Dissemination of information in event-based surveillance, a case study of Avian Influenza.

Event-Based Surveillance (EBS) tools, such as HealthMap and PADI-web, monitor online news reports and other unofficial sources, with the primary aim to provide timely information to users from health agencies on disease outbreaks occurring worldwide. In this work, we describe how outbreak-related information disseminates from a primary source, via a secondary source, to a definitive aggregator, an EBS tool, during the 2018/19 avian influenza season. We analysed 337 news items from the PADI-web and 115 news articles from HealthMap EBS tools reporting avian influenza outbreaks in birds worldwide between July 2018 and June 2019. We used the sources cited in the news to trace the path of each outbreak. We built a directed network with nodes representing the sources (characterised by type, specialisation, and geographical focus) and edges representing the flow of information. We calculated the degree as a centrality measure to determine the importance of the nodes in information dissemination. We analysed the role of the sources in early detection (detection of an event before its official notification) to the World Organisation for Animal Health (WOAH) and late detection. A total of 23% and 43% of the avian influenza outbreaks detected by the PADI-web and HealthMap, respectively, were shared on time before their notification. For both tools, national and local veterinary authorities were the primary sources of early detection. The early detection component mainly relied on the dissemination of nationally acknowledged events by online news and press agencies, bypassing international reporting to the WAOH. WOAH was the major secondary source for late detection, occupying a central position between national authorities and disseminator sources, such as online news. PADI-web and HealthMap were highly complementary in terms of detected sources, explaining why 90% of the events were detected by only one of the tools. We show that current EBS tools can provide timely outbreak-related information and priority news sources to improve digital disease surveillance.

Эпизоотическая обстановка по инфекционному ринотрахеиту крупного рогатого скота в Карагандинской области Республики Казахстан в 2021–2022 гг.

A comprehensive serological monitoring is currently underway in the Republic of Kazakhstan to detect the circulation of the infectious bovine rhinotracheitis pathogen. To conduct a full-fledged and resultful study, the principles of sampling size representativeness provision and mathematical calculations were observed. The sampling size of the total number of epizootological units included mainly the raions and settlements in which (or near which) infectious bovine rhinotracheitis cases had been previously recorded. The sampling size of livestock population included in the study was determined in accordance with the recommendations of the World Organization for Animal Health. Thus, the study covered 7 (out of 13) raions of the Karaganda Oblast in 2021 and 2022. The other 6 raions and cities of regional significance will be included in the research in 2023. The paper presents the results of infectious bovine rhinotracheitis retrospective analysis and own studies conducted in 2021–2022. Statistical analysis and graphical visualization of investigation results were performed using Statistica, Excel, and QGIS programs. It was established that the epizootic situation for this disease was unfavourable in the Karaganda Oblast in 2021–2022. The data and results of serological studies presented by the Veterinary Control and Surveillance Committee of the Ministry of Agriculture of the Republic of Kazakhstan confirm the circulation of the infectious bovine rhinotracheitis virus in the following raions of the Karaganda Oblast: Abaisky, Aktogaisky, Bukhar-Zhyrausky, Karkaralinsky, Nurinsky and Osakarovsky.

Effective Treatment of Lumpy Skin Disease by Serum Therapy

Lumpy skin disease is a World Organization for Animal Health (OIE) listed disease and causes high morbidity around the globe. It causes damage to hide and affects the fertility and productivity of domestic animals. This case report describes the clinical management of lumpy skin disease in a local Sahiwal bull. Bull with the complaint of nodular eruptions on different body parts was presented at the veterinary clinic of University of Veterinary and Animal Sciences, Lahore, Pakistan. A comprehensive medical examination indicated that the bull was feverish, with a rectal body temperature of 105°F and had a burst of small to large-sized circumscribed nodules on various body regions, including the neck area. The diagnosis of lumpy skin disease was verified based on the history, clinical observations, and ELISA results. The case was addressed with a mix of medication (20% oxytetracycline, 1% doramectin, and 5% mepyramine maleate) and serum treatment. The bull recovered and is now healthy.

Paratuberculosis Cases in the Livestock Industry in Nigeria: A Review

Paratuberculosis, also known as Johne’s (Yo’-ness) disease, is a significant bacterial disease of large and small ruminants, including other animals, caused by Mycobacterium avium subspecies paratuberculosis (MAP). Due to its impact on animal health and severe economic loss, the disease is recognized and reported in developed countries as a significant disease of livestock importance. The paper aims to review the current information about paratuberculosis in Nigeria. About 95 research articles were downloaded from various online journals databases such as Scopus, Google Scholar, Research Gate, PubMed, and CABI abstracts based on the following keywords: Johne’s disease, paratuberculosis, Africa, prevalence, Nigeria, report, occurrence, and Mycobacterium avium subspecies paratuberculosis. In addition to the perusal of organization databases, 60 research articles were used in writing this review article. Few cases and suspected cases of paraTB were reported in Nigeria by the World Organization for Animal Health (OIE). Despite growing global concerns, Johne's disease can be considered an unreported disease of livestock animals in Nigeria based on the knowledge gap and lack of reports about this significant disease of livestock animals. Studies need to be carried out to address the knowledge gaps before the disease becomes endemic and causes severe economic and public health menace in Nigeria and, by extension, Sub-Saharan Africa.

Prevalence of Infectious Hypodermal and Hematopoietic Necrosis Virus (IHHNV) in Farmed Procambarus clarkii of the Middle and Lower Reaches of the Yangtze River in China.

Procambarus clarkii is an important economic aquaculture species worldwide. Infectious hypodermal and hematopoietic necrosis virus (IHHNV) infects numerous crustacean hosts, including P. clarkii. However, there have been few reports on the prevalence of IHHNV in P. clarkii. In this study, 200 farmed P. clarkii were collected from Anhui, Jiangsu, Zhejiang, Hunan, Hubei, and Sichuan provinces in China. PCR detection was employed per the protocol by the World Organization for Animal Health (WOAH) to identify and detect the presence of IHHNV. The positive rate of IHHNV in different provinces ranged from 16.7 to 56.7%, and the overall IHHNV-positive rate was 38.5%. IHHNV strains isolated in this study related closely to infectious IHHNV and split into two major distinct branches. Besides, the IHHNV strains shared a high homology (93.4-99.4%). These findings suggest that a high prevalence of IHHNV was established in farmed P. clarkii in the middle and lower reaches of the Yangtze River.

Import Risk Analysis of the Decapod Iridescent Virus 1 (DIV1) Applied in South Korea: Qualitative Risk Review and Institutional Improvement Plans

Decapod iridescent virus 1 (DIV1) is a dangerous new virus that causes mortality to shrimp and crayfish. This virus has been reported in China, which is a major exporter of decapods to South Korea and its neighboring countries, Taiwan and Thailand. This virus can be regarded as a pathogen with a high risk of entry into Korea. Therefore, South Korea has been conducting emergency quarantine for DIV1 since 2020, and in this study, an import risk analysis was performed following the World Organization of Animal Health and domestic laws and regulations to present scientific grounds and validity for South Korea’s DIV1 emergency quarantine. Import risk analysis was performed by hazard identification, entry assessment, exposure assessment, and consequence assessment. As a result of the import risk analysis, it was confirmed that the current DIV1 quarantine can significantly reduce the import risk. However, it was also established that the management of prawns and polychaetes intended for animal feed or bait and currently not subjected to quarantine is also required. Additionally, two species of crabs that would meet the criteria for a completely susceptible species and two species of crayfish that required surveillance of the import amount or route of exposure were also identified. In conclusion, these uses and susceptible species were designated as quarantine targets for nonhuman consumption requiring risk management and import sanitary conditions to be performed before export and after import. Finally, alternatives to reduce the cost burden of stakeholders following the introduction of the system were also presented.

Regulations on the Use of Antibiotics in Livestock Production in South America: A Comparative Literature Analysis.

As a global health problem, antimicrobial resistance (AMR) crosses national borders, leading UN (United Nations) multilateral agencies to call for all countries to improve the stewardship of antibiotics in humans and animals. South American countries have changed their regulations regarding antibiotic use in livestock production. This literature review examines how far the five largest meat-producing countries in South America (Argentina, Brazil, Chile, Colombia, and Uruguay) have come in terms of the relevant legislation. Rules on market entry (marketing authorization and official distribution systems) are already set in all countries examined. Four countries do not allow growth promoters based on critically important antibiotics, and countries have also begun to set guidelines and minimum welfare and biosecurity requirements to reduce the therapeutic demand for antibiotics. Nonetheless, there are aspects related to the distribution, use, and disposal of antibiotics that need to be developed further. In conclusion, legislation in South American countries is moving towards the goals set by UN multilateral agencies, but more can be done. Differences between countries' rules and the gold standards set by the World Organization for Animal Health (OIE), World Health Organization (WHO), and Food and Agriculture Organization of the United Nations (FAO) reveal possible adaptations to the countries' realities. Further studies must examine compliance with the legislation already set and investigate other tools that can be used alongside legislation as a driving force to change stakeholder behaviour.

Resistance profiles and genotyping of extended-spectrum beta-lactamase (ESBL) -producing and non-ESBL-producing E. coli and Klebsiella from retail market fishes.

Studies on antimicrobial resistance (AMR) profiles and epidemiological affirmation for AMR transmission are limited in fisheries and aquaculture settings. Since 2015, based on Global Action Plan on AMR by World Health Organization (WHO) and World Organization for Animal Health (OIE), several initiatives have been under taken to enhance the knowledge, skills and capacity to establish AMR trends through surveillance and strengthening of epidemiological evidence. The focus of this study was to determine the prevalence of antimicrobial resistance (AMR), its resistance profiles and molecular characterization with respect to phylogroups, antimicrobial resistance genes (ARGs), virulence genes (VGs), quaternary ammonium compounds resistance (QAC) genes and plasmid typing in retail market fishes. Pulse field gel electrophoresis (PFGE) to understand the genetic lineage of the two most important Enterobacteriaceae members, E. coli and Klebsiella sp. was performed. 94 fish samples were collected from three different sites viz., Silagrant (S1), Garchuk (S2) and North Guwahati Town Committee (NGTC) Region (S3) in Guwahati, Assam. Out of the 113 microbial isolates from the fish samples, 45 (39.82%) were E. coli; 23 (20.35%) belonged to Klebsiella genus. Among E. coli, 48.88% (n=22) of the isolates were alerted by the BD Phoenix M50 instrument as ESBL, 15.55% (n=7) as PCP and 35.55% (n=16) as non-ESBL. E. coli (39.82%) was the most prevalent pathogen among the Enterobacteriaceae members screened and showed resistance to ampicillin (69%) followed by cefazoline (64%), cefotaxime (49%) and piperacillin (49%). In the present study, 66.66% of E. coli and 30.43% of Klebsiella sp. were categorized as multi drug resistance (MDR) bacteria. CTX-M-gp-1, with CTX-M-15 variant (47%), was the most widely circulating beta-lactamase gene, while other ESBL genes blaTEM (7%), blaSHV (2%) and blaOXA-1-like (2%) were also identified in E. coli. Out of the 23 isolates of Klebsiella, 14(60.86%) were ampicillin (AM)-resistant (11(47.82%) K. oxytoca, 3(13.04%) K. aerogenes), whereas 8(34.78%) isolates of K. oxytoca showed intermediate resistance to AM. All Klebsiella isolates were susceptible to AN, SCP, MEM and TZP, although two K. aerogenes were resistant to imipenem. DHA and LAT genes were detected, respectively, in 7(16%) and 1(2%) of the E. coli strains while a single K. oxytoca (4.34%) isolate carried MOX, DHA and blaCMY-2 genes. The fluoroquinolone resistance genes identified in E. coli included qnrB (71%), qnrS (84%), oqxB (73%) and aac(6)-Ib-cr (27%); however, in Klebsiella, these genes, respectively, had a prevalence of 87%, 26%, 74% and 9%. The E. coli isolates belonged to phylogroup A(47%), B1(33%) and D(14%). All of the 22(100%) ESBL E. coli had chromosome-mediated disinfectant resistance genes viz., ydgE, ydgF, sugE(c), mdfA while 82% of ESBL E. coli had emrE. Among the non-ESBL E. coli isolates, 87% of them showed the presence of ydgE, ydgF and sugE(c) genes, while 78% of the isolates had mdfA and 39% had emrE genes respectively. 59% of the ESBL and 26% of the non-ESBL E. coli had showed the presence of qacEΔ1. The sugE(p) was present in 27% of the ESBL-producing E. coli and in 9% of non-ESBL isolates. Out of the 3 ESBL-producing Klebsiella isolates, 2(66.66%) K. oxytoca isolates were found harboring plasmid-mediated qacEΔ1 gene while one (33.33%) K. oxytoca isolate had sugE(p) gene. IncFI was the most prevalent plasmid type detected in the isolates studied, with A/C (18%), P (14%), X, Y (9% each) and I1-Iγ (14%, 4%). 50% (n=11) of the ESBL and 17% (n=4) of the non-ESBL E. coli isolates harboured IncFIB and 45% (n=10) ESBL and one (4.34%) non-ESBL E. coli isolates harboured IncFIA. Dominance of E. coli over other Enterobacterales and diverse phylogenetic profiles of E. coli and Klebsiella sp. suggests the possibility of contamination and this may be due to compromised hygienic practices along the supply chain and contamination of aquatic ecosystem. Continuous surveillance in domestic markets must be a priority in addressing antimicrobial resistance in fishery settings and to identify any unwarranted epidemic clones of E. coli and Klebsiella that can challenge public health sector.

Infectious Bronchitis Virus (Gammacoronavirus) in Poultry: Genomic Architecture, Post-Translational Modifications, and Structural Motifs

Infectious bronchitis virus (IBV) is an avian coronavirus (CoV) that belongs to the genus Gammacoronavirus and has been listed as an important disease by the World Organization for Animal Health (WOAH). It causes highly contagious respiratory, reproductive, and renal diseases in commercial poultry farms. Multiple IBV serotypes and genotypes have been identified in many countries and many detected variants do not provide cross-protection against infection, resulting in repeated outbreaks and significant economic losses worldwide. In addition, the high genetic mutations and recombination events in the prominent genomic regions of IBV, particularly in the spike glycoprotein (S) and nucleocapsid (N) proteins, are directly involved in the evolutionary processes of IBV and lead to increased pathogenicity and tissue tropism. The characterization of the different genotypes and the relationship between the structure, function, post-translational modifications (PTMs), and structural motifs will elucidate the mechanisms that promote replication and pathogenicity and affect the host’s immune response during infection. In this review, we discuss the molecular features of various IBV genes and proteins that contribute to the infection process. We also highlight the common PTMs and structural motifs that occur during protein synthesis and are essential components of IBV ecology.

Detection and identification of foot-and-mouth disease O/ME-SA/Ind-2001 virus lineage, Indonesia, 2022

ABSTRACT Foot-and-mouth disease (FMD) is a contagious disease affecting cloven-hoofed animals, which has been well-documented as one of the major animal diseases that causes severe economic loss in livestock sectors. The disease is endemic in many countries, particularly in Asia and Africa. Indonesia has been declared a disease-free country since 1986, and the World Organization of Animal Health (WOAH/OIE) recognized Indonesia as an FMD-free country without vaccination in 1990. However, the FMD virus was detected in many disease outbreaks in cattle and goats in Indonesia in May 2022. This study reports the detection and identification of FMD serotype O viruses in Indonesia. Although these viruses appeared to belong to the ME-SA/Ind-2001e lineage, they formed a unique cluster with 95.3% average nucleotide sequence similarity of the FMD VP1 gene to Ind-2001e viruses from other Asia countries. The illegal trade of live animals from endemic areas in Southeast Asia is one of the possible routes regarding the incursion of FMD in Indonesia, however, it requires further investigation.

Efficacy of a plant-produced infectious bronchitis virus-like particle vaccine in specific pathogen-free chickens

Infectious bronchitis (IB) Gammacoronavirus causes a highly contagious respiratory disease in chickens that is listed by the World Organisation for Animal Health (WOAH). Its high mutation ability has resulted in numerous variants against which the commercially available live or recombinant vaccines singly offer limited protection. Agrobacterium-mediated transient expression in Nicotiana benthamiana (tobacco) plants was used here to produce a virus-like particle (VLP) vaccine expressing a modified full-length IBV spike (S) protein of a QX-like IB variant. In a challenge study with the homologous live IB QX-like virus, VLP-vaccinated birds produced S protein-specific antibodies comparable to those produced by live-vaccinated birds seroconverting with mean geometric titers of 6.8 and 7.2 log2, respectively. The VLP-vaccinated birds had reduced oropharyngeal and cloacal viral shedding compared to an unvaccinated challenged control and were more protected against tracheal ciliostasis than the live-vaccinated birds. While the results appeared similar, plant-produced IB VLPs are safer, more affordable, easier to produce and update to antigenically match any emerging IB variant, making them a more suitable alternative to IBV control than live-attenuated vaccines.

‘One Health’ Approach to Infectious Diseases and Prevention of Antimicrobial Resistance: A Review

This review paper aims to provide an understanding on current concepts of ‘One Health’ approach in the field of public health with a special focus on infectious diseases and prevention of antimicrobial resistance. 'One Health' is an approach to designing and implementing programmes, policies, legislation, and research in which multiple sectors communicate and work together to achieve better public health outcomes. The scope of ‘One Health’ includes zoonotic diseases, antimicrobial resistance, food safety and security, vector-borne diseases that come from insect bites or animals, environmental contamination, and other health threats shared by people, animals, and the environment. The World Health Organization (WHO) is working closely with the Food and Agriculture Organization of the United Nations (FAO) and the World Organization for Animal Health (OIE) to promote multi-sectoral responses to food safety hazards, risks from zoonoses, and other public health threats at the human-animal-ecosystem interface and provide guidance on how to reduce those risks for a better living in our planet Earth. Our medical education curriculum should also offer a first exposure to both the concepts of ‘One Health’ and the collaborative processes required to manage issues associated with human, animal, and environmental health. Surveillance and control of Trypanosoma evansi in the canary Islands: A descriptive analysis

This study examines the occurrence of Surra, a disease caused by Trypanosoma evansi, in camels in the Canary Islands. The 1997 detection of T. evansi in camels in the Canary Islands led to the implementation of an initial control program, resulting in a decrease in prevalence. Following an outbreak in 2014, and due to the impossibility of eradicating it using the conventional measures, a lazaret was set up to separate positive and suspicious animals, in addition to the control measures previously implemented. Stomoxys calcitrans was the only vector captured, and no other animals tested were found to be positive for T. evansi. In November 2019, the last camels that tested serologically positive were detected; however, since February 2018, no camels that tested positive for PCR have been found in the farms were the outbreak was detected, suggesting that the sanitary measures implemented are adequate. The duration of the outbreak control and potential eradication for the disease has yet to be established. This study provides evidence to facilitate the control of African Animal Trypanosomosis in endemic areas of the world, which may contribute to revise the World Organization for Animal Health (WOAH) protocol to implement recommendations of surveillance and control strategies for animal Trypanosomosis in camels.

A quadruple fluorescence quantitative PCR method for the identification of wild strains of african swine fever and gene-deficient strains

BackgroundOriginating in Africa, African swine fever (ASF) was introduced to China in 2018. This acute and highly virulent infectious disease affects domestic pigs. The World Organization for Animal Health has listed it as a statutory reportable disease, and China has listed it as a category A infectious disease.MethodsPrimers and probes were designed for four ASFV genes (B646L, EP402R, MGF505-3R, and A137R). The primers/probes were highly conserved compared with the gene sequences of 21 ASFV strains.ResultsAfter optimization, the calibration curve showed good linearity (R2 > 0.99), the minimum concentration of positive plasmids that could be detected was 50 copies/µL, and the minimum viral load detection limit was 102 HAD50/mL. Furthermore, quadruple quantitative polymerase chain reaction (qPCR) with nucleic acids from three porcine-derived DNA viruses and cDNAs from eight RNA viruses did not show amplification curves, indicating that the method was specific. In addition, 1 × 106, 1 × 105, and 1 × 104 copies/µL of mixed plasmids were used for the quadruple qPCR; the coefficient of variation for triplicate determination between groups was < 2%, indicating the method was reproducible.ConclusionsThe results obtained by testing clinical samples containing detectable EP402R, MGF505-3R, and A137R strains with different combinations of gene deletions were as expected. Therefore, the established quadruple qPCR method was validated for the molecular diagnosis of ASF using gene-deleted ASFV strains.

Assessment of Peste Des Petits Ruminants (PPR) in Turkey Between 2017-2019

Peste des petits ruminants (PPR) is a viral disease affecting small ruminants and causes significant agricultural damage. The World Organization for Animal Health (OIE) has classified PPR as an animal disease that must be reported immediately. In this study was aimed to understand of spatial and seasonal epidemiology of PPR in Turkey and provide information by using Geographical Information Systems (GIS). Descriptive and analytical statistics were used in the study, logistic regression analysis was applied to assess the effect of season and species factors on PPR outbreaks occurring in Turkey. Data were obtained from the World Animal Health Information System (WAHIS) database of OIE. Between 2017-2019, 337 outbreaks, 18.467 cases and 11.526 deaths were reported 57 provinces of Turkey and the highest number of PPR outbreaks, cases and deaths were reported in The Central Anatolia Region. On time-wise examination, the PPR risk-areas revealed changes with different levels of endemicity. The Central Anatolia Region were the top outbreaks reported region during 2017-2018, whereas The Marmara Region reported more outbreaks during 2019. PPR outbreaks occurred throughout the year in all seasons, but were most frequently encountered during the winter season (December to February) and the results showed that seasonal and species factors are important in the spread of the disease. PPR has widely distributed throughout the country, and the epidemiological picture suggests that the disease has become an endemic infection in the country. The spatial and seasonal distribution of PPR in Turkey gives useful information on hotspot areas, allowing managers to make informed decisions about prevention and control in different parts of the country. In accordance with the PPR Global Control and Eradication Strategy, the study also outlines when and where extensive surveillance, vaccination, and biosecurity measures are required for the control and eradication of the disease in Turkey.

In-house developed Bacillus anthracis-specific synthetic peptide-based latex agglutination test (LAT) assay was comparatively evaluated with World Organisation for Animal Health (WOAH)-recommended polymerase chain reaction (PCR)/real-time PCR (qPCR) methods for the screening of B. anthracis spores from the soil to provide a simple, rapid, and economical immunodiagnostic test for field application.

Contagious Bovine Pleuropneumonia: A Passage to India.

The World Organization for Animal Health (WOAH)-listed contagious bovine pleuropneumonia (CBPP) emerged first in Europe and then spread to Eastern Asia, including Japan and China, from the Northern Territories of Australia at the end of the 19th century. Its route to India, however, is less well known as there is little evidence for large importations of cattle from Australia. The lack of accurate diagnostic tests at this time meant veterinary authorities relied solely on clinical and pathological signs, many of which were non-specific. Consequently, any diagnoses of CBPP reported in the early 20th century must be viewed with caution. More convincing reports of CBPP confirmed by laboratory tests were made in the 1930s and 1940s in the Indian state of Assam. Eradication campaigns began in the 1940s with immunizations of live attenuated vaccines and then more comprehensively in the 1950s and 1960s, supplemented with serological screening and the establishment of quarantine centres at international borders. The last case of CBPP, reported to WOAH, was seen in 1990, but the launch of a new awareness campaign in Assam in 2002 and recent reports of the disease in Pakistan suggests the disease has persisted in the Indian subcontinent well into the 21st century.

Coreceptor AXL Facilitates African Swine Fever Virus Entry via Apoptotic Mimicry.

African swine fever (ASF) is an acute and hemorrhagic infectious disease caused by African swine fever virus (ASFV), which is listed as an animal epidemic disease that must be reported by The World Organization for Animal Health and that causes serious economic losses to China and even the whole world. Currently, the entry mechanism of ASFV is not fully understood. Especially in the early stages of virus entry, the host factors required for ASFV entry have not yet been identified and characterized. In this study, we demonstrated that ASFV externalized phosphatidylserine (PS) on the envelope functioned as viral apoptotic mimicry, which interacts with AXL, a tyrosine kinase receptor, to mediate ASFV entry into porcine alveolar macrophages (PAMs). We found that AXL was the most pronounced phosphatidylserine receptor (PSR) affecting ASFV entry in PAMs by RNA interference screening. Knockout AXL gene expression remarkably decreased ASFV internalization and replication in MA104 cells. Furthermore, the antibody against AXL extracellular domains effectively inhibited the ASFV entry. Consistent with these results, the deletion of the intracellular kinase domain of AXL and the treatment of the AXL inhibitor, R428, significantly inhibited the internalization of ASFV. Mechanistically, AXL facilitated the internalization of ASFV virions via macropinocytosis. Collectively, we provide evidence that AXL is a coreceptor for ASFV entry into PAMs, which expands our knowledge of ASFV entry and provides a theoretical basis for identifying new antiviral targets. IMPORTANCE African swine fever (ASF) is a highly contagious infectious disease caused by the ASF virus (ASFV), with a mortality rate of up to 100%. ASFV has caused huge economic losses to pig farming worldwide. Specific cellular surface receptors are considered crucial determinants of ASFV tropism. However, the host factors required for ASFV entry have not yet been identified, and the molecular mechanism of its entry remains unclear. Here, we found that ASFV utilized phosphatidylserine (PS) on the surface of virions to masquerade as apoptotic mimicry and facilitated virus entry by interacting with host factor AXL. We found that knockout of AXL remarkably decreased ASFV internalization and replication. The antibody against AXL extracellular domains and AXL inhibitor R428 significantly inhibited the internalization of ASFV via macropinocytosis. The current work deepens our understanding of ASFV entry and provides clues for the development of antiviral drugs to control ASFV infection.
