Fish as model systems for the study of vertebrate apoptosis

Apoptosis is a process of pivotal importance for multi-cellular organisms and due to its implication in the development of cancer and degenerative disease it is intensively studied in humans and mammalian model systems. Invertebrate models of apoptosis have been well-studied, especially in C. elegans and D. melanogaster, but as these are evolutionarily distant from mammals the relevance of findings for human research is sometimes limited. Presently, a non-mammalian vertebrate model for studying apoptosis is missing. However, in the past few years an increasing number of studies on cell death in fish have been published and thus new model systems may emerge. This review aims at highlighting the most important of these findings, showing similarities and dissimilarities between fish and mammals, and will suggest topics for future research. In addition, the outstanding usefulness of fish as research models will be pointed out, hoping to spark future research on this exciting, often underrated group of vertebrates.

Perchlorate and ethylenethiourea induce different histological and molecular alterations in a non-mammalian vertebrate model of thyroid goitrogenesis

Despite evidence for a conserved role of thyroid-stimulating hormone (TSH) in regulating vertebrate thyroid function, molecular data on thyroid responses to TSH are mainly limited to mammalian species. In this study, we examined histological and molecular changes in the thyroid of Xenopus laevis tadpoles during a 12-day treatment with 20mg/l perchlorate (PER) and 50mg/l ethylenethiourea (ETU). Inhibition of thyroid hormone (TH) synthesis by PER and ETU was evident from developmental retardation, reduced expression of TH-regulated genes and up-regulation of tshb-A mRNA. Thyroid histopathology revealed goiters with strikingly different follicular morphologies following PER and ETU treatment. Using real-time PCR, we analyzed thyroids sampled on day 12 for differential expression of 60 candidate genes. Further temporal analyses were performed for a subset of 14 genes. Relative to the control, PER and ETU treatment modulated the expression of 51 and 49 transcripts, respectively. Particularly genes related to TH synthesis and protein metabolism were similarly affected by PER and ETU. However, several genes were differentially expressed in PER- and ETU-treated tadpoles. Specifically, goiter formation in the PER treatment was associated with low expression of genes related to DNA replication but high expression of negative growth regulators. Results from this work provide for the first time a characterization of gene expression profiles during goitrogenesis in a non-mammalian vertebrate model. Overall, our data suggest that, in addition to TSH over-stimulation, further mechanisms related to the mode of goitrogen action contribute to the regulation of thyroid gene expression.

Non-mammalian vertebrate models and the endocannabinoid system: Relationships with gonadotropin-releasing hormone

Endocannabinoids, via cannabinoid receptors (CB1 and CB2), affect reproductive functions at both local and central level. Due to the high complexity of the endocannabinoid system, to the widespread distribution outside the nervous system and to the high degree of evolutionary conservation, a deep CB1 molecular characterization among species may be useful to elucidate the activity of endocannabinoids at multiple levels. In this review we report CB1 characterization in non-mammalian animal models and, in particular, in the anuran amphibian, the frog, Rana esculenta; we also describe its expression during the annual sexual cycle. Moreover, since reproductive functions are under control of gonadotropin releasing hormone (GnRH), cb1 mRNA and protein expression profile in the forebrain has been compared to those of GnRH-I, the mammalian form primarily involved in gonadotropin release.

Fishing for an ECG: a student-directed electrocardiographic laboratory using rainbow trout

Cardiac physiology is emphasized in many undergraduate physiology courses, but few nonmammalian vertebrate model systems exist that 1) can be studied fairly noninvasively, 2) are well suited for controlled experimentation, and 3) emphasize principles characteristic of the vertebrate heart. We have developed an inquiry-based undergraduate/graduate-level laboratory in cardiac physiology and electrocardiography using rainbow trout (Oncorhynchus mykiss Walbaum) and the BioPac MP30 data-acquisition system (other fish species and/or electrocardiographic recording devices can be substituted). This laboratory facilitates intensive study of vertebrate electrocardiograms (ECGs) under a variety of environmental and physiological perturbations and is ideal for use in multi-session, inquiry-based laboratory projects in animal physiology. Furthermore, students gain valuable experience in scientific inquiry, study design, following and/or developing scientific protocols, and animal care. This laboratory requires the ability to keep captive fish of at least 100 g and equipment to record ECGs. Departments meeting these requirements can adopt this technique at modest expense. Student enthusiasm and feedback were positive, and several students commented that the nonlethal methods used added to the laboratory's perceived value.

Glutamic acid decarboxylase 65, 67, and GABA-transaminase mRNA expression and total enzyme activity in the goldfish ( Carassius auratus) brain

GAD65 and GAD67 are the two major isoforms of the enzyme that converts glutamate into GABA in a single step reaction. Despite studies describing GAD65 and GAD67 mRNA expression in the mammalian brain, both GAD65 and GAD67 mRNA expression has not yet been fully described for a non-mammalian vertebrate model. Similarly, the expression patterns of GABA-T mRNA, the major enzyme involved in metabolizing GABA, have not been described for any vertebrate. In the present study, we utilized non-radioactive in situ hybridization to localize GAD65, GAD67, and GABA-T in the adult goldfish brain and complimented this with an in vitro assessment of total GAD and GABA-T enzyme activities. A partial fragment of goldfish GABA-T was cloned for a riboprobe that showed approximately 92% deduced amino acid identity to zebrafish GABA-T and 78% identity to human GABA-T. Transcripts for GAD65, GAD67, and GABA-T were detected throughout the brain and were detected largely in the medial and ventral regions of the telencephalon, nucleus preopticus, nucleus recessus lateralis of the hypothalamus, and Purkinje cell layer of the cerebellum. GAD65 mRNA was significantly more abundant in the nucleus recessus posterioris of the hypothalamus than GAD67 and GABA-T mRNA. Total GAD and GABA-T specific enzyme activity was highest in the hypothalamus and optic tectum and GABA-T activity was significantly higher than total GAD enzyme activity. Our results show that GAD65, GAD67, and GABA-T mRNAs are generally correlated with total GAD and GABA-T activity and all three transcripts have a largely overlapping mRNA distribution in the goldfish forebrain.

ACE2 orthologues in non-mammalian vertebrates ( Danio, Gallus, Fugu, Tetraodon and Xenopus)

Angiotensin-converting enzyme 2 (ACE2), a newly identified member in the renin–angiotensin system (RAS), acts as a negative regulator of ACE. It is mainly expressed in cardiac blood vessels and the tubular epithelia of kidneys and abnormal expression has been implicated in diabetes, hypertension and heart failure. The mechanism and physiological function of this zinc metallopeptidase in mammals are not yet fully understood. Non-mammalian vertebrate models offer attractive and simple alternatives that could facilitate the exploration of ACE2 function. In this paper we report the in silico analysis of Ace2 genes from the Gallus (chicken), Xenopus (frog), Fugu and Tetraodon (pufferfish) genome assembly databases, and from the Danio (zebrafish) cDNA library. Exon ambiguities of Danio and Xenopus Ace2s were resolved by RT-PCR and 3′RACE. Analyses of the exon–intron structures, alignment, phylogeny and hydrophilicity plots, together with the conserved synteny among these vertebrates, support the orthologous relationship between mammalian and non-mammalian ACE2s. The putative promoters of Ace2 from human, Tetraodon and Xenopus tropicalis drove the expression of enhanced green fluorescent protein (EGFP) specifically in the heart tissue of transgenic Xenopus thus making it a suitable model for future functional genomic studies. Additionally, the search for conserved cis-elements resulted in the discovery of WGATAR motifs in all the putative Ace2 promoters from 7 different animals, suggesting a possible role of GATA family transcriptional factors in regulating the expression of Ace2.

Estrogen-induced Akt-1 activity in the lizard (Podarcis s. sicula) testis

There are always more evidences indicating that 17beta-estradiol (E2) is to be necessary for normal male fertility. Here we report the expression of the most ubiquitously expressed member of the akt family of genes, akt1, in the lizard (Podarcis s. sicula) testis. We have used a nonmammalian vertebrate model (the lizard P. s. sicula) to investigate the regulation of the serine/threonine kinase Akt activity, implicated in the control of cell proliferation, survival, and metabolism, in the testis during the annual sexual cycle and to study whether E2 exerts a role in the spermatogenesis through Akt-1 activity. Immunocytochemistry analysis show that Akt-1 proteins are present in the spermatogonia (SPG), and spermatocytes (SPC), and spermatids (SPT). The annual E2 profile shows a progressive increase during the active spermatogenesis (from April to June) and a peak in the month of August (spermatogonial mitosis). In parallel, Akt-1 (molecular weight 60 kDa) are highly phosphorylated during the period of active spermatogenesis and in post-refractory period (August) compared with the winter stasis (from November to March). Present results demonstrate that E2 treatment induces the activation of Akt-1, and this effect is counteracted by the anti-estrogen ICI 182-780.

Estrogen receptor beta localization in the lizard (Podarcis s. sicula) testis.

There is increasing evidence that 17beta-estradiol is necessary for normal male fertility. The aim of the present study was to characterize estrogen receptor beta (ERbeta) expression in a non-mammalian vertebrate model, the lizard (Podarcis s. sicula) testis. Immunocytochemical analysis shows that ERbeta proteins are present among germ cells in the nucleus of the spermatogonia, in primary spermatocytes and spermatids. Western blot analysis with antibodies against the ERbeta gene product revealed an isoform with a specific weight of 55 kDa. In conclusion, the widespread expression of ERbeta in the Podarcis s. sicula testis is consistent with a role for estrogens in modulating spermatogenesis in the male.

Changes in JNK1 activity in the frog (Rana esculenta) testis.

Several lines of evidences support a key role of mitogen-activated protein kinases (MAPK) in male fertility. It has been used as a non-mammalian vertebrate model, the frog Rana esculenta, to investigate the regulation of Jun amino-terminal kinase 1 (JNK1) activity in the testis in two different periods of the annual reproductive cycle: winter stasis and breeding season. JNK1 protein was localized by immunohistochemistry in the cytoplasm and nucleus of spermatogonia (SPG) and spermatocytes (SPC) during all the year. Immunoprecipitation analysis of JNK1 clearly shows that this isoform presents a strong phosphorylation status (thr183/Tyr185) in the breeding period, in contrast with only very low activity during the winter stasis. Simultaneously, during the breeding period c-Jun protein is highly phosphorylated on Ser-63. These data suggest that the increase of JNK1 activity, with strong c-Jun phosphorylation (Ser-63) during the breeding period, could be the signal of increasing transcriptional activity in the frog testis.

Characterization of zebrafishmerlot/chablisas non-mammalian vertebrate models for severe congenital anemia due to protein 4.1 deficiency

The red blood cell membrane skeleton is an elaborate and organized network of structural proteins that interacts with the lipid bilayer and transmembrane proteins to maintain red blood cell morphology, membrane deformability and mechanical stability. A crucial component of red blood cell membrane skeleton is the erythroid specific protein 4.1R, which anchors the spectrin-actin based cytoskeleton to the plasma membrane. Qualitative and quantitative defects in protein 4.1R result in congenital red cell membrane disorders characterized by reduced cellular deformability and abnormal cell morphology. The zebrafish mutants merlot (mot) and chablis (cha) exhibit severe hemolytic anemia characterized by abnormal cell morphology and increased osmotic fragility. The phenotypic analysis of merlot indicates severe hemolysis of mutant red blood cells, consistent with the observed cardiomegaly, splenomegaly, elevated bilirubin levels and erythroid hyperplasia in the kidneys. The result of electron microscopic analysis demonstrates that mot red blood cells have membrane abnormalities and exhibit a severe loss of cortical membrane organization. Using positional cloning techniques and a candidate gene approach, we demonstrate that merlot and chablis are allelic and encode the zebrafish erythroid specific protein 4.1R. We show that mutant cDNAs from both alleles harbor nonsense point mutations, resulting in premature stop codons. This work presents merlot/chablis as the first characterized non-mammalian vertebrate models of hereditary anemia due to a defect in protein 4.1R integrity.

Annual profile of mitogen-activated protein kinase (extracellular signalregulated kinase 1 and 2) in the frog(Rana esculenta) testis

Using a non-mammalian vertebrate model, the frogRana esculenta, we have investigated the regulation of ERK1/2 activity in the testis during the annual sexual cycle. ERK1/2 proteins (molecular weight 44 and 42 kDa respectively) are present in the cytoplasm and nucleus of the primary (I) and secondary (II) spermatogonia (SPG), and in the nucleus of I spermatocytes (SPC). The annual plasma 17β-estradiol profile shows a progressive increase during the active spermatogenesis with a peak in the month of June. Concomitantly, ERK 1/2 are highly phosphorylated during the period of active spermatogenesis (from April to July) respect to the regressive period (September/October) and winter stasis (from November to March). In conclusion these data suggest that ERK1/2 activity have a role, possibly via 17β-estradiol activation, in the regulation of germinal epithelium progression.

C-fos activity in Rana esculenta testis: seasonal and estradiol-induced changes.

Estradiol-17beta (E2) is suspected to exert a role in the regulation of testicular activity. Using a nonmammalian vertebrate model (the frog, Rana esculenta), we have investigated whether c-fos activity is detectable in the testis during the annual sexual cycle and whether E2 exerts a regulatory role on spermatogenesis through fos activity. FOS protein is available in testicular nuclear extracts (about 60 kDa) and, surprisingly, also in cytosolic extracts (about 60, 80, and 100 kDa). Estradiol induces primary spermatogonia (ISPG) proliferation [this effect is counteracted by antiestrogens (Tamoxifen and ICI 182-780)] and FOS appearance in testicular cytosolic extracts as well as c-fos transcription. Also, this effect is counteracted by ICI 182-780. Interestingly, the number of FOS immunopositive nuclei of ISPG strongly increases after E2 treatment, whereas a great increase of immunopositivity in the cytoplasm of ISPG is observed with the contemporaneous treatment with antiestrogens. In conclusion, our results demonstrate that E2 induces ISPG multiplication in the frog, R. esculenta, and, for the first time in a vertebrate species, that it triggers c-fos activity in the testis. Moreover, E2 may be involved in mechanisms related to FOS transport in the nucleus of ISPG to induce the mitotic activity.

Nonmammalian vertebrate models in studies of brain-steroid interactions

Estrogen formation in brain and pituitary mediates certain androgen actions in central targets. Goldfish (Carassius auratus) and quail (Coturnix coturnix japonica) have been advantageous for studying the role of locally formed estrogen in autoregulating aromatization and in controlling estrogen receptor occupancy, androgen receptor levels, and behavioral expression. Data from these two experimental models reveal a molecular basis for androgen-estrogen synergism in neuroendocrine tissues and for alterations in androgen sensitivity/responsiveness. These mechanisms are essential components of seasonal reproduction in the test species and may have wider relevance for cyclicity in other vertebrates, including mammals.
