Aervalanata L. belongs to the family of Amaranthaceae, found in the tropical regions. Traditionally, this plant is known for antimicrobial, anthelminthic, antiparasitic, antidiabetic, diuretic, nephroprotective, cytotoxic, and antihyperlipidemic activities. As yet no further characteristic study has been conducted from ethanolic extract of this species, therefore in this present study we seek to identify and evaluate the bioactive compounds from the ethanolic extracts of Aervalanata L whole plant by using the GCMS. Result of this research work reports twenty-eight compounds. The identified chemical compounds were correlated with the NIST Mass Spectrum Library.In conclusion, we seek to provide additional information on the clinical significance and pharmacological information associated with this plant.

Pharmacological approach on kalanchoe pinnata – a concise report

The herbal extract of different parts of the kalanchoe pinnata plant are being used for the treatment of injuries, respiratory infections, diabetes, wound healing, diabetic wound healing, bug stings, and pain and inflammation. This genus has about 125 species. Alkaloids, flavonoids, phenolic compounds, tannins, macro elements, microelements, iron, zinc, vitamins, ascorbic acid, riboflavin, thiamine, and niacin are all present in the plant. It is mainly used in treatment of urinary bladder stones, diarrhea, cold, coughs, fever, headache, pain. It has wide pharmacological properties such as antihypertensive, anti-mutagenic, antibacterial, wound healing, antidiabetic, hepatoprotective, nephroprotective and antioxidant activities.

Nephroprotective Activity of Green Microalgae, Chlorella sorokiniana Isolated from Jordanian Water

Nephropathy is a global health issue that affects more than 20% of the adult population. Nephropathy is expected to be the fifth leading cause of death worldwide over the coming two decades. The introduction of green microalgae in nutrition and therapeutics for their biological activities is increasing. The current study examined the effect of Chlorella sorokiniana on renal health after inducing nephrotoxicity in mice. Preliminary screening of the algal aqueous extract revealed the presence of soluble polyphenols and triterpenoids. Successive intraperitoneal doses of gentamicin were administered to mice to induce nephrotoxicity. Concurrent intraperitoneal doses of the algal extract were administered to the infected mice to evaluate their nephroprotective activity. Two different concentrations of the treatment agent were administered in successive doses to two groups of mice. The tested concentrations were 150 and 300 mg/kg of mouse weight, respectively. The other two groups were either left untreated (normal control) or treated only with antibiotics (negative control). Creatinine, urea, and uric acid levels were analyzed in both serum and urine samples to evaluate the renal health of each animal group. Histochemical examination of the renal tissues was performed to assess the damage and improvement status. In vivo studies revealed a promising and significant nephroprotective activity of C. sorokiniana.

Metabolite profiling and nephroprotective potential of Glycyrrhiza glabra L. roots against cisplatin-induced nephrotoxicity in vitro and in vivo.

The present study was conducted to investigate the phytochemical analysis and demonstrate the nephroprotective potential of root extract of Glycyrrhiza glabra L. against cisplatin (CP) -induced nephrotoxicity in vitro and in vivo. The HPTLC analysis and UPLC-MS were carried out for standardizing and metabolite profiling of methanolic extract of roots of G. glabra (GGE). Further, in vitro studies were conducted in human embryonic kidney (HEK)-293 cells to evaluate the cytotoxicity and anti-oxidant potential of GGE with CP as a toxicant and ascorbic acid as standard. Also, in vivo nephroprotective potential at doses of 31.5, 63, and 126 mg/kg/day on CP (6 mg/kg, bw, IP) induced nephrotoxicity was evaluated on rodents. Phytochemical analysis by HPTLC and UPLC-MS revealed the presence of glycyrrhizin, glabridin, and liquiritin along with other bioactive constituents. The in vitro assay of GGE showed significant (P<0.001 nephroprotective, cellular anti-oxidant potential and improvement in morphological changes induced by CP. Further, administration of CP caused significant (P<0.001) elevation in biochemical, inflammatory, oxidative stress, caspase-3, as well as histopathological changes in kidney tissue. Pre-treatment with GGE attenuated the elevated biochemical markers significantly, improved histopathological damage, and showed a comparable result to ascorbic acid and α-ketoanalogue. Present study concluded the nephroprotective potential of GGE which supports the traditional claim of G. glabra roots in various kidney and its related disorders. The nephroprotective activity may be attributed to its anti-oxidant, anti-inflammatory, and anti-apoptosis effects. Thus, it holds promising potential in management of nephrotoxicity.

Ameliorative effect of Azima tetracantha extract on Cadmium induced Hepato-Renal oxidative stress: An in vivo approach

Cadmium (Cd), an environmental pollutant affects humans through contaminated foodstuffs and industrial processes. Cd accumulates in various organs upon exposure and the primary targets are kidney and liver. Its toxicity induces free radicals, creating oxidative stress, defects in antioxidant mechanism, damage to membrane proteins, DNA damage, necrosis, and apoptosis. In this study hepatoprotective and nephroprotective activity of Azima tetracantha towards Cd-induced toxicity was investigated by in vivo approach. Ultra Violet (UV) spectral and Fourier Transform Infra-Red (FTIR) analysis of methanol extract of the Azima tetracantha leaves revealed phenolic compounds. Cadmium Chloride (CdCl2) exposure in Wister Albino rats showed a marked increase in lipid profile, enzymatic antioxidants, and non-enzymatic antioxidants. Co-administration of a methanol extract of Azima tetracantha leaves (500 mg/kg/day) with CdCl2 resulted in the reversal of toxicity effects in the kidney and hepatic cells by maintaining the normal histological architecture. Further, reduced lipid peroxidation and increased antioxidant defense system activity were observed. Hence, the current study suggests that the nephroprotective and hepatoprotective efficacy of Azima tetracantha in Cd-induced toxicity might result from its antioxidant and metal chelating characteristics that might benefit in reaching the best remedy in Cd-induced damage.

Metabolite Profiling and Nephroprotective Potential of the Zea mays L. Silk Extract against Diclofenac-Induced Nephrotoxicity in Wistar Rats.

The lack of sufficient scientific evidence prompted the analytical investigation of nephroprotective potential of the silk extract of Zea mays L., which is traditionally and ethnomedicinally used for various disorders including kidney dysfunction. The present study was conducted to investigate the phytochemical analysis and demonstrate the nephroprotective potential of the methanolic silk extract of Z. mays L. using a rodent model. High-performance thin-layer chromatography (HPTLC) analysis was carried out to standardize the methanolic silk extract of Z. mays (ZME) using naringenin as a marker. The metabolite profiling of the ZME was carried out using ultrahigh-performance liquid chromatography mass spectrometry (UPLC-MS) on a monolithic capillary silica-based C18 column to identify bioactive compounds and for confirmation of the identified markers. Furthermore, for acute toxicity study, a single dose (2000 mg/kg bw) of the ZME was administered orally to Wistar rats. Also, nephrotoxicity was induced in Wistar rats by injecting diclofenac (DC) (50 mg/kg, bw, i.p.) at a single dose. The efficacy of the ZME as a nephroprotective agent was then evaluated at doses of 100, 200, and 400 mg/kg/day, bw, p.o. Furthermore, the kidney, liver, antioxidant, inflammatory, and apoptotic biochemical markers and histopathological and immunohistochemical alterations (caspase-3 and reduced nicotinamide adenine dinucleotide phosphate (NADPH) oxidase-4 (NOX-4)) were evaluated. Phytochemical analysis by HPTLC and UPLC-MS revealed the presence of naringenin, vanillic acid, ferulic acid, gallic acid (GA), ellagic acid, quercetin, and morin, along with other bioactive constituents exhibiting multiple pharmacological properties. The acute toxicity study of the ZME showed no mortality or any clinical signs of toxicity through all the 14 days of the toxicity study at a dose of 2000 mg/kg. Also, administration of DC caused a significant elevation (P < 0.001) in kidney biochemical parameters and also caused oxidative, inflammatory, and apoptotic stress. Furthermore, DC also caused histopathological and immunohistochemical changes. Pretreatment with the ZME attenuated the elevated biochemical markers significantly at medium and high doses along with improvement in histopathological and immunohistochemical damages and showing comparable results to those of α-ketoanalogue. The present study verifies the traditional claims of Z. mays silk alleviating various kidney and related disorders by concluding the nephroprotective potential of the ZME. The nephroprotective activity of the ZME is attributed to the phytoconstituents present, acting as potent restoring antioxidants and preventing inflammatory and apoptotic cellular damages in rats. Thus, it holds promising potential in the management of nephrotoxicity.

Daphnetin: A bioactive natural coumarin with diverse therapeutic potentials.

Daphnetin (DAP), a coumarin derivative extracted from Daphne species, is biologically active phytochemical with copious bioactivities including anti-inflammatory, anti-oxidant, neuroprotective, analgesic, anti-pyretic, anti-malarial, anti-bacterial, anti-arthritic, neuroprotective, hepatoprotective, nephroprotective, and anti-cancer activities. A wide range of studies have been conducted exploring the significance and therapeutic potential of DAP. This study reviewed various databases such as NCBI, PubMed, Web of Science, Scopus and Google Scholar for published research articles regarding the sources, synthesis, and various bioactivities of DAP using different key words, including but not limited to “pharmacological activities,” “sources,” “neuroprotective effect,” “synthesis,” “cancer,” “anti-inflammatory effect” of “daphnetin.” Furthermore, this review encompasses both in-vivo and in-vitro studies on DAP for treating various diseases. A comprehensive review of the literature revealed that the DAP had a promising pharmacological and safety profile, and could be employed as a pharmaceutical moiety to treat a variety of illnesses including microbial infections, cancer, arthritis, hepatic damage, inflammation and neurological anomalies. The current review intends to provide an in-depth focus on all pharmacological activities and therapeutic approaches for the pharmaceutical and biomedical researchers.

Tunisian Nephroprotective Plants: A Review

In Tunisian folk medicine, several herbs are prescribed for reducing renal damage and to avoid kidney related complications. These can be of immense value in combating renal damage. In this review, we provide a description of the current literature on the use of indigenous herbs as alternative medicine for treating renal damage. The aim of this review was to collect information on promising active phytoconstituents such as organosulfur compounds, polyphenols, terpenes, alkaloids phenylpropanoids, and polysaccharides from Tunisian plants that have been scientifically examined for their nephroprotective capacities. Twenty-nine Tunisian medicinal plants have been reported for their significant nephroprotective activities against renal toxicities in animal models. Lamiaceae was the most commonly used Tunisian plant family used for renal protection. The leaves were maximally used for nephroprotection compared to the other plant parts. Nephrotoxicity is commonly the result of several nephrotoxins. Many studies have focussed on drug-caused renal failure which is one of the major problems in medical practice. Other studies focused on other important nephrotoxicity factors, including drugs and industrial chemicals. This literature review highlights the use of some medicinal plants as nephroprotective agents. To defend against this nephrotoxicity, some medicinal plants, known as nephroprotective agents, have been highlighted in this review.

Pioglitazone in diabetic kidney disease: forgotten but not gone.

Diabetic kidney disease (DKD) is described in approximately 20–40% of all diabetic patients and is associated with significant cardiovascular and all-cause mortality. The involvement of multiple metabolic, haemodynamic, inflammatory, and tubular pathways in the pathophysiology of DKD generates the need for multitargeted treatment approaches to improve its development at all levels and delay or even reverse its progression. Thiazolidinediones are potent exogenous agonists of PPAR-γ, which augment the effects of insulin on its cellular targets, mainly at the level of adipose tissue. Pioglitazone, currently the main thiazolidinedione in clinical practice, has achieved significant improvements of albuminuria in patients with type 2 diabetes. It can also interfere with most cellular pathways involved in the development and evolution of DKD. This paper explores the pathophysiological mechanisms governing its possible nephroprotective activity during a diabetic state. It also discusses its future role to ameliorate the global burden of DKD.

Standardized herbal remedies are highly effective and relatively safe. That is why it is suggested to pay attention to herbal remedies already known for the treatment of genitourinary diseases to expand the indications for their use. The aim of the research is to study the nephroprotective properties of the standardized herbal composition BNO 2103, which is the active ingredient of Canephron N ®, in a model of renal failure in rats. Materials and methods. An experimental study was performed on 42 male white outbred rats weighing 150–200 g, which were divided into 4 groups. Nephropathy was induced by a single subcutaneous injection of 0.7 ml/kg of 2.5 % potassium chromate solution. Animals in 3 and 4 groups received BNO 2103 at a dose of 33.0 mg/kg and prednisolone at a dose of 1.9 mg/kg intragastrically daily for 20 days. After animals were sacrificed, their kidneys were obtained and studied with standard electronic microscopy methods. Results. BNO 2103 normalized the ultrastructural picture of the kidneys, however, with the presence of minor pathological signs. BNO 2103 reduced the destruction of basement membranes, improved organelle structure and increased metabolic processes in podocytes, and endotheliocytes, reducing the manifestations of degenerative-dystrophic changes in them. BNO 2103 was superior to the reference drug prednisolone. Conclusions. BNO 2103 showed a significant nephroprotective effect, normalizing the ultrastructural picture of the kidneys in rats with renal failure. This effect is most likely related to the presence of biologically active substances with a powerful antioxidant, anti-inflammatory, nephroprotective activity. The obtained results allow us to consider BNO 2103 as a promising nephroprotective agent for the treatment of chronic kidney disease.

Pharmacological Evaluation of Polyherbal Formulation for Nephroprotective Activity

The kidney plays an essential role in removing waste products and drugs from the body and maintaining balanced body fluids. It gets affected due to many factors, notably, diabetes and high blood pressure. Nowadays, naturally, derived products are essential in curing various ailments and are safe and cost-effective. The purpose of this study is to assess the toxicity profile and nephroprotective effect of a proprietary polyherbal formulation in Wistar albino rats for gentamicin-induced nephrotoxicity. The Polyherbal formulation was procured from Rumi Herbals Private Limited. Acute toxicity experiments were conducted in Wistar rats using the Gentamicin induced nephrotoxicity model as per OECD standards 423, and the efficacy was assessed using the Gentamicin induced nephrotoxicity model. The formulation was proven safe up to 2000mg/kg orally in an acute toxicity study, with no behavioral abnormalities and no fatality. The gentamicin 80 mg/kg i.p for 7 days induced nephrotoxicity in rats showed a significant (P&lt;0.05) increase in the renal parameters and reduction in antioxidant levels compared with day 0. Whereas test drug-treated groups at a low dose (200 mg/kg) and high dose (400 mg/kg) showed significant (P&lt;0.05) reduction in elevated renal parameters and improvement in antioxidant levels compared with the disease control group. According to the histopathological interpretation of isolated kidneys, this formulation protects from kidney damage and restores typical kidney architecture. From the results, the proprietary polyherbal formulation has shown effective nephroprotective activity may be due to the presence of secondary metabolites/phytoconstituents. Further investigation is essential to focus on the mechanism involved and standardize the active phytoconstituents responsible for the nephroprotective activity.

The liver and kidneys are the vital organs of the body and perform important life-sustaining functions in the body. Synthetic drugs used in the treatment of liver and kidney diseases are sometimes inadequate and can lead to serious side effects. Medicinal herbs and plants were used to combat diseases for a long time and combination therapy is preferred over single plant therapy. In the current study, the Asparagus racemosus, Mucuna pruriens, Anacyclus pyrethrum, and Tribulus terrestris polyherbal preparation (PHP) was selected to evaluate its hepatoprotective, antioxidant, and anti-nephrotoxic potential. The methanolic extract of PHP was prepared following standard protocols. Fifty-six albino rats were divided into 7 groups (n = 8). The negative control (NC) having the healthy rats and the remaining 6 groups were induced liver toxicity by intraperitoneally injecting 0.5 mL/kg of 50% CCl4 in olive oil. Group 2 was positive control and group 3 and 4 received silymarin standard drug at the dose of 100 and 200 mg/kg body weight. Groups 5, 6, and 7 (PHP-1, PHP-2, PHP-3) were the liver-damaged rats receiving the PHP at a dose of 50, 100, and 150 mg/kg body weight. Blood samples were collected at 21 of the trial, to evaluate oxidative stress, hepatoprotective and anti-nephrotoxic potential. Results of liver function tests revealed significant (P < .05) hepatoprotective activities of PHP after intoxication with CCl4 of albino rats as compared to standard groups. Moreover, results of renal functions also showed that PHP has a significant (P < .05) restoring the capacity of blood urea, creatinine, and uric acid in intoxicated rats as compared with the control group. The PHP also reduced the oxidative stress in the treatment groups by increasing the total antioxidant capacity and reducing the total oxidative status. It can be concluded that selected medicinal plants have a potential role in the management of liver and kidney disorders. So, by running the clinical trial on a large scale and by isolating the phytochemical constituents responsible for hepatoprotective and nephroprotective activities, locally prepared drugs could be developed to manage liver and renal disorders.

Tribulus Terrestris: An Important Medicinal Herb of Indian Desert

Abstract: Tribulus terrestris L. (Family Zygophyllaceae), commonly known as Small Caltrops or Chhota Gokhru, growing wildly in sandy areas is known for its medicinal uses in traditional medicine system. The fruit, leaves, stem and root of the plant are used medicinally. T. terrestris possessed many pharmacological attributes such as nephroprotective, aphrodisiac, antiinflammatory, anti-oxidant, antimicrobial and insecticidal activities. These marvellous bioactivities of the T. terrestris can be attributed to the presence of a wide range of phytochemicals including flavonoids, flavonol glycosides, steroidal saponins, and alkaloids. The dried fruit of the herb is very effective in most of the genitourinary tract disorders. The present study highlights the potency of T. terrestris in controlling the reproductive disorders, mainly impotence, nocturnal emissions, erectile dysfunction in men and gonorrhoea in women. In addition, it acts as a diuretic and provides remedy for urinary tract disorders. The present study provides a comprehensive overview of the phytochemistry, folk medicinal uses and pharmacological aspects of T. terrestris. The information will be helpful in developing the new formulation with therapeutic and economic value in the future.

Antioxidative stress and nephroprotective activity of leaf fractions of Setaria megaphylla in alloxan-induced diabetic Wistar rats

Setaria megaphylla (Steud) Dur &amp; Schinz (Poaceae), a medicinal plant used traditionally to treat diabetes was investigated for antioxidative stress and renoprotective potentials against alloxan-induced kidney injuries in diabetic rats. Standard biochemical and histological methods were used to determine oxidative stress markers levels, kidney function indices and kidney histopathology, which were used as parameters to evaluate antioxidative stress and renoprotective activities of the leaf fractions (200 mg/kg). Significantly (p&lt;0.05 – 0.001) increased levels of kidney oxidative stress markers (SOD, CAT, GPx, GSH) and decreased MDA level were caused by the leaf fractions in the treated-diabetic rats. Creatinine and urea levels were also significantly (p&lt;0.05-0.001) reduced, while electrolytes levels were not significantly (p&gt;0.05) reduced when compared to control. Kidney histology revealed absence or significant reductions in pathological features in the treated diabetic rats relative to untreated diabetic rats. The results suggest that the leaf fractions of Setaria megaphylla possess antioxidative stress and nephroprotective potentials due to the antioxidant activities of their phytochemical constituents.

The study evaluated the nephroprotective activity of hydroalcoholic extracts of Trachyspermum ammi leaves and Citrus paradisi fruits against prednisolone-induced ADPKD in experimental rats. Discovery of a chalcone derivative as potent necroptosis inhibitor for the treatment of acute kidney injury.

Necroptosis, a form of inflammation-related programmed cell death, is a major mechanism of proximal tubular cell injury in acute kidney injury (AKI). Blockade of necroptosis signalling represents a promising strategy for clinical therapy of AKI. Previously, we identified a small molecular receptor-interacting protein kinases (RIPK)1 inhibitor Cpd-71 with nephroprotective activities. To discover more nephroprotective agents, in this study, 20 chalcone derivatives were synthesized and evaluated for their anti-necroptosis and nephroprotective activities. Among the chalcone derivatives, Cpd-2 exhibited the most potent anti-necroptosis activity (IC50 =1.08 μM) and protective activity (EC50 =1.49 μM) through directly binding to RIPK1 and blocking RIPK1-RIPK3-mixed-lineage kinase domain-like protein (MLKL) signalling pathway. Furthermore, Cpd-2 effectively attenuated cisplatin or hypoxia/reoxygenation (H/R)-induced injury and necroptotic inflammation in renal cell models. Moreover, in cisplatin- or ischemia/reperfusion (I/R) induced AKI mouse model, detection of creatinine and urea nitrogen in blood showed that Cpd-2 improved kidney function. Periodic acid-Schiff (PAS) staining and immunofluorescence analysis indicated that Cpd-2 also reduced pathological damage and inhibited inflammatory development in kidney tissues. In summary, although some chalcone derivatives have been reported to prevent kidney injury previously, our present study not only discovered a promising leading compound Cpd-2, but also provided a novel and successful practice for the development of necroptosis inhibitors from natural products derivatives as AKI therapeutic agents.

A A Synergistic Nephroprotective Activity of A Miracle Drink Ayurvedic Proprietary Medicine Renal Support A Novel Therapy for Chronic Kidney Disease in Human

Miracle drink “Renal Support (S-5)” an Ayurvedic formulation in conjugation with other cardiovascular support (S3), Sugar Care (S10), and liver health support (S4) was scientifically evaluated on 12 humans subjects for its therapeutic potential in treating chronic kidney diseases caused due to: a) Induce of pain killers medication, and other medications, b) Chronic diabetes, c) Blood pressure. Patients suffering from renal failure due to over medications, pain killer medication and BP were advised to take 15ml of Renal Support and S3 twice a day morning and evening before food, and 15ml of S4 trice a day. As the main biomarker of kidney disease, creatinine was monitored every month till three months of treatment whereas; blood urea and hemoglobin were screened at month end. Cytotoxicity and nephroprotective activity of Renal Support were evaluated on Baby Hamster Kidney Fibroblast cells (BHK-21). Radical decline in serum creatinine content was observed from 6.31mg/dl to 1.80mg/dl (68%), 1.20mg/dl (79%), and 0.84mg/dl (82%) on 30, 60, and 90 days of treatment respectively and in 90 days of treatment most of the patients showed 50 to 83% creatinine reduction. A significant decrease in the blood urea from 91mg/dl to 30mg/dl(67%) and hemoglobin content from 7.27 to 11.77g% was observed in 30days of treatment and the majority of patients showed &gt;50% of blood urea reduction. No toxicity of Renal Support towards BHK-21 was noticed and showed 40.92% and 47.54% nephroprotective activity. A novel, natural-based, and safe Ayurveda formulation with significant nephroprotective potential for CKD treatment was proposed in the present study.

Asparagus falcatus L. (Asparagaceae) leaf extracts attenuate doxorubicin-induced renal toxicity via antioxidant, anti-inflammatory, and anti-apoptotic pathways

The search for therapeutic agents that improve kidney function against doxorubicin-induced renal toxicity is important. Herein, the potential nephroprotective activity by Asparagus falcatus L. (AF, Asparagaceae) leaf extracts against doxorubicin-induced renal toxicity (5 mg/kg, ip) in Wistar rats (n = 6/group) after oral administration of hexane (55 mg/kg), ethyl acetate (35 mg/kg), butanol (75 mg/kg), and aqueous (200 mg/kg) extracts of AF for 28 consecutive days was investigated. It was noticed that the treatment with the selected extracts of AF significantly attenuated doxorubicin-induced elevations of serum creatinine, urea nitrogen, β2-microglobulin, cystatin C, and proteinuria in experimental rats. The histology showed attenuation of the features of acute tubular injury. Treatment regimens significantly reversed the doxorubicin-induced reduction in total antioxidant status, glutathione peroxidase, and glutathione reductase activity in renal tissue homogenates. A suppression in lipid peroxidation was noted with hexane, ethyl acetate, and butanol extracts of AF. Moreover, a reduction in the concentration of the pro-inflammatory mediator TNF-α (p < 0.05), and immunohistochemical expression of COX-2 were observed. The immunohistochemical expression of pro-apoptotic Bax protein was decreased and the anti-apoptotic BCL-2 was increased in renal tissues following the treatments. In conclusion, it was revealed that, hexane, ethyl acetate, butanol, and aqueous extracts of AF attenuate doxorubicin-induced renal toxicity in Wistar rats through antioxidant, anti-inflammatory, and anti-apoptotic pathways. The plant, AF could be recommended as a promising therapeutic agent to minimize renal toxicity induced by doxorubicin in cancer patients, however, subsequent clinical trials are warranted.
