The problem of vestibular stability displaying is considered depending on individual parameters of motor-coordination abilities of those practicing martial arts. Methods and organization of the research. 161 male athletes engaged in kickboxing, boxing, karate, hand-to-hand fighting, sports wrestling, etc. took part in the testing. The average age was 21.3 years old, the average experience – 9.5 years. This group of athletes was divided into two subgroups: the first one with a high level of vestibular stability, the other one – with a low level of vestibular stability. Studying the parameters of motor-coordination abilities was conducted according to a ten-point scale; symmetry-asymmentry of athletes’ limbs was conducted according to a six-point scale and the subjective assessment of an athlete. The integral indicator of motor-coordination abilities was determined by the sum of points of all parameters studied. Sportsmanship was assessed according to one hundred-point scale. Research results. It is revealed that the group with a higher level of vestibular stability is different from the group with a low level of vestibular stability at a reliable level towards higher values in the following parameters: speed of change of motor actions; time differentiation of individual phases of technical action; orientation in space; coordination of different body parts’ movements; stability of the kinematic movement structure; accuracy of spatial characteristics; preservation of stability; integral indicator of movement coordination. There were also significant differences in the parameters of symmetry and asymmetry of performing technical actions in a duel, mainly with both arms or both legs. In regard to the influence of the level of sportsmanship and the experience of sports on the display of vestibular static-dynamic stability, there were no significant differences between the groups. Conclusion. The features of vestibular stability manifestation in the structure of individual parameters of motor-coordination abilities of those engaged in martial arts are revealed.

Differences in the level of development and proficiency in coordination motor abilities among female and male athletes of selected combat sports

Aim. The aim of this study was to determine gender differences in the level of development and proficiency in coordination motor abilities (CMAs) in athletes of selected combat sports. Materials and Methods. We analyzed the results of our experimental studies and the data and analyses of the material collected by other authors on the differences in the level of development and proficiency in CMAs in female and male wrestlers, taekwondo athletes, and kickboxers. Results. It was found that with age, training experience, and increasing competencies, gender differences in the level of most CMAs tend to decrease. Each athlete has their individual dynamics of development and proficiency in CMAs, which is not consistent for each ability and does not always coincide with the general trend of changes with age. Conclusions. The conclusion from the findings of the author’s empirical research is that the level of various CMAs can be improved across different age categories including preschool children, schoolchildren, and qualified athletes aged 15-29 years. With proper coordination training, younger and middle school children can reach, and often exceed, the CMAs levels of adolescents (boys and girls) at secondary school age. The rates of improvement of various CMAs over the same period in children of the experimental groups and in young competitive athletes may be 10-20% higher than in children or athletes of control groups of similar age. A comparison of the different CMAs in young athletes aged from 11 to 15 years, in whom these abilities have been developed in a systematic manner for one to four years with peers not involved in regular sports training showed that trained boys and girls outperformed untrained individuals by 10-20, 20-40, and even 60-100 and more percent. The analysis of the collected material shows that the assumption that, with equal trainability of girls and boys of preschool age, younger and older schoolchildren, and young and competitive wrestlers, higher responsiveness to identical coordination training programs, however, is observed in girls and adolescent and adult competitive athletes compared to their male peers.

Analysis of developmental coordination disorder in Chinese children

Objective: To investigate the prevalence of children's developmental coordination disorder (DCD) and its distribution based on different family socioeconomic characteristics in China, in order to provide a theoretical basis for early prevention, diagnosis, and intervention for DCD. Methods: From June to October, 2016, 1 887 children aged 3-10 years from 20 kindergartens and 10 elementary schools from 8 cities in seven geographic areas of China using a stratified cluster sampling method were recruited. With a cross-sectional design, parents were asked to report on their basic information. Children' s motor ability was assessed using the movement assessment battery for children-second edition (MABC-2). Children were grouped by age, sex, body mass index (BMI), one-child status, and family structure. Chi-square test and one-way ANOVA were used to compare family socioeconomic characteristics of children between different groups. Results: Among the 1 887 children, there were 1 110 (58.8%) preschool children (3-6 years of age) and 777 (41.2%) school-aged children (7-10 years of age). There were 982 males (52.0%) and 905 females (48.0%). A total of 5.5% (104 cases) children were diagnosed with DCD, 10.4% (197 cases) with suspected DCD, and 84.1% (1 586 cases) as typical motor developing children. There were no significant differences in prevalence of diagnosed and suspected DCD among different regions (χ²=17.342 and 4.877, P=0.173 and 0.560), total motor coordination score (F=2.759, P<0.05), and the scores of all dimensions (manual dexterity, positioning and grabbing, balance: F=9.276, 5.277, 3.706, all P<0.01). The prevalence of DCD in preschool children was significantly higher than that in school-age children (χ²=11.891, P<0.01). Girls were significantly better than boys in total motor coordination, manual dexterity, and balance (all P<0.01). Boys were significantly better than girls in positioning and grabbing (P<0.01). The prevalence of DCD in boys was significantly higher than that in girls (70 boys (7.1%) and 34 girls (3.8%), χ²=28.508, P<0.01). The total motor coordination ability, manual dexterity and balance of children who are overweighted (BMI>18 kg/m2) were significantly lower than those of children of normal weight (BMI≤18 kg/m2) (all P<0.01). The prevalence of suspected DCD children who are overweighted was significantly higher than that of children of normal weight (χ²=4.369, P<0.05). The difference of total motor coordination ability (F=6.811, P<0.01) and the prevalence of DCD (χ²=14.902, P<0.01) among different family structures were statistically significant. The total motor coordination ability and balance ability of children from multi-child family were better than those of children from one-child family (both P<0.05). Conclusion: The motor coordination ability of Chinese children is well-developed, with differences among different regions, gender, age, BMI, and family structure.

Playing the piano with a robotic third thumb: assessing constraints of human augmentation

Contemporary robotics gives us mechatronic capabilities for augmenting human bodies with extra limbs. However, how our motor control capabilities pose limits on such augmentation is an open question. We developed a Supernumerary Robotic 3rd Thumbs (SR3T) with two degrees-of-freedom controlled by the user’s body to endow them with an extra contralateral thumb on the hand. We demonstrate that a pianist can learn to play the piano with 11 fingers within an hour. We then evaluate 6 naïve and 6 experienced piano players in their prior motor coordination and their capability in piano playing with the robotic augmentation. We show that individuals’ augmented performance with the SR3T could be explained by our new custom motor coordination assessment, the Human Augmentation Motor Coordination Assessment (HAMCA) performed pre-augmentation. Our work demonstrates how supernumerary robotics can augment humans in skilled tasks and that individual differences in their augmentation capability are explainable by their individual motor coordination abilities.

The m6 A Readers YTHDF1 and YTHDF2 Synergistically Control Cerebellar Parallel Fiber Growth by Regulating Local Translation of the Key Wnt5a Signaling Components in Axons.

Messenger RNA m6A modification is shown to regulate local translation in axons. However, how the m6A codes in axonal mRNAs are read and decoded by the m6A reader proteins is still unknown. Here, it is found that the m6A readers YTHDF1 and YTHDF2 are both expressed in cerebellar granule cells (GCs) and their axons. Knockdown (KD) of YTHDF1 or YTHDF2 significantly increases GC axon growth rates in vitro. By integrating anti‐YTHDF1&2 RIP‐Seq with the quantitative proteomic analysis or RNA‐seq after KD of YTHDF1 or YTHDF2, a group of transcripts which may mediate the regulation of GC axon growth by YTHDFs is identified. Among them, Dvl1 and Wnt5a, encoding the key components of Wnt pathway, are further found to be locally translated in axons, which are controlled by YTHDF1 and YTHDF2, respectively. Specific ablation of Ythdf1 or Ythdf2 in GCs increases parallel fiber growth, promotes synapse formation in cerebellum in vivo, and improves motor coordination ability. Together, this study identifies a mechanism by which the m6A readers YTHDF1 and YTHDF2 work synergistically on the Wnt5a pathway through regulating local translation in GC axons to control cerebellar parallel fiber development.

The benefit of assessing implicit sequence learning in pianists with an eye-tracked serial reaction time task.

Playing piano professionally has been shown to benefit implicit motor sequence learning. The aim of the current study was to determine whether this advantage reflects generally enhanced implicit sequence learning unrelated to pianists' higher motor and/or visual-motor coordination abilities. We examined implicit sequence learning using the ocular serial reaction time (O-SRT) task, a manual-free eye-tracked version of the standard SRT, in 29 pianists and 31 controls. Reaction times (RT) and correct anticipations (CA) of several phases describing implicit sequence learning were analyzed. Furthermore, explicit sequence knowledge was compared between the groups, and relationships between implicit sequence learning with explicit sequence knowledge or demographic measures were evaluated. Pianists demonstrated superiority in all critical phases of implicit sequence learning (RT and CA). Moreover, pianists acquired higher explicit sequence knowledge, and only in pianists was explicit sequence knowledge related to implicit sequence learning. Our results demonstrate that pianists' superiority in implicit sequence learning is due to a higher general implicit sequence learning ability. Hence, we can exclude that higher motor and/or visual-motor coordination abilities are related to pianists' higher implicit sequence learning. Furthermore, the significant relationship of implicit sequence learning and explicit sequence knowledge suggests that pianists either used explicit strategies to support implicit sequence learning, had better explicit access to sequence knowledge, or both.

Effect of long-term exposure to acrylamide on endoplasmic reticulum stress and autophagy in rat cerebellum

Acrylamide (ACR) is a widely used chemical compound that has neurotoxicity in human, but whether ACR could impair the cerebellum and the related mechanism were still unknown. This study aimed to observe the changes in behavioral performance and cerebellar morphology caused by chronic ACR exposure, and to evaluate its influence on apoptosis, endoplasmic reticulum stress (ERS) and autophagy. Rats were treated with 0, 0.5 and 5 mg/kg ACR by drinking water for 12 months. Results showed that 5 mg/kg ACR treatment damaged the gait, balance ability, hindlimb muscle strength and motor coordination ability of rats. The results of hematoxylin and eosin and Nissl staining indicated that ACR impaired the structures of all three layers of the cerebellum, especially the Purkinje cell layer, showing abnormal morphology with nucleus condensation and pyknosis. Accumulation of autophagosomes, dilated endoplasmic reticulum and swollen mitochondria were observed in neurons under transmission electron microscopy. The enhanced apoptotic rates and the increased Bax expression indicated the elevated level of apoptosis. The results of Western blot showed that ACR treatment elevated protein levels of Beclin1, LC3-II/LC3-I, p-PERK/t-PERK, ATF4 and CHOP, indicating the initiation of autophagy, the activation of PERK pathway in ERS. This work helps to demonstrate the ACR neurotoxicity on cerebellum under chronic treatment and its underlying mechanism.

18F]PBR146 and [18F]DPA-714 in vivo Imaging of Neuroinflammation in Chronic Hepatic Encephalopathy Rats.

Neuroinflammation is an important pathogenesis of hepatic encephalopathy (HE). The upregulation of translocator protein (TSPO) during neuroinflammation provides an imaging molecular target to evaluate the severity of neuroinflammation in chronic HE rats. [18F]DPA-714 and [18F]PBR146 targeting TSPO are often used for neuroinflammation imaging. This study performed bile duct ligation (BDL) in rats to simulate chronic HE model, tested the behavioral experiments, and conducted [18F]PBR146 and [18F]DPA-714 micro-PET/CT scans followed analyzing the average %ID/g values of the whole brain, brain regions and main organs of subjects. After sacrifice the rats, the blood plasma samples were taken for blood biochemical indexes and plasma inflammatory factor levels examination, the liver and brain specimens were obtained for pathological analysis. The BDL rats showed chronic liver failure with defects in cognition, motor coordination ability and mental state. [18F]PBR146 and [18F]DPA-714 micro-PET/CT imaging results were similar in whole brain of BDL group and Sham group. Besides, some regional brain areas in BDL rats were found abnormal uptakes mainly located in basal ganglia area, auditory cortex, motor cortex, cingulate gyrus, somatosensory cortex, hippocampus, thalamus, midbrain, and medulla oblongata, and these regions also correlated with behavioral alterations. In conclusion, both [18F]PBR146 and [18F]DPA-714 had the similar imaging effects in hepatic encephalopathy models could quantitatively evaluate neuroinflammation load and distribution. The difference brain regions with higher uptake values of radiotracers in BDL rats were correlated with behavioral alterations.

O-EP004. Targeting epilepsy with novel therapeutic entities to investigate the biochemical and molecular mechanisms

Introduction . Analysis of anticonvulsant action of novel therapeutic entity (NTE) in kindling (Pentylenetetrazole (PTZ)) and acute (Kainic Acid (KA)) seizure mice (male swiss albino) models. Methods . The NTE (NP-VM1) was screened from a class of 5 hydrazone derivatives of 2,4-disubstituted thiazole through subcutaneous PTZ and neurotoxicity screening followed by log-probit analysis for determining ED50, TD50 and protective-index (PI). Neuroprotective potential of the NTE was analyzed in both models and was validated through behavioral screening tests and oxidative stress estimation (MDA, GPx and SOD levels). Results were analyzed by ANOVA single factor and Tukey’s post-hoc test. Results . NP-VM1 showed significant biological activity in screening. It significantly reduced the seizure activity in both models. Significant increase in the spatial learning ability in Morris Water Maze test was further confirmed by increased recognition index in Object Recognition Test. Reduced transfer latency and time spent in open arm in Elevated Plus Maze test along with increased rearings and movement through the centre in Open Field Test signified restoration of behavior normalcy and locomotion ability. Rotarod test showed that NTE significantly negated the neurotoxic and ataxic effects which was further proved by enhanced GPx and SOD levels and attenuated MDA levels in the hippocampus. The results of sucrose consumption test were however not statistically significant. Conclusion . NP-VM1 significantly augmented the ROS detoxification thereby fortifying the neurons from chronic damage, which was evident by the restoration of behavioral normalcy and learning, motor coordination and locomotion abilities.

Human urine-derived stem cell-derived exosomal miR-21-5p promotes neurogenesis to attenuate Rett syndrome via the EPha4/TEK axis

Rett syndrome (RTT) is a rare neurodevelopmental disorder that results in multiple disabilities. Exosomal microRNA (miRs) from urine-derived stem cells (USCs) have been shown to induce neurogenesis and aid in functional recovery from brain ischemia. In the present study, we sought to determine whether that exosomal miR-21-5p from USCs could promote early neural formation in a model of RTT. USCs were isolated and evaluated by flow cytometry. Exosomes were analyzed by transmission electron microscopy, tunable resistive pulse sensing (TRPS), and western blotting. PKH26 fluorescent dyes were used to observe intake of exosomes in vivo and in vitro. An RTT mouse model was treated with exosomes for behavioral studies. Dual‐luciferase report gene assays were conducted to evaluate the relationship between miR-21-5p and Eph receptor A4 (EphA4). In vitro, treatment with exosomes from human urine‐derived stem cells (USC-Exos) increased the percentage of neuron-specific class III beta-tubulin (Tuj1)+ nerve cells as well as the transcription levels of β-III tubulin and doublecortin (DCX). A higher level of miR-21-5p was observed in USC-Exos, which promoted differentiation in NSCs by targeting the EPha4/TEK axis. In vivo, exosomal miR-21-5p improved the behavior, motor coordination, and cognitive ability of mice, facilitated the differentiation of NSCs in the subventricular zone of the lateral ventricle and promoted a marked rise in the number of DCX+ cells. Our data provide evidence that exosomal miR-21-5p from human USCs facilitate early nerve formation by regulating the EPha4/TEK axis.Rett syndrome (RTT) is a rare neurodevelopmental disorder that results in severe multiple disabilities. This study provides evidence that exosomal miR-21-5p from human urine-derived stem cells facilitates early neural formation by regulating the EPha4/TEK axis, and may be a promising therapeutic option for this devastating disease.

The Effect of a Traditional Preparation Containing Piper nigrum L. and Bunium persicum (Boiss.) B.Fedtsch. on Immobility Stress-Induced Memory Loss in Mice.

Objective Alzheimer's disease is a progressive, age-related, and neurodegenerative disease characterized by mental decline. The exact cause of Alzheimer's disease is unclear, but cholinergic dysfunction, protein accumulation, and oxidative stress are among the most important hypotheses. The main purpose of our study was to investigate the effects of aqueous and hydroalcoholic extract combination of these two medicinal plants, black pepper and cumin (as a related formulation in traditional Persian medicine), on memory and learning of an immobilized stress animal model. Methods In this study, hydroalcoholic and aqueous extracts of cumin and black pepper fruits were prepared. Six groups of mice were treated orally for 2 weeks: control group, immobility stress, and stress-induced immobility mice received different doses of the hydroalcoholic extract (100 and 200 mg/kg) and aqueous extract (100 and 200 mg/kg). The shuttle box, novel object detection, and rotarod test were used to evaluate memory and learning. The activities of acetylcholinesterase, catalase (CAT), and superoxide dismutase (SOD) and the level of reduced glutathione (GSH) and malondialdehyde (MDA) were measured in the brain tissue. Results Immobility stress significantly reduced learning and motor coordination. Furthermore, MDA levels and acetylcholinesterase activity were significantly increased, while CAT and SOD activities were significantly reduced in the brain of immobility-induced stress mice. Other findings indicated that hydroalcoholic and aqueous extracts (100 and 200 mg/kg) of cumin and black pepper fruits have an improving effect on animal motor coordination and learning ability, GSH content, and CAT, SOD, and acetylcholinesterase enzyme function in comparison with stress groups (p < 0.05). Conclusion The hydroalcoholic and aqueous extracts of cumin and black pepper fruits have protective effects against stress-induced memory deficit and oxidative stress and may have beneficial therapeutic effect in the treatment of neurodegenerative diseases.

Rg1 exerts protective effect in CPZ-induced demyelination mouse model via inhibiting CXCL10-mediated glial response.

Myelin damage and abnormal remyelination processes lead to central nervous system dysfunction. Glial activation-induced microenvironment changes are characteristic features of the diseases with myelin abnormalities. We previously showed that ginsenoside Rg1, a main component of ginseng, ameliorated MPTP-mediated myelin damage in mice, but the underlying mechanisms are unclear. In this study we investigated the effects of Rg1 and mechanisms in cuprizone (CPZ)-induced demyelination mouse model. Mice were treated with CPZ solution (300 mg· kg-1· d-1, ig) for 5 weeks; from week 2, the mice received Rg1 (5, 10, and 20 mg· kg-1· d-1, ig) for 4 weeks. We showed that Rg1 administration dose-dependently alleviated bradykinesia and improved CPZ-disrupted motor coordination ability in CPZ-treated mice. Furthermore, Rg1 administration significantly decreased demyelination and axonal injury in pathological assays. We further revealed that the neuroprotective effects of Rg1 were associated with inhibiting CXCL10-mediated modulation of glial response, which was mediated by NF-κB nuclear translocation and CXCL10 promoter activation. In microglial cell line BV-2, we demonstrated that the effects of Rg1 on pro-inflammatory and migratory phenotypes of microglia were related to CXCL10, while Rg1-induced phagocytosis of microglia was not directly related to CXCL10. In CPZ-induced demyelination mouse model, injection of AAV-CXCL10 shRNA into mouse lateral ventricles 3 weeks prior CPZ treatment occluded the beneficial effects of Rg1 administration in behavioral and pathological assays. In conclusion, CXCL10 mediates the protective role of Rg1 in CPZ-induced demyelination mouse model. This study provides new insight into potential disease-modifying therapies for myelin abnormalities.

The Impact of Age, Gender and Technical Experience on Three Motor Coordination Skills in Children Practicing Taekwondo.

The study aim was to investigate the age, gender and technical level on motor coordination abilities of Taekwondo children. One hundred and fifteen children (83 male, 32 female), aged 7.76 ± 1.71 years, divided in three different groups, under 8 (5–7 years), under 10 (8–9 years) and under 12 (10–11 years), underwent three coordination skills tests: the ruler drop test (RDT), assessing visual reaction time, the hexagonal test (HT), assessing agility, and the target kick test (TKT), assessing kicking ability. MANOVA showed significant gender differences for TKT, in which females showed higher scores than males (p = 0.033). Significant differences were found in HT and TKT, where the under 12 group showed higher scores than younger athletes (p < 0.001). No differences amongst different age groups were found in RDT, showing that this could be a good predictor of Taekwondo performance, assessed at an early stage. High-level athletes showed better scores in all the tests than the low levels, as it was expected. Coordinative performance improves with age and is positively influenced by practicing a sports activity. The predisposition to a particular sport with a well-planned training may lead to a motor proficiency comparable to that reached by older athletes and better than same-age athletes.

Slc20a2-Deficient Mice Exhibit Multisystem Abnormalities and Impaired Spatial Learning Memory and Sensorimotor Gating but Normal Motor Coordination Abilities.

BackgroundPrimary familial brain calcification (PFBC, OMIM#213600), also known as Fahr’s disease, is a rare autosomal dominant or recessive neurodegenerative disorder characterized by bilateral and symmetrical microvascular calcifications affecting multiple brain regions, particularly the basal ganglia (globus pallidus, caudate nucleus, and putamen) and thalamus. The most common clinical manifestations include cognitive impairment, neuropsychiatric signs, and movement disorders. Loss-of-function mutations in SLC20A2 are the major genetic causes of PFBC.ObjectiveThis study aimed to investigate whether Slc20a2 knockout mice could recapitulate the dynamic processes and patterns of brain calcification and neurological symptoms in patients with PFBC. We comprehensively evaluated brain calcifications and PFBC-related behavioral abnormalities in Slc20a2-deficient mice.MethodsBrain calcifications were analyzed using classic calcium-phosphate staining methods. The Morris water maze, Y-maze, and fear conditioning paradigms were used to evaluate long-term spatial learning memory, working memory, and episodic memory, respectively. Sensorimotor gating was mainly assessed using the prepulse inhibition of the startle reflex program. Spontaneous locomotor activity and motor coordination abilities were evaluated using the spontaneous activity chamber, cylinder test, accelerating rotor-rod, and narrowing balance beam tests.ResultsSlc20a2 homozygous knockout (Slc20a2-HO) mice showed congenital and global developmental delay, lean body mass, skeletal malformation, and a high proportion of unilateral or bilateral eye defects. Brain calcifications were detected in the hypothalamus, ventral thalamus, and midbrain early at postnatal day 80 in Slc20a2-HO mice, but were seldom found in Slc20a2 heterozygous knockout (Slc20a2-HE) mice, even at extremely old age. Slc20a2-HO mice exhibited spatial learning memory impairments and sensorimotor gating deficits while exhibiting normal working and episodic memories. The general locomotor activity, motor balance, and coordination abilities were not statistically different between Slc20a2-HO and wild-type mice after adjusting for body weight, which was a major confounding factor in our motor function evaluations.ConclusionThe human PFBC-related phenotypes were highly similar to those in Slc20a2-HO mice. Therefore, Slc20a2-HO mice might be suitable for the future evaluation of neuropharmacological intervention strategies targeting cognitive and neuropsychiatric impairments.

Squamosamide Derivative FLZ Diminishes Aberrant Mitochondrial Fission by Inhibiting Dynamin-Related Protein 1.

Mitochondrial dysfunction is involved in the pathogenesis of Parkinson’s disease (PD). Mitochondrial morphology is dynamic and precisely regulated by mitochondrial fission and fusion machinery. Aberrant mitochondrial fragmentation, which can result in cell death, is controlled by the mitochondrial fission protein, dynamin-related protein 1 (Drp1). Our previous results demonstrated that FLZ could correct mitochondrial dysfunction, but the effect of FLZ on mitochondrial dynamics remain uncharacterized. In this study, we investigated the effect of FLZ and the role of Drp1 on 1-methyl-4-phenylpyridinium (MPP+)–induced mitochondrial fission in neurons. We observed that FLZ blocked Drp1, inhibited Drp1 enzyme activity, and reduced excessive mitochondrial fission in cultured neurons. Furthermore, by inhibiting mitochondrial fission and ROS production, FLZ improved mitochondrial integrity and membrane potential, resulting in neuroprotection. FLZ curtailed the reduction of synaptic branches of primary cultured dopaminergic neurons caused by MPP+ exposure, reduced abnormal fission, restored normal mitochondrial distribution in neurons, and exhibited protective effects on dopaminergic neurons. The in vitro research results were validated using an MPTP-induced PD mouse model. The in vivo results revealed that FLZ significantly reduced the mitochondrial translocation of Drp1 in the midbrain of PD mice, which, in turn, reduced the mitochondrial fragmentation in mouse substantia nigra neurons. FLZ also protected dopaminergic neurons in PD mice and increased the dopamine content in the striatum, which improved the motor coordination ability of the mice. These findings elucidate this newly discovered mechanism through which FLZ produces neuroprotection in PD.

DEVELOPMENT OF BALANCE IN CHILDREN PARTICIPATING IN DIFFERENT RECREATIONAL PHYSICAL ACTIVITIES

Balance, an important motor coordination ability, underlies the performance of variousmotor skills and allows for participation in common childhood activities. Research indicatesthat sport participation could support balance development in children. To confirm the aboveeffect, this study investigated whether participation in different recreational physicalactivities could induce differences in children’s balance ability. Specifically, 138 children, 5-11 years old (Mage=8.4±1.3), classified into four groups according to the activity they tookpart in (basketball, track and field, rhythmic gymnastics, contemporary dance), were assessedby the balance subset of the KörperKoordinationstest für Kinder. Pearson correlations wereapplied to detect associations of balance scores with age, height, body mass index (BMI) andshowed significant correlations for BMI (p&lt; .001). The analyses of covariance (covariate:BMI) that were computed on children’s balance scores showed statistically significant effectsof group (p&lt; .001). Sidak post hoc tests indicated that children participating in rhythmicgymnastics surpassed all three other groups; whereas those taking part in basketball had thelowest scores in almost every balance item. It seems that the type of physical activity a childparticipates in could be a significant contributor to the development of his/her balance.Physical activities encompassing the execution of various balance tasks, such as rhythmicgymnastics, seem to offer children greater opportunities to make improvements in this areacompared to children engaging in activities of low balancing requirements. This findinghighlights the potential that is available to coaches: they can contribute to children’s balancedevelopment by implementing sport-specific programs that target this human ability.

Germacrone alleviates neurological deficits following traumatic brain injury by modulating neuroinflammation and oxidative stress

BackgroundGermacrone (GM) is a terpenoid compound which is reported to have anti-inflammatory and anti-oxidative effects. However, its role in treating traumatic brain injury (TBI) remains largely unknown.MethodsMale C57BL/6 mice were divided into the following groups: control group, TBI group [controlled cortical impact (CCI) model], CCI + 5 mg/kg GM group, CCI + 10 mg/kg GM group and CCI + 20 mg/kg GM group. GM was administered via intraperitoneal injection. The neurological functions (including motor coordination, spatial learning and memory abilities) and brain edema were measured. Nissl staining was used to detect the neuronal apoptosis. Colorimetric assays and enzyme linked immunosorbent assay (ELISA) kits were used to determine the expression levels of oxidative stress markers including myeloperoxidase (MPO), malondialdehyde (MDA) and superoxide dismutase (SOD), as well as the expressions of inflammatory markers, including tumor necrosis factor α (TNF-α), interleukin-1β (IL-1β) and interleukin-6 (IL-6). Additionally, protein levels of Nrf2 and p-p65 were detected by Western blot assay.ResultsGM significantly ameliorated motor dysfunction, spatial learning and memory deficits of the mice induced by TBI and it also reduced neuronal apoptosis and microglial activation in a dose-dependent manner. Besides, GM treatment reduced neuroinflammation and oxidative stress compared to those in the CCI group in a dose-dependent manner. Furthermore, GM up-regulated the expression of antioxidant protein Nrf2 and inhibited the expression of inflammatory response protein p-p65.ConclusionsGM is a promising drug to improve the functional recovery after TBI via repressing neuroinflammation and oxidative stress.

Music primarily affects the auditory analyzer in children, helps to increase auditory attention in children with dementia. Thus, the child perceives sounds better over time and develops the ability to differentiate them. Improved hearing allows children to understand the speech of adults better. Over time, it accelerates the process of sound imitation, allows the child to pronounce the initial sounds and syllables. Musical-rhythmic is based on the combination of sounds with movements, which leads to the improvement of children's motor skills. Nrf2 knockout altered brain iron deposition and mitigated age-related motor dysfunction in aging mice

The transcription factor NF-E2-related factor 2 (Nrf2) is a central regulator of cellular antioxidant and detoxification response. The association between Nrf2 activity and iron-related oxidative stress in neurodegenerative diseases has been studied, and Nrf2 was found to transcriptionally regulate the expression of iron transporters and ferroptosis-related factors. However, the role of Nrf2 in age-related motor dysfunction and its link to iron metabolism dysregulation in brain have not been fully elucidated. In this study, with different ages of Nrf2 knockout (KO) and wild type (WT) mice, we investigated the effects of Nrf2 deficiency on brain oxidative stress, iron metabolism and the motor coordination ability of mice. In contrast to the predicted neuroprotective role of Nrf2 in oxidative stress-related diseases, we found that Nrf2 KO remarkably improved the motor coordination of aged mice, which was associated with the reduced ROS level and decreased apoptosis of dopaminergic neurons in substantia nigra (SN) of 18-month-old Nrf2 KO mice. With high-iron and Parkinson's disease (PD) mouse models, we revealed that Nrf2 KO prevented the deposition of brain iron, particularly in SN and striatum, which may subsequently delay motor dysfunction in aged mice. The regulation of Nrf2 KO on brain iron metabolism was likely mediated by decreasing the ferroportin 1 (FPN1) level on brain microvascular endothelial cells, thus hindering the process of iron entry into the brain. Nrf2 may be a potential therapeutic target in age-related motor dysfunction diseases for its role in regulating brain iron homeostasis.

Functional Development of Children 6–7 years old with Different Levels of Informatization of Living Conditions

Currently, world science is actively conducting research aimed at analyzing the influence of the modern digital environment on motor fitness, physical development and functional capabilities of preschoolers. Despite the available data, the problem under consideration remains poorly understood, especially in relation to preschool children. The aim of the study is to identify the features of the functional development of children 6–7 years old, taking into account the level of informatization of living conditions. Research methodology. The study involved practically healthy children 6–7 years old. To assess the information load of children when using digital technologies, a modified index of informatization of living conditions was used. The set of physical fitness tests included control exercises to assess speed, speed-strength, coordination motor abilities, flexibility, general and strength endurance. According to the well-known formulas, a double product, Kerdo’s vegetative index, Myznikov’s index, an indicator of the body’s adaptive potential and body mass index were determined. Results. The data obtained indicate that boys, on average, are characterized by a higher indicator of informatization of living conditions in comparison with girls. The differences were revealed, determined by the degree of informatization of living conditions, in terms of general and strength endurance, speed, speed-strength and coordination abilities, physical development, as well as the functional capabilities of the organism. The results of the study give grounds to believe that an adequate mode of physical education, ensuring the satisfaction of the biological needs of children for physical activity, may be the most important factor in increasing their motor fitness, preventing adverse changes in functional development with the active use of digital technologies in everyday life. Conclusions. Analysis of the influence of digital technologies on the functional development of children aged 6–7 years has shown that the features of motor fitness, functional capabilities of the body and physical development of boys and girls aged 6-7 years depend on the level of informatization of living conditions. It turned out that with an increase in the time of using digital technologies in the daily routine, there is a decrease in the considered indicators of the morphological and functional development of children.
