Identification of outbreak-associated and other strains ofClostridium difficileby numerical analysis of SDS-PAGE protein patterns

Seventy-three cultures of Clostridium difficile isolated both during, and in the period immediately following, an outbreak of infection in a group of three hospitals, were characterized by one-dimensional sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) of whole-cell proteins. Each protein pattern was characterized by the presence of one or two dense bands which were highly reproducible. The protein patterns were used as the basis for a numerical analysis which divided the strains into five phenons (electrophoretic or EP types). The majority, 60 of the 73 cultures, belonged to a single phenon which included strains from both patients and the environment. We conclude that high-resolution SDS-PAGE of proteins provides an effective method for typing C. difficile and therefore for tracing the possible spread of epidemic strains in hospitals and other institutions, thereby allowing a better understanding of the epidemiology of the organism.

Identification of proteins in polyacrylamide gels by mass spectrometric peptide mapping combined with database search

Mass spectrometric peptide mapping of proteins separated by one-dimensional sodium dodecyl sulphate polyacrylamide gel electrophoresis has been investigated. The best results are obtained after blotting of the proteins onto polyvinylidene difluoride membranes followed by enzymatic digestion of the protein on the membrane. The peptide maps were investigated in terms of completeness and applicability for protein identification using a previously developed database search program as well as for the possibility for full characterization of covalent modifications in the proteins. The most complete peptide maps were obtained when the proteins were reduced and alkylated on the membrane prior to enzymatic digestion followed by separation of the resulting mixture by high performance liquid chromatography prior to mass spectrometric analysis. Such peptide maps cover up to 98% of the sequence and consequently may allow complete characterization of post-translational modifications in proteins for which the amino acid sequence is known. The fastest and most sensitive procedure to obtain peptide maps sufficient for protein identification was direct analysis of the extracted peptide mixture by matrix-assisted laser desorption ionization (MALDI) mass spectrometry. The use of external and internal calibration of MALDI spectra for database searches is evaluated as well as the possibility of including a post-calibration routine within the search program.

Mycoplasma pneumoniae lacks immunologically-active eukaryotic actin-like antigens.

Mycoplasma pneumoniae was tested for immunologically active eukaryotic actin-like antigens with the use of both polyclonal and monoclonal anti-actin antibodies. No reactivity was demonstrable. Monoclonal antibody OC2F5, which reacts with a M. pneumoniae antigen that co-migrates with actin in one-dimensional sodium dodecyl sulphate polyacrylamide electrophoresis, did not recognize actin. Organism-specific actin antigens are not likely to be responsible for the development of smooth muscle antibodies during acute M. pneumoniae infection.

Expression and translocation of the chlamydial major outer membrane protein in Escherichia coli

The entire gene encoding the major outer membrane protein (MOMP) from Chlamydia psittaci strain GPIC has been cloned and expressed in Escherichia coli . A tightly regulated T7 promoter is used to control expression of the protein in Escherichia coli. Upon induction of expression, the precursor (pre-MOMP) is synthesized in the cell. This is followed by the appearance of a lower molecular weight protein that comigrates with mature MOMP from chlamydial elementary bodies by both one-dimensional sodium dodecyl sulfate polyacrylamide gel electrophoresis and two-dimensional gel electrophoresis. When E. coli cells expressing MOMP are converted to spheroplasts and subjected to protease treatment, MOMP is quantitatively degraded while cytoplasmic pre-MOMP is protected from degradation. Whole cells subjected to the same protease treatment show no degradation of MOMP. Furthermore, MOMP is not detected in surface-labeling experiments using several MOMP-specific antibodies. These data indicate that pre-MOMP is translocated to the periplasmic space and processed but is not surface exposed in E. coli. Expression of MOMP in this system causes a significant reduction in cell viability. In addition, coexpression in E. coli of MOMP or a MOMP-PhoA fusion with various chaperone proteins does not alter the level of MOMP translocation.

Purification and Partial Amino Acid Sequence of the Major 70,000-Dalton Heat Shock Protein in Neurospora crassa

Experimental Mycology , 17, 362-367. The major heat shock protein of 70 kDa (hsp70) from heat-shocked mycelial extracts of Neurospora crassa was purified to near homogeneity employing DEAE anion-exchange chromatography followed by affinity chromatography on ATP-agarose. The isolated hsp70 migrates as a single band on one-dimensional sodium dodecyl sulfate polyacrylamide gels (SDS-PAGE), with a molecular mass of ∼69 kDa. On two-dimensional gels it is resolved into two polypeptides with isoelectric points in the acidic range of ∼pH 5.2. The first 53 amino terminal amino acids of the major protein were sequenced and compared with hsp70 of other species. The amino acids aspartic acid, arginine, and phenylalanine occur at positions 27, 28, and 44 (from the methionine terminus) in contrast to the main consensus sequence. These three differing amino acids are shared by yeast, and, in addition, the first two by Arabidopsis, petunia, and maize.

Identification and Intra-specific Heterogeneity of Campylobacter hyointestinalis Based on Numerical Analysis of Electrophoretic Protein Profiles

Sixty-six strains received as Campylobacter hyointestinalis, along with twelve type or reference strains of other Campylobacter species, were characterized using high-resolution one-dimensional sodium dodecyl sulfate polyacrylamide gel electrophoresis. The resulting protein patterns were analyzed numerically, by computer, and revealed that when only background patterns were analyzed, 57 of the 66 strains formed a single cluster which included the type strain of C. hyointestinalis, thus confirming their identity. This cluster could be divided into two sub-clusters (a and b) which could also be differentiated phenotypically. When the full patterns were analyzed, 17 electrophoretypes were recognized among the 57 C. hyointestinalis strains. Evidence for the co-existence of strains of different protein types in the digestive tract of a single animal is presented.

Anti-endothelial antibodies and coronary artery disease after cardiac transplantation.

Accelerated coronary artery disease is the most serious complication after cardiac transplantation. The disease has a multifactorial aetiology, with little agreement about the relative importance of the various risk factors. We have investigated the frequency of anti-endothelial antibodies against human umbilical vein endothelial cells by one-dimensional sodium dodecyl sulphate polyacrylamide gel electrophoresis and western blotting. Peptide-specific anti-endothelial antibodies were found in 15/21 heart transplant recipients with accelerated coronary artery disease, and 1/20 transplant patients who had not developed the disease. Positive immunofluorescence of patients' serum on frozen sections of coronary vessels confirmed the endothelial specificity of antibodies. These results provide evidence of an immune aetiology for transplant-associated coronary artery disease and could have important implications for its diagnosis and therapy.

Antiheart antibodies following open heart surgery: incidence and correlation with postpericardiotomy syndrome.

One-dimensional sodium dodecyl sulphate polyacrylamide gel electrophoresis of myocardial proteins followed by Western blotting is a sensitive method for the detection of antiheart antibodies after cardiac transplantation. In a previous study we found that the majority of patients made antiheart antibodies after cardiac transplantation. It is possible that these antibodies were formed in response to cardiac damage caused during the surgical procedure rather than being specific to the transplantation situation. In this study we have evaluated the role of open cardiac surgery in the formation of antiheart antibodies for the first 9 months of the postoperative period using the Western blotting technique and correlated that with the development of post-pericardiotomy syndrome. Only 25% (9/36) of patients showed any increase in the pre-existing level of antiheart antibodies or developed antiheart antibodies with new reactivities. None of the patients in the study developed manifestations specific for post-pericardiotomy syndrome during the period of follow-up. The results support the contention that the high incidence of antiheart antibodies formed after cardiac transplantation is due to a humoral immune response to the presence of alloantigens on the grafted heart rather than as a result of the surgical procedure itself.

The Actin Cytoskeleton in Hyphal Cells ofSclerotium Rolfsii

The distribution of the actin cytoskeleton was examined in hyphal tips of the plant pathogenic fungus Sclerotium rolfsii. Actin was visualized with light microscopy using immunofluorescence techniques and with ultrastructural immunocytochemistry. Computer-aided, three-dimensional reconstruction analysis of ultrathin serial sections and immunoblot analysis techniques were also employed in this study. Immunofluorescence data indicated that the majority of actin was localized in brightly fluorescent plaques that were principally localized in the first 10 to 12 gm of the hyphal tip. Focusing through the hyphal cell revealed that most plaques were positioned in the peripheral cytoplasmic regions. Actin microfilaments were not resolved at the light microscopic level; however, coarse actin fibers oriented parallel to the long hyphal axis were observed in subapical regions and in association with septa. A diffuse, amorphous cytoplasmic staining was present in the apical regions of the hypha except for that area occupied by the Spitzenk6rper. Ultrastructural immunocytochemistry and three-dimensional reconstruction analysis suggested that the fibrillar coating of filasomes contained actin and that filasomes represented the ultrastructural equivalent of actin plaques. Immunoblot analysis of total protein extracts separated by one-dimensional sodium dodecyl sulfate polyacrylamide gel electrophoresis identified two actin related proteins, one with a relative molecular mass of 43,000 and the other migrating with a slightly higher molecular mass of 47,500.

The effect of castration on the synthesis and secretion of proteins in 7,12-dimethylbenz(a)anthracene-induced rat mammary tumors.

The effect of castration on the incorporation of [35S]methionine into secreted proteins in 7,12-dimethylbenz(a)anthracene (DMBA)-induced rat mammary tumors, was investigated. Biopsy specimens were obtained from 19 tumors, 0, 24, 48, 72, and 96 h after castration. In 14 tumors, castration induced an increase in the incorporation (mean of 5-fold), reaching the maximal level after 24 h (3 tumors), 48 h (7 tumors), 72 h (3 tumors), and after 96 h (1 tumor). In three tumors castration did not alter the incorporation rate, while in two tumors incorporation declined immediately after castration. One-dimensional sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) analysis of the labeled secreted protein showed that castration did not decrease or increase significantly the incorporation of [35S]methionine into any of the major labeled proteins. tumor regression following hormonal deprivation is apparently preceded by an increased synthesis of secreted proteins. However, no qualitative differences in any major labeled proteins could be observed.

Molecular Techniques for Studying the Epidemiology of Infection byHelicobacter pylori

The efficacy of one-dimensional sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) of whole-cell protein patterns for fingerprinting isolates of Helicobacter pylori was assessed by means of computerized numerical analysis. Virtually all strains were found to have unique, stable, and reproducible protein profiles. The application of this technique to a collection of isolates from eight patients showed clearly that each harboured a distinct strain that was present before treatment and persisted after treatment. This suggests that relapse was due to recrudescence of the same strain rather than re-infection with a different strain. Minor differences in protein banding profiles within sets of isolates from the same patient were evident, and this was confirmed by means of both two-dimensional PAGE protein patterns and restriction endonuclease analysis of DNA on the same strains.

Numerical analysis of SDS–PAGE protein patterns ofSerratia marcescens: a comparison with other typing methods

Twenty-five cultures comprising 18 clinical isolates of Serratia marcescens from two hospitals, the type strain of S. marcescens, two reference strains of S. marinorubra, the type or a reference strain of three other Serratia species and a reference strain of undetermined species, were characterized by one-dimensional sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) of whole-cell proteins. The protein patterns were highly reproducible and were used as the basis of a numerical analysis which divided the clinical isolates into eight protein types. Comparison with O-serotyping indicated that the level of discrimination by SDS-PAGE was similar. As with O-serotyping, a secondary scheme, such as phage typing, is necessary to differentiate strains of the same protein type. We conclude that high-resolution SDS-PAGE of proteins provides an effective adjunct to other methods for typing isolates of S. marcescens.

Investigation of a pseudo-outbreak of ‘Pseudomonas thomasii’ in a special-care baby unit by numerical analysis of SDS–PAGE protein patterns

Forty-two cultures of pseudomonas comprising 28 clinical isolates from a pseudo-outbreak on a Special-Care Baby Unit and 14 reference strains, including 9 type strains, of various Pseudomonas species, were characterized by one-dimensional sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) of whole-cell proteins. The protein patterns were highly reproducible and were used as the basis for a numerical analysis which divided the strains into 9 phenons. Two of the 28 clinical isolates were identified by biochemical tests as P. pickettii and their identification was confirmed by SDS-PAGE as they fell in the same phenon as the type strain of the species. The remaining 26 isolates, which could not be identified on phenotypic tests, fell in the same phenon as three reference strains of 'P. thomasii'. The protein patterns provided the first clear evidence that P. pickettii and 'P. thomasii' were separate taxa and that the 'outbreak' was polymicrobial in origin, in line with the probable aqueous source of contamination. We conclude that high-resolution SDS-PAGE of proteins provides an effective method of identifying and differentiating pseudomonads, especially where this cannot be done adequately using conventional biochemical tests.

Characterization of Four Major Allergens of Hen Egg-White by IEF/SDS-PAGE Combined with Electrophoretic Transfer and IgE-Immunoautoradiography

The antigenic and allergenic epitope maps of the hen egg-white ovalbumin, ovomucoid, ovotransferrin and lysozyme were obtained using one-dimensional sodium dodecyl sulfate polyacrylamide gel electrophoresis and two-dimensional sodium dodecyl sulfate gel electrophoresis following isoelectric focusing. The isoelectric points of egg-white proteins were assigned by the isoelectric focusing technique. Two-dimensional electrophoresis following isoelectric focusing provided further information regarding epitope mapping. Furthermore, electrophoretic transfer of the proteins to nitrocellulose and subsequent immunoautoradiography, clearly demonstrated the allergenicity of these proteins. An important benefit of these methods was confirming that lysozyme bound strongly to IgE in all the human sera from egg-allergic individuals and that lysozyme, in addition to ovalbumin, ovomucoid and ovotransferrin, was one of the major allergens of hen egg-white.

Novel Phaseolin types in wild and cultivated common bean (Phaseolus vulgaris, Fabaceae)

Forty-one wild types and 41 cultivars of common bean (Phaseolus vulgaris) from Meso-and South America were screened for variability of phaseolin seed protein using one-dimensional sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS/PAGE) and two-dimensional isoelectric focusing SDS/PAGE. Wild accessions from the Andean region showed phaseolin types which had not been previously identified in wild material from that region. Other wild accessions from Argentina exhibited novel phaseolin patterns collectively designated as ‘J’ (‘Jujuy’) phaseolin types, and one accession from northern Peru exhibited a novel phaseolin type, the ‘I’ (‘Inca’) type. The ‘H’ and ‘C’ phaseolins, previously identified only in cultivars, were observed in several wild accessions from Argentina. Among cultivars, two minor variants of the ‘S’ phaseolin type were identified. The ‘Sb’ (‘S Brazil’) was characteristic of a limited number of cultivars from Brazil whereas the ‘Sd’ (‘S Durango 222’) predominated in cultivars of the Mexican central highlands. The distribution of the previously described ‘B’ phaseolin appeared to be larger than formerly known as it extended not only in Colombia but also in Central America. It is possible to correlate the ‘Sb’, ‘Sd’, and ‘B’ phaseolin types with certain agronomic traits.

Numerical analysis of sodium dodecyl sulphatepolyacrylamide gel electrophoretic protein patterns for the classification, identification and typing of medically important bacteria

A numerical analysis of one-dimensional sodium dodecyl sulphate polyacrylamide gel electrophoretic protein patterns of three separate bacterial groups is described. Similarity between patterns was estimated using a correlation coefficient and is represented diagramatically by the unweighted pair-group with arithmetic averages method of clustering. Protein patterns were scanned using a laser densitometer interfaced directly to a microcomputer, where calculations were performed semi-automatically with specially written software. A method for the normalization of patterns from different gels is utilized. Whole cell protein patterns clearly distinguished strains representing various groups of Achromobacter. Similarly the two biovars of the Group EF-4 bacteria could be separated after analysis of partial "background patterns" as could the different species of Providencia. In addition, whole patterns could be used to type the various strains of P. rettgeri. These examples serve to illustrate the power, versatility and potential of the technique when applied to problems of classification, identification and typing of bacteria of medical interest. Its advantages and disadvantages, when compared to other available electrophoretic methodologies, are discussed, as is the future application of such technology to the hospital laboratory.

Partial amino acid sequence determination of Bovine Corneal Protein 54 K (BCP 54)

The most abundant soluble protein of bovine cornea, BCP 54 (Bovine Corneal Protein, molecular weight 54 kD) was isolated and digested under both limited and complete digestion conditions with Staphylococcus aureus V8 protease. The fragments resulting from limited digestion were separated by one-dimensional sodium dodecyl sulfate polyacrylamide gel electrophoresis, transferred to a polyvinylidene difluoride membrane, visualized by Coomassie Blue staining, cut out, and submitted to N-terminal protein sequence analysis. Complete digestion fragments were separated on a Vydac C18 reverse-phase HPLC, collected, and concentrated prior to sequencing. Using this method, we obtained amino acid sequence data from three internal V8 protease derived fragments of BCP 54 and a number of HPLC fragments. Comparison of these amino acid sequences, corresponding to 30% of the BCP 54 molecule, to those sequences contained within release 22 of the National Biomedical Research Foundation Protein Identification Resource revealed no extended sequence similarity of known proteins to BCP54.

Human Vascular Endothelial Cells Express a Membrane Protein Complex Immunochemically Indistinguishable From the Platelet VLA-2 (Glycoprotein Ia-IIa) Complex

Endothelial cells express surface molecules that are involved in cell-matrix interaction, including the vitronectin receptor and the fibronectin receptor, both members of a family of cell adhesion receptors (integrins). Here we provide evidence that endothelial cells express a membrane molecule, indistinguishable from the platelet VLA-2 complex, which is a collagen receptor and a member of the integrin family. To identify this endothelial molecule, we have used a monoclonal antibody, CLB-10G11, which recognizes the VLA-2 complex from platelets. The molecule recognized by CLB-10G11 from endothelial cells was characterized as follows. (1) The monoclonal antibody precipitated two proteins from surface-labeled endothelial cells that corresponded to the platelet VLA-2 subunits (glycoprotein Ia and IIa) as judged by one-dimensional sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) and two-dimensional nonreduced/reduced SDS-PAGE. (2) Preclearing of endothelial cells with monoclonal antibody A-1A5, an antibody that is directed against the common VLA beta subunit, removed all the CLB-10G11-binding material. (3) Crossed immunoelectrophoresis revealed that CLB-10G11 recognizes a single precipitation arc from either platelets or endothelial cells. Analysis of these two cell types in one gel again revealed one precipitation arc. The antigen of either cell type, recognized by CLB-10G11 could be precipitated by either polyclonal antiplatelet or polyclonal antiendothelial cell antiserum. Hence, it appears that endothelial cells express at least three different surface molecules (the vitronectin receptor, the fibronectin receptor and a collagen receptor), which may play an important role in controlling the anchorage of endothelial cells to the extracellular matrix.
