Influence of Aeration, Introduction of Probiotics, and Supply of Water on Landfill Gas Production—Study of Models

When municipal solid waste (MSW) is placed in a landfill, it undergoes anaerobic decomposition, leading to the production of landfill gas, which primarily consists of methane (CH4) and carbon dioxide (CO2). Reducing methane emissions is essential in the fight against climate change. It must be implemented at global and European levels, as set out in 2030 in the impact assessment of the climate goal plan. This assessment states that to achieve the goal by 2030 and to reduce greenhouse gas emissions by at least 55%, the methane emissions must be reduced, considering the goals of the Paris Agreement. The Glasgow Climate Pact includes a global mitigation target of the year 2030: to reduce CO2 emissions by 45%, and the emissions of methane and other greenhouse gasses. For that purpose, looking for new, more advanced ways of managing such waste is necessary. The main objective of this experimental study was to evaluate the influence of aeration, probiotic introduction, and water supply on the production of landfill gasses (CO2, CH4, N2, H2, etc.) in five different landfill models during the management of MSW and to propose the best solutions for reducing environmental pollution. The results of the research showed that the first and second models of landfills, using only anaerobic conditions, can be used for the treatment of MSW for the production of biogas (CH4, CO2), as up to 40–60% of it was released during the 120-experiment period. The third landfill model can be applied in old, already closed landfills, where the rapid stabilization and aeration of MSW are required to minimize pollutant emissions (N2, etc.) and unwanted odors and shorten biodegradation processes. The results of the fourth and fifth landfill models, in which aerobic–anaerobic conditions were applied, showed that the developing nitrification–denitrification processes resulted in complete nitrogen removal (from 20% to 0%), and overall waste stabilization improved the biodegradation of the MSW. Later, relatively good (on average, 30%) results of biogas (CH4, CO2) emissions are achieved during anaerobic condition formation results. Summarizing all experiment results of all landfill models for the further evaluation of the processes, all models can be applied in real practice depending on where they will be used and what result they want to achieve.

Nitrogen removal strategy for real swine wastewater by combining partial nitrification-denitrification process with anammox

Anammox process offers reduced operational cost and energy requirement compared to nitrification-denitrification methods due to lower biomass generation and no need for external carbon sources and aeration. High ammonia concetration and low biodegradable anaerobic digester of swaine wastewater provided an advantage for the growth of anammox microorangism. An anoxic/oxic (A/O) SBR and an anammox SBR were implemented parallelly to treat the same swine wastewater with partial nitrification/denitrification and partial nitrification/anammox process, respectively, and to compare their nitrogen removal efficiency. The nitrogen removal rates (NRRs) of the A/O SBR and anammox SBR were 0.054 and 0.26 kg-N/m3/day, respectively. The lower NRR of the A/O SBR could be attributed to insufficient biodegradable organic carbon sources in the denitrification process. The kinetic parameters obtained from the two SBRs were applied to estimate the time required for using the A/O process and partial nitrification/anammox process to treat the same amount of ammonia with the same reaction volume. Results showed that the A/O process required 3.3 times the reaction time of the partial nitrification/anammox process, suggesting that the partial nitrification/anammox process is a more efficient and economic nitrogen removal process for swine wastewater treatment. The next generation sequencing results revealed that Candidatus Brocadia, ranging from 10 to 23%, was the predominant anammox bacteria in the anammox SBR. More than 78.2 % of nitrite in the anammox SBR was removed through the anammox reaction.

Exploring Aeration Strategies for Enhanced Simultaneous Nitrification and Denitrification in Membrane Aerated Bioreactors: A Computational Approach.

In this study we employ computational methods to investigate the influence of aeration strategies on simultaneous nitrification-denitrification processes. Specifically, we explore the impact of periodic and intermittent aeration on denitrification rates, which typically lag behind nitrification rates under identical environmental conditions. A two-dimensional deterministic multi-scale model is employed to elucidate the fundamental processes governing the behavior of membrane aerated biofilm reactors (MABRs). We aim to identify key factors that promote denitrification under varying aeration strategies. Our findings indicate that the concentration of oxygen during the off phase and the duration of the off interval play crucial roles in controlling denitrification. Complete discontinuation of oxygen is not advisable, as it inhibits the formation of anaerobic heterotrophic bacteria, thereby impeding denitrification. Extending the length of the off interval, however, enhances denitrification. Furthermore, we demonstrate that the initial inoculation of the substratum (membrane in this study) influences substrate degradation under periodic aeration, with implications for both nitrification and denitrification. Comparison between continuous and periodic/intermittent aeration scenarios reveals that the latter can extend the operational cycle of MABRs. This extension is attributed to relatively low biofilm growth rates associated with non-continuous aeration strategies. Consequently, our study provides a comprehensive understanding of the intricate interplay between aeration strategies and simultaneous nitrification-denitrification in MABRs. The insights presented herein can contribute significantly to the optimization of MABR performance in wastewater treatment applications.

Moving-bed biofilm reactor (MBBR) alleviates the negative effect of methylisothiazolinone on the nitrification-denitrification process

Methylisothiazolinone (MIT) is an emerging contaminant used as a biocide in housecleaning and skin care products. Therefore, MIT can reach civil wastewater treatment plants due to domestic usage, thus potentially inhibiting the biological process. This study aims to investigate, for the first time, the effect of MIT on the nitrification-denitrification process in a moving-bed biofilm reactor (MBBR). After a preliminary study performed in a fed-batch reactor, two MBBRs were simultaneously operated in semi-continuous mode for 136 days evaluating a concentration of MIT in the influent of 0.1 and 1.0 mg/L. A parallel study was also conducted under anaerobic conditions to evaluate the effect of MIT on methane production and volatile fatty acid accumulation. At a concentration of 0.1 mg MIT/L, the nitrification process experienced an initial complete inhibition. Subsequently, nitrifying bacteria gradually recovered, reaching approximately 49 % oxidation of ammonia nitrogen, a removal improved up to 64 % at 1.0 mg MIT/L. Although denitrification dealt with a temporary inhibition, the process ultimately achieved a 100 % decrease in nitrite and nitrate concentrations when the MIT concentration was increased from 0.1 to 1.0 mg/L. The cumulative methane yield reached approximately 134 mL CH4/g VS for the control group and was similar to the methane production obtained in the presence of MIT. A MIT removal of 100 % was achieved under aerobic, anoxic, and anaerobic conditions.

Harnessing the Potential of Extracellular Polymeric Substances in Enhancing ANAMMOX Processes: Mechanisms, Strategies, and Perspectives

Anaerobic ammonium oxidation (ANAMMOX) has emerged as a promising sustainable nitrogen removal technology that offers significant advantages over conventional nitrification–denitrification processes, such as reduced energy consumption, a 60% reduction in oxygen demand, and a 90% reduction in sludge production. However, the practical application of ANAMMOX is hindered by several challenges, including the slow growth of ANAMMOX bacteria, long start-up periods, and high sensitivity to environmental disturbances. Recent studies have highlighted the crucial role of extracellular polymeric substances (EPSs) in the formation, activity, and stability of ANAMMOX biofilms and granules. An EPS is a complex mixture of high-molecular-weight polymers secreted by microorganisms, mainly composed of polysaccharides, proteins, nucleic acids, and lipids. The diverse physicochemical properties and functional groups of EPSs enable them to serve as a structural scaffold, protective barrier, sorption site, electron shuttle, and nutrient source for ANAMMOX bacteria. This review aims to provide an overview of the latest research progress on harnessing the potential of EPSs to enhance the ANAMMOX process. The characteristics, compositions, and extraction methods of ANAMMOX-derived EPSs are summarized. The mechanisms of how EPSs facilitate the enrichment, immobilization, aggregation, and adaptation of ANAMMOX bacteria are elucidated. The strategies and effects of EPS supplementation on improving the performance and robustness of ANAMMOX reactors under various stresses are critically reviewed. The challenges and future perspectives of the EPS-mediated optimization of the ANAMMOX process are also discussed. This review sheds new light on exploiting EPSs as a renewable bioresource to develop more efficient and stable ANAMMOX applications for sustainable wastewater treatment.

Ecological filter walls for efficient pollutant removal from urban surface water

Urban surface water pollution poses significant threats to aquatic ecosystems and human health. Conventional nitrogen removal technologies used in urban surface water exhibit drawbacks such as high consumption of carbon sources, high sludge production, and focus on dissolved oxygen (DO) concentration while neglecting the impact of DO gradients. Here, we show an ecological filter walls (EFW) that removes pollutants from urban surface water. We utilized a polymer-based three-dimensional matrix to enhance water permeability, and emergent plants were integrated into the EFW to facilitate biofilm formation. We observed that varying aeration intensities within the EFW's aerobic zone resulted in distinct DO gradients, with an optimal DO control at 3.19 ± 0.2 mg L−1 achieving superior nitrogen removal efficiencies. Specifically, the removal efficiencies of total organic carbon, total nitrogen, ammonia, and nitrate were 79.4%, 81.3%, 99.6%, and 79.1%, respectively. Microbial community analysis under a 3 mg L−1 DO condition revealed a shift in microbial composition and abundance, with genera such as Dechloromonas, Acinetobacter, unclassified_f__Comamonadaceae, SM1A02 and Pseudomonas playing pivotal roles in carbon and nitrogen elimination. Notably, the EFW facilitated shortcut nitrification-denitrification processes, predominantly contributing to nitrogen removal. Considering low manufacturing cost, flexible application, small artificial trace, and good pollutant removal ability, EFW has promising potential as an innovative approach to urban surface water treatment.

Revealing the differences in metabolisms and nitrogen removal mechanisms of the full and shortcut nitrification-denitrification processes for the treatment of livestock wastewater

The full nitrification-denitrification process (FN) and shortcut nitrification-denitrification process (SN) are both important approaches for treating livestock wastewater. However, most existing studies focus on pollutant removal performance in wastewater treatment, while the differences between the two processes in terms of bacterial communities, metabolic pathways, nitrogen removal mechanisms, and potential pathogen threats have been poorly investigated. In this study, the SN process exhibited lower oxygen demand, reduced sludge production, higher nitrogen removal rate, and lower pathogen threat for treating livestock wastewater. The bacterial communities, the primary participants of pollutant removal in wastewater treatment, were significantly different between the two processes. In the FN process, 15 genera were significantly enriched, including Saccharimonadales, Micropruina, Ferruginibacter, etc. On the other hand, the SN process showed enrichment of 10 genera, including Brachymonas, Tessaracoccus, Ottowia, etc. These enriched genera, such as Brachymonas, Tessaracoccus, and Ottowia in the SN process, were positively correlated with the denitrification genes, facilitating a higher nitrogen removal rate. Moreover, the FN process exhibited significantly higher amino acid metabolism and reproduction-related pathways, contributing to its higher alpha-diversity of the bacterial community. The significantly enriched genes involved in assimilatory nitrate reduction and nitrogen assimilation pathway suggested that nitrogen assimilation played a crucial role in ammonia nitrogen removal, but it also increased sludge production in the FN process. Additionally, the FN process had a higher number and abundance of potential pathogens, indicating a greater pathogen threat. In conclusion, this study provides valuable insights into the differences between the FN and SN processes in terms of bacterial communities, nitrogen removal mechanisms, and potential pathogen threats. It also offers theoretical guidance for adopting appropriate strategies in wastewater treatment.

Metabolic Rates of Rainbow Trout Eggs in Reconstructed Salmonid Egg Pockets

In situ evaluations of the metabolic rates (i.e., respiration and excretion) of salmonid eggs are mostly indirect, focusing on the sampling of hyporheic water from wild or artificial nests. Comparatively, experimental studies carried out under controlled, laboratory conditions are less abundant due to methodological difficulties. This study presents a novel experimental setup aimed to address this issue and enable the measurement of oxygen and dissolved inorganic nitrogen fluxes in simulated rainbow trout (O. mykiss) egg pockets. The experimental setup consists of reconstructed egg pockets in cylindrical cores under flow-through conditions. Live and dead eyed-stage eggs were incubated in a natural, sterilised gravel substrate. Hyporheic water circulation was ensured using peristaltic pumps, with the possibility to collect and analyse inflowing and outflowing water for chemical analyses. Microcosm incubations, with closed respirometry of eggs in water alone, were also carried out in order to infer the importance of microbial respiration in the simulated egg pockets. The results show an increasing trend in oxygen demand, due to the development of biofilm in the reconstructed egg pockets and increased egg respiration rates. Moreover, egg pockets showed positive ammonium net fluxes connected with the advancing developmental egg stage, while nitrate removal peaked during the last phase of the experiment, mainly due to the formation of oxic-hypoxic interfaces, leading to couple nitrification–denitrification processes. The suggested approach enables to test a number of in situ situations, including the effects of extreme hydrological conditions, sediment clogging and sudden changes in water chemistry or temperature on the survival and metabolic performances of nests, at different egg development stages.

Potential Contributions of Ammonia‐Oxidizing Microorganisms to the Distributions of Nitrous Oxide in the Northern Bering Sea

AbstractOceanic N2O is a major source of atmospheric N2O gas and is involved in global warming and ozone depletion. It is thought to be mainly produced by nitrification, denitrification and nitrifier denitrification processes mediated by ammonia‐oxidizing bacteria, ammonia‐oxidizing archaea (AOA) and denitrifying bacteria. The Bering Sea, especially its continental shelf area, is considered a typical source of atmospheric N2O. During the 7th Chinese National Arctic Research Expedition (CHINARE2016), the distributions of N2O and ammonia‐oxidizing microorganisms (AOOs) in the Bering Sea continental shelf and abyssal basin water were investigated. At a depth of 50∼900 m within the abyssal basin, the in‐situ ammonia oxidation process, particularly performed by AOA, exhibits considerable potential for the formation of supersaturated N2O. Meanwhile, beneath the oxygen minimum zone (depth range is approximately 800∼1,000 m), supersaturated N2O is primarily driven by mixing processes, while the ammonia oxidation mediated by AOA also contributes to a certain extent. In addition, the N2O distribution characteristic exhibits a substantial disparity between the southern and northern Bering Sea shelves, with the former characterized as a mild sink and the latter as a weak source. The water column of the Bering Sea demonstrates a considerable potential for generating supersaturated N2O through ammonia oxidation, as corroborated by the current study.

Full nitrogen and phosphorus removal in the PASDEBPR system

Photo-activated sludge (PAS) system aims to utilize microalgae to deliver oxygen for bacterial respiration, eliminating the need for external aeration. However, research on the treatment potential of PAS systems in the removal of nutrients is limited. In this context, a research study was devised to evaluate the possibility of developing a microalgae-bacteria consortium to achieve the simultaneous removal of organic carbon, nitrogen, and phosphorus. A successful PAS system capable of removing phosphorus was established at the end of the first phase, with an effluent phosphorus (P) concentration of 1.6 mg P L−1. In the subsequent stage, during the introduction of the nitrification-denitrification process, the system lost stability and deteriorated. Interestingly the system recovered via the sparging of nitrogen gas reaching effluent concentrations of 1.22 mg P L−1 and 0.88 mg N L−1. Thus, the system was capable of removing phosphorus and nitrogen via biological means without the need for external aeration. It is hypothesized that the inhibition caused was due to the production of a gaseous compound during the nitrification/denitrification process.

Study of bacterial population dynamics in seed culture developed for ammonia reduction from synthetic wastewater.

The waterbodies have been polluted by various natural and anthropogenic activities. The aquatic waste includes ammonia as one of the most toxic pollutants. Several biological treatment systems involving anoxic and semi anoxic bacteria have been proposed for reducing nitrogen loads from wastewater and increasing the efficiency and cost effectiveness. These bacteria play a vital role in the processes involved in the nitrogen cycle in nature. However, the enrichment, sustainability and identification of bacterial communities for wastewater treatment is an important aspect. Most of the chemolithotrophs are unculturable hence their identification and measurement of abundance remains a challenging task. In this study the different bacteria involved in total nitrogen removal from the wastewater are enriched for 700days under anoxic condition. The synthetic wastewater containing 0.382g/L of ammonium chloride was used. Molecular identification of the bacteria involved in various steps of the nitrogen cycle was carried out based on amplification of functional genes and 16S rRNA gene Polymerase chain reaction followed by DNA sequencing. Change in the abundance of chemolithotrophs was studied using qPCR. The mutual growth of various nitrifiers along with anaerobic bacteria were identified by molecular characterisation of DNA at various time intervals with the different genes involved in the nitrogen cycle. Nitrosomonas species like Nitrosomonas europaea were identified throughout the batch scale studies possessing the genes associated with ammonia oxidizing bacteria and nitrite oxidizing bacteria which act as a complete ammonia oxidizer. The uncultured species of Nitrospira and anammox bacteria were also observed which predicts the coexistence of the anammox and comammox bacteria in a batch scale study. The coexistence of the semi anoxic and anoxic bacteria helped in the growth of these bacteria for a longer duration of time. The nitrite produced by the comammox during nitrification can be utilized by anammox as an electron carrier. The other species of denitrifiers like Pseudomonas denitrificans and Aminobacter aminovorans were also observed. It is concluded that the enrichment of semi anoxic and anoxic bacteria was faster with the increase in growth of the bacteria involved in nitrification, comammox, anammox and partial denitrification process. The bacterial growth is enhanced and the efficiency is increased which can be further used in the development of small pilot scale bioreactor for total nitrogen removal.

Simulation assisted process simplification and energy recovery from a cryptic biological nutrient removal plant

In this study, simulation-based analysis is conducted for nitrogen removal mechanism in a biological nutrient removal plant designed in an encrypted manner. In the first phase, the simulation study elucidates how nitrogen removal occurs in a complex design using a series of 4 carousel bioreactors with additional nitrate recycle. It is demonstrated that pre-anoxic volume fed with limited internal recirculation only would not be sufficient to meet the discharge Total Nitrogen (TN) limit of 10 mg N/L. However, the addition of simultaneous nitrification-denitrification (SND) processes in aerobic reactors enables compliance with TN discharge limits. In the second phase, the configuration is simplified by means of process simulation, where the complex design is streamlined by eliminating internal recycle while achieving nitrogen removal through the SND process. Plant data confirmed that the simplified configuration can achieve nitrogen removal providing the same discharge quality. The simulations resulted that TN concentration achieved at the plant effluent is comparable, while effluent total phosphorus concentration is decreased by approximately 50 % due to increased activity of Phosphorus Accumulating Organisms (PAOs) under reduced sludge retention time (SRT). In the third phase, the application of the SND process in the field using a simplified and new activated sludge configuration yields similar results to the simulation. According to plant records, 28 % energy savings and an equivalent chemical cost reduction of €945,000 per year were achieved with the new process configuration.

Bioelectricity recovery through isolation and removal of nitrogen from wastewater by shortcut nitrification-denitrification in a coupled microbial fuel cell system

Achieving both sustainable power recovery and efficient nitrogen removal in a single microbial fuel cell (MFC) reactor is a big challenge in the selection of external resistance and choosing favorable conditions. This study introduced a coupled MFC system (N-MFC and D-MFC) with a unique approach of isolating nitrogen input from wastewater and removing it via shortcut nitrification–denitrification, achieving treatment of wastewater along with improved energy recovery. The configuration of the N-MFC facilitated it to contribute significantly to the 98 % COD removal and to the 1.27 W m−3 energy recovered of the coupled MFC system. The results highlighted that operational circumstances affected MFC performance and offered insights into power generation, ammonium isolation, and shortcut nitrification in the N-MFC. The ratio of nitrite to nitrate was maximized at 2 under the dissolved oxygen (DO) condition of 0.6 mg/L, as opposed to 1.44 under the DO condition of 1.2 mg/L, during the shortcut nitrification process in the cathode chamber of N-MFC. The total nitrogen removal efficiency of the coupled MFC system was obtained at 50.2–71.2 %, proportionate to the COD influent and nitrite-to-nitrate ratio. The nitrogen removal of the coupled MFC system was significantly aided by autotrophic denitrification in the cathode chamber of D-MFC. The finding of this study emphasized the potential for optimizing the nitrogen removal in the D-MFC to increase the performance of N-MFC in power recovery by implementing the shortcut nitrification–denitrification process. This concept can be applied in practical for meeting the goals of SDG6 (Sustainable Development Goals) and clean energy production.

Adaptive evolutionary strategies of microbes to manganese in nitrogen-culturing sludge

Human activities have significantly increased the accessibility of nitrogen and manganese, which has had an impact on the evolution of microorganisms. To investigate the effect of increasing manganese accessibility on microbial nitrogen cycle, Mn-HA (Manganese -Humus chelates), a naturally occurring compound in the form of organochelate, was introduced into the conventional nitrification-denitrification process. The findings revealed that the addition of Mn-HA significantly facilitated the removal of ammonium and nitrate potentially by Mnammox (anaerobic ammonium oxidation mediated by Mn (IV)-oxide reduction) and Mn-based autotrophic denitrification. Metagenomic analyses suggested the presence of manganese-redox microorganisms involved in nitrification and denitrification, indicating the occurrence of manganese redox-coupled nitrogen metabolism processes. Additionally, the production and consumption of bio-MnOx (bio-Manganese Oxides) were observed in conjunction with denitrification and nitrification. The production of unequal amounts of bio-MnOx suggested that bio-MnOx may have been deposited in the sediment. This was confirmed by the BCR sequential extraction experiment. Coupling Mn metabolism with N metabolism is an adaptive strategy for nitrogen metabolizing microorganisms. For Mn-sensitive populations, they gain respite from Mn-rich environments by adopting strategies such as manganese oxidation, regulating manganese transport, pumping out intracellular manganese and evolving other manganese resistance genes.

Impacts of substrate properties and aquatic nutrient concentrations on the relative abundance of nitrifying/denitrifying genes and the associated microbes in epilithic biofilms.

Substrates like sand or gravels and aquatic nutrient concentrations of rivers are highly heterogeneous, influencing the abundance of functional genes in epilithic biofilms where nitrification-denitrification processes take place. To analyze how the relative abundance of nitrifying/denitrifying genes and the associated microbes changes with the physical properties of substrates and aquatic concentrations of nutrients, this paper utilized metagenomics to comprehensively characterize these functional genes (i.e., amoA, hao, and nxrB involved in nitrification, and napA, narG, nirS, norB, and nosZ associated with denitrification) from epilithic biofilms collected along the Shitingjiang River in Southwest China and further obtained the relative abundance of major nitrifiers and denitrifiers. The results show that substrate size most significantly affects the relative abundance of hao and norB by altering the hydrodynamic conditions. In sampling sites with high heterogeneity in substrate size distribution, the relative abundance of most denitrifying genes is also higher. The carbon-nitrogen ratio negatively correlates with the relative abundance of all the nitrifying genes, while ammonium, total inorganic carbon, and total organic carbon concentrations positively affect the relative abundance of amoA and nxrB. As to the relative abundance of nitrifiers and denitrifiers, mainly belonging to phyla Proteobacteria and Actinobacteria, substrate heterogeneity and the aquatic concentrations of nutrients have greater influences than substrate size. Also, the substrate heterogeneity exerted positive influence on functional species of Pseudogemmobacter bohemicus and Paracoccus zhejiangensis. Considering the genes' functions and the dominant species linked to denitrification, nitrous oxide is more likely to occur in rivers with higher heterogeneity and larger substrates.

Mechanisms and responses of nitrogen transformations mediated by microbial communities in constructed tailwater wetlands to salinity stress

The long-term salinity stress may affect the nitrogen transformation processes and microbial community assembly patterns in constructed wetlands treating tailwater, influencing their operational effectiveness. In this study, we employ 16S rRNA gene sequencing, combined with multivariate ecological and statistical methods, to investigate interactions between bacterial community diversity and nitrogen transformation genes in constructed tailwater wetlands under salinity stress. The results revealed that long-term salinity stress led to reduced microbial diversity and increased community heterogeneity. Additionally, salinity stress led to decreased functionality of microbial community, intensified competition, and elevated expression of functional genes. Furthermore, as predicted by PICRUSt2, long-term salinity stress resulted in overall inhibition of community nitrogen conversion functions, while short-cut nitrification-denitrification processes played a bigger part in nitrogen conversion. Moreover, the symbiotic nitrogen fixation process was promoted, while the competing dissimilatory nitrate reduction process was inhibited. Salinity can greatly explain the apparent differences in microbial communities and nitrogen transformation potential in saline constructed wetland systems compared to common wetlands. These findings provide novel insights into the mechanisms underlying nitrogen transformation processes in constructed tailwater wetlands under long-term salinity stress and important implications for redesign and renovations of constructed wetlands.

Deciphering antibiotic resistome variations during nitrogen removal process transition under mixed antibiotics stress: Assembly process and driving factors

Antibiotic resistome, which encompasses all types of antibiotic resistance genes (ARGs) in a given environment, has received increasing attention in research on different wastewater treatment processes. However, the variation in antibiotic resistome during the transition from the full nitrification–denitrification to the shortcut nitrification–denitrification process remains unclear. In this study, a total of 269 targeted gene subtypes were identified, along with 108 genes were consistently present in all samples. The introduction of mixed antibioticsrapidly increased the abundance of corresponding and non-corresponding ARGs, as well as that of mobile genetic elements.The variations in of the antibiotic resistome were primarily driven by dissolved oxygen and nitrite accumulation rate. Moreover, 34 bacterial genera were identified as potential ARG hosts, with most denitrifiers considered as potential antibiotic-resistant bacteria, including Branchymonas, Rhodobacter, and Thauera. This study provides a method for controlling antibiotic resistance by regulating the changes in environmental variables and bacterial communities.

Prolonged flooding followed by drying increase greenhouse gas emissions differently from soils under grassland and arable land uses

Several climate change scenarios have predicted that heavy precipitation could result in prolonged flooding (PF) and flooding–drying (FD) of soils under agriculture. The influence of PF and FD on soil greenhouse gas (GHG) fluxes and ammonium‑nitrogen (NH4+-N) and nitrate‑nitrogen (NO3−-N) dynamics of arable and grassland soils, the dominant land-use types in the UK, remain unclear. A two-month soil incubation experiment was conducted to determine the impact of PF and FD on soil N dynamics and GHG fluxes from arable and grassland soils. Arable soil emitted more N2O-N when soil moisture exceeded 100% water-holding capacity (WHC) compared to grassland soil under PF. Grassland soils exhibited increased N2O-N emissions than arable soils when soil moisture was lower than 100% WHC under FD. When soil moisture exceeded 100% WHC, the available NO3−-N in the soil contributed 58% of N2O-N emissions potentially by denitrification from grassland. When soil moisture was lower than 100% WHC, soil NH4+-N and NO3−-N contributed 71% of N2O-N emissions, which suggests coupling of nitrification-denitrification processes in driving high emissions from grassland soils. The N2O-N and CO2-C emissions increased with the incubation time under FD. Moreover, FD significantly increased N2O-N, CO2-C, and CH4-C emissions in grassland soil by 0.93, 2.15, and 37.29 times more than arable soil, respectively. These findings points to important tipping points in the source strengths of GHG fluxes from the two land use types differently. Future land use changes should consider the contribution of the changing dynamics of GHG fluxes in light of climate extremes and its implications for net zero greenhouse gas emission ambitions.

Pilot-scale demonstration of self-enrichment of anammox bacteria in a two-stage nitrification-denitrification suspended sludge system treating municipal wastewater under extremely low nitrogen loading rate

In suspended sludge system, efficient enrichment and retention of anammox bacteria are crucial obstacles in mainstream wastewater treatment by anammox process. In this study, anammox bacteria was self-enriched in a pilot-scale suspended sludge system of two-stage nitrification–denitrification process serving municipal wastewater treatment. With the low ammonia (NH4+-N) of 9.3 mg/L, nitrate (NO3–-N) of 15.6 mg/L and COD/NO3–-N of 2.2 under extremely low nitrogen loading rate of 0.012 kg N/m3/d, anammox activity bloomed after its abundance increasing from 5.9 × 107 to 4.6 × 109 copies/g dry sludge. Significant NH4+-N removal was occurred and maintained stably in the denitrification reactor with anammox bacteria accounting for 1.13%, even under temperature decreasing to 20.0℃. The adequately anoxic environment, efficient retention with the static settlement, and NO2– production via NO3– reduction provided favorable environment for anammox bacteria. This study demonstrated the feasibility and great potential in mainstream anammox application without seeding specific sludge.
