Optimization and Multimachine Learning Algorithms to Predict Nanometal Surface Area Transfer Parameters for Gold and Silver Nanoparticles.

Interactions between gold metallic nanoparticles and molecular dyes have been well described by the nanometal surface energy transfer (NSET) mechanism. However, the expansion and testing of this model for nanoparticles of different metal composition is needed to develop a greater variety of nanosensors for medical and commercial applications. In this study, the NSET formula was slightly modified in the size-dependent dampening constant and skin depth terms to allow for modeling of different metals as well as testing the quenching effects created by variously sized gold, silver, copper, and platinum nanoparticles. Overall, the metal nanoparticles followed more closely the NSET prediction than for Förster resonance energy transfer, though scattering effects began to occur at 20 nm in the nanoparticle diameter. To further improve the NSET theoretical equation, an attempt was made to set a best-fit line of the NSET theoretical equation curve onto the Au and Ag data points. An exhaustive grid search optimizer was applied in the ranges for two variables, 0.1≤C≤2.0 and 0≤α≤4, representing the metal dampening constant and the orientation of donor to the metal surface, respectively. Three different grid searches, starting from coarse (entire range) to finer (narrower range), resulted in more than one million total calculations with values C=2.0 and α=0.0736. The results improved the calculation, but further analysis needed to be conducted in order to find any additional missing physics. With that motivation, two artificial intelligence/machine learning (AI/ML) algorithms, multilayer perception and least absolute shrinkage and selection operator regression, gave a correlation coefficient, R2, greater than 0.97, indicating that the small dataset was not overfitting and was method-independent. This analysis indicates that an investigation is warranted to focus on deeper physics informed machine learning for the NSET equations.

Nanometal surface energy transfer-based lateral flow immunoassay for T2 toxin detection

In this study, we incorporated nanometal surface energy transfer (NSET) in lateral flow immunoassay (LFIA) and explored the relationship between fluorescence quenching efficiency and detection sensitivity to improve sensitivity of NSET-LFIA system. We developed nine gold nanoparticles (GNPs) with absorption spectrum in the range of 520–605 nm as acceptors and quantum dot microspheres (QDMs) with emission spectrum of 530, 570, and 610 nm as donors. By analyzing the overlap integral area, fluorescence quenching efficiency, and detection sensitivity of 27 donor-acceptor pairs, we observed that the larger overlap integral area led to higher fluorescence quenching efficiency and detection sensitivity. A maximum fluorescence quenching efficiency of 91.0% was obtained from the combination of GNPs at 605 nm and QDMs at 610 nm, achieving the highest detection sensitivity. We developed NSET-LFIA for the detection of T2 toxin with a limit of detection of 0.04 ng/mL, which was 10-times higher than that obtained via conventional GNP-LFIA. NSET-LFIA represents a versatile, ultrasensitive and valuable screening tool for small molecules in real samples.

Single-Nucleobase-Resolved Nanoruler Determines the Surface Energy Transfer Radius on the Living Cell Membrane.

Investigations about surface energy transfer radius (r0) are limited to the aqueous solution system, and it is quite limited on experimental values of r0 between dyes and the corresponding gold particle (AuNP) sizes, especially for living cell systems. Hence, the selection of suitable AuNP-dye pairs is restricted when designing nanometal surface energy transfer (NSET) strategies in analytical sciences. Here, we developed a single-nucleobase-resolved NSET strategy to in situ measure the r0 value between a specific dye and different-sized AuNPs on the living cell membrane. Using the aptamer-dye complex (XQ-2d-nTA-FAM) and antiCD71 antibody-coupled AuNP conjugate (Au@antiCD71) as two working elements to bind two different sites on CD71 receptors on living cell membranes, we modified the nTA spacer between FAM and the terminal of aptamer to change the distance (r) from FAM to AuNP center and further adjusted the quenching efficiency (Φ) between them. Different r0 values of various AuNP-FAM pairs in living cells are determined by this in situ detection strategy. Based on this single-nucleobase-resolved NSET strategy, we established a simple and efficient universal method for measuring r0 in the living cell system, which greatly expanded the selection range of AuNP-dye pairs during the construction of the NSET model at the nanoscale.

Nanometal Surface Energy Transfer (NSET) from Biologically Active Heterocyclic Ligands to Silver Nanoparticles Induces Enhanced Antimicrobial Activity against Gram-positive Bacteria

Herein we report the formation of a nanometal surface energy transfer (NSET) pair between a donor biologically active heterocyclic luminescent ligand such as 3-(1,3-Dioxoisoindolin-2-yl)-N, N-dimethylpropan-1-ammonium perchlorate (S4PNL; λem-408nm) and an acceptor silver nanoparticle (Ag NP; λabs-406nm). When the S4PNL ligand interacts with Ag NPs, the quenching in their luminescence intensity at 408nm is noticed, with a Stern-Volmer constant of 0.8×104 M-1. The present donor-acceptor pair displays a binding constant of 2.8×104 M-1 and binding sites of 1.12. The current work shows the energy transfer from a molecular dipole (S4PNL) to a nanometal surface (Ag NP) and thus follows the nanometal surface energy transfer (NSET) ruler with an energy transfer efficiency of 80.0%, 50% energy transfer efficiency distance (d0) of 4.9nm, donor-acceptor distance of 3.4nm. The alteration in the zeta potential value of S4PNL upon interaction with AgNP clearly demonstrates the strong electrostatic interaction between donor and acceptor. Importantly, the current NSET pair shows enhanced antimicrobial activity against gram-positive bacteria such as Bacillus cereus (B. cereus) in comparison to their parent components i.e. S4PNL ligand and Ag NP. The NSET pair shows maximum inhibition against B. cereus (9202.21±463.26 CFU/ml.) at 10% while minimum inhibition is observed at 0.01% of it (39,887.19±242.67 CFU/ml.).

Record Resolution of Nanometal Surface Energy Transfer Optical Nanoruler Projects 3D Spatial Configuration of Aptamers on a Living Cell Membrane.

Decrypting the in situ three-dimensional spatial configuration of an aptamer is of considerable significance; however, suitable nanoscale resolution tools are lacking. Herein, we show that a new nanometal surface energy transfer (NSET) optical nanoruler has a record resolution, down to single-nucleobase levels. We labeled fluorophores on different T bases of XQ-2d, including 5', 3', 6T, 22T, 38T, and 52T positions. The NSET nanoruler in situ decrypted the base sequence-dependent distance projection on the nanogold surface, demonstrating that 5', 3', stem, and loop structures are symmetrical in three-dimensional spatial configuration. The orientation of the 5' and 3' stem was toward the antiCD71-binding site, whereas the loop was in the opposite direction at a considerable distance. Molecular docking simulation was performed to list all of the possible conformations; however, all base distance parameters projecting on the nanogold surface determined a single conformation of XQ-2d. The specific binding sites of XQ-2d were Lys477, Ser691, and Arg698 on the CD71 receptor.

Principles and Applications of Resonance Energy Transfer Involving Noble Metallic Nanoparticles.

Over the past several years, resonance energy transfer involving noble metallic nanoparticles has received considerable attention. The aim of this review is to cover advances in resonance energy transfer, widely exploited in biological structures and dynamics. Due to the presence of surface plasmons, strong surface plasmon resonance absorption and local electric field enhancement are generated near noble metallic nanoparticles, and the resulting energy transfer shows potential applications in microlasers, quantum information storage devices and micro-/nanoprocessing. In this review, we present the basic principle of the characteristics of noble metallic nanoparticles, as well as the representative progress in resonance energy transfer involving noble metallic nanoparticles, such as fluorescence resonance energy transfer, nanometal surface energy transfer, plasmon-induced resonance energy transfer, metal-enhanced fluorescence, surface-enhanced Raman scattering and cascade energy transfer. We end this review with an outlook on the development and applications of the transfer process. This will offer theoretical guidance for further optical methods in distance distribution analysis and microscopic detection.

Polyaniline and Copper based nanometal surface energy transfer probe for efficient turn-on bovine serum albumin sensing

In the present study, we have investigated the energy transfer mechanism owing to the interaction between colloidal Cu nanorods and polyaniline as a function of varying amounts of Cu. XRD, FESEM, and HRTEM examined the sample's structural and morphological characteristics, and the optical characteristics were explored by UV–Vis absorbance, photoluminescence, and time-resolved photoluminescence measurements. The steady-state photoluminescence exhibits quenching behaviour and time-resolved photoluminescence measurements exhibit lifetime reduction in accordance with the nanometal surface energy transfer theory, following 1/d4 distance dependency from polyaniline to the surface of Cu. With the increasing quencher (Cu) concentration, quenching efficiency and rate of energy transfer increase, whereas a decrease in the distance between Cu and polyaniline has been observed. Our research attempts to see if such nanohybrid comprising polymer–noble metal nanoparticles could be used as sensitive biosensors. Hence, the nanohybrid system consisting of Cu and polyaniline has been employed in the potential bovine-serum albumin sensing, which showed an excellent detection limit of 15.3 nM. The Cu- PANI pair also exhibits excellent selectivity amidst the other possible interfering compounds.

Single-nucleobase resolution of a surface energy transfer nanoruler for in situ measurement of aptamer binding at the receptor subunit level in living cells.

In situ identification of aptamer-binding targets on living cell membrane surfaces is of considerable interest, but a major challenge, specifically, when advancing recognition to the level of membrane receptor subunits. Here we propose a novel nanometal surface energy transfer (NSET) based nanoruler with a single-nucleobase resolution (SN-nanoruler), in which FAM-labeled aptamers and single-sized gold nanoparticle (GNP) antibody conjugates act as a donor and an acceptor. A single nucleobase resolution of the SN-nanoruler was experimentally illustrated by molecular size, orientation, quenching nature, and other dye-GNP pairs. The SN-nanoruler provides high reproducibility and precision for measuring molecule distance on living cell membranes at the nanometer level owing to only the use of single-sized antibody-capped GNPs. In situ identification of the aptamer binding site was advanced to the protein subunit level on the living cell membrane for the utilization of this SN-nanoruler. The results suggest that the proposed strategy is a solid step towards the wider application of optical-based rulers to observe the molecular structural configuration and dynamic transitions on the membrane surface of living cells.

Probing nanometal surface energy transfer between nanocopper and MoS2 for glutathione sensing

Cu nanoparticles and 2D-MoS2 nanosheets have been synthesised and the morphology of the prepared samples has been characterised using high-resolution transmission electron microscopy and field emission scanning electron microscopy. Steady-state fluorescence studies exhibit quenching of fluorescence intensity upon the addition of quencher in varying amounts. Time-resolved fluorescence studies show a reduction of fluorescence lifetime in the presence of quencher. A thorough investigation of energy transfer between MoS2 and copper nanoparticles has been accomplished using steady-state and time-resolved fluorescence studies. The results are more consistent with the nanometal surface energy transfer (NSET) theory, which follows a 1/d 4 distance dependence than the Förster resonance energy transfer theory exhibiting 1/d 6 distance dependence. For the MoS2–Cu pair, the NSET efficiency, the proximal distance between donor–acceptor pairs and the rate of energy transfer has been explored. Energy transfer-based fluorescent biosensors have been regarded as potential candidates in biosensing applications. Glutathione, an endogenous tripeptide compound, plays a vital role as an antioxidant in the human body. Irregular glutathione levels have been linked to several adverse illnesses. Henceforth, detection of glutathione levels is indispensable. The Cu–MoS2 pair was used for glutathione sensing in nanomolar concentration (nM), which demonstrated a turn-off sensing behaviour.

Concentration-dependent β-cyclodextrin-promoted refolding of gold nanoparticles-conjugated bovine serum albumin complexed with gemini surfactants with different spacer groups

This work demonstrates refolding of unfolded bioconjugated bovine serum albumin [(BSA, 10 μM):gold nanoparticles (AuNPs, 0.0124 μM)] using β-cyclodextrin (β-CD). Protein bioconjugates are first unfolded with 0.2 mM of each of gemini surfactants, 12–4–12,2Br and 12–8–12,2Br- that have hydrophobic spacer groups of different lengths. At this concentration of a gemini surfactant, protein bioconjugates remain in compressed form due to strong hydrophobic interactions imparted by highly hydrophobic microdomain within the core of the gemini micelles formed along the chain of a bioconjugate. The compressed protein bioconjugates get relaxed in presence of a low concentration of β-CD which strips off surfactant molecules from bioconjugate-micelles complexes. Relaxed denatured protein bioconjugates then get refolded once remaining surfactant molecules are stripped off at a higher concentration of β-CD. A step-wise refolding process of bioconjugated protein chains exactly opposite to the unfolding is noted. Binding isotherms are constructed using Förster’s resonance energy transfer (FRET) and nanometal surface energy transfer (NSET) parameters calculated based on energy transfer process occurs between Trp residues and AuNPs. α-helix of protein bioconjugates remains in inverse relationship to β-turns during refolding by β-CD. Refolding is found to be more in presence of 12–8–12,2Br- than 12–4–12,2Br-. Secondary/tertiary structural changes of refolded BSA, on being bioconjugated, have been determined through various spectroscopic analysis. An approach towards construing the refolding of denatured AuNPs-bioconjugates as potential materials and impacts of AuNPs on conformations of refolded protein is presented here.

One-donor-two-acceptors coupled energy transfer nanoprobe for recording of viral gene replication in living cells

Highly efficient Förster resonance energy transfer (FRET) is in great request for sensitive and high signal-to-background ratio imaging of biomolecule in cells, which is limited in many simple energy transfer system. Herein we report a sensitive one-donor-two-acceptors (DAA*) nanoprobe for live-cell imaging by assembling quantum dots (QDs) and BHQ-2 loaded gold nanoparticles (AuNPs) through molecular beacon. Both nanometal surface energy transfer (NSET) from QDs to AuNPs through the exciton-plasmon interaction and FRET from QDs to BHQ-2 through the exciton-dipole interaction can synergistically occur, significantly enhancing the energy transfer efficiency. The DAA* nanoprobe with strong photostability was then successfully applied to record the spatiotemporal distribution and dynamics of genomic RNA (gRNA) replication of human respiratory syncytial virus (RSV) in live cells by fluorescence imaging with improved signal-to-background ratio. Furthermore, dark-field microscopic imaging (iDFM) was also used to confirm the fluorescence imaging results after scattering signal amplification of AuNPs in the nanoprobe. This multiple energy transfer strategy with universal applicability for other donor-acceptor system required for higher energy transfer efficiency is promising in long-term bioimaging and biosensing of low-abundance biomolecules.

Refolding of denatured gold nanoparticles-conjugated bovine serum albumin through formation of catanions between gemini surfactant and sodium dodecyl sulphate.

The present work elucidates binding interactions of sodium dodecyl sulphate (SDS) with the conjugated gold nanoparticles (AuNPs)-bovine serum albumin (BSA), unfolded by each of two gemini surfactants, 1,4-bis(dodecyl-N,N-dimethylammonium bromide)-butane (12-4-12,2Br−) or 1,8-bis(dodecyl-N,N-dimethylammonium bromide)-octane (12-8-12,2Br−). Initially, at a low concentration of SDS there is a relaxation of bioconjugates from their compressed form due to the formation of catanions between SDS and gemini surfactants. On moving towards higher concentrations of SDS, these relaxed unfolded bioconjugates renature by removal of residual bound gemini surfactants. Mixed assemblies of SDS and gemini surfactants formed during refolding of bioconjugates are characterized by DLS and FESEM measurements. A step-by-step process of refolding observed for these denatured protein bioconjugates is exactly the inverse of their unfolding phenomenon. Parameters concerning nanometal surface energy transfer (NSET) and Förster's resonance energy transfer (FRET) phenomenon were employed to develop a binding isotherm. Moreover, there remains an inverse relationship between α-helix and β-turns of bioconjugates during the refolding process. Significantly, in the presence of 12-8-12,2Br−, SDS induces more refolding as compared to that for 12-4-12,2Br−. Bioconjugation shows an effect on the secondary structures of refolded BSA, which has been explored in detail through various studies such as Fourier transform infrared spectroscopy, fluorescence, and circular dichroism (CD). Therefore, this approach vividly describes the refolding of denatured bioconjugates, exploring structural information regarding various catanions formed during the process that would help in understanding distance-dependent optical biomolecular detection methodologies and physicochemical properties.

Binding interactions of cationic gemini surfactants with gold nanoparticles-conjugated bovine serum albumin: A FRET/NSET, spectroscopic, and docking study

This work demonstrates binding interactions of two cationic gemini surfactants, 12-4-12,2Br- and 12-8-12,2Br- with gold nanoparticles (AuNPs)-conjugated bovine serum albumin (BSA) presenting binding isotherms from specific binding to saturation binding regions of surfactants. The binding isotherm has been successfully constructed using Förster's resonance energy transfer (FRET) and nanometal surface energy transfer (NSET) parameters calculated based on fluorescence quenching of donor, tryptophan (Trp) residue by acceptor, AuNP. Energy transfer efficiency (ET) changes due to alteration in the donor-acceptor distance when surfactants interact with bioconjugates. A solid reverse relationship between α-helix and β-turn contents of BSA-AuNPs-conjugates is noted while interacting with surfactants. 12-8-12,2Br- shows stronger binding interactions with BSA-bioconjugates than 12-4-12,2Br-. The effect of bioconjugation on secondary/tertiary structures of BSA in the absence and presence of a surfactant is studied through circular dichroism, fluorescence, and Fourier transform infrared spectroscopic measurements. Motional restrictions imposed by AuNPs on Trp residues of folded and unfolded BSA have been investigated using red edge emission shift (REES) measurements. Finally, the molecular docking results present the modes of interactions of 12-4-12,2Br- and 12-8-12,2Br-, and Au-nanoclusters (Au92) with BSA. An approach to describe the binding isotherms of surfactants using AuNPs-bioconjugates as optical-based molecular ruler and possible effects of AuNPs on microenvironment and conformations of the protein is presented.

DNA Logic Nanodevices for Real-Time Monitoring of ATP in Lysosomes.

DNA logic nanodevices have prospects in molecular recognitions but still face challenges in achieving DNA computation-controlled regulation in specific compartments of living cells. By incorporating the i-motif sequence and ATP aptamers into a Y-shaped DNA (Y-DNA) structure, and applying gold nanoparticles (AuNPs) as the transporting carrier, herein we present a new type of DNA logic nanodevices to monitor the ATP levels in lysosomes of living cells. Triple energy transfers including dual fluorescent resonance energy transfers (FRETs) and a nanometal surface energy transfer (NSET) occurred in the DNA logic nanodevices. It was identified that the proposed nanodevices perform an AND logic operation to output FRET signals only when an endogenous proton and ATP simultaneously exist in the cellular microenvironment. Owing to the use of the i-motif sequence, the nanodevices have lysosome-recognizing capacity without causing alkalization of the acidic organelle, making DNA computation-controlled regulation at the level of cellular organelles achievable. These DNA logic nanodevices show high application prospects in lysosome-related cellular function and disease treatment.

Preparation of Nanosensors for Detecting the Activity of Glycosaminoglycan Cleaving Enzymes.

Glycosaminoglycans (GAGs) play crucial roles in several biological processes including cell division, angiogenesis, anticoagulation, neurogenesis, axon guidance and growth, and viral and bacterial infections among others. The GAG cleaving hydrolases/lyases play a major role in the control of GAG structures, functions, and turn over. Dysregulation of GAG cleaving enzymes in vivo are linked to a number of human diseases including cancer, diabetes, atherosclerosis, arthritis, inflammation, and cardiovascular diseases. Several GAG cleaving enzymes are widely used for studying GAG glycobiology: heparitinases, chondroitinases, heparanases, hyaluronidases, and keratanases. Herein, we describe a method to synthesize four distinct nanometal surface energy transfer (NSET)-based gold-GAG-dye conjugates (nanosensors). Heparin, chondroitin sulfate, heparan sulfate, and hyaluronic acid are covalently linked with distinct fluorescent dyes and then immobilized on gold nanoparticles (AuNPs) to build nanosensors that serve as excellent substrates for GAG cleaving enzymes. Upon treatment of nanosensors with their respective GAG cleaving enzymes, dye-labeled oligosaccharides/disaccharides are released from AuNPs resulting in enhanced fluorescence recovery. These nanosensors have a great promise as diagnostic tools in various human pathophysiological conditions for detecting dysregulated expression of GAG cleaving enzymes and also as a sensitive analytical tool for assessing the quality control of pharmaceutical grade heparin polysaccharides that are produced in millions of small- and medium-sized animal slaughter houses worldwide.

Enzyme-free and label-free detection of Staphylococcus aureus based on target-inhibited fluorescence signal recovery

In this work, a one-step fluorometric strategy based on nanometal surface energy transfer (NSET) between carbon dots (CDs) and gold nanoparticles (AuNPs) was developed for facile detection of Staphylococcus aureus (S. aureus). The fluorescence of CDs was quenched up to 63.5% by AuNPs due to nucleic acid hybridization in the presence of linker DNA, which contained the complementary sequences of S. aureus-specific aptamer, and the fluorescence signal was in the “off” state. Upon aptamer addition, the CDs was released from linker DNA through the strong competitiveness of aptamer, leading to the notable fluorescence recovered. Once S. aureus is introduced, aptamer preferentially bind to the bacterial surface and cannot hybridize with complementary sequences in the linker DNA, resulting in the fluorescence signal with “off” state. Based on these findings, the performance and reliability of the fluorescence-based assay were evaluated. Compared to direct hybridization of complementary DNA on the surface of CDs and AuNPs, our sensing strategy has enhanced detection limit (10 cfu⋅mL−1) and improved linear range (10 to 106 cfu⋅mL−1) for S. aureus. Therefore, our proposed enzyme-free and label-free strategy may become a promising method for ease of operation, sensitive and selective S. aureus detection.

Selective uptake and modulation of nanometal surface energy transfer from quantum dot to Au nanoparticle across lipid bilayer of liposomes

Understanding the initial adsorption and subsequent translocation of nanoparticles (NPs) across the phospholipid bilayer of liposome find importance in drug delivery and in vivo bio-imaging applications. Herein, we have investigated the time-dependent selective uptake of 2.0 ± 0.3 nm-sized CdTe quantum dots (QDs) into small unilamellar vesicles (SUV) of dipalmitoylphoshatidylcholine (DPPC) in its gel phase and subsequent alteration of nanometal surface energy transfer (NSET) in the presence of 18.0 ± 0.2 nm-sized citrate-capped gold NPs (Au NPs). The interactions between CdTe QD and Au NP across the lipid bilayer of DPPC liposome has been explored by monitoring the NSET process using fluorescence spectroscopy. The time-dependent selective partitioning of QDs into the liposomal phase has been demonstrated using confocal laser scanning microscopy (CLSM) and atomic force microscopy (AFM). It has been observed that instant mixing of QDs and Au NPs with liposomes does not alter the extent of NSET between QD-Au NP pair relative to that in the absence of liposome. However, equilibrating the binary mixture of liposome and QDs for 24 h at room temperature leads to the formation of liposome encapsulated QD (LipQD) which does not take part in NSET with aqueous phase Au NPs. In contrast, equilibrating the ternary mixture of QDs, liposomes, and Au NPs for 24 h results in the formation of Au NP adsorbed QD-encapsulated liposomes (AuLipQD) which exhibit moderate amount of NSET from encapsulated QDs to the adsorbed Au NPs at the surface of liposomes. It has been observed that the efficiency of NSET decreases from 62% in bulk aqueous medium to 24% in AuLipQD complex. Our present findings may be useful to understand the fundamental interaction of other metal and semiconductor NPs with liposomes and cell membrane for various drug-delivery and bio-imaging applications.

Programmed Dual-Functional DNA Tweezer for Simultaneous and Recognizable Fluorescence Detection of microRNA and Protein.

A programmed dual-functional DNA tweezer (DFDT) as a signaling molecule is reported for the simultaneous and recognizable fluorescence detection of microRNA 21 (miRNA 21) and mucin 1 (MUC1). This unique DFDT is assembled from two Au-NP-attached central strands (C1 and C2) and an arm strand (A) dually ended by fluorophores Cy3 and Cy5, which are spatially separated from Au NP in the originally opened state. Through the competitive affinity interaction between targets and their complementary and aptamer sequences tethered in two recognition strands (R1 and R2), miRNA 21 and MUC1 are respectively converted into two dependently displaced fuel strands (F1 and F2). The next hybridization with two pairs of unpaired segments overhung in open DFDT leads to its conformational closure, resulting in the approach of Cy3 and Cy5 to Au NP. On the basis of the nanometal surface energy transfer scheme, the fluorescence emission of Cy3 or Cy5 is cooperatively quenched by Au NPs attached in C1 and C2. The significant variation of fluorescence intensity enables one-step, cost-effective, and specific quantization of miRNA 21 and MUC1 with high sensitivity down to 32 fM and 2.6 fg·mL-1 (8.5 pM), respectively. The novel DFDT-based assay route of multiplex analytes is promising and has the potential for rapid and reliable diagnosis and treatment of cancer-related diseases.

Dual Energy Transfer-Based Fluorescent Nanoprobe for Imaging miR-21 in Nonalcoholic Fatty Liver Cells with Low Background.

Nonalcoholic fatty liver disease (NAFLD) can progress gradually to liver failure, early warning of which is critical for improving the cure rate of NAFLD. In situ imaging and monitoring of overexpressed miR-21 is an advanced strategy for NAFLD diagnosis. However, this strategy usually suffers from the high background imaging in living cells owing to the complexity of the biological system. To overcome this problem, herein, we have developed a one-donor-two-acceptor nanoprobe by assembling gold nanoparticles (AuNPs) coupled with BHQ2 (AuBHQ) and quantum dots (QDs) through DNA hybridization for imaging of miR-21 in living cells. The fluorescence of QDs was quenched up to 82.8% simultaneously by the AuNPs and the BHQ2 via nanometal surface energy transfer and fluorescence resonance energy transfer, reducing the background signals for target imaging. This low background fluorescent nanoprobe was successfully applied for imaging the target miR-21 in nonalcoholic fatty liver cells by catalyzing the disassembly of QDs with the AuBHQ and the fluorescence recovery of QDs. In addition, the sensitivity of this nanoprobe has also been enhanced toward detecting miR-21 in the range of 2.0-15.0 nM with the detection limit (LOD, 3σ) of 0.22 nM, which was 13.5 times lower than that without BHQ2. The proposed approach provides a new way for early warning, treatments, and prognosis of NAFLD.

Fluorometric determination of morphine via its effect on the quenching of fluorescein by gold nanoparticles through a surface energy transfer process.

A method is described for sensitive and selective fluorometric determination of morphine. It is based on the effect of morphine on quenching of the fluorescence of fluorescein by gold nanoparticles (AuNPs) via surface energy transfer. When fluorescein is added to solutions of colloidal AuNPs, its fluorescence becomes quenched due to nanometal surface energy transfer (NSET) because the absorption of AuNPs strongly overlaps the emission spectrum of fluorescein. In the presence of morphine, which contains both a tertiary nitrogen ring atom and a phenolic hydroxy group, it will coordinate to the AuNPs, and this causes recovery of fluorescence. The presence of a tertiary nitrogen ring atom and a phenolic hydroxy group (both required for the effect to occur) in morphine make the probe highly selective and sensitive for morphine. A paper strip assay also was developed by utilizing this detection scheme. The turn-on fluorescent probe was successfully applied to the determination of morphine in spiked serum and urine samples. The method has a 53 pM limit of detection. The paper strip was applied tothe determination of morphine in sweat, urine and other biological fluids. It is perceived to be useful for early detection of drug abuse by adolescent.
