The delay in the enhancement of PSII photochemistry by the calcium hydroxide NPs was less at the GI than at the HI. The enhancement of PSII function by calcium hydroxide NPs is suggested to be triggered by the NPQ mechanism that intensifies ROS generation, which is considered to be beneficial. Calcium hydroxide nanoparticles, in less than 72 h, activated a ROS regulatory network of light energy partitioning signaling that enhanced PSII function. Therefore, synthesized Ca(OH)2@OAm NPs could potentially be used as photosynthetic biostimulants to enhance crop yields, pending further testing on other plant species.

Modulation of Photosystem II Function in Celery via Foliar-Applied Salicylic Acid during Gradual Water Deficit Stress.

Water deficit is the major stress factor magnified by climate change that causes the most reductions in plant productivity. Knowledge of photosystem II (PSII) response mechanisms underlying crop vulnerability to drought is critical to better understanding the consequences of climate change on crop plants. Salicylic acid (SA) application under drought stress may stimulate PSII function, although the exact mechanism remains essentially unclear. To reveal the PSII response mechanism of celery plants sprayed with water (WA) or SA, we employed chlorophyll fluorescence imaging analysis at 48 h, 96 h, and 192 h after watering. The results showed that up to 96 h after watering, the stroma lamellae of SA-sprayed leaves appeared dilated, and the efficiency of PSII declined, compared to WA-sprayed plants, which displayed a better PSII function. However, 192 h after watering, the stroma lamellae of SA-sprayed leaves was restored, while SA boosted chlorophyll synthesis, and by ameliorating the osmotic potential of celery plants, it resulted in higher relative leaf water content compared to WA-sprayed plants. SA, by acting as an antioxidant under drought stress, suppressed phototoxicity, thereby offering PSII photoprotection, together with enhanced effective quantum yield of PSII photochemistry (ΦPSII) and decreased quantity of singlet oxygen (1O2) generation compared to WA-sprayed plants. The PSII photoprotection mechanism induced by SA under drought stress was triggered by non-photochemical quenching (NPQ), which is a strategy to protect the chloroplast from photo-oxidative damage by dissipating the excess light energy as heat. This photoprotective mechanism, triggered by NPQ under drought stress, was adequate in keeping, especially in high-light conditions, an equal fraction of open PSII reaction centers (qp) as of non-stress conditions. Thus, under water deficit stress, SA activates a regulatory network of stress and light energy partitioning signaling that can mitigate, to an extent, the water deficit stress on PSII functioning.

Mechanistic Insights on Salicylic Acid-Induced Enhancement of Photosystem II Function in Basil Plants under Non-Stress or Mild Drought Stress.

Photosystem II (PSII) functions were investigated in basil (Ocimum basilicum L.) plants sprayed with 1 mM salicylic acid (SA) under non-stress (NS) or mild drought-stress (MiDS) conditions. Under MiDS, SA-sprayed leaves retained significantly higher (+36%) chlorophyll content compared to NS, SA-sprayed leaves. PSII efficiency in SA-sprayed leaves under NS conditions, evaluated at both low light (LL, 200 μmol photons m-2 s-1) and high light (HL, 900 μmol photons m-2 s-1), increased significantly with a parallel significant decrease in the excitation pressure at PSII (1-qL) and the excess excitation energy (EXC). This enhancement of PSII efficiency under NS conditions was induced by the mechanism of non-photochemical quenching (NPQ) that reduced singlet oxygen (1O2) production, as indicated by the reduced quantum yield of non-regulated energy loss in PSII (ΦNO). Under MiDS, the thylakoid structure of water-sprayed leaves appeared slightly dilated, and the efficiency of PSII declined, compared to NS conditions. In contrast, the thylakoid structure of SA-sprayed leaves did not change under MiDS, while PSII functionality was retained, similar to NS plants at HL. This was due to the photoprotective heat dissipation by NPQ, which was sufficient to retain the same percentage of open PSII reaction centers (qp), as in NS conditions and HL. We suggest that the redox status of the plastoquinone pool (qp) under MiDS and HL initiated the acclimation response to MiDS in SA-sprayed leaves, which retained the same electron transport rate (ETR) with control plants. Foliar spray of SA could be considered as a method to improve PSII efficiency in basil plants under NS conditions, at both LL and HL, while under MiDS and HL conditions, basil plants could retain PSII efficiency similar to control plants.

A SIF-based approach for quantifying canopy photosynthesis by simulating the fraction of open PSII reaction centers (qL)

Advances in retrieval of solar-induced chlorophyll fluorescence (SIF) provide a promising and independent approach for quantifying gross primary production (GPP) across spatial scales. Recent studies have highlighted the prominent role of qL, the fraction of open Photosystem II (PSII) reaction centers, in mechanistically modeling GPP from remote sensing SIF. However, due to the limited availability of simulated and experimental data, a comprehensive understanding of qL responses to environmental and physiological variations has yet to emerge, and as a consequence, prediction of qL across leaf and canopy scales is still in an early stage. Based on a global sensitivity analysis of a recently developed mechanical model of photosynthesis, we find that the broadband total SIF emitted from PSII (SIFTOT_FULL_PSII) and leaf temperature (TLeaf) are the two major predictors of qL. A leaf-level instrument is designed to obtain concurrent measurements of qL, SIFTOT_FULL_PSII, and TLeaf over a wide range of environmental conditions. From these measurements, we show that qL can be modelled as a hyperbolic function of SIFTOT_FULL_PSII with only one temperature-related parameter m which increases with temperature, but decreases rapidly as temperatures exceed the optimum temperature. It is suggested that m can be mathematically modelled by a peaked function. The results of the leaf-level experiments on winter wheat demonstrate that the proposed model predicts qL with high accuracy (R2 ≥ 0.91, rRMSE ≤ 8.46%) under diverse light and temperature conditions. The essential steps necessary to apply it at canopy scale, including estimating the escape fraction, removing fluorescence emitted from Photosystem I, and reconstructing SIFTOT_FULL_PSII from top-of-canopy (TOC) narrowband SIF, are also presented. Our results confirm that estimated GPP using SIF-informed qL agrees well with measured GPP at a winter wheat site (R2 = 0.81, rRMSE = 12.03%). The key benefit of SIF-informed approach is that SIFTOT_FULL_PSII provides critical information on the collective influence of the sub-canopy light environment on qL, avoiding the requirement to explicitly estimate qL at different canopy depths, potentially promoting the ability of SIF to mechanistically quantify photosynthetic CO2 assimilation at large scales.

Salinity-induced changes in plastoquinone pool redox state in halophytic Mesembryanthemum crystallinum L.

We have analyzed the effect of salinity on photosystem II (PSII) photochemistry and plastoquinone (PQ) pool in halophytic Mesembryanthemum crystallinum plants. Under prolonged salinity conditions (7 or 10 days of 0.4 M NaCl treatment) we noted an enlarged pool of open PSII reaction centers and increased energy conservation efficiency, as envisaged by parameters of the fast and slow kinetics of chlorophyll a fluorescence. Measurements of oxygen evolution, using 2,6-dichloro-1,4-benzoquinone as an electron acceptor, showed stimulation of the PSII activity due to salinity. In salt-acclimated plants (10 days of NaCl treatment), the improved PSII performance was associated with an increase in the size of the photochemically active PQ pool and the extent of its reduction. This was accompanied by a rise in the NADP+/NADPH ratio. The presented data suggest that a redistribution of PQ molecules between photochemically active and non-active fractions and a change of the redox state of the photochemically active PQ pool indicate and regulate the acclimation of the photosynthetic apparatus to salinity.

Mechanistic Insights on Salicylic Acid Mediated Enhancement of Photosystem II Function in Oregano Seedlings Subjected to Moderate Drought Stress.

Dramatic climate change has led to an increase in the intensity and frequency of drought episodes and, together with the high light conditions of the Mediterranean area, detrimentally influences crop production. Salicylic acid (SA) has been shown to supress phototoxicity, offering photosystem II (PSII) photoprotection. In the current study, we attempted to reveal the mechanism by which SA is improving PSII efficiency in oregano seedlings under moderate drought stress (MoDS). Foliar application of SA decreased chlorophyll content under normal growth conditions, but under MoDS increased chlorophyll content, compared to H2O-sprayed oregano seedlings. SA improved the PSII efficiency of oregano seedlings under normal growth conditions at high light (HL), and under MoDS, at both low light (LL) and HL. The mechanism by which, under normal growth conditions and HL, SA sprayed oregano seedlings compared to H2O-sprayed exhibited a more efficient PSII photochemistry, was the increased (17%) fraction of open PSII reaction centers (qp), and the increased (7%) efficiency of these open reaction centers (Fv'/Fm'), which resulted in an enhanced (24%) electron transport rate (ETR). SA application under MoDS, by modulating chlorophyll content, resulted in optimized antenna size and enhanced effective quantum yield of PSII photochemistry (ΦPSII) under both LL (7%) and HL (25%), compared to non-SA-sprayed oregano seedlings. This increased effective quantum yield of PSII photochemistry (ΦPSII) was due to the enhanced efficiency of the oxygen evolving complex (OEC), and the increased fraction of open PSII reaction centers (qp), which resulted in an increased electron transport rate (ETR) and a lower amount of singlet oxygen (1O2) production with less excess excitation energy (EXC).

A Hormetic Spatiotemporal Photosystem II Response Mechanism of Salvia to Excess Zinc Exposure.

Exposure of Salvia sclarea plants to excess Zn for 8 days resulted in increased Ca, Fe, Mn, and Zn concentrations, but decreased Mg, in the aboveground tissues. The significant increase in the aboveground tissues of Mn, which is vital in the oxygen-evolving complex (OEC) of photosystem II (PSII), contributed to the higher efficiency of the OEC, and together with the increased Fe, which has a fundamental role as a component of the enzymes involved in the electron transport process, resulted in an increased electron transport rate (ETR). The decreased Mg content in the aboveground tissues contributed to decreased chlorophyll content that reduced excess absorption of sunlight and operated to improve PSII photochemistry (ΦPSII), decreasing excess energy at PSII and lowering the degree of photoinhibition, as judged from the increased maximum efficiency of PSII photochemistry (Fv/Fm). The molecular mechanism by which Zn-treated leaves displayed an improved PSII photochemistry was the increased fraction of open PSII reaction centers (qp) and, mainly, the increased efficiency of the reaction centers (Fv′/Fm′) that enhanced ETR. Elemental bioimaging of Zn and Ca by laser ablation–inductively coupled plasma–mass spectrometry (LA–ICP–MS) revealed their co-localization in the mid-leaf veins. The high Zn concentration was located in the mid-leaf-vein area, while mesophyll cells accumulated small amounts of Zn, thus resembling a spatiotemporal heterogenous response and suggesting an adaptive strategy. These findings contribute to our understanding of how exposure to excess Zn triggered a hormetic response of PSII photochemistry. Exposure of aromatic and medicinal plants to excess Zn in hydroponics can be regarded as an economical approach to ameliorate the deficiency of Fe and Zn, which are essential micronutrients for human health.

Harnessing the Role of Foliar Applied Salicylic Acid in Decreasing Chlorophyll Content to Reassess Photosystem II Photoprotection in Crop Plants.

Salicylic acid (SA), an essential plant hormone, has received much attention due to its role in modulating the adverse effects of biotic and abiotic stresses, acting as an antioxidant and plant growth regulator. However, its role in photosynthesis under non stress conditions is controversial. By chlorophyll fluorescence imaging analysis, we evaluated the consequences of foliar applied 1 mM SA on photosystem II (PSII) efficiency of tomato (Solanum lycopersicum L.) plants and estimated the reactive oxygen species (ROS) generation. Tomato leaves sprayed with 1 mM SA displayed lower chlorophyll content, but the absorbed light energy was preferentially converted into photochemical energy rather than dissipated as thermal energy by non-photochemical quenching (NPQ), indicating photoprotective effects provided by the foliar applied SA. This decreased NPQ, after 72 h treatment by 1 mM SA, resulted in an increased electron transport rate (ETR). The molecular mechanism by which the absorbed light energy was more efficiently directed to photochemistry in the SA treated leaves was the increased fraction of the open PSII reaction centers (qp), and the increased efficiency of open reaction centers (Fv’/Fm’). SA induced a decrease in chlorophyll content, resulting in a decrease in non-regulated energy dissipated in PSII (ΦNO) under high light (HL) treatment, suggesting a lower amount of triplet excited state chlorophyll (3Chl*) molecules available to produce singlet oxygen (1O2). Yet, the increased efficiency, compared to the control, of the oxygen evolving complex (OEC) on the donor side of PSII, associated with lower formation of hydrogen peroxide (H2O2), also contributed to less creation of ROS. We conclude that under non stress conditions, foliar applied SA decreased chlorophyll content and suppressed phototoxicity, offering PSII photoprotection; thus, it can be regarded as a mechanism that reduces photoinhibition and photodamage, improving PSII efficiency in crop plants.

Induction of a Compensatory Photosynthetic Response Mechanism in Tomato Leaves upon Short Time Feeding by the Chewing Insect Spodoptera exigua

Simple SummaryInsects such as beet armyworm (Spodoptera exigua) can cause extensive damage to tomato plants (Solanum lycopersicum). Tomato photosynthesis was clearly reduced directly at S. exigua feeding spots. However, neighboring zones and the rest of the leaf compensated through increased light energy use in photosystem II, possibly trigged by singlet oxygen from the feeding zone. Three hours after feeding, whole-leaf photosynthetic efficiency was as before feeding, demonstrating the compensatory ability. Thus, chlorophyll fluorescence imaging analysis could contribute to understanding the effects of herbivory on photosynthesis at a detailed spatial and temporal pattern.In addition to direct tissue consumption, herbivory may affect other important plant processes. Here, we evaluated the effects of short-time leaf feeding by Spodoptera exigua larvae on the photosynthetic efficiency of tomato plants, using chlorophyll a fluorescence imaging analysis. After 15 min of feeding, the light used for photochemistry at photosystem II (PSII) (ΦPSII), and the regulated heat loss at PSII (ΦNPQ) decreased locally at the feeding zones, accompanied by increased non-regulated energy losses (ΦNO) that indicated increased singlet oxygen (1O2) formation. In contrast, in zones neighboring the feeding zones and in the rest of the leaf, ΦPSII increased due to a decreased ΦNPQ. This suggests that leaf areas not directly affected by herbivory compensate for the photosynthetic losses by increasing the fraction of open PSII reaction centers (qp) and the efficiency of these centers (Fv’/Fm’), because of decreased non-photochemical quenching (NPQ). This compensatory reaction mechanism may be signaled by singlet oxygen formed at the feeding zone. PSII functionality at the feeding zones began to balance with the rest of the leaf 3 h after feeding, in parallel with decreased compensatory responses. Thus, 3 h after feeding, PSII efficiency at the whole-leaf level was the same as before feeding, indicating that the plant managed to overcome the feeding effects with no or minor photosynthetic costs.

Changes in Light Energy Utilization in Photosystem II and Reactive Oxygen Species Generation in Potato Leaves by the Pinworm Tuta absoluta.

We evaluated photosystem II (PSII) functionality in potato plants (Solanum tuberosum L.) before and after a 15 min feeding by the leaf miner Tuta absoluta using chlorophyll a fluorescence imaging analysis combined with reactive oxygen species (ROS) detection. Fifteen minutes after feeding, we observed at the feeding zone and at the whole leaf a decrease in the effective quantum yield of photosystem II (PSII) photochemistry (ΦPSII). While at the feeding zone the quantum yield of regulated non-photochemical energy loss in PSII (ΦNPQ) did not change, at the whole leaf level there was a significant increase. As a result, at the feeding zone a significant increase in the quantum yield of non-regulated energy loss in PSII (ΦNO) occurred, but there was no change at the whole leaf level compared to that before feeding, indicating no change in singlet oxygen (1O2) formation. The decreased ΦPSII after feeding was due to a decreased fraction of open reaction centers (qp), since the efficiency of open PSII reaction centers to utilize the light energy (Fv′/Fm′) did not differ before and after feeding. The decreased fraction of open reaction centers resulted in increased excess excitation energy (EXC) at the feeding zone and at the whole leaf level, while hydrogen peroxide (H2O2) production was detected only at the feeding zone. Although the whole leaf PSII efficiency decreased compared to that before feeding, the maximum efficiency of PSII photochemistry (Fv/Fm), and the efficiency of the water-splitting complex on the donor side of PSII (Fv/Fo), did not differ to that before feeding, thus they cannot be considered as sensitive parameters to monitor biotic stress effects. Chlorophyll fluorescence imaging analysis proved to be a good indicator to monitor even short-term impacts of insect herbivory on photosynthetic function, and among the studied parameters, the reduction status of the plastoquinone pool (qp) was the most sensitive and suitable indicator to probe photosynthetic function under biotic stress.

GROWTH AND PHOTOSYNTHETIC ACTIVITY OF G REE N ALGAE CHLORELLA VULGARIS BEI JER. IN THE PRESENCE OF SELENIUM NANOAQUA CHELATES

Aim. Study of the influence of the novel form of selenium-containing additives, selenium nanoaquachelates carboxylated with citric acid, on biomass accumulation and efficiency of the reactions of the light stage of photosynthesis in unicellular green algae Chlorella vulgaris. Materials and methods. Algae were grown under sterile conditions in 1 L flasks on liquid mineral medium at 25–26 °С. Cultures were provided with continuous irradiation with 40–42 μmol m-2·s-1 photon flux density. The solution of selenium nanoaquachelates carboxylated with citric acid was added to algal cultures immediately after inoculation. The dry mass of algae was determined gravimetrically. Modulated chlorophyll a fluorescence was measured at room temperature, and common fluorescence parameters were calculated. Results. Selenium nanoaquachelates at 0.4–4 mg/L concentrations stimulated the growth of C. vulgaris, and the increase in biomass reached 40–45%. The addition of Se nanoparticles at smaller concentrations (0.07 or 0.2 mg/L) at first caused the retardation of culture growth, but that effect disappeared after 18–24 days of cultivation. When added to C. vulgaris cultures at 0.4–4 mg/L concentrations, selenium nanoaquachelates evoked the initial increase in maximal efficiency of Photosystem II (PSII ) photochemical reactions and in the efficiency of photochemical reactions in open PSII reaction centres. After 6 days of the addition of 2 or 4 mg/L selenium nanoparticles, the fluorescence photochemical quenching coefficient also increased. As a result, the quantum yield of PSII electron transport has risen. Conclusion. Selenium nanoaquachelates carboxylated with citric acid, being added to C. vulgaris cultures at 0.4–4 mg/L concentrations, have positive effect on biomass accumulation, and transiently improve the efficiency of Photosystem II photochemical reactions as well.

Effects of Nitrogen Deficiency on Leaf Chlorophyll Fluorescence Parameters in Two Olive Tree Cultivars ‘Meski’ and ‘Koroneiki’

Effects of nitrogen (N) deficiency on photosynthetic carbon dioxide (CO2) assimilation, photosystem II (PSII) photochemistry and photoinhibition were investigated in young trees of two olive cultivars ‘Meski’ and ‘Koroneiki’ grown in a greenhouse under controlled conditions. The trees were subjected to four different levels of N supply. N deficient trees had a significantly smaller CO2 assimilatory capacity, but showed little changes in maximum quantum efficiency of PSII photochemistry. However, modifications in PSII photochemistry induced by N deficiency were observed. This was reflected in decreases in quantum yield of PSII electron transport (ΦPSII) and efficiency of excitation energy capture by open PSII reaction centres (Fv’/Fm’) and in an increase in non-photochemical quenching (NPQ). These results suggest that modifications in PSII photochemistry might be a mechanism to down-regulate photosynthetic electron transport so that production of adenosine triphosphate (ATP) and nicotinamide adenine dinucleotide phosphate (NADPH). would be in equilibrium with the decreased demand in the Calvin cycle in the N deficient trees. Therefore, both CO2 assimilation rate and total electron flow (Jt) with its compound electron flows devoted to either carboxylation (Jc) or oxygenation (Jo) can be considered as useful tools to assess the N nutrition status of the trees. Clear relationships were found between Amax and the nitrogen nutrition index (NNI) on the one hand, and between Jt and NNI on the other hand. The results demonstrate that ‘Meski’ is more efficient than ‘Koroneiki’ when subjected to N deficiency.

Photoprotective mechanism of the non-target organism Arabidopsis thaliana to paraquat exposure

The response of photosystem II (PSII), of the non-target organism Arabidopsis thaliana, to paraquat (Pq) exposure was studied by chlorophyll fluorescence imaging. Effects of 1mM Pq application by spray on A. thaliana leaves were monitored as soon as 20min after application at the deposit areas of the droplets. A decline in the effective quantum yield of photochemical energy conversion in PSII (ΦPSII) was accompanied by an increase in the quantum yield for dissipation by down regulation in PSII (ΦNPQ). The concomitant decrease in the quantum yield of non-regulated energy loss in PSII (ΦNO) pointed out a quick effective photoprotection mechanism to Pq exposure. Even 1h after Pq spray, when the maximum Pq effect was observed, the decrease of electron transport rate (ETR) and the increase in non-photochemical quenching (NPQ) resulted to maintain almost the same redox state of quinone A (QA) as control plants. Thus, maximal photoprotection was achieved since NPQ was regulated in such a way that PSII reaction centers remained open. Arabidopsis plants were protected from Pq exposure, by increasing NPQ that dissipates light energy and decreases the efficiency of photochemical reactions of photosynthesis (down regulation of PSII) via the “water–water cycle”. PSII photochemistry began to recover 4h after Pq exposure, and this was evident from the increase of ΦPSII, the simultaneous decrease of ΦNPQ, and the concomitant decrease of ΦNO. Yet, ETR began to increase, as well as the fraction of open PSII reaction centers.

Chloroplast ultrastructure regeneration with protection of photosystem II is responsible for the functional ‘stay‐green’ trait in wheat

CN17 is a functional stay-green wheat variety that exhibits delayed leaf senescence and enhanced photosynthetic competence. To better understand these valuable traits, levels of chlorophyll a and b, soluble proteins, unsaturated fatty acids, and other components of CN17 were assayed. In addition, chloroplast ultrastructure, chloroplast number, and differences in gene expression between CN17 and a control variety, MY11, were examined. By 21 d post-anthesis (DPA), CN17 leaves exhibited a significantly higher maximal photochemical efficiency for photosystem II (PSII) (F(v) /F(m) ) and a significantly higher efficiency of excitation capture by open PSII reaction centres (F(v) '/F(m) '). In addition, chlorophyll degradation in CN17 was delayed by approximately 14 d, and was not blocked as observed in cosmetic stay-green phenotypes. The soluble protein content (Ps) of CN17 was higher than MY11 at all timepoints assayed, and the ratio of unsaturated to saturated fatty acids was significantly higher. CN17 also exhibited isolated granal lamellae associated with vesicles and diminished peroxidation, and between 35 and 42 DPA, a sharp decrease in chloroplast number was detected. Taken together, these results strongly support the hypothesis that chloroplast ultrastructure regeneration is responsible for the functional stay-green trait of CN17, and gene expression data provide insight into the mechanistic details.

Photosynthetic characterization of a rolled leaf mutant of rice (Oryza sativa L.)

A new rolling leaf rice mutant was identified which showed an apparently straighter longitudinal shape normal transverse rolling characters at all developing stages. The chlorophyll contents per fresh weight of this mutant leaves were lower than those of wild-type. The electron transfer rate (ETR) and photochemical quenching (qP) were a little higher than those of wild-type. However, because of significant increase of non-photochemical quenching (NPQ), the maximal photosystem II (PSII) photochemistry (Fv/Fm) and the efficiency of excitation energy trapping by open PSII reaction centers in the light–adapted state (Fv’/Fm’) were lower than those of wild-type. Low temperature fluorescence analysis showed that rolling leaf mutant assigned more excited energy to photosystem I (PSI) than to PSII. The superoxide dismutase (SOD) content, soluble sugar content, proline content and malonaldehyde (MDA) content of the rolling leaf mutant were nearly 39.4, 91.2, 96.7 and 143.7% of those of wild-type, respectively. The great increase of MDA content suggests that membrane lipid system was damaged in rolling leaf mutant leaves. These results indicate that rolling leaf mutant decrease the efficiency of light utilization compared to the wild-type. This was because of the reduction of leaf area and chlorophyll contents, and the dissipation of more excitation energy as NPQ as a result of avoiding potential damage of membrane structure.

Effects of Salt Stress on Photosystem II Activity in Sweet Sorghum Seedlings Grown in Pots Outdoors

The effects of the different NaCl treatments (100 mmol L-1, 200 mmol L-1) on photosynthetic characteristics and the photosystem Ⅱ (PSⅡ) activity in sweet sorghum seedlings grown in pots outdoors were carefully investigated in this study. The Na+ content and the ratio of Na+/K+ in leaves increased significantly with increases of NaCl concentration and treatment time. The net photosynthetic rate (Pn), the efficiency of excitation energy captured by open PSII reaction centers (Fv /Fm ), the photochemical quenching (qP) and the actual photosystem II efficiency (ΦPSII) all decreased while the non-photochemical quenching (NPQ) increased greatly with the increasing of NaCl concentration. In addition, Pn, Fv /Fm , qP, and ΦPSII had an ameliorative trend in the 100 mmol L-1 NaCl treatment, but this phenomenon did not appear in 200 mmol L-1 NaCl treatment. The maximum quantum yield of photosystem II photochemistry (Fv/Fm) was slightly affected in 100 mmol L-1 NaCl treatment, whereas distinct decreased in 200 mmol L-1 NaCl treatment. The chlorophyll fluorescence kinetic curves exhibited no changes at the beginning of NaCl treatment, while the rising speeds of O-K and O-J phases increased markedly in the 200 mmol L-1 NaCl treatment after five days. Furthermore, the photoinactivation rate of PSII did not change in both treatments while the repair rate was depressed significantly after five days treatment of 200 mmol L-1 NaCl. Therefore, we suggest that under salt stress the decrease of carbon assimilation in leaves of sweet sorghum seedlings grown in pots outdoors is mainly due to the accumulation of Na+, and salt stress alters the excited energy distribution. During salt stress outdoors, the decline of PSII activity is majorly attributed to the depression of the repair rate of the photoinactivated PSII, rather than the photoinactivation rate under high light. It is helpful to understand the mechanisms of photoinhibition in C4 crop outdoors under salt stress to some extent.

The effect of zearalenone on PSII photochemical activity and growth in wheat and soybean under salt (NaCl) stress

The effects of mycotoxin zearalenone (ZEN) on the photochemical activity of photosystem II (PSII) in wheat and soybean leaf discs incubated in ZEN solutions as well as the after-effects of pre-sowing soaking of seeds in solutions containing ZEN on the photochemical activity of PSII and on the seedlings growth under salt stress (NaCl solutions were investigated). The incubation of wheat leaf discs in ZEN solutions strongly inhibited the energy flux per cross section (CS) for absorption (ABS/CS), trapping (TRo/CS) and electron transport (ETo/CS), while the effects of ZEN action on soybean discs were opposite and the values of those parameters significantly increased with the increase in ZEN concentration. Incubation of seeds in a ZEN solution resulted in an increase in photochemical efficiency of PSII in soybean seedlings, but did not induce any response of PSII in those of wheat at medium illuminations. Only at the stronger illumination for both species did ZEN induce an increase in efficiency of excitation energy capture by open PSII reaction centers, photochemical quenching of chlorophyll a fluorescence and quantum yield of PSII electron transport. Pre-sowing soaking of seeds in a ZEN solution decreased the photoinhibitory injuries of PSII in wheat and soybean due to safe scattering of the excess excitation energy through an increase in energy-dependent quenching (qE) and state transition quenching (qT). ZEN when added to NaCl solutions during the period of germination contributed to reduction in the growth inhibition of wheat seedlings. The incubation of wheat leaf discs in ZEN solutions strongly inhibited CS, ABS/CS, TRo/CS and ETo/CS. Possible effects of ZEN on some physiological processes in plants have been discussed especially in the context with photochemical activity of PSII and a salt stress.
