Periselective, Lewis acid-induced intramolecular Diels–Alder reaction of conjugated carbodiimides: efficient synthesis of nitrogen heterocycles, indolo[2,3-b]quinolines and pyrido[2,3-b]indole

A suitable Lewis acid has been found that accelerates the intramolecular Diels–Alder reaction of conjugated carbodiimides with high periselectivity control, thus providing an efficient and straightforward procedure for constructing indolo[2,3-b]quinoline and pyrido[2,3-b]indole frameworks.

Azabicyclo[3.3.0]octenones and azabicyclo[4.3.0]nonenones have been obtained by cyclisation of hexacarbonyldicobalt complexes of aza-heptenynes and -octenynes respectively. An unusual feature of the cyclisation of N-acyl-4-azahept-1-en-6-ynes is the extensive formation of the reduced products, N-acyl-3-azabicyclo[3.3.0]octan-7-ones under most reaction conditions.

Trithiocyclopropenium ion as a building block for nitrogen heterocycle synthesis

A new methodology for heterocycle synthesis using trithiocyclo-propenium salt as a building bock is described. Tris( tert -butylthio)cyclopropenium perchlorate ( 1 ) reacts with β-amino acids under basic conditions to give 1,2-dihydropyridines, where cyclopropenium salt serves as the three carbon homologator in heterocyclic ring formation. The selective syntheses of 1,5-benzodiazepines and benzimidazoles from 1 are also described. Reaction of 1 with o-phenylenediamines in dimethylformamide gives 1,5-benzodiazepines as a single product but in methanol benzimidazoles. The selective formation of 1,5-benzodiazepines and benzimidazoles could be accounted for by the solvent participation in the cyclization step.

Synthesis of nitrogen heterocycles via palladium-catalyzed intramolecular cyclization

Catalytic amounts of Pd(OAc) 2 in the presence of n -Bu 4NCl, DMF and an appropriate base (Na 2CO 3, (NaOAc or Et 3N) cyclize nitrogen-containing o -iodoaryl alkenes to indoles, indolines,3 oxindoles, quinolines, isoquinolines and isoquinolones in short reaction times, under mild temperatures, and in high yields.

The data on the synthesis of nitrogenous heterocycles on the basis of mononitroalkanes, polynitroalkanes, and alkenes have been generalized in this review. Only reactions in which the oxygen and nitrogen atoms of a nitro group appear in the composition of the heterocycle formed have been considered.

Synthesis of nitrogen heterocycles by condensation of the conjugate base of open‐chain reissert compound analogs with vinyltriphenylphosphonium bromide (schweizer's salt)

AbstractReaction of the conjugate base of several open‐chain Reissert compound analogs with vinyltriphenylphosphonium bromide is shown to afford a convenient route to substituted pyrroles. The condensation is a two‐step process involving an initial reversible addition to form an unstabilized Wittig reagent followed by an intramolecular Wittig reaction upon the carbonyl group of the tertiary amide functionality with concomitant elimination of HCN. Several applications to the synthesis of 1,2,5‐trisubstituted pyrroles are presented, together with attempts to expand this methodology to the synthesis of other heterocyclic systems.

Reactions of Aroylhydrazones, IV: Synthesis of Nitrogen Heterocycles from Opianic Acid Hydrazones

AbstractOpianic acid reacts with various hydrazines to yield either hydrazone (3, 6) or phthalazinone (4) derivatives depending upon the electronic properties of the substituent attached to the hydrazine molecule. Oxidative cyclization of opianic acid aroylhydrazones with iodine and yellow mercuric oxide gave the 1,3,4‐oxadiazole derivatives 21. The aroylhydrazones were also condensatively cyclized by heating with acetic anhydride to give the 1,3,4‐oxadiazoline derivatives 24. Structures assigned to the prepared compounds were corroborated by studying their chemical as well as their spectral characteristics.
