Effects of Acetogenins from Annona coriacea on the in Vitro Reactions of Photosynthesis.

Our search for candidates for photosynthesis inhibitors is allowing us to report the effect of two acetogenins identified in Annona coriacea Mart. leaves, ACG-A and ACG-B, a non-adjacent bis-THF and a mono-THF types, respectively. This is an important class of natural products which presents biological properties such as anticancer, neurotoxic, larvicidal and insecticidal. However, this is only the second report associated to its herbicidal activity. Their mechanisms of action on the light reactions of the photosynthesis were elucidated by polarographic techniques. Compounds inhibited the noncyclic electron transport on basal, phosphorylating, and uncoupled conditions from H2 O to methyl viologen (MV); therefore, they act as Hill reaction inhibitors. Studies on fluorescence of chlorophyll a (ChL a) indicated that they inhibited the acceptor side of PSII between P680 and PQ-pool, exactly as the commercial herbicide DCMU does.

Biological effects of the free and embedded metribuzin and tribenuron-methyl herbicides on various cultivated weed species

The present study addresses the herbicidal activity and biological effects of the metribuzin (MET) and tribenuron-methyl (TBM) herbicides used to control various weed species (Amaranthus retroflexus, Sinapis arvensis, and Leucanthemum maximum). The effects of the free herbicides and the herbicides embedded in granules of degradable polymer poly-3-hydroxybutyrate [P(3HB)] blended with birch wood flour were compared. Metribuzin, regardless of the form, caused 100% mortality of the three weeds by day 21. The herbicidal activity of tribenuron-methyl was lower than that of metribuzin, but the embedded TBM was superior to the free herbicide in the length and strength of its action on the weeds. Both metribuzin forms dramatically decreased the main parameters of fluorescence: maximum quantum yield of photosystem-II [Y(II)max], maximum quantum yield of non-photochemical quenching [Y(NPQ)max], and maximum rate of non-cyclic electron transport [ETRmax] and concentrations of chlorophyll a and b. The effect of the embedded TBM on the photosynthetic activity of the weeds was lower in the first two weeks of the growth of herbicide-treated plants but lasted longer than the effect of the free TBM and increased over time. Embedding of metribuzin in the matrix of degradable blend did not decrease its herbicidal activity.

Drought response of cowpea (Vigna unguiculata (L.) Walp.) landraces at leaf physiological and metabolite profile levels

Drought threatens the world’s food production, particularly in Sub Saharan Africa low external input and rain fed agricultural systems, where cowpea (Vigna unguiculata (L.) Walp.) is an important food crop. In the context of growing concerns regarding climate changes implications on water availability, this study aimed at 1) to evaluate the drought responses in cowpea landraces with contrasting drought tolerance levels (A55 – high sensitivity; A80 - mild sensitivity; A116 - tolerant), 2) using an integrated physiological (leaf gas exchanges; chlorophyll a fluorescence) and biochemical (photoprotective pigments; RuBisCO activity; primary metabolite profiling) analysis to identify drought tolerance probes, in plants submitted to three water availability levels (well-watered, WW; mild drought, MD; severe drought, SD). A116 plants maintained a better water status under drought, what could justify the higher Pn and Pnmax values in MD, as well as higher photochemical use of energy (reflected in the photochemical quenching (qL) and in the quantum yield of non-cyclic electron transport (Y(II))), and the lower need of photoprotective thermal dissipation mechanisms (given by the non-photochemical quenching (qN), and the quantum yield of regulated energy dissipation at photosystem PSII (Y(NPQ))), in MD and SD plants. Greater declines of net (Pn) and potential (Pnmax) photosynthesis were observed in A55 plants, which frequently showed significant impacts already under MD conditions in most parameters, whereas A80 usually displayed and intermediate behaviour. Still, even A55 showed some acclimation response, regarding photoprotective mechanisms associated with high contents of zeaxanthin, lutein, and carotenes, and high Y(NPQ), and qN values, supporting the absence of an increase in the non-regulated energy dissipation at PSII (Y(NO) did not increased) even in SD plants. Additionally, A55 was not significantly affected in RuBisCO activity, which showed to be quite resilient in cowpea. A primary metabolite profiling, complemented with a partial least square discrimination analysis (PLS-DA), allowed a better separation of A116 and A55 plants according to their degree of drought tolerance. In response to drought, A116 showed the greatest accumulation of most responsive metabolites, 14 in total, with sucrose, fucose, urea, alanine and putrescine being exclusively increased in this genotype, suggesting that they can be candidates as drought tolerance proxies. Other compounds, as proline, valine, isoleucine (among amino acids), and rhamnose and raffinose (among sugars) showed close increase patterns across landraces, thus they would be involved in the common drought response of cowpea plants. This pioneer approach to characterize Africa’s and Central Mozambique highly diverse cowpea germplasm, highlights the advantages of an integrated physiological and biochemical analysis, to identify response mechanisms and potential probes for drought tolerance in cowpea, aiming at to support selection and breeding strategies to obtain high yield and drought tolerant elite varieties.

Physiological effects of cadmium stress on Astragalus membranaceus seedlings and alleviative effects of attapulgite clay on cadmium stress

In this study we investigate the effects of cadmium stress on Astragalus membranaceus seedlings and the alleviative effects of attapulgite clay in growth substrate on cadmium stress to A. membranaceus seedlings. The results showed that the Y (Ⅱ) (effective photochemical quantum yield of PSⅡ photosynthetic), qP(photochemical quenching coefficient), ETR(the rate of non-cyclic electrontransport through PSⅡ), and chlorophyll content of the leaves were significantly decreased with the increase of cadmium concentrations, while the cadmium content, non-photochemical quenching(NPQ, qN) of the leaves and cadmium content, MDA content, plasma membrane permeability, and the damage degree of root apical membrane of the roots were significantly increased. Simultaneously, the activities of APX(ascorbate peroxidase), SOD(superoxide dismutase), POD(peroxidase), CAT(catalase), soluble protein content, and soluble sugar content of roots were increased first but then decreased with the increasing cadmium concentration. Under the condition of without Cd stress, the attapulgite clay into the growth substrate did not significantly affect above physiological indexes of leaves, but significantly increased SOD activity and soluble sugar content of roots and decreased the MDA content, damage degree of root apical membrane of roots, while other physiological indexes did not significantly change. Under cadmium stress, the presence of attapulgite clay in the growth substrate significantly alleviated the cadmium-induced decreases Y (Ⅱ), qP, ETR and chlorophyll content of leaves, and the CAT activity, soluble protein content, and soluble sugar content of roots. Under condition with cadmium stress, the presence of attapulgite clay significantly alleviated the cadmium-induced increases of leaves cadmium content, qN and NPQ, and the cadmium content, MDA content, plasma membrane permeability, damage degree of root apical membrane, SOD, POD, and APX activity of the roots. And, the alleviative effects of attapulgite clay on cadmium stress to A. membranaceus roots were more obvious with the increase of cadmium stress time. The above results showed that the addition of attapulgite clay into the growth substrate has certain alleviative effect on the cadmium stress to A. membranaceus seedlings.

Natural products from Pluchea sagittalis act as inhibitors of photosynthesis in vitro

Four compounds were isolated from roots and aerial parts of Pluchea sagittalis (Asteraceae), 3, 5-dihydroxy-6, 7, 3′, 4′-tetramethoxiflavunol (1), 5-hydroxymethylfurfural (2), 3, 4-dimethoxybenzaldehyde (3) and 2, 3, 4-trihydroxybenzaldeyde (4). Their herbicidal potential was detected by polarographic techniques. All of them inhibited the non-cyclic electron transport on basal, phosphorylating and uncoupled conditions from H2O to methylviologen (MV); thus, they act as Hill reaction inhibitors. Studies on fluorescence of chlorophyll a (ChL a) indicated they have different modes of interaction and inhibition sites on the photosystem II electron transport chain; 1–3 have interacted with the acceptor side while 4 has interacted at the donor side.

Effect of phytotoxic secondary metabolites and semisynthetic compounds from endophytic fungus Xylaria feejeensis strain SM3e-1b on spinach chloroplast photosynthesis

We investigated the mechanism of action on the photosynthesis light reactions of three major secondary metabolites produced by the endophytic fungus Xylaria feejeensis strain SM3e-1b, isolated from Sapium macrocarpum; and four novel derivatives of coriloxine, a major compound produced by X. feejeensis. The natural phytotoxins include one epoxycyclohexenone derivative, coriloxine (1), and two quinone derivatives (2–3). The semisynthetic derivatives of coriloxine are two cyclohexenone (4–6) and two quinone compounds (5–7). Cyclohexenone (4), (4R,5S,6R)-6-chloro-4,5-dihydroxy-3-methoxy-5-methylcyclohex-2-enone, inhibited ATP synthesis in freshly lysed spinach chloroplasts from water to MV; it also partly inhibited the basal and uncoupled photosynthetic electron transport, and significantly enhanced the phosphorylating electron transport and Mg2+-ATPase activity, thus demonstrating its action as an uncoupler agent. On the other hand, quinone (7), 2-((4-butylphenyl)amino)-5-methoxy-3-methylcyclohexa-2,5-diene-1,4-dione, inhibited ATP synthesis, and non-cyclic electron transport from water to MV in basal, phosphorylating and uncoupled conditions in a concentration-dependent manner. Hence, (7) behaves as a Hill reaction inhibitor at the PSII electron transport on the water splitting enzyme (OEC), and on the acceptor side between P680 and QA. This mechanism of action was confirmed by chlorophyll a fluorescence measurements. These results indicate that coriloxine derivatives 4 and 7 could work as prototype structures for the development of new herbicides. Contrastingly, natural products 1–3, and derivatives 5 and 6 did not show a significant inhibitory effect on ATP synthesis.

Effects of elevated ozone on physiological, anatomical and ultrastructural characteristics of four common urban tree species in China

Fast urbanization has led to ozone (O3) being the main pollutant in summer in most of China. To assess future ground-level O3 effects on the service of urban greening species and clarify the underlying mechanism of O3 damage, four common urban greening species, Ailanthus altissima (AA), Fraxinus chinensis (FC), Platanus orientalis (PO) and Robinia pseudoacacia (RP) were exposed to non-filtered air (NF) and to elevated O3 (E-O3) in open-top chambers. E-O3 induced visible injury in all species as well as microscopic alterations such as collapse of the palisade parenchyma cells, callose accumulation, or chloroplast and mitochondrial accelerated senescence. E-O3 significantly reduced light-saturated CO2 assimilation (Asat), the maximum activity of Rubisco (Vcmax), the maximum electron transport rate (Jmax), and fluorescence parameters such as the quantum yield of noncyclic electron transport (ϕPSII), and the quenching of photochemical efficiency of PSII (qP). It also increased total antioxidant capacity, phenolics and ascorbate contents. No significant interaction between O3 and species was found in photosynthetic performance and antioxidant systems, suggesting that the four species selected were sensitive to O3. Of all four species, AA was the most sensitive species due to a combination of earlier injury onset, anatomical features, lower antioxidant responses and higher stomatal conductance. The sensitivity of tree species to O3 is a factor to be considered for urban greening. Ozone may affect important urban forest ecosystem services by reducing CO2 assimilation.

Acridone Alkaloids from Swinglea glutinosa (Rutaceae) and Their Effects on Photosynthesis.

Continuing our search for herbicide models based on natural products, we investigated the action mechanisms of five alkaloids isolated from Swinglea glutinosa (Rutaceae): Citrusinine-I (1), glycocitrine-IV (2), 1,3,5-trihydroxy-10-methyl- 2,8-bis(3-methylbut-2-en-1-yl)-9(10H)-acridinone (3), (2R)-2-tert-butyl-3,10-dihydro-4,9-dihydroxy-11-methoxy-10-methylfuro[3,2-b]acridin-5(2H)-one (4), and (3R)-2,3,4,7-tetrahydro-3,5,8-trihydroxy-6-methoxy-2,2,7-trimethyl-12H-pyrano[2,3-a]acridin-12-one (5) on several photosynthetic activities in an attempt to find new compounds that affect photosynthesis. Through polarographic techniques, the compounds inhibited the non-cyclic electron transport in the basal, phosphorylating, and uncoupled conditions from H2 O to methylviologen (=MV). Therefore, they act as Hill reaction inhibitors. This approach still suggested that the compounds 4 and 5 had their interaction site located at photosystem I. Studies on fluorescence of chlorophyll a suggested that acridones (1-3) have different modes of interaction and inhibition sites on the photosystem II electron transport chain.

Effect of methylmercury on the light dependence fluorescence parameters in a green alga Chlamydomonas moewusii

The effect of a dangerous toxic substance, methylmercury, on light dependence curves of chlorophyll fluorescence in Chlamydomonas moewusii was studied. We found low concentration of methylmercury (10(-7) M) to cause a decrease in the relative rate of the non-cyclic electron transport activity of PS 2, a decline in the maximum utilization of light energy (α), and a decline in the saturation light intensity (E(s)). Non-photochemical fluorescence quenching increased after short-term exposure and decreased in the course of prolonged incubation. These parameters were more sensitive to the action of the toxic substance than the widely used parameter F(V)/F(M), which reflects the maximum quantum yield of PS 2. We propose the use of the method of fast measurement of light dependence curves of fluorescence to detect the changes in algal cells at the early stages of exposure to mercury salts.

Selected phytotoxins and organic extracts from endophytic fungus Edenia gomezpompae as light reaction of photosynthesis inhibitors

In a search for natural herbicides, we investigated the action mechanism of the naphthoquinone spiroketals, isolated from the endophytic fungus Edenia gomezpompae: preussomerins EG1 (1) and EG4 (2), and palmarumycins CP17 (3), and CP2 (4) on the photosynthesis light reactions. The naphthoquinone spiroketals 1–4 inhibited the ATP synthesis in freshly lysed spinach thylakoids from water to MV, and they also inhibited the non-cyclic electron transport in the basal, phosphorylating and uncoupled conditions from water to MV. Therefore, they act as Hill reaction inhibitors. The results suggested that naphthoquinone spiroketals 1–4 have two interactions and inhibition site on the PSII electron transport chain. The first one involves the water splitting enzyme inhibition; and, the second on the acceptor site of PSII in a similar way that herbicide Diuron, studied by polaroghaphy and corroborated by fluorescence of the chlorophyll a of PSII. The culture medium and mycelium organic extracts from four morphological variants of E. gomezpompae were phytotoxic, and the culture medium extracts were more potent than mycelium extracts. They also act as Hill reaction inhibitors.

Moderate water stress causes different stomatal and non‐stomatal changes in the photosynthetic functioning of Phaseolus vulgaris L. genotypes

The impact of moderate water deficit on the photosynthetic apparatus of three Phaseolus vulgaris L. cultivars, Plovdiv 10 (P10), Dobrudjanski Ran (DR) and Prelom (Prel), was investigated. Water shortage had less impact on leaf hydration, RWC (predawn and midday) and predawn water potential in Prel. RWC and Ψ(p) were more reduced in P10, while there was no osmotic adjustment in any cultivar. Although drought drastically reduced stomatal opening in P10 and DR, reduced A(max) indicated non-stomatal limitations that contributed to the negligible P(n). These limitations were on potential thylakoid electron transport rates of PSI and II, pointing to photosystem functioning as a major limiting step in photosynthesis. This agrees with decreases in actual photochemical efficiency of PSII (F(v)'/F(m)'), quantum yield of photosynthetic non-cyclic electron transport (ϕ(e)) and energy-driven photochemical events (q(P)), although the impact on these parameters would also include down-regulation processes. When compared to DR, Prel retained a higher functional state of the photosynthetic machinery, justifying reduced need for photoprotective mechanisms (non-photochemical quenching, zeaxanthin, lutein, β-carotene) and maintenance of the balance between energy capture and dissipative pigments. The highest increases in fructose, glucose, arabinose and sorbitol in Prel might be related to tolerance to a lower oxidative state. All cultivars had reduced A(max) due to daytime stomatal closure in well-watered conditions. Under moderate drought, Prel had highest tolerance, higher leaf hydration and maintenance of important photochemical use of energy. However, water shortage caused appreciable non-stomatal limitations to photosynthesis linked to regulation/imbalance at the metabolic level (and growth) in all cultivars. This included damage, as reflected in decreased potential photosystem functioning, pointing to higher sensitivity of photosynthesis to drought than is commonly assumed.

Furoquinoline alkaloids isolated from Balfourodendron riedelianum as photosynthetic inhibitors in spinach chloroplasts

In the search for natural inhibitors of plant growth, we investigate the mechanism of action of the natural furoquinoline alkaloids isolated from Balfourodendron riedelianum (Rutaceae): evolitrine (1), kokusaginine (2), γ-fagarine (3), skimmianine (4) and maculosidine (5) on the photosynthesis light reactions. Their effect on the electron transport chain on thylakoids was analyzed. Alkaloids 1, 2, 4 and 5 inhibited ATP synthesis, basal, phosphorylating and uncoupled electron transport acting as Hill reaction inhibitors on spinach chloroplasts. Alkaloid 3 was not active. The inhibition and interaction site of alkaloids 1, 2, 4 and 5 on the non-cyclic electron transport chain was studied by polarography and fluorescence of the chlorophyll a (Chl a). The results indicate that the target for 1 was localized on the donor and acceptor side of PS II. In addition alkaloids 2 and 5 affect the PS I electron acceptors on leaf discs.

Ecophysiological responses of Caragana korshinskii Kom. under extreme drought stress: Leaf abscission and stem survives

Caragana korshinskii Kom. is a perennial xerophytic shrub, well known for its ability to resist drought. In order to study ecophysiological responses of C. korshinskii under extreme drought stress and subsequent rehydration, diurnal patterns of gas exchange and chlorophyll (Chl) fluorescence parameters of photosystem II as well as Chl content were analyzed. Plant responses to extreme drought included (1) leaf abscission and using stem for photosynthesis, (2) improved instantaneous water-use efficiency, (3) decreased photosynthetic rate and partly closed stomata owing to leaf abscission and low water status, (4) decreased maximum photochemical efficiency of photosystem II (PSII) (variable to maximum fluorescence ratio, Fv/Fm), quantum efficiency of noncyclic electron transport of PSII, and Chl a and Chl b. Four days after rehydration, new leaves budded from stems. In the rewatered plants, the chloroplast function was restored, the gas exchange and Chl fluorescence returned to a similar level as control plant. The above result indicated that maintaining an active stem system after leaf abscission during extreme drought stress may be the foundation which engenders these mechanisms rapid regrowth for C. korshinskii in arid environment.

Intraspecific variation in Pinus pinaster PSII photochemical efficiency in response to winter stress and freezing temperatures.

As part of a program to select maritime pine (Pinus pinaster Ait.) genotypes for resistance to low winter temperatures, we examined variation in photosystem II activity by chlorophyll fluorescence. Populations and families within populations from contrasting climates were tested during two consecutive winters through two progeny trials, one located at a continental and xeric site and one at a mesic site with Atlantic influence. We also obtained the LT50, or the temperature that causes 50% damage, by controlled freezing and the subsequent analysis of chlorophyll fluorescence in needles and stems that were collected from populations at the continental trial site. P. pinaster showed sensitivity to winter stress at the continental site, during the colder winter. The combination of low temperatures, high solar irradiation and low precipitation caused sustained decreases in maximal photochemical efficiency (Fv/Fm), quantum yield of non-cyclic electron transport (ΦPSII) and photochemical quenching (qP). The variation in photochemical parameters was larger among families than among populations, and population differences appeared only under the harshest conditions at the continental site. As expected, the environmental effects (winter and site) on the photochemical parameters were much larger than the genotypic effects (population or family). LT50 was closely related to the minimum winter temperatures of the population's range. The dark-adapted Fv/Fm ratio discriminated clearly between interior and coastal populations.In conclusion, variations in Fv/Fm, ΦPSII, qP and non-photochemical quenching (NPQ) in response to winter stress were primarily due to the differences between the winter conditions and the sites and secondarily due to the differences among families and their interactions with the environment. Populations from continental climates showed higher frost tolerance (LT50) than coastal populations that typically experience mild winters. Therefore, LT50, as estimated by Fv/Fm, is a reliable indicator of frost tolerance among P. pinaster populations.

Synthetic Auxin 3,5,6-TPA Provokes Citrus clementina (Hort. ex Tan) Fruitlet Abscission by Reducing Photosynthate Availability

The aim of this study was to determine the effects of the synthetic auxin 3,5,6-trichloro-2-pirydiloxyacetic acid (3,5,6-TPA) on photosynthetic activity, photosynthate transport to the fruit, and fruitlet abscission to further explain the physiological basis of auxin-mediated citrus fruit thinning. Applying 15 mg l−1 3,5,6-TPA to trees during the fruit cell division stage significantly increased fruitlet abscission of Clementine mandarin. On treated trees, abnormal foliar development and photosynthetic damage were observed at the same time as 3,5,6-TPA reduced fruitlet growth rate. Briefly, treatment reduced chlorophyll and carotenoid concentrations and modified chlorophyll a fluorescence parameters, that is, reduced the quantum yield (ФPSII) of the noncyclic electron transport rate, diminished the capacity to reduce the quinone pool (photochemical quenching; qp), and increased nonphotochemical quenching (qN), thereby preventing the dissipation of excess excitation energy. In addition, the net photosynthetic flux (μmol CO2 m−2 s−1) and leaf photosynthate content decreased in treated trees. As a result, the 3,5,6-TPA treatment significantly reduced the photosynthate accumulation in fruit from day 3 to day 8 after treatment, thus reducing fruitlet growth rate. Hence, treated fruitlets significantly increased ethylene production and abscised. Twenty days after treatment, chlorophyll a fluorescence parameters and fruitlet growth rate were reestablished. Accordingly, the thinning effect of 3,5,6-TPA may be due to a temporarily induced photosynthetic disorder that leads to reduction in photosynthate production and fruitlet uptake that temporarily slows its growth, triggering ethylene production and fruitlet abscission. Afterward, the remaining treated fruit overcame this effect, increased growth rate, and reached a larger size than control fruit.

Effects of inhibitors and cofactors of cyclic and noncyclic electron transport on nonphotochemical quenching of chlorophyll fluorescence induced by action potential (AP) was investigated in Chara corallina cells. Under control conditions, energy-dependent quenching (qE) develops upon the increase in photosynthetically active radiation (PAR); it also arises and reversibly disappears after AP generation at moderate irradiances. The treatment of cells with diuron (DCMU) completely eliminated qE established at high irradiances and qE induced by AP generation. The activation of cyclic electron transport by DCMU in combination with phenazine methosulfate restored qE at high irradiances but did not restore qE imposed after AP generation. The presence in the medium of a PSI acceptor, methyl viologen at concentrations from 100 μM to 0.83 mM had no effect on fluorescence and photosynthetic activity of chloroplasts until the application of a single excitatory stimulus. Once a single AP was generated in the presence of methyl viologen, it induced irreversible qE at a wide range of irradiances, which indicates the AP-triggered redirection of a part of electron flow from the main pathway to the artificial acceptor. It is concluded that AP generation opens access for permeation of a divalent cation methyl viologen from the medium to the chloroplast stroma across two membrane barriers, the plasmalemma and the inner membrane of the chloroplast envelope.

Alternative pathways of photoinduced electron transport in chloroplasts

The influence of the alternative pathways of electron transport in photosynthetic systems of the oxygen type on the kinetics of the photoinduced redox transitions of P700, ferredoxin, NADP, pH of the intrathylakoid space or lumen, and relative concentration of ATP was studied. The oxygen effect on the kinetics of photooxidation of P700 was analyzed. The retardation of the photooxidation of P700 at low oxygen concentrations can be explained by the “over-reduction” of the acceptor side of PS1 as a result of a decrease in the electron outflow from PS1 to oxygen during hypoxia. The results of numerical experiments are in good agreement with known experimental data that the withdrawal of electrons from PS1 (on the ferredoxin-NADP segment of the chain) can be the limiting stage in the noncyclic electron transport chain. The functioning of the cyclic electron transport chain provides additional synthesis of ATP molecules and weakens the excess reduction of the acceptor segment of PS1. The alternative pathway of electron transport, namely, electron outflow from PS1 to oxygen also favors the optimum conditions for the functioning of the photosynthetic electron transport chain.

Oxygen stress in Salvinia natans: Interactive effects of oxygen availability and nitrogen source

The ability of Salvinia natans (L.) All. to tolerate growth in oxic, hypoxic and anoxic nutrient solutions when supplied with either NH 4 + or NO 3 − were studied in the laboratory to test the hypothesis that inorganic N-source affects the response of the plants to O 2 deprivation. The relative growth rate (RGR) was significantly reduced in the anoxic treatment, but in the hypoxic treatment RGR was only slightly affected. The NH 4 + fed plants generally had a higher shoot to root ratio than the NO 3 − fed plants, and highest in the anoxic treatment. Plants had more roots and larger leaves when supplied with NH 4 + as compared with NO 3 −, particularly in the oxic treatment, and root length was most affected by O 2 deprivation for NO 3 − fed plants. Cell walls in the endodermis, the bundle sheath and the cortex adjacent to endodermis developed thickened sclerenchymatous walls when deprived of O 2, and more so in plants supplied with NO 3 −. Plants lost chlorophylls, had lower rates of photosynthetic electron transport (ETR max) and lower quantum yields (Fv/Fm ratios) when grown in anoxic solutions, and the negative effects were mildest for NO 3 − fed plants suggesting that NO 3 − may be used as an alternative e −-acceptor in non-cyclic electron transport in the chloroplasts. Overall S. natans grew best on NH 4 +, but it also grew well on NO 3 −, and the O 2 stress symptoms differed somewhat between NH 4 + fed and NO 3 − fed plants. However, because N-form itself significantly influenced morphology and cell metabolism, it was impossible to conclusively identify the role of N-form for the O 2 stress reactions. S. natans is not well-adapted to grow in O 2 deficient waters and will not tolerate completely anoxic conditions as will prevail in waters receiving high loadings of organic pollutants such as livestock wastewater.

Facilitated permeation of methyl viologen into chloroplasts in situ during electric pulse generation in excitable plant cell membranes

Generation of action potential (AP) in plasma membranes of characean algae has a strong impact on photoreactions occurring in chloroplasts. Under physiological conditions, AP suppresses electron transport in alkaline and acidic regions, although to a different extent; these changes are transient and reversible. In the presence of the artificial electron acceptor, methyl viologen (MV2+), AP-induced changes in electron transport in photosystem II become irreversible. Incubation of Chara corallina internodal cells with MV2+ has no effect on the chlorophyll P700 photooxidation kinetics in photosystem I reaction centers, suggesting that MV2+ is inaccessible for interactions with photosystem I, because its permeation into chloroplasts of a resting cell is hindered by membrane barriers. At the same time, AP generation in the presence of MV2+ is accompanied by irreversible modification of P700 photooxidation kinetics, as can be evidenced from differences in absorption changes at 810 and 870 nm (ΔA810 signals). These findings suggest that MV2+ permeation into chloroplasts in situ is facilitated during or after the AP generation. Similar to the ΔA810 signals, light-induced changes in membrane potential do not depend on the presence of MV2+ in the external medium until the first excitatory stimulus is applied. Electric photoresponses of the cell are irreversibly modified by AP generated in the presence of MV2+ at the expense of non-cyclic photosynthetic electron transport redirected to the MV2+ reduction. It is concluded that AP effects on chloroplast photosynthesis in situ are complex and involve permeability changes for MV2+ in membrane barriers of the “plasmalemma-chloroplast envelope” system.

Photosynthetic activity of poikilochlorophyllous desiccation tolerant plant Reaumuria soongorica during dehydration and re-hydration

Diurnal patterns of gas exchange and chlorophyll (Chl) fluorescence parameters of photosystem 2 (PS2) as well as Chl content were analyzed in Reaumuria soongorica (Pall.) Maxim., a perennial semi-shrub during dehydration and rehydration. The net photosynthetic rate (P N), maximum photochemical efficiency of PS2 (variable to maximum fluorescence ratio, Fv/Fm), quantum efficiency of non-cyclic electron transport of PS2, and Chl content decreased, but non-photochemical quenching of fluorescence and carotenoid content increased in stems with the increasing of drought stress. 6 d after re-hydration, new leaves budded from stems. In the re-watered plants, the chloroplast function was restored and Chl a fluorescence returned to a similar level as in the control plants. This improved hydraulic adjustment in plant triggered a positive effect on ion flow in the tissues and increased shoot electrical admittance. Thus R. soongorica plants are able to sustain drought stress through leaf abscission and keep part of Chl content in stems.
