Monoclonal antibodies to chromaffin cells can distinguish proteins specific to or specifically excluded from chromaffin granules

I have prepared a number of monoclonal antibodies to chromaffin cell membranes. One of these antibodies recognizes a number of antigenically related proteins that are present in all tissues examined. In the adrenal, these proteins are completely excluded from chromaffin granules but are present in other subcellular membrane fractions. This non-granule membrane-specific antibody has been designated NG3. A second antibody, CG7, binds to a single protein which segregates specifically into chromaffin granules. The protein recognized by CG7 is cytochrome b 561, or chromomembrin B, one of the major protein components of chromaffin granule membranes. CG7 also labels a protein (the identical cytochrome b 561) in bovine posterior pituitary neurosecretory vesicle membranes indicating that it functions in both peptidergic and catecholaminergic secretory granules. These two monoclonal antibodies provide useful probes of both granule and extra-granule membrane proteins for studies of membrane trafficking in chromaffin cells

Ultrastructural localization of gamma amino butyric acid immunoreactivity in B cells of the rat pancreas.

The purpose of this study was to determine the ultrastructural localization of gamma amino butyric acid (GABA) within the B cell of isolated rat islets, particularly with respect to the B cell secretion granules. GABA immunoreactivity was localized ultrastructurally, with colloidal gold, in the B cells and absent in the A cells and D cells. Quantitative analysis of the colloidal gold particle distribution in the B cell revealed 29.5 +/- 5.2 gold particles/micron 2 in the nuclei, 29.3 +/- -6.9 gold particles/micron 2 in the mitochondria and 4.9 +/- 1.0 gold particles/micron 2 in the secretion granules. Particle density in the remainder of the cytoplasm was 41.9 +/- 4.1 particles/micron 2. The paucity of gamma amino butyric acid in the B cell secretion granules differs from observations on gamma amino butyric acidergic neurons, where there is an accumulation of gamma amino butyric acid within the neurosecretory vesicles. These findings indicate that if gamma amino butyric acid is released from the islet, then it is by a nongranular mechanism. In addition, the results are compatible with the hypotheses that gamma amino butyric acid within the B cell functions in the regulation of insulin biosynthesis, and/or functions as an alternative energy source for the B cell through the gamma amino butyric acid shunt.

Ultrastructural Features of the Cells of Dufour's Gland and Associated Structures in the Carpenter Bee, Xylocopa virginica virginica (L.) (Hymenoptera: Anthophoridae)

The cells of Dufour's gland and of the associated visceral muscle and nerve of the carpenter bee, Xylocopa virginica virginica (L.), were studied with light and electron microscopes. The gland cells revealed short microvilli, interdigitations of plasma membrane-limited projections, associated glycocalyx and cuticle, a single nucleus with one to four nucleoli, many free ribonucleoprotein particles, septate junctions, and numerous mitochondria and microtubules. Many tracheoles accompanied the gland cells, the associated visceral muscle, and unmyelinated nerve. The muscle cells contained myofibrils, composed of thin and thick myofilaments. The myofilaments were somewhat greater in diameter than usually encountered, and their disposition patterns were irregular. Transverse tubules, A and I bands, and Z lines were present, but no H bands or dyads were seen. The nerve cell processes contained mitochondria, neurotubules, and, as they approached muscle cells, neurosecretory vesicles with electron-dense or electron-transparent contents. Carpenter bee Dufour's glands are compared with those of the megachilid Osmia cornifrons (Radoszkowski).

CRF: its regulation of ACTH and pro-opiomelanocortin peptide release and its extra hypothalamic occurrence

CRF-41 and vasopressin are the major part of the hypothalamic complex responsible for the release of ACTH. The potentiated response of the corticotroph to mixtures of the peptides and their co-localization in the same neurosecretory vesicles would indicate their obligatory co-secretion in many stress situations. The occurrence of CRF at peripheral sites especially in the placenta and in the plasma of women in their third trimester of pregnancy is suggestive of further roles for this peptide although care should be exercised in interpreting simple immunohistochemical staining in certain tissues. When ACTH is released from the pituitary several other peptides which form part of its precursor are co-released. The effect of these peptides on the adrenal gland and the way their activity is expressed has led to a post-secretional proteolytic control mechanism being proposed.

The Subgenus Persicargas (Ixodoidea: Argasidae: Argas) 44. Ultrastructure of the lateral organs of A. (P.) arboreus

The structure of the lateral organs of adult Argas (Persicargas) arboreus is described with the aid of the transmission electron microscope. Four pairs of small, ganglionlike bodies are attached to the hemal nerve branches arising from the pedal nerves near their origin from the central nerve mass. Each organ is ensheathed by a thin neural lamella continuous with that of the corresponding nerve. Neuronal cell bodies with axons probably extending into the nerve are present in the organ. The cell cytoplasm is rich in smooth endoplasmic reticulum and contains dispersed rough endoplasmic reticulum, free ribosomes, mitochondria, coated vesicles in the Golgi area, membrane-bound vacuoles, and lysosomelike bodies. Some cells contain neurosecretory vesicles of variable electron densities. These substructures, which have not previously been observed, suggest the neuroendocrine nature of the lateral organs of A. (P.) arboreus .

Effect of monensin on synthesis, post-translational processing, and secretion of dopamine beta-hydroxylase from PC12 pheochromocytoma cells.

Monensin was used to ascertain the location in the biosynthetic pathway where the 77,000-Mr membrane-bound subunit form of dopamine beta-hydroxylase is post-translationally converted to the 73,000-Mr soluble form. Treatment with low concentrations of monensin (less than or equal to 50 nM) completely depleted the cells of the norepinephrine and dopamine, had a small effect on protein synthesis, and enhanced post-translational processing of only dopamine beta-hydroxylase which was previously synthesized and presumably packaged into neurosecretory vesicles. At these low concentrations, exit from the Golgi apparatus did not appear to be blocked since stimulated secretion of a group of high molecular weight [35S]methionine-labeled proteins was not inhibited. Treatment with higher concentrations of monensin (200 nM) prevented the secretion of the [35S] methionine-labeled proteins normally released with a secretagogue, and also prevented the secretion of [3H] mannose-labeled proteins including dopamine beta-hydroxylase. Surprisingly, a group of lower molecular weight [35S]methionine-labeled proteins was now released from monensin-treated cells. Treatment with high concentrations of monensin (greater than or equal to 200 nM) appeared to block the secretory pathway prior to the packaging step, probably in the Golgi apparatus. If the proteins were packaged prior to monensin treatment, they were released upon stimulation with secretagogues. Monensin treatment (200 nM) enabled the post-translational processing of newly synthesized dopamine beta-hydroxylase, from the 77,000-Mr to the 73,000-Mr subunit form, to go to completion. The susceptibility of this 73,000-Mr subunit form to endoglycosidase H digestion was unaltered, suggesting that dopamine beta-hydroxylase from monensin-treated cells may have the same high mannose oligosaccharide content as native dopamine beta-hydroxylase. These experiments indicate that the post-translational processing of dopamine beta-hydroxylase occurs in the Golgi apparatus and may continue in immature granules prior to their acidification.

Annulate lamellae and whorl bodies in the diffuse supraoptic nucleus of the hamster.

The ultrastructure of neurons of the diffuse supraoptic nucleus of the hamster has been studied. These neurons show two specializations of the endoplasmic reticulum: annulate lamellae and whorl bodies. From one to three whorl bodies are found in the same neuron. The annulate lamellae and the whorl body cisterns are continuous with the cisterns of the rough endoplasmic reticulum. These neurons present an extraordinarily developed rough endoplasmic reticulum, small mitochondria, neurosecretory vesicles and a Golgi complex filled with electron-dense material. Astrocytic processes of different thickness surround the neurosecretory cells.

Coordinate Expression of Hypothalamic Pro-Dynorphin and Pro-Vasopressin mRNAs with Osmotic Stimulation

Peptides derived from pro-dynorphin and pro-vasopressin precursors coexist within neurosecretory vesicles of magnocellular neurons in the rat hypothalamus projecting to the posterior pituitary. The secretory activity of these neurons can be stimulated using physiological manipulations known to increase plasma vasopressin levels, such as dehydration and salt-loading. With chronic osmotic challenge, the mRNAs for both pro-dynorphin and pro-vasopressin increase in parallel in the supraoptic and paraventricular nuclei of the hypothalamus, but not within the nonmagnocellular suprachiasmatic nucleus cell groups projecting elsewhere than the neural lobe. The results indicate an example of coordinate regulation of mRNA expression for coexisting peptides within the brain.

Ultrastructural immunolocalization of vasopressin and neurophysin in neurosecretory cells of dehydrated rats

It has been proposed that neurosecretory material may be transported in a non-vesicular compartment in magnocellular neurons in the hypothalamus of osmotically stimulated rats. We have reexamined this issue using postembedding electron microscopic immunoperoxidase labeling of the secreted peptides (vasopressin and neurophysin) found in these neurons. Reaction product was found exclusively over neurosecretory vesicles in cell bodies, and in axons in the median eminence and neurohypophysis. The findings are consistent with an exclusive secretory vesicle location of the neuropeptides in the hypothalamo-neurohypophysial system of both normal and dehydrated rats.

Dual ultrastructural localization of two neurotransmitter-related antigens: colloidal gold-labeled neurophysin-immunoreactive supraoptic neurons receive peroxidase-labeled glutamate decarboxylase- or gold- labeled GABA-immunoreactive synapses

To study the morphological substrate for interaction between two chemically distinct neuronal types, two double ultrastructural immunolabeling strategies were employed. In the first, two different electron-dense markers were used to examine simultaneously two different neurotransmitter-related antigens in the hypothalamic supraoptic nucleus in the same thin section. Results obtained with the first method were confirmed with a second approach based on postembedding immunostaining of alternate serial thin sections with different antisera. Antiserum against glutamate decarboxylase, the enzyme responsible for the synthesis of the inhibitory amino acid transmitter gamma-aminobutyric acid (GABA), or antisera against GABA, was used to localize immunoreactive axons in the hypothalamic supraoptic nucleus. With light microscopy, glutamate decarboxylase- and GABA-immunoreactive axon terminals immunostained with peroxidase were found arborizing throughout all areas of the nucleus; terminal boutons were found adjacent to unlabeled somata within the nucleus. Cells containing immunoreactive oxytocin, vasopressin, and neurophysin were localized with peroxidase. Glutamate decarboxylase-immunoreactive axons stained with peroxidase prior to embedding in plastic were demonstrated to contact neurons which contained vesicles immunostained with neurophysin antiserum by a post-embedding immunocytochemical procedure which used immunoglobulins or protein A adsorbed to colloidal gold as a second ultrastructural marker. Quantitative evaluation of post-embedding staining with colloidal gold using a neurophysin primary antiserum indicated a specific antigen localization in neurosecretory vesicles. A critical factor in this double-labeling paradigm was that immunological reagents used in the second series did not cross-react with those used in the first series, regardless of the species of origin of antisera. To provide further verification of GABAergic synapses on neurophysin-containing neurons, alternate serial ultrathin sections were stained with colloidal gold using antisera against either neurophysin or GABA; boutons immunoreactive for GABA made synaptic contact with supraoptic neurons containing neurophysin immunoreactivity. Converging results obtained with these two procedures indicate that GABAergic axons synapse directly on neurons containing oxytocin or vasopressin in the rat hypothalamic supraoptic nucleus.(ABSTRACT TRUNCATED AT 400 WORDS)

Co-localization of corticotropin-releasing factor and vasopressin in median eminence neurosecretory vesicles.

Vasopressin (VP) potentiates the effect of corticotropin-releasing factor (CRF) on the secretion of adrenocorticotropic hormone (ACTH) from anterior pituitary cells in vitro, and both CRF and VP have been found in portal blood. These data support the hypothesis that VP acts synergistically with CRF to cause the secretion of ACTH in vivo but the origin of the CRF and VP, and the physiology of their release, have not been precisely defined. Parvocellular cell bodies in the paraventricular nucleus (PVN) which project to the external zone of the median eminence can be stained for both CRF and VP after adrenalectomy, and there is light microscopic immunocytochemical evidence that neurophysin (NP) may be located within some of the CRF-containing axons. Electron microscopic immunocytochemical studies have demonstrated the presence of CRF, VP and its 'carrier' protein, VP-associated neurophysin (NP-VP) in 100-nm neurosecretory vesicles (NSVs) in axons terminating near the portal capillary plexus in the external zone of the median eminence. If these peptides are extensively co-localized in the same NSVs in the median eminence, then coordinate secretion of CRF and VP in vivo is obligatory, at least in some physiological circumstances. We demonstrate in this report, using post-embedding electron microscopic immunocytochemistry on serial ultrathin sections, that CRF, VP and NP-VP are contained not only in the same axons and terminals, but in the same 100-nm NSVs in the median eminence of both normal and adrenalectomized rats. In addition, in the normal rat median eminence 44% of the CRF-positive axons and terminals stained strongly for VP and NP-VP, whereas in the adrenalectomized rat virtually all the CRF-positive structures in the median eminence showed strong staining for VP and NP-VP, indicating a transformation of one subpopulation of CRF-positive axons and terminals by adrenalectomy.

Mucosal nerves and smooth muscle relationships with gastric glands of the opossum: An ultrastructural and three‐dimensional reconstruction study

The precise anatomical relation by which autonomic nerve endings contact gastric epithelial cells to enhance the rate of gastric secretions is not fully understood. The aim of the present study was to clarify this issue by using the technique of serial section reconstruction of areas of the gastric mucosa. The work also explored the possibility of a functional role for a system of smooth muscle strands in the gastric mucosa that emanate from the muscularis mucosa, run in the lamina propria, and are associated in a unique manner with the gastric glands. Electron microscopic serial sections of the gastric mucosa were performed to visualize the entire limiting membrane of gastric epithelial cells to determine any nerve associations (especially varicose endings) with these cells. Evaluation of serial sections of five separate parietal cells showed that their basal membrane did not come in close contact (nearest distance 500 nm) with any nerve axon or varicosity. Moreover, the axons passing in the area of these cells ultimately showed varicose endings associated with smooth muscle cells in the adjacent connective tissue (often separated by only 20 nm), with mast cells or with vascular elements. Additionally, the lateral membrane of these five parietal cells did not contact any endocrine cell in the epithelium, although other parietal cells in the area were adjacent to endocrine cells. Chief cells in the immediate area also did not form any close associations with nerve varicosities. Random analysis of 5,000 additional epithelial cells in these sections showed no close associations to nerve elements with significant accumulations of neurosecretory vesicles (varicosities). Because of the observed existence of innervation to the smooth muscle strands in the area of the gastric glands, serial 1-micron epoxy sections of the gastric mucosa were prepared, and profiles of smooth muscle and gastric glands were entered into a computer-assisted reconstruction system. Three-dimensional reconstruction techniques were employed to reveal the existence of a unique association between the mucosal smooth muscle strands and the gastric glands. The muscle strands arose from the muscularis mucosa at regular intervals and became branched to form an intricate wrap around a series of gastric glands that empty into one gastric pit.(ABSTRACT TRUNCATED AT 400 WORDS)

Immunocytochemical identification of dynorphin-containing vesicles in brattleboro rats

Vasopressin and its carrier protein, vasopressin-associated neurophysin, are co-packaged together with an opioid peptide, dynorphin, into 160 nm diameter neurosecretory vesicles in the normal rat hypothalamo-neurohypophysial system. The homozygous Brattleboro rat lacks vasopressin and vasopressin-associated neurophysin, but contains substantial amounts of dynorphin in the vasopressin-deficient neurosecretory cells. We used post-embedding electron microscopic immunocytochemistry to determine the subcellular location of dynorphin in Brattleboro rats. The results show that dynorphin is present within 100 nm neurosecretory vesicles in homozygous Brattleboro cell bodies and axons, and within 160 nm vesicles in heterozygous (control) neurosecretory cell bodies and axons. Oxytocin-associated neurophysin is present in a separate population of magnocellular neurons in both homozygous and heterozygous rats, and is contained within 160 nm vesicles in both cases. Therefore, the absence of synthesis of the vasopressin prohormone results in a dramatic reduction of neurosecretory vesicle size, despite the continued synthesis and packaging of dynorphin peptides.

Opioid biology: the next set of questions.

A range of biologically different opioid peptides are synthesised as components of three distinct precursors, pro-opiomelanocortin, proenkephalin, and prodynorphin. They interact with a number of receptors which have so far been characterised as mu, delta, kappa, sigma, and epsilon. It is unclear which ligands bind to which receptors under physiological circumstances, but preferential in vitro interactions include enkephalins with delta receptors, dynorphin with kappa receptors, and beta-endorphin with epsilon receptors. Post-translational processing determines which of several opioid products are produced from each precursor, but the identity of the enzymes involved and regulation of processing is unknown. Opioid involvement in the neuroendocrine and cardiovascular systems is reviewed. Naloxone-sensitive opioid mechanisms are implicated in the control of gonadotrophin and adrenocorticotropic hormone secretion and in the hypotension of various types of shock. The investigation of possible dynorphin involvement in neurohypophysial function is taking place because vasopressin and dynorphin A (1-8) have been shown to coexist in the neurosecretory vesicles of magnocellular neurons.

Electron transfer across posterior pituitary neurosecretory vesicle membranes.

Secretory vesicles from the neurohypophysis have a transmembrane electron carrier very similar to that found in adrenal medullary chromaffin granules. Two different tests show that ascorbic acid contained in the vesicles will reduce an external electron acceptor. First, reduction of cytochrome c or ferricyanide in the medium by a neurosecretory vesicle suspension can be followed spectrophotometrically. Second, the membrane potential (inside positive) generated by electron transfer can be monitored using the membrane potential-sensitive optical probe Oxonol VI. As in chromaffin granules, this electron transfer is probably mediated by cytochrome b561. It may function to regenerate internal ascorbic acid and to provide reducing equivalents needed by the intravesicular amidating enzyme.

Angiotensin I-generating acid endopeptidase activity in neurosecretory vesicles isolated from bovine pituitary

Secretory vesicles purified from the neural and intermediate lobes of the bovine pituitary contain acidic endopeptidases which are capable of converting renin tetradecapeptide (RTD) substrate to Angiotensin I (AI). Preliminary characterization of the neurosecretory vesicle (NSV) endopeptidase showed that it had a pH optimum of 4.0, and unlike renin was inactive at pHs > 6.0. It is inhibited by 10 −6M pepstatin A, but not by PMSF, leupeptin, PMBS, or the specific renin inhibitor H-142. This NSV endopeptidase differed from cathepsin D in that it was unable to degrade α-casein, but was quite active in generating AI from RTD (V max = 5 moles/g protein/hour). No enzyme activity that could convert AI to Angiotensin II could be detected in the NSVs suggesting that the acidic endopeptidase is involved in processing neurosecretory vesicle proteins other than those associated with the renin angiotensin system in the brain.

Delta pH, H+ diffusion potentials, and Mg2+ ATPase in neurosecretory vesicles isolated from bovine neurohypophyses.

The proton gradient (delta pH) and electrical potential (delta psi) across the neurosecretory vesicles were measured using the optical probes 9-aminoacridine and Oxanol VI, respectively. The addition of neurosecretory vesicles to 9-aminoacridine resulted in a rapid quenching of the dye fluorescence which was reversed when the delta pH was collapsed with ammonium chloride or K+ in the presence of nigericin. From fluorescence quenching data and the intravesicular volume, delta pH across the membrane was calculated. Mg2+ ATP caused a marked carbonyl cyanide p-trifluoromethoxyphenylhydrazone-sensitive change in the membrane potential measured using Oxanol VI (plus 100 mV inside positive), presumably due to H+ translocation across the neurosecretory vesicle membrane. Imposition of this membrane potential was responsible for the lysis of vesicles in the presence of permeant anions. The effectiveness of these anions to support lysis reflected the relative permeability of the anion which followed the order acetate greater than I- greater than Cl greater than F- greater than SO4- = isethionate = methyl sulfate. These data showed that the neurosecretory vesicles possess a membrane H+-translocating system and prompted the study of Mg2+-dependent ATPase activities in the vesicle fractions. In intact vesicles a Mg2+ ATPase appeared to be coupled to electrogenic proton translocation, since the enzyme activity was enhanced by uncoupling the electrical potential, using proton ionophores. Inhibition of this enzyme with dicyclohexylcarbodiimide also inhibited the carbonyl cyanide p-trifluoromethoxyphenylhydrazone-sensitive delta psi across the vesicle membrane caused by H+ translocation. A second Mg2+ ATPase was also found on the vesicle membranes which is sensitive to vanadate. Complete inhibition of this enzyme with vanadate had little effect on the proton ionophore-uncoupled ATPase activity or on the Mg2+ ATP-induced membrane potential change.

An identical cytochrome b561 is present in bovine adrenal chromaffin vesicles and posterior pituitary neurosecretory vesicles.

An identical cytochrome b561 was found to be an integral component of both chromaffin vesicles from adrenal medulla and neurosecretory vesicles from posterior pituitary by spectrophotometric and immunological techniques. The neurosecretory vesicles had 6.8 micrograms of cytochrome/mg of membrane protein versus 69 micrograms/mg in chromaffin vesicles. This cytochrome was also immunologically detected in various regions of bovine brain and was immunologically distinct from the cytochrome found in serotonin-containing vesicles from platelets. Dopamine beta-hydroxylase involved in the biosynthesis of catecholamines was not present in neurosecretory vesicles, suggesting an alternative functional role for the cytochrome in these vesicles. Neurosecretory vesicles do contain a mixed function oxidase (peptidyl alpha-amidase) which appears to be involved in alpha-amidation of the carboxyl termini of vasopressin and oxytocin. We suggest that cytochrome b561 in the two vesicles may be functionally associated with different ascorbic acid-dependent, copper-containing mixed function oxidases: dopamine beta-hydroxylase and peptidyl alpha-amidase.

Lenticles: Innervated secretory structures that are expressed at every other larval moult

Lenticles are dome-shaped circles or ovals of cuticle with a dark rim. They occur with a precise segmental arrangement in the larvae and pupae of lycaenid and hesperiid butterflies. In Calpodes ethlius (Lepidoptera, Hesperiidae) each lenticle is secreted by a pair of large polyploid epidermal cells. The dark rim or annulus is formed from a ring-shaped cell. The dome, which consists of an epicuticle with a perforate intermediate layer like a pepper-pot, is formed by a central goblet cell. Between the perforate intermediate layer and the cell surfaces there is a cavity that contains material presumed to be secretion. Both cells have elaborate basal plasma membrane reticular systems and the apical microvilli associated with an extensive smooth endoplasmic reticulum that is typical of lipid-secreting cells. In addition, there is a plasma membrane reticular system in the ring cell and between it and the goblet cell that contains the endings of nerves having neurosecretory vesicles. Lenticles thus have a structure appropriate for an innervated organ of lipid secretion. However, in their development, lenticles arise from bristles that are presumed to be sensory. Lenticles or their precursors are segmentally arranged in the five larval instars and the pupa, but the pattern changes at each moult. The cells that form a lenticle at one moult have a rest period at the next one when they only secrete surface cuticle. Many lenticles are paired in their cycle of development, with only one of the pair making a lenticle at a particular moult. For example, the dorsal and lateral lenticles alternate in position between anterior and posterior. The second and fourth instar segments have anterior and the third and fifth instars have posterior lenticles. In the first instar the cells that will make lenticles for the second and third instars both make bristles. Lenticles are thus formed by cells that not only change their response to ecdysone qualitatively by switching from bristle to lenticle but also alternate in their later responses, switching back and forth at alternate moults between the formation of a lenticle and the secretion of surface cuticle.

A fine structural study of the formation of temporary swellings (Herring bodies) and reversible degeneration of neurosecretory axons following microiontophoretic ejection of vinblastine into the hypothalamo-neurohypophysial tract of the frog (Rana pipiens).

The microiontophoretic ejection of vinblastine into the median eminence effectively blocked axonal transport in the hypothalamoneurohypophysial system of the frog. Proximal to the ejection site, large axon swellings (Herring bodies) were found that contained mainly abundant neurosecretory granulated vesicles and axonal smooth endoplasmic reticulum. At advanced post ejection times, autophagic events were obvious. Subsequently, preexperimental conditions were reestablished, indicative of the resumption of axonal transport. Distal to the ejection site, i.e. in the neural lobe, the relatively long block of axonal transport subsequent to the vinblastine ejection caused most axons to degenerate and to be phagocytized by pituicytes. The degenerative process was reversible since upon reestablishment of the axonal transport regeneration of the neurosecretory fibers occurred.
