In "Immunization of normal mouse spleen cell suspensions in vitro" (26 Aug., p. 1004) by R. I. Mishell and R. W. Dutton, the nutritional cocktail in reference 9 should have included 2.5 ml of 200 m M glutamine.
Dissociated cells from the spleens of unimmunized mice were cultured with and without various mammalian erythrocytes. Spleen cell suspensions cultured with heterologous red cells developed levels of hemolytic plaque-forming cells only one log(2) less than those seen in vivo. The reaction is specific for the in vitro immunizing erythrocytes. Antibody was demonstrated in the culture fluids.
DIFFERENTIATION of nucleated, red blood cells has been observed when mouse bone marrow was cultivated in vivo in diffusion chambers1. Recently, during investigations of the survival of thymus and spleen cells similarly cultured, we found that differentiation of nucleated red blood cells also occurred. This was indicated by an increase in the number of erythrocytes after cultivation. The present report describes these findings.
