The Effect of Holder Pasteurization and Different Variants on Breast Milk Antioxidants.

After birth, breast milk (BM) is a known essential source of antioxidants for infants. We analyzed the non-enzymatic total antioxidant capacity (TAC), oxygen radical absorbance capacity (ORAC), and glutathione, calcium, transferrin, and total protein levels of human breast milk before and after Holder pasteurization (HoP). The collected donor BM samples were pasteurized with HoP. HoP decreased TAC (-12.6%), ORAC (-12.1%), transferrin (-98.3%), and total protein (-21.4%) levels; HoP did not influence the glutathione concentration, and it increased the total calcium (+25.5%) concentration. Mothers who gave birth via Cesarean section had significantly lower TAC in their BM. TAC and glutathione levels were elevated in the BM of mothers over the age of 30. BM produced in the summer had higher glutathione and calcium levels compared to BM produced in the winter. The glutathione concentration in term milk samples was significantly higher in the first two months of lactation compared to the period between the third and sixth months. The transferrin level of BM for female infants was significantly higher than the BM for boys, and mothers with a BMI above 30 had increased transferrin in their samples. Antioxidant levels in human milk are influenced by numerous factors. Environmental and maternal factors, the postpartum age at breast milk collection, and Holder pasteurization of the milk influence the antioxidant intake of the infant.

Serum Total Antioxidant Capacity (TAC) and TAC/Lymphocyte Ratio as Promising Predictive Markers in COVID-19.

SARS-CoV-2 infection might cause a critical disease, and patients' follow-up is based on multiple parameters. Oxidative stress is one of the key factors in the pathogenesis of COVID-19 suggesting that its level could be a prognostic marker. Therefore, we elucidated the predictive value of the serum non-enzymatic total antioxidant capacity (TAC) and that of the newly introduced TAC/lymphocyte ratio in COVID-19. We included 61 COVID-19 (n = 27 ward, n = 34 intensive care unit, ICU) patients and 29 controls in our study. Serum TAC on admission was measured by an enhanced chemiluminescence (ECL) microplate assay previously validated by our research group. TAC levels were higher (p < 0.01) in ICU (median: 407.88 µmol/L) than in ward patients (315.44 µmol/L) and controls (296.60 µmol/L). Besides the classical parameters, both the TAC/lymphocyte ratio and TAC had significant predictive values regarding the severity (AUC-ROC for the TAC/lymphocyte ratio: 0.811; for TAC: 0.728) and acute kidney injury (AUC-ROC for the TAC/lymphocyte ratio: 0.747; for TAC: 0.733) in COVID-19. Moreover, the TAC/lymphocyte ratio had significant predictive value regarding mortality (AUC-ROC: 0.752). Serum TAC and the TAC/lymphocyte ratio might offer valuable information regarding the severity of COVID-19. TAC measured by our ECL microplate assay serves as a promising marker for the prediction of systemic inflammatory diseases.

Does the presence of heavy metal and catechol contaminants in organic waste challenge the physiological performance of the bioconverter Hermetia illucens?

The increased human activities and the worldwide population growth are constantly increasing the production of solid wastes. Over the years, waste management has thus become a prominent issue for several companies and municipalities, and several engineering techniques have been developed over the years in order to convert wastes into other solid materials or fuels. Yet, several techniques are important contributors to environmental pollution, and biological-based solutions have thus become progressively very popular. In particular, insect-based conversion of organic wastes represent eco-friendly tools, and the growth and development of insect species such as the black soldier fly have been tested and improved for a large diversity of organic wastes. However, organic wastes, including food wastes, may contain several pollutants such as heavy metals and catechol which could affect the bioconversion efficiency by incurring physiological costs that would be undetectable at the organismal level, i.e. have null to little effects on the life cycle of Hermetia illucens. In this context, assessments of antioxidant capacities can provide a rapid and low-cost evaluation of the capability of insects to handle exposure to heavy metals and catechol. Here, we aimed at measuring the physiological responses of the black soldier fly H. illucens grown on food wastes (kitchen, fruit or vegetable wastes) contaminated by cadmium, iron, lead or catechol. Biomarkers of oxidative stress (concentrations of hydrogen peroxide and protein carbonyls), non-enzymatic total antioxidant capacity (ascorbic acid amounts) and activity of enzymatic antioxidants (activities of superoxide dismutase and polyphenoloxidase) were measured from the gut of the larvae. We found no evidence of deleterious impacts of food waste contamination by catechol or heavy metals on H. illucens. In most experimental treatments, the array of physiological endpoints we measured for evaluating the degree of oxidative stress experienced by the larvae remained similar to controls. Possible physiological effects were reported for cadmium and catechol only, which tended to increase the oxidation of proteins and hydrogen peroxide in the larvae. Finally, our results suggested that the nature of the food waste could equally affect the physiological responses of the insect.

Maternal Apical Periodontitis Increases Insulin Resistance and Modulates the Antioxidant Defense System in the Gastrocnemius Muscle of Adult Offspring

IntroductionMaternal apical periodontitis (AP) is associated with insulin resistance (IR) in adult offspring. Oxidative stress has been linked to IR. This study investigated insulin sensitivity (IS) and oxidative stress in the gastrocnemius muscle (GM) of adult offspring of rats with AP. MethodsFifteen female Wistar rats were distributed into a control group, a group with 1 tooth with AP, and a group with 4 teeth with AP. Thirty days after AP induction, female rats were mated with healthy male rats. When male offspring reached 75 days of age, glycemia, insulinemia, and IS were determined. In the GM, the oxidative damage products (thiobarbituric acid reactive substances and carbonyl protein) and activities of enzymatic (superoxide dismutase, catalase, and glutathione peroxidase) and nonenzymatic (glutathione and total antioxidant capacity) antioxidants were quantified. Analysis of variance was performed followed by the Tukey post hoc test (P < .05). ResultsMaternal AP was associated with decreased IS and changes in antioxidant activities (reduced superoxide dismutase and increased catalase, glutathione peroxidase, and glutathione) and decreased thiobarbituric acid reactive substance concentration in the GM of their adult offspring. However, maternal AP does not appear to affect glycemia, carbonyl protein concentration, and the nonenzymatic total antioxidant capacity in the GM of this offspring. ConclusionsMaternal AP modulates the antioxidant defense system in the GM of their adult offspring, attenuating lipid peroxidation in this tissue. This reflects part of an adaptive response of the offspring to the stimulation of the maternal chronic oral inflammatory process in which the organism acts by decreasing oxidative tissue damage in the postnatal stage. The present study improves knowledge about the impact of maternal oral inflammation on healthy offspring.

Cardioprotective effects of severe calorie restriction from birth in adult ovariectomized rats

AimsMenopause is a female condition induced by a reduction of ovarian hormone and is related to an increase in cardiovascular diseases in women. We have shown that severe calorie restriction (SCR) from birth reduces the cardiometabolic risk in adult male Wistar rats. In this study, we investigated the effects of SCR from birth to adulthood on cardiovascular function of ovariectomized rats. Main methodsFrom birth to adulthood, rats were daily fed ad libitum (control group - C) or with 50% of the amount consumed by the control group (calorie-restricted group - R). At 90 days, half of the rats in each group underwent bilateral ovariectomy (OVX), totaling 4 groups: C-Sham, C-OVX, R-Sham, R-OVX. Systolic blood pressure (SBP), heart rate (HR) and, double product (DP) index were recorded by tail-cuff plethysmography. Cardiac function was analyzed by the Langendorff technique and cardiomyocyte diameter was accessed by histologic analysis. Additionally, cardiac SERCA2 content and redox status were evaluated. Key findingsC-OVX rats exhibited reduced cardiac function and cardiac non-enzymatic total antioxidant capacity (TAC). R-Sham animals showed reduced SBP, DP, HR, improved cardiac function, reduced cardiac protein carbonyl derivatives and increased TAC, catalase, and superoxide dismutase activities. R-OVX rats maintained reduced SBP, DP, HR, and increased contractility and relaxation indexes. R-Sham and R-OVX rats exhibited preserved heart mass and reduced cardiomyocyte diameter. Cardiac SERCA2 content did not differ between the groups. SignificanceTaken together, our findings show cardioprotective effects of SCR from birth in adult ovariectomized rats.

A single session of high-intensity interval exercise increases antioxidants defenses in the hippocampus of Wistar rats

It is known that a single session of high-intensity interval exercise (HIIE) contributes to the increase of the reactive species of oxygen, accompanied by a greater antioxidant activity. However, it is poorly understood if a single session of HIIE has similar effects on the brain tissue. This study evaluated the effects of a single HIIE on the hippocampal redox status. Sixteen males Wistar rats were allocated into HIIE (n = 8) and control (n = 8) groups. Maximum oxygen consumption (VO2max) was evaluated using a treadmill at 10° inclination in a metabolic chamber. HIIE group was submitted to a single run on the treadmill composed by 10 bouts of high-intensity exercise of 1 min each (85–100% of VO2max), at 28 m/min, 10° inclination, interspersed by 2 min of active recovery, at 10 m/min, with no inclination. Analysis of the redox status at the hippocampus were conducted 24 h after the HIIE session. It was not identified lipid peroxidation in the hippocampus of the HIIE group (Control 1.9 ± 0.31, vs HIIT 2.2 ± 0.53 nmol MDA/mg protein (p > .05). However, the activity of the superoxide dismutase (Control 2.614 ± 0.225 vs HIIT 3.718 ± 0.4589 U/mg protein), and the non-enzymatic total antioxidant capacity (Control 1584 ± 75.88 vs HIIT 1984 ± 137.7 nM FeSO4/mg protein) were enhanced (p < .05) after the exercise session. These results indicate that the antioxidant mechanisms are enhanced even after a single session of HIIE. A single session of HIIE does not induce lipid peroxidation and improves the antioxidant defenses in Wistar rats' hippocampus.

Effects of the Urban Environment on Oxidative Stress in Early Life: Insights from a Cross-fostering Experiment

As urban areas expand rapidly worldwide, wildlife is exposed to a wide range of novel environmental stressors, such as increased air pollution and artificial light at night. Birds in highly polluted and/or urbanized habitats have been found to have increased antioxidant protection, which is likely important to avoid accumulation of oxidative damage, which can have negative fitness consequences. Yet, the current knowledge about the ontogeny of antioxidant protection in urban areas is limited; i.e., is the capacity to up-regulate the antioxidant defences already established during pre-natal development, or does it manifest itself during post-natal development? We cross-fostered great tit (Parus major) nestlings within and between urban and rural habitats, to determine if oxidative stress (measured as non-enzymatic total antioxidant capacity, superoxide dismutase (SOD), and plasma lipid peroxidation) is affected by habitat of origin and/or by habitat of rearing. The results demonstrate that being reared in the urban environment triggers an increase in SOD (an intracellular, enzymatic antioxidant) independent of natal habitat. Oxidative damage increased with hatching date in urban-reared nestlings, but there was little seasonal change in rural-reared nestlings. Total antioxidant capacity was neither affected by habitat of rearing or habitat of origin, but we observed a decline with hatching date in both rearing habitats. Taken together, our results support the growing evidence that the urban environment induces a direct plastic adjustment in antioxidant protection, but that up-regulation is not sufficient to avoid increased oxidative damage in late-hatched broods. Future studies should explore the underlying causes for this effect in late-hatched broods and whether it has any negative long-term implications, both at the individual- and the population level.

Increased Total Antioxidant Capacity in Seminal Plasma of Varicocele Patients: A Multivariate Analysis

To investigate seminal antioxidant systems and their correlation with hormonal pattern in varicocele patients, we studied 33 varicocele (VAR) patients (12 oligozoospermic, 21 normozoospermic) and 34 non-VARs (10 idiopathic oligozoospermic, 24 normozoospermic). Non-enzymatic total antioxidant capacity (TAC) was measured using H(2)O(2)-metamyoglobin, which generates the radical form, spectroscopically detectable, of the chromogen 2,2',-azinobis-3-ethylbenzothiazoline-6-sulphonate (ABTS): time till appearance (Lag) of ABTS*(-) signifies antioxidant concentration. Lag was significantly longer in VARs than controls suggesting ineffective utilization of antioxidants. A significant direct correlation (r = 0.65, p < 0.01) of Lag with sperm count was observed in non-VARs, while in VARs it was inverted, as well as with hematic FSH levels. A multivariate analysis including FSH, Lag, progressive spermatozoa, oligozoospermia and varicocele indicated a strong inverse correlation between FSH and motility (r(2) = 0.31, p > F = 0.0007), not modified by Lag (r(2) = 0.31, p > F = 0.002). Their inverse correlation with Lag may suggest that higher FSH levels, improving sperm antioxidant efficiency, counterbalance varicocele-induced dyspermia.

Coenzyme Q10 effects on non-enzymatic total antioxidant capacity in seminal plasma of varicocele patients

In order to investigate antioxidant systems in varicocele (VAR), we studied the correlation between non-enzymatic total antioxidant capacity (TAC) and seminal parameters and subsequently the effect of supplementation of antioxidant, coenzyme Q10 (CoQ10). TAC was determined in seminal plasma of 27 VAR patients (10 oligospermic and 17 normospermic) and 24 non-VAR controls (7 with idiopathic oligospermia and 17 normospermic). We measured the interaction of system H 2O 2-metamyoglobin with chromogen ABTS, whose radical cation is spectroscopically detectable. The length of latency phase (Lag) in ABTS accumulation is proportional to antioxidant concentration. Lag was significantly greater in all VAR vs. control subjects; a multivariate analysis indicated a very high inverse correlation between FSH and the percentage of motile progressive cells, which was not further modified by addition of Lag length to the statistical model. CoQ10 treatment (100 mg/daily for 3 months) significantly increased the Lag phase. We maintain that greater FSH levels, achieved by inducing a more efficient utilisation of antioxidants by spermatozoa, may counterbalance VAR-induced spermatozoa deficit.
