IN SILICO DOCKING STUDY OF HERBAL COMPOSITION AND DEVELOPMENT OF MOUTH DISSOLVING TABLETS FOR COMMON RESPIRATORY DISEASES THEREOF

Objective: The present study aims to develop a novel mouth dissolving tablets containing a combination of herbal extracts and a bioactive constituent and evaluating it for activity against common respiratory diseases (in silico studies). Methods: Docking study was done to provide a scientific foundation, keeping the traditional knowledge as a base. Four trial batches were developed. The final batch was then formulated and various pre and post-compression and assays were performed to evaluate the formation of good quality of product. The final batch was prepared by the method of direct compression and taken for accelerated stability studies. Results: The final batch containing 10 % active ingredients, 7.5 % super-disintegrant and 47 % diluent was found to be stable, easily producible and economic. Conclusion: This research work grasps possibilities for researchers in the development and evaluation of mouth-dissolving tablets with significant bioactive potential against common respiratory diseases.

Solid dispersion with β-cyclodextrin and skimmed milk powder is one way to increase disintegration time by increasing the water solubility of inadequately water-soluble FCD. In-vitro dissolution experiments of FCD mouth-dissolving tablets revealed significant differences. Stability studies should evaluate drug product characteristics that are susceptible to change during storage and are anticipated to impact quality, safety, and efficacy to demonstrate that the optimal formulations remain stable over the course of the study. The results of stability experiments were statistically significant at p<0.05 using one-way ANOVA followed by Dunnet's test. Preparation and Evaluation of Mouth Dissolving Tablet of Albendazole Using Different Concentrations of Super-Disintegrant

Preparation and Optimization of Ivabradine Hydrochloride Mouth-Dissolving Tablet

The aim of the present work was the preparation and optimization of mouth-dissolving tablets (MDTs) of Ivabradine hydrochloride by using natural super disintegrants. The tablets were prepared using microcrystalline cellulose as diluent and aspartame as a sweetening agent along with Natural super disintegrants. The natural super disintegrants used in this study were Guar Gum and Banana powder. By using natural superdisintegrants we had saved the environment and protected our bodies from the harmful effect of synthetic superdisintegrants. The tablet was prepared by the direct compression method. The 6mm of punch was used and the tablet weight is 110 mg. The tablets were evaluated for weight variation, hardness, friability, wetting time, disintegration time (DT), and dissolution study. Different concentration of superdisintegrants was used in this formulation as 6%, 8%, and 10%. The three batches of guar gum and 3 batches of banana powder were prepared i.e., a Total of six batches were prepared. From the results obtained, it can be concluded that the tablet formulation prepared with 10% banana powder i.e., 10 mg showed fast and higher drug release (98.66%) during in vitro dissolution study. Also, the hardness, friability, dissolution rate, and assay of prepared tablets (batch F6) were found to be acceptable according to standard limits. The result was the F6 batch was optimized batch from all the batches.

Objective: The rationale of the current work is to design, develop and optimize of mouth dissolving tablet of ambrisentan to treat hypertension. Methods: Sodium starch glycolate and crospovidone were used as the super disintegrants in the direct compression method to create nine ambrisentan mouth-dissolving tablet formulations. Wetting time, drug content, in vitro disintegration time, dispersion time, and dissolution time were all assessed for the produced formulations. Results: Based on the results obtained, formulation F6 containing 30 mg of crospovidone exhibited good wetting time, dispersion time, disintegration time and drug release. The hardness of formulations AS1 to AS9 was found to be in the range of 2.5 to 3.11 Kg/cm2. The friability of formulations AS1 to AS9 was found to be less than 1%. A water absorption ratio was performed for ensuring the moisture sorption and water uptake properties of super disintegrants. The in vitro drug release of formulation AS6 containing a concentration of Crospovidone 30 mg, shows 91.30% drug release respectively at the end of 12 min. Conclusion: The mouth-dissolving tablets of ambrisentan were successfully designed, developed, and fabricated. It can be reasonably concluded that the AS6 batch of mouth-dissolving tablets of ambrisentan with 30 mg of crospovidone exhibited maximum cumulative drug release in 12 min.

DESIGN, DEVELOPMENT AND FABRICATION OF MOUTH-DISSOLVING TABLETS CONTAINING EXTRACT OF TRIBULUS TERRESTRIS FOR THE TREATMENT OF HYPERTENSION

Objective: The present work is aim to design, development and fabrication of mouth dissolving tablets containing extract of Tribulus terrestris for the treatment of hypertension. Methods: The extract of fruits of Tribulus terrestris was formulated as mouth dissolving tablets (MDTs) by full factorial design at 32 levels and prepared by direct compression method using super integrants like sodium starch glycolate and cross povidone. Furthermore, the tablet was evaluated for thickness, hardness, weight variation, wetting time, disintegration time, and in vitro drug release study. Results: The tablets were analyzed for a variety of characteristics, such as hardness (2.4-2.9 kg/cm2), friability (0.33-1.7%), disintegration time (20-34 s), drug content (95.32-99.09%), water uptake ratio (26-48%), wetting time (29-69 s), and in vitro drug release illustrated in 5 min (99.04-68.21%). There was no interaction between both the drug and the polymer, according to FTIR and DSC studies. Conclusion: The research revealed that Tribulus terrestris fruits extract can be designed, developed and fabricated into mouth dissolving tablet for the treatment of hypertension with improved bioavailability and expected patient compliance.

Formulation and Evaluation of Gastro Retentive Drug Delivery System of Domperidone Mouth Dissolving Tablet by using Super Disintegrating Agents

Direct compression method can be considered as an important method for the formulation of fast dissolving tablets of Domperidone compare to wet granulation method. Higher the concentration of the lubricating agent (Magnesium Stearate or Talc), higher will be the disintegration time. DESIGN AND IN VITRO EVALUATION OF ACRIVASTINE AS ORODISPERSIBLE TABLET USING DIRECT COMPRESSION METHOD.

The aim: This study aimed to develop mouth-dissolving tablets of Acrivastine, an antihistamine medication, in order to increase its oral bioavailability. Materials and methods: Different super disintegrants, such as crospovidone, croscarmellose sodium, and sodium starch glycolate, were used to make Acrivastine oral dispersible tablets (ODTs). These super disintegrants were utilized in various concentrations. The formulation (F3) with 6% w/w crospovidone had a fast disintegration time (less than 30 seconds) and practically total drug release within 10 minutes. All of the formulations were made using the direct compression method and proper diluents, binders, and lubricants. Fourier transform infrared spectroscopy (FTIR) tests were used to investigate the drug-ex¬cipient interaction, and all formulations demonstrated improved drug-excipient compatibility. Results: The average weight of all formulations was between 175 and 180 mg. All formulations' hardness and friability were within acceptable ranges. Direct compression tablets had a hardness of 3.2 to 4 kg/cm2. All formulations were determined to have a friability of less than 1.0%. For oral dissolving tablets, the in vitro disintegration time is critical, and this time preferred to be < 60 seconds. The results also showed that crospovidone disintegrated after 24 seconds and sodium starch glycolate disintegrated in 40 seconds in vitro. Conclusions: When compared to croscarmellose sodium and sodium starch glycolate, crospovidone performs better as a super disintegrant. In comparison to other formula, tablets breakdown in the mouth in 30 seconds and have a maximum in vitro drug release time in 1-3 minutes.

ABSTRACT: The foremost intention of present research was the preparation and assessment of mouth dissolving formulation Rivaroxaban and its validation. During present work, this tablet was formulated by straight compression technique by means of Cros-carmellose sodium and Sodium starch glycolate as super-disintegrants (concentration of 2, 4, 6%) and Avicel 102 as a binder. The calibration curve of API using solvent phosphate buffer pH 6.8 was carried out. All prepared formulations exposed to different assessment parameters have shown the findings within prescribed limit. Due to the large concentration of super disintegrants in F8, disintegration time can reach 29±0.06 seconds. In used buffer, drug release was calculated at intervals of 0, 2, 4, 6, 8, 10, and 12 minutes. The F8 demonstrates 96.5±0.567 percent medication release. UV spectrophotometric validation was performed for the quantification of Rivaroxaban in bulk. Rivaroxaban was estimated at 247nm in phosphate buffer 6.8. The linearity range was observed 2–12µg/ml.

Methods: Mouth Dissolving Tablets (MDTs) of Tofacitinib Citrate were developed by full factorial design at 32levelsand prepared by direct compression method using super integrants like sodium starch glycolate, Ludiflash. The tablets were compressed into compacts on a 10 station tablet machine. The bulk drug was characterised by determining, MP, Solubility, pH and FTIR spectra. Results: The weight variation, hardness and diameter, thickness, friability, drug content, wetting time, in vitro disintegration time and in vitro dissolution studies, and stability study, tablet thickness, weight variation and drug content post compression parameters remained consistent and reproducible. All the formulations showed, almost 100 percent of drug release within 75 min. Formulations F1, F2 and F3 were prepared with 5 mg of SSG and 20 mg, 30 mg, and 40 mg Ludiflash which shows % release of drug in the order of F1&lt;F2&lt;F3. Formulations F4, F5 and F6 were prepared with 10 mg of SSG and 20 mg, 30 mg, and 40 mg Ludiflash which shows % release of drug in the order of F4&lt;F5&lt;F6. Formulations F7, F8 and F9 were prepared with 15 mg of SSG and 20 mg, 30 mg, and 40 mg Ludiflash which shows % release of drug in the order of F7&lt;F8&lt;F9. Conclusion: It is concluded that the amount of superdisintegrants decreases disintegration time of tablets, decreases wetting time, increases the cumulative % drug release causes better absorption.
