Evaluation of Polypeptide Complexes from Marine Sources with Relevance in Initiation and Propagation Mechanisms of Osteoarticular Dysfunctions

The aim of the present study was to obtain a protein fraction rich in enzymes with proteolytic activity such as serine proteases, chymotrypsins and cysteine proteases and to investigate preliminary pharmacological effects with therapeutic potential in osteoarticular diseases. The processing of marine raw material by applying established and optimized sequences of operations (selective precipitation followed by fractionation by FPLC) led to well-defined protein fractions rich in proteolytic enzymes. To evaluate the biological effect of the isolated polypeptide fractions, an in vitro model on normal human osteoblast (HOB) and osteoarthritic (HOB-OA) cell lines was carried out in which the modulation of cell proliferation under the action of the polypeptide complexes was followed.

Preparation, Optimisation, and In Vitro Evaluation of [18F]AlF-NOTA-Pamidronic Acid for Bone Imaging PET.

[18F]sodium fluoride ([18F]NaF) is recognised to be superior to [99mTc]-methyl diphosphate ([99mTc]Tc-MDP) and 2-deoxy-2-[18F]fluoro-D-glucose ([18F]FDG) in bone imaging. However, there is concern that [18F]NaF uptake is not cancer-specific, leading to a higher number of false-positive interpretations. Therefore, in this work, [18F]AlF-NOTA-pamidronic acid was prepared, optimised, and tested for its in vitro uptake. NOTA-pamidronic acid was prepared by an N-Hydroxysuccinimide (NHS) ester strategy and validated by liquid chromatography-mass spectrometry analysis (LC-MS/MS). Radiolabeling of [18F]AlF-NOTA-pamidronic acid was optimised, and it was ensured that all quality control analysis requirements for the radiopharmaceuticals were met prior to the in vitro cell uptake studies. NOTA-pamidronic acid was successfully prepared and radiolabeled with 18F. The radiolabel was prepared in a 1:1 molar ratio of aluminium chloride (AlCl3) to NOTA-pamidronic acid and heated at 100 °C for 15 min in the presence of 50% ethanol (v/v), which proved to be optimal. The preliminary in vitro results of the binding of the hydroxyapatite showed that [18F]AlF-NOTA-pamidronic acid was as sensitive as [18F]sodium fluoride ([18F]NaF). Normal human osteoblast cell lines (hFOB 1.19) and human osteosarcoma cell lines (Saos-2) were used for the in vitro cellular uptake studies. It was found that [18F]NaF was higher in both cell lines, but [18F]AlF-NOTA-pamidronic acid showed promising cellular uptake in Saos-2. The preliminary results suggest that further preclinical studies of [18F]AlF-NOTA-pamidronic acid are needed before it is transferred to clinical research.

MiR-382-5p’s Function as a Suppressor in Osteosarcoma (OS) by Targeting PDPK1

ABSTRACT miR-382-5p engages in development of osteosarcoma (OS). However, the regulatory system of miR-382-5p in osteosarcoma remains to be revealed. This research studied the interplay between PDPK1 and miR- 382-5p in OS. RT-PCR was used to evaluate miR-382-5p and PDPK1 expression in OS cells and normal human osteoblast cells. Dual-luciferase reporter assay validated PDPK1 as a miR-382-5p target. CCK-8 evaluated the cell viability. Flow cytometric method determined cell apoptosis rate. Transwell and Scratch assays estimated the cell metastasis. miR-382-5p was inhibited in OS cells. Further functional results showed miR-382-5p upregulation reduced cell viability, and mobility by mediating PDPK1 in OS cells

Tannic acid enhances cisplatin effect on cell proliferation and apoptosis of human osteosarcoma cell line (U2OS).

The increase in cases of chemoresistance of cisplatin for osteosarcoma treatment has called for the need to establish a new treatment regime. Tannic acid (TA) possesses a potent antiproliferative effect against various cancers. Therefore, this study investigated the effect of TA combined with cisplatin on human osteosarcoma cell lines (U2OS). MTT assay was used to determine the half-maximal inhibitory concentration (IC50), while the combination index (CI) value was utilized to analyze the interaction within each combination. The antiproliferative effect of the treatment was evaluated by trypan blue exclusion assay. The morphological changes of cells were observed under a phase-contrast inverted microscope. The nuclear morphology and percentage of apoptosis cells were evaluated by using the Hoechst 33258 staining and annexin V/PI assay, respectively. The U2OS cells showed cytotoxic effect when treated with TA and cisplatin, with IC50 at 4.47µg/mL and 16.25µg/mL, respectively. The TA demonstrated no significant inhibition effect on the normal human fetal osteoblast cells (hFOB 1.19); yet, interestingly, a potent proliferative effect was indicated. Synergistic interaction was triggered when TA was combined with cisplatin at percentage ratios of 90:10 and 85:15. Meanwhile, antagonistic interaction was induced in the combination at percentage ratios of 75:25 and 50:50. On the other hand, a significant antiproliferative effect with prominent morphological alteration was detected in the cells treated with a combination of TA and cisplatin at the percentage ratio of 90:10. Additionally, combination-treated cells demonstrated the highest percentage of apoptosis cells, with distinct chromosomal condensation, nuclear fragmentation, reduction of nuclear volume, and notable apoptotic body. Therefore, there is a high potential for the inclusion of TA in the cisplatin-based chemotherapeutic regimen of osteosarcoma.

Synthesis of Gold Nanoparticles by Using Green Machinery: Characterization and In Vitro Toxicity.

Green synthesis of gold nanoparticles (GNPs) with plant extracts has gained considerable interest in the field of biomedicine. Recently, the bioreduction nature of herbal extracts has helped to synthesize spherical GNPs of different potential from gold salt. In this study, a fast ecofriendly method was adopted for the synthesis of GNPs using fresh peel (aqueous) extracts of Benincasa hispida, which acted as reducing and stabilizing agents. The biosynthesized GNPs were characterized by UV–VIS and Fourier transform infrared spectroscopy, transmission electron microscopy (TEM), and dynamic light scattering. In addition, the in vitro antibacterial and anticancer activities of synthesized GNPs were investigated. The formation of gold nanoparticles was confirmed by the existence of a sharp absorption peak at 520 nm, corresponding to the surface plasmon resonance (SPR) band of the GNPs. TEM analysis revealed that the prepared GNPs were spherical in shape and had an average particle size of 22.18 ± 2 nm. Most importantly, the synthesized GNPs exhibited considerable antibacterial activity against different Gram-positive and Gram-negative bacteria. Furthermore, the biosynthesized GNPs exerted remarkable in vitro cytotoxicity against human cervical cancer cell line, while sparing normal human primary osteoblast cells. Such cytotoxic effect was attributed to the increased production of reactive oxygen species (ROS) that contributed to the damage of HeLa cells. Collectively, peel extracts of B. hispida can be efficiently used for the synthesis of GNPs, which can be adopted as a natural source of antimicrobial and anticancer agent.

Key Properties of a Bioactive Ag-SiO2/TiO2 Coating on NiTi Shape Memory Alloy as Necessary at the Development of a New Class of Biomedical Materials.

Recent years have seen the dynamic development of methods for functionalizing the surface of implants using biomaterials that can mimic the physical and mechanical nature of native tissue, prevent the formation of bacterial biofilm, promote osteoconduction, and have the ability to sustain cell proliferation. One of the concepts for achieving this goal, which is presented in this work, is to functionalize the surface of NiTi shape memory alloy by an atypical glass-like nanocomposite that consists of SiO2-TiO2 with silver nanoparticles. However, determining the potential medical uses of bio(nano)coating prepared in this way requires an analysis of its surface roughness, tribology, or wettability, especially in the context of the commonly used reference coat-forming hydroxyapatite (HAp). According to our results, the surface roughness ranged between (112 ± 3) nm (Ag-SiO2)—(141 ± 5) nm (HAp), the water contact angle was in the range (74.8 ± 1.6)° (Ag-SiO2)—(70.6 ± 1.2)° (HAp), while the surface free energy was in the range of 45.4 mJ/m2 (Ag-SiO2)—46.8 mJ/m2 (HAp). The adhesive force and friction coefficient were determined to be 1.04 (Ag-SiO2)—1.14 (HAp) and 0.247 ± 0.012 (Ag-SiO2) and 0.397 ± 0.034 (HAp), respectively. The chemical data showed that the release of the metal, mainly Ni from the covered NiTi substrate or Ag from Ag-SiO2 coating had a negligible effect. It was revealed that the NiTi alloy that was coated with Ag-SiO2 did not favor the formation of E. coli or S. aureus biofilm compared to the HAp-coated alloy. Moreover, both approaches to surface functionalization indicated good viability of the normal human dermal fibroblast and osteoblast cells and confirmed the high osteoconductive features of the biomaterial. The similarities of both types of coat-forming materials indicate an excellent potential of the silver-silica composite as a new material for the functionalization of the surface of a biomaterial and the development of a new type of functionalized implants.

MicroRNA-208a-3p promotes osteosarcoma progression via targeting PTEN.

Osteosarcoma (OS) is a malignant bone tumor with a poor prognosis. Accumulated evidence has suggested that microRNAs (miRNAs/miRs) may function as either oncogenes or tumor suppressors, which are associated with tumorigenesis and the progression of different types of cancer. In the present study, the role of miR-208a-3p in OS was investigated. The expression levels of miR-208a-3p in OS tissues and cell lines were determined via reverse transcription-quantitative PCR (RT-qPCR). MTT and colony formation assays were performed to verify the proliferation rate of OS cells. In addition, the effects of miR-208a-3p on the migration and invasion of OS cells were revealed using wound-healing and Transwell assays, respectively. Furthermore, the association between miR-208a-3p and phosphatase and tensin homolog (PTEN) 3'-untranslated region was determined via luciferase reporter assays, western blot and RT-qPCR analysis. The results indicated that miR-208a-3p was upregulated in OS tissues and cell lines compared with adjacent normal tissues and human osteoblastic cells, respectively. miR-208a-3p overexpression promoted and miR-208a-3p knockdown inhibited OS cells proliferation and metastatic potential. Additionally, PTEN was validated as a direct target of miR-208a-3p and its expression was negatively associate with that of miR-208a-3p in OS cells. Taken together, these results may suggest that miR-208a-3p promoted OS cells proliferation and metastatic potential via targeting PTEN. Therefore, miR-208a-3p may be considered as a diagnostic biomarker for OS.

Hsa_circ_0008934 promotes the proliferation and migration of osteosarcoma cells by targeting miR-145-5p to enhance E2F3 expression.

To investigate the role of hsa_circ_0008934 in osteosarcoma and the molecular mechanism involved in the regulation of the occurrence and development of osteosarcoma METHODS: Differentially expressed circRNAs in the osteosarcoma cell lines SaOS2 and MG63 and in the normal human osteoblast cell line hFOB1.19 were identified via next-generation RNA sequencing. The expression and circular morphology of hsa_circ_0008934 were analyzed via quantitative real-time polymerase chain reaction (qRT-PCR) and RT-PCR analysis, respectively. Proliferation, apoptosis, cell cycle progression, migration, and invasion of SaOS2 and MG63 cells with hsa_circ_0008934 silencing or overexpression were assessed using the MTS method, colony formation assay, flow cytometry, and the transwell system, respectively. The subcellular distribution of hsa_circ_0008934 was revealed via fluorescence in situ hybridization. The binding of hsa_circ_0008934 with microRNAs was confirmed using the dual-luciferase reporter assay. The oncogenic roles of hsa_circ_0008934 in osteosarcoma were determined using an in vivo tumorigenesis assay with nude mice. qRT-PCR, western blotting, TUNEL assay, and immunohistochemistry (IHC) were used to detect the tumorigenicity of hsa_circ_0008934 in osteosarcoma cells. Many circRNAs were differentially expressed in SaOS2 and MG63 cells than in hFOB1.19 cells. Hsa_circ_0008934 expression was significantly elevated in SaOS2 and MG63 cells. Hsa_circ_0008934 silencing significantly reduced proliferation, enhanced apoptosis, blocked cell cycle progression, and impaired migration and invasion capacities of SaOS2 cells. Opposite cellular alterations were achieved by overexpressing hsa_circ_0008934 in MG63 cells. Hsa_circ_0008934 was mainly distributed in the cytosol and positively regulated E2F3 expression in osteosarcoma cells. In addition, it directly bound with miR-145-5p to repress E2F3 expression and enhanced the tumorigenesis of MG63 cells in nude mice. qRT-PCR revealed that the intracellular injection of hsa_circ_0008934 lentivirus resulted in hsa_circ_0008934 overexpression and miR-145-5p downregulation. Western blotting confirmed that E2F3 was upregulated. Moreover, the TUNEL assay showed that hsa_circ_0008934 overexpression inhibited the apoptosis of tumor cells. IHC detection revealed that the hsa_circ_0008934 overexpression could promote the expression of Ki67 and PCNA. Elevated hsa_circ_0008934 expression promotes the proliferation and migration of osteosarcoma cells by sponging miR-145-5p to enhance E2F3 expression.

In vitro bioactivity and osteoblast cell viability studies of hydroxyapatite-incorporated silica aerogel

Bioactivity and osteoblastic growth of hydroxyapatite (HA)-incorporated silica aerogels (SA) (HAESA) with different ratio of HA/SiO2 (0.1, 0.5, 1.0, and 1.3 HA/SiO2) were investigated. Bioactivity of HAESA was studied by immersing all samples in simulated body fluids (SBF) for 7 and 14 days. The recovered samples were then characterised using Fourier transform infrared spectroscopy, X-ray diffraction, field emission scanning electron microscopy combined with energy dispersive X-ray analyser (FESEM–EDX) and the analysis of phosphate loss in the solution. These samples were also investigated for their effect on cell viability and proliferation abilities against normal human osteoblast cells in vitro after 24 and 48 h of exposure times. The HA/SiO2 ratio affected its bioactivity in which the bioactivity increased to 0.5 (HAESA-0.5) and declined at higher ratios (1.0 and 1.3). A similar trend was observed for cell viability and proliferation assays. HAESA-0.5 (HA/SiO2 ratio of 0.5), which possesses the optimal characteristics of SA and HA, resulted in the highest osteoblastic growth due to the synergistic effects between SA and HA. Thus, HAESA-0.5 could be an alternative biomaterial compared to silica- and HA-based biomaterials for bone implant application due to its higher bioactivity in SBF with the improved performance of osteoblast growth. Hydroxyapatite-incorporated silica aerogel with different weight ratio of HA/SiO2 (HAESA) was studied for their bioactivity in simulated body fluid and in vitro osteoblastic growth.

MicroRNA-652 promotes cell proliferation and osteosarcoma invasion by directly targeting KLF9.

Previous studies have demonstrated that various microRNAs (miRNAs or miRs) are abnormally expressed in osteosarcoma (OS) and serve roles in its malignant development. An in-depth understanding of the specific roles of dysregulated miRNAs in OS may be important for cancer research and the identification of novel therapeutic targets. In the current study, reverse transcription-quantitative PCR was performed to determine miR-652 expression in OS tissues and cell lines. Cell Counting Kit-8 and Transwell invasion assays were used for assessing the effect of miR-652 on the proliferation and invasion of OS cells. Herein, miR-652 expression was assessed in OS and the effects and molecular mechanisms of miR-652 in OS cells were examined. The results revealed that miR-652 expression was significantly upregulated in OS tissues and cell lines compared with adjacent normal tissues and a normal human osteoblast cell line. Furthermore, miR-652 downregulation inhibited the proliferation and invasion of OS cells. miR-652 was also demonstrated to directly interact with the 3'-untranslated region of kruppel-like factor 9 (KLF9) and miR-652 negatively regulated KLF9 expression in OS cells. miR-652 and KLF9 mRNA levels were also revealed to be inversely correlated in OS tissues. Treatment with KLF9 small interfering RNA abolished the suppression of OS proliferation and invasion induced by miR-652 downregulation. miR-652 may serve an oncogenic role in OS cells by targeting KLF9 directly. The results also indicated that miR-652 may be an effective novel therapeutic target for the treatment of patients with OS.

Hypoxia-induced let-7f-5p/TARBP2 feedback loop regulates osteosarcoma cell proliferation and invasion by inhibiting the Wnt signaling pathway.

Osteosarcoma (OS) is the most common bone tumor in children and adolescents and is characterized by high metastatic and recurrence rates. In the past, it has been shown that microRNAs may play critical roles in hypoxia-related OS proliferation and invasion. However, the mechanisms by which OS cells acquire this malignant phenotype have remained largely unknown. In the present study, we report that let-7f-5p and TARBP2 were expressed in lower amounts in human OS cell lines when compared with the hFOB normal human osteoblastic cell line; however, both types of cells were repressed by hypoxia. let-7f-5p and TARBP2 significantly inhibited the proliferation and invasion of OS cells. Furthermore, TARBP2 as a downstream and functional target of let-7f-5p regulated the expression of let-7f-5p, and there was a regulatory feedback loop between let-7f-5p and TARBP2. This loop reduced the expression of let-7f-5p and TARBP2 in OS cells to a very low level, which was induced by hypoxia. Furthermore, the hypoxia-induced let-7f-5p/TARBP2 feedback loop contributed to activation of the Wnt signaling pathway. Taken together, our data clearly showed that the feedback loop between let-7f-5p and TARBP2 induced by the hypoxia-promoted OS cell malignant phenotype increased with activation of the Wnt signaling pathway.

Long noncoding RNA OR3A4 promotes the proliferation and invasion of osteosarcoma cells by sponging miR-1227-5p

BackgroundLong noncoding RNAs (lncRNAs) have been identified as key players in promoting tumourigenesis in osteosarcoma. LncRNA OR3A4 (OR3A4) has been reported as an oncogene in a number of tumours. However, the clinical value of OR3A4 in osteosarcoma and the role of OR3A4 in osteosarcoma progression are still unknown. MethodsThe expression levels of OR3A4 in the tumour tissue of osteosarcoma patients and osteosarcoma cell lines were detected by RT-PCR. Kaplan-Meier analysis and log-rank test were performed to evaluate the relationship between the level of OR3A4 expression and the prognosis of osteosarcoma patients. We investigated the association between the tissue expression levels of OR3A4 and different clinicopathological characteristics of osteosarcoma patients by χ2 tests. Bioinformatic databases and luciferase reporter assays were used to predict and validate the target microRNA of OR3A4. Finally, the role of OR3A4 in the proliferation and invasion of osteosarcoma cells was tested by MTT and Transwell assays, respectively. ResultsWe observed that the expression level of OR3A4 was upregulated in the tumour tissue of osteosarcoma patients (p < 0.001) and osteosarcoma cell lines (p < 0.01) compared with the normal adjacent tissue and a normal human foetal osteoblastic cell line, respectively. The survival curve revealed that patients with high expression levels of OR3A4 had lower overall survival. Increased OR3A4 expression in osteosarcoma patients was associated with distant metastasis (p = 0.02) and advanced clinical stage (p < 0.001). In addition, bioinformatics analysis and luciferase reporter assays verified the complementary binding between OR3A4 and miR-1227-5p. Furthermore, we found that OR3A4 acted as a miR-1227-5p “sponge” to modulate osteosarcoma cell proliferation and invasion via downregulation of miR-1227-5p. ConclusionOR3A4 promotes osteosarcoma cell proliferation and invasion by sponging miR-1227-5p, which might be related to the metastasis of osteosarcoma and could be used as a potential prognostic biomarker and therapeutic target in osteosarcoma.

MicroRNA-9-5p inhibits osteosarcoma cell promotion, metastasis and resistance to apoptosis via negatively targeting Grb2-associated binding protein 2

The study explores the inhibition effects of MicroRNAs in osteosarcoma, as a means of suggesting it as treatment for bone cancer. MicroRNAs (miRNAs) are a sort of noncoding RNA molecules that regulates gene expression by targeting mRNAs and play critical roles in tumor development. This study probed the effect of miR-9-5p on osteosarcoma development. Human osteosarcoma cell lines U2-OS, 143B, MG63 and HOS and normal human osteoblast cell line hFOB were cultivated and expression of miR-9-5p and Grb2-associated binding protein 2 (Gab2) measured. The binding of miR-9-5p and Gab2 was confirmed using a bio-information program and dual luciferase reporter gene assay. Loss-of-functions of miR-9-5p and Gab2 were performed to measure their roles in osteosarcoma cell proliferation, invasion, migration and resistance to death. Result showed high miR-9-5p expression and low Gab2 expression in osteosarcoma cells, particularly in U2-OS cells. miR-9-5p directly bound to the 3untranslated region of Gab2. Down-regulated miR-9-5p induced U2-OS cell proliferation, invasion and the resistance to death, while conversely, silenced Gab2 led to an opposite trend on U2-OS cell growth and metastasis. Moreover, co-effect of inhibited miR-9-5p and silenced Gab2 led to decreased cell proliferation but promoted cell apoptosis compared to inhibited miR-9-5p alone, while it led to enhanced cell proliferation and invasion, but reduced cell apoptosis compared to silenced Gab2 alone. To conclude, this study demonstrated that miR-9-5p could inhibit osteosarcoma cell proliferation, invasion, migration and resistance to death via negatively targeting Gab2.

Mechanisms of microRNA-19b on osteosarcoma cell proliferation and apoptosis by Targeting Krueppel-like factor 13

Objective To investigate the mechanisms of microRNA-19b on osteosarcoma cell proliferation and apoptosis by Targeting Krueppel-like factor 13 (KLF13). Methods The expression of miR-19b and KLF13 was detected by quantitative Real-time polymerase chain reaction in three human osteosarcoma cell lines (MG-63, US-OS and Saos-2) and one normal human osteoblast cell line (hFOB 1.19). KLF13 as a potential target of miR-19b was predicted by sequence analysis. The viability and apoptosis effects of miR-19 inhibitor on MG-63 cell was assessed by Cell Proliferation Assay and flow cytometry. Luciferase assay and Western blotting were used to examine the regulatory effect. Results The relative expression of miR-19b and KLF13 in osteosarcoma cells (665.66±39.74, 517.75±26.82, 564.88±41.20) were significantly higher than that in normal human osteoblast cell (16.09±4.80) (both P<0.05). Luciferase reporter vector system confirmed that KLF13 was one of the target genes of miR-19b. The absorbance (A) of experiment groups and control group was 0.424±0.021, 0.265±0.016 and 0.783±0.053 respectively by Western blotting. 3-(4, 5-Dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assay and flow cytometry demonstrated that miR-19b inhibitor significantly inhibited proliferation and promoted apoptosis of MG-63 cell (both P<0.01). Moreover, the anti-proliferation and pro-apoptotic effect by miR-19b inhibitor could be consolidate d by KLF13 knock down. Conclusion Suppression the expression of miR-19b can effectively inhibit the proliferation and promote apoptosis of osteosarcoma cells. miR-19b may become a new target for the regulation of gene expression in osteosarcoma. Enhanced antibacterial and corrosion resistance properties of Ag substituted hydroxyapatite/functionalized multiwall carbon nanotube nanocomposite coating on 316L stainless steel for biomedical application.

The present study reports the fabrication of silver substituted hydroxyapatite/functionalized multiwall carbon nanotube (Ag-HA/f-MWCNT) on 316L stainless steel (SS) implant by spray pyrolysis technique. XRD results show an enhanced crystallinity and crystallite sizes with increasing concentration of silver in HA/f-MWCNT. The vibrational spectral analysis revealed the presence of P-O stretching vibration of phosphate group (PO43-) in all the samples. The morphology of Ag substituted HA/f-MWCNT coatings revealed regular rod-like particles arranged in the form of sheet exhibiting slight variation in the size of the particle with increasing the Ag concentration. All the samples indicate the presence of calcium, phosphor, carbon, silver and oxygen constituents in the coating surface. The minimum inhibitory concentration of the nanocomposite decreased from 0.25 mg to 0.125 mg with the increase of Ag concentration, and AO/EB results confirmed the mode of cell distraction. The 1 and 3 wt% Ag-HA/f-MWCNT nanocomposite revealed less toxic effect to the normal human osteoblast cells. The corrosion efficiencies of the fabricated films in the stimulated body fluid reveal the increase in polarization resistance with a decrease in current density (icorr) from 3.9 to 3.5 μA due to the increase of Ag concentration. The estimated hemolysis rate for 1 and 3 wt% Ag substituted HA/f-MWCNT was less than 10%. Therefore, it can be concluded that 3 wt% Ag substituted HA/f-MWCNT coating on passivated 316L SS is nonhemolytic and most suited as a novel alternative to dental and orthopaedic implants.

Circular RNA 0001785 regulates the pathogenesis of osteosarcoma as a ceRNA by sponging miR-1200 to upregulate HOXB2

ABSTRACTCircular RNAs (circRNAs) are recently emerged to be promising therapeutic targets of tumors. Osteosarcoma is the most prevalent primary bone tumor and the third most prevalent cancer in children and adolescents. This study firstly analyzed circRNA microarray of osteosarcoma and selected circ-0001785 as the study object. We aimed to comprehensively investigate the expression pattern and biological function of circ-0001785 in the progression of osteosarcoma. Relative levels of circ-0001785 and miR-1200 in the normal human osteoblast cell line and osteosarcoma cell lines were determined. Bioinformatics analyses predicted the binding relationship between miR-1200 to HOXB2 and circ-0001785, while dual-luciferase reporter gene assay further verified this relationship. Flow cytometry and EdU assay were used for evaluating the regulatory effects of circ-0001785/miR-1200/HOXB2 axis on osteosarcoma cells. Consistent with the microarray analysis, circ-0001785 was highly expressed in osteosarcoma cell lines. Knockdown of circ-0001785 attenuated proliferative ability, but induced the apoptosis of osteosarcoma cells. Furthermore, we confirmed that circ-0001785 competitively bound to miR-1200, thus up-regulating its target gene HOXB2. Western blot analyses revealed that circ-0001785 regulated the PI3K/Akt signaling and Bcl-2 family pathway in osteosarcoma. In conclusion, circ-0001785 regulates the pathogenesis of osteosarcoma by sponging miR-1200 to up-regulate HOXB2 expression.

The lncRNA HOXA11-AS regulates Rab3D expression by sponging miR-125a-5p promoting metastasis of osteosarcoma.

Objective: Many studies have shown that long non-coding RNAs (lncRNAs) are closely related to various cancers. This study aims to explore the roles of lncRNA HOXA11-AS in the development and progression of osteosarcoma (OS).Methods: The expression levels of HOXA11-AS and miR-125a-5p in tumor tissues and the adjacent tissues were detected by RT-PCR method. The proliferation, migration and invasion of MG-63 and KHOS cells were determined.Results: It was found that HOXA11-AS expression levels in OS tissues and OS cell lines were higher than those in OS adjacent tissues and normal human osteoblast cell lines. The higher expression level of HOXA11-AS was positively correlated with more severe clinical stage, distant metastasis and poor prognosis of OS. Inhibition of HOXA11-AS expression could reduce metastasis and invasion of OS cell lines. In addition, HOXA11-AS was found to be an endogenous inhibitor of miR-125a-5p, it down regulated the expression level of miR-125a-5p, and this process could promote the expression of Rab3D, the target gene of miR-125a-5p.Conclusion: Our study elucidated the role of a new HOXA11-AS/miR-125a-5p/Rab3D regulatory pathway in promoting OS metastasis.

DEAD-Box Helicase 5 Interacts With Transcription Factor 12 and Promotes the Progression of Osteosarcoma by Stimulating Cell Cycle Progression.

Osteosarcoma (OS) is a common malignant primary bone tumor. Its mechanism of development and progression is poorly understood. Currently, there is no effective therapeutic regimens available for the treatment of OS. DEAD-box helicase 5 (DDX5) is involved in oncogenic processes. This study aimed to explore the role of DDX5 in the development and progression of OS and its relationship with transcription factor 12 (TCF12), which is as an important molecule of Wnt signaling pathway. We found that the expressions of DDX5 and TCF12 protein were significantly higher in OS patients tissues and in the MG63 cells than in the corresponding normal tissues and human osteoblast cell hFOB 1.19. Overexpressions of both DDX5 and TCF12 were associated with clinicopathological features and poor prognosis of OS patients. siRNA based knockdown of DDX5 inhibited the proliferation of MG63 cells as demonstrated by an in vitro MTS assay and 5-ethynyl-2-deoxyuridine DNA proliferation detection, and promoted apoptosis of MG63 cells measured by flow cytometry. In addition, DDX5 knockdown inhibited the MG63 cell migration and invasion on transwell assays. Further experiments showed that DDX5 knockdown not only inhibited the expression of TCF12 but also decreased the mRNA and protein levels of Cyclin E1, an important regulator of G1–S phase progression, suggesting that DDX5 was required for the entry of cells into S phase. Overexpression of TCF12 reversed the cell proliferation, migration and invasion in MG63 cells induced by DDX5 knockdown accompanied by the upregulation of Cyclin E1. Additionally, we observed that DDX5 interacted with TCF12 in both OS tissues and MG63 cells by Co-immunoprecipitation assays. Taken together, our study revealed that DDX5 interacts with TCF12 and promotes the progression of OS by stimulating cell cycle progression. Our results suggest that DDX5 and TCF12 could be potential biomarkers for the diagnosis and treatment of OS.

Osteoblast adhesion and proliferation on porous chitosan/polycaprolactone scaffolds for bone tissue engineering application

Tissue engineering is an evolving technique to reduce the limitations of the bone graft. It provides diversity to improve the healing process of bone fractures and defect by combining the use of 3D scaffolds, healing promoting factors, gene therapy and different drugs. Flexibility to use a different technique to fabricate scaffolds lead to the new insight into bone healing future. A bone graft is defined as promoted bone healing through osteogenesis, osteoinduction, and osteoconduction by the implanted material alone or with other materials. Ideal bone graft depends on several factors such as biomechanical characteristic, tissue viability, the morphological structure as well as biological characteristics. In this work, we have investigated adhesion, proliferation, and differentiation of poly (ɛ-caprolactone) / chitosan scaffolds with the incorporation of hydroxyapatite and tetracycline HCl on normal human osteoblast cells. Both of the polymers were blended without a cross-linking agent to form porous scaffolds by freeze-drying technique. From the results, it was observed that the compressive modulus increased from 4.0 MPa to 12.5 MPa and the yield strength increased from 0.48 MPa to 0.75 MPa for the PCL/CS scaffold and nHA/PCL/CS scaffold, respectively. Scanning electron microscopy study revealed that the cells successfully adhered to the surface of scaffolds after 24 hours incubation. Proliferation analysis exhibited the increasing trend of growth of cells. This study indicated that the scaffold fabricated using this technique was able to promote adhesion, proliferation, and differentiation of normal human osteoblast cells.

Long non-coding RNA XIST serves an oncogenic role in osteosarcoma by sponging miR-137.

The long non-coding RNA X inactive-specific transcript (XIST) has been implicated in certain human cancers, including osteosarcoma (OS), but the molecular mechanism of XIST underlying OS progression remains to be fully uncovered. In the present study, reverse transcription-quantitative polymerase chain reaction data demonstrated that XIST was significantly upregulated in OS tissues and cell lines (Saos-2, U2OS, HOS and MG63) compared with adjacent non-tumour tissues and normal human osteoblast cell line HFOB1.19. Bioinformatics analysis and luciferase reporter gene assay data demonstrated that XIST could directly target microRNA (miR)-137 and negatively regulate the expression of miR-137 in Saos-2 and U2OS cells. Furthermore, miR-137 was markedly downregulated in OS tissues and cell lines. An inverse association between XIST and miR-137 expression was observed in OS tissues. Knockdown of XIST caused a significant reduction in cell proliferation and invasion and suppressed matrix metalloproteinase (MMP2) and MMP9 protein levels in Saos-2 and U2OS cells. Furthermore, inhibition of miR-137 expression abolished the effects of XIST downregulation on the proliferation and invasion of OS cells. In summary, the present study suggests that XIST promotes OS cell proliferation and invasion by inhibition of miR-137 expression. Thus, XIST may be a potential therapeutic target for the treatment of OS.
